研究了米根霉的固定化方法,研制了用吸附法固定米根霉的生物转盘反应器,考察了在该反应器中培养基组织及其它操作条件对L-乳酸发酵的影响。实验结果表明,应用吸附法固定化的米根霉及生物转盘反应器进行L-乳酸发酵具有发酵速率快,L-乳酸得率高及既能用于连续发酵又能用于间歇发酵等优点。 相似文献

以聚乙烯醇(PVA)丝为吸附材料的立体载体,固定化米根霉菌种As3.3462发酵生产乳酸。与游离发酵相比,摇瓶单批固定化发酵乳酸的最终质量浓度提高了25%,发酵时间缩短了33%。分别在摇瓶和鼓泡式反应器中进行了载体的稳定性研究,摇瓶中可稳定发酵10批次,平均转化率为65%,平均产率为1.1g/(L.h);1L鼓泡式生物反应器的放大实验中进行了9批半连续发酵实验,乳酸质量浓度最高达到55.68g/L,转化率69.60%。 相似文献

经过紫外诱变获得一株来根霉菌株R8416产L-乳酸高,产酸稳定.探讨了不同氮源、通气量、培养基配方、中和剂等发酵条件对其产L-乳酸的影响。试验结果表明.该菌株最适发酵培养基组成(%):淀粉水解糖11,MgSO4、0.025.KH2PO4 0.06.ZnSO4 0.004.CaCO3 6.pH自然。8批平均产L-乳酸85.9g/L,对糖转化率80.3%,发酵周期50h,几乎不舍杂酸.L-乳酸旋光纯度与Purac公司乳酸相当。 相似文献

Low product concentration and high cost of recovery process have motivated the study of in situ removal of fumaric acid from fermentation broth. After a series of screenings, an integrated process with fumaric acid fermentation and extraction was developed. Under this integrated process, fumaric acid production and productivity increased from 18.06 g/L to 34.23 g/L, and from 0.27 g/L/h to 0.57 g/L/h, respectively. Neutralizing agent utilization decreased by 58.75%, and 140.7 g/L ammonium fumarate was obtained after back extraction, which could be directed used to produce related derivatives without concentration. 利用果胶酶处理烟梗纤维生产乳酸等对环境友好,而应用新的发酵组合方式和优化发酵条件是提高产酶量,降低成本的有效途径。本文采用实验室研发的棉布支架固定化米根霉细胞的技术,优化了底物为果胶或烟梗的发酵产果胶酶的培养条件,还优化了处理烟梗果胶的条件。底物为果胶的最佳产酶条件为:转速190r/min,装液量50mL/250mL,发酵温度30℃,初始pH值5,初始孢子浓度0.75×106个/mL。底物为烟梗的最佳产酶条件为:初始pH值4.6、初始孢子浓度0.5×106个/mL等。在优化处理烟梗果胶的条件下,固定化米根霉产果胶酶连续法比游离的果胶酶法降解烟梗果胶的效果好,果胶降解率提高18.5%,达到74.1%。棉布支架固定化米根霉利用果胶发酵产果胶酶量较高,利用烟梗发酵脱胶效果较好。 相似文献

采用添加钙离子的方法,考察钙离子对米根霉菌体形态及L-乳酸发酵合成的影响. 结果表明,添加钙离子有利于球状米根霉形成. 可溶性钙CaCl2浓度越低,越有利于形成较小直径的菌球,利于L-乳酸生成;添加较高浓度的难溶性钙CaCO3 (8.0 g/L除外)能形成较大菌球(约1.5 mm),更有利于L-乳酸合成,产量达58.6 g/L,高于CaCl2的57.2 g/L,平均生产强度为1.22 g/(L×h),高于CaCl2的1.02 g/(L×h). 但过量的CaCO3会包裹菌球,影响L-乳酸合成. 相似文献

Lipolytic enzymes are the subject of great industrial and academic interest. For this reason, a detailed study of lipolytic enzyme production by the fungus Rhizopus oryzae is tackled, and several steps from plate to shake flasks and bioreactor cultures are investigated in order to propose an optimized strategy to perform the biological process. The suitability of several lipidic compounds and surfactants is assessed. Triton X‐100 (5 g/L) gives the highest activities with a maximum value of 6320 U/L which is 10‐fold the value attained in cultures without addition of lipidic compounds. As there are almost no studies on bench‐scale bioreactors, two bioreactor configurations, stirred tank and air‐lift, are investigated to determine the most suitable one to carry out the biological reaction. It is demonstrated that the lipolytic activity is strongly enhanced when a stirred‐tank bioreactor is used with a maximum value of 3521 U/L within two days which is clearly higher than the values produced by other recently reported species. 相似文献
