Effects of delta-aminolevulinic acid dehydratase (ALAD) polymorphisms on the renal and neurobehavioral functions were investigated in Chinese workers from a storage battery plant exposed to inorganic lead. Blood and urine were collected from each worker to determine the ALAD genotypes, blood lead levels (PbB), urinary β2-MG and urinary NAG activity. The World Health Organization Neurobehavioral Core Test Battery (WHO-NCTB) was used. Of the 135 lead workers tested for ALAD genotype, 126 were ALAD1-1, 9 were ALAD1-2 but none were ALAD2-2. The gene frequencies of ALADl-1 and ALADl-2 were 93.33% and 6.67%, respectively. The workers with ALAD1-2 genotype had significantly higher concentrations of PbB (62.52 µg/dl vs. 41.02 µg/dl), urinary NAG (22.01 U/gCr vs. 13.49 U/gCr), urinary β2-MG (194.98 μg/gCr vs. 112.88 μg/gCr), and digit span backward (DSB) score (6.67 vs. 5.33) than those of ALAD1-1 genotype. Urinary NAG of ALAD1-2 genotype carriers was significantly higher than that of ALAD1-1 genotype under the same blood lead level (bi 0.75 vs. bi 0.29). Interaction between PbB and ALAD genotypes has a significant influence on NAG (= 0.02) and β2-MG (= 0.01). It is postulated that the workers with the ALAD2 allele appear to be more susceptible to the effects of lead on renal injury, whereas neurobehavioral functions in ALAD1 homozygote tend to be more vulnerable.  相似文献

Lead (Pb) levels, delta-aminolevulinic acid dehydratase (ALAD) activities, zinc protoporphyrin (ZPP) levels in blood, and urinary delta-aminolevulinic acid (ALA) and coproporphyrin (CP) concentrations were measured for 318 environmental Pb exposed children recruited from an area of southeast China. The mean of blood lead (PbB) levels was 75.0 μg/L among all subjects. Benchmark dose (BMD) method was conducted to present a lower PbB BMD (lower bound of BMD) of 32.4 μg/L (22.7) based on ALAD activity than those based on the other three haematological indices, corresponding to a benchmark response of 1%. Childhood intelligence degrees were not associated significantly with ALAD activities or ALA levels. It was concluded that blood ALAD activity is a sensitive indicator of early haematological damage due to low-level Pb exposures for children.  相似文献

The refining of silver from old silver ornaments, articles and jeweller's waste by smelting these with lead scraps for the fabrication of new jewellery is an important small scale industry in India. The present survey and clinical investigations have shown that 31 out of 50 silver refiners with a mean blood lead level of 32.84+/-1.78 microg/dl (range 20.3-64.9), decrease in blood delta-aminolevulinic acid dehydratase (ALAD) activity and thiamine (as pyruvate) level and an enhanced urinary excretion of ALA as compared to control, were suffering from lead poisoning. Most of these workers have shown anaemia, abdominal colic, blue lining of gum and muscular wasting indicative of lead toxicity. Twenty-four workers with relatively high blood lead levels were equally divided into two groups and given either vitamin B1 (75 mg, once a day) or vitamin C (250 mg. twice a day) for 1 month. The treatment with both the vitamins significantly lowered the blood lead levels and reduced blood thiamine and copper deficiency. In addition, vitamin C was also effective in reversing the inhibition of blood ALAD activity while the effect of vitamin B1 on its activity was marginal. The daily intake of vitamin B1 and vitamin C may prevent the accumulation of lead and reduce its toxic effects particularly in those regularly exposed to lead.  相似文献

