A study of 234 Holstein cows was conducted to determine if early postpartum treatments of gonadotropin-releasing hormone, prostaglandin F2 alpha, or both would alter frequency and occurrence of estrus and ovulation as well as subsequent fertility. Cows in groups 1 and 2 received gonadotropin-releasing hormone (200 micrograms) between 10 and 14 d postpartum, and cows in groups 3 and 4 received saline. Ten days later, cows in groups 2 and 3 received prostaglandin F2 alpha (25 mg), and cows in groups 1 and 4 received saline. Treatment with gonadotropin-releasing hormone reduced intervals to first ovulation and first detected estrus as well as increasing the proportion of cows with three or more ovulations before first service from 57% for saline-treated controls to 83%. Treatment with prostaglandin F2 alpha reduced intervals to second and third ovulation and shortened the first estrous cycle. Treatments for cows in groups 1 and 3 increased the proportion of cows having estrous cycles of normal duration. Interval from calving to conception was reduced by 43 to 48 d for cows with an abnormal puerperium treated with either gonadotropin-releasing hormone or prostaglandin F2 alpha compared with controls (group 4) and by 27 to 29 d overall in all cows in groups 1 and 3. Cows with normal or abnormal puerperium in groups 1 and 3 required 26 to 41% fewer services per conception than controls. We conclude that treatments of gonadotropin releasing hormone or prostaglandin F2 alpha, but not the treatment combination, improved fertility of dairy cows, especially those that experienced puerperal problems.  相似文献

Prostaglandin F2 alpha was used to induce estrus in postpartum dairy cows to decrease calving interval and time spent on heat checks. The amount of prostaglandin residue in milk after treatment also was investigated. Twenty-two control cows, checked for heat twice daily, were bred at estrus after 55 days postpartum. Sixty-three cows checked on days 55 to 60 were either bred if in estrus or treated with 30 mg of prostaglandin F2 alpha injected intramuscularly on day 60 and bred at induced estrus or at 75 h posttreatment. Twelve cows were in heat between days 55 and 60 and were bred. Twenty-nine of 51 treated cows were seen in estrus; all of these were bred and 51.7% conceived at this time. Treated cows had fewer days to first service than did controls, but days open were not different, 105.7 versus 101.8. Fertility was similar. Controls required more heat checks than did treated cows (39 to 8.2). Milk from 17 cows at two milkings before and ten milkings after treatment had similar concentrations of prostaglandin (698 +/- 27 pg/ml) except for the first posttreatment milking when prostaglandin concentration almost doubled (1.293 +/- 143 pg/ml).  相似文献

Two experiments were performed in two different herds to determine if utilizing prostaglandin F 2 alpha to induce estrus for first services would be effective in reducing the duration and variability of calving intervals. In Experiment 1, cows were assigned randomly as controls (n = 217) to be inseminated as they were detected in estrus (beginning d 42 to 53 postpartum depending on replicate) or treated with prostaglandin F2 alpha (n = 185). In Experiment 2, the same treatments were utilized, except control cows (n = 124) were inseminated at their first detected estrus after d 40 postpartum, and treated cows received either one injection of prostaglandin F2 alpha between d 54 and 63 (n = 116) or were given progesterone (via a progesterone-releasing intravaginal device) for 7 d, with the device removed 24 h after prostaglandin F2 alpha (n = 116). More cows were inseminated and pregnancy rates were higher within 5 d after treatment with prostaglandin F2 alpha, and interval from prostaglandin F2 alpha to first service was reduced compared with that of control cows. Duration and variation of calving intervals were unaffected in either experiment, despite the fact the elective waiting period was 6 to 23 d longer for treated cows than for controls. Prebreeding treatment with progesterone failed to improve conception rates, but the efficiency of estrous expression increased from 54% in prostaglandin F2 alpha-treated cows to 71% in those cows also receiving prebreeding progesterone. We conclude that prostaglandin F2 alpha was ineffective in improving reproductive performance of these herds under good herd management. However, prostaglandin F2 alpha offers the convenience of inseminating small groups of cows, controlling when breedings occur during the work week, and prolonging the elective waiting period without extending the calving interval.  相似文献

