Leaves and products made from them are the major focus of research on bioactive compounds of mate (Ilex paraguariensis). However, studies on other parts of the plant, are scarce. The aim of this study was to determine the methylxanthines, phenolic compounds, and antioxidant activity of mate bark (residual biomass) and compare with those of mate leaves. The high antioxidant activity of mate bark and its high concentration of total polyphenols were apparent in both the aqueous and the methanolic extracts, the values of which were greater than those detected in the leaves. Of the phenolic acids identified, the levels of chlorogenic acid and 4,5-dicaffeoylquinic acid in the samples were significantly higher in the methanolic bark extract. With regard to methylxanthines, considerable concentrations were detected in the samples. The results obtained contribute to the viability of exploiting this residue, broadening the use of the mate plant.  相似文献

The aim of this study was to evaluate the effects of freeze concentration of strawberry juice on content of total phenolic, anthocyanins content and antioxidant activity of the concentrated fluid and the ice obtained. An increase in the values of phenolic content was observed in the concentrate fraction for all the freeze concentration stages (five stages). The process efficiency presented a reduction when compared with the first stage, and however, the average efficiency remained around 65% in all the stages. The concentration of pelargonidin‐3‐O‐glycoside present in samples was increased in all stages. Furthermore, the antioxidant activity of the concentrated fluid of each stage, measured by DPPH and ABTS methods, was significantly higher in comparison with the juice feed. Also, antioxidant activity can be correlated with the content of pelargonidin‐3‐O‐glycoside presented in the strawberry juice. Freeze concentration can be considered a good alternative for improving the antioxidant activity of strawberry juice.  相似文献

The aim of this study was to characterise the bioactive compounds in mate (Ilex paraguariensis St. Hil) extract and in concentrated mate extract obtained by nanofiltration (NF). Also, the impact of NF on the antioxidant activity of both mate extracts was evaluated in vitro and using eukaryotic cells of Saccharomyces cerevisiae (yeast assay). The results showed a significant increase in the contents of total phenolics (338%), chlorogenic acid (483%), theobromine (323%), caffeine (251%), chlorophyll (321%), condensed tannins (278%) and saponins (211%) in the concentrated mate extract. The concentrated mate extract showed higher in vitro antioxidant activity than the mate extract. According to the results obtained, it can be stated that the use of nanofiltration membrane is a valid approach for the concentration of biologically active compounds in aqueous extract of mate.  相似文献

ABSTRACT: Ilex paraguariensis is known to contain compounds with antioxidant properties, such as phenolic acids, and its stimulant properties are attributed to methylxanthines, such as caffeine. The aims of this study were to evaluate the phenolic, methylxanthinic, and tannin composition of a mate residue (mate powder), to compare the quali-quantitative phenolic composition and the antioxidant potential of extracts obtained from distinct solvent systems. Among the extracts prepared with different solvents, the 80% methanol extract showed the highest total polyphenol content (11.51 g/100 g) and antioxidant activity. HPLC analysis showed that 4,5 dicaffeoylquinic acid is the major component of the phenolic fraction of mate powder. The caffeine, theobromine, and tannin contents in mate powder were 1.01, 0.10, and 0.29 g/100 g, respectively. Consumption of mate powder would significantly contribute to antioxidant and stimulant intake, providing high amounts of phenolic acids, tannins, and methylxanthines with biological effects potentially beneficial for human health. Practical Application: This article contributes to the minimization of residues in yerba-mate processing.  相似文献

The nutrient profile and the phytochemical composition of the sapucaia nut cake milk (SNCM) were evaluated after applying five consecutive stages of block freeze concentration (C1, C2, C3, C4 and C5). The profile of phenolic compounds by LC-ESI-MS/MS and the antioxidant activity (ABTS and FRAP) of SNCM were also determined. The freeze concentration increased proteins, lipids, carbohydrates, ash, soluble solids (°Brix) and total phenolic compounds (TPC) in all stages and in the ice fractions obtained from C1, C2 and C3 stages. The antioxidant activity (ABTS) in C3, C4 and C5 fractions was highly significant (P < 0.05). The antioxidant activity (FRAP) and the phenolic compounds (gallic, vanillic, ferulic, sinapic and salicylic acids, catechin, taxifolin and sinapaldehyde) increased significantly (P < 0.05) for all five fractions. The freeze concentration was successfully used for enriching the SNCM, generating a promising ingredient for the food industry.  相似文献

