The methodology used by the author consists of the following elements: Expose the design team to the user environment, followed by question and answer periods with users while still in the use environment. Place biomedical engineers in the leading teaching institutions where they will have day-to-day exposure to the use of products similar to the one being designed. Bring biomedical engineers back into the company as part of the design team. Expose concepts to focus groups while the product is in the definition stage. Bring a select group of users into the design review process. Evaluate the ease of use of the device as part of clinical trials. Establish a means of monitoring product performance after the product has been released. How well such a methodology will work in any particular environment is a function of management's recognition of the concept that ease of operator use is a vital element to the overall success of the product. 相似文献

随着蛋白质组学的产生和发展,对蛋白质组表达水平的差异和变化进行体内的和动态的分析已势所必然地成为蛋白质组学研究的发展趋势。但是传统差异蛋白质组学方法只能提供细胞或组织内各种蛋白累积总量的信息,而不是真正意义上的蛋白质差异表达分析。 SiLAD(~(35)S in vivo Labeling Analysis for Dynamic Proteomics)技术正是基于这种需求而在本实验室创立的。SiLAD技术由于其在差异检出的灵敏度和时间分辨率方面的明显优势,以及除提供蛋白质组表达动态以外,可以提供蛋白质代谢等相关动态变化的更多信息,所以可适用于对多种生物过程进行的动态蛋白质组学研究。本文对SiLAD技术的原理及特点进行简要综述。 相似文献

Cataract, the opacification of the eye lens, is the leading cause of blindness worldwide--it accounts for approximately 42% of all cases. The lens fibers have the highest protein content within the body, more than 35% of their wet weight. Given the eye lens pure composition of highly abundant structural proteins crystallins (up to 90%), it seems to be an ideal proteomic entity to study and might be also hypothesized to model the other protein conformational diseases. Crystallins are extremely long-lived, and there is virtually no protein turnover. This provides great opportunities for post-translational modifications (PTM) to occur and to predispose lens to the cataract formation. Despite recent progress in proteomics, the human lens proteome remains largely unknown. Mass spectrometry hold great promise to determine which crystallin modifications lead to a cataract. Quantitative analysis of PTMs at the peptide level with proteomics is a powerful bioanalytical tool for lens-tissue samples, and provides more comprehensive results. New mass spectrometry-based approaches that are being applied to lens research will be highlighted. Finally, the future directions of proteomics cataract research will be outlined. 相似文献

AB SCIEX公司最新发布全新的高分辨液相色谱质谱联用系统——TripleTOF5600是在享有盛名的API 5500三重四极杆系统和QSTAR Elite(QqTOF)系统这两个技术平台的基础上开发出来的。TripleTOF 5600高分辨液相色谱质谱联用系统能够对复杂的蛋白质组学样品进行高通量分析,并能同时提供高质量的定性和定量结果。为评估这款仪器的性能,设计基于纳升液相色谱和TripleTOF 5600联用系统的一系列实验。首先,应用这一拥有高分辨率、高灵敏度和高质量准确度的系统对酵母细胞裂解物的胰酶消化产物进行分析。实验中采用每秒最多采集50幅串联质谱谱图的信息相关采集模式,单次LC-MS/MS实验就鉴定10179条独特肽段和1174种蛋白(at 1%global false discoveryrate)。而实际达到平均每秒34幅串联谱图的采集速度是在单次实验获得如此优异的鉴定结果的决定性因素。此外,也对一组含有5种已知蛋白以及部分重同位素标记肽段的样品进行分析,以检验TripleTOF 5600的定量能力。实验中这5种蛋白质都获得很高的序列覆盖率,其中进样量仅有100amol的Peroxiredoxin 1蛋白的序列覆盖率也高达53%。而测试中得到肽段的平均质量偏差仅为1.7 ppm,展示TripleTOF 5600的高质量准确度和稳定性。此外还获得高度准确的定量结果,充分证明TripleTOF 5600能够提供三重四极杆系统级别的定量结果。 相似文献

简要概述了加强群塔集中管理的必要性以及通用无线分组业务(general packet radio service,GPRS)的应用现状;通过群塔系统的特点分析,提出基于GPRS的群塔数据无线传输方案,解决了工地监控中心与远程群塔集中管理中心的通信问题;然后从软硬件两方面完成基于GPRS技术的群塔数据传输终端设计,并且对基于GPRS网络的无线数据传输系统进行调试,根据调试结果对软件进行了修改完善,最终实现了数据的可靠传输. 相似文献

The systematic analysis of biological processes requires an understanding of the quantitative expression patterns of proteins, their interacting partners and their subcellular localization. This information was formerly difficult to accrue as the relative quantification of proteins relied on antibody‐based methods and other approaches with low throughput. The advent of soft ionization techniques in mass spectrometry plus advances in separation technologies has aligned protein systems biology with messenger RNA, DNA, and microarray technologies to provide data on systems as opposed to singular protein entities. Another aspect of quantitative proteomics that increases its importance for the coming few years is the significant technical developments underway both for high pressure liquid chromatography and mass spectrum devices. Hence, robustness, reproducibility and mass accuracy are still improving with every new generation of instruments. Nonetheless, the methods employed require validation and comparison to design fit for purpose experiments in advanced protein analyses. This paper introduces an innovative one column, many pencil local scanning maskless lithography technology used in dynamic mask and applies it to rapid prototyping (RP) system. By using this technology in micro RP system, the resolution of the 3-D microstructures can be upgraded remarkably. In addition, some relevant experiments are conducted on this micro rapid prototyping system. In the experiment, light beam is projected onto a digital mirror device (DMD) chip [Hornbeck LJ. Digital light processing: A new MEMS-based display, 4th Sensor Symp, p. 297–304 (1996)], and the image is reflected through a micro mirror. Meanwhile, by making use of zoom lens and pins array mask (PAM), the continuing pixels can be scattered. Moreover, in order to project scattered pixels onto the resin surface, the optical system is exploited. Additionally, by using a micro-stage, it is able to control the micro X-Y movement between the scattered pixels. Furthermore, this paper makes use of the spray coating technology to improve the build up capability of the overhand structures of the traditional deep dip liquid-type RP system. Finally, combining the fore-mentioned technologies, one can produce 3-D prototypes with overhand structures in higher resolution. 相似文献

Progress in proteomics research has led to a demand for powerful analytical tools with high separation efficiency and sensitivity for confident identification and quantification of proteins, posttranslational modifications, and protein complexes expressed in cells and tissues. This demand has significantly increased interest in capillary electrophoresis‐mass spectrometry (CE‐MS) in the past few years. This review provides highlights of recent advances in CE‐MS for proteomics research, including a short introduction to top‐down mass spectrometry and native mass spectrometry (native MS), as well as a detailed overview of CE methods. Both the potential and limitations of these methods for the analysis of proteins and peptides in synthetic and biological samples and the challenges of CE methods are discussed, along with perspectives about the future direction of CE‐MS. @ 2019 Wiley Periodicals, Inc.
