3-(3′,4′-Dihydroxyphenyl)-l-alanine (l-DOPA), which is synthesized in velvet bean (Mucuna pruriens), inhibits plant growth. The concentration of l-DOPA in soil is reduced by adsorption and transformation reactions, which can result in the reduction of its plant-growth-inhibitory activity. To determine which part of the l-DOPA structure is involved in the adsorption and soil transformation reactions, we compared the kinetics of l-DOPA disappearance in a volcanic ash soil with that of l-phenylalanine (3-phenyl-l-alanine) and l-tyrosine (3-(4′-hydroxyphenyl)-l-alanine), compounds that are similar in structure to l-DOPA but do not have a catechol (o-dihydroxybenzene) moiety. l-Phenylalanine and l-tyrosine were not adsorbed and transformed in the soil at equilibrium pH values between 4 and 7. These results suggest that the adsorption and transformation reactions of l-DOPA in the soil involve the catechol moiety and not the amino and carboxylic acid groups, which are common to all three compounds. Like l-DOPA, (+)-catechin, another allelochemical that contains a catechol moiety, underwent adsorption and soil transformation reactions. Thus, we concluded that the concentrations of allelochemicals bearing a catechol moiety in soils will decrease rapidly owing to adsorption and transformation reactions, and this decrease will be faster in soils with a high pH value or high adsorption ability. Owing to this decrease in concentration, allelopathic phenomena may not occur.  相似文献

l-DOPA is an active allelochemical that inhibits plant growth. To determine whether the phytotoxicity is due to the reactive oxygen species generated during its oxidation to melanin, oxidative damage, melanin accumulation, and the effect of antioxidants on its phytotoxicity were examined in l-DOPA-tolerant (barnyard grass) and -susceptible (lettuce) plants, and in suspension-cultured carrot cells. l-DOPA suppressed root elongation in lettuce compared to barnyard grass. Levels of melanin and thiobarbituric acid reactive substances (TBARS) increased remarkably in l-DOPA-treated lettuce roots, but not in barnyard grass. l-DOPA also suppressed carrot cell growth to 60% of the control at 1 mM. Melanin content in 1 mM l-DOPA-treated carrot cells increased continuously; however, ascorbic acid and -tocopherol suppressed accumulation. When melanin formation was inhibited by ascorbic acid and -tocopherol, growth of l-DOPA-treated cells was restored. TBARS levels were higher in 1 mM l-DOPA-treated carrot cells than in untreated control cells 2 d after treatment, but not at 4 or 6 d. Ascorbic acid and -tocopherol suppressed the production of lipid peroxide during the initial 2 d. These results suggest that the phytotoxicity of l-DOPA is due to oxidative stress caused by reactive oxygen species from the melanin synthesis pathway.  相似文献

The sex pheromone of Phyllophaga (Phytalus) georgiana was characterized as valine methyl ester, tentatively the l-enantiomer. This is the first sex pheromone identified from the Phyllophaga subgenus Phytalus. The pheromone was extracted from female glands, the active component isolated by coupled gas chromatography–electroantennogram detection analysis, characterized by mass spectrometry, and shown to be active in field tests. The seasonal flight pattern was determined for P. georgiana as well as for three other species, P. anxia (both northern and southern genitalic forms), P. gracilis, and P. postrema. We examined the allelochemical effects of control soil, native soil (treated soil), and leaf extracts of Phytolacca americana (pokeweed) on the germination rate and seedling growth of Cassia mimosoides var. nomame. We also studied the resulting changes in root-tip ultrastructure and peroxidase isozyme biochemistry. P. americana leaf extract inhibited seed germination, seedling growth, and biomass when compared to control and treated soil. Root and shoot growth in treated soil was stimulated relative to control soil, but root growth was inhibited by 50% in the leaf extract treatment. Biomass of C. mimosoides seedlings grown on leaf extract was reduced sevenfold when compared to the control seedlings. The amounts of total phenolic compounds in the leaf extract, treated soil, and control soil were 0.77, 0.14, and 0.03 mg l−1, respectively. The root tips of C. mimsoides treated with leaf extracts of P. americana showed amyloplasts and large central vacuoles with electron-dense deposits inside them when compared to control root tips. The activity of guaiacol peroxidase (GuPOX) in whole plant, roots, and shoots of C. mimosoides increased as leaf extract increased; maximum activity was observed in extract concentrations of 75% and higher. Root GuPOX activity was three times higher than in shoots. Therefore, we conclude that inhibition of C. mimosoides growth is related to the phenolic compounds in the P. americana leaf extract and the ultrastructure changes in root-tip cells and increased GuPOX activity is a response to these allelochemicals.  相似文献

