The present work examines the in vitro antioxidant and antimicrobial properties of the essential oil and various extracts from the herbal parts of Origanum syriacum L var bevanii. Polar subfractions of methanol extracts from both deodorised and non‐deodorised materials showed the highest DPPH (2,2‐diphenyl‐l‐picrylhydrazyl) radical‐scavenging activity, with IC50 values of 21.40 and 26.98 µg ml?1 respectively, whereas the IC50 of the essential oil was 134.00 µg ml?1. The antioxidant potential of the extracts appeared to be closely related to the presence of polar phenolics. However, the inhibitive effect on linoleic acid oxidation might be promoted by the presence of non‐polar phenolics, as both hexane and dichloromethane extracts showed high antioxidant activities. The antimicrobial activity of the essential oil was superior to those of the other extracts. Nineteen compounds representing 962 g kg?1 of the essential oil were identified; carvacrol (669 g kg?1) was the main component. Overall, the results suggest that the essential oil and extracts from the herbal parts of O syriacum could be used as natural preservative ingredients in the food industry. The present investigation was to evaluate the antioxidant and antibacterial activities of hexane and methanol extracts from Origanum vulgare ssp. hirtum and Thymus vulgaris (Komotini, Greece). The methanol extracts of oregano and thyme against DPPH radical were more active than the hexane extracts (mean values 0.94 and 0.47, respectively) and oregano exhibited stronger activity than thyme (mean values 0.82 and 0.55, respectively). The results from the β-carotene/linoleic acid assay showed that all plant extracts inhibited linoleic acid oxidation up to 70.78±1.17%. The oregano extracts exhibited the strongest inhibition against Escherichia coli, Listeria monocytogenes, and Staphylococcus aureus using the disc diffusion assay. The chemical composition of the hexane and methanol extracts, by using GC-MS, showed that carvacrol, thymol, and p-cymene were the most prominent compounds. The methanol extract of oregano was found the most potent antioxidant with the highest content of total phenolics (138.92 mg GA/g extract) and carvacrol (76.7%).  相似文献

ABSTRACT:  GC-FID and GC-MS analysis of essential oils of  Juniperus phoenicea  resulted in the identification of 30 compounds, representing more than 98% of the total composition. α-pinene (55.7% and 80.7%), δ-3-carene (10.7% and 4.5%), and γ-cadinene (2.9% and 5.1%) were the main components, respectively, in leaves and berries essential oil. Extracts of  J. phoenicea  were obtained by different extraction solvents: methanol, ethanol, ethyl acetate, and dichloromethane and evaluated composition for polyphenols (gallic acid equivalent 52 to 217 g/kg), tannins (catechin equivalent 6.5 to 60.2 g/kg), antocyanins (cyanidin equivalent 84 to 373 mg/kg), and flavonoids (quercetin equivalent 6.4 to 29.3 g/kg). The samples (essential oils and extracts) were subjected to a screening for their antioxidant activity by using DPPH and ABTS assays; antimicrobial activity was tested with 6 bacteria (3 Gram-positive and 3 Gram-negative), 1 yeast, and 2 fungi. The strongest antioxidant activity was obtained by the methanolic extract ( IC 50= 6.5 ± 0.3 mg/L). Flavonoids are likely to contribute to the antifungal activity against  Saccharomyces cerevisiae.  Correlations were studied between chemical composition and antioxidant and antimicrobial activities.  相似文献

