New interpenetrating polymeric network (IPN) hydrogels based on chitosan (C), poly(N‐vinyl pyrrolidone) (PVP) and poly(acrylic acid) (PAAc), crosslinked with glutaraldehyde (G) and N,N‘‐methylenebisacrylamide (MBA), were prepared and investigated for potential gastrointestinal drug delivery vehicles utilizing a model drug, amoxicillin. IPN hydrogels were synthesized by simultaneous polymerization/crosslinking of acrylic acid monomer in the presence of another polymer (C) and crosslinker (G, MBA). Three different concentrations of glutaraldehyde were used (0.5, 1.0 and 2.0 w/w) to control the overall porosity of the hydrogels, named C‐P‐AAc/0.5, C‐P‐AAc/1.0 and C‐P‐AAc/2.0, respectively. Spectroscopic and thermal analyses such as Fourier transform infrared spectroscopy, thermogravimetric analysis and thermomechanical analysis were performed for IPN characterization. Equilibrium swelling studies were conducted for pH and temperature response behavior. Swelling studies were also carried out in simulated gastric fluid of pH = 1.1 and simulated intestinal fluid of pH = 7.4 to investigate possible site‐specific drug delivery. It was found that the release behavior of the drug from these IPN hydrogels was dependent on the pH of the medium and the proportion of crosslinker in the IPN. It was observed that amoxicillin release at pH = 7.4 was higher than at pH = 1.1. These interpolymeric hydrogels were synthesized by a γ‐irradiation polymerization technique. The degree of gelation was over 96% and increased as the chitosan or acrylic acid (AAc) content increased. The equilibrium swelling studies of hydrogels prepared under various conditions were carried out in an aqueous solution, and the pH sensitivity in a range of pH 1–12 was investigated. The AAc/chitosan hydrogels showed the highest water content when 30 vol % AAc and 0.1 wt % chitosan were irradiated with a 30 kGy dose of radiation. In addition, an increase of the degree of swelling with an increase in the pH was noticed and it had the highest value at pH 12. The drug 5‐fluorouracil was loaded into these hydrogels and the release studies were carried out in simulated gastric and intestinal fluids. The in vitro release profiles of the drugs showed that more than 90% of the loaded drugs were released in the first 1 h at intestinal pH and the rest of the drug was released slowly. © 2003 Wiley Periodicals, Inc. A series of pH‐responsive hydrogels were studied as potential drug carriers for the protection of insulin from the acidic environment of the stomach before releasing in the small intestine. Hydrogels based on poly(vinyl alcohol) networks grafted with acrylic acid or methacrylic acid were prepared by a two‐step process. Poly(vinyl alcohol) hydrogels were prepared by gamma ray irradiation (50 kGy) and then followed by grafting either acrylic acid or methacrylic acid onto these poly(vinyl alcohol) hydrogels with subsequent irradiation (5–20 kGy). These graft hydrogels showed pH‐sensitive swelling behavior and were used as carriers for the controlled release of insulin. The in vitro release of insulin was observed for the insulin‐loaded hydrogels in a simulated intestinal fluid (pH 6.8) but not in a simulated gastric fluid (pH 1.2). The release behavior of insulin in vivo in a rat model confirmed the effectiveness of the oral delivery of insulin to control the level of glucose. © 2003 Wiley Periodicals, Inc. A series of novel hydrogels were prepared from acrylic acid (AA), N‐vinyl pyrrolidone (NVP), and chitosan by photopolymerization. The swelling behavior, gel strength, and drug release behavior of the poly(AA/NVP) copolymeric hydrogels and corresponding interpenetrating polymer network hydrogels were investigated. Results showed that the swelling ratios for the present hydrogels decreased with an increase of NVP content in the gel, but the gel strength increased with an increase of NVP content in the gel. Results also showed that the drug‐release behavior for the gels is related to the ionicity of drug and the swelling ratio of the gel. © 2003 Wiley Periodicals, Inc. Semi‐interpenetrating polymer network hydrogels with different compositions of chitosan (Cs), acrylic acid, and citraconic acid were synthesized via free‐radical polymerization with ethylene glycol dimethacrylate as a crosslinker. The variations of the swelling percentages of the hydrogels with time, temperature, and pH were determined, and Cs–poly(acrylic acid) (PAA) hydrogels were found to be most swollen at pH 7.4 and 37°C. Scanning electron micrographs of Cs–PAA and Cs–P(AA‐co‐CA)‐1 (Cs‐poly(acrylicacid‐co‐citraconir acid)?1) were taken to observe the morphological differences in the hydrogels. Although the less swollen hydrogel, Cs–P(AA‐co‐CA)‐1, had a sponge‐type structure, the most swollen hydrogel, Cs–PAA, displayed a uniform porous appearance. Fluconazole was entrapped in Cs–P(AA‐co‐CA)‐1 and Cs–PAA hydrogels, and the release was investigated at pH 4.0 and 37°C. The kinetic release parameters of the hydrogels (the gel characteristic constant and the swelling exponent) were calculated, and non‐Fickian diffusion was established for Cs–PAA, which released fluconazole much more slowly than the Cs–P(AA‐co‐CA)‐1 hydrogel. BACKGROUND: A considerable amount of research has been focused on smart hydrogels that can respond to external environmental stimuli, especially temperature and pH. In this study, fast responsive thermo‐ and pH‐sensitive poly[(N,N‐diethylacrylamide)‐co‐(acrylic acid)] hydrogels were prepared by free radical copolymerization in aqueous solution using poly(ethylene glycol) (PEG) as a pore‐forming agent. RESULTS: Swelling studies showed that the hydrogels produced had both temperature and pH sensitivity. The deswelling kinetics at high temperature demonstrated that the shrinking rates were influenced by the addition of the pore‐forming agent and the amount of acrylic acid in the initial total monomers. The deswelling curves in low‐buffer solutions had two stages. Pulsatile swelling studies indicated that the PEG‐modified hydrogels were superior to the normal ones. These different swelling properties were further confirmed by the results of scanning electron microscopy. CONCLUSION: Such fast responsive thermo‐ and pH‐sensitive hydrogels are expected to be useful in biomedical fields for stimuli‐responsive drug delivery systems. Novel pH‐sensitive chitosan‐poly(acrylamide‐co‐itaconic acid) hydrogels were prepared by free radical copolymerization of acrylamide and itaconic acid (IA) in chitosan solution. The hydrogels were characterized using Fourier transform infrared spectroscopy, scanning electron microscopy, thermogravimetric analysis, differential scanning calorimetry and the swelling ratios of the hydrogels in water (pH 6.8) and pH 1.2. The influence of composition on the thermal properties of the hydrogels was assessed. The glass transition temperatures of the samples increased with IA content, ranging from 110 to 136 °C. Swelling of the hydrogels was found to obey second‐order kinetics with respect to the remnant swelling, indicating that diffusion is controlled by the relaxation of chains. The equilibrium swelling degree was strongly dependent on pH and composition. At both pH values the highest water uptake was obtained for the IA‐free sample M1. From the equilibrium swelling results the average molar mass between crosslinks, Mc, and the crosslink density of the chitosan‐poly(acrylamide‐co‐itaconic acid) samples were calculated. The results evidenced the reinforcing effect of IA on the hydrogel structure. It is concluded that these highly swellable pH‐sensitive hydrogels can be useful for applications in biomedicine and pharmacy. © 2013 Society of Chemical Industry  相似文献