In order to establish the parameters to obtain aqueous mate extract with the highest content of total phenolic compounds (TPC), the response surface methodology (RSM) was used. The extract obtained at 100 °C, within 3 min and with pH of 6.0 was submitted to nanofiltration to concentrate the phenolic compounds of mate (Ilex paraguariensis A. St. Hill). The total phenolic content values in the permeate and in the concentrate collected in different volumetric reduction factors (VRF) were different from those detected in the feed extract. The concentration of phenolic compounds increased when VRF was increased, reaching the highest retention index (R) of 0.99 when VRF was 4.0. The permeate and the concentrate obtained in VRF 4.0 were verified through HPLC. 4,5-dicaffeoylquinic acid and gallic acid showed retention index of 1.00 and 0.95, respectively, while 3,4-dihydroxybenzoic acid and chlorogenic acid reached 0.99 and 0.98, respectively.  相似文献

The aim of this research was to investigate the effect of the addition of peel powder and/or its aqueous extract on physical and antioxidant characteristics of carboxymethyl cellulose (CMC) edible films. Prickly pear peel powder and aqueous extract (2 and 4%) were characterized in color, bioactive compounds (betalains and total phenolic compounds), antioxidant capacity and reducing power. Edible films were prepared with CMC, glycerol, varying the dissolution medium (0, 2, or 4% aqueous extract from prickly pear peel) and peel powder (0, 1, or 2%) using a face-centered central composite design. Physical, mechanical, bioactive compounds, and antioxidant capacity properties were investigated in edible films. Prickly pear peel powder presented 58.8 ± 0.08 mg of betacyanins/100 g, 53.8 ± 0.2 mg of betaxanthins/100 g, 967.8 ± 20 mg Gallic acid equivalent (GAE)/100 g for total phenolic compounds, an antioxidant capacity equivalent to 420.9 ± 4.9 mg GAE/100 g and a reducing power of 879.1 ± 115.9 mg ascorbic acid equivalent (AAE)/100 g. High concentration of aqueous extract and peel powder in CMC films increased the bioactive compounds content, antioxidant capacity and reducing power. Films mechanical properties were not affected by the aqueous extract; nevertheless, were strongly affected by the peel powder. Response surface methodology was used to optimize edible films formulation with high antioxidant characteristics, being the optimal formulation 1.7% of peel powder plus 3.3% of aqueous extract. This study may be the base for exploitation of prickly pear peel as source of bioactive compounds with antioxidant capacity for their use in edible coatings.  相似文献

The popularity of tea is increasing on the global aspect because of its role as a significant source of phenolic compounds in human diet. The purpose of this study was to determine and compare the phenolic and methylxanthine composition as well as the antioxidant capacity of white, green, Oolong and black teas, and chamomile and linden infusions depending on the extraction conditions (water temperature and multiple extractions). The content of total phenols and total flavonoids in teas and herbal infusions was determined by using UV/vis spectrophotometric methods, whilst individual polyphenols (phenolic acids and flavan-3-ols) and methylxanthines were identified and quantified by using high performance liquid chromatography coupled with photodiode array detection. In order to determine the antioxidant capacity of teas the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) radical scavenging assays were applied. The highest content of phenolic compounds was determined in green tea, which also demonstrated the highest antioxidant capacity, whilst herbal infusions were characterised with the lowest content of phenolic compounds, as well as the lowest antioxidant capacity. The highest content of caffeine, as the most abundant methylxanthine, was determined in black tea. Extraction at 100 °C is the most effective to extract the highest content of polyphenols and methylxanthines in all studied teas.  相似文献

Like tea, the leaves of Turkish thyme (Thymus vulgaris) can be boiled in water to produce an extract. This is widely used as syrup for the treatment of coughs and bronchitis at alternative medicine clinics in many parts of the world. In the current study, we assessed the phenolic content and antioxidant activity of thyme. The antioxidant activities of both ethanol and aqueous extracts of thyme were determined using various in vitro methods. The total phenolic and total flavonoid contents were determined to be a gallic acid equivalent and a quercetin equivalent, respectively. Finally, the quantities of the phenolic compounds were detected using high-performance liquid chromatography and tandem mass spectrometry. The total phenolic compounds in the aqueous extract and ethanol extracts of Turkish thyme were 256.0 μg gallic acid equivalent/mg dried extract and 158.0 μg gallic acid equivalent/mg dried extract, respectively. Conversely, the total flavonoid compounds in both extracts were 44.2 μg and 36.6 μg quercetin equivalent/mg dried extract, respectively. For the first time, we determined phenolic contents and investigated the antioxidant potential of thyme. The results indicate that Turkish thyme is a good dietary source with phenolic properties.  相似文献

