A fractionation method for isolating the polycyclic aromatic sulphur heterocycles from complex mixtures of polycyclic aromatic compounds was evaluated with respect to recovery for selected standard compounds. It was found that recovery varied from 0 to 70% depending on the structure of the compound. Although quantitative recovery for all components was not obtained, isolation of the sulphur heterocycle fractions has allowed the identification of many new sulphur compounds previously not identified in coal liquids and shale oils. Capillary column gas chromatography with flame photometric detection and mass spectrometry were used for the identification of individual sulphur heterocycles.  相似文献

Advances in analytical methods and bioassay development have helped to push forward the research in natural products. In plant extracts and nutraceuticals, bioactive compounds are part of a complex mixture. The development of high-resolution methods related to HPLC for both chemical and biological profiling has significantly increased the efficiency of classical bioactivity-guided fractionation procedures. Furthermore, the level of sensitivity obtained by these methods give the possibility to work with few micrograms of compound. This represents a key advantage for rapid localisation of the biological activity and subsequent identification of the compounds of interest. The same methods are also used to study the extracts from a metabolomic view point. The possibility to study them as a whole can highlight synergetic effects, which are likely to occur in plant extracts and nutraceuticals. In this paper, the main trends are summarised and the developments made in our laboratory on profiling crude extracts with UHPLC-TOF-MS, natural product identification at the microgram level using microflow NMR and integration of these methods with biological evaluation are highlighted.  相似文献

Detailed identification of sulfur-containing polycyclic aromatic compounds (S-PAC) was accomplished for solvent extracts of five different coals. The S-PAC were isolated by ligand-exchange chromatography. Due to the great complexities of these samples, capillary gas chromatography (GC) was employed for their analysis. An element selective radio frequency plasma detector was used to provide sulfur selective detection. Gas chromatography-mass spectrometry was used to identify individual compounds in isolated sulfur fractions. Condensed thiophenic compounds were found to be the major constituents in all five S-PAC fractions. Diaryl sulfides were also detected in the high volatile bituminous Illinois #6 coal. The prevalence of various S-PAC in different coal extracts was observed as a function of their rank.  相似文献

The chemical basis of feeding responses to the acceptable nonhost plantVigna sinensis (cowpea) by larvae ofManduca sexta was investigated using chemical isolation techniques directed by a novel chemosensory-based bioassay. The presence of feeding stimulatory and inhibitory compounds in leaves or leaf extracts was determined in a two-choice preference test using leaf disks or glass fiber filter paper disks laced with leaf extract as test substrate and filter paper disks laced with water as control. Larvae strongly prefer the control disks over leaf disks, indicating the presence of feeding inhibitory compounds in the leaf. An ethanol extract of both fresh and dried leaves neither stimulated nor inhibited feeding. The cause of this inactivity was examined by using larvae that respond strongly to either feeding stimulatory or inhibitory compounds due to selective chemosensory deprivation. Larvae having chemosensory organs remaining only on the maxillary palps are stimulated to feed by whole leaf disks and by the ethanol extracts. In contrast, larvae having only the medial and lateral maxillary sensilla styloconica and the epipharyngeal sensilla remaining are strongly inhibited by whole leaf disks and the ethanol extract of fresh leaves. Thus, the ethanol extract contains both feeding stimulatory and inhibitory compounds, which elicit opposite behavioral effects in unoperated larvae, therefore nullifying any stimulatory and inhibitory activity. These compounds can only be demonstrated by using discrimination-enhanced larvae in the choice tests. Further isolation of the feeding stimulatory principle inV. sinensis yielded two separate fractions of neutral compounds, suggesting at least two different chemicals belonging to two different classes: nonpolar and polar lipids. Feeding inhibitory chemicals have apparently polar properties because strong activity was found in the ethyl acetate and methanol extracts of dried leaves. The role of feeding stimulatory and inhibitory compounds in food selection ofM. sexta larvae is discussed.  相似文献

The attractiveness of representative host materials, host extracts, and individual host volatiles (primarily carboxylic acids, alcohols, and esters) toCarpophilus hemipterus (L.) (Coleoptera: Nitidulidae) adults in wind-tunnel bioassays was examined. Attractiveness of the materials was examined alone and in combination with the aggregation pheromone. Host materials and extracts were often attractive on their own, and the attractancy was synergized when they were combined with the pheromone. Propanoic and butanoic acids, methanol, 2-propanol, 1-heptanol, methyl butanoate, and propanal were among the most effective attractants relative to the pheromone, but many other compounds significantly synergized the pheromone (typically three- to four fold). Attractiveness and synergism were influenced by the carbon chain length and branching of the substitutents. Straight-chain compounds that had at least three carbon atoms were generally effective as synergists. Many branched-chain compounds were also effective synergists. In general, the degree of attractiveness and synergism could be predicted fairly well with the physicochemical steric (Es) parameter, although the lipophilicity (Pi) parameter also appeared to be useful in explaining the lower activity of short-chain substituents. Thus, many compounds that had only limited attractiveness on their own may nevertheless play and important role in synergizing the pheromone. Structure-activity studies appear to be appropriate not only for determining optimal attractants for these insects, but also for determining effective synergists for the pheromone.The mention of firms or trade names does not imply that they are endorsed or recommended by the U.S. Department of Agriculture over other firms or similar products not mentioned.  相似文献

