The male sexual behavior-stimulating and inhibiting properties of a series of analogs of the European corn borer sex pheromone were determined in a flight tunnel. The structural requirements for inhibition of pheromonal response were far less restrictive than those for elicitation of that response. Analogs that by themselves elicited upwind flight response from males at a low dose were generally less inhibitory to male response than many of the analogs that had no pheromonal activity. These findings suggest that many pheromone analogs bind to pheromone receptors without provoking behavioral response and possibly undergo slower degradation on the antenna than pheromonally active compounds. The disparity of response to analogs by two pheromonal types of the European corn borer indicates that the pheromone receptor and pheromone catabolic systems are biochemically very different in the two types.  相似文献

Isomers of [11,12-3H2]-11-tetradecenyl acetate (57 Ci/mM) were synthesized. Behavioral assay of the two compounds using Z- and E-type European corn borer (ECB) males showed that introduction of tritons into the double bond of the pheromone caused a significant isotope effect in the E-type ECB but not in the Z-type ECB. Measurements of tritium associated with the male antennae after a 3-min exposure showed that radioactivity equivalent to 10–7 mol pheromone was adsorbed onto male antennae. Time-course in vivo metabolic studies with picogram amounts of compound applied topically to antennae of E- and Z-type males and Z-type females showed that they metabolized pheromone similarly but females degraded pheromone more slowly than males. Pheromone was hydrolyzed, and the only other major radiolabeled metabolite observed by combined high-pressure liquid chromatography-radiodetection was tritiated water. Capillary gas chromatography and radiomonitoring permitted detection of a trace amount of 11-tetradecenoic acid, which indicated alcohol oxidase activity is associated with the antennae. Evidence shows that clearing of pheromone from the ECB male antennae involves hydrolysis and oxidation of the alcohol to fatty acid, which in turn is degraded, probably via -oxidation, to carbon dioxide and water.  相似文献

Sex gland extracts ofOstrinia nubilalis females collected in the wild or laboratory-reared from Switzerland, Italy, and Hungary were analyzed. Individuals collected in the north of Switzerland contained (Z)- and (E)-11-tetradecenyl acetate at the approximate ratio of 973 (Z type), in accordance with field responses of males and previous findings. On the other hand, females from a laboratory culture derived from field collections made in the same area and reared for four to five generations contained theZ andE isomers at ratios of ca. 397 and 3565, respectively. In the south, one of the eight wild females analyzed was of theZ type and the rest intermediate, whereas males were predominantly trapped with blends of the two isomers containing 60 to 97E. In a laboratory culture reared for one to two generations from corn borers collected in Hungary, three of nine females were of the intermediate type and the restZ. Small amounts of (Z)-11-hexadecenyl acetate were detected in female glands of theE strain; however, no effect of this compound could be observed in the field.  相似文献

The biological activity of analogs of the pheromone components of the European corn borer,Ostrinia nubilalis, (Z)- and (E)-11-tetradecen1-ol acetate, in which modifications were made in the terminal alkyl portion were studied in the three pheromonal types of the insect. European corn borer males respond to pheromonal stimuli at three levels of behavioral activity, i.e., short-range sexual stimulation, activation in the flight tunnel, and response in the field. Structural requirements for elicitation of response at these levels were found to be increasingly restrictive, respectively. Flighttunnel activity was induced only by compounds that had a total chain length of 13 or 14 carbons and in which branching at carbon 13 was limited to one methyl group or a cyclopropyl group. Three new analogs were active in the flight tunnel, viz., (E and (Z)-13-methyl-11-tetradecen-1-ol acetate and (Z)-12-cyclopropyl-11-dodecen-1-ol acetate. The cyclopropyl analog was the most active analog against theZZ type of the European corn borer. TheE isomer, however, was pheromonally inactive in theEE type and was shown to be a pheromone antagonist. This dissimilarity is most likely due to differences in structure of the receptors in the European corn borer strains. Analogs that were biologically active against the European corn borer were tested against the redbanded leafroller,Argyrotaenia velutinana, which also uses (Z)- and (E)-11-tetradecen-1-ol acetate as part of its pheromone. Results showed that the redbanded leafroller pheromone acceptor system is different from that of the European com borer; marginal behavioral response was elicited by only one of the new analogs. Thus, although both species use 11-tetradecen-1-ol acetate isomers as their pheromone, the mechanisms by which they are perceived are different.Mention of a commercial product does not constitute endorsement of that product by USDA.  相似文献