A comparative study was conducted to assess the contact and fumigant toxicities of eleven monoterpenes on two important stored products insects—, Sitophilus oryzae, the rice weevil, and Tribolium castaneum, the rust red flour beetle. The monoterpenes included: camphene, (+)-camphor, (−)-carvone, 1-8-cineole, cuminaldehyde, (l)-fenchone, geraniol, (−)-limonene, (−)-linalool, (−)-menthol, and myrcene. The inhibitory effect of these compounds on acetylcholinesterase (AChE) activity also was examined to explore their possible mode(s) of toxic action. Although most of the compounds were toxic to S. oryzae and T. castaneum, their toxicity varied with insect species and with the bioassay test. In contact toxicity assays, (−)-carvone, geraniol, and cuminaldehyde showed the highest toxicity against S. oryzae with LC50 values of 28.17, 28.76, and 42.08 μg/cm2, respectively. (−)-Carvone (LC50 = 19.80 μg/cm2) was the most effective compound against T. castaneum, followed by cuminaldehyde (LC50 = 32.59 μg/cm2). In contrast, camphene, (+)-camphor, 1-8-cineole, and myrcene had weak activity against both insects (i.e., LC50 values above 500 μg/cm2). In fumigant toxicity assays, 1-8-cineole was the most effective against S. oryzae and T. castaneum (LC50 = 14.19 and 17.16 mg/l, respectively). Structure-toxicity investigations revealed that (−)-carvone—, a ketone—, had the highest contact toxicity against the both insects. 1-8-Cineole—, an ether—, was the most potent fumigant against both insects. In vitro inhibition studies of AChE from adults of S. oryzae showed that cuminaldehyde most effectively inhibited enzyme activity at the two tested concentrations (0.01 and 0.05 M) followed by 1-8-cineole, (−)-limonene, and (l)-fenchone. 1-8-Cineole was the most potent inhibitor of AChE activity from T. castaneum larvae followed by (−)-carvone and (−)-limonene. The results of the present study indicate that (−)-carvone, 1,8-cineole, cuminaldehyde, (l)-fenchone, and (−)-limonene could be effective biocontrol agents against S. oryzae and T. castaneum.  相似文献

This study offers experimental evidence for the existence of three pheromone races of the northern genitalic form of Phyllophaga anxia: one race in which females produce and males respond mainly to l-valine methyl ester, a second producing and responding to l-isoleucine methyl ester, and a third producing and responding to an intermediate range of blends of the two compounds. At Franklinville, NY, pheromone gland contents of females were analyzed using coupled gas chromatography–electroantennogram detection. Two types of females were found, one that produced greater than 99% l-valine methyl ester and another that produced greater than 99% l-isoleucine methyl ester. Capture–mark–release–recapture field tests with males at Franklinville established that most males were recaptured in traps baited with the same blends with which they were originally captured. The populations characterized at Franklinville, NY, have also been found at numerous locations from eastern Canada and the northeast and north central USA, sometimes in allopatry and sometimes in sympatry. At a site in Carver, MA, P. anxia males responded to blends of the methyl esters of l-valine and l-isoleucine, and Carver females produced blends similar to those to which the males responded. Populations responding to blends have been identified only from southeastern Massachusetts and Rhode Island. At a field site near Waterloo, NY, the addition of small proportions of l-isoleucine methyl ester to lures containing l-valine methyl ester did not affect trap captures, but higher proportions of l-isoleucine methyl ester were inhibitory, decreasing trap captures.  相似文献