The allelopathic potential of a cattail species (Typha domingensis) towards the common water fern (Salvinia minima) was studied in a short-term bioassay. Aqueous extracts of cattail plants (roots, stems, and leaves) as well as two of its phenolic compounds (2-chlorophenol and salicylaldehyde) were found to inhibit the growth of Salvinia when incorporated in the growth medium. After a period of seven days, chlorophyll concentration, fresh weight, dry weight, and number of fronds of Salvinia were compared and correlations were found among the different parameters. The results suggest an inhibitory effect by components of Typha, especially of the root fractions. Results from pure chemicals are in qualitative and quantitative agreement with those obtained from the cattail extracts.  相似文献

Bioassay-guided isolation of bioactive natural substances requires monitoring of all fractionation and purification steps using a bioassay system. This is often a long and tedious process, especially with insect feeding bioassays. We report here a new method, based on the principle of bioautography, for a quick isolation of insect antifeedant compounds. TLC plates, after development, are coated with a thin layer of artificial diet and fed toSpodoptera litura larvae. The location of uneaten areas is then compared with theR fvalues of the TLC spots, in order to determine rapidly the active fractions. This methods allows for a very fast determination of the most active antifeedant compounds in a complex mixture and considerably speeds up the isolation process. This new method was successfully applied in the study of antifeedant activity of several plant samples, and results are presented here for a model plant,Skimmia japonica (Rutaceae). Using this new method, the compounds responsible for the feeding-deterrent activity, three furanocoumarins (bergapten, xanthotoxin, and oxypeucedanin), were quickly and efficiently identified.  相似文献

Mice can discriminate samples of urine obtained from two groups of inbred mice that are genetically identical except in their major histocompatibility complex (MHC) haplotype (congenic mice), whereas they cannot distinguish urine samples from two genetically identical groups of mice. Chemical fractions of urine samples obtained from MHC congenic mice were tested in a Y-maze olfactometer using a method modified to accommodate the bioassay to chemical fractions that might differ in sensory properties from the unfractionated urine. Fractions depleted in protein by several methods were consistently discriminable by mice in the Y maze, providing a direct demonstration that the airborne MHC genotype information can be conveyed by volatile compounds alone.  相似文献

Bioassay-guided fractionations of a dichloromethane extract of wheat epicuticular wax allowed the identification of 1-octacosanal and 6-methoxy-2-benzoxazolinone (MBOA) as the major components that stimulate oviposition by the Hessian fly, Mayetiola destructor. These compounds were identified by their mass spectral fragmentation patterns and by comparison of their gas chromatographic retention times with synthetic samples. Synthetic samples of 1-octacosanal or MBOA stimulated significant oviposition when compared with solvent controls. In combination, these compounds elicited a synergistic effect on the number of eggs laid by females compared to when they were presented alone. In a choice bioassay, a mixture of synthetic 1-octacosanal and MBOA in the approximate concentrations determined to be present in one plant equivalent of crude extract stimulated the same amount of oviposition as one plant equivalent of extract. This showed that together these two compounds appear to be responsible for the major proportion of the ovipositional stimulatory activity of the wheat epicuticular wax extract. Comparison of the activity of five straight-chain primary aldehydes with chain lengths from C22 to C30 revealed a relationship between chain length and the number of eggs laid by female Hessian flies, with 1-hexacosanal and 1-heptacosanal the most active of the aldehydes tested.  相似文献

Plant extracts are complex matrices and, although crude extracts are widely in use, purified compounds are pivotal in drug discovery. This study describes the application of automated preparative-HPLC combined with a rapid off-line bacterial bioassay, using reduction of a tetrazolium salt as an indicator of bacterial metabolism. This approach enabled the identification of fractions from Dodonaea viscosa that were active against Staphylococcus aureus and Escherichia coli, which, ultimately, resulted in the identification of a clerodane type diterpenoid, 6β-hydroxy-15,16-epoxy-5β, 8β, 9β, 10α-cleroda-3, 13(16), 14-trien-18-oic acid, showing bacteriostatic activity (minimum inhibitory concentration (MIC) = 64–128 µg/mL) against test bacteria. To the best of our knowledge, this is the first report on antibacterial activity of this metabolite from D. viscosa.  相似文献