Data from previous experiments in our laboratory indicated that estrous cycles following synchronization with two injections of prostaglandin F2 alpha often averaged more than 21 d. To investigate whether this could be an effect of prostaglandin F2 alpha, 54 mature heifers and nonlactating Holstein cows were used in a crossover experiment in which estrous cycles either occurred spontaneously or followed two injections of prostaglandin F2 alpha at a 14-d interval (induced estrus). One estrous cycle of each type was recorded for each animal. Blood samples were collected twice weekly for analysis of progesterone. Cattle were observed twice daily for estrus with the aid of tail chalking and an androgenized cow. Estrous cycles following induced estrus were longer than those following spontaneous estrus (24.2 vs. 22.7 d). Ninety-two percent of the cycles following spontaneous estrus were 17 to 25 d in duration compared with 71% of the cycles following induced estrus. If the first prostaglandin F2 alpha was given when progesterone was greater than 1 ng/ml, the estrous cycle following treatment was longer than if the first prostaglandin F2 alpha was given when progesterone was less than 1 ng/ml (26.0 vs. 22.6 d). This experiment demonstrates that two injections of prostaglandin F2 alpha increase the incidence of estrous cycles greater than 25 d. Stage of the estrous cycle at time of the first injection of prostaglandin F2 alpha may contribute to this effect.  相似文献

A field study using 322 lactating dairy cows in seven commercial and two university herds was conducted to determine if treatment with estradiol benzoate 40 to 48 h after treatment with prostaglandin F2 alpha would enhance synchronization of estrus. Estrogen treatment tended to increase the proportion of cows in estrus within 5 d (synchronized) after prostaglandin treatment (66.9% versus 58.9%). Of synchronized cows, a greater proportion treated with estrogen (66.9%) were in estrus on d 3 than those not receiving estrogen (48.2%). First service conception rate (31.9%) and interval to second service (35.6 d) were not affected by treatment with estrogen. Milk progesterone was measured in university herds. More cows with milk progesterone concentrations greater than or equal to 8 ng/ml were synchronized (75.4%) than those with less than 8 ng/ml (63.3%). Treatment with estrogen increased synchrony of cows with high progesterone (90.3%) more than prostaglandin alone (60.0%). Based on progesterone concentrations at breeding and 22 to 24 d later, estimated conception rate was 58.7%, and net conception rate based on palpation was 41.3%. Tighter synchrony of estrus can be achieved by using estradiol benzoate 40 to 48 h after prostaglandin. Concentrations of milk progesterone might predict success of treatment.  相似文献

Concentrations of reproductive hormones and ovarian changes were monitored every 4 days during the postpartum period in 35 dairy cows. The cows were classified as either control (22 cows) or cystic (13 cows) based on per rectum examination of the ovaries. Control cows were detected with a corpus luteum after the first postpartum ovulation while in cystic cows, ovarian follicles of at least 2.5 cm in diameter persisted in the absence of a corpus luteum for at least 14 days. The mean postpartum interval to the first follicle 1.5 to 2.0 cm in diameter was about 16 days for both groups. Ovulation was detected earlier postpartum for cows in the control group (18.4 +/- 1.7 days) than for cows in the cystic group (36.3 +/- 2.7 days). Luteinizing hormone in plasma increased during the early postpartum period (days 1 to 10) for both groups, but mean concentrations of luteinizing hormone, progesterone, and estradiol-17 beta in plasma were not different between groups then or on days -8, -4, or 0 (day of the first postpartum follicle 1.5 to 2.0 cm in diameter). During this period (days -8 to 0), estradiol-17 beta and luteinizing hormone in plasma were correlated positively, but regression coefficients differed between groups. These results suggest a partial hypothalamic and/or pituitary failure in releasing pituitary luteinizing hormone as a cause for postpartum ovarian cysts.  相似文献