In this study, a sensor for the sensitive determination of ascorbic acid (AA) has been fabricated based on meso-tetra-(3,5-dibromo-4-hydroxydroxyphenyl) porphyrin copper (II) (T(DBHP)P-Cu) modified Au electrode through l-cysteine (l-cys). Firstly, l-cys modified Au electrode was prepared through self-assembled technology. Then T(DBHP)P-Cu was adsorbed on l-cys/Au through covalent binding. The fabrication process and electrochemical behavior of T(DBHP)P-Cu/l-cys/Au were studied by cyclic voltammetry and differential pulse voltammetry. The results showed that AA exhibited good electrochemical activity at T(DBHP)P-Cu/l-cys/Au. The oxidation peak current increased linearly with AA concentration in the range of 1.00 × 10−3–1.02 × 10−5 mol L−1 with a detection limit of 5.41 × 10−7 mol L−1. Additionally, the modified electrode could be applied to the detect AA in practical samples.  相似文献

The supramolecular recognition of Ca(II) and N α-4-tosyl-l-arginine methyl ester hydrochloride (TAME) with ATP were investigated using 1H and 31P NMR spectra. In the Ca(II)–ATP–TAME ternary system, Ca2+ and TAME bind with ATP via the phosphate chain and adenine ring of ATP. The binding forces are mainly electrostatic and cation (Ca2+)–π and π–π stacking interaction. Furthermore, the hydrolysis of ATP catalyzed by Ca(II) and TAME was studied at pH 7.0 and 60 °C using 31P NMR spectra. Kinetics studies show that the ATP hydrolysis rate constant is 0.1035 h−1 in the Ca(II)–TAME–ATP ternary system, whereas the value is 8.5 × 10−3 h−1 under the same conditions without TAME and Ca2+. The Ca(II) ions and TAME accelerate the ATP hydrolysis process about 12-fold. The proposed mechanism of ATP hydrolysis catalyzed by Ca2+–TAME occurs through an addition–elimination reaction sequence. These results can help us get more useful information at the molecular level about the key amino acid residue(s) and metal ions that serve as cofactors in the ATPase effect on ATP hydrolysis/synthesis.  相似文献

Saturated acyl (6-O-caproyl, lauroyl, and myristoyl) and unsaturated acyl (6-O-oleoyl, linoleoyl, and arachidonoyl) l-ascorbates were continuously synthesized at 50°C using a system where a column packed with ascorbic acid powder and a packed-bed reactor with an immobilized lipase from Candida antarctica were connected in series. A productivity of 1.6–1.9 kg/L reactor·d was achieved for at least 11 d. The surface tension of the caproyl or lauroyl l-ascorbate in aqueous solution was measured at various temperatures and pH to estimate the critical micelle concentration (CMC) of the acyl l-ascorbate. The CMC values were independent of temperature but dependent on the pH. The value of the caproyl ascorbate increased with an increase in pH.  相似文献

Apple replant is a widespread agricultural problem documented in all of the major fruit-growing regions of the world. In order to better understand the phytotoxic mechanisms induced by allelochemicals involved with this problem, Malus prunifolia plants were grown hydroponically to the six-leaf-stage in the presence of phthalic acid (0 or 1 mM) for 5, 10, or 15 days. Apple plants were evaluated for: shoot and root length, fresh and dry weight, malondialdehyde (MDA) content, hydrogen peroxide (H2O2) content, superoxide radical (O2 ·−) generation rate, and antioxidant enzyme activities. Shoot and root lengths and fresh and dry weights of M. prunifolia decreased in plants exposed to phthalic acid. MDA and H2O2 content increased in phthalic acid-treated plants as did the generation rate of O2 ·− in M. prunifolia roots. The activities of superoxide dismutase (EC 1.15.1.1), peroxidase (EC 1.11.1.7), catalase (EC 1.11.1.6), ascorbate peroxidase (EC 1.11.1.11), glutathione reductase (EC 1.6.4.2), dehydroascorbate reductase (EC 1.8.5.1), and monodehydroascorbate reductase (EC 1.6.5.4) increased in phthalic acid-stressed roots compared with control roots. These results suggest that activation of the antioxidant system by phthalic acid led to the formation of reactive oxygen species that resulted in cellular damage and the decrease of M. prunifolia growth.  相似文献