Poly(2‐ethyl‐2‐oxazoline) and acrylic acid were copolymerized in different compositions using γ‐rays‐induced polymerization and cross‐linking to obtain a series of pH‐sensitive hydrogels. The preparation parameters that may affect the copolymerization process such as the feed solution composition and irradiation dose were optimized. Swelling characteristics of the obtained polymeric hydrogels were evaluated. The results show the significant effects of the hydrogel composition, soaking time, and pH on the swelling equilibrium. The diffusion parameters obtained at pH 1 and 7 show the possibility of using the prepared hydrogels in the field of colon‐specific drug delivery systems. Ibuprofen as a model drug was loaded into (poly(2‐ethyl‐2‐oxazoline)/acrylic acid) copolymer hydrogel to investigate their drug release behavior at different pH values. © 2011 Wiley Periodicals, Inc. To reach sustained drug release, a new composite drug‐delivery system consisting of poly(d,l ‐lactide‐co‐glycolide) (PLGA) nanoparticles (NPs) embedded in thermosensitive poly(N‐isopropyl acrylamide) (PNIPAAm) hydrogels was developed. The PNIPAAm hydrogels were synthesized by free‐radical polymerization and were crosslinked with poly(ethylene glycol) diacrylate, and the PLGA NPs were prepared by a water‐in‐oil‐in‐water double‐emulsion solvent‐evaporation method. The release behavior of the composite hydrogels loaded with albumin–fluorescein isothiocyanate conjugate was studied and compared with that of the drug‐loaded neat hydrogel and PLGA NPs. The results indicate that we could best control the release rate of the drug by loading it to the PLGA NPs and then embedding the whole system in the PNIPAAm hydrogels. The developed composite hydrogel systems showed near zero‐order drug‐release kinetics along with a reduction or omission of initial burst release. The differential scanning calorimetry results reveal that the lower critical solution temperature of the developed composite systems remained almost unchanged (<1°C increase only). Such a characteristic indicated that the thermosensitivity of the PNIPAAm hydrogel was not distinctively affected by the addition of PLGA NPs. In conclusion, an approach was demonstrated for the successful preparation of a new hybrid hydrogel system having improved drug‐release behavior with retained thermosensitivity. Hydrogels, composed of poly(N‐vinyl‐2‐pyrrolidone) and crosslinked polyacrylamide, were synthesized and the release of vitamin B12 from these hydrogels was studied as a function of the degree of crosslinking and pH of the external swelling media. The three drug‐loaded hydrogel samples synthesized with different crosslinking ratios of 0.3, 0.7, and 1.2 (in mol %) follow different drug‐release mechanisms, that is, chain relaxation with zero‐order, non‐Fickian and Fickian, or diffusion‐controlled mechanisms. To establish a correlation between their swelling behavior and drug‐release mechanism, the former was studied by the weight‐gain method and, at the same time, the concentration of the drug released was studied colorimetrically. Various swelling parameters such as the swelling exponent n, gel‐characteristic constant k, penetration velocity v, and diffusion coefficient D were evaluated to reflect the quantitative aspect of the swelling behavior of these hydrogels. This paper investigates the pH‐dependent micellization of (hydroxyethyl cellulose)‐graft‐poly(acrylic acid) (HEC‐graft‐PAA), the crosslinking of the pH‐induced nanoparticles and the drug loading of the crosslinked multi‐morphological nanoparticles. The hydrodynamic diameter (〈Dh〉) of the pH‐induced micelles was found dependent on both temperature and concentration in dynamic light scattering studies. After the crosslinking of PAA segments in the micelles, shape‐persistent nanoparticles were obtained, which exhibited multiple morphologies from nano‐cage to hollow sphere, and a homogeneous swollen microgel in neutral medium with an increase of the degree of crosslinking. To investigate the drug loading of these shape‐persistant nanoparticles, diminazene aceturate was chosen as a model drug. In this study, we facilely introduce silver nanoparticles into Poly(N‐isopropylacrylamide‐co‐acrylic acid)(Poly(NIPAM‐co‐AA)) microgels and specially focus on the effect of hydrophilic acrylic acid segments on the responsive catalytic performance of silver nanoparticles. The obtained Poly(NIPAM‐co‐AA)/AgNPs composites are characterized by Fourier transform infrared spectra, X‐ray diffraction, X‐ray photoelectron spectroscopy, and transmission electron microscopy. The composites as catalysts are applied to the hydrogenation reaction of p‐nitrophenol and the related conditions such as reaction temperature, concentration of