This study reported the chemical composition, phenolic content, antioxidant and anti‐lipase activity of oregano and Lippia essential oils. The major compounds found in oregano essential oil were γ‐terpinene (32.10%), α‐terpinene (15.10%), p‐cymene (8.00%) and thymol (8.00%). In Lippia essential oil, α‐limonene (76.80%) and 1,8‐cineole (4.95%) represented the major compounds. Oregano essential oil had higher phenolic content (12.47 mg gallic acid mL?1) and DPPH scavenging activity (IC50 0.357 μg mL?1) than Lippia essential oil (7.94 mg gallic acid mL?1 and IC50 0.400 μg mL?1, respectively). Both essential oils had similar antioxidant indexes (about 1.2) determined by Rancimat. Moreover, oregano essential oil had also higher anti‐lipase activity (IC50 5.09 and 7.26 μg mL?1). Higher phenolic content in the essential oils was related with higher scavenging and anti‐lipase activities. Oregano and Lippia essential oils could be used as natural antioxidants on food products.  相似文献

BACKGROUND: The aim of this research was to determine essential oil composition, phenolic constituents and antioxidant properties of Turkish oregano (Origanum onites L.) leaves harvested during the months of June to September. RESULT: The maximum essential oil yield in the leaves appeared in the middle of July. The main components of oregano oil were carvacrol, thymol, γ‐terpinene, p‐cymene, α‐terpinene and α‐pinene. Carvacrol was highest in the July harvest. The maximum extract yield was found in September. Oil distilled from early‐season (June) harvested leaves had the highest antioxidant ability, expressed as low concentration providing 50% inhibition of free radical scavenging activity and high levels of reducing/antioxidant capacity. Twelve phenolic compounds of oregano extract were identified and the main components were found to be rosmarinic acid and acecetin. The maximum rosmarinic acid and acecetin were found in harvests of July and June, respectively. Total phenolic contents, free radical scavenging activities and reducing/antioxidant capacities were found to be highest in the July harvest. DISCUSSION: All yields, chemical compositions, free radical scavenging activities and reducing/antioxidant capacities of extracts and essential oils of Turkish oregano changed importantly depending on vegetative periods of growing season. This study reports a comparative screening of four species of Origanum in Turkey, based on their essential oil composition, total phenolic content, antioxidant and antibiofilm activities. The major components of essential oils were p-cymene, linalool, and thymol. The total phenolic contents differed from 3.81 to 47.54 mg of GAE/g of extract. The concentrations of flavonoids varied from 12.74 to 58.39 mg of Ru/g of extract. Antioxidant activity was determined in vitro using DPPH reagent and expressed as concentration of each extract required to inhibit radical by 50% (IC50) values that ranged from 16.03 to 48.94 μg/ml. Our results indicated that chloroform extracts of species O. majorana and O. onites, with a total content of polyphenols (47.54 mg of GAE/g and 45.17 mg of GAE/g, respectively) and an IC50 of 16.03 μg/ml and 16.89 μg/ml, respectively were more antioxidant. Among the essential oil concentrations tested, maximum antibiofilm activity was found as 92.24% against M. luteus NRRL-B 1013 by O. majorana essential oil at 50 mg/ml.  相似文献

Antibacterial and antioxidant potential of essential oil, extract and its fractions of Bidens frondosa Linn were evaluated. Sixty‐one components representing 95.41% of the total oil were identified. The essential oil (7.5 μL disc?1), methanol extract and its different organic subfractions (0.5 μg disc?1) of B. frondosa displayed a great potential of antibacterial activity against Staphylococcus aureus (ATCC 6538 and KCTC 1916), Listeria monocytogenes ATCC 19116, Bacillus subtilis ATCC 6633, Pseudomonas aeruginosa KCTC 2004, Salmonella enteritidis KCTC 12021 and Enterobacter aerogenes KCTC 2190. Antioxidant activity was evaluated by using 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) assay. The free radical scavenging activity of ethyl acetate (EtOAc) fraction was superior to all other fractions (IC50 = 11.96 μg mL?1), which was higher than synthetic antioxidant butylated hydroxyanisole, (IC50 = 18.27 μg mL?1). Furthermore, the amount of total phenolic compounds was determined and its content in EtOAc fraction was the highest as compared to methanol extract or other fractions. The results indicate that the oil and extracts of B. frondosa could serve as an important bio‐resource of antimicrobial agents and antioxidants for using in the food industries.  相似文献

