Two variants of a continuous recognition training procedure were designed in order to query 2 forms of spatial memory. A continuous reinforcement condition (reflecting perceptual memory) and a differential reinforcement condition (reflecting episodic-like memory) were used to test rats on a 12-arm radial maze. After total hippocampal lesions, rats demonstrated intact performance on the continuous reinforcement condition, but impaired performance on the differential reinforcement condition. After parietal lesions, rats demonstrated the reverse pattern of performance: impaired performance on the continuous reinforcement condition and intact performance on the differential reinforcement condition. Thus, a double dissociation appears to exist between parietal cortex and hippocampus for the continuous reinforcement condition (reflecting perceptual memory) versus the differential reinforcement condition (reflecting episodic memory) for spatial location information. In rapid serial visual presentation tasks, correct identification of a target triggers a deficit for reporting a 2nd target appearing within 500 ms: an attentional blink (AB). A different phenomenon, termed repetition blindness (RB), refers to a deficit for the 2nd of 2 stimuli that are identical. What is the relationship between these 2 deficits? The present study obtained a double dissociation between AB and RB. AB and RB followed different time courses (Experiments 1 and 4A), increased target-distractor discriminability alleviated AB but not RB (Experiments 2 and 4A), and enhanced episodic distinctiveness of the two targets eliminated RB but not AB (Experiments 3 and 4B). Groups of normal old people living in institutions or in the community and young adults were administered tests of implicit (IM) and explicit (EM) memory with word-stem (WSC) and word-fragment (WFC) completion paradigms. Neuropsychological tests sensitive to frontal and medial temporal lobe function were also administered. Age differences were observed on both tests of EM and on all neuropsychological tests. Priming effects on WSC were smaller in the institutionalized group than the other 2 groups. Comparisons of EM and IM test results with neuropsychological test scores revealed several effects, including significant correlations (a) between EM scores on both tests and performance on standard memory tests in both aged groups and (b) between IM scores of both aged groups on WSC and frontal-lobe test performance. The results provide evidence of a double dissociation with respect to involvement of brain regions in EM and IM. They also indicate that repetition priming in WSC and WFC involve different mechanisms and that frontal-lobe dysfunction is a factor in reduced priming on the WSC test. Animals with medial prefrontal cortex or parietal cortex lesions and sham-operated and non-operated controls were tested for the acquisition of an adjacent arm task that accentuated the importance of egocentric spatial lateralization and a cheese board task that accentuated the importance of allocentric spatial localization. Results indicated that relative to controls, animals with medial prefrontal cortex lesions are impaired on the adjacent arm task but displayed facilitation on the cheese board task. In contrast, relative to controls, rats with parietal cortex lesions are impaired on the cheese board task but show no impairment on the adjacent arm task. The data suggest a double dissociation of function between medial prefrontal cortex and parietal cortex in terms of coding of egocentric versus allocentric spatial information. Carried out 3 experiments, using 101 female rats of the CFE strain. Bilateral parasagittal transections were made at different lateral and dorsal-ventral positions in and between the ventromedial hypothalamus (VMH) and lateral hypothalamus (LH). Cuts through or just lateral to the VMH produced hyperphagia and obesity, while cuts through or medial to the LH produced, depending upon their exact position and the diet, either temporary aphagia-hypophagia, mild hyperphagia, or no effect. The cuts medial to the LH, but not the others, abolished insulin-induced eating. All cuts produced deficits in drinking regulation that varied according to their exact position. These deficits included hypodipsia, mild hyperdipsia, and reduced water/food ratios, as well as reduced drinking responses to food deprivation, water deprivation, hypertonic sodium chloride injections, and polyethylene glycol injections. The knife cuts also produced changes in gustatory reactivity. The hypothalamic pathways responsible for these effects are discussed. (54 ref.) Using the peak procedure, rats with aspiration lesions to the medial prefrontal cortex (PFC) or the hippocampus were tested for the acquisition of timing behavior and temporal memory. After surgery, rats were trained to discriminate a 40-sec interval and then tested for temporal memory with gap trials. Results indicated that lesions to the medial PFC disrupted the acquisition of timing behavior. Medial PFC animals needed significantly more trials to reach criterion, and their temporal discrimination function was less uniform and steep, indicating a general deficit in timing ability. In hippocampal rats, the ability to estimate the duration of the discriminative stimulus was unaffected by the lesion. It was concluded that the hippocampus is not necessary for the acquisition of timing behavior in this task. Gap trials failed to produce a deficit in the memory for temporal events for either lesion. Patients with Alzheimer's disease (AD) and healthy control participants performed 2 conceptual repetition priming tasks, word-associate production and category -exemplar production. Both tasks had identical study-phases of reading target words aloud, had the most common responses as target items, and required production of a single response. Patients with AD showed normal priming on word-associate production but impaired priming on category-exemplar production. This dissociation in AD suggests that conceptual priming is not a unitary form of memory but rather is mediated by separable memory