目的:探讨不同质量浓度的海带多糖对人肝癌细胞Bel-7402增殖的抑制作用。方法:分别以质量浓度为5、10、20、30 mg/mL的海带多糖处理Bel-7402细胞,48 h后观察海带多糖对细胞增殖的抑制情况,Annexin V-FITC试剂盒检测细胞凋亡状况,酶联免疫吸附(enzyme-linked immunosorbent assay,ELISA)法检测Caspase-3活性,Western blotting法检测细胞内Caspase-3和甲胎球蛋白(alpha-fetoprotein,AFP)表达量。结果:海带多糖对Bel-7402细胞的增殖有明显抑制作用,最佳抑制剂量为30 mg/mL;随着海带多糖质量浓度的升高,细胞凋亡率明显增加(P<0.05),海带多糖处理组Bel-7402细胞内的Caspase-3表达水平同正常对照组细胞相比呈上升趋势,差异具有统计学意义(P<0.05);AFP蛋白表达水平呈下降趋势。结论:海带多糖对人肝癌细胞Bel-7402增殖具有抑制作用,其机制可能是激活Caspase-3,引起细胞凋亡和AFP蛋白表达水平下降所致。  相似文献

为了探讨姜黄素对LoVo细胞凋亡与凋亡相关蛋白Caspase-3和多聚腺苷酸二磷酸核糖聚合酶(PARP)表达的影响,明确其治疗大肠癌的生物学基础。用不同浓度姜黄素处理体外培养的人大肠癌LoVo细胞,采用MTT法检测姜黄素对LoVo细胞的增殖抑制作用,倒置显微镜观察姜黄素对LoVo细胞集落形成的影响;Hoechst33258染色法观察细胞凋亡;采用分光光度法检测Caspase-3酶活性;Western blotting检测姜黄素作用前后Caspase-3和PARP蛋白表达的差异。结果显示姜黄素可抑制人大肠癌LoVo细胞的生长増殖,并能抑制细胞集落的形成,降低细胞增殖生存率,激活Caspase-3的表达,呈量效和时间依赖性,20μmol/L以上浓度姜黄素能显著诱导LoVo细胞凋亡,上调Caspase-3和PARP蛋白表达。这表明姜黄素能显著抑制LoVo细胞的增殖并诱导其发生凋亡,其分子机制与调节Caspase-3和PARP的表达相关。  相似文献

为改善传统牛排煎制后汁液流失、肉质干硬、咀嚼性差的现状,本文首先通过对牛排蒸煮损失、色差、质构以及感官评价等指标的测定,来筛选洋葱汁预制牛排中洋葱汁的添加量(0、3%、6%、9%、12%、15%),然后对最佳洋葱汁添加量下预制牛排的滚揉时间(0、5、10、15、20、25 min)进行优化,最后对洋葱汁预制牛排的贮藏稳定性进行分析。结果表明,洋葱汁的添加和滚揉操作的进行均对牛排的食用品质有显著影响(p<0.05)。随着洋葱汁添加量的增加以及滚揉时间的延长,牛排的蒸煮损失、硬度和咀嚼性均呈现先下降后上升趋势,感官评分总体可接受性与之完全相反,先上升后下降。当洋葱汁添加量为9%,滚揉时间为10 min时,调理牛排的蒸煮损失达到最小值,分别为13%和14.33%;弹性达到最大值,分别为0.95、0.94 mm;此时牛排的口感以及总体可接受性评分最佳(p<0.05)。此外,添加了洋葱汁的牛排硫代巴比妥酸值(thiobarbituric acid values,TBARS)和过氧化值(peroxide values,POV)都有明显下降(p<0.05)。综合蒸煮损失、色差、质构和感官评分的结果,选择调理牛排的洋葱汁最佳添加量为9%,最佳滚揉时间为10 min。本研究结果为牛排规模化生产提供有效参考。  相似文献

目的:探索黄参提取物黄参多糖(SGP)对肝癌细胞HepG2和淋巴白血病细胞P388增殖的抑制与诱导细胞凋亡的效应,并初步探讨其对生命延长的作用。方法:应用SGP与肝癌细胞HepG2和淋巴白血病细胞P388共培养,采用MTT比色法测定不同剂量SGP对HepG2和P388细胞增殖的抑制;通过对小鼠皮下移植性肿瘤HepG2生长抑制实验,用流式细胞术分析SGP诱导细胞凋亡和细胞周期分布的变化;探讨SGP对荷淋巴白血病小鼠生命延长的作用。结果:SGP可明显抑制体外培养HepG2和P388细胞的增殖,200mg/(kg ·d)剂量作用48h,对HepG2的抑制率可达37.05%(P<0.01),而150、200mg/(kg ·d)剂量作用P388细胞48h后抑制率均超过38%(P<0.01);SGP在小鼠体内也能明显抑制HepG2移植性肿瘤生长,其抑制率超过40%;可诱导移植性肝癌HepG2细胞凋亡,并使细胞时相分布在G1期发生阻滞;对荷P388(腹水)小鼠生命有延长作用,延长率达10%。结论:SGP对小鼠有良好的抗癌变效果以及对小鼠原发性肝癌和淋巴白血病的良好抑制作用。  相似文献