The behaviour of blood (PbB) and two biological indicators of effect: erythrocyte protoporphyrin (EP) and urinary aminolaevulinic acid (ALAU), were studied in subjects who had ceased working with inorganic lead for at least one year. The relationships between these indices and chelatable lead (PbU-EDTA) were also analysed. As a comparison, a group of subjects currently exposed to inorganic lead was studied. At a given PbU-EDTA level, the PbB values were significantly lower in the workers with past exposure than those found in currently exposed subjects. The relationship between EP and PbU-EDTA illustrates that, for corresponding levels of chelatable lead, the values of the erythrocyte metabolite are identical in the two groups. Considering the EP-PbB relationship, however, at a given PbB level the protoporphyrin values appear distinctly more altered in the subjects with past exposure. The relationship between ALAU and PbU-EDTA shows that, at a given PbU-EDTA level the urinary metabolite in past exposed subjects is distinctly lower than in currently exposed workers, while the relationship between ALAU and PbB shows that, at similar blood lead values, the ALAU levels are identical in both groups. On the basis of these results, it is concluded that in subjects with past exposure, erythrocyte protoporphyrin can be used in establishing the persistence and extent of an "active deposit" of lead in the organism, while PbB and ALAU are of very limited value.  相似文献

Biological monitoring of lead effects in specific population groups living in the vicinity of a lead smelter has been followed up after effective emission control, in order to estimate the length of time necessary before the characteristic indicators of increased lead absorption normalize. The period established could be a realistic basis for predicting the time necessary to elapse before a lead contaminated area can be proclaimed as safe for normal living. Three groups of subjects were examined: preschool children, school children and mothers. The biological monitoring included the measurement of δ-aminolevulinic acid dehydratase activity (ALAD) and the concentration of zinc-protoporphyrin (ZPP), blood lead (Pb-B) and hemoglobin (Hb). The results obtained demonstrated a gradual normalization of the characteristic indicators of lead absorption, although they were still outside corresponding values in the control group even four years after the installation of effective emission control. These findings emphasize the advantages of biological monitoring over environmental for objective assessment of internal and external lead exposure following effective engineering control of the emission source.  相似文献

Secondary copper smelters, which primarily utilize the waste materials that contain organic impurities, and the zinc recovery plant, which handles mostly fly ash and slag from the iron and steel industry, are major emission sources of polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDD/Fs) in Taiwan. In this study, we compared the levels of erythrocyte glutathione (GSH), erythrocyte superoxide dismutase (SOD) and plasma malondialdehyde (MDA) in workers at a secondary copper smelting plant and a zinc recovery plant who may have been exposed to PCDD/Fs. Though the PCDD/F levels were higher in workers of zinc recovery plant than those of secondary copper smelting plant, no significant difference was found for serum PCDD/F levels between the two kinds of plants. We observed a significant difference in plasma MDA levels between workers at the zinc recovery plant (2.54 microM) and those at the copper smelting plant (1.79 microM). There was and a significant positive correlation between plasma MDA levels and the PCDD/Fs levels. In addition, we observed that the MDA levels were not affected by smoking and exercise status. Therefore, the data suggest that the MDA levels of the metal recovery workers are influenced by their PCDD/F exposure. The erythrocyte SOD activity in workers from the zinc recovery plant was marginally higher than that from the secondary copper plant (196 vs. 146 units/ml, p<0.06). In both plants, large variations in the MDA and SOD levels were found, especially in the high-PCDD/Fs-exposure group, which may be attributed, at least partially, to the differences in smoking status and the number of cigarettes smoked. Overall, our results indicate a higher oxidative stress in workers of the zinc recovery plant than in workers of the secondary copper smelting plant in Taiwan.  相似文献

In an observational cross-sectional design, a sample of 292 children aged 6-11 years from two primary schools around a shipyard area, known to be an area contaminated with lead (from the industry), were examined to verify the cariogenicity of lead. The number of decayed and filled surfaces on deciduous teeth (dfs), and the number of decayed, missing, and filled surfaces on permanent teeth (DMFS), the salivary flow rate, pH, buffer capacity, oral hygiene, Lactobacillus spp. and mutans streptococci counts were recorded. The mean (range) of DMFS and dfs were respectively 1.3 (range 0-17) and 13.2 (range 0-45); and the geometric mean blood lead level (PbB) and SD were 7.2 and 1.5 microg/dl. The level of dfs, but not DMFS correlated with the blood lead level (R(s)=0.25, p=0.00 / R(s)=0.09, p=0.14). The odds ratio for DMFS>or=1 and dfs>5 for a doubling of PbB after adjusting for other factors were 1.28 (95%CI, 0.81-2.04; p value=0.35) and 2.39 (95%CI, 1.36-4.20; p value=0.004), respectively. The cariogenicity of lead is evident in deciduous teeth but not in permanent teeth for this age group.  相似文献