systems. These experiments illustrate 2 new dissociations in word-recognition tasks. In one, relatedness facilitated lexical decision but impaired searching for a common letter in the same pairs of words (a cross-over interaction between relatedness and task). In the other dissociation, lexicality facilitated performance (words processed faster than nonwords) while relatedness impaired performance (related words processed slower than unrelated words) in the letter search task. Two classes of explanation are discussed. In the first, the perception of relatedness serves to focus attention to the word level, thereby making explicit letter level processing more difficult and/or increasing the number of competing lexical entries via priming. The purpose of this research was to determine whether brain stimulation reward and exploration are induced by activation of the same set of neurons along the medial forebrain bundle. The behavioral version of the collision test was utilized with electrodes in the lateral hypothalamus (LH) and the ventral tegmental area (VTA). A collision effect obtained between LH and VTA in one behavior at the exclusion of the other was treated as evidence of the involvement of two different sets of fibers. In 4 rats, a collision effect was observed only in self-stimulation, whereas in 1 rat, a collision was obtained in exploration at the exclusion of self-stimulation. Three animals showed no collision in either behavior. These data suggest that coexistence of self-stimulation and exploration following medial forebrain bundle stimulation can be explained by current spread on two different sets of fibers. In 2 experiments single or multiple sets of bilateral knife cuts were made in a total of 73 female CFE rats just lateral to the ventromedial hypothalamus (VMH) and/or just medial to the lateral hypothalmus (LH). The lateral VMH knife cuts by themselves produced greater hyperphagia and obesity than did the medial LH cuts. The lateral VMH knife cuts also significantly increased food intake and body weight in Ss previously given bilateral cuts along the medial LH border. Findings indicate that the feeding inhibitory fibers responsible for the hyperphagia syndrome do not project from the VMH to the LH, and this calls for a reevaluation of hypothalamic circuitry. It was also discovered that sham surgery in 7 Ss had a significant suppressive effect on the hyperphagia syndrome produced by hypothalamic knife cuts. Complete and dorsal hippocampal lesions impaired spatial performance on 2 working memory tasks: rewarded alternation on the T maze and matching to position in the water maze. In contrast, ventral hippocampal lesions had no effect on these tasks, even when task difficulty was increased by the introduction of delays. Ventral lesions did resemble complete lesions in reducing anxiety in 3 commonly used tests of anxiety (social interaction, plus-maze, and hyponeophagia). Dorsal lesions also appeared to be anxiolytic in the social interaction and plus-maze tests, but they did not affect hyponeophagia. Complete- and dorsal-lesioned rats displayed hyperactivity, whereas ventral-lesioned rats did not. These results show a double dissociation between dorsal and ventral hippocampal lesions (hyponeophagia vs. spatial memory), suggesting differentiation of function along the septotemporal axis of this structure. Correlated attitudes concerning the job in general and specific aspects of the work situation with termination decisions and recorded frequency of absences during the year following administration of job attitude questionnaire to 160 nonsupervisory female employees. Overall satisfaction and satisfaction with several intrinsic aspects of the work situation were significantly related to the termination criterion. None of the extrinsic variables were related. For the frequency of absence criterion, attitudes toward specific aspects of the work situation showed no consistent relationships when nonattitudinal variables were partialed out, but overall satisfaction was significantly related. In 4 experiments, 61 undergraduates viewed rapidly presented series of color photographs (9/sec) and were required to indicate which photograph appeared within a black outline rectangle (the frame). Exp I demonstrated that Ss were often confident and wrong, reporting the immediately preceding or following picture in the sequence. Exp II showed that migration of the frame to other pictures could not be attributed to spatial separation, because the same effect occurred when a small frame was presented in the center of the picture itself. Exp III ruled out masking of the framed picture as the cause of the illusion by showing that the framed picture was identified on those trials in which the frame appeared to be elsewhere. Exp IV showed that when simpler, more familiar stimuli (numbers) were presented, a more rapid presentation rate (18/sec) was required to obtain the effect. It is proposed that the illusion reflects the action of integrative processes in a very short-term buffer and that it may provide a new tool with which to study the integration of features within scenes. Amyloid deposition is a neuropathological hallmark of Alzheimer's disease. The principal component of amyloid deposits is beta amyloid peptide (Abeta), a peptide derived by proteolytic processing of the amyloid precursor protein (APP). APP is axonally transported by the fast anterograde component. Several studies have indicated that Abeta deposits occur in proximity to neuritic and synaptic profiles. Taken together, these latter observations have suggested that APP, axonally transported to nerve terminals, may be processed to Abeta at those sites. To examine the fate of APP in the CNS, we injected [35S]methionine into the rat entorhinal cortex and examined the trafficking and processing of de novo synthesized APP in the perforant pathway and at presynaptic sites in the hippocampal formation. We report that both full-length and processed APP accumulate at presynaptic terminals of entorhinal neurons. Finally, we demonstrate that at these synaptic sites, C-terminal fragments of APP containing the entire Abeta domain accumulate, suggesting that these species may represent the penultimate precursors of synaptic Abeta.  相似文献

