以新疆马奶酒中自行分离得到的副干酪乳杆菌(Lactobacillus paracasei)M5L为对象,研究该菌株对结肠癌Caco-2细胞的抑制作用并分析发酵乳贮藏品质变化。结果表明,副干酪乳杆菌M5L可以在一定的盐浓度、胆盐浓度及酸性环境中较好的生长。当菌体以100∶1的感染系数作用于Caco-2细胞72 h后抑制率最佳;显微镜下观察活菌处理后的细胞发生明显的凋亡现象;流式细胞仪检测到副干酪乳杆菌M5L可以诱导Caco-2细胞内活性氧显著增加,并将细胞周期阻滞在合成期,且通过增大Caspase-3和Bax基因表达量,降低Bcl-2基因表达量来促进肿瘤细胞的凋亡。其发酵酸乳贮藏5 d时品质较佳,长时间的贮存会导致酸乳品质的劣变。 相似文献

本研究基于盐酸咯哌丁胺建模制造小鼠便秘模型,考察副干酪乳杆菌 N1115发酵乳对治疗小鼠便秘的 作用。低、中、高剂量副干酪乳杆菌N1115发酵乳(活菌含量分别为1×107、1×108、1×109 CFU/mL)连续灌胃 15 d后,与对照组相比小鼠的排便量及粪便含水量均显著升高(P<0.05);肠道推进率分别为57.3%、67.3%和 71.3%,显著高于对照组(P<0.05)。气相色谱仪分析小鼠粪便中的短链脂肪酸发现,灌胃副干酪乳杆菌N1115 发酵乳后,小鼠粪便中的乙酸、丙酸、丁酸、戊酸含量显著升高(P<0.05),且短链脂肪酸种类较模型组更为丰 富。对小鼠远端结肠的组织形态观察发现,灌胃副干酪乳杆菌N1115发酵乳的小鼠结肠黏膜完整,细胞排列整齐, 而对照组的肠黏膜厚度降低,凹窝变浅。通过c-kit抗体对小鼠结肠中的肠间质细胞(interstitial cells of Cajal,ICC)进 行免疫组化分析,结果显示,灌胃副干酪乳杆菌N1115发酵乳后小鼠的ICC含量显著增加,表明副干酪乳杆菌N1115发 酵乳通过提高ICC活性,促进肠道蠕动。综上,副干酪乳杆菌N1115发酵乳能够有效缓解小鼠的便秘症状。 相似文献

以副干酪乳杆菌作为发酵剂,研究其在滋味(可溶性物质)和香气(挥发性化合物)两方面对发酵香肠风味的改善情况。采用电子鼻分析未添加发酵剂的香肠—自然发酵(1#)与添加发酵剂的香肠(2#)的香气组成,并利用气相色谱-质谱法(GC-MS)和氨基酸自动分析仪测定其挥发性成分及可溶性呈味物质的含量。结果表明,添加副干酪乳杆菌的发酵香肠较未添加的氨基酸的含量有所提高,谷氨酸、天门冬氨酸、甘氨酸、丝氨酸、丙氨酸等呈味氨基酸含量增加明显。醇、酸、酯、酮类等挥发性成分物质增加,说明副干酪乳杆菌可以很好的改善发酵香肠的风味。 相似文献

This study investigated the profiles of volatile compounds produced by Lactobacillus paracasei during oat fermentation using gas chromatography‐mass spectrometry coupled with headspace solid‐phase microextraction method. A total of 60 compounds, including acids, alcohols, aldehydes, esters, furan derivatives, hydrocarbons, ketones, sulfur‐containing compounds, terpenes, and other compounds, were identified in fermented oat. Lipid oxidation products such as 2‐pentylfuran, 1‐octen‐3‐ol, hexanal, and nonanal were found to be the main contributors to oat samples fermented by L. paracasei with the level of 2‐pentylfuran being the highest. In addition, the contents of ketones, alcohols, acids, and furan derivatives in the oat samples consistently increased with the fermentation time. On the other hand, the contents of degradation products of amino acids, such as 3‐methylbutanal, benzaldehyde, acetophenone, dimethyl sulfide, and dimethyl disulfide, decreased in oat samples during fermentation. Principal component analysis (PCA) was applied to discriminate the fermented oat samples according to different fermentation times. The fermented oats were clearly differentiated on PCA plots. The initial fermentation stage was mainly affected by aldehydes, whereas the later samples of fermented oats were strongly associated with acids, alcohols, furan derivatives, and ketones. The application of PCA to data of the volatile profiles revealed that the oat samples fermented by L. paracasei could be distinguished according to fermentation time. 相似文献

