以红曲米为原料采用两步提取法提取红曲色素,第一步提取去除桔霉素,第二步提取红曲色素。实验结果表明,第一步提取的最佳条件为提取溶剂乙酸乙酯、提取溶剂pH2.5、提取温度50℃、提取时间60min、料液比1:15(g:mL)、提取次数3次。第二步提取的最佳条件为乙醇浓度85%、提取温度60℃、提取时间50min、料液比1:20(g:mL)、提取次数2次。采用上述最佳条件两步提取可得到低桔霉素高色价的红曲色素产品,红曲色素得率8.31%、色价18625U/kg、桔霉素含量1.37mg/kg,桔霉素含量低于红曲色素中桔霉素最低限量指标。  相似文献

以红曲菌丝体为原料,通过单因素实验及正交设计优化了超声辅助乙醇提取红曲色素的工艺条件。结果表明,最佳提取工艺条件为:超声波功率200 W,乙醇浓度70%,液料比10∶1,提取温度50℃。在该工艺条件下提取40 min,红曲色素的提取率达到92.43%,3次平行实验标准相对偏差RSD为1.58%。该提取工艺简便、高效、重复性好。  相似文献

以荞麦皮为原料,色素得率为指标,在单因素试验基础上,采用正交试验设计法优化超声波辅助提取荞麦皮色素工艺,考察料液比、超声时间、乙醇体积分数、超声功率等因素对荞麦皮色素提取率的影响。结果显示,在超声温度80℃下,超声波辅助提取荞麦皮色素最佳提取工艺为:乙醇体积分数50%,超声时间30 min,超声波功率500W,料液比1︰50 (g/mL)。在该优选工艺条件下,荞麦皮色素得率为5.81%(RSD=0.18%)。该工艺条件操作稳定,有较好重复性。  相似文献

通过Plackett-burman设计法研究料液比、超声功率、乙醇体积分数、提取温度和时间5个因素对芡实叶黄酮提取效果的影响,选择影响效果较显著的4个因素——料液比、超声功率、乙醇体积分数、提取时间通过正交试验优化得出超声提取芡实叶黄酮最佳工艺条件为料液比1:25(g/mL)、超声功率175W、乙醇体积分数85%、提取时间40min,在此条件下芡实叶黄酮提取量为90.28mg/g。  相似文献

以蒲公英根为原料,采用单因素和正交试验优化超声波复合酶法提取蒲公英根总黄酮工艺。结果表明:最佳工艺参数为超声功率90 W、超声温度50℃、料液比1∶60(g/m L)、p H 4.5、乙醇浓度75%时,此条件下黄酮得率最高,为18.24 mg/g。影响超声波复合酶法提取蒲公英根总黄酮工艺的因素由强到弱为超声功率超声温度料液比p H乙醇浓度。  相似文献

采用超声波辅助提取技术对松籽壳色素进行分离。首先,对松籽壳进行预处理,考察乙醇体积分数、液料比、超声温度、超声功率4个因素对色素提取率的影响;然后,利用正交设计法优化提取条件,确定最佳提取条件为:乙醇体积分数50%、液料比25∶1(mL/g)、超声温度60℃、超声功率200W。在此条件下松籽壳色素的提取率为3.304%。该结果为松籽壳资源的综合利用和产品开发提供科学参考。  相似文献

为优化超声波辅助提取桑叶槲皮素工艺,以桑叶槲皮素提取量为指标,通过单因素试验,探讨液料比、乙醇体积分数、超声时间、超声功率及超声温度等对槲皮素提取量的影响,利用响应面法对影响槲皮素提取量的4个主要因素进行优化,分别为乙醇体积分数、液料比、超声功率、超声温度。结果表明,最佳提取工艺条件为乙醇体积分数51%、液料比26∶1(m L/g)、超声功率200 W、超声温度70℃,在此条件下,做3次平行实验进行验证,桑叶槲皮素提取量为11.13 mg/g,与模型预测值11.31 mg/g基本相符。模型可较好地预测桑叶槲皮素的提取量,响应面法对桑叶槲皮素提取条件参数优化具有可行性。  相似文献

