为提高右旋糖酐的产率,对肠膜明串珠菌CICC-21725发酵培养基进行优化。在单因素试验的基础上,根据中心设计原理,采用响应面法确定培养基成分间交互作用及最优组成。结果表明:优化后各培养基成分为蔗糖80.0 g/L,蛋白质7.0 g/L,磷酸氢二钠1.5 g/L,磷酸二氢钾0.3 g/L,在p H7.1、25℃、150 r/min时发酵21 h,右旋糖酐产率可达92.79%,与单因素优化后的右旋糖酐产率相比,提高了7.2%。红外光谱及核磁共振分析表明,发酵产物为右旋糖酐。 相似文献

为优化紫兰草紫色素提取工艺并对其稳定性进行研究,在单因素试验基础之上,以提取温度、料液比和提取时间为影响因素,蒸馏水为提取剂,吸光度值为响应值,利用Box-Behnken中心组合法进行三因素三水平响应面分析试验;同时,研究pH、光照、蔗糖、NaCl、金属离子、氧化剂和还原剂等因素对紫色素稳定性的影响。结果表明:经响应面法优化得到最佳提取工艺条件为提取温度38℃、料液比1∶24g/mL、提取时间60min,该工艺条件所得响应值与理论值相对误差为0.28;同时,此条件下提取的紫色素具耐酸性,易受光照影响,耐受蔗糖能力较强,NaCl具增色作用,除了Cu^2+以外,对大部分金属离子较稳定;在氧化剂中稳定性较好。本研究用响应面法优化了紫兰草紫色素的提取工艺,并验证出所提取的紫色素具有良好的稳定性。 相似文献

为了提高啤酒有害菌的检出率和检测的灵敏性,采用Plackett-Burman设计法和响应面分析法(Response Surface Analysis)对CNFF-A培养基组分和配比进行优化。先用Plackett-Burman设计从7个因子中筛选出对菌落数有显著影响的因素,再用最陡爬坡试验及Box-Behnken设计进一步优化。结果表明,蜂蜜、叶酸和乙酸钠是影响菌落数的显著因素,优化后的培养基配方为:蜂蜜0.1g/L、牛肉浸粉7.0g/L、酵母浸出物5.0g/L、精氨酸1.0g/L、叶酸2.0g/L和乙酸钠1.5g/L。此条件下,培养基对啤酒有害菌的检出率大大提高,且检测时间缩短了24h。 相似文献

以紫甘薯酒为研究对象,在单因素实验的基础上,应用Box-Behnken方法进行三因素三水平实验,以液料比、发酵初始p H和发酵温度为影响因素,花色苷含量为响应值,进行响应面分析。确定最佳发酵工艺条件为液料比1.4∶1,p H4.15,发酵温度24℃。在此条件下发酵,可得酒度为12.3°,花色苷含量为123.34mg/L的紫甘薯酒,花色苷含量与其理论值124.54mg/L接近,相对误差小于0.96%。结果表明,采用响应面分析法对紫甘薯酒中的花色苷含量进行优化合理可行,可为进一步研究提供依据。 相似文献

为提高粘质沙雷氏菌(Serratia marcescens)PG12的灵菌红素产量,以K2HPO4浓度、溶氧量和发酵时间为考察因素,以灵菌红素产量为评价指标,对灵菌红素发酵工艺条件进行研究。在单因素试验基础上,通过响应面法对发酵工艺条件进行优化。结果表明,菌株产灵菌红素的最佳的发酵工艺条件为K2HPO4浓度28.45 mmol/L,溶氧量47%,发酵时间29 h。在最佳条件下,灵菌红素产量达到1 636.5 mg/L,较原始条件下的灵菌红素产量提高了13.14%。该研究可为灵菌红素发酵工艺条件的优化及生产实践提供有价值的参考。 相似文献