Capillary gas chromatography with columns of different polarity and two-dimensional fractionation of effluents were used with novel subtrac-tive-combination bioassays to rigorously isolate host- and insect-produced pheromone synergists of the bark beetlePityogenes chalcographus (Coleop-tera: Scolytidae). Methyl (E,Z)-2,4-decadienoate (E,Z-MD) and the previously identified chalcogran were found to be synergistically attractive to both sexes.E,Z-MD was produced sex-specifically in males, and only when they had fed on host-plant tissue. A Norway spruce monoterpene fraction (including -pinene, -pinene, and camphene) increased the attractive response to the pheromone components. Dose-response curves forE,Z-MD and chalcogran in the laboratory bioassay indicated the two components are highly synergistic. The isolation methods are important for further progress in identifying certain semiochemical synergists found in trace amounts in complex chemical mixtures, such as when insects must feed in host plants in order to produce pheromone.  相似文献

More than twice the number of mated female Hessian flies,Mayetiola destructor (Say) entered a zone within 1 cm of a paper strip treated with one plant equivalent (PE) of a chloroform extract of wheat foliar waxes compared to a strip treated with solvent only; females also stayed six times longer and laid 10 times more eggs on the strip treated with the wheat extract. Column chromatographic fractionation of the wheat extract and application of these fractions onto filter paper strips showed four fractions elicited significant numbers of eggs to be laid. Single, binary, and tertiary combinations of three of these fractions (two of the four fractions apparently contained similar compounds) were tested. The greatest numbers of eggs were laid on strips treated with the tertiary combination or the binary combination conaining the two most active fractions (3 and 6); three times the number of eggs were laid on strips treated with this binary combination than the sum of eggs laid on strips treated with these two fractions separately. A comparison of grasses and their extracts showed female Hessian flies laid greater numbers of eggs on wheat or rye than on barley or oat. Fractionated barley and oat extracts were tested for activity as for wheat, and a similar pattern was observed, i.e., the greatest numbers of eggs were laid on fractions 3 and 6. Dose-response tests, using these two fractions of wheat, barley, or oat showed the same threshold of activity for fraction 3 for all three extracts, i.e., 2 PE. In contrast, fraction 6 of wheat was active at the lowest dosage tested, 0.25 PE, while the same fraction of either barley or oat was not active until tested at a dosage of 2 PE. It appears that (at least) two chemicals in the foliar waxes of these grasses influence ovipositional behavior of female Hessian flies. Furthermore, given the similar foliar chemistry of these grasses and the strong synergistic interaction between fractions 3 and 6 shown for wheat extract, it is likely that the ovipositional preferences exhibited by female Hessian flies towards these grasses may be explained by quantitative differences in the amount(s) of the active chemical(s) in their respective fraction 6 (most polar fraction tested).  相似文献

Three types of combustion sample extracts, smokeless coal, smoky coal, and wood, were analyzed for a range of polycyclic aromatic hydrocarbons (PAHs) and polycyclic aromatic sulfur heterocycles (PASH). Standard Reference Material (SRM) 1597, Complex Mixture of PAHs from Coal Tar, was also analyzed as a control sample and for the determination of a larger number of PAHs relative to those determined previously. Target analytes included many alkyl-substituted PAHs such as dimethylphenanthrenes, methylfluoranthenes, and methylpyrenes. The analytical methods included sample clean-up and the selection of specific stationary phases to accomplish unique separations of PAHs. Clean-up involved the use of normal-phase liquid chromatographic isolation of PAHs based on the number of aromatic carbons and a total PAH fraction, PAHs in the resulting fractions were separated by gas chromatography using two stationary phases with different selectivities and analyzed using mass spectrometry. These methods are discussed below and results are presented with an emphasis on the relative concentrations and distribution of PAHs in the combustion samples.  相似文献