Poly(l-lactic acid)/poly(l-lactic acid)-grafted SiO2 nanocomposites were prepared by in situ melt polycondensation, in which “free” poly(l-lactic acid) and poly(l-lactic acid)-grafted SiO2 nanoparticles were formed simultaneously. The maximum values of grafting ratio and grafting efficiency of poly(l-lactic acid) were up to 37.67% and 26.60%, respectively. In the polycondensation system, SiO2 content was a critical parameter of getting nanocomposites with uniformly dispersed SiO2 nanoparticles. At lower SiO2 content, Mn of grafted poly(l-lactic acid) was close to that of “free” poly(l-lactic acid), and poly(l-lactic acid)-grafted SiO2 nanoparticles could be well dispersed in poly(l-lactic acid) matrix. While at higher SiO2 content, Mn of “free” poly(l-lactic acid) and grafted poly(l-lactic acid) decreased seriously, especially GPC curves of “free” poly(l-lactic acid) exhibited two peaks due to the aggregation of SiO2 nanoparticles during the polycondensation process. The grafting ratio and SiO2 content exhibited a clear effect on the thermo-oxidative degradation of nanocomposites. The existence of poly(l-lactic acid)-grafted SiO2 nanoparticles dramatically improved the thermo-oxidative stability of poly(l-lactic acid). Compared with that of pure poly(l-lactic acid), T g, T c, and T m of nanocomposites varied slightly.  相似文献

The behavioral responses of the potato tuberworm moth Phthorimaea operculella and the polyphagous predator Orius insidiosus to volatiles emanating from exposed tubers were studied by four-arm olfactometer bioassays. Mated females of P. operculella distinguished volatiles released by intact potato tubers from volatiles damaged mechanically or by conspecific larvae. Volatiles from intact potato tubers were attractive to them. On the other hand, unmated females of P. operculella did not respond to tuber volatiles. Adults of O. insidiosus were attracted to volatiles from tubers damaged by P. operculella larvae, but did not respond to intact or mechanically damaged tubers. Methyl jasmonate (MeJA) was the only compound identified from the headspace of potato tubers (GC-MS of direct headspace sampling). The amount varied with the type of induction, being 0.001 ± 0.0003 ng g−1 in tissues of intact fresh tubers, 0.002 ± 0.0007 ng g−1 in mechanically damaged tubers, and showing a six- to tenfold increase in P. operculella damaged tubers (0.090 ± 0.006 ng g−1). Behavioral bioassays with synthetic MeJA confirmed that the response of the insects is dependent on MeJA concentration. Mated females of P. operculella showed the highest response at 0.001 ng g−1 (concentration released by intact tubers), whereas O. insidiosus showed the highest response, between 0.01 and 0.05 ng g−1, which is close to the concentration released by P. operculella damaged tubers. Based on these results, we postulate that P. operculella and O. insidiosus have adapted their responses to plant volatiles differently, enabling them to locate suitable hosts or prey.  相似文献

