In this study several oil press cakes were investigated as exclusive substrates for different moulds and yeasts for the production of flavor‐active decalactones via solid‐state fermentation (SSF). Experiments are focused on pre‐treatment methods for olive cake to remove antimicrobial phenolic substances contained in the oil cake disturbing or even inhibiting microbial growth. Choosing Ceratocystis moniliformis as the reference microorganism best results were obtained by a combination of hot water flushes and enzymatic treatment of the cake. Fermentation with sunflower, olive and linseed cake did not lead to lactone formation in detectable amounts although all of these substrates provided good microbial growth. On castor cake, however, five microorganisms have synthesized the requested decalactones via SSF. Thus fermentation of the fungus Moniliella suaveolens on this substrate resulted in a maximum concentration of γ‐decalactone of 180.7 mg·kg?1 dry matter without optimization. Another δ‐lactone, 6‐pentyl‐α‐pyrone, was produced in small amounts by the fungus Trichoderma harzianum on castor cake.  相似文献

A Box–Behnken experimental design was used to investigate the effects of five factors—ie oxygen content in the gas phase; concentrations of C, N and P; and fermentation time—on the concentrations of biomass and lovastatin produced in batch cultures of Aspergillus terreus. The values of the various factors in the experiment ranged widely, as follows: 20–80% (v/v) oxygen in the aeration gas; 8–48 g dm?3 C‐concentration; 0.2–0.6 g dm?3 N‐concentration; 0.5–2.5 g dm?3 phosphate‐concentration; and 7–11 days fermentation time. No previous work has used statistical analysis in documenting the interactions between oxygen supply and nutrient concentrations in lovastatin production. The Box–Behnken design identified the oxygen content in the gas phase as the principal factor influencing the production of lovastatin. Both a limitation and excess of oxygen reduced lovastatin titers. A medium containing 48 g dm?3 C supplied as lactose, 0.46 g dm?3 N supplied as soybean meal, and 0.79 g dm?3 phosphate supplied as KH2PO4, was shown to support high titers (~230 mg dm?3) of lovastatin in a 7‐day fermentation in oxygen‐rich conditions (80% v/v oxygen in the aeration gas). Under these conditions, the culture medium had excess carbon but limiting amounts of nitrogen. The optimized fermentation conditions raised the lovastatin titer by four‐fold compared with the worst‐case scenario within the range of factors investigated. BACKGROUND: Succinic acid is a valuable four‐carbon organic chemical with applications in many fields. It was found that cell mass was an important factor in succinic acid production by metabolically engineered Escherichia coli strains. In this work, high cell density fermentation was investigated for succinic acid production by a metabolically engineered strain SD121 with ldhA, pflB, ptsG mutation and heterogenous cyanobacterial ppc overexpression. RESULTS: Under two‐stage cultivation, the controlled DO feeding strategy during the aerobic growth phase facilitated biomass up to a dry cell weight of 19.6 g L?1, and enhanced succinic acid production in the following anaerobic fermentation phase to a concentration of 116.2 g L?1. A near theoretical maximum succinic acid yield of 1.73 mol mol?1 glucose was achieved with an average productivity of 1.55 g L?1 h?1. CONCLUSION: The results indicated the potential advantage of high cell density fermentation for improvement of succinic acid production by E. coli. Tomography, an efficient nonintrusive technique, was employed to visualize the flow in continuous‐flow mixing and to measure the cavern volume (Vc) in batch mixing. This study has demonstrated an efficient method for flow visualization in the continuous‐flow mixing of opaque fluids using two‐dimensional (2‐D) and 3‐D tomograms. The main objective of this study was to explore the effects of four inlet‐outlet configurations, fluid rheology (0.5–1.5% xanthan gum concentration), high‐velocity jet (0.317–1.660 m s?1), and feed flow rate (5.3 × 10?5?2.36 × 10?4 m3 s?1) on the deformation of the cavern. Dynamic tests were also performed to estimate the fully mixed volume (Vfully mixed) for the RT, A310, and 3AM impellers in a continuous‐flow mixing system, and it was found that Vfully mixed was greater than Vc. Incorporating the findings of this study into the design criteria will minimize the extent of nonideal flows in the continuous‐flow mixing of complex fluids and eventually improve the quality of end‐products. © 2013 American Institute of Chemical Engineers AIChE J, 60: 315–331, 2014  相似文献