研究Lactobacillus paracasei W2分批发酵过程中细胞生长、苯乳酸(PLA)的积累、葡萄糖消耗的变化规律。基于Logistic方程和Luedeking-Piret方程,建立了PLA分批发酵过程中细胞生长、产物合成及基质消耗随时间变化的数学模型。通过Matlab 7.0 软件进行最优参数估计和非线性拟合,拟合模型R2均大于0.955,显示了模型与实验数据能较好地吻合,反映了Lactobacillus paracasei W2分批发酵合成PLA过程的动力学特征。 相似文献

用气相色谱-质谱联用(GC-MS)技术建立了拟干酪乳杆菌发酵过程中胞内代谢物的代谢组学研究方法。考察了不同溶解剂(吡啶、四氢呋喃、二甲基甲酰胺)和衍生化试剂(N-甲基叔丁基二甲基硅基三氟乙酰胺(MTBDSTFA)、N,O-双(三甲基硅烷基)三氟乙酰+三甲基氯硅烷(BSTFA+TMCS)、N-甲基-N-三甲基硅烷基三氟乙酰胺(MSTFA)),对质谱峰的信号强度和个数的影响,确定使用吡啶作为溶解剂、BSTFA+TMCS作为衍生剂的两步衍生法效果最佳。以拟干酪乳杆菌为分析对象,共鉴定出38种胞内中间代谢物。方法学考察结果表明,该方法具有良好的精密度、稳定性和重现性。本方法已成功地应用于拟干酪乳杆菌发酵过程研究,并且发现了发酵过程中可能的生物标志物。 相似文献

旨在研究副干酪乳杆菌VL8胞外多糖(Lactobacillus paracasei VL8 exopolysaccharides,VL8-EPS)对巨噬细胞RAW264.7的免疫激活作用。试验采用不同浓度的VL8-EPS作用于RAW264.7细胞,设空白对照组,脂多糖(lipopolysaccharide,LPS)处理组(终浓度为10μg/mL)和VL8-EPS处理组(终浓度为50、100、200、400、800μg/mL)。迪夫快速染色法观察细胞形态;比色法测定诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)活力;WST-1法测定超氧化物歧化酶(superoxide dismutase,SOD)活力;微量法检测超氧阴离子(O2-)的含量;钼酸显色法检测过氧化氢(H2O2)的分泌量。结果表明,VL8-EPS能够改变细胞形态,剂量依赖性提高RAW264.7细胞内iNOS、SOD酶活力,促进细胞分泌O2^-和H2O2。在800μg/mL时,较空白对照组分别提高了104.64%、10.30%、666.87%、81.78%。说明VL8-EPS通过提高胞内酶活性及释放活性氧激活RAW264.7细胞从而发挥免疫增强作用。 相似文献

目的:分析和预测副干酪乳杆菌(Lactobacillus paracasei)乳酸脱氢酶(lactate dehydrogenase,LDH)基因及其编码蛋白的结构和特性,以指导其生物学功能的实验研究。方法:利用美国国家生物技术信息中心(NCBI)和瑞士生物信息学研究所的蛋白分析专家系统(ExPASY)中有关基因和蛋白的序列和结构信息分析的各种工具,结合其他生物信息学分析软件,如ClustalX、VMD,从数据库中得到乳酸脱氢酶基因,分析、预测该基因编码的蛋白理化性质、翻译后的修饰位点、拓扑结构、二级结构、三级结构和功能域。并与其他微生物的乳酸脱氢酶蛋白序列进行比对,得出它们的保守序列。结果:该基因编码335个氨基酸,其蛋白理论分子质量为36608.8u,预测该蛋白有3个跨膜区。与干酪乳杆菌的乳酸脱氢酶进化关系最近。结论:应用生物信息方法可预测得到副干酪乳杆菌的乳酸脱氢酶的结构与功能方面的信息。 相似文献

Abstract: A dried tomato‐flavored probiotic cream cheese (P) containing Lactobacillus paracasei Lpc‐37 was developed for the purpose of this study. The same product, but without probiotic addition (C) was used as control. Lactococcus lactis subsp. lactis and Lactococcus lactis subsp. cremoris were used as lactic starter cultures. Chemical composition analyses and sensory tests were performed on days 1 and 7, respectively. Titratable acidity, pH value and L. paracasei population were determined every 7 d during the refrigerated storage (21 d) of the cream cheeses. The experiment and analyses were performed in triplicate, using standard methods. Probiotic population remained greater than 107 CFU/g throughout the storage period, thereby characterizing the product as potentially probiotic. Cream cheeses C and P did not differ on the sensory tests, both obtaining good overall acceptance by the consumers, of which 82.6% stated that they certainly or probably would buy the product. Practical Application: Lactobacillus paracasei Lpc‐37 is a probiotic bacterium and clinical studies have shown that this microorganism beneficially affects its host. In general, dried tomato‐flavored products and cream cheese are products with good acceptance by the consumers. Thus, regular consumption of the probiotic cream cheese developed in this study may have positive effects on health and well being of people if incorporated into their diet. 相似文献