Paired-pulse plasticity is typically used to study the mechanisms underlying synaptic transmission and modulation. An important question relates to whether, under physiological conditions in which various opposing synaptic properties are acting in parallel, the net effect is facilitatory or depressive, that is, whether cells further or closer to threshold. For example, does the net sum of paired-pulse facilitation (PPF) of excitatory postsynaptic potentials (EPSPs), paired-pulse depression (PPD) of inhibitory postsynaptic potentials (IPSPs), and the hyperpolarizing slow IPSP result in depression or facilitation? Here we examine how different time-dependent properties act in parallel and examine the contribution of gamma-aminobutyric acid-B (GABAB) receptors that mediate two opposing processes, the slow IPSP and PPD of the fast IPSP. Using intracellular recordings from rat CA3 hippocampal neurons and L-II/III auditory cortex neurons, we examined the postsynaptic responses to paired-pulse stimulation (with intervals between 50 and 400 ms) of the Schaffer collaterals and white matter, respectively. Changes in the amplitude, time-to-peak (TTP), and slope of each EPSP were analyzed before and after application of the GABAB antagonist CGP-55845. In both CA3 and L-II/III neurons the peak amplitude of the second EPSP was generally depressed (further from threshold) compared with the first at the longer intervals; however, these EPSPs were generally broader and exhibited a longer TTP that could result in facilitation by enhancing temporal summation. At the short intervals CA3 neurons exhibited facilitation of the peak EPSP amplitude in the absence and presence of CGP-55845. In contrast, on average L-II/III cells did not exhibit facilitation at any interval, in the absence or presence of CGP-55845. CGP-55845 generally "erased" short-term plasticity, equalizing the peak amplitude and TTP of the first and second EPSPs at longer intervals in the hippocampus and auditory cortex. These results show that it is necessary to consider all time-dependent properties to determine whether facilitation or depression will dominate under intact pharmacological conditions. Furthermore our results suggest that GABAB-dependent properties may be the major contributor to short-term plasticity on the time scale of a few hundred milliseconds and are consistent with the hypothesis that the balance of different time-dependent processes can modulate the state of networks in a complex manner and could contribute to the generation of temporally sensitive neural responses.  相似文献

In 5 experiments, the authors examined the perceptual and cognitive processes used to track the locations of objects during locomotion. Participants learned locations of 9 objects on the outer part of a turntable from a single viewpoint while standing in the middle of the turntable. They subsequently pointed to objects while facing the learning heading and a new heading, using imagined headings that corresponded to their current actual body heading and the other actual heading. Participants in 4 experiments were asked to imagine that the objects moved with them as they turned and were shown or only told that the objects would move with them; in Experiment 5, participants were shown that objects could move with them but were asked to ignore this as they turned. Results showed that participants tracked object locations as though the objects moved with them when shown but not when told about the consequences of their locomotion. Once activated, this processing mode could not be suppressed by instructions. Results indicated that people process object locations in a body- or an environment-stabilized manner during locomotion, depending on the perceptual consequences of locomotion. Several forms of long-term potentiation (LTP), a putative cellular mechanism for memory storage, have been described in the hippocampus. In this review, I discuss the mechanisms of induction and expression of LTP at the Schaffer collateral synapses and at the mossy fiber pathway. The early biochemical steps responsible for LTP at these two pathways are well understood. However, future studies should transcend the study of signal transduction systems and focus on the identification of the synaptic proteins that experience activity-dependent modifications, ultimate effectors of the plastic changes.  相似文献

Extracellular investigation in unanaesthetized rabbits revealed low reactivity of the lateral entorhinal cortex neurones (lEC, field 28b) to visual and auditory stimuli. Only 25% of cells responded to them by diffuse long-latency reactions, while in medial EC (mEC, field 28a) 70% of neurones responded to these stimuli with short-latency patterned reactions. The neurones of lEC were selectively responsive to different somatosensory stimuli. Among reactions short-latency on-effects were observed. In some cells application of these stimuli switched on rhythmic bursts of spikes (frequency about 3-4 Hz) or regular activity of pacemaker type. The lEC neurones responded by short-latency driving to electrical stimulation of prefrontal cortex, while mEC neurones were responsive to stimulation of posterior non-primary neocortical areas. The data are discussed in the light of recent morphological findings on cortico-cortical connections of EC.  相似文献

We characterized the neuronal properties of the anterior cingulate cortex (ACC) evoked by electrical stimulation of the medial thalamus (MT). MT stimulation sites were found by their neuronal responses to noxious stimuli. Of 487 units identified histologically in the rat ACC, 94% were activated trans-synaptically at different areas of the ACC. Six percent of MT-evoked ACC units were activated antidromically and all of these units projected to a specific nucleus of MT. We suggest that MT nuclei mediate different aspects of nociceptive information to specific ACC areas, and that nociceptive information in the MT is modulated reciprocally by activities from the ACC.  相似文献