Reproductive efficiencies were compared among herds with (treated, n = 25) or without (visited control, n = 25) routine synchronization of estrus and herds without investigator visits (unvisited control, n = 22). Treatment groups were subdivided based on participation in veterinary herd health programs. Reproductive tracts were palpated rectally at 21-d intervals for 6 mo in two sets of treated and visited control herds. Data were collected during the visitation period and from the 6-mo period before visits. In treated herds, cows more than 40 d postpartum with a corpus luteum received prostaglandin F2 alpha. In visited control herds, cows received prostaglandin F2 alpha after 82 d postpartum if they were not observed in estrus. Routine synchronization of estrus did not enhance herd reproductive efficiency compared with no synchronization of estrus in visited herds. Small changes in reproductive efficiency occurred during visits in herds with an established reproductive health program. However, herds not using a health program had 15.2 fewer d from calving to insemination, and the pregnancy rate at 120 d postpartum was 13.0 percentage units higher than before herd visits. Consequently, reproductive performance of those herds during visits was similar to that of contemporaries under a herd health program.  相似文献

The complex interplay of physiological factors that underlies fertility in dairy cows was investigated using a mechanistic mathematical model of the dynamics of the bovine estrous cycle. The model simulates the processes of follicle and corpus luteum development and its relations with key hormones that interact to control these processes. Several factors may perturb the regular oscillatory behavior of a normal estrous cycle, and such perturbations are likely the effect of simultaneous changes in multiple parameters. The objective of this paper was to investigate how multiple parameter perturbation changes the behavior of the estrous cycle model, so as to identify biological mechanisms that could play a role in the development of cystic ovaries. Cystic ovaries are a common reason for reproductive failure in dairy cows, but much about the causes of this disorder remains unknown. We investigated in which region of the parameter space the model predicts a normal cycle, and when a progesterone pattern occurred with delayed ovulation (indicating a cystic follicle) or delayed luteolysis (indicating a persistent corpus luteum). Perturbation of the initial values for all parameters simultaneously showed 2 specific parameter configurations leading to delayed ovulation or delayed luteolysis immediately. The most important parameter changes in these 2 configurations involve the regulation of corpus luteum functioning, luteolytic signals, and GnRH synthesis, suggesting that these mechanisms are likely involved in the development of cystic ovaries. In the multidimensional parameter space, areas exist in which the parameter configurations resulted in normal cycles. These areas may be separated by areas in which irregular cycle patterns occurred. These irregular patterns thus mark the transition from one stable (normal) situation to another. Interestingly, within a series, there were some cycles with delayed ovulation and some with delayed luteolysis in these patterns. This could represent a situation of resumption of normal cyclicity (e.g., after parturition). In conclusion, the method of parameter perturbation used in the present study is an effective tool to find parameter configurations that lead to progesterone profiles associated with delayed ovulation and delayed luteolysis. Thereby, the model helps to generate hypotheses regarding the underlying cause of the development of cystic ovaries, which could be investigated in future experiments.  相似文献

