Because of the potentially high arsenic concentrations found in soils immediately adjacent to chromated copper arsenate (CCA)-treated wood structures and utility poles, CCA-contaminated soil ingestion may be a significant exposure route to arsenic for children. Therefore, a strong need exists to provide accurate data on oral relative bioavailability (RBA) of arsenic (in vivo or in vitro) in field-collected CCA-contaminated soils. The objectives of this study were (1) to assess arsenic bioaccessibility in contaminated soils collected near in-service CCA-treated utility poles, (2) to determine the influence of soil properties and arsenic fractionation on arsenic bioaccessibility, and (3) to estimate an average daily arsenic intake from incidental soil ingestion. Arsenic bioaccessibility (in vitro gastrointestinal (IVG) method) was determined on surface soil samples collected immediately adjacent to 12 CCA-treated utility poles after 18 months of service. Bioaccessible arsenic was also determined in 3 certified reference materials. Total arsenic concentrations in soils (<300 microm) varied from 37 +/- 2 to 251 +/- 12 mg/kg, irrespective of soil organic matter contentwith the major soil-bound arsenic species being As(V). Arsenic bioaccessibility ranged between 25.0 +/- 2.7 and 66.3 +/- 2.3% (mean value 40.7 +/- 14.9%). The mean value was in agreement with the in vivo arsenic RBA reported by Casteel et al. (2003) in soil near CCA-treated utility poles. Bioaccessible arsenic was positively correlated with total organic carbon content (r2 = 0.36, p < 0.05) and with water-soluble arsenic (2 = 0.51, p < 0.01), and was negatively correlated with clay content (r2 = 0.43, p < 0.05). Using conservative exposure parameters, the mean daily arsenic intake from incidental ingestion of contaminated soil near CCA-treated utility poles was 0.18 +/- 0.09 microg As kg(-1) d(-1). This arsenic intake appeared negligible compared to the daily intake of inorganic arsenic from water and food ingestion for children.  相似文献

Tortillas were prepared using commercial hydrocolloids, stored for 7 and 14 days and their available, resistant and retrograded resistant starches, were evaluated alongside their in vitro starch digestibility. Available starch (AS) decreased with storage time and tortillas with hydrocolloids had lower values than the control sample. Tortillas elaborated with TC-20 gum did not present substantial differences in AS. Control tortilla had resistant starch (RS) content that increased with storage time but, in general, tortillas with hydrocolloids did not show any change in RS values with storage time, except tortillas with TC-1 gum that presented a slight increase after 7 storage days. Approximately 50% of RS is due to the retrogradation phenomenon as it was shown by the amount of retrograded resistant starch (RRS). Tortillas with added hydrocolloids had lower hydrolysis percentage and the hydrolysis was slower than in the control. In general, tortillas prepared with hydrocolloids had a lower tendency for retrogradation than control tortillas; it is important to consider this to obtain tortillas with better texture and lower RS content.  相似文献

Cereals are a staple food in both developed and developing countries, and are considered to be the best vehicle for iron and calcium fortification, as an important strategy for combating dietary deficits. Inadequate dietary intake of iron and calcium is related to a number of disease conditions such as anemia, osteoporosis, hypertension, and different cancers. From a nutritional point of view, it is interesting to know not only the amount of minerals consumed, but also their bioavailability. The present study reviews the current knowledge on the in vitro bioavailability of iron and calcium in cereals, placing emphasis on the methodologies used and on the influence of dietary factors and food processing.  相似文献

The objective of the present investigation was to analyze the relative influence of oxalic acid, phytic acid, tannin and dietary fiber on in vitro availability of iron and calcium from green leafy vegetables (GLV). Thirteen GLV were selected and analyzed for iron, calcium, oxalic acid, phytic acid, tannin and dietary fiber contents using standard methods. The bioavailability of calcium and iron in the GLV was estimated by equilibrium dialysis. Oxalic acid content was less than 1 g kg?1 in four greens and ranged between 1.22 to 11.98 g kg?1 in the remaining. Dietary fiber ranged from 19.5 to 113.7 g kg?1. Tannin content ranged between 0.6138 and 2.1159 g kg?1 with the exception of two GLV that had 0.1332 and 14.8619 g kg?1. Four GLV were found to have approximately 40% bioavailable iron, while the others were in the range of 6–30%. In vitro available calcium was less than or equal to 25% in eight GLV and between 34% and 52% in five GLV. Multiple regression analysis revealed that these factors together accounted for 53% (r2 = 0.53) and 45% (r2 = 0.45) inhibition of iron and calcium absorption, respectively. Lead (Pb) bioaccessibility was assessed using 2 in vitro methods in 12 Pb-contaminated soils and compared to relative Pb bioavailability using an in vivo mouse model. In vitro Pb bioaccessibility, determined using the intestinal phase of the Solubility Bioaccessibility Research Consortium (SBRC) assay, strongly correlated with in vivo relative Pb bioavailability (R(2) = 0.88) following adjustment of Pb dissolution in the intestinal phase with the solubility of Pb acetate at pH 6.5 (i.e., relative Pb bioaccessibility). A strong correlation (R(2) = 0.78) was also observed for the relative bioaccessibility leaching procedure (RBALP), although the method overpredicted in vivo relative Pb bioavailability for soils where values were <40%. Statistical analysis of fit results from X-ray absorption near-edge structure (XANES) data for selected soils (n = 3) showed that Pb was strongly associated with Fe oxyhydroxide minerals or the soil organic fraction prior to in vitro analysis. XANES analysis of Pb speciation during the in vitro procedure demonstrated that Pb associated with Fe minerals and the organic fraction was predominantly solubilized in the gastric phase. However, during the intestinal phase of the in vitro procedure, Pb was strongly associated with formation of ferrihydrite which precipitated due to the pH (6.5) of the SBRC intestinal phase. Soils where Fe dissolution was limited had markedly higher concentrations of Pb in solution and hence exhibited greater relative bioavailability in the mouse model. This data suggests that coexistence of Fe in the intestinal phase plays an important role in reducing Pb bioaccessibility and relative bioavailability.  相似文献