The occurrence of a genital sex pheromone in the anterior reproductive tract of partially fed femaleDermacentor variabilis was demonstrated by extraction and bioassay. A new type of bioassay, the neutered female assay, was developed to test the potency of extracts or chemically defined fractions to stimulate males to copulate. Electrophysiological tests confirmed the ability of males to detect the pheromone with sensilla on their cheliceral digits. Males of bothD. variabilis andD. andersoni exhibited neuronal excitation when stimulated with extracts of theD. variabilis reproductive tissues. The pheromone, which is soluble in methanol, was fractionated and found to contain at least two fractions that stimulated copulation by sexually excited males. Evidently, the pheromone is a mixture of two or more compounds. Histologic, ultrastructural, and histochemical studies suggest the vestibular vagina as the site of genital sex pheromone occurrence, presumably from secretions of the surrounding lobular accessory gland. Egg-laying females of the pine weevil, Hylobius abietis (L.), regularly deposit feces adjacent to each egg. Egg cavities are gnawed in the bark of roots of recently dead conifer trees. After egg deposition, the cavity is sealed by feces and a plug of bark fragments. Root bark containing egg cavities with feces is avoided as food by pine weevils, which indicates the presence of natural antifeedants. Here we present the first results of the isolation and chemical analyses of antifeedant compounds in the feces of H. abietis. In feeding bioassays, methanol extracts of the feces revealed strong antifeedant properties. Methanol extracts were fractionated by medium-pressure liquid chromatography and the antifeedant effects were mainly found in the fractions of highest polarity. Volatile compounds in the active fractions were identified by gas chromatography–mass spectrometry (GC–MS) and the nonvolatile compounds were characterized by pyrolysis–GC–MS. Based on mass spectra, a number of compounds with various chemical structures were selected to be tested for their antifeedant properties. Antifeedant effects were found among compounds apparently originating from lignin: e.g., a methylanisol, guaiacol, veratrol, dihydroxybenzenes, and dihydroconiferyl alcohol. A weak effect by fatty acid derivatives was found. The types of naturally occurring antifeedant compounds identified in this study may become useful for the protection of planted conifer seedlings against damage by H. abietis.  相似文献

Low-pressure liquid chromatography, high-performance liquid chromatography, and field ionization mass spectrometry (f.i.m.s.) were used to obtain compositional information on large-ring number polycyclic aromatic hydrocarbon (PAH) present in a non-distillable coal liquid sample. A highly selective h.p.l.c. method for the separation of (PAH) from polar compounds was applied to nitrogen-compound fractions derived from a Wyodak non-distillable ( > 427 °C) coal-liquid sample. F.i.m.s. analyses revealed that the PAH subfractions isolated by the h.p.l.c. procedure contained large-ring number PAH and relatively few nitrogen compounds. The methods developed can generally be applied to the analyses of complex organic mixtures, and in conjunction with other methods, can yield detailed polycyclic aromatic hydrocarbon compositional information.  相似文献

Sources of Apple Odor Attractive to Adult Plum Curculios   总被引:2,自引:0,他引:2  
We evaluated olfactory attraction of overwintered plum curculio (PC) adults, Conotrachelus nenuphar, to volatiles from extracts of host and nonhost plant tissues in the laboratory using a still-air dual-choice bioassay system. In experiments evaluating four different solvents, hexane and water proved to be better than dichloromethane and methanol for extracting nonpolar and polar attractive compounds, respectively, from host McIntosh apple tissues. Significantly more PCs were attracted to volatiles from hexane extracts of host wild plum fruit at bloom, and host wild plum and McIntosh fruit at two weeks after bloom, than to volatiles from hexane extracts of nonhost honeysuckle fruit at either phenological stage. In every case, for hexane-extracted or water-extracted volatiles from McIntosh apple tissues (twigs, leaves, or fruit) at seven phenological stages of development (pink through five weeks after bloom), greatest numerical responses were recorded to volatiles from extracts made between bloom and two weeks after bloom. We conclude that source material used to identify volatile components of McIntosh apple odor attractive to PCs should be collected between bloom and two weeks after bloom.  相似文献

Tree shrews of both sexes exhibit marking behavior (chinning) in response to scent marks made by the urine of fertile male conspecifics. To isolate the effective odor components, the urine was fractionated by liquid-liquid extraction and TLC, and the fractions were tested by bioassay. The results show that chinning is elicited by several lipophilic urine fractions, which are more effective in combination than alone. To characterize the complex scent signal, the lipophilic extracts from urine of the two sexes were analyzed by GC-MS and compared. The GC profile of the males is distinguished by pyrazine compounds not detected in the profile of the females. The profiles of the sexes also differ with regard to several volatile monocarboxylic acids, which are present at higher concentrations in male than in female urine. More than 30 urine components have been identified. Synthetic equivalents of these urine components were bioassayed for effectiveness in eliciting chinning and compared with one another as well as with scent substances not normally present in tree-shrew urine. Strong chinning responses were elicited by (1) certain pyrazine compounds and (2) some monocarboxylic acids, when presented at the high concentrations specific to male urine. Marking behavior is usually not elicited by scent substances not contained in tree-shrew urine or by urine components common to mammals in general. The data so far available indicate that the male-specific scent signal of tree shrews is based less on a single unique component than on the qualitative and quantitative characteristics of a multicomponent mixture. In the bioassay, tree shrews of both sexes respond equally to the male specific substances by chinning. As the scent signal represented by these substances has a different meaning to males and females (rival or potential mate, respectively), chinning probably serves several different functions.  相似文献