Although Trifolium pratense (Red Clover) is considered to be one of the leading crops for livestock grazing, it could also be used as a potential source of bioactive compounds in phytopharmacy. The aim of this study was to investigate the phenolic content and its biological activity at the growth phases (30 cm, 50 cm, and bud) of this plant. The phenolic compounds in methanolic extracts of T. pratense leaves at three growth stages, obtained by Microwave Assisted Extraction, were quantified using the HPLC-ESI-MS/MS technique, and their antioxidant and antimicrobial activity were assessed. Isoflavonoids, genistein, and daidzein, as well as other phenols, p-hydroxybenzoic and caffeic acids, kaempferol 3-O-glucoside, quercetin 3-O-glucoside, and hyperoside were found in all the extracts, but the content of these compounds was the highest in the extract of the plant at the lowest growth stage (30 cm, vegetative). Therefore, this extract showed the best antioxidant potential and it was most effective against bacterial strains such as Escherichia coli and Enterobacter aerogenes. These results indicated that red clover has potential health benefits, and that growth phase contributes to its biological activity. The extract of red clover at the growth stage of 30 cm is a great source of bioactive compounds and could be used in phytotherapy and nutrition.  相似文献

The aim of this study is to investigate the influence of certain factors on the yield, antioxidant activity (AA) and total phenolic content (TPC) of guava leaf extract. The effects of pretreatment of leaf sample prior to extract, extraction method, and the leaf age were investigated. Folin-Ciocalteau was used to determine the TPC. Trolox equivalent antioxidant capacity (TEAC) and equilibrium concentration (EC) were used for evaluation of AA. Results indicated that ultrasonication is the most suitable method for guava leaf extraction as it yielded the extract with the significantly highest TPC and AA. Blanching followed by ice water cooling (BCD) was suggested for the pretreatment process of guava leaves. The study of leaf maturity demonstrated that the highest activity was from the young leaves. Hot water was the best solvent to extract the active principles. The extract of BCD pretreated young leaves, extracted by hot water exhibited the highest TPC and AA with the TEAC and EC values of 24.30 ± 0.50 and 20.41 ± 0.67 mM/mg, respectively. These values are 1.88 and 8.72 times higher than the synthetic antioxidant butylated hydroxy toluene (BHT) and 1.75 and 1.21 times higher than vitamin E, respectively. It was concluded that pretreatment and drying process, method of extraction and leaf maturity played important roles on the bioactive compounds and their antioxidant power of guava leaf extract.  相似文献

A purified olive extract (POE) rich in phenolic and oleosidic compounds was prepared from olive mill wastewaters by adsorption onto an amphoteric polymer resin with the yield of 2.2% (w/v). In addition to complex and simple phenolic compounds found in the POE, the highly hydrophilic elenolic acid and DEDA (dialdehydic form of decarboxymethyl elenolic acid, a non-phenolic secoiridoids) were also present in high concentration (45%, w/w). Among the phenolic constituents, the major compounds are hydroxytyrosol and its glucoside, DEDA linked to hydroxytyrosol (Hy-DEDA), p-coumaric acid and caffeic acid, verbascoside and some flavonoids. The high content (23%, w/w in gallic acid equivalent) and structural diversity of these phenolic compounds confer superior antioxidant potential of POE with an IC50 less than 8.5 μg/ml determined by DPPH assay. The stability of bioactive components during storage of the extract and changes in antioxidant properties were studied under different conditions. Determination of kinetics of degradation revealed that air/oxygen is the determinant factor which influences the stability of POE under low temperature storage conditions, while exposure to sunlight did not have significant effect on their stability. Whilst an increase in storage temperature decreased the total content of phenolic compounds (20-24% reduction) and non-phenolic secoiridoid aglycons, a high antioxidant capacity was still observed particularly in the nitrogen-protected POE samples (93-95% of the initial value). This was believed to be due to the transformation of hydroxytyrosol-containing complex phenols, especially the secoiridoid ester Hy-DEDA, into free phenolic monomer. This contributed significantly to the maintenance of a high antioxidant potency of the whole extract during storage. The high antioxidant, radical scavenging activity and stability of the extract suggest potential applications in the food and pharmaceutical industries.  相似文献