The objective of this study was to characterize the estrous cycle of cows with similar proportions of Holstein genetics, similar genetic merit for milk production traits, but with good (Fert+) or poor (Fert-) genetic merit for fertility traits. In total, 37 lactating cows were enrolled on a protocol to synchronize estrus. Nineteen Fert+ and 12 Fert- cows that successfully ovulated a dominant follicle and established a corpus luteum underwent daily transrectal ultrasonography. Blood sampling was carried out at 8-h intervals from d 0 to 6 and from d 15 to ovulation, and once daily from d 7 to 15. Blood samples were analyzed for progesterone, estradiol, follicle stimulating hormone, and luteinizing hormone. Estrus behavior was recorded using neck activity collars and mounting pads. The Fert+ cows tended to have fewer follicular waves (2.2 vs. 2.7) and had a shorter estrous cycle (21.0 vs. 25.1 d) than Fert- cows. We observed no effect of genotype on day of first-wave emergence or day of first-wave dominant follicle peak diameter, but the peak diameter of the first-wave dominant follicle tended to be larger in Fert- cows. During the first 13 d of the cycle, Fert+ cows developed a corpus luteum that was 16% larger than that in Fert- cows. Circulating progesterone concentrations were 34% greater in Fert+ than in Fert- cows (5.15 vs. 3.84ng/mL, respectively) from d 5 to 13. During the final follicular wave, the interval from preovulatory follicle emergence to ovulation and the interval from preovulatory follicle dominance to ovulation were similar in both genotypes. Maximum preovulatory follicle diameter was larger in Fert+ than Fert- cows (17.9 vs. 16.8mm, respectively); however, circulating concentrations of estradiol were not different between genotypes. A greater proportion of Fert- cows ovulated to a silent heat than Fert+ cows (22 vs. 2%, respectively). Of cows that showed behavioral estrus, Fert+ cows had 41% greater mean activity count; however, no difference was seen in mounting behavior between genotypes. These results demonstrate, for the first time, that genetic merit for fertility has pronounced effects on corpus luteum development, progesterone concentration, preovulatory follicle diameter, and behavioral estrus.  相似文献

Two experiments examined physiological changes throughout estrous cycles of Holstein cows. In experiment one, changes were characterized for physical activity, vaginal pH, vaginal temperature, milk yield, heart rate, and concentration in blood plasma of progesterone and 13,14-dihydro-15-keto-prostaglandin F2 alpha. Variables were measured daily in 11 cows through a total of 28 estrous cycles. Cows in estrus at least once postpartum were used. Observations for estrus were at least twice daily. Variables changed significantly with day of estrous cycle. Changes of progesterone, physical activity, and 13,14-dihydro-15-keto-prostaglandin F2 alpha were more marked around estrus than were changes of the other variables. Peaks of physical activity coincided with estrus in 75% of the cases. Experiment two was to determine if increased physical activity, measured with pedometers, coincided with estrus in commercial dairy herds. Cows on 14 farms in New York were fitted with mechanical pedometers, and the pedometers were read at each milking. Readings from 3 days before until 3 days after estrus were used from 55 estrous periods identified by the producers. Physical activity was maximal on day of estrus in 73% of the estrous periods. Increased physical activity of commercial dairy cows agreed well with the producers' diagnoses of estrus. Pedometers could be a valuable component of an estrous detection program.  相似文献

The objectives of this study were to evaluate the effect of 3 methods of resynchronization of estrus and ovulation for lactating dairy cows of unknown pregnancy status on conception rate and time to pregnancy. Holstein cows (n = 495) were randomly assigned to 1 of 3 treatments: 1) control (n = 167), resynchronization with a timed AI protocol upon diagnosis of nonpregnancy on d 31 after preenrollment AI (PAI); 2) CIDR-G (n = 159), use of an intravaginal progesterone insert from d 14 to 21 after AI, with AI at estrus from d 21 to 24 and initiation of a timed AI protocol on d 24 after AI in cows not reinseminated; 3) CIDR-G + ECP (n = 169), the same treatment as CIDR-G but with an injection of 1 mg of estradiol cypionate at the time of progesterone insert removal. Cows were continuously reenrolled in the same treatment until diagnosed as pregnant, which resulted in a total of 1,148 AI (495 PAI and 653 resynchronized AI; RAI). Blood was collected from 1,001 cows on d 14, 21, and 24 after each AI for analysis of progesterone, and ovaries were scanned on d 21, 24, and 31 after AI. The presence of an active corpus luteum was presumed based on progesterone ≥1 ng/mL. Pregnancy was diagnosed by ultrasonography on d 31 and 61 after AI. The presence of an active corpus luteum and the incidence of luteolysis were similar for all treatments from d 14 to 24; however, luteolysis increased in the CIDR-G + ECP treatment from d 21 to 24. Conception rates for the PAI and all AI were similar on d 31 and 61 after insemination. Conception rates at 31 and 61 d after the RAI were also similar among treatments. Overall pregnancy loss for the PAI, RAI, and all AI were similar for all treatments. The accuracy of estrous detection, based on progesterone concentration within 2 d of detection of estrus, was similar for all treatments for the RAI and averaged 95.3%. The estrus-detection rate (EDR) decreased for the CIDR-G and CIDR-G + ECP treatments from d 14 to 21, but increased from d 21 to 24 compared with control cows; however, the EDR was smaller for cows in the CIDR-G treatment during the entire resynchronization period compared with those in the CIDR-G + ECP and control groups. The reinsemination interval was reduced in cows receiving the CIDR-G + ECP treatment compared with control cows because of increased EDR after removal of the intravaginal insert; however, the interval from study enrollment to pregnancy was not different among treatments. These results indicate that the reproductive performance of dairy cows did not differ among the 3 resynchronization treatments evaluated.  相似文献