Experiments were conducted to (1) determine whether the electroantennogram (EAG) can detect differences among the responses of antennae from males derived from the three strains ofOstrinia nubilalis (Hübner), and (2) characterize the EAG responses of each strain to isomeric forms of the natural pheromone, (E)- and (Z)-11-tetradecen-1-ol acetate (TDA), and analogs possessing differences in the terminal alkyl group, cyclopropyl (CPA), ortert-butyl (TBA).EAG responses differed among the strains in two ways: (1) Antennae fromZZ males always produced an EAG to (Z)-TDA with an extended duration of response. This signature EAG response was found to be unique to the antennal response ofZZ males to (Z)-TDA, thus providing a relatively easy method of distinguishing liveZZ males fromEE orZE males. Correlated with this longer EAG response was a longer disadaptation time, i.e., the EAG response ofZZ antennae disadapted more slowly (ca. 10 min) than the response ofEE antennae. (2) Strain differences in the relative EAG amplitudes to isomers and analogs were observed at the stimulus amounts eliciting the peak EAG amplitude as follows: TDA CPA > TBA forZZ males and both isomers; TDA > CPA TBA and CPA TDA > TBA forEE males and theE andZ isomers, respectively; CPA > TBA TDA forZE males and both isomers. Dose—response relationships were seen for all compounds if amplitude (peak height) of the EAG was used as a measure of response. However, if width of the EAG at half the peak height (peak width) was used, then only theZZ antennal response to (Z)-TDA resulted in a meaningful dose-response relationship. For all strains, the EAG amplitudes elicited by theZ isomers of any of the tested compounds were greater than those elicited by the correspondingE isomers. Therefore, correlations between the relative EAG and upwind flight responses were observed in theZZ (r = 0.86) andZE (r = 0.80) strains but were not correlated in theEE strain (r = 0.18). Temporal studies showed that adaptation, not postexcision deterioration, was responsible for the observed decreases in the EAG amplitude after repetitive stimulation or after stimulation with amounts in a descending order. Disa-daptation required at least 20 min for a moderate dose (10 g for 1 sec). Developmental studies showed that antennae from 2-day-old adults had the greatest EAG response.  相似文献

When physiologically excessive amounts of the female sex pheromone of the European corn borer (ECB) or esters analogous to the pheromone were applied to the antennae of males, their behavioral responsiveness to pheromone in a flight tunnel was significantly impaired for 2 hr. Concurrent quantitative analyses of heptane extracts of the male antennae by gasliquid chromatography showed that the compounds applied to antennae were hydrolyzed and, at 2 and 4 hr posttreatment, little or none of the compound applied or hydrolysis product was detectable in the antennal extracts. After 4 hr of in vivo incubation, male responsiveness to pheromone was restored among moths treated with the analogs but not among moths treated with pheromone. Esterase activity on the antennae was moderately inhibited in vivo by a pheromone analog that is a so-called transition-state esterase inhibitor, 1,1, 1-trifluoro-14-heptadecen-2-one. However, the analog did not inhibit male behavior when it was coevaporated with pheromone in a flight-tunnel assay. Therefore, in the presence of pheromone, the analog did not compete well for esterase or the pheromone receptor. Treating the antennae of intact males with tetrahydrofuran obliterated sex pheromone response capability in males, but the treatment did not significantly attenuate esterase and other catabolic activity of the antennae. Indications are that degradation of esters on the ECB antennae involves substrate-nonspecific esterase activity and other metabolic processes that in turn remove hydrolysis products from the antennae. Maintenance of a male's ability to respond to pheromone is linked to these processes.  相似文献

Structure-activity relationship (SAR) observations were made for the bagworm moth pheromone, (R)-2-pentyl decanoate, and a series of analogs with modifications in the alcohol portion of the molecule. Observed attractiveness of these analogs was related to molecular structure and their physical attributes using computational chemistry. Electrostatic potential and Van der Waals (VdW) electrostatic coded surface three-dimensional (3D) maps of the molecular mechanics (MM) minimized lowest energy conformation of the pheromone show that size, shape, charge distribution, and chirality of the molecule are related to attractiveness.  相似文献

The responsiveness pf 3-day-old European corn borer males to three concentrations of 973 ZE 11-tetradecenyl acetate, the female sex pheromone, was studied over a range of relative humidities (43–100%) in a wind tunnel. The proportion of males taking flight and reaching the source decreased, while the proportion exhibiting in-flight arrestment of upwind progress increased under high humidity conditions at all three concentrations of pheromone tested. The relationships between relative humidity and these behaviors were best described by polynomial equations.  相似文献

