Microbial source tracking (MST) has become a focus of some recreational beach monitoring programs. Suspected sources of contamination include human sewage, agricultural runoff, and feces from wildlife and domestic animals, depending on beach location. Waterfowl have been suggested as a primary source of fecal contamination at many beaches, but techniques to “prove” contaminating microbes are of avian origin are mostly unsubstantiated. Researchers often rely on bird counts to measure the impact of waterfowl on beach health. Since waterfowl populations at Door County, Wisconsin (USA) beaches are transitory, this study focused on enumeration of avian waste material along beach transects, rather than on once per day “snapshot” bird counts. Escherichia coli (E. coli) concentration in beach water was not correlated with avian waste counts at the ten beaches studied in 2004 or the 13 studied in 2005 (rural to semi-urban). Bird counts correlated with E. coli concentrations in beach water at 30% of the sample sites in 2004 and at only one site in 2005. During the 2004 swimming season avian waste counts correlated with bird counts at only one beach and there was no correlation in 2005. These results indicate that neither avian waste enumeration nor bird counts can successfully be used to predict microbial contamination of recreational water at selected Great Lakes beaches.  相似文献

Studies to examine the prevalence of water‐related diseases, cyanobacteria toxins and other health risks at landing beaches in the Ugandan portion of the Lake Victoria basin were carried out. Based on surveillance studies and other data sources in the region, water samples were analysed for indications of faecal contamination. The coliform numbers were generally high for most sampling sites, indicating significant water contamination. The wet seasons exhibited significantly higher coliform counts than the dry seasons for all lakeshore sample sites. This seasonal variation in coliform counts correlated positively with the incidence of waterborne diseases, which are typically higher in the wet season. The water supply for domestic consumption for the Lake Victoria riparian communities is mainly the lake itself. The most prevalent diseases associated with the landing sites include malaria, dysentery, diarrhoea and bilharzia. Malaria was most prevalent, followed by dysentery, in all the studied districts. Many people in the catchment dispose of faecal wastes in lakeshore area bushes, or in polythene bags, contaminating water sources with faecal material, and resulting in waterborne diseases. The vulnerability of lakeside communities to water‐related diseases is further aggravated by low accessibility to health facilities and personnel. Further, cyanobacteria (potentially toxic to humans and animals) dominate other algal species in Lake Victoria, contributing >50% of the algal biomass. Algal blooms occurred frequently in Murchison Bay, a source of drinking water for the city of Kampala and surrounding urban centres. Algal blooms can cause unpleasant odours and tastes in domestic water supplies, clog pump and machinery filters, increase chlorine demands for water disinfection, and necessitate more complex and expensive treatment processes. The findings of this study suggest improved water quality, changes in sanitation and hygiene behaviour could significantly reduce the prevalence of water‐related diseases and cyanotoxins in the Ugandan portion of the Lake Victoria basin.  相似文献

Using a combination of chemical and microbiological (culture-dependent and -independent) approaches, sources of human faecal contamination were identified in two water reservoirs in Puerto Rico – Guajataca and La Plata. Fluorescence from optical brighteners (OB) – commonly found in laundry detergents – was used as an indicator of contamination from septic systems and other household discharges. Traditional indicators of faecal contamination (e.g. Escherichia coli; faecal enterococci) were enumerated, and human faecal contamination was confirmed through detection of Bifidobacterium adolescentis utilizing polymerase chain reaction (PCR)-based analyses. For Guajataca Reservoir, four of 19 sampling sites (21%) were positive for the presence of B. adolescentis under baseflow conditions. The OB fluorescence data suggested that the most likely source for three of these sites was residential, whereas the source of contamination at the remaining site, although of human origin, was probably non-residential. B. adolescentis was present in 83% (19 of 23) of the sampling sites in La Plata. The La Plata sources were more difficult to identify because samples were taken under stormflow conditions, although the presence of OB fluorescence suggested a residential origin in a number of instances. OB fluorescence and traditional bacterial indicators of faecal contamination produced a number of false positive and negative findings for both reservoirs, pointing to the importance of understanding the limitations of these tools for tropical freshwater systems. The results of this study should be useful in developing a weight-of-evidence approach for the identification of potential sources and extent of human faecal contamination in similar tropical reservoirs, a necessary step in the development of management plans to reduce or eliminate these sources.  相似文献