Our objective was to determine if calving intervals could be shortened and made less variable by using prostaglandin F2 alpha to control the occurrence of first services. Holstein cows (n = 348) were assigned at calving to four treatment groups. Control cows (n = 88) were inseminated at their first observed estrus after 40 d postpartum. Estrous cycles of the remaining cows were synchronized with prostaglandin F2 alpha to allow insemination (first services) 80 h after the second injection (n = 86), insemination at 80 h preceded by gonadotropin-releasing hormone at 72 h (n = 86), or insemination at 72 and 96 h (n = 88) after the second injection (51 to 57 d postpartum). By design, interval to first service was reduced to 57 d for treated cows (63 d for controls) and was less variable (12% of that for controls). Conception rate at first service was lower after timed inseminations than that of controls. Intervals to conception and subsequent calving were similar in all treated cows and controls. Of cows sampled, 23 of 176 (13%) failed to respond with luteolysis when progesterone in serum exceeded 1 ng/ml, and 26 of 176 (15%) had low concentrations of progesterone in serum and could not respond to prostaglandin F2 alpha. Poor response to timed inseminations may have occurred because only 72% of 176 cows sampled responded with luteolysis. Dairy cows (n = 571) were treated with 1) one or two injections of prostaglandin F2 alpha given 11 or 14 d apart, 2) two injections given 14 d apart and a progesterone coil inserted 8 d after the first injection for a 7-d period; or 3) a coil inserted for 7 d and prostaglandin injected 1 d before its removal. Cows given two prostaglandin injections that conceived had higher progesterone concentrations during the luteal phase preceding AI than did cows that did not conceive. Cows with progesterone coils that conceived did not have higher progesterone concentrations than did cows that did not conceive. Older cows had lower progesterone concentrations than younger ones, and they appeared in estrus earlier after treatment. In four daily observations, 75% of cows were seen in estrus within 7 d after treatment. Cows mounting two or more times had a conception rate (62%) similar to that of cows that stood to mount. Fewer cows (56%) treated with prostaglandin that had low concentrations of progesterone appeared in estrus than did cows with high progesterone (84%). Cows treated with prostaglandin differ from cows treated with progesterone coils in respect to manifestation of estrus and to the relationship between plasma progesterone and conception.  相似文献

Testosterone in plasma was measured by radioimmunoassay in cows with ovarian cysts and in cows during the estrous cycle. Concentrations of testosterone in plasma (60.8 +/- 2.7 pg/ml; mean +/- standard error) in cows with ovarian cysts were variable but within the range in cows during the estrous cycle. Mean concentrations of testosterone in plasma increased from 46.3 pg/ml at estrus to 85.1 pg/ml on day 13 of the estrous cycle. In cows with ovarian cysts, concentrations of testosterone in plasma declined within 24 h following prostaglandin F2 alpha and tended to increase by 9 days following gonadotropin releasing hormone. The role of testosterone in estrual behavior and inhibiting resumption of ovarian cycles is likely minimal.  相似文献