p‐nitrophenol, and the loadings of Ag nanoparticles are studied in detail. NIPAM segments of composites conveniently give silver nanoparticles a controllable characteristic for catalytic reaction by their conformation variation. AA segments of composites not only provide good stability and dispersibility for silver nanoparticles but also favor an easier diffusion of p‐nitrophenol to Ag NPs. POLYM. In this study, a series of poly(acrylamide‐co‐acrylic acid‐co‐2‐hydroxy ethyl actylate) [AM‐co‐AA‐co‐HEA] hydrogels have been synthesized by varying the acrylic acid (AA)content over eightfold in feed in the range of 33.34–93.76% by keeping other monomer constant. These hydrogels were characterized by FTIR, SEM analysis, elemental analysis, residual acrylic acid analysis, network parameters, and dynamic swelling behavior. The swelling study showed that equilibrium swelling ratio was nonlinearly increased with increasing AA content. Interestingly, the equilibrium swelling ratio decreased from 53.42 to 48.52 for 75–80% AA content hydrogel. The swelling data were found to satisfactorily fit Fick's second law, demonstrating that diffusion rate of water uptake was primarily Fickian. From model fitting, it was observed that early model was applicable for first 30% water absorption, and late model was applicable for latter 70% water absorption for increasing AA content from 33.34–90.90%. For 93.76% AA, early‐time model was extended up to first 50% of water absorption and late model was contracted for latter 50% water absorption, indicating that excessive AA content affects the applicability range of early‐time and late‐time diffusion models for water absorption. Hard gelatin capsules, containing riboflavin‐loaded poly (N‐viny1–2‐pyrrolidone)‐polyacrylamide cylindrical hydrogels, were modified chemically by treating with an aqueous formaldehyde solution for the purpose of delayed release of drug along the gastrointestinal tract. The tdis (disintegration time) of capsules was studied as a function of concentration of formaldehyde solution and the treatment time. The dynamic release of vitamin B2 was studied as a function of crosslinking ratio of the hydrogels. An effective method was developed to isolate toxic heavy metal ions from the aqueous solution by the magnetic nanopolymers. The magnetic sorbent was prepared with radiation‐induced crosslinking polymerization of chitosan (CS), 2‐acrylamido‐glycolic acid (AMGA), and acrylic acid (AAc), which stabilized by magnetite (Fe3O4) as nanoparticles. The formation of magnetic nanoparticles (MNPs) into the hydrogel networks was confirmed by Fourier transform infrared spectroscopy, X‐ray diffraction, transmission electron microscopy, and Scanning electron microscopy, which revealed the formation of MNPs throughout the hydrogel networks. The swelling behavior of the hydrogels and magnetic ones was evaluated at different pH values. The adsorption activity for heavy metals such as Cu2+ and Co2+ by nonmagnetic and magnetic hydrogels, Fe3O4/CS/(AMGA‐co‐AAc), in terms of adsorption amount was studied. It was revealed that hydrogel networks with magnetic properties can effectively be used in the removal of heavy metal ions pollutants and provide advantageous over conventional ones. POLYM. ENG. Hydrogels have been widely used as mild biomaterials due to their bio‐affinity, high drug loading capability and controllable release profiles. However, hydrogel‐based carriers are greatly limited for the delivery of hydrophobic payloads due to the lack of hydrophobic binding sites. Herein, nano‐liposome micelles were embedded in semi‐interpenetrating poly[(N‐isopropylacrylamide)‐co‐chitosan] (PNIPAAm‐co‐CS) and poly[(N‐isopropylacrylamide)‐co‐(sodium alginate)] (PNIPAAm‐co‐SA) hydrogels which were responsive to both temperature and pH, thereby establishing tunable nanocomposite hydrogel delivery systems. Nano‐micelles formed via the self‐assembly of phospholipid could serve as the link between hydrophobic drug and hydrophilic hydrogel due to their special amphiphilic structure. The results of transmission and scanning electron microscopies and infrared spectroscopy showed that the porous hydrogels were successfully fabricated and the liposomes encapsulated with baicalein could be well contained in the network. In addition, the experimental results of response release in vitro revealed that the smart hydrogels showed different degree of sensitiveness under different pH and temperature stimuli. The results of the study demonstrate that combining PNIPAAm‐co‐SA and PNIPAAm‐co‐CS hydrogels with liposomes encapsulated with hydrophobic drugs is a feasible method for hydrophobic drug delivery and have potential application prospects in the medical field. © 2018 Society of Chemical Industry  相似文献