The supply of unfiltered disinfected drinking water from Melbourne's fully protected catchments means that the water-quality managers must ensure that the source water poses no public health risk. High turbidity is currently used as a surrogate of pathogens, and harvesting of water is based on its measurement. The work presented here summarises suspended particle and associated pathogen, microbial indicator and faecal biomarker concentrations collected to (a) quantify turbidity in an Australian water supply system and (b) assess the possibility of increasing water harvesting from selected tributaries. Pathogens and microbial indicators were present in low numbers in these source waters; increased turbidity during storm events was not associated with an increase in pathogen concentration. The results confirmed that protected catchments, along with good management, were effective barriers to pathogen contamination. Aesthetic issues still need to be addressed, but no measurable increase in microbiological risk was associated with storm-generated particles.  相似文献

Perceptions of the quality of roof harvested rainwater remain an impediment to widespread implementation of rainwater tanks on urban allotments. Previous literature reports on roof water quality have given little consideration to the relative significance of airborne environmental micro-organisms to roof catchment contamination and the issue of tank water quality. This paper outlines the findings of a recent study into the influence of weather on roof water contamination conducted at an urban housing development in Newcastle, on the east coast of Australia. Samples of direct roof run-off were collected during a number of separate rainfall events, and microbial counts were matched to climatic data corresponding to each of the monitored events. Roof run-off contamination was found to be under the strong influence of both wind speed and direction. The preliminary findings of an investigation currently under way into the microbial diversity of rainwater harvesting systems have also been presented. The results indicate that the composition of organisms present varied considerably from source to source and throughout the collection system. In all cases, evidence of faecal contamination was found to be negligible. The implications of these findings to the issues of tank water quality, health risk analysis and monitoring protocols have been discussed.  相似文献

Standard PCR (SPCR) and quantitative PCR (QPCR) assays using primers for general and for human-specific Bacteroides 16S rRNA markers were selected as the molecular tests to assess sewage contamination in recreational waters of Hawaii and these same water samples were assayed for culturable concentrations of selected faecal microbial indicators. The results of this study showed that the general primer for Bacteroides was not useful because ambient and polluted water samples were positive for this marker. However, use of human-specific primers reliably detected sewage contamination. The human-specific Bacteroides detection data supported previously reported conclusions that concentrations of alternative faecal indicators (C. perfringens, FRNA coliphages) but not traditional faecal indicators (faecal coliform, E. coli, enterococci) are reliable indicators of faecal contamination in Hawaii's environmental waters. The QPCR assay for the human-specific Bacteroides 16S rRNA marker was faster, more sensitive and more reliable than comparable SPCR assay because OPCR assay provided additional information such as melting temperatures, which confirmed that the right amplicons were being measured and Ct values, which indicated the relative level of faecal contamination.  相似文献

Microplastic pollution of the environment is ubiquitous, but the processes by which microplastics accumulate within beach sediments are not yet well understood. We isolate microplastic pollution from the sediments at three western Lake Superior beaches. Samples of both surface and subsurface sediments are considered. We find that the average microplastic contamination is 65 microplastic particles kg?1 sediment across our sites with significant variability across beaches, but the microplastic composition is always dominated by polyester fibers. The variation across beaches does not seem to relate to the distance from suspected sources of microplastics to the lake. Within each beach, we find no significant variation of the mean microplastic concentration in the cross-shore direction or in the surface vs. subsurface sediments at the wrack line. We interpret this mean microplastic concentration in the sediment as a measure of the bulk microplastic concentration in the nearshore water at each beach. In subsurface sediments, we observe a significant difference in the variance of microplastic concentrations, and we attribute this variation to the intermittency of the extreme hydrodynamic conditions that deposit microplastics deeper into the sediment.  相似文献