Lead is pervasive environmental pollutant with potential public health hazard as a contaminant of food from animal origin. The present study examines the blood and milk lead level in animals reared in areas around different industrial activities and to find out correlation between blood and milk lead levels in lactating cows. Blood and milk samples (n = 149) were collected from animals reared around steel processing unit (n = 22), lead-zinc smelter (n = 21), aluminum processing plant (n = 25), rock phosphate mining area cum phosphate fertilizer plant (n = 21), coal mining areas (n = 46) and closed lead but functional zinc smelter (n = 14). Samples were also collected from randomly chosen 52 lactating cows reared in non-polluted areas to serve as controls. Significantly (P < 0.05) higher blood lead level was recorded in animals reared around lead-zinc smelting factories followed by closed lead but functional zinc smelter, aluminum processing unit and steel manufacturing plant, as compared to values recorded for control animals. The highest milk lead level (0.84 +/- 0.11 microg/ ml) was detected in animals reared in the vicinity of lead-zinc smelting unit followed by aluminum processing plant and steel processing unit. Analysis of correlation between blood lead levels and lead excretion in milk through sorting the blood lead values into 9 different ranges irrespective of site of collection of samples (n = 201) revealed significant correlation (r = 0.469 at P < 0.01) between blood and milk lead concentrations. The lactating cows with blood lead levels above 0.20 microg/ml (groups 5-9) had significantly (P < 0.05) higher milk lead excretion than those with blood lead levels from non detectable to 0.20 microg/ml (groups 1-4). Pearson correlation analysis between blood and milk lead concentrations in 122 animals with blood lead <0.20 microg/ml showed non-significant correlation (r = 0.030 at P < 0.05) but a significant correlation was observed between these two parameters with blood lead level above >0.02 microg/ml indicating that the excretion of lead through milk increases with the increased in blood lead level above 0.20 microg/ml.  相似文献

As part of the assessment of a site in northern France polluted by metals from two smelters (in particular, lead, cadmium and mercury), a cross-sectional study was carried out which intended to estimate the levels of the lead burden of the adult population living on the site and the factors associated with these levels. The exposed zone included 10 municipalities in the Nord-Pas de Calais region, located in the vicinity of two non-ferrous metal smelters. The soils in these municipalities contained between 100 and 1700 ppm of lead. The non-polluted zone contained 20 municipalities from the same region, drawn randomly from those in the region of comparable size but free from any industrial lead exposure. The adult study population (301 men and 300 women) was stratified according to age, sex, employment status and exposure level. The inclusion criteria required subjects who were aged between 20 and 50 years and had been living in the exposed zone for at least 8 years; the exclusion criteria were pregnancy, cancer, kidney disease and diabetes. No more than 10% of the subjects participating could work at one of the two smelters. Data collection took place at home; visiting nurses interviewed subjects to complete a questionnaire and also took blood samples. The lead assay was performed by atomic absorption spectrometry. The geometric mean of the blood-lead levels was 74 microg/l, 95% CI = 69-80 among men and 49 microg/l, 95% CI = 46-53 among women. Blood-lead levels exceeding 100 microg/l were found among 30% of men and 12% of women. Several factors were associated with variation of the mean blood-lead level: the blood-lead level was significantly higher among the men for subjects living less than 1 km from the smelters (geometric mean x 1.3, 95% CI = 1.1-1.6), for those who drink alcoholic beverages (x 1.1, 95% CI = 1.0-1.2 for consumption of 30 g/day), those who smoke (x 1.2, 95% CI = 1.0-1.3 for 20 cigarettes/day), and for subjects with occupational exposure; among the women, for subjects living less than 1 km from the smelters (geometric mean x 1.5, 95% CI = 1.2-1.7), for those who drink alcohol (x 1.1, 95% CI = 1.1-1.2 for a daily consumption of 10 g), and for women living in a building constructed before 1948 (x 1.2, 95% CI = 1.0-1.4).  相似文献

