响应面法优化金蝉花多糖提取工艺及抗氧化活性分析   总被引:3,自引:0,他引:3  
通过考察液料比、浸提时间及浸提温度对金蝉花多糖含量的影响,在单因素试验基础上进行响应面优化提取工艺条件,并通过测定金蝉花多糖总还原力、清除1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-trinitrophenylhydrazine,DPPH)自由基、羟自由基(·OH)和超氧阴离子自由基(O2-·)的能力研究其体外抗氧化活性。结果表明,金蝉花多糖适宜的提取工艺参数为浸提时间130min、浸提温度80℃、液料比50∶1(mL/g),在此条件下金蝉花多糖含量实际值为26.14mg/g。金蝉花多糖具有较好的抗氧化能力,其清除DPPH自由基、·OH、O2-·的半抑制质量浓度(IC50)分别为28.99μg/mL、0.19mg/mL和0.30mg/mL。  相似文献

选用回流法提取槐角中黄酮类化合物,采用DPPH·法、邻苯三酚自氧化法和邻二氮菲法,测定了槐角黄酮对DPPH·、O2-·、·OH的清除率,并与维生素C(VC)比较,进行槐角黄酮的体外抗氧化活性研究.结果表明,槐角黄酮具有一定的抗氧化能力,在一定浓度范围内,清除自由基能力与其浓度呈正相关性.与阳性对照VC相比,槐角黄酮对DPPH·的清除作用较弱,IC50为0.0192mg/mL;而对O2-、、·OH的清除作用较强,IC50分别为0.0127、0.0160mg/mL.这表明槐角黄酮具有一定的抗氧化能力.  相似文献

通过水提法探讨蕨麻多糖适宜的提取工艺,并研究其抗氧化活性。考察料液比、浸提温度、浸提时间对蕨麻多糖含量的影响,在单因素试验的基础上做L9(34)正交试验优化提取工艺参数。通过测定蕨麻多糖总抗氧化能力,清除DPPH、·OH、O2-·自由基的能力来评价其抗氧化活性。研究结果表明,蕨麻多糖适宜的提取工艺参数是:浸提温度90℃、浸提时间2 h、料液比1∶30。在此条件下蕨麻多糖含量为2.54%。蕨麻多糖具有较好的抗氧化能力,对DPPH、·OH、O2-·自由基的IC50分别为5.47,2.62,27.53 mg/mL。本研究结果为蕨麻开发利用奠定理论基础。  相似文献

采用水提醇沉法提取密楝和吴茱萸果实中粗多糖,苯酚-硫酸法测定粗多糖含量。在体外化学模拟条件下,测定密楝和吴茱萸果实粗多糖对羟自由基(·OH)、H2O2和DPPH自由基的清除作用。细胞培养板液体稀释法测定密楝和吴茱萸粗多糖最小抑菌浓度。同时,将密楝果实多糖的抗氧化活性和抑菌活性与吴茱萸果实多糖的活性进行对比。体外实验结果表明:蜜楝果实多糖对·OH、H2O2和DPPH自由基清除能力EC50分别为0.8、0.25mg/mL和0.9mg/mL;吴茱萸果实中粗多糖对·OH、H2O2和DPPH自由基清除能力EC50分别为1.0、0.1mg/mL和0.8mg/mL。两种多糖均表现出较强的清除活性,对H2O2和·OH清除活性尤其明显,而且在相同质量浓度下对H2O2的清除活性明显高于VC。抗菌实验表明,吴茱萸粗多糖的抑菌、杀菌活性强于密楝粗多糖的活性。  相似文献

以还原能力和金属离子螯合能力、清除DPPH·、O2-·、OH·自由基能力为指标,考察鲨鱼肉酶解物的抗氧化活性,同时以清除自由基DPPH·的活性为指标,分析温度和pH值对其抗氧化作用稳定性的影响。结果表明:鲨鱼肉酶解液产物具有一定的还原能力和金属离子螯合能力,其清除DPPH·、O2-·、OH·自由基的IC50值分别为10.16、23.71、19.82mg/mL。酶解产物耐热性强;在酸性环境中,鲨鱼肉酶解液能较好地保持其抗氧化活性,但在碱性环境中抗氧化活性丧失较快。  相似文献