BACKGROUND: The use of olive‐oil mill wastewater (OMW) from a three‐phase centrifugation process used in the olive‐oil industry, has been studied in relation to the production of the microalga Scenedesmus obliquus CCAP 276/3A. The chemical characteristics of OMW indicated nitrogen deficiency. RESULTS: S. obliquus is able to assimilate nutrients present in a culture medium (water‐OMW 5%) and grow at its maximum specific growth rate of 0.026 h?1, both under mixotrophic as well as heterotrophic conditions. The different daily doses of light (DDL) used, in the range 0–36 E m?2 d?1, determined light‐limited and light‐inhibited cultures. The light‐inhibited mixotrophic cultures bore characteristics similar to those of the heterotrophic cultures, and became more so when the dose of light received was higher. The low protein yield (258 mg g?1) and high percentage of carbohydrates of the biomass (65.8%) confirmed a nutritional‐stress situation associated with nitrogen limitation. CONCLUSION: The similarity between the fatty‐acid composition of the heterotrophic and mixotrophic cultures strongly inhibited by light appeared to indicate the cancelling of the photosynthetic behaviour of the cells at high DDL values. The biomass generated can be used for biofuels. The maximum elimination of biological oxygen demand (BOD5) per unit of biomass was achieved in the heterotrophic cultures. New data are presented on drop size distribution at high dispersed phase fractions of organic‐in‐water mixtures, obtained with a light back scattering technique (3 Dimensional Optical Reflectance Measurement technique, 3D ORM). The 3D ORM technique, which provides fast, in‐situ and on‐line drop distribution measurements even at high concentrations of the dispersed phase, is validated using an endoscope attached to a high‐speed video recorder. The two techniques compared favourably when used in a dispersion of oil (density (ρ) = 828 kg m?3, viscosity (µ) = 5.5 mPa s, interfacial tension (σi) = 44.7 mN m?1) in water for a range of 5–10% dispersed phase fractions. Data obtained with the ORM instrument for dispersed phase fractions up to 60% and impeller speeds 350–550 rpm showed a decrease in the maximum and the Sauter mean drop diameters with increasing impeller speed. Phase fractions did not seem to significantly affect drop size. Both techniques showed that drop size distributions could be fitted by the log‐normal distribution. Textile (eg cotton) finishing industry wastewater is characterised by high concentrations of surfactants (up to 2 g dm?3) and of readily biodegradable biopolymers (COD 5–15 g dm?3). The anionic surfactant decyl sulfate (DS) was chosen as model surfactant and soluble starch (size) as model compound for the readily biodegradable fraction of the wastewater. Twenty‐two batch experiments with increasing DS/biomass ratio (and starch/biomass ratio) were started simultaneously. Biomass concentrations ranged from 50 to 15 000 mg dm?3. Minor inhibition effects were found for the surfactant degradation itself at all DS/biomass ratios (maximum biodegradation rate 7.7 mgDS gbiomass?1 h?1). The starch hydrolysis started without a lag‐phase at DS/biomass ratios of up to 0.15 gDS gbiomass?1. The lag‐phase was prolonged to about 100 h at a very high DS/biomass ratio (3 gDS gbiomass?1). The relative importance of the accumulated intermediates was dependent on the DS/biomass ratio. Above 0.3 gDS gbiomass?1 10% of the substrate organic carbon accumulated as ethanol, but no ethanol accumulation was observed at low DS/biomass ratios. Moderate DS/biomass ratios caused a considerable delay of the methanogenesis; high DS/biomass ratios prevented the methanogenesis almost completely. © 2002 Society of Chemical Industry  相似文献