Grape seed extracts (GSEs,) obtained from Italian and Rhine Rieslings, were examined for polyphenolic composition and antioxidant activities using HPLC and ESR spectrometry. The seed extraction was carried out with ethyl acetate and ethanol. The contents of polyphenols, flavan-3-ols and antioxidant activities were found to be higher in ethyl acetate than in ethanolic extracts. IC50 values were 0.1045 mg/mL and 0.0599 mg/mL for the stable DPPH radical in ethanolic and ethyl acetate extracts, respectively. The values for the short-lived OH radical were 0.1989 mg/mL and 0.0362 mg/mL, in the given order. The significant correlations between the antioxidant activities of GSEs and polyphenols were established (P < 0.05). Owing to their antioxidant activities, the cultivars could be used as a source to produce a GSE.  相似文献

The methanolic extracts of 16 Helichrysum species were investigated for their in vitro antioxidant, radical scavenging and antimicrobial activities. All the extracts showed strong antioxidant and radical scavenging activity. The highest total antioxidant capacity as ascorbic acid equivalents (AAE) of 194.64 mg/g dry extract was obtained for Helichrysum noeanum in the phosphomolybdenum assay. The highest IC50 value (7.95 μg/ml) was observed for the extract of Helichrysum stoechas subsp. barellieri in the DPPH assay. The total phenolic contents of the extracts ranged from 66.74 to 160.63 mg gallic acid equivalents (GAE)/g dry extract. The major component present in the extracts was identified as chlorogenic acid followed by apigenin-7-glucoside and apigenin by HPLC analysis. All the extracts showed significant antimicrobial activity against microorganisms containing 13 bacteria and two yeasts in the agar diffusion method.  相似文献

The effect of 30 kGy of gamma radiation on the antioxidant activity of methanolic and aqueous extracts prepared from dry gamma irradiated sage and oregano were analysed. The antioxidant activity was characterised by DPPH radical test and by reducing power assay within an extract concentration range of 0–40 mg/ml of methanol or water. In addition, the total phenolic content in the extracts was determined. From the EC50 estimated values, it was established that, by both methods, oregano extracts showed a higher antioxidant activity than sage extracts. Oregano extracts also showed a phenolic content significantly higher than sage extracts. A good correlation between the phenolic content and the antioxidant activity was also observed. The 30 kGy dose applied to dry sage and oregano for sanitization did not significantly affect the capacity to inhibit the DPPH radical or the reducing power, nor did it affect the total phenolic content of the methanolic and aqueous extract.  相似文献

Composition and antioxidant and antimicrobial activities of essential oil and methanol extract polar and nonpolar subfractions of Stachys inflata were determined. GC and GC/MS analyse of the essential oil showed 45 constituents representing 95.46% of the oil, the major components linalool (28.55%), α-terpineol (9.45%), spathulenol (8.37%) and (2E)-hexenal (4.62%) constituted 50.99% of it. Essential oil and extracts were also tested for their antioxidant activities using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and β-carotene/linoleic acid assays. In the DPPH test, IC50 value for the polar subfraction was 89.50 μg/ml, indicating an antioxidant potency of about 22% of that of butylated hydroxytoluene (IC50 = 19.72 μg/ml) for this extract. In β-carotene/linoleic acid assay, the best inhibition belonged to the nonpolar subfraction (77.08%). Total phenolic content of the polar and nonpolar extract subfractions was 5.4 and 2.8% (w/w), respectively. The plant also showed a week antimicrobial activity against three strains of tested microorganisms. Linalool and α-terpineol were also tested as major components of the oil and showed no antioxidant but considerable antimicrobial activities.  相似文献