In this study, 15 soils ranging in Pb content from 32 to 6330 mg kg(-1) were subjected to in vitro gastrointestinal extractions with and without added powdered milk. Before and after treatment, Pb in the soils was fractionated according to a 7-step sequential extraction procedure. A subset of five soils and Pb acetate was used for a minipig dosing study. The amount of bioaccessible Pb determined with the in vitro system ranged from 3 to 20% without powdered milk and from 11 to 56% with powdered milk. The higher bioaccessibility of Pb in the in vitro model with addition of powdered milk was related to a depletion of Pb in the organic Pb pool and indicates that soluble milk constituents compete with soil organic ligands for Pb. The absolute and relative bioavailabilities of Pb in the minipig dosing experiment were not related to bioaccessible Pb determined in any of the two in vitro systems. However, relative bioavailabilities in liver, kidney, and total uptake were highly correlated to Pb in the third fraction of the sequential extraction that is attributed to easily reducible Mn oxides. These results indicate that reductive processes in the intestine may be more relevant for Pb absorption than the initial solubilization in the acidic stomach.  相似文献

食物摄入是许多化学物质进入人体的主要渠道.只有当化学物质能够被释放进入消化液, 才能够发挥毒性作用.介绍了建立体外消化模型用于大米中无机砷生物可给性研究工作.结果显示:无机砷标准溶液能全部进入消化糜中;不同浓度的加标大米样品中无机砷的生物可给性随着加标浓度的上升而下降;现场采集的三种大米样品的无机砷的生物可给性基本一致.体外消化模型对于丰富暴露评估技术和健全风险评估制度将产生良好的影响.  相似文献

A quantitative framework based on a set of dimensionless numbers was developed to capture the effects of competing interfacial and biokinetic processes and define limits on the application of in situ bioremediation. An integrated numerical modeling and experimental approach was utilized to evaluate the quantitative framework. Experiments were conducted to examine the transport and biodegradation of naphthalene in a saturated, heterogeneous intermediate-scale flow cell with two layers of contrasting hydraulic conductivities. The experiments were carried out in two phases: Phase I, simulating intrinsic biodegradation; and Phase II, simulating an engineered in situ bioremediation. In Phase I, dispersion was identified as the overall rate-limiting process based on the proposed quantitative framework. Two engineered perturbations to the system were selected in Phase II to examine their abilities to enhance in situ biodegradation. In the first perturbation, nitrogen and phosphorus were spiked into the influent solution in excess of the required stoichiometric amounts. This perturbation did not have a significant impact because dispersion, not biokinetics, was the overall rate-limiting process. However, in the second perturbation, advection was increased, resulting in increased longitudinal and vertical transverse dispersion, thereby alleviating the rate-limiting process, and enhancing the overall biotransformation rate.  相似文献

Bioavailability of total arsenic, toxic (arsenite, As(III); and arsenate, As(V)), and non-toxic (monomethylarsonic acid, MA; dimethylarsonic acid, DMA; arsenobetaine, AB; and arsenocholine, AC) arsenic species has been assessed in different raw seafood samples (white fish, cold water fish and molluscs) by using an in vitro model that combines simulated gastric and intestinal digestion/dialysis methods. Correlations between arsenic species bioavailability and seafood nutrient contents (fat and protein) have also been established. Total arsenic content in seafood samples, and dialyzable and non-dialyzable fractions, were analyzed by inductively coupled plasma – mass spectrometry (ICP–MS) after a microwave-assisted acid digestion treatment. The determination of the different arsenic species concentrations in the samples (after an optimised matrix solid phase dispersion (MSPD) approach) and in the dialyzable fraction was done by high performance liquid chromatography (HPLC) coupled to ICP-MS as a selective detector. Accuracy of the procedure (total arsenic determination) was assessed by analyzing DORM-2 and BCR-627 certified reference materials. The accuracy of the in vitro procedure was established through a mass-balance study. After statistical evaluation (95% confidence interval), good accuracy of the whole in vitro process, for total arsenic and for arsenic speciation, was observed. High dialyzability percentages for total arsenic and for arsenic species were found (i.e. from 84.6 ± 1.7% to 106 ± 2.6%). Bioavailability of arsenic exhibits a negative correlation with the fat content of the seafood. However, no correlation was observed between the bioavailable fraction of total arsenic and arsenic species and the protein content of the seafood studied.  相似文献