The effect of different extraction conditions and storage time of prepared infusions on the content of bioactive compounds of green teas and their antioxidant capacity were investigated. The content of total phenols, total flavonoids and total non-flavonoids in green teas was determined spectrophotometrically, while 7 flavan-3-ols, 6 phenolic acids and 3 methylxanthines were identified and quantified by using high performance liquid chromatography (HPLC–PDA). Among the tested green teas bagged green tea Twinings of London was recognized as the richest source of phenolic compounds (3585 mg/L GAE of total phenols). The most abundant phenolic constituents of green tea were flavan-3-ols, of which EGCG was prevailing in all teas (94.54–357.07 mg/L). The highest content of caffeine, as the most abundant methylxanthine, was determined in powdered green tea. The findings of this investigation suggest that extraction efficiency of studied bioactive compounds from green tea depends on the extraction conditions and that maximum extraction efficiency is achieved during aqueous extraction at 80 °C, for 5′ (powder), 15′ (bagged) and 30′ (loose leaf). In order to determine the antioxidant capacity of teas the DPPH, ABTS and FRAP assays were applied. Regardless of the extraction conditions all green teas exhibited significant antioxidant capacity in vitro, which was in correlation with their phenolic content, confirming that green tea is one of the best dietary sources of antioxidants.  相似文献

为探究干燥方式(冷冻真空干燥、热风干燥)和干燥温度(冻干、40、60、80、100℃)对余甘子多酚活性成分的影响,利用高效液相色谱(HPLC)检测不同干燥处理后余甘子醇提液中没食子酸、柯里拉京、鞣花酸、诃子鞣酸的含量,并测定其总多酚含量和抗氧化活性(ABTS+自由基清除能力和DPPH自由基清除能力)。结果表明,随着干燥温度的升高,余甘子中没食子酸、柯里拉京、鞣花酸、诃子鞣酸含量均增加,100℃热风干燥时达最高(分别为12.83、6.04、16.89、13.48 mg/g)。总多酚含量与抗氧化活性(ABTS+·清除能力和DPPH·清除能力)也随干燥温度升高而增加,80℃热风干燥时达最高(分别为200.92、695.76、725.05 mg/g)。此外,试验结果表明柯里拉京、诃子鞣酸和总多酚的含量与余甘子醇提样品ABTS+自由基清除能力呈显著或极显著正相关(P<0.05或P<0.01)。本次试验中,与热风干燥相比,冷冻干燥的样品中4种多酚单体和总多酚含量以及抗氧化活性无明显优势,综合考虑,80℃热风干燥为余甘子的最佳干燥方式。  相似文献

Dried mate leaves or aqueous extract of dried mate leaves added to chicken meat balls prior to cooking for antioxidant protection was found not to affect taste or smell of the product significantly for addition corresponding to 0.050% of dried mate leaves, while 0.10% changed the smell but not the taste and more significantly for dried leaves. Addition of aqueous mate extract corresponding to 0.050% of mate is recommended as an alternative to more strongly flavoured herbs for oxidative protection of meat products.  相似文献

This study was undertaken to evaluate the functional properties of two of the most popular species of edible bamboo shoots in Korea (Phyllostachys pubescens and Phyllostachys nigra). Powdered bamboo shoots were extracted with methanol and an aqueous suspension of the obtained methanol extract was partitioned successively with chloroform, ethyl acetate, and butanol, leaving a residual water extract. All obtained extracts were evaluated for their antioxidant capacity and antimicrobial activity, angiotensin converting enzyme (ACE) inhibition activity, and ascorbic acid and phenolic compound content. Methanol and water fractions showed a particularly high ascorbic acid contents. The ethyl acetate fraction contained a high concentration of phenolic compounds. Among all extracts, the ethyl acetate and butanol fractions showed particularly high antioxidant activity. Methanol extract had a significantly higher ACE inhibitory activity than other extracts. None of the extracts inhibited the tested bacteria.  相似文献