The conventional steam distillation process for oregano (Lippia berlandieri v. Shauer) essential oil extraction produces large volumes of mother liquor. This residual liquid represents a potential value because the soluble antioxidants it contains. Essential oil and ethyl acetate mother liquor extracts (MLEs) were evaluated for antioxidant activity. Total phenolic content and antioxidant activities by the 2-2′-diphenyl-1-picrylhydrazyl (DPPH) method, by the deoxyribose degradation assay, and by oxidation of low density lipoproteins (LDL) with CuSO4 were evaluated. Oil yield was 4.34%. Total phenolic content was 151 ± 2.00 and 150.5 ± 0.98 mg of GAE (gallic acid equivalents)/mL for the essential oil and MLEs, respectively. DPPH assay showed a low radical scavenging activity (RSA) for oregano essential oil. Meanwhile MLEs exhibited no significant RSA at low concentrations, but at higher concentrations (100 μg/mL), it was superior to those exhibited by the controls ascorbic acid and butylated hydroxytoluene (BHT). Deoxy-d-ribose assay results for both essential oil and MLEs showed a good hydroxyl radical RSA at the concentrations tested. Essential oil and MLEs delayed induction time effectively. Solubility problems, chemical constituents, and their hydrophilic–lipophilic distribution are key factors that explain samples behavior for an eventual use of these natural products.  相似文献

In this study, antioxidant activities of aqueous and ethanolic extracts of Brassica oleracea L. var. italica were investigated. The antioxidant properties of both extracts of Brassica oleracea L. var. italica were evaluated using different antioxidant tests, including 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, superoxide radical scavenging, inhibition of microsomal lipid peroxidation, reduction of power, and metal ion chelating activities. Inhibition of superoxide scavenging by aqueous and ethanolic extracts showed an IC50 of 0.93 and 0.25 mg/ml, respectively. Metal ion chelation showed an IC50 of 0.35 mg/ml of both the extracts and was equipotent to positive control, ethylenediamine tetra-acetic acid. The ethanolic extract of Brassica oleracea L. var. italica exhibited higher antioxidant activity in DPPH radical and superoxide anion scavenging than that of aqueous extract. The results obtained in the in vitro models clearly suggest that, Brassica oleracea L. var. italica is a natural source for antioxidants, which could serve as a nutraceutical with potential applications in reducing the level of oxidative stress and related health benefits. However, comprehensive studies need to be conducted to ascertain the in vivo safety of such extracts in experimental animal models.  相似文献

采用DPPH·法及Fe3+ 还原力法对蛇菰提取物的抗氧化活性进行研究,分别测定蛇菰乙醇提物、石油醚提取物、氯仿提取物、乙酸乙酯提取物、正丁醇提取物、水层部分、水提粗多糖及碱提粗多糖的抗氧化作用,同时以VC 作为阳性对照。结果表明:蛇菰提取物具有较强的抗氧化活性,尤其是乙酸乙酯提取物、正丁醇提取物、总醇提取物及水提粗多糖;它们对DPPH·的清除作用与VC 相当,其中乙酸乙酯提取物IC50 值为6.0μg/mL,活性略高于VC,同时以上提取物对Fe3+ 也具有很强的还原能力。  相似文献