The pathophysiology underlying follicular cysts appears to be lack of an estradiol (E2)-induced GnRH/LH surge due to hypothalamic insensitivity to E2. In addition, progesterone (P4) can cause animals with follicular cysts to resume normal cyclicity and normal hypothalamic responsiveness to E2. We postulated that follicular cysts may be a pathological manifestation of a physiological state that cows, and possibly other species, go through during the normal estrous cycle but the rise in P4 following ovulation induces them back to normal hypothalamic responsiveness to E2. Based on this hypothesis, we expected that removal of the ovary containing the corpus hemorrhagicum would prevent the normal rise in P4 following ovulation and induce development of follicular cysts. Cows (n = 24) on day 7 of the estrous cycle were treated with prostaglandin F2alpha (PGF2alpha) and time of ovulation was detected by ovarian ultrasonography every 8 h. Immediately following detection of ovulation, cows were randomly but unequally assigned to have the ovary containing the corpus hemorrhagicum removed (TRT; n = 16) or the ovary opposite to the corpus hemorrhagicum removed (CON; n = 8). Cows were subsequently evaluated by daily ultrasound and blood sampling to determine follicular dynamics. Ovulation was detected at 93.7 +/- 4.5 h after PGF2alpha injection. All CON cows had a normal estrous cycle length (22.0 +/- 0.6 days) after ovariectomy (OVX). Half of the TRT cows became anovular (TRT-ANO; n = 8) after OVX with large anovular follicles developing on the ovary (maximal size, 25.4 +/- 1.4 mm; range, 20-32 mm). However, eight TRT cows ovulated (TRT-OV; n = 8) 7.3 +/- 0.6 days after OVX. Control cows had a normal P4 rise after ovulation. Removal of the newly formed corpus hemorrhagicum prevented the rise in circulating serum P4 in TRT-ANO cows throughout the 25-day experimental period. The TRT-OV cows had a delayed increase in circulating P4 but it was normal in relation to time of ovulation. Serum E2 concentrations were similar among groups (TRT-OV, TRT-ANO and CON cows) until 7 days after OVX. Serum E2 concentrations then decreased in TRT-OV and CON cows but remained elevated (>5 pg/ml) in TRT-ANO cows. Thus, the endogenous increase in circulating E2 that induces the GnRH/LH surge and estrus is sufficient to induce cows into a physiological state that resembles follicular cysts if it is not followed by increased circulating P4.  相似文献

The objective of this study was to determine whether organically complexed Co, Cu, Mn, and Zn would improve the reproductive performance and milk and milk component production in lactating dairy cows that began receiving bovine somatotropin in the ninth week of lactation. Holstein (n = 50) and Jersey (n = 10) cows were blocked by breed, lactation number, and incidence of retained fetal membranes. Two diets assigned within blocks and fed from parturition until 154 d of lactation were control or control supplemented daily with 26 mg of Co as Co glucoheptonate, 125 mg of Cu as Cu-Lys, 199 mg of Mn as Mn-Met, and 359 mg of Zn as Zn-Met. Cows were fitted with electronic pressure-sensing devices in the second week of lactation for detection of estrus. Ovarian structures were determined via transrectal ultrasonography at 7-d intervals from parturition until observation of the first corpus luteum. Blood samples were taken at 7-d intervals and analyzed for plasma concentrations of progesterone, insulin, and urea nitrogen. Onset of luteal activity was identified by progesterone concentrations > or = 1 ng/ml. Retained fetal membranes increased days to first estrus (detected via electronic estrous detection), first luteal activity, and first corpus luteum in control cows but not in supplemented cows. Days to first observed estrus were greater for control cows than for supplemented cows. Days to first service, days open, days from first service to conception, services per conception, milk yield, milk components, and somatic cell counts were similar for control and supplemented cows. Supplementation with complexed trace minerals effectively reduced days to first estrus.  相似文献