The contents of the elements antimony, arsenic, cadmium, copper, lead, mercury, selenium, silver, tin and zinc in bone tissue from autopsy specimens of the femur of workers who had been exposed to a large number of metals in a smeltery and refinery in Northern Sweden as well as of a control group have been quantitatively assayed.The analytical techniques used were atomic absorption spectrophotometry, neutron activation analysis and particle induced X-ray emission analysis (PIXE) in a proton microprobe. Increased levels of lead in the bone tissue of exposed workers compared to a non-exposed control group were observed. The median level of lead in the group of exposed workers exceeded the corresponding value of the control group about 5 times.Using the proton microprobe in the PIXE-mode, concentration profiles of copper, lead and zinc were examined within the Haversian system of the bone samples.  相似文献

Lead is a natural component of aquatic ecosystems with no known biological role and is highly toxic. Its toxicity stems from its ability to mimic biologically important metals and to produce membrane damage through lipid peroxidation (LPO). Most lead poisoning symptoms are thought to occur by interfering with an essential enzyme, delta-aminolevulinic acid dehydratase (ALAD), the activity of which is markedly inhibited by lead. The purpose of this work was to study the levels and effects of lead pollution (responses of ALAD and oxidative stress biomarker LPO) in the freshwater bivalve Corbicula fluminea along the lower Guadiana River (Portugal and Spain); a major river system impacted by historic mining pollution and more recent anthropogenic inputs. The results show that the enzymatic activity of ALAD is negatively correlated with the total Pb concentration of the whole tissue suggesting that ALAD has considerable potential as a biomarker of lead exposure in C. fluminea. To identify the sources of lead to which bivalves have been exposed, high precision (206)Pb/(204)Pb, (207)Pb/(204)Pb, (208)Pb/(204)/Pb ratios for C. fluminea confirm that historical mining activities in the Iberian Pyrite Belt are the dominant source of lead pollution in the lower Guadiana River. The isotope patterns however exhibit marked seasonal and geographic variation in response to rainfall and river water management. Locally, other anthropogenic sources of lead have been detected in C. fluminea close to population centres, thus adding to its versatility as a freshwater bio-indicator. Overall, the study highlights the value of natural ecosystems as monitors of water quality and their importance for public health assessment and surveillance.  相似文献

Foreign workers employed in industries in Taiwan have been found at elevated risk of injuries. Less well known is whether the elevated risk persists in chemical exposure such as lead exposure at battery manufacturing. A cohort of 70 Thai workers and 55 native workers employed at a battery plant were followed up, after an education of job safety, from 2000 until 2002. This study compared the change of blood lead levels (BLLs) between these two groups of workers. With informed consent, BLLs were measured annually for participants and compared. The average baseline BLLs were approximately at similarly high levels between Thai workers and native workers with means +/- standard deviations of 36.9 +/- 16.4 and 36.2 +/- 12.4 microg/dl, respectively (p = 0.79). At the end of 2002, the average concentration was higher in Thai workers than in native workers. Using mixed models, Thai workers had an average of 5.95 microg/dl increase in BLLs over native workers during the 3-year study. Further measurements revealed that the average BLL for workers in the assembly department was 3.57 +/- 1.83 microg/dl in excess, compared with workers in the plate engineering department. Thai workers were more likely to drink alcohol but less likely to wear gloves at work and wash hands before meals. The BLL disparities between Thai workers and native workers can partly be explained by differences in risk-taking behaviors. Higher BLLs in Thai workers suggest the need of language appropriate health education to improve their personal hygiene. Workplace smoking ceasing program may be needed both in Thai and native workers.  相似文献