BACKGROUND: An unstructured mathematical model was developed to understand information on the relationship between Bacillus circulans growth and metabolism‐related protease production (using logistic and Luedeking–Piret equations respectively) in a batch reactor with respect to glucose consumption and fermentation time. The objective was to develop an indispensable tool for the optimisation, control, design and analysis of alkaline protease production. RESULTS: Biomass growth and enzyme production titres changed with a change in substrate concentration. Modelling analysis of biomass and enzyme production titres at different substrate concentrations revealed significant accuracy in terms of statistical consistency and robustness with respect to fermentation kinetic profiles. CONCLUSION: With the B. circulans strain used, an economic protease yield (2837 × 103 U g?1) with respect to biomass and glucose ratio was achieved at low substrate concentration (10 g L?1). The developed model could be effectively utilised for designing, controlling and up‐scaling the protease production process in high‐density fermentation in selected bioreactors with statistical consistency. A new chitosan molecular‐imprinted adsorbent was prepared from the mycelium of waste biomass. The results showed that an adsorbent using Penicillium chrysogenum mycelium as the core material was better than one derived from peanut coat. The adsorption capacity of the surface‐imprinted adsorbent for Ni2+ was enhanced by increasing the chitosan concentration in the imprinting process. Epichlorohydrin was better than glutaraldehyde as a cross‐linking agent; the optimal imprinted Ni2+ concentration for preparing the surface‐imprinted adsorbent was 2 mg (Ni2+) g?1 of mycelium. The adsorption capacity of the surface‐imprinted adsorbent was 42 mg g?1 (at 200 mg dm?3 initial metal ions concentration) and twice that of the mycelium adsorbent. Increasing population has resulted in overexploitation of conventional seeds. The limited supply of water and salinization of agricultural lands are threats to crop production. This creates food insecurity and results in ever‐increasing prices of crops and edible oils. Halophytes that produce high‐quality seeds can serve as sources of oil and edible products. We analyzed the chemical composition and antioxidant activity of seeds from 5 halophytic grasses, i.e., Aeluropus lagopoides, Eragrostis ciliaris, Eragrostis pilosa, Panicum antidotale, and Sporobolus ioclados. These seeds contained crude protein (10–29%), carbohydrates (32–55%), crude fiber (4–21%), minerals (3.8–9.2%), and oil (4–11%), indicating their nutritional potential. Oils of these seeds had suitable fatty‐acid composition with 62–82% unsaturation and only 17–24% saturation. Out of this, 91–94% of the total oil constituted by linoleic, oleic, and palmitic acids. High contents of total phenols (2.8–4.2 mg gallic acid equivalent [GAE] g?1), flavonoids (0.5–1.3 mg Quercetin equivalent [QE] g?1), and tannins (0.3–1.3 mg catechin equivalent [CE] g?1) supported their high antioxidant activity (1,1‐Diphenyl‐2‐picryl‐hydrazyl (DPPH) activity in terms of half maximal inhibitory concentration‐IC50 1.1–5.86 mg mL?1; 2,2′‐azino‐bis3‐ethylbenzothiazoline‐6‐sulphonic acid (ABTS) 18.8–72.8 mmol Trolox g?1; ferric‐reducing antioxidant power 2.0–4.4 mmol Fe+2 g?1). The reverse phase‐high‐pressure liquid chromatography analysis identified the presence of bioactive phenolic antioxidants (mainly gallic acid, chlorogenic acid, coumaric acid, ferulic acid, kaempferol, and quercetin). Due to these characteristic composition and salt tolerability, these plants can serve as potential sources of industrial raw materials for food, edible oil, phytochemicals, and oliochemicals.  相似文献