A comparison was made between two different methods of reproductive management in the dairy cow. One protocol administered prostaglandin to open cows weekly; the other administered prostaglandin to open cows with a corpus luteum identified by rectal palpation. Survival analysis was used to analyze the data. Cows receiving weekly doses of prostaglandin had a 30% higher pregnancy rate (number of pregnancies per time) than the cows receiving prostaglandin based on rectal palpation of a corpus luteum. The average number of days to first insemination was shorter in cows given prostaglandin weekly.  相似文献

The objectives of this retrospective observational study were to determine the associations of anogenital distance (AGD) with (a) postpartum estrous activity, (b) diameter of the preovulatory follicle, (c) intensity of estrous expression, (d) postestrus ovulation, (e) corpus luteum (CL) size, and (f) concentrations of progesterone at estrus and on d 7 after estrus. Lactating Holstein cows (n = 178; 55 primiparous, 123 multiparous) were enrolled into the study during the first postpartum week. All cows were continuously monitored by a pedometer-based automated activity monitoring (AAM) system for estrus. Postpartum estrous activity was assessed using the AAM estrus alerts, in which cows with at least one true estrus alert (i.e., a relative increase in steps from each cow's baseline detected by the AAM and the presence of at least one follicle >15 mm, a CL <20 mm, or no CL detected by ultrasound) by the first 50 d in milk (DIM) were considered to have commenced estrous activity. At the estrus alert >60 DIM, ovulation was determined by ultrasound at 24 h, 48 h, and 7 d after estrus, and blood samples were collected at estrus alert and on d 7 after estrus for progesterone analysis. The AGD was measured from the center of the anus to the base of the clitoris and classified as either short- or long-AGD using 2 cut-points of 148 mm (predictive of the probability of pregnancy to first insemination; short-AGD, n = 115; long-AGD, n = 63) and 142 mm (the median AGD; short-AGD, n = 90; long-AGD, n = 88). Regardless of the cut-point used, early postpartum estrous activity by 50 DIM (67 vs. 54%), duration of estrus (11.6 vs. 9.7 h), and preovulatory follicle diameter (20 vs. 19 mm) were greater in short-AGD than in long-AGD cows. Increased peak of activity at estrus in short-AGD cows (354 vs. 258% mean relative increase) was affected by an interaction between AGD and parity in which multiparous long-AGD cows had lesser relative increase in activity than primiparous cows (217 vs. 386%, respectively). Mean progesterone concentration at estrus was lesser in short-AGD (0.47 vs. 0.61 ng/mL) than in long-AGD cows. The ovulatory response at 24 h did not differ, but at 48 h (91 vs. 78%) and on d 7 after estrus (97 vs. 84%) it was greater in short-AGD cows. Although CL diameter on d 7 after estrus did not differ, short-AGD cows had greater progesterone concentration 7 d after estrus than long-AGD cows (4.1 vs. 3.2 ng/mL, respectively). In conclusion, greater proportions of short-AGD cows commenced estrous activity by 50 DIM, had larger preovulatory follicles, exhibited greater duration of estrus, had reduced progesterone concentration at estrus, had greater ovulation rates and progesterone concentration 7 d after estrus compared with long-AGD cows, with no difference in CL size between AGD groups. Because all the differences in physiological characteristics of short-AGD cows reported herein favor improved reproductive outcomes, we infer that these are factors contributing to improved fertility reported in short-AGD cows compared with long-AGD cows.  相似文献