Cylindrical hydrogels, composed of starch and poly(acrylic acid), were synthesized, and their swelling behavior was studied as a function of the pH of the medium. The gels underwent a sharp transition from Fickian swelling behavior (swelling exponent n = 0.30) to non‐Fickian swelling behavior (n = 0.96) as the pH of the swelling medium changed from 2.0 to 7.4. The hydrogels also underwent partial enzymatic degradation in an amylase‐containing medium of pH 7.4 at 37°C. The effects of the enzyme concentration in the swelling media, the amount of starch present in the gel, the initial water content, the degree of crosslinking, and the diameter of cylindrical hydrogels on the degradation behavior were studied. The degradation of the gels followed Michaelis–Menten kinetics, and the value of the Menten constant was 41.62 × 10?2. The gels exhibited minimum swelling in an acidic pH medium through the formation of a complex hydrogen‐bonded structure and underwent enzymatic degradation in a medium of pH 7.4 (i.e., simulating intestinal fluid) along with chain‐relaxation‐controlled swelling. Polymer solutions that gel in response to changes in temperature and pH are of interest for various forms of drug delivery, and it is desirable to increase swelling for diffusion‐controlled release without bringing the lower critical solution temperature (LCST) above 37°C. N‐isopropylacrylamide (NIP) was polymerized with maleic acid (MAc), a diprotic acid, and acrylic acid (AAc), a monoprotic acid, to compare swelling and temperature response with changes in pH. For samples with equal acid contents and almost identical LCST responses to pH, poly(N‐isopropylacrylamide‐co‐maleic acid) (pNIP MAc) demonstrated greater swelling than poly(N‐isopropylacrylamide‐co‐acrylic acid) (pNIP AAc). The LCST increase for MAc occurred at a pH corresponding to the deprotonation of almost all of the first acid groups. Further increases in pH led to the deprotonation of the second ? COOH and only served to increase the charge concentration at a given location. These results provide strong support for the theory that LCST results largely from uninterrupted chain lengths of NIP and that swelling results from the actual charge density of acid groups along the chain. Because the use of a diprotic acid copolymer allows swelling to be decoupled from LCST, pNIP MAc may be an appropriate candidate for pH‐sensitive drug‐delivery applications. © 2004 Wiley Periodicals, Inc. The current study focuses on the alteration of properties of the gelatin hydrogels using polysaccharides (e.g., maltodextrin, dextran, and sodium carboxymethyl cellulose) for probable use in vaginal delivery of antimicrobials. The hydrogels were prepared by varying the proportions of gelatin and polysaccharides and were characterized by microscopy, mechanical testing, and impedance spectroscopy. Metronidazole (MZ), drug of choice for the treatment of bacterial vaginosis, was incorporated within the hydrogels. In vitro release studies of MZ from the hydrogels was studied in‐depth using modified Franz's diffusion cell. Antimicrobial efficiency of the MZ‐loaded hydrogels was tested against E. coli and B. subtilis. The results suggested that the incorporation of polysaccharides resulted in the phase‐separated hydrogels. The properties of the hydrogels was found be suitable for vaginal delivery. The drug release and antimicrobial efficiency from the hydrogels suggested that the developed hydrogels may be used for the delivery of antimicrobials in the vaginal lumen. © 2014 Wiley Periodicals, Inc.