Behavioral responses of males from three New York races of European corn borer,Ostrinia nubilalis (Hübner), to various suspected sexpheromone-response antagonists were investigated with a flight tunnel protocol. Males from a bivoltine (Z) race and from a univoltine (Z) race utilizing a natural pheromone blend of 982 (Z)-(E)-11-tetradecenyl acetates and males from a bivoltine (E) race utilizing a 199 (Z)-(E)-11-tetradecenyl acetate ratio were flown to their natural blends and to blends containing 1 % of one of the following additional compounds: Z9–12OAc,E9–12OAc,Z9–14OAc, andE9–14OAc. In each race, the added components lowered significantly the number of individuals completing the behavioral sequence. The only exception was bivoltineE males flown to a blend withE9–12OAc added. The number completing the sequence in this case was statistically not significantly lower than the number that completed the sequence to the standard blend. In all three races, theZ9–14OAc produced the most dramatic reduction in completed flights, and in the univoltineZ race, this added component was significantly more effective in reducing completed flights than any other added component. The response to the Z9–14OAc is understandable given recently published findings of an electrophysiological study of single sensilla in the European corn borer.  相似文献

Sex pheromone behavioral responses were analyzed in a flight tunnel with European corn borer,Ostrinia nubilalis (Hübner), males from three distinct populations. Males from a bivoltine and a univoltine biotype using a 97.82.2 blend (Z strains) of (Z)- and (E)-11-tetradecenyl acetate were assayed with treatments containing 0, 0.5, 1, and 3% of theE isomer. Males from neither population oriented in the plume to the 100%Z treatment, but bivoltine males oriented and flew to the source to the other three treatments, whereas univoltine males oriented and flew to the source only to the 1 % and 3 %E treatments. Males from a bivoltine biotype using a 199 blend ofZ/E isomers (E strain) were assayed with sources containing 0, 0.5, 1, and 3% of theZ isomer. Males did not orient to the 0%Z source, but oriented and flew to the source to the other three treatments. In addition to using opposite geometric isomers for the main pheromone component, the bivoltineE strain differed from the bivoltineZ strain by producing and responding better to 1 % of the minor component and by storing 3 times more pheromone in the female glands. Contrary to previous reports, the blend of two pheromone components is significantly better than the main component alone in eliciting oriented flight and close-range behavior with males from all three populations of European corn borer.  相似文献

Pheromone blend analyses of glands from individual female European corn borers,Ostrinia nubilalis (Hübner), from field-collected larvae or pupae associated with bivoltine flights in June and August and a univoltine flight in July have shown that: (1) a site in western New York has a bivoltine biotype utilizing (Z)-11-tetradecenyl acetate as its primary pheromone component (designatedZ), (2) two sites in central New York have mixed populations consisting of a bivoltine biotype utilizing theE pheromone isomer (designatedE) and a univoltine biotype utilizingZ, and (3) one site in central New York was found to have only the univoltineZ biotype. The combinations of voltine biotypes and pheromone strains found in New York support the existence of three European corn borer populations designated bivoltineE, bivoltineZ, and univoltineZ.  相似文献

The feeding preference of European corn borer larvae for immature whorl tissue of maize was examined by conducting leaf bioassays and quantifying resistance factors along the length of mid-whorl leaves from the maize synthetic BS9(C4) developed by recurrent selection for resistance. Potential resistance factors that were quantified included percent foliar nitrogen, gravimetric determination of soluble metabolites and fiber, soluble phenolics and hydroxamic acids, cell-wall-bound phenolics, leaf toughness, and UV absorbance of the epidermal cell wall determined by microspectrophotometry. Larvae consumed immature tissue at a higher rate than more mature tissue outside of the whorl, despite higher levels of DIMBOA in immature tissue. Consumption rate was highly negatively correlated with epidermal cell wall absorbance and leaf toughness. Fiber content and phenolic fortification of cell walls are proposed as the major resistance components that influence European corn borer feeding preference within the resistant synthetic BS9(C4).PRC contribution no. 1562.  相似文献

Sex pheromone activity ofEriborus terebrans (Gravenhorst) (Hymenoptera: Ichneumonidae) was recovered from acetone rinses of flasks that previously contained females. The acetone flask rinses elicited the following male responses: upwind anemotaxis, casting, hovering, landing, wingfanning, and mating attempts with other nearby males. Activity of the acetone flask rinse lasted up to four days on a glass substrate. Polar component and nonpolar components were demonstrated in the acetone flask rinse. The polar component elicited male behavioral responses similar to those by the acetone flask rinse, although retention of males at the pheromone source and the period of wing-fanning were of shorter duration. Chromatography data and chemical derivatization indicated that the polar component had the properties of a carboxylic acid with an additional oxygen-containing functional group. The non-polar component acted as a synergist since it was inactive alone but increased male behavioral responses when added to the polar component. Florisil open column chromatography suggested that the nonpolar component was a hydrocarbon(s).  相似文献

The female sex pheromone of the Asian com borer,Ostrinia furnacalis, widespread in Taiwan, was confirmed as (Z)-12-tetradecenyl acetate and its geometric isomer (E)-12-tetradecenyl acetate in a ratio of ca. 31 by gas chromatography and gas chromatography-mass spectrometry in selected ion monitoring mode. Males were attracted by the mixture of these two synthetic components in the field, but the attractiveness was less than by virgin females. The presence of minor components in the sex pheromone was therefore suggested.  相似文献