Effect of smelter emission reductions on children's blood lead levels   总被引:2,自引:0,他引:2  
Trail, British Columbia has been the site of an active lead-zinc smelter for approximately 95 years. Since 1989, the community has been monitoring blood lead levels in children, studying exposure pathways and conducting comprehensive education and case management programs. From 1989 through 1996, mean blood lead levels of pre-school children declined at an average rate of 0.6 microg/dl per year. From 1996 to 1999, mean blood lead levels fell at an average rate of 1.8 microg/dl per year, from 11.5 in 1996 to 5.9 in 1999. The recent rapid decline appears to be mainly attributable to the start-up of a new lead smelter using modern flash-smelting technology in May of 1997. In 1998, the annual arithmetic mean air lead level in Trail was 0.28 microg/m(3), compared with 1.1 microg/m(3) in 1996. Reductions of approximately 50% were observed in lead loadings and concentrations in outdoor dustfall, street dust and indoor dustfall after smelter emissions were reduced. Slight reductions (statistically insignificant) have been observed in carpet dust and soil lead concentrations. During the summer of 2001, the smelting and refining operations at Trail were shut down completely for 3 months. During this period, average air lead levels in Trail dropped to 0.03 microg/m(3). The average blood lead level in Trail pre-school children at the end of the shutdown was 4.7 microg/dl. These results challenge prevailing theories about the relative importance of various environmental lead sources. For example, the US EPA Integrated Biokinetic Uptake Model for Lead (IEUBK), with its emphasis on soil concentrations, would not have predicted the dramatic decline in children's blood lead levels seen in Trail following the reductions in air lead levels. The Trail experience suggests that increased attention should be paid to the importance of active sources of highly bioavailable and mobile lead bearing dusts.  相似文献

Blood lead levels are reported for a total of 421 schoolchildren in 13 schools in rural and urban environments in Jamaica, including one highly contaminated community. In the rural areas blood lead levels ranged from 3 to 28.5 microg dl(-1), with a median of 9.2 microg dl(-1); the range and median in the urban schools were 4-34.7 and 16.6 microg dl(-1), respectively. Forty-two percent of the rural and 71% of the urban blood lead levels exceeded the US Centers for Disease Control and Prevention intervention level of 10 microg dl(-1). Except in the contaminated area, the relationship between soil lead levels, which in Jamaica are in general typical of tropical lateritic soils, and blood lead levels is not clear-cut. Very high blood lead levels of 18 to > 60 microg dl(-1) with a median of 35 microg dl(-1) were observed among children in the contaminated area, the site of a former lead ore processing plant. These high blood lead levels were significantly reduced, by the implementation of relatively simple mitigation strategies which involved isolation of the lead, education, and a food supplementation programme, to levels similar to those observed in the urban schools. These values, however, remain higher than are desirable and unfortunately, all the sources of lead are not yet identified. The recent discontinuation of the use of leaded petrol is expected to result in significant reductions in exposure to lead.  相似文献

The present study was designed to determine variations in blood lead levels during pregnancy and the influence of physiological anemia during this period on these findings. An exploratory study was made of a cohort of 38 pregnant women whose blood was tested for lead level and erythrocyte counts at different stages of pregnancy. The values obtained for lead were adjusted to take account of the erythrocyte count values, in order to control the influence of pregnancy-related physiological anemia on blood lead levels. A statistically significant increase in blood lead levels at a 5% level was observed between the first and third trimester. The median blood lead levels for the first, second and third trimester were 5.1 microg/dl, 5.9 microg/dl, and 8.25 microg/dl, respectively (Kruskal-Wallis=11.9, P<0.002). Statistical significance was also shown for raw data, when blood lead levels were not adjusted to erythrocyte count values. The median blood lead levels observed in this latter situation were 5.55 microg/dl, 5.65 microg/dl and 7.3 microg/dl, respectively (Kruskal-Wallis=8.3, P<0.02). It is concluded that there was a significant increase in blood lead concentration during pregnancy in this group of women.  相似文献

The phase-out of leaded gasoline began in Jakarta, Indonesia on July 1, 2001. We evaluated mean blood lead levels (BLLs) and the prevalence of elevated BLLs of Jakarta school children and assessed risk factors for lead exposure in these children before the beginning of the phase-out activities. The study involved a population-based, cross-sectional blood lead survey that included capillary blood lead sampling and a brief questionnaire on risk factors for lead poisoning. A cluster survey design was used. Forty clusters, defined as primary schools in Jakarta, and 15 2nd- and 3rd-grade children in each cluster were randomly selected for participation in the study. The average age of children in this study was 8.6 years (range 6-12) and the geometric mean BLL of the children was 8.6 microg/dl (median: 8.6 microg/dl; range: 2.6-24.1 microg/dl) (n=397). Thirty-five percent of children had BLLs > or =10 microg/dl and 2.4% had BLLs > or =20 microg/dl. Approximately one-fourth of children had BLLs 10-14.9 microg/dl. In multivariate models, level of education of the child's primary caregiver, water collection method, home varnishing and occupational recycling of metals, other than lead, by a family member were predictors of log BLLs after adjustment for age and sex. BLLs of children who lived near a highway or major intersection were significantly higher than those of children who lived near a street with little or no traffic when level of education was not included in the model. Water collection method was a significant predictor of BLLs > or =10 microg/dl after adjustment for age and sex. BLLs in children in this study were moderately high and consistent with BLLs of children in other countries where leaded gasoline is used. With the phase-out of leaded gasoline, BLLs of children in Jakarta are expected to rapidly decline as they have in other countries that have phased lead out of gasoline.  相似文献