Sawfly larvae of the tribe Phymatocerini (Hymenoptera: Tenthredinidae), which are specialized on toxic plants in the orders Liliales and Ranunculales, exude a droplet of deterrent hemolymph upon attack by a predator. We investigated whether secondary plant metabolites from Ranunculaceae leaves are sequestered by phymatocerine Monophadnus species, i.e., Monophadnus alpicola feeding upon Pulsatilla alpina and Monophadnus monticola feeding upon Ranunculus lanuginosus. Moreover, two undescribed Monophadnus species were studied: species A collected from Helleborus foetidus and species B collected from Helleborus viridis. Comparative high-performance liquid chromatographic–photodiode array detection–electrospray ionization–mass spectrometric analyses of plant leaf and insect hemolymph extracts revealed the presence of furostanol saponins in all samples. Larvae of species A and B actively sequestered (25R)-26-[(α-l-rhamnopyranosyl)oxy]-22α-methoxyfurost-5-en-3β-yl O-β-d-glucopyranosyl-(1→3)-O-[6-acetyl-β-d-glucopyranosyl-(1→3)]-O-β-d-glucopyranoside (compound 1). This compound occurred at a 65- to 200-fold higher concentration in the hemolymph of the two species (1.6 and 17.5 μmol/g FW, respectively) than in their host plant (0.008 and 0.268 μmol/g FW, respectively). In M. monticola, compound 1 was found at a concentration (1.2 μmol/g FW) similar to that in the host plant (1.36 μmol/g FW). The compound could not be detected consistently in M. alpicola larvae where, however, a related saponin may be present. Additional furostanol saponins were found in H. foetidus and H. viridis, but not in the two Monophadnus species feeding on them, indicating that sequestration of compound 1 is a highly specific process. In laboratory bioassays, crude hemolymph of three Monophadnus species showed a significant feeding deterrent activity against a potential predator, Myrmica rubra ant workers. Isolated furostanol saponins were also active against the ants, at a concentration range similar to that found in the hemolymph. Thus, these compounds seem to play a major role for chemical defense of Monophadnus larvae, although other plant secondary metabolites (glycosylated ecdysteroids) were also detected in their hemolymph. The oxidation processes of linoleic acid (LA) in the presence of l-ascorbic acid or saturated acyl l-ascorbate additives were measured at various temperatures and molar ratios of the additive to LA. Higher oxidative stability of LA was observed at higher additive levels for all additives. The addition of the ascorbates lengthened the induction period for the oxidation of LA. An autocatalytic kinetic rate equation was used to model the oxidation processes of LA mixed with the ascorbates, and the dependence of the rate constant, k, on acyl-chain carbon number was determined. At any temperature, the use of ascorbate additives decreased the k value for LA, and there was a slight tendency for k values to decrease with increasing acyl-chain length. The apparent activation energy, E a, and the frequency factor, k 0, for the rate constant were determined from Arrhenius plots. The calculated E a and k 0 values also decreased with increasing ascorbate acyl-chain length.  相似文献

Fatty acid amides (FAAs) are known elicitors that induce plants to release volatile compounds that, in turn, attract foraging parasitoids. Since the discovery of volicitin [N-(17-hydroxylinolenoyl)-l-glutamine] in the regurgitant of larval Spodoptera exigua, a series of related FAAs have been identified in several other species of lepidopteran caterpillars. We screened 13 non-lepidopteran insects for the presence of FAAs and found that these compounds were present in adults of two closely related cricket species, Teleogryllus taiwanemma and T. emma (Orthoptera: Gryllidae), and larvae of the fruit fly, Drosophila melanogaster (Diptera: Drosophilidae). When analyzed by liquid chromatography/mass spectrometry-ion trap-time-of-flight (LCMS-IT-TOF), the gut contents of both crickets had nearly identical FAA composition, the major FAAs comprising N-linolenoyl-l-glutamic acid and N-linoleoyl-l-glutamic acid. There were also two previously uncharacterized FAAs that were thought to be hydroxylated derivatives of these glutamic acid conjugates, based on their observed fragmentation patterns. In addition to these four FAAs containing glutamic acid, N-linolenoyl-l-glutamine and a small amount of volicitin were detected. In D. melanogaster, N-linolenoyl-l-glutamic acid and N-linoleoyl-l-glutamic acid were the major FAAs found in larval extracts, while hydroxylated glutamic acid conjugates, volicitin and N-linolenoyl-l-glutamine, were detected as trace components. Although these FAAs were not found in ten of the insects studied here, their identification in two additional orders of insects suggests that FAAs are more common than previously reported and may have physiological roles in a wide range of insects besides caterpillars.  相似文献