A glass tube olfactometer bioassay was used to examine pheromone response of males of the (Z)-pheromone strain ofOstrinia nubilalis (Hubner). The presence of (E)-11-tetradecenyl acetate at the natural ratio to (Z)-11-tetradecenyl acetate (973; ZE) did not consistently elevate wing-fanning, upwind walking, or clasper extrusion over (Z)-11-tetradecenyl acetate alone. Four C14 straight-chain acetate esters—(Z)-11-tetradecenyl acetate, (E)-11-tetradecenyl acetate, (E)-9-tetradecenyl acetate, and tetradecyl acetate—were isolated and identified from the diethyl ether extracts of the abdominal tips of adult female European corn borers,Ostrinia nubilalis (Hübner). The compounds were isolated using a combination of high-resolution gel-permeation, adsorption highpressure liquid, and micropreparative gas chromatography. The pheromone-mediated flight and landing behaviors of maleOstrinia nubilalis were studied in a wind tunnel. The pheromone source was placed in the middle of an 18 × 18-cm horizontal surface, and a smaller surface placed 4, 18, or 36 cm downwind. The smaller surface did not appear to affect significantly the flight tracks or position of landing of males on the upwind surface, and it allowed the positions and altitudes of males as they passed over the downwind surface to be estimated. The flight altitude and position of males as they passed over the downwind surface related to where males landed on the upwind surface. Regardless of the downwind position of the downwind surface, most males flew over its center (i.e., in line with the source) and landed in line with the source on the upwind surface. When a small 2.5 × 10-cm vertical object was placed on the upwind surface, just upwind and to one side of the source, males flew over the downwind surface in positions skewed toward the vertical object and in broader distributions than for the comparable situation without an object: males landed on the upwind surface on positions skewed toward, or on, the object and with a broader distribution (laterally). Flight altitude also corresponded with landing position. Thus, when there was no vertical object, most males flew just above the downwind surface and landed on the downwind edge of the upwind surface. In contrast, with the vertical object, males flew significantly higher and tended to land past the downwind edge of the upwind surface. With a taller object (20 cm), males flew even higher, past the downwind edge and most landed on the vertical object. These data show the close relationship between flight and landing behaviors of maleO. nubilalis and suggest that flight maneuvers that determine track and altitude largely govern where a male lands.  相似文献

The cocoa pod borer,Conopomorpha cramerella (Snellen) (Lepidoptera: Gracillariidae), is the most serious pest of cocoa in Southeast Asia. Analyses of ovipositor washings and entrained volatiles from virgin female moths by gas chromatography (GC) linked to electroantennography (EAG), and comparison of EAG responses from the male moth to synthetic compounds indicated the presence of theE,Z,Z andE,E,Z isomers of 4,6,10-hexadecatrienyl acetate and the corresponding alcohols, and of hexadecyl alcohol. Amounts of pheromone produced were less than 0.1 ng/female, and no peaks for the unsaturated components were observed on GC analysis. Extensive field testing of synthetic mixtures in Sabah, East Malaysia, showed that traps baited with a polyethylene vial impregnated with 1.2 mg of a mixture of the above five components in 40604610 ratio caught more maleC. cramerella moths than traps baited with a virgin female moth.  相似文献

The volatile plant sesquiterpenoids farnesene, nerolidol, and farnesol were tested to determine their effect on European corn borer,Ostrinia nubilalis, oviposition during the first six nights of the adult stage. Adult European corn borers were released into cages designed to encourage oviposition on eight glass plates randomly arranged on top of the cage: four coated with either 80µg/cm2 farnesene, nerolidol, or farnesol, and four coated with the solvent methylene chloride. The setup was used as a binary choice test. Farnesene was preferred by females because they deposited significantly more egg masses on plates coated with the compound. Nerolidol had no effect. Farnesol, by contrast, deterred oviposition: the number of egg masses was significantly lower on plates covered with that compound. These data show that structural modification at a single terminal functional group in these compounds affects the ovipositional behavior of European corn borer females.This paper reports the results of research only. Five maytansinoids fromMaytenus (Celastraceae) andPutterlickia (Rhamnaceae) species were tested for biological activity against the European corn borerOstrinia nubilalis. Maytanbutine, maytansine, and maytanvaline, all of which contain an amino acid residue at C-3, were active and comparable in their effect on larvae to trewiasine, a known active, amino acid-containing maytansinoid fromTrewia nudiflora. Maytanacine, which has an acetate group at C-3, was not as active as maytansine, maytanvaline, maytanbutine, or trewiasine, but significantly retarded the development of the larvae. Normaysine, which has no oxygen substituent at C-3, had no significant effect on mortality and only moderate effect on development of the larvae.