Incubation of human whole blood for 24 h at 37 degrees C in the presence of 100-400 microg/dl lead chloride or lead acetate caused a concentration-dependent decrease in the level of reduced glutathione up to 40%. Similarly, the activities of glutathione reductase, glutathione peroxidase and glutathione S-transferase were decreased up to 25%, 50%, and 19%, respectively. Moreover, 100 microg/dl lead chloride or lead acetate slowed the process of glutathione regeneration, and delayed the time for complete regeneration from 20 to 40 min. When glutathione S-transferase was purified by affinity chromatography on Sepharose-linked glutathione, incubated with lead chloride or lead acetate, a concentration-dependent inhibition of the enzymatic activity was observed reaching 50% inhibition at a lead salt concentration of 6000 microg/dl.  相似文献

The 21 square mile Bunker Hill Superfund Site in northern Idaho includes several thousand acres of contaminated hillsides and floodplain, a 365-acre abandoned lead/zinc smelter and is home to more than 7000 people in 5 residential communities. Childhood lead poisoning was epidemic in the 1970s with >75% of children exceeding 40 microg/dl blood lead. Health response activities have been ongoing for three decades. In 1991, a blood lead goal of 95% of children with levels less than 10 microg/dl was adopted. The cleanup strategy, based on biokinetic pathways models, was to reduce house dust lead exposure through elimination of soil-borne sources. An interim health intervention program, that included monitoring blood lead and exposures levels, was instituted to reduce exposures through parental education during the cleanup. In 1989 and 2001, 56% and 3% of children, respectively, exceeded the blood lead criteria. More than 4000 paired blood lead/environmental exposure observations were collected during this period. Several analyses of these data were accomplished. Slope factors derived for the relationship between blood lead, soil and dust concentrations are age-dependent and similar to literature reported values. Repeat measures analysis assessing year to year changes found that the remediation effort (without intervention) had approximately a 7.5 microg/dl effect in reducing a 2-year-old child's mean blood lead level over the course of the last ten years. Those receiving intervention had an additional 2-15 microg/dl decrease. Structural equations models indicate that from 40 to 50% of the blood lead absorbed from soils and dusts is through house dust with approximately 30% directly from community-wide soils and 30% from the home yard and immediate neighborhood. Both mean blood lead levels and percent of children to exceed 10 microg/dl have paralleled soil/dust lead intake rates estimated from the pathways model. Application of the IEUBK model for lead indicates that recommended USEPA default parameters overestimate mean blood lead levels, although the magnitude of over-prediction is diminished in recent years. Application of the site-specific model, using the soil and dust partitions suggested in the pathways model and an effective bioavailability of 18%, accurately predicts mean blood lead levels and percent of children to exceed 10 microg/dl throughout the 11-year cleanup period. This reduced response rate application of the IEUBK is consistent with the analysis used to originally develop the cleanup criteria and indicates the blood lead goal will be achieved.  相似文献

Renal function was studied in 40 workers chronically exposed to lead at a smelter. The geometric means of lead were 40 (range 24.0 - 63.0) micrograms/dl in blood and 47 (range 19 - 115) micrograms/g creatinine or 71 (range 20 - 250) micrograms/l in urine, respectively. There were no significant differences between the lead-exposed group and a control group in the concentrations of beta 2-microglobulin in serum, urinary total protein and IgG, whereas the level of beta 2-microglobulin in urine was significantly higher in the lead-exposed group (p less than 0.01). No individual lead worker showed abnormally increased level of urinary beta 2-microglobulin.