A perfluorocarbon (PFC), namely perfluorodecalin, was added to fermentation medium to increase the medium's oxygen solubility. The antibiotic concentration obtained in the absence of PFC was 45 mg dm?3, whereas it was 90 mg dm?3 in the presence of 10% (v/v) PFC. On the other hand, biomass concentration decreased from 5.7 kg m?3 to 2.9 kg m?3 by adding 10% PFC. The use of PFC in the fermentation medium also reduced the formation of mycelial pellets. The values of the mass transfer coefficient, kLa, measured in the medium with PFC were found to be in the range of 122–175 h?1 during the active growth phase which were two to three times higher than those in the medium containing no PFC. Furthermore, the maximum oxygen uptake rates obtained at the stationary phase with and without PFC were 7 mmol dm?3 h?1 and 4.9 mmol dm?3 h?1, respectively. The actual effect of PFC on actinorhodin fermentation was demonstrated by applying different operational strategies to the system. © 2001 Society of Chemical Industry  相似文献

Reduced graphene oxide nanosheets modified by conductive polymers including polythiophene (GPTh), polyaniline (GPANI) and polypyrrole (GPPy) were prepared using the graphene oxide as both substrate and chemical oxidant. UV–visible and Raman analyses confirmed that the graphene oxide simultaneously produced the reduced graphene oxide and polymerized the conjugated polymers. The prepared nanostructures were subsequently electrospun in mixing with poly(3‐hexylthiophene) (P3HT)/phenyl‐C71‐butyric acid methyl ester (PC71BM) and poly[bis(triisopropylsilylethynyl)benzodithiophene‐bis(decyltetradecylthien)naphthobisthiadiazole] (PBDT‐TIPS‐DTNT‐DT)/PC71BM components and embedded in the active layers of photovoltaic devices to improve the charge mobility and efficiency. The GPTh/PBDT‐TIPS‐DTNT‐DT/PC71BM devices demonstrated better photovoltaic features (Jsc = 11.72 mA cm?2, FF = 61%, Voc = 0.68 V, PCE = 4.86%, μh = 8.7 × 10?3 cm2 V–1 s?1 and μe = 1.3 × 10?2 cm2 V–1 s?1) than the GPPy/PBDT‐TIPS‐DTNT‐DT/PC71BM (Jsc = 10.30 mA cm?2, FF = 60%, Voc = 0.66 V, PCE = 4.08%, μh = 1.4 × 10?3 cm2 V–1 s?1 and μe = 8.9 × 10?3 cm2 V–1 s?1) and GPANI/PBDT‐TIPS‐DTNT‐DT/PC71BM (Jsc = 10.48 mA cm?2, FF = 59%, Voc = 0.65 V, PCE = 4.02%, μh = 8.6 × 10?4 cm2 V–1 s?1 and μe = 7.8 × 10?3 cm2 V–1 s?1) systems, assigned to the greater compatibility of PTh in the nano‐hybrids and the thiophenic conjugated polymers in the bulk of the nanofibers and active thin films. The volumetric mass transfer coefficient kLa in a 0.1 m‐diameter bubble column was studied for an air‐slurry system. A C9‐C11 n‐paraffin oil was employed as the liquid phase with fine alumina catalyst carrier particles used as the solid phase. The n‐paraffin oil had properties similar to those of the liquid phase in a commercial Fischer‐Tropsch reactor under reaction conditions. The superficial gas velocity UG was varied in the range of 0.01 to 0.8 m/s, spanning both the homogeneous and heterogeneous flow regimes. The slurry concentration ?S ranged from 0 to 0.5. The experimental results obtained show that the gas hold‐up ?G decreases with an increase in slurry concentration, with this decrease being most significant when ?S < 0.2. kLa/?G was found to be practically independent of the superficial gas velocity when UG > 0.1 m/s is taking on values predominantly between 0.4 and 0.6 s–1 when ?S = 0.1 to 0.4, and 0.29 s–1, when ?S = 0.5. This study provides a practical means for estimating the volumetric mass transfer coefficient kLa in an industrial‐size bubble column slurry reactor, with a particular focus on the Fischer‐Tropsch process as well as high gas velocities and high slurry concentrations.  相似文献

Thin films of different thicknesses were prepared through glow discharge of 2‐(diethylamino)ethyl methacrylate (DEAEMA) using a capacitively coupled reactor. Current density–voltage (J–V) characteristics for plasma polymerized (PP) DEAEMA thin films of thicknesses 100, 200, 250, and 300 nm in aluminum/PPDEAEMA/aluminum sandwich configuration were studied over the temperature range from 298 to 423 K. JV curves reveal that in the low‐voltage region, the conduction current obeys Ohm's law while in the high‐voltage region the behavior attributed to be space charge‐limited conduction in PPDEAEMA thin films. The carrier mobility was calculated to be about 6.80 × 10?19 to 2.38 × 10?18 m?2 V?1s?1 for various thicknesses. The free carrier density was found to be about 1.78 × 1023 to 2.04 × 1023 m?3, and the trap density was found to be about 6.93 × 1023 to 15.9 × 1023 m?3 