开发了一种测定超临界二氧化碳中大分子溶质的溶解度的方法。这一方法将微型超微界流体萃取直接与超临界色谱相耦合,超临界流体色谱采用FID作为检测器。实验中两者具有同一压力、温度及同样的CO2流速度。  相似文献

制备生物柴油的超临界流体技术是使用动、植物油脂与超临界甲醇反应生产脂肪酸甲酯的新工艺.与传统的催化反应技术相比,超临界技术生产生物柴油的反应时间由1~8 h缩短至3~4min,而且反应不需催化剂,简化了产品提纯过程,提高了酯交换反应后脂肪酸甲酯的产率,并对环境友好.  相似文献

A supercritical fluid extraction (SFE) method has been developed for the extraction of lipids in bilberry. Experimental design was used to optimize pressure, temperature and extraction time using CO2 as solvent. Best SFE condition for total lipids was 450 bar, 60 °C and 45 min. The SFE method was compared to conventional Bligh & Dyer (B&D) extraction. The amount of fatty acid methyl esters (FAME) was found to be 4.84 ± 0.06 mg and 4.564 ± 0.003 mg per g of the freeze‐dried bilberry sample for the developed SFE and B&D methods, respectively, while the amount of total lipids was found to be 54.40 ± 6.06 mg and 65.70 ± 0.67 mg per g of sample for SFE and B&D, respectively. This discrepancy between FAME and total lipids could be explained by the presence of wax esters, sterol esters, carotenoids and phospholipids, as determined by supercritical fluid chromatography.  相似文献

This review enlightens the role of supercritical fluid chromatography (SFC) in the field of lipid analysis. SFC has been popular in the late 1980s and 1990s before almost disappearing due to the commercial success of liquid chromatography (LC). It is only 20 years later that a regain of interest appeared when new commercial instruments were introduced. As SFC is fully compatible with the injection of extracts in pure organic solvent, this technique is perfectly suitable for lipid analysis and can be coupled with either highly universal (UV or evaporative light scattering) or highly specific (mass spectrometry) detection methods. A short history of the use of supercritical fluids as mobile phase for the separation oflipids will be introduced first. Then, the advantages and drawbacks of SFC are discussed for each class of lipids (fatty acyls, glycerolipids, glycerophospholipids, sphingolipids, sterols, prenols, polyketides) defined by the LIPID MAPS consortium.  相似文献

用气相色谱法测定了50-100℃下烷烃和芳烃分别在正十六烷、正十七烷、正二十烷、正二十四烷、正三十烷中的亨利系数考虑了固定液流失量和上非理想性的校正。讨论了亨利系数H与溶质碳数和溶剂碳数之间的关系。结果证明:亨利系数随着溶碳数和溶剂碳数的增加而减少。  相似文献

Triacylglycerol (TAG) isomers have been reported to have differing physical and nutritional properties. The analysis of TAG isomers is therefore important for understanding the physical properties of lipids as well as their digestion and absorption. However, methods for the quantitative analysis of TAG regioisomers and enantiomers in vegetable oils and biological samples are still under development. Recently, methods using recycle high-performance liquid chromatography (HPLC) and silver ion column-HPLC have been reported. However the recycle HPLC method requires more than 1 hour, in general, for each sample that is analyzed. Furthermore, existing methods are unable to quantify regioisomers and enantiomers simultaneously. Thus, we aimed to develop a practical method to simultaneously quantify regioisomers and enantiomers of TAG. Three isomers of sn-POO, OPO, and OOP were separated by supercritical fluid chromatography coupled with triple quadrupole mass spectrometer (SFC/MS/MS) using a CHIRALPAK® IG-U column with acetonitrile and methanol as mobile phase. The separation was completed in 40 min, which is a shorter run time than the conventional techniques published to date. Linear calibration curves with standards were obtained and used to quantify sn-OPO, sn-POO, and sn-OOP in extra virgin olive oil, refined olive oil, palm oil, palm olein, and interesterified palm olein.  相似文献

The separation method using chiral stationary phase (CSP) for the preparation of enantioselective compound was widely used. In this work, supercritical fluid chromatography(SFC) was proposed to resolve the chiral mixtures. To determine the optimum operating conditions for the chiral separation of the racemic ibuprofen, the retention factors and resolutions with the change in pressure, temperature and the content of IPA (%, by volume) in supercritical CO2 were investigated. Experiments showed that the retention factor decreased with the increase of pressure and decrease in temperature. The retention factor was also influenced by the content of IPA in mobile phase, as the content of IPA in the supercritical fluid increased, the retention factor decreased. The resolution of the enantiomers became worse with the increase of IPA in the supercritical fluid. Through optimizing the experimental conditions, a SFC procedure with 13MPa, 311.15K and 4% IPA in CO2 was obtained. The peak shape of the enantiomers was symmetric with supercritical fluid chromatography when compared to the asymmetric peak shape obtained by the conventional liquid chromatography. This work demonstrated that the developed supercritical fluid chromatography procedure was suitable for the chiral separation of ibuprofen enantiomers.  相似文献