The amount of phenolics, flavonoids, and anthocyanins in ethanol extracts and antioxidant activity of both ethanol and lipophilic extracts of common fruity vegetables in Bangladesh were studied. Among the ethanol extracts of 15 fruity vegetables, M. oleifera had the highest total polyphenol content (85.05 mg gallic acid equivalent/g extract) followed by L. acutangula (61.74 mg gallic acid equivalent/g extract) and A. esculentus (48.92 mg gallic acid equivalent/g extract). L. acutangula had high a content of flavonoids (14.46 mg (+)-catechin equivalent/g extract), which was almost similar to L. siceraria (13.67 mg catechin equivalent/g extract) followed by A. esculentus (11.95 mg catechin equivalent/g extract) and S. melongena (11.42 mg catechin equivalent/g extract). Highest anthocyanins content was in F. hispida (2.22 μmol/g extract) followed by S. melongena (1.04 μmol/g extract). Ethanol extracts of A. esculentus, F. hispida, L. acutangula, L. siceraria, and S. melongena exhibited high DPPH free radical scavenging activity with IC50 of 70.4, 64.9, 70.4, 64.9, and 94.3 μg/mL respectively, whereas for the same lipophilic extracts of F. hispida and S. melongena showed lowest (37 μg/mL) IC50 followed by M. oleifera (47.6 μg/mL), L. siceraria (57.5 μg/mL), and A. esculentus (63.3 μg/mL). These vegetables also showed high reducing powers, NO scavenging and total antioxidant capacity. Therefore, the top five potential fruity vegetables consist of both hydrophilic and lipophilic antioxidant(s), the order being F. hispida > M. oleifera > A. esculentus, L. acutangula > L. siceraria > and S. melongena.  相似文献

The objective of the present research work was to evaluate the antioxidant, antimicrobial and antimalarial activities of essential oil and various extracts from O. sanctum. Gas chromatography-mass spectrometry analysis identified the following major compounds with their quantification as: eugenol (22.0%), β-elemene (19.2%), β-caryophyllene (19.1%), and Germacrene D (5.03%). HPLC analysis of O. sanctum extracts revealed that gallic acid, chlorogenic acid, p-hydroxy benzoic acid, caffeic acid, vanillic acid, p-coumeric acid, sinapic acid, and ferulic acid were the important phenolic acids. The methanol extract exhibited highest level of total phenolic (1.36 g/100 g dry plant material) and total flavonoid (0.67 g/100 g dry plant material) followed by ethanol and n-hexane extracts. The oil and extracts exhibited excellent free radical scavenging potential as measured by 2,2-diphenyl-1-picrylhydrazyl radical free radical-scavenging ability, and antioxidant activity as measured by inhibition of linoleic acid oxidation. Essential oil, n-hexane, methanol, and ethanol extracts exhibited moderate antimalarial potential in term of anti-haem biocrystallization activity. In the resazurin microtitre plate and disc diffusion assays, the essential oil of O. sanctum showed better antibacterial activity than various extracts. The results of the present investigation demonstrated significant (p < 0.05) variations in the antioxidant, antimicrobial, and antimalarial activities of essential oil and extracts from O. sanctum.  相似文献

Black rice bran contains phenolic compounds of a high antioxidant activity. In this study, the 40% acetone extract of black rice bran was sequentially fractionated to obtain 5 fractions. Out of the 5 fractions, ethyl acetate fraction was subfractionated using the Sephadex LH‐20 chromatography. The antioxidant activity of phenolic compounds in the extracts was investigated by 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) radical assay, 2,2‐azino‐bis‐(3‐ethylenebenzothiozoline‐6‐sulfonic acid) (ABTS) radical cation assay, reducing power. The subfraction 2 from ethyl acetate fraction had the highest total phenolic contents (TPC) (816.0 μg/mg) and the lowest EC50 values (47.8 μg/mL for DPPH radical assay, 112.8 μg/mL for ABTS radical cation assay, and 49.2 μg/mL for reducing power). These results were 3.1, 1.3, and 2.6 times lower than those of butylated hydroxytoluene (BHT), respectively. At a concentration of 100 μg/mL, the antioxidant activity and TPC of various extracts was closely correlated, with correlation coefficients (R2) higher than 0.86. The major phenolic acid in subfraction 2 was identified as ferulic acid (178.3 μg/mg) by HPLC and LC‐ESI/MS/MS analyses. Our finding identified ferulic acid as a major phenolic compound in black rice bran, and supports the potential use of black rice bran as a natural source of antioxidant.  相似文献