目的:评价槲皮素对前列腺癌PC-3细胞凋亡和热休克蛋白27(HSP27)表达的影响,探讨槲皮素抗前列腺癌的可能作用机制。方法:体外培养人前列腺癌RWPE-1、LNCa P、PC-3和TSU-Pr1细胞株,分别随机分为4组:空白对照组;阴性对照组:给予二甲亚砜(DMSO);给药高剂量组:给予0.3mg/m L槲皮素150μL;给药低剂量组:给予0.6mg/m L槲皮素150μL。以TUNEL法检测给予槲皮素后PC-3细胞凋亡、RT-PCR检测HSP27的表达情况。结果:给药组可见大量PC-3细胞凋亡,且呈剂量依赖性,与空白对照组比较,差异具有统计学意义(P〈0.05);给药组PC-3细胞HSP27的表达明显降低,且呈剂量依赖性,与空白对照组比较,差异具有统计学意义(P〈0.05)。结论:槲皮素可诱导前列腺癌PC-3细胞凋亡和抑制HSP27的表达,可能是其抗前列腺癌的机制之一。  相似文献

本研究以人类非小细胞肺癌A549细胞系为模型,采用藻蓝蛋白处理体外培养的细胞,分别从细胞存活率、细胞形态、细胞凋亡程度以及周期分布4?个方面阐述了藻蓝蛋白对A549细胞的影响。结果表明,藻蓝蛋白能够显著降低A549细胞的存活率与生长速率(P<0.05),引起细胞形态发生非正常改变;同时上调促凋亡基因Bax、Bad,下调抑凋亡基因Bcl-2、Bcl-xl的表达而诱导细胞凋亡;此外,藻蓝蛋白能够异常调控细胞周期G1/S转换点相关基因的表达而引起细胞发生G1期阻滞。研究结果揭示了藻蓝蛋白对A549肺癌细胞具有显著的抑制作用,为开发新型抗癌类功能性食品着色剂提供了必要的理论依据。  相似文献

反相柱层析法从大豆胚轴提取单糖链大豆皂甙和双糖链大豆皂甙,研究其抗结肠癌细胞增殖的作用。MTT比色法观察大豆皂甙对结肠癌HT-29细胞增殖的影响,TUNEL法检测其对HT-29细胞凋亡的影响。结果表明,不同糖链结构的大豆皂甙均可时间和浓度依赖性地抑制结肠癌细胞增殖和诱导细胞凋亡,其中单糖链大豆皂甙的诱导细胞凋亡作用更明显。大豆皂甙可通过诱导结肠癌细胞凋亡发挥抗结肠癌作用,其抗肿瘤作用机制以及抗肿瘤作用与糖链结构之间关系需进一步的探讨  相似文献

目的:研究毛兰素对人结肠癌Caco-2细胞增殖的抑制作用及其诱导细胞凋亡的分子机制。方法:采用SRB法检测毛兰素对Caco-2细胞增殖的抑制作用,用Hoechst33342染色法观察细胞的形态学改变,流式细胞术检测细胞的凋亡率和细胞周期;蛋白免疫印迹法(Western Blot)检测细胞凋亡相关蛋白Caspase-3的表达水平。结果:与对照相比,毛兰素能抑制结肠癌细胞Caco-2的增殖,且抑制率随着药物浓度与时间增加,呈剂量时间效应,48 h半数抑制浓度IC50为0.845μg/m L;毛兰素能诱导Caco-2细胞凋亡,并诱导细胞周期阻滞于G2期;Caspase-3活性裂解片段表达增高。结论:毛兰素对肿瘤细胞的增殖具有一定抑制作用且通过线粒体途径诱导Caco-2细胞凋亡。   相似文献

以黑脉羊肚菌多糖为原料,研究羊肚菌多糖对人乳腺癌细胞MDA-MB-231(以下简称MDA)增殖和凋亡的影响。结果表明:在无细胞毒性范围内,羊肚菌多糖能显著抑制人乳腺癌细胞MDA的增殖,半数有效浓度(median effective concentration,EC50)值为0.096 mg/mL,同时人乳腺癌细胞MDA表现出多种细胞凋亡的形态学变化。免疫印迹检测实验结果显示,羊肚菌多糖能抑制B淋巴细胞瘤-2(B-cell lymphoma-2,Bcl-2)蛋白表达,促进Bcl-2相关X蛋白(Bcl-2 associated X protein,Bax)表达,同时增加Bax/Bcl-2蛋白表达比值,并呈现出剂量依赖效应。说明羊肚菌多糖能抑制人乳腺癌细胞MDA增殖,促进细胞凋亡。  相似文献