The aim of this research was to investigate the levels of phenolic compounds and antioxidant activities in different tissues including leaves, stems, and roots from baekseohyang (Daphne kiusiana). The highest contents of total phenolics (43.59 mg gallic acid equivalent, GAE/g) and flavonoids (15.73 mg rutin equivalents, RE/g) were observed in the 75% methanol extract of leaves. Moreover, this extract had the predominant antioxidant capacity, DPPH (85.91%) and ABTS (92.57%) radical scavenging activities as well as reducing power (7.20%) at a concentration of 5 mg/mL. The highest content of phenolic compounds was also exhibited in this extract with an increasing order in leaves, roots, and stems and their major components were vanillic acid (6.37 mg/g), tannic acid (1.91 mg/g), and p-hydroxybenzoic acid (3.96 mg/g). Thus, the strong antioxidant activities of the 75% methanol extract are correlated with high phenolic compound contents. This study suggests that baekseohyang leaves may potentially be used as an accessible source of natural antioxidants.  相似文献

Carqueja (Pterospartum tridentatum) is an endemic species and various bioactive compounds have been identified in its aqueous extract. The aim of this study was to protect the natural antioxidants from the aqueous extract of carqueja by encapsulation in Ca–alginate microbeads and Ca–alginate microbeads containing 10% and 20% (w/v) of inulin. The microbeads produced by electrostatic extrusion technique had an average diameter from 625 μm to 830 μm depending on the portion of inulin. The sphericity factor of the hydrogel microbeads had values between 0.014 and 0.026, while freeze dried microbeads had irregular shape, especially those with no excipient. The reduction in microbeads size after freeze drying process (expressed as shrinkage factor) ranged from 0.338 (alginate microbeads with 20% (w/v) of inulin) to 0.523 (plain alginate microbeads). The expressed radical scavenging activity against ABTS and DPPH radicals was found to be between 30% and 40% for encapsulated extract, while the fresh extract showed around 47% and 57% of radical scavenging activity for ABTS and DPPH radicals, respectively. The correlation between antioxidant activity and the total phenolic content were found to be positive (in both assay methods, DPPH and ABTS), which indicate that the addition of inulin didn't have influence on antioxidant activity. The presence of inulin reduced stiffness of the hydrogel, and protected bead structure from collapse upon freeze‐drying. Alginate–inulin beads are envisaged to be used for delivery of aqueous P. tridentatum extract in functional food products.  相似文献

本文研究了藜蒿叶提取物在模拟体外消化过程中酚酸含量、抗氧化活性以及对酪氨酸酶活力抑制率的变化。研究表明,经过模拟体外消化后藜蒿叶提取物中的绿原酸、1,5-双咖啡酰奎宁酸、3,4-双咖啡酰奎宁酸、3,5-双咖啡酰奎宁酸、4,5-双咖啡酰奎宁酸5种酚酸含量均显著性降低。总酚含量和DPPH、FRAP、ABTS抗氧化活性经过模拟体外消化后均显著性增加。消化后粗提物和纯化样总酚含量分别升高了73.57%、23.79%,DPPH抗氧化能力分别增强了36.17%、53.29%,FRAP抗氧化能力分别增强了28.71%、15.45%,ABTS清除自由基能力分别增强了163.67%和49.50%。此外,藜蒿叶粗提物对酪氨酸酶活力的抑制率在体外消化过程中也显著性增加,消化后粗提物的抑制率可达到49.88%。从而可有效抑制黑色素生成,具有较好的美白潜力。因此,藜蒿叶提取物经体外消化后具有较好的抗氧化活性和抑制酪氨酸酶活力的能力,有望应用于美白产品的开发,该研究为藜蒿茎加工副产物-藜蒿叶的开发提供了借鉴。  相似文献

Ilex paraguariensis St. Hil. (Aquifoliaceae) (yerba mate) is one of the most used plant species in South America due to its nutritional and medicinal properties. The industrial processing involves different stages (green leaves, zapecado, drying, forced or natural aging) which can modify the qualitative and quantitative composition and the pharmacological activities. In this work, the main compounds, caffeoyl derivatives (caffeic acid, mono- and dicaffeoylquinic acids), methylxanthines (caffeine and theobromine) and flavonoids (rutin, quercetin and kaempferol) were studied by HPLC in extracts obtained by decoction of samples of a large amount of I. paraguariensis during its industrial process stages. The comparative quantitative analysis of all the samples indicated that those obtained after the zapecado, drying and aging stages possess higher content of biologically active principles when compared with green leaves. No differences were found between the natural and forced aging processes. This is the first complete report on the quantitative variation of the bioactive compounds of I. paraguariensis during each step of the industrial processing. The results obtained in this work provide a guideline for the obtention of extracts enriched in biological principles for the pharmaceutical, food and cosmeceutical industry.  相似文献