The 'H2S test' is being advanced for microbiological water quality testing where conventional coliform based methods are impractical or too expensive. It involves ambient temperature incubation of water samples with nutrient formulated to generate hydrogen sulphide when 'faecal' bacteria are present. Recently a WHO review identified several concerns including the limited number of comparative studies, formulation variability, and false positives and negatives. In response we have compared the H2S test's ability to detect and quantify faecal contamination in an aquifer impacted by septic tank leachfields with measurements obtained concurrently using conventional bacterial indicators, coliphages, faecal sterol biomarkers, Cryptosporidium and Giardia. Like these other analytes, H2S testing detected a contamination gradient ranging from high (septic liquid) to moderate (exfiltration zones), to background (e.g. domestic bores), corresponding to indicator removal + dilution by factors > 10(6). Presence/absence tests could not distinguish between heavily and slightly contaminated waters, whereas multi-tube testing (e.g. 10 x 10 mL arrays) did. It was concluded that while the WHO review concerns are justified, the H2S test performance shows promise in sanitary survey work, can be improved by employing an mpn approach and has potential to aid in the protection of source water and identifying contaminated groundwater.  相似文献

The first subsurface flow wetland (SSFW) system for about 1,000 PE, was constructed in Nicaragua in 1996 to apply this technology in the form of an integral project, combining the treatment of domestic wastewater with its reuse for crop production in small and medium size communities. The SSFW-effluent meets all standards established in the national regulations for wastewater reuse in agriculture, except for faecal coliforms, existent at an average concentration of 7 x 10(4) MPN/100 ml. A conventional surface irrigation method was used to irrigate different crop species selected to establish their risk of contamination. To judge the potential health risk for consumers and farmers, samples of vegetables and fruits harvested in the dry seasons of the years 1997 to 2002, were analyzed for the presence of pathogenic microorganisms like faecal coliforms, salmonella and shigella. In addition, a yield comparison between crops irrigated with well water using chemical fertilizers, and crops irrigated with the effluent of the SSFW-system was made, to analyze the economical benefits of the wastewater reuse.  相似文献

以水动力计算为基础,依据正向浓度概率密度与逆向位置概率密度的耦合关系,构建了以污染源位置和释放时间为参数的相关性优化模型,并利用微分进化算法实现了模型的求解。在此基础上,根据污染物正向浓度概率密度函数构建优化模型确定了污染物强度。耦合概率密度方法将突发水污染溯源问题转换为两个最小值优化问题,原理简单,求解方便,实现了污染源参数的解耦。在求解优化模型时,将梯度概念引入到微分进化算法中,提高了搜索效率。将所提出的溯源方法运用于仿真实验案例以及南水北调中线应急示范工程实例,结果表明,模型溯源效果好、模拟精度高,溯源结果对于河渠突发水污染事件应急处置具有一定指导意义。  相似文献

The soil environment in Hawaii is generally characterised as sub-optimal but permissive to support the in situ growth of E. coli and enterococci. However, soil desiccation and competition for nutrients by major indigenous soil microflora have been identified as potential factors that could limit a rapid and continual growth of faecal indicator bacteria in this soil environment. Despite these limitations, the genetic capacities of E. coli and enterococci are robust enough to enable these bacteria to become established as minor populations of Hawaii's soil microflora. Although the concentrations of E. coli and enterococci may have represented a fraction of the total soil microbiota, their presence in this habitat was very significant, for two important reasons: (a) soil was a major environmental source of E. coli and enterococci, and (b) the elevated counts of these bacteria in streams that routinely exceeded the EPA standards were due to run-off from soil. As a result, E. coli and enterococci were inadequate indicators to measure the degree of faecal contamination and potential presence of sewage-borne pathogens in Hawaiian streams.  相似文献