for different thicknesses. The activation energies were estimated to be about 0.005–0.016 eV for 2 and 30 V of PPDEAEMA thin films of different thicknesses. The low‐activation energies indicate that the thermally activated hopping conduction is operative in PPDEAEMA thin films. POLYM. ENG. Morphological and photovoltaic stabilities of poly(3‐hexylthiophene) (P3HT):phenyl‐C61‐butyric acid methyl ester (PC71BM) solar cells were investigated in pristine and modified states. To this end, four types of patterned/assembled nanostructures, namely reduced graphene oxide (rGO)‐g‐poly(3‐dodecylthiophene)/P3HT patched‐like pattern, rGO–polythiophene/P3HT/PC71BM nanofiber, rGO‐g‐P3HT/P3HT cake‐like pattern and supra(polyaniline (PANI)‐g‐rGO/P3HT), were designed on the basis of rGO and various conjugated polymers. Intermediately covered rGO nanosheets by P3HT crystals (supra(PANI‐g‐rGO/P3HT)) performed better than sparsely (patched‐like pattern) and fully (cake‐like pattern) covered ones in P3HT:PC71BM solar cell systems. Supra(PANI‐g‐rGO/P3HT) nanohybrids largely phase‐separated in active layers (root mean square = 0.88 nm) and also led to the highest performance (power conversion efficiency of 5.74%). The photovoltaic characteristics demonstrated decreasing trends during air aging for all devices, but with distinct slopes. The steepest decreasing plots were obtained for the unmodified P3HT:PC71BM devices (from 1.77% to 0.28%). The two supramolecules with the most ordered structures, that is, cake‐like pattern (10.12 mA cm?2, 51%, 0.58 V, 2.2 × 10?6 cm2 V?1 s?1, 4.3 × 10?5 cm2 V?1 s?1, 0.69 nm and 2.99%) and supra(PANI‐g‐rGO/P3HT) (12.51 mA cm?2, 57%, 0.63 V, 1.2 × 10?5 cm2 V?1 s?1, 3.4 × 10?4 cm2 V?1 s?1, 0.82 nm and 4.49%), strongly retained morphological and photovoltaic stabilities in P3HT:PC71BM devices after 1 month of air aging. According to the morphological, optical, photovoltaic and electrochemical results, the supra(PANI‐g‐rGO/P3HT) nanohybrid was the best candidate for stabilizing P3HT:PC71BM solar cells. © 2020 Society of Chemical Industry  相似文献

The effect of aeration rate on pullulan production and the rheological properties of the fermentation broth in an airlift reactor was investigated. An airlift fermenter was shown to be an appropriate fermentation system for the production of pullulan. A maximum pullulan concentration (30 kg m?3), biomass concentration (6.0 kg m?3), pullulan yield (60%, w/w) and sugar utilization (100%, w/w) was obtained at an aeration rate of 2 vvm. The mycelium and the yeast‐like cells were the morphological forms responsible for pullulan production. The highest polysaccharide concentration was obtained when the mycelial forms and the yeast‐like cells were 60% and 40% (w/w) of the total biomass, respectively. The apparent viscosity of the broth was increased with the increase of the aeration rate from 1 to 2 vvm and then decreased at higher vvms. On the other hand, the dissolved oxygen concentration and the volumetric mass transfer coefficient continually increased with the increase of the aeration rate. The mycelial forms and the production of extracellular polysaccharide were responsible for the non‐Newtonian flow behaviour of the fermentation broth. The rheological behaviour can be characterized by a power law type of equation. The relationship between shear rate/shear stress and shear rate/apparent viscosity showed a non‐Newtonian behaviour of the fermentation broth. © 2001 Society of Chemical Industry  相似文献

A pilot‐plant membrane bioreactor (MBR) and two pilot‐plant hybrid moving bed biofilm reactor–membrane bioreactors (MBBR–MBRs), divided into three aerobic and one anoxic chambers, were started up for the treatment of salinity‐amended urban wastewater. The MBBR–MBR systems worked with and without carriers in the anoxic zone (MBBR–MBRanox and MBBR–MBRn/anox, respectively). The systems were operated from start‐up to stabilization, showing high removal of organic matter—a maximum of 90% chemical oxygen demand and 98% biochemical oxygen demand on the fifth day for MBBR–MBRn/anox in the stabilization phase—but low nitrogen elimination—30% maximum for MBBR–MBRn/anox in the stabilization