The extract yields obtained by three methods in different solvents from chilli were compared. The antioxidant activities of chilli extracts vs. butylated hydroxytoluene (BHT) was determined through the peroxide value (POV), the beta‐carotene bleaching assay and the 2,2‐diphenyl‐1‐picrylhydrazyl radical scavenging method. The results showed that the extract yields by three extract methods, namely, Soxhlet, ultrasonic wave and saturation, were 16.32%, 15.78% and 11.62%, respectively, when 85% ethanol acted as solvent. The extracting efficiency of solvent from high to low was 95% ethanol > ethyl acetate > anhydrous ethanol > distilled water. The total phenol content of chilli extracts from ultrasonic wave with 85% ethanol as solvent was 73.91 mg gallic acid equivalents (GAE) per gram extracts and the value of inhibitory capacity IC50 on free radical (DPPH) was 38.99 mg mL?1. The POV of chilli extracts was close to that of BHT. The ultrasonic wave was more helpful for the extraction of antioxidant substance from chilli than other methods. In addition, the composition of chilli extracts was analysed by gas chromatography‐mass spectrum (GC‐MS).  相似文献

The crude methanol extract (ME) from camellia oil was fractionated by serial benzene, ether, EtOAc and n-BuOH, divided into five subfractions designated as SF1–5. The obtained fractions as well as ME were evaluated for their antioxidant activities using DPPH assay, and their phenolic constituents were isolated using silica gel column chromatography and analysed by TLC, RP-HPLC–PDA, UPLC-ESI–MS and GC–MS. Results showed that the content of total phenolics (TPs) of ME, determined by the Folin–Ciocalteu method, was estimated to be 79.5?±?10.2?mg of p-hydroxybenzoic acid equivalent/kg oil, while that of the total flavonoids was 8.98?±?0.54?mg of rutin equivalent/kg oil. ME exhibited pronounced radical-scavenging activity against the stable DPPH radical (with an antioxidant capacity IC50 value of 52.37?μg/mL), and three subfractions (SF1–3) separated from ME contributed the most significant activity with IC50 of 28.41, 17.42 and 43.17?μg/mL, respectively. Consistent with the different capacity of scavenging DPPH radicals, the subfractions from ME showed different composition and contents of phenolic compounds as detected by TLC, RP-HPLC–PDA, UPLC-ESI–MS and GC–MS. Kaempferol, as well as a number of volatile phenolic compounds, was first identified from camellia oil. Results showed that the presence of phenolic compounds and flavonoids in each extracts may be responsible for its individual antioxidant activity, and the lipophilic phenolic fractions would likely account for a great part of the antioxidant properties of the oil.  相似文献

Aqueous extract, proanthocyanidin rich extract, and organic extracts of Cymbopogon schoenanthus L. Spreng (lemon grass) shoots from three different locations in South Tunisia were screened for their antioxidant, acetylcholinesterase and antimicrobial activities. In addition to the evaluation of these activities, the contents of flavonoids and total phenolic compounds were determined.Antioxidant activity measured by DPPH assay showed that the proanthocyanidin extract exhibited higher antioxidant activity than the aqueous extract. Extract concentration providing 50% inhibition (IC50) ranged from 16.4 ± 6.8 μg/mL to 26.4 ± 6.8 μg/mL. The antioxidant activity was also determined using the β-carotene/linoleic acid bleaching test. The best results (IC50 = 0.11 ± 0.10 mg/mL) were obtained with the proanthocyanidin extract of the plants collected from the desert region (Dhibat).The greatest acetylcholinesterase inhibitory activity (IC50 = 0.23 ± 0.04 mg/mL) was exhibited by the ethyl acetate and methanol extracts of the plants collected from the mountainous region. It seems that extracts obtained with more polar solvents gave better results.The proanthocyanidin extracts showed a good antimicrobial activity against Streptococcus sobrinus at low concentration (MIC = 4 mg/mL). Therefore, these extracts could be used to prevent carious lesions by inhibiting S. sobrinus growth.  相似文献