采用响应曲面法优化海红果水溶性多糖(MPB)提取工艺的条件。在单因素实验基础上依据回归分析确定最佳工艺条件为:提取温度86℃,提取时间4h,料液比1∶28g/mL,MPB的实际提取率可达8.33%。用MPB对羟基自由基(·OH)、超氧阴离子自由基(O2-·)、DPPH自由基进行清除能力和还原力的测定,研究MPB的抗氧化能力。结果表明,MPB对·OH、O2-·、DPPH均有一定的清除能力,对DPPH的清除作用尤为明显,当质量浓度达到1mg/mL时,清除率达到87.12%,接近VC的94.47%,且MPB清除DPPH的IC50值为1.181mg/mL。  相似文献

以猕猴桃籽油为原料,采用体外抗氧化实验模型研究其抗氧化活性。通过测定猕猴桃籽油还原力和DPPH自由基、超氧阴离子自由基(O2-·)、H2O2以及羟自由基(·OH)清除实验,以VC为阳性对照,探讨猕猴桃籽油的抗氧化性能。结果表明:猕猴桃籽油的还原能力随其质量浓度的增加而增强,对DPPH自由基、O2-·、H2O2以及·OH的清除率随质量浓度的增加而升高,其中对O2-·的清除效果比VC更明显,对其他自由基的清除效果却比VC弱;猕猴桃籽油对O2-·、H2O2、·OH的IC50分别为0.078、0.1、0.2mg/mL,表明猕猴桃籽油清除O2-·的效果最好。  相似文献

以火龙果果皮为原料,采用乙醇为提取剂对火龙果皮色素进行提取,利用正交设计实验优化其提取工艺,并采用体外模型从羟自由基(·OH)的清除能力,DPPH自由基(DPPH·)的清除能力和超氧阴离子清除能力三个角度评价其抗氧化活性.结果表明,色素提取液清除DPPH自由基的最优工艺条件是:选用90%的乙醇作提取溶剂,提取时间为180min,料液比为1∶16(g/mL),温度为25℃,此条件下的DPPH·清除率可达到94.68%;火龙果果皮色素清除50%DPPH·、·OH与O2-·的有效浓度(IC50)分别为2.09、46.0、2.44mg/mL.火龙果皮色素提取液对DPPH自由基的清除作用在一定的范围内呈对数递增;对超氧自由基的清除作用和羟基自由基的清除作用与其浓度呈正相关.  相似文献

对羊肚菌菌丝体液体深层发酵富硒条件进行优化,考察培养基中硒浓度、温度、装液量和p H值对富硒率的影响。采用Fenton法、邻苯三酚自氧化法和DPPH(1,1-二苯基-2-苦基肼)还原法对羊肚菌菌丝体多糖的体外抗氧化活性进行了研究。羊肚菌菌丝体富硒的适宜培养条件为:亚硒酸钠质量浓度为10 mg/L,温度25℃,装液量100 m L/250 m L,初始p H值7。在此条件下,富硒率最大达9.10%。羊肚菌菌丝体多糖和羊肚菌菌丝体硒多糖清除羟自由基(·OH)IC50分别为:0.62 mg/m L和0.42 mg/m L;清除超氧阴离子(O2-·)IC50分别为:0.85mg/m L和0.59 mg/m L;清除DPPH自由基IC50分别为:0.66 mg/m L和0.49 mg/m L。羊肚菌菌丝体多糖具有较强的体外抗氧化活性,且随着多糖浓度的增大其抗氧化活性逐渐增强,羊肚菌菌丝体硒多糖抗氧化作用更强。  相似文献

采用多个抗氧化能力评价体系,包括清除DPPH?自由基、羟自由基(.OH)和超氧阴离子自由基(O2ˉ.)体系,评价玉米须中黄酮类化合物的抗氧化活性。结果表明:玉米须黄酮具有较强的抗氧化活性,对DPPH?自由基、羟自由基和超氧阴离子自由基的半清除率IC50分别为0.057 mg/mL、0.38 mg/mL和0.56 mg/mL。玉米须黄酮的浓度与其活性大小呈明显的量效关系。  相似文献