phase. Biofilm attached to carriers reached less than 50 mg L?1 in the hybrid system. MBR showed faster kinetics than the two MBBR–MBR systems during start‐up, but the opposite occurred during stabilization. In the present work, water and three phase compositions of Solka-Floc, a cellulose fiber for simulating the biomass in bacteria, yeast, and fungal fermentation were studied in a 1.4?m3 stirred airlift reactor. The fractional dispersed phase holdup and the overall volumetric mass transfer coefficients were measured. The dispersed phase riser gas holdup and overall volumetric mass transfer coefficients both increased with increasing riser superficial dispersed phase velocity (0.02–0.1?ms?1) and agitator speed in the range of 0–5?rs?1. An increase in the Solka-Floc concentration (1–3% w/v) was found to reduce ?GR and K L a L . Empirical correlations have been developed for fractional dispersed phase gas holdup and overall volumetric mass transfer coefficients.  相似文献

This study reports on the effects of internal fermenter and external in‐line agitation and fed‐batch mode of operation on citric acid production from Candida lipolytica using n‐paraffin as the carbon source. An optimum range of fermenter agitation speeds in the range 800–1000 rpm corresponding to Reynolds numbers of 50433–62947 (based on initial batch conditions) seemed to give the best balance between substrate utilization for biomass growth and citric acid production. Proof of concept evidence is presented that indicates that an external in‐line agitator could be used in place of high speed internal agitation to increase citric acid production. However, more work is required to optimize the external agitator concept. Application of multiple fed‐batch feedings can be used to extend the batch fermentation and increase final citric acid concentrations and product yield. Experiments were conducted implementing a three‐cycle fed‐batch process which increased overall citric acid yields to 0.8–1.0 g citric acid g?1 n‐paraffin, approximately 200% improvement from those found in the normal batch process. The three‐cycle fed‐batch mode of operation also increased the final citric acid concentration to 42 g dm?3 from about 6 g dm?3 for normal batch operation. Injected chemical flooding systems with high salinity tolerance and fast‐dissolving performance are specially required for enhancing oil recovery in offshore oilfields. In this work, a new type of viscoelastic‐surfactant (VES) solution, which meets these criteria, was prepared by simply mixing the zwitterionic surfactant N‐hexadecyl‐N,N‐dimethyl‐3‐ammonio‐1‐propane sulfonate (HDPS) or N‐octyldecyl‐N,N‐dimethyl‐3‐ammonio‐1‐propane sulfonate (ODPS) with anionic surfactants such as sodium dodecyl sulfate (SDS). Various properties of the surfactant system, including viscoelasticity, dissolution properties, reduction of oil/water interfacial tension (IFT), and oil‐displacement efficiency of the mixed surfactant system, have been studied systematically. A rheology study proves that at high salinity, 0.73 wt.% HDPS/SDS‐ and 0.39 wt.% ODPS/SDS‐mixed surfactant systems formed worm‐like micelles with viscosity reaching 42.3 and 23.8 mPa s at a shear rate of 6 s?1, respectively. Additionally, the HDPS/SDS and ODPS/SDS surfactant mixtures also exhibit a fast‐dissolving property (dissolution time <25 min) in brine. More importantly, those surfactant mixtures can significantly reduce the IFT of oil–water interfaces. As an example, the minimum of dynamic‐IFT (IFTmin) could reach 1.17 × 10?2 mN m?1 between the Bohai Oilfield crude oil and 0.39 wt.% ODPS/SDS solution. Another interesting finding is that polyelectrolytes such as sodium of polyepoxysuccinic acid can be used as a regulator for adjusting IFTmin to an ultralow level (<10?2 mN m?1). Taking advantage of the mobility control and reducing the oil/water IFT of those surfactant mixtures, the VES flooding demonstrates excellent oil‐displacement efficiency, which is close to that of polymer/surfactant flooding or polymer/surfactant/alkali flooding. Our work provides a new type of VES flooding system with excellent performances for chemical flooding in offshore oilfields.  相似文献