Green leafy vegetables have the potential to contribute significant amounts of iron to the diet, if the bioavailability of iron from these foods is improved. In order to determine the potential for genetic manipulation of this trait, 46 lines from 12 species of Amaranthus were evaluated for total and bioavailable iron in greenhouse and field experiments. Bioavailable iron was estimated using an in vitro assay for dialysable, low-molecular-weight iron compounds. Significant differences (P < 0·01) were detected among lines and species for total and bioavailable iron. Total iron ranged from 358 to 880 ppm (dry weight) in field-grown plants and 55 to 123 ppm (dry weight) in greenhouse-grown plants. Bioavailable iron ranged from 41 to 63 ppm in field-grown plants and from 24 to 51 ppm in greenhouse-grown plants. Amaranthus tricolor and A lividus had the highest total and bioavailable iron; A hypochondriacus had the lowest levels of the species tested. Although field-grown Amaranthus accumulated higher levels of total and bioavailable iron, a greater proportion of the total iron was sequestered in insoluble, unavailable forms. Generally, species with higher total iron had higher levels of bioavailable iron. The assessment of selenium and selenium species bioavailability in foodstuff is of special concern on the context of human nutrition. In vivo (human and animal), and in vitro tests are important approaches for estimating the bioavailability of toxic and essential compounds to humans. An overview on in vivo and in vitro bioavailability assays for releasing selenium and selenium species in foodstuffs is summarized. Se and Se species content in a foodstuff critically influence Se bioavailability and bioactivity to humans and animals. Se bioavailability is affected by foodstuff-matrix major composition and minor components. Foodstuffs processing and/or treatments could enhancement or decrease Se bioavailability. Experimental conditions such as the selection of healthy status of examined people (in in vivo humans approaches), the selection of animal model (in vivo animals approaches), or the selection of GI conditions (in in vitro tests) could determines the results. Thus, international standardized protocol for in vivo and in vitro approaches assessment is mandatory.  相似文献

The food preservatives, sulphur dioxide and its salts, are known to present some toxic, mutagenic and antinutritional effects; in fact they interact with a number of nutrients, e.g. some vitamins, notably thiamine (Th) and folic acid (FA). The effect of different concentrations of sodium bisulphite in cell culture media has been studied in vitro on a human cell line, HEp-2, deriving from a carcinoma of the larynx. Moreover, the sulphites have been tested with different levels of Th and FA with the aim of elucidating how much the cellular response depended on either the anti-nutritional effect or the toxicity of sulphites. Cell growth has been taken as an index of cytotoxicity and measured both as total protein content and as colony-forming ability. With no Th and FA in the culture medium, a clear decrease of cell growth was observed either with or without addition of sodium bisulphite. A dose-dependent reduction of protein content was detected in cells treated with 10, 50, 100, 200, 250 or 500 microM sodium bisulphite. Moreover, when the cells were treated with 10 or 100 microM of this compound, the colony-forming ability was reduced both in number and colony size. As far as the interaction of the two vitamins with sodium bisulphite is concerned, when these nutrients were present in the medium at 0.5, 1.0, 1.5, 2.0 or 2.5 mg/l, a similar growth profile, determined from their concentration, was observed in treated and control cells, the growth levels being affected by the sodium bisulphite contents. At higher levels of Th and FA, the growth index was still increasing only in treated cells, this phenomenon being particularly evident in cultures treated with 200 microM sodium bisulphite. The colony-forming ability was reduced in controls but still increased in treated cells at the highest concentration of vitamins.  相似文献

Iron deficiency anemia is the most common nutritional deficiency in humans. Not all dietary ingested iron, heme or nonheme, will be available to absorption and negative imbalance between iron requirements and absorption leads to iron deficiency and/or anemia. The recommended iron values usually are based on the genetic and on diet iron-bioavailability, which can be considered as the principal factor that change among the cultures and influences the distinct levels of recommendation among countries. Dietary changes present practical limitations due to be difficult to change food habits. The iron food fortification is considered more cost effective and economically more attractive than iron supplementation. There are many iron compounds available to be used in iron fortification. Cereals represent a target food group to iron fortification programs due to high consumption and the in vitro studies can be useful to estimate the relative iron bioavailability in large number of products in short time and with a low cost. Wheat flour baked into bread or not was the main product tested in in vitro bioavailability studies and ferrous sulfate was the principal iron compound used in the fortification studies. However, iron bioavailability from ferrous sulfate is lower than from other compounds, such FeNaEDTA or ferric pyrophosphate. The variables level of fortification, storage, level of extraction, baking and also the association or not with other chemical compound seems to influence the results obtained.  相似文献