The objective of this study was to examine the association between increased physical activity at the moment of timed artificial insemination (AI), detected by an automated activity monitor (AAM), and fertility outcomes. This paper also investigated factors affecting estrous expression in general. A total of 1,411 AI events from 1,040 lactating Holstein cows were recorded, averaging 1.3 ± 0.6 (±standard deviation) events per cow. Activity (measured as steps/h) was monitored continuously by a leg-mounted AAM located on the rear leg of the cow. Ovulation was synchronized by a timed AI protocol based on estradiol and progesterone. Ovarian ultrasonography was performed in all cows on d ?11 (AI = d 0) and in a subset of cows on d 0 (n = 588) and d 7 (n = 819) to determine the presence of a corpus luteum and follicles. The body condition score (1 to 5 scale) was assessed on d 0 and a blood sample was collected for progesterone measurement on d 7. Using the AAM, an estrus event was determined when the relative increase (RI) in physical activity of the cow exceeded 100% of the baseline activity. The physical activity was classified as strong RI (≥300% RI), moderate RI (100–300% RI), or no estrus (<100% RI). Milk production was measured daily and averaged between d ?11 and 0. Pregnancy was diagnosed at 32 and 60 d post-AI and pregnancy losses were calculated. The mean RI at estrus was 328.3 ± 132.1%. Cows with strong RI had greater pregnancy per AI than those with moderate RI and those that did not express estrus (35.1 vs. 27.3 vs. 6.2%). When including only cows that successfully ovulated after timed AI, those that displayed strong intensity RI still had greater pregnancy per AI than those with moderate intensity RI or those that did not express estrus (45.1 vs. 34.8 vs. 6.2%). Cows expressing strong RI at timed AI had greater ovulation rates compared with moderate RI and cows that did not express estrus (94.9 vs. 88.2 vs. 49.5%). Furthermore, pregnancy losses were reduced in cows with strong RI compared with cows expressing moderate RI (13.9 vs. 21.7%). Cows with a strong RI at estrus were more likely to have a corpus luteum at the beginning of the protocol and had greater concentration of progesterone 7 d post-AI. Multiparous cows expressed lower RI compared with primiparous cows. Cows with lower body condition score tended to have decreased RI at estrus. No correlation between estrous expression and pre-ovulatory follicle diameter was observed. Also, no correlation was observed between milk production at AI and RI. In conclusion, strong intensity RI of estrus events at timed AI was associated with improved ovulation rates and pregnancy per AI, and reduced pregnancy losses. These results provide further evidence that measurements of estrous expression can be used to predict fertility at the time of AI and possibly be used as a tool to assist decision making strategies of reproduction programs.  相似文献

The objective of this field trial was to compare the effect of intrauterine (i.u.) antibiotic or intramuscular (i.m.) prostaglandin F2 alpha (PGF2 alpha) on time to pregnancy in dairy cows diagnosed with clinical endometritis between 20 and 33 days in milk (DIM). The case definition of endometritis was the presence of purulent uterine discharge or cervical diameter > 7.5 cm, or the presence of muco-purulent discharge after 26 DIM. There were 316 cows with endometritis from 27 farms assigned randomly within herd to receive 500 mg of cephapirin benzathine intrauterine (i.u.), 500 micrograms of cloprostenol i.m., or no treatment. The rate of resolution of clinical signs 14 d after treatment was 77% and was not affected by treatment. Reproductive performance was monitored for a minimum of 7 mo after treatment. Survival analysis (multivariable proportional hazards regression) was used to measure the effect of treatment on time to pregnancy. There was no benefit of treatment of endometritis before 4 wk postpartum. Administration of PGF2 alpha between 20 and 26 DIM to cows with endometritis that did not have a palpable corpus luteum was associated with a significant reduction in pregnancy rate. Between 27 and 33 DIM, cows with endometritis treated with cephapirin i.u. had a significantly shorter time to pregnancy than untreated cows (hazard ratio = 1.63). In this time period, there was no difference in pregnancy rate between PGF2 alpha and untreated cows, but the difference in pregnancy rate between cows treated with cephapirin i.u. and with PGF2 alpha was not statistically significant. Treatment of postpartum endometritis should be reserved for cases diagnosed after 26 DIM, based on criteria that are associated with subsequent pregnancy rate.  相似文献