文中探讨了洋葱汁在酱油中的抑菌特性及洋葱汁代替部分苯甲酸钠用于酱油的防腐保鲜,同时,利用加速破坏性实验(accelerated shelf life testing,ASLT)预测加入复配防腐剂酱油的保质期。结果表明:40℃,添加6.25%的洋葱汁能把酱油的微生物菌落总数控制在3×10~4CFU/m L以内,且能达到1 g/kg苯甲酸钠的抑菌效果;洋葱汁与苯甲酸钠的最适复配比例为1.9%洋葱汁,0.7 g/kg苯甲酸钠;ASLT实验得到25℃条件下添加复配防腐剂酱油的保质期为544 d,添加1 g/kg苯甲酸钠酱油的保质期为547 d。  相似文献

American India Pale Ales (IPAs) with and without addition of dark/roasted malts (DRM) and dry hopping (DP) were analysed to determine whether these processes will increase total phenolic content (TP), antioxidant capacity (AA) and the levels of bioactive compounds (xanthohumol, XN and isoxanthohumol, IX). In addition, bioactivity of whole beer matrices, that is, the ‘phytochemical team approach,’ was compared to isolated compounds, the ‘silver bullet approach,’ by measuring antiproliferative and pro‐apoptotic properties using HCT 116 human colon cancer cells. DP and addition of DRM elevated the XN, IX, TP and AA. Dark malts reduced losses in XN and TP due to filtration. Xanthohumol content positively correlated with phenolic content (= 0.88, = 0.0002), indicating that the processes which increased xanthohumol content also elevated other bioactive compounds in beer. However, whole extract from IPAs were more potent in suppressing proliferation and elevating apoptosis in colon cancer cells compared with xanthohumol alone.  相似文献

Hypercholesterolemia is known to be associated with enhanced oxidative stress related to increased lipid peroxidation. Several epidemiological and clinical studies have reported the benefits of consuming fruits and vegetables, leading to reduced risks for cardiovascular diseases and several types of cancers. Flavonoids are potential antioxidants found in foods such as onions. The aim of this study was to investigate the effect of decocted onion juice (about 300 g onion) supplementation for 10 weeks on antioxidant status in mild hypercholesterolemic subjects. Twenty-seven participants were studied in a randomized, single-blind, placebo-controlled, crossover design. Indices of antioxidant status used were plasma total antioxidant capacity, antioxidant vitamins, LDL oxidation, erythrocyte antioxidant enzyme activity, and leukocyte DNA damage. After 10 weeks of consuming decocted onion juice, there were no significant differences in all the tested parameters. In conclusion we failed to find any antioxidant effect of onion supplementation in subjects with mild hypercholesterolemia.

The cellular effects of nobiletin on human pancreatic cancer cells (PANC-1) and the mechanisms by which nobiletin inhibits the proliferation of these cells were investigated. A MTT assay and flow cytometry were used to examine cell proliferation and apoptosis, respectively. A Western blot assay was used to examine expression levels of the apoptotic proteins bax, bcl-2, and p53. NOB induced apoptosis in these cells via up-regulation of the proapoptotic protein bax and down-regulation of the antiapoptotic proteins bcl-2 and p53. The normal cell cycle of PANC-1 cells was arrested by NOB with a significant increase in the proportion of G0/G1 phase cells (p<0.05) and a significant decrease in the proportion of S phase cells (p<0.05). NOB can inhibit the proliferation of human pancreatic carcinoma cells by inducing apoptosis and arresting the cell cycle progression.  相似文献

Three fruit beverages: Fb (grape–orange–apricot), FbFe (Fb + iron sulphate) and FbFeM (FbFe + skimmed milk) were subjected to in vitro gastrointestinal digestion. Addition of Fe and milk decreased the phenolic content of Fb by 13% and 55%, respectively. The digestion process decreased the phenolic content by 47%, 60%, and 70% in Fb, FbFe and FbFeM with respect to the initial, non-digested, Fb. Caco-2 cells were incubated for 4 h daily for four days or continuously for 24 h with bioaccessible fractions obtained after the digestion (digests). Polyphenols were not metabolised by Caco-2 cells. Fb digest (∼50 μM total phenolics) was the sample that led to the highest inhibition of cell proliferation under the two experimental conditions. Fb digest did not induce apoptosis but ceased cell-cycle in the S-phase which is associated to a decrease in B1 and D1 cyclin levels upon incubation for 24 h.  相似文献

Pulsatilla koreana has been used as a traditional medicine for the treatment of several diseases. The purpose of this study was to determine if SB365, Pulsatilla saponin D isolated from the root of P. koreana inhibits the progression of colon cancer. We found that SB365 strongly suppressed the growth and proliferation of colon cancer cells and induced their apoptosis. Also, SB365 showed anti-angiogenic activity by decreasing the expression of HIF-1α and VEGF. These results were confirmed by an in vivo study showing that SB365 significantly inhibited tumor growth by the induction of apoptosis and inhibition of angiogenesis with stronger anticancer activity than 5-FU. When further examined for its anticancer mechanism, SB365 effectively suppressed the AKT/mTOR pathway both in vitro and in vivo. Taken together, our study demonstrated that SB365 inhibits the AKT/mTOR pathway, leading to the suppression of tumor growth and angiogenesis together with induction of apoptosis. Therefore, SB365 is a good candidate as a natural product for use in the treatment of colon cancer.  相似文献