In this study, we synthesized the new kinds of semiconducting polymeric gels having negative (n-type) and positive (p-type) counter ions as charge carriers. The polyacrylamide gel was doped with pyranine (8-hydroxypyrene-1,3,6-trisulfonic acid, trisodium salt), having \textSO3 - {\text{SO}}_{3}^{ - } ions as side groups and Na+ as counter ions, so-called p-type semiconducting gel. The doping process was performed during the polymerization where the pyranine binds to the polymer strands over OH group chemically via radical addition. In a similar way, N-isopropylacrylamide (NIPA) gel was doped with methacrylamidopropyltrimethyl ammonium chloride (MAPTAC), having Cl as counter ions, so-called n-type semiconducting gel. Here MAPTAC was embedded by copolymerization within the polymer network (NIPA). These semiconducting gels can show different electrical properties by changing the concentration of the doping agents, swelling ratio etc. We have shown that the pn junction, formed by combining p-type and n-type gels together in close contact, rectifies the current similar to the conventional Si and Ge diodes.  相似文献

N-trimelliticimido-l-aspartic acid (1) was prepared from the reaction of trimellitic anhydride with l-aspartic acid in a mixture of glacial acetic acid and pyridine solution (3/2 ratio) under refluxing conditions. The solution polycondensation of the corresponding activated monomer with eight aromatic diamines were carried out in DMAc. The resulting poly(amide-imide)s were obtained in quantitative yields, showed admirable inherent viscosities (0.20–0.36 dl g−1), good optical activity (+7.32o to +15.24o), and were readily soluble in polar aprotic solvents. They start to decompose (T 10%) above 170 °C and display glass-transition temperatures at 120–237 °C. All of the above polymers were fully characterized by UV, FT–IR, and 1HNMR spectroscopy, elemental analysis, thermogravimetric analyses, DSC, inherent viscosity measurement, and specific rotation.  相似文献

Biodegradable star-shaped PLLA–ESO copolymers were synthesized by the bulk copolymerization of l-lactide (l-LA) and epoxidized soybean oil (ESO) with stannous octanoate as the catalyst. Effects of molar ratios of monomer to catalyst, and various amounts of ESO on copolymerization were studied. The resulting copolymers were characterized by FTIR, 1H NMR, GPC, etc., which confirmed the successful synthesis of star-shaped copolymers of l-LA and ESO. The thermal and mechanical properties of samples were also investigated by means of DSC, TGA and tensile testing. The results showed that the PLLA–ESO copolymers possed lower glass transition temperature, melting point, crystallinity, and maximum decomposition temperature than those of neat polylactide. Tensile testing demonstrated that PLLA–ESO copolymer had better ductility than linear PLLA. It was also found that the amount of catalyst almost had no influence on the weight average molecular weight of PLLA–ESO copolymers, but which could be controlled by variation of molar ratios of l-LA to ESO.  相似文献

The possibility of using xylitol, D-mannitol and D-sorbitol as Cu(II) ligands in electroless copper baths was demonstrated. The ligands mentioned showed good chelating properties for Cu(II) ions in alkaline media (pH > 11.5), i.e. under conditions of the use of traditional formaldehyde-containing solutions. Electroless copper plating solutions containing the chelators xylitol, D-mannitol and D-sorbitol are stable and, under the optimal conditions selected, copper coatings up to 3 m thick can be obtained in 1 h at ambient temperatures.  相似文献

Identification of molecular species of various N-acylated bacteriohopanehexol-mannosamides from the thermophilic bacterium Alicyclobacillus acidoterrestris by semipreparative HPLC and by RP-HPLC with ESI is described. We used triple-quadrupole type mass spectrometer, 1H and 13C NMR for analyzing this complex lipid. CD spectra of two compounds (model compound—7-deoxy-d-glycero-d-allo-heptitol obtained by stereospecific synthesis, and an isolated derivative of hopane) were also measured and the absolute configuration of both compounds was determined. On the basis of all the above methods, we identified the full structure of a new class of bacteriohopanes, represented by various N-acylated bacteriohopanehexol-mannosamides.  相似文献