Since grapevine ( Vitis spp .) rootstock material is being traded increasingly as disbudded woody material a lack of distinctive morphological features on such material necessitates an alternative and reliable means of identification. Methods described here were developed for rapid and efficient extraction of DNA from woody samples rich in phenolic compounds and polysaccharides, and for subsequent identification of varieties by RAPD PCR. Using these methods, and with the application of only one selected RAPD primer, we were able to differentiate sixteen rootstock varieties, including the seven varieties most commonly used in Germany. Problems commonly encountered with reproducibility of RAPD patterns were avoided by choosing primers with a dinucleotide sequence and a high G/C content that allowed a rather high annealing temperature of 45°C. Methods described here should also be useful for other horticultural crops, especially those with woody tissues rich in phenolic compounds and polysaccharides.  相似文献

The characterization of the aromatic profile of several apricot cultivars with molecular tracers in order to obtain objective data concerning the aromatic quality of this fruit was undertaken using headspace–solid phase microextraction (HS–SPME). Six apricot cultivars were selected according to their organoleptic characteristics: Iranien, Orangered, Goldrich, Hargrand, Rouge du Roussillon and A4025. The aromatic intensity of these varieties measured by HS–SPME–Olfactometry were defined and classified according to the presence and the intensity of grassy, fruity and apricot like notes. In the six varieties, 23 common volatile compounds were identified by HS–SPME–GC–MS. Finally, 10 compounds, ethyl acetate, hexyl acetate, limonene, β-cyclocitral, γ-decalactone, 6-methyl-5-hepten-2-one, linalool, β-ionone, menthone and (E)-hexen-2-al were recognized by HS–SPME–GC–O as responsible of the aromatic notes involved in apricot aroma and considered as molecular tracers of apricot aromatic quality which could be utilized to discriminate apricot varieties.  相似文献

The advent of the functional barrier concept in food packaging has brought with it a requirement for fast tests of permeation through potential barrier materials. In such tests it would be convenient for both foodstuffs and materials below the functional barrier (sub-barrier materials) to be represented by standard simulants. By means of inverse gas chromatography, liquid paraffin spiked with appropriate permeants was considered as a potential simulant of sub-barrier materials based on polypropylene (PP) or similar polyolefins. Experiments were performed to characterize the kinetics of the permeation of low molecular weight model permeants (octene, toluene and isopropanol) from liquid paraffin, through a surrogate potential functional barrier (25 μm-thick oriented PP) into the food simulants olive oil and 3% (w/v) acetic acid. These permeation results were interpreted in terms of three permeation kinetic models regarding the solubility of a particular model permeant in the post-barrier medium (i.e. the food simulant). The results obtained justify the development and evaluation of liquid sub-barrier simulants that would allow flexible yet rigorous testing of new laminated multilayer packaging materials.  相似文献

This paper describes the second part of a project undertaken to develop certified mussel reference materials for paralytic shellfish poisoning toxins. In the first part two interlaboratory studies were undertaken to investigate the performance of the analytical methodology for several PSP toxins, in particular saxitoxin and decarbamoyl-saxitoxin in lyophilized mussels, and to set criteria for the acceptance of results to be applied during the certification exercise. Fifteen laboratories participated in this certification study and were asked to measure saxitoxin and decarbamoyl-saxitoxin in rehydrated lyophilized mussel material and in a saxitoxin-enriched mussel material. The participants were allowed to use a method of their choice but with an extraction procedure to be strictly followed. The study included extra experiments to verify the detection limits for both saxitoxin and decarbamoyl-saxitoxin. Most participants (13 of 15) were able to meet all the criteria set for the certification study. Results for saxitoxin.2HCl yielded a certified mass fraction of <0.07 mg/kg in the rehydrated lyophilized mussels. Results obtained for decarbamoyl-saxitoxin.2HCl yielded a certified mass fraction of 1.59+/-0.20 mg/kg. The results for saxitoxin.2HCl in enriched blank mussel yielded a certified mass fraction of 0.48 +/- 0.06 mg/kg. These certified reference materials for paralytic shellfish poisoning toxins in lyophilized mussel material are the first available for laboratories to test their method for accuracy and performance.  相似文献