The objective was to investigate the potential risk of faecal indicator organism (FIO) bacteriological contamination of river catchments and coastal bathing waters from farm management practices and to develop practices to reduce the risk. A risk assessment on 117 farms was carried out in two river catchments in south-west Scotland. Manure storage facilities, farming practices, field conditions and catchment characteristics were assessed. River samples at 33 locations were regularly taken and analysed for FIOs. Available manure storage capacity and farm management practices are inadequate on a high proportion of farms and FIO contamination of watercourses was likely the result of effluent transported into watercourses due to non-collection or poor containment. In addition, surface run-off or leaching following land application of manure or intensive stocking in adverse conditions was a high risk on up to 50% of farms. The concentrations of FIOs in the streams of two sub-catchments with high livestock intensity was 4 to 8 times higher compared to the two sub-catchments which had a low livestock intensity. The majority of potential risks of agricultural pollution to watercourses may be eliminated through improved manure and dirty water management, forward planning of manure spreading activities and improved operational procedures.  相似文献

E. coli is an indicator of recent fecal contamination of freshwater beaches around the Great Lakes region. Elevated concentrations indicate that a fecal contamination has occurred, and that the risk for contact with fecal pathogenic organisms is heightened. The green algae, Cladophora, harbors populations of E. coli and potentially allows for prolonged survival and even replication of the bacterium in the lake environment. If presence of Cladophora mats on beaches is associated with persistence of E. coli in beach water, then E. coli would be a useful indicator organism only if pathogens also were able to survive and persist in the algae. This study utilized lab microcosms to study the persistence of E. coli, and of the fecal pathogens, Salmonella and Shigella, in lake water with and without the presence of Cladophora. E. coli was able to persist for extended periods in the presence of Cladophora (attached to algal mats for 45 days). Salmonella and Shigella, however, were unable to persist for this time period while in the presence of Cladophora (Salmonella attached to Cladophora was detectable for 10 days and Shigella was detectable for only 2 days). These data imply that E. coli is able to survive in the presence of Cladophora for greater times than are the fecal pathogens and that E. coli may not be an appropriate indicator organism for beaches with accumulations of algal material.  相似文献

This research used a weight-of-evidence approach to evaluate sources of contaminants in a drinking water watershed that serves as part of the City of Boston's water supply. The approach incorporated land use analysis using GIS, sanitary surveys, traditional water quality monitoring and microbial source tracking (MST) tools. Case-study tributaries were selected based on elevated faecal coliform counts. Land use analysis and sanitary surveys were used to identify suspected microbial sources, including residential septic systems, agricultural animal operations, commercial/industrial operations and wildlife activity. Sampling sites were selected to hydrologically isolate potential contamination sources. Samples were collected seasonally over 1 year and analysed for traditional and MST parameters. Results demonstrated that both septic systems and a horse stable were contributing microbial loads in the first tributary. In the second tributary, septic systems from the townhouses were contributing microbial loads while a plant nursery was contributing organic matter. This evidence was used to evaluate best management practices to mitigate the contamination.  相似文献

Antibiotic resistance profiling (ARP) is a potentially useful method for distinguishing faecal bacteria according to host source. This phenotypic approach has cost benefits over genotypic methods, but existing protocols are time-consuming and manual data handling is open to human error. A simplified, low-cost approach to the ARP technique was developed that used automated data recording techniques combined with simple statistical analyses to compare isolates of the genus Enterococcus from various faecal sources. An initial battery of 21 antibiotics (at up to four concentrations) was chosen for source discrimination. Images of growth or non-growth in microplate wells were stored as bitmaps and converted to binary data to form a database of known antibiotic resistance profiles. Discriminant function analysis (DFA) showed that the average rate of isolates correctly classified by the database (wastewater vs non-wastewater) was 86%. Once the more discriminating antibiotics and their concentrations had been identified, it was possible to reduce the number of tests from 80 to 18 whilst increasing the number of correctly classified human isolates. ARP could offer a low-cost and rapid means of identifying sources of faecal pollution. As such, the technique may be of particular benefit to developing countries, where water quality may have a significant impact on health and where cost is a major factor when choosing environmental management technology.  相似文献