Since grapevine ( Vitis spp .) rootstock material is being traded increasingly as disbudded woody material a lack of distinctive morphological features on such material necessitates an alternative and reliable means of identification. Methods described here were developed for rapid and efficient extraction of DNA from woody samples rich in phenolic compounds and polysaccharides, and for subsequent identification of varieties by RAPD PCR. Using these methods, and with the application of only one selected RAPD primer, we were able to differentiate sixteen rootstock varieties, including the seven varieties most commonly used in Germany. Problems commonly encountered with reproducibility of RAPD patterns were avoided by choosing primers with a dinucleotide sequence and a high G/C content that allowed a rather high annealing temperature of 45°C. Methods described here should also be useful for other horticultural crops, especially those with woody tissues rich in phenolic compounds and polysaccharides.  相似文献

The characterization of the aromatic profile of several apricot cultivars with molecular tracers in order to obtain objective data concerning the aromatic quality of this fruit was undertaken using headspace–solid phase microextraction (HS–SPME). Six apricot cultivars were selected according to their organoleptic characteristics: Iranien, Orangered, Goldrich, Hargrand, Rouge du Roussillon and A4025. The aromatic intensity of these varieties measured by HS–SPME–Olfactometry were defined and classified according to the presence and the intensity of grassy, fruity and apricot like notes. In the six varieties, 23 common volatile compounds were identified by HS–SPME–GC–MS. Finally, 10 compounds, ethyl acetate, hexyl acetate, limonene, β-cyclocitral, γ-decalactone, 6-methyl-5-hepten-2-one, linalool, β-ionone, menthone and (E)-hexen-2-al were recognized by HS–SPME–GC–O as responsible of the aromatic notes involved in apricot aroma and considered as molecular tracers of apricot aromatic quality which could be utilized to discriminate apricot varieties.  相似文献

The advent of the functional barrier concept in food packaging has brought with it a requirement for fast tests of permeation through potential barrier materials. In such tests it would be convenient for both foodstuffs and materials below the functional barrier (sub-barrier materials) to be represented by standard simulants. By means of inverse gas chromatography, liquid paraffin spiked with appropriate permeants was considered as a potential simulant of sub-barrier materials based on polypropylene (PP) or similar polyolefins. Experiments were performed to characterize the kinetics of the permeation of low molecular weight model permeants (octene, toluene and isopropanol) from liquid paraffin, through a surrogate potential functional barrier (25 μm-thick oriented PP) into the food simulants olive oil and 3% (w/v) acetic acid. These permeation results were interpreted in terms of three permeation kinetic models regarding the solubility of a particular model permeant in the post-barrier medium (i.e. the food simulant). The results obtained justify the development and evaluation of liquid sub-barrier simulants that would allow flexible yet rigorous testing of new laminated multilayer packaging materials.  相似文献

This paper describes the second part of a project undertaken to develop certified mussel reference materials for paralytic shellfish poisoning toxins. In the first part two interlaboratory studies were undertaken to investigate the performance of the analytical methodology for several PSP toxins, in particular saxitoxin and decarbamoyl-saxitoxin in lyophilized mussels, and to set criteria for the acceptance of results to be applied during the certification exercise. Fifteen laboratories participated in this certification study and were asked to measure saxitoxin and decarbamoyl-saxitoxin in rehydrated lyophilized mussel material and in a saxitoxin-enriched mussel material. The participants were allowed to use a method of their choice but with an extraction procedure to be strictly followed. The study included extra experiments to verify the detection limits for both saxitoxin and decarbamoyl-saxitoxin. Most participants (13 of 15) were able to meet all the criteria set for the certification study. Results for saxitoxin.2HCl yielded a certified mass fraction of <0.07 mg/kg in the rehydrated lyophilized mussels. Results obtained for decarbamoyl-saxitoxin.2HCl yielded a certified mass fraction of 1.59+/-0.20 mg/kg. The results for saxitoxin.2HCl in enriched blank mussel yielded a certified mass fraction of 0.48 +/- 0.06 mg/kg. These certified reference materials for paralytic shellfish poisoning toxins in lyophilized mussel material are the first available for laboratories to test their method for accuracy and performance.  相似文献
