Triton X-100 inhibits the NADH oxidase and rotenone-sensitive NADH-Q1 reductase activities of bovine heart submitochondrial particles (SMP) with an apparent Ki of 1x10-5 M (pH 8.0, 25 degrees C). The NADH-hexammineruthenium reductase, succinate oxidase, and the respiratory control ratio with succinate as the substrate in tightly coupled SMP are not affected at the inhibitor concentrations below 0.15 mM. The succinate-supported aerobic reverse electron transfer is less sensitive to the inhibitor (Ki=5x10-5 M) than NADH oxidase. Similar to rotenone, limited concentrations of Triton X-100 increase the steady-state level of NAD+ reduction when the nucleotide is added to tightly coupled SMP oxidizing succinate aerobically. Also similar to rotenone, Triton X-100 partially protects Complex I against the thermally induced deactivation and partially activates the thermally deactivated enzyme. The rate of the NADH oxidase inhibition by rotenone is drastically decreased in the presence of Triton X-100 which indicates a competition between these two inhibitors for a common specific binding site. In contrast to rotenone, the inhibitory effect of Triton X-100 is instantly reversed upon dilution of the reaction mixture. The NADH-Q1 reductase activity of SMP is inhibited non-competitively by added Q1 whereas a simple competition between Q1 and the inhibitor is seen for isolated Complex I. The results obtained show that Triton X-100 is a specific inhibitor of the ubiquinone reduction by Complex I and are in accord with our previous findings which suggest that different reaction pathways operate in the forward and reverse electron transfer at this segment of the mammalian respiratory chain.  相似文献

AIM: To study the relationship between the calcium and the release of platelet-activating factor (PAF) from rat peritoneal macrophages (PM?) stimulated by lipopolysaccharides (LPS). METHODS: The effects of tetrandrine (Tet), Fura 2-AM, and Bay k 8644 on the PAF release from rat PM? was investigated by the bio-assay of PAF. RESULTS: LPS stimulated PM? to release PAF, without increasing intracellular Ca2+ of PM?, Tet at 0.1, 1.0, 10, 100 mumol.L-1 and Fura 2-AM at 0.01, 0.1, 1.0, 10 mumol.L-1 could dose-dependently decrease the release of PAF (9.8 +/- 1.2, 6.5 +/- 1.6, 4.7 +/- 0.8, 3.4 +/- 0.4 microgram.L-1 and 9.2 +/- 1.7, 5.2 +/- 1.3, 3.7 +/- 0.4, 3.2 +/- 0.3 microgram.L-1, respectively, no drugs 11.8 +/- 1.2 micrograms.L-1), Bay k 8644 at 1.0, 5.0, 10 mumol.L-1 could dose-dependently increase the release of PAF (13.2 +/- 1.7, 16.2 +/- 1.4, 17.6 +/- 1.5 micrograms.L-1), and the effects of Tet and Fura 2-AM were completely or partly reversed by Bay k 8644 at 5.0 mumol.L-1. CONCLUSION: Although LPS did not increase intracellular Ca2+ of PM?, intracellular Ca2+ was necessary for PAF release from rat PM? stimulated by LPS.  相似文献

Cibacron Blue F3GA (CB) inhibited the activities of wheat leaves NADH:nitrate reductase and NADH:cytochrome-c reductase in a time-independent and concentration dependent manner. The methyl viologen:nitrate reductase activity of the enzyme was unaffected by various CB concentrations used in the experiment. Inhibition of NADH:nitrate reductase was of mixed type (partial competitive and pure noncompetitive) with respect to NADH and noncompetitive with respect to nitrate. The estimated inhibition constant (Ki) values were 1 microM for NADH and 8.4 microM for nitrate. The secondary plots of inhibition with respect to NADH, indicated a dissociation constant (KI) of 8.8 microM for the enzyme-NADH-CB complex. This KI being greater than the Ki suggested that the noncompetitive inhibition is predominant over the competitive inhibition at the NADH binding site.  相似文献

应用含有梯度外源镧(La)(0、16mg/L、64mg/L、256mg/L、512mg/L和1024 mg/L)的液体培养基培养黑曲霉,每间隔8h取样测定黑曲霉对还原糖和蛋白质的吸收和利用率。培养3天后,测定黑曲霉质膜三磷酸腺苷酶(ATPase),胞内乳酸脱氢酶(LDH)和淀粉酶活性,以及脂质过氧化产物(MDA)的变化。结果表明,在0~64mg/L外源La作用下,黑曲霉对还原糖和蛋白质的吸收与利用率、质膜Na+,K+-ATPase、Ca2+-ATPase、Mg2+-AT-Pase、胞内LDH酶和淀粉酶活性均随La3+的增加而升高,而MDA呈现下降趋势。在64mg/L~1024mg/L剂量范围内,上述生理指标则呈现相反的变化趋势。因此,La3+诱导了黑曲霉生理代谢的Hormesis效应。  相似文献

Experiments were performed on eight subjects affected by peripheral arterial occlusive disease (PAOD) of the lower limbs. Each patient was submitted to Ecodoppler, angiography and the "Treadmill test". Two bioptic muscle of these patients. A sample was used for the spectrophotometric and spectrophotofluorimetric determinations of: glycogen, pyruvate, lactate, citrate, alpha-ketoglutarate, malate, aspartate, glutamate, AMP, ADP, ATP and creatine phosphate (CP). The other bioptic sample was used to determine the following enzyme activities: hexokinase, phosphofructokinase, pyruvate kinase, lactate dehydrogenase, citrate synthase, succinate dehydrogenase, malate dehydrogenase, total NADH cytochrome c reductase, cytochrome oxidase, aspartate aminotransferase and alanine aminotransferase. Patients showed an increase in lactate dehydrogenase, total NADH cytochrome c reductase and succinate dehydrogenase activities, a decrease in glycogen, ATP and CP concentrations. Telethermographic data showed patient muscle thermic emission quantitatively different from control group. The telethermographic test can be used as an additional diagnostic tool to determine and monitor the efficiency of a muscle undergoing metabolic failure.  相似文献

通过模拟试验研究了镧对太湖地区水稻土酸性磷酸酶、脲酶及蔗糖酶活性的影响。镧对水稻土酸性磷酸酶活性表现出抑制作用 ,当镧浓度大于 6 0 mg/kg时 ,抑制作用达到显著水平 ,最大抑制率达到 13%;镧对水稻土脲酶活性影响不显著 ;镧对水稻土蔗糖酶活性表现出一定的刺激作用 ,当镧浓度为 30 0 mg/kg时 ,刺激作用达到显著水平 ,最大刺激率为 10 %。  相似文献