The World Health Organisation's (WHO) Water Safety Plans highlight the need for preventative risk management when managing water contamination risks. As part of this approach, a management framework incorporating multiple barriers is necessary and there is a need to validate those barriers through scientific evidence. This paper reports on a study undertaken to validate the effectiveness, in terms of pathogen numbers, of having protected watersheds. The study aimed to determine if the deer population in a protected watershed carried Cryptosporidium and whether or not it was human infectious. Deer faecal samples were collected from the protected watersheds over a 12 month period and analysed using a new method, developed as part of this project, for genotyping Cryptosporidium. Early results showed the presence of Cryptosporidium, but following a refinement in the method no human infectious Cryptosporidium was detected. The results give some confidence that having protected watersheds is an effective barrier against pathogen contamination. They do not, however, imply that continued monitoring and management of the deer should cease. To maintain compliance with the Water Safety Plans, continual validation of barrier effectiveness is required.  相似文献

To ensure optimum quality at Bathing Waters, the control of diffuse sources of bacterial contamination is receiving increasing attention. As part of an initiative to improve the quality of the EU designated bathing waters on the Fylde Coast (North West England), a project was undertaken to quantify the faecal indicator load from birds and assess the significance on water quality. High bird counts are encountered on the Fylde Coast with gulls, feral pigeons and starlings being prominent. The piers at Blackpool make an attractive roost for starlings with numbers peaking at over 30,000 in late summer. Systematic recording of bird numbers and locations was undertaken during 2001/2. Estimates were also made of the daily faecal organism production by the different species. The spatial distribution of faecal organisms from the bird population was statistically linked to synoptic water quality data. This allowed estimates to be made of the contribution from birds to the faecal pollution load at the bathing waters. The work confirmed a statistically significant link between bird populations and water quality with a marked seasonal bias.  相似文献

Although contamination by organic pollutants has previously been reported to occur in the Haihe River basin, few studies have been carried out on the levels of source water reservoir contamination and the health risk in the Haihe River basin. To understand the organic pollution status of the reservoirs in the Haihe River basin, samples were collected from 16 source water reservoirs. The samples were analyzed for the representative organic pollutants, which included benzene homologues, chlorobenzene compounds, organophosphorus pesticides, and nitrobenzene compounds, a total in all of 17 compounds. It was observed that the concentrations of the 17 compounds in the 16 reservoirs were all less than the limit laid down by Chinese surface water quality standards. In addition, benzene, toluene, nitrobenzene, p-nitrochlorobenzene, 2,4-dinitrotoluene and 2,4-dinitrochlorobenzene, dichlorvos, demeton, dimethoate methyl parathion, malathion and parathion were frequently detected in the 16 source water reservoirs, especially the organophosphorus pesticides; the detection rates of dichlorvos, dimethoate, methyl parathion, malathion and parathion were all 100% in the 16 source water reservoirs. The detection rate of target compounds suggested that organic pollution had been common in the source water of the Haihe River basin. The health risk assessment results suggested that the noncarcinogenic risk hazard quotient values of the target compounds were less than one, and the cancer risk values were all below 1 × 10(-6), which indicated that the heath risk produced by the target compounds in the 16 reservoirs was at an acceptable level.  相似文献

Impairment of surface water quality by faecal coliform bacteria is an issue of great importance across the globe. A water quality model, Hydrological Simulation Program FORTRAN, was used to predict the impacts of farming and climate change on faecal coliform loads and concentrations in streams of the Lis River watershed, in the Leiria region, Portugal. The calibrated faecal coliform model simulated well the patterns and range of observed faecal coliform concentrations. The accuracy of the model was evaluated by the per cent bias coefficient and the coefficient of determination. The results indicate a general deterioration of the water quality regarding faecal contamination in Lis River. Maximum daily loads were calculated for each of the impaired streams; an average of 77% reduction in the current faecal coliform load from the watershed is necessary to achieve the established water quality goals by the Council Directive 75/440/EEC ( 1975 ). Climate change scenarios (increments on temperature and precipitation) were assumed to predict the behaviour of faecal coliform bacteria in the watershed. The simulated results showed that an increase of 1°C in air daily temperature results in an increase of water temperature of 1.1°C and a 1.5% decrease on faecal coliform bacteria in stream concentration. The combined effect of air temperature (+1°C) and precipitation (+7%) increment leads to an increase of ~2% in bacteria inflow to the basin. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献