Optimization for purple sweet potato (Ipomoea batatas (L.) Lam) anthocyanins (PSPAs) extraction was investigated using response surface methodology in this paper. PSPAs were extracted using acid-ethanol at different extraction temperature (40-80 °C), time (60-120 min) and solid-liquid ratio (1:15-1:30). The combined effects of extraction conditions on PSPAs yield and color attributes (expressed as L*, C* and H) were studied using a three-level three-factor Box-Behnken design. The results showed that The highest yield (158 mg/100 g dw) of PSPAs were reached at the temperature 80 °C, extraction time 60 min, and solid-liquid ratio 1:32. PSPAs yield indicated a high and significant correlation with L* (P<0.05; r=−0.961) and it was significantly affected by extraction temperature (P<0.01) and solid-liquid ratio (P<0.05). 相似文献

采用响应面法对一株普鲁兰酶产生菌的产酶条件进行优化。首先,结合单因素实验,通过Plackett-Burmen试验设计,以P0.05为准,筛选出玉米淀粉、玉米浆、初始p H对普鲁兰酶产量有显著影响。根据响应面分析得到普鲁兰酶酶活最佳工艺参数为:玉米淀粉浓度3.49%,玉米浆浓度2.48%,初始p H为6.7。最后,优化后的培养基成分如下:玉米淀粉浓度3.49%,葡萄糖0.4%,玉米浆浓度2.48%,Fe SO40.1%,K2HPO4 0.1%,Mg SO4·7H2O 0.1%,初始p H为6.7,培养温度37℃,初始接种量为6%,酶活达到11.633U/m L。 相似文献

A central composite design was applied to assess some basic factors that affect profoundly the production of dextrose equivalent from banana pulp. Optimal conditions for the conversion of starch banana pulp to glucose were found to be a banana pulp concentration of 8.5%, reaction time of 90 min, and α‐amylase concentration of 0.04% for liquefaction and reaction time of 24 h with a glucoamylase concentration of 0.05% for saccharification. Only 16 and 12 experiments, respectively, were needed to assess these conditions. The adequacy of model was highly satisfactory, because the coefficients of determination (R2) were 0.93 and 0.91, respectively. The glucose syrup (GS) obtained was slightly yellowish with a hint of brown and slightly sweet. The syrup had a protein of 0.63% and an ash content of 0.60%, with a pH of 4.8. The characteristics of the GS produced met all the recommended standards, except for that of colour. 相似文献

利用响应面分析法,研究蔗糖、牛肉膏和pH值对虫草菌生长生物量的影响。结果表明:营养源的种类和浓度对虫草菌生物量有很大的影响,在所选用的蔗糖、葡萄糖、柠檬酸铵、淀粉和牛肉膏、蛋白胨、硫酸铵七种营养源中,蔗糖和牛肉膏最利于虫草菌的生长,浓度以2%左右为宜;当蔗糖的浓度为2.74%、牛肉膏浓度为1.86%、pH值为7.37时,虫草菌C.FFCC5124的生物量为10.2g/L。响应面试验的方差分析表明:一次项对响应值的影响很显著,其次为平方项,而交互项影响较为微弱。 相似文献

Response surface methodology (RSM) was used to determine the optimum experimental conditions to extract methylmercury from fish samples for GC analysis. The influence of four variables – acid concentration (3–12 M), cysteine concentration (0.5–2% w/v), solvent volume (3–9 ml) and extraction time (10–30 min) – on recovery of methylmercury was evaluated. The detection limit for methylmercury analysis using a microelectron capture detector was 7 ng g?1 in fish samples. The mean recovery under optimum conditions was 94%. Experimental data were adequately fitted into a second-order polynomial model with multiple regression coefficients (r 2) of 0.977. The four variables had a significant effect (p < 0.05) on the recovery of methylmercury from a reference material (BCR-463). Optimum conditions for methylmercury extraction were found using an acid concentration of 12.2 M, cysteine concentration of 2.4%, solvent volume of 1.5 ml and extraction time of 35 min. The validation of the developed method to analyze methylmercury in fish samples exhibited good agreement with mercury content in the samples. 相似文献

采用Plackett—Burman设计(Plackett—BurmanDesign,P-B)对影响玉米黑粉菌(Ustilago maydis)AS 5.128发酵胞外多糖的内在和外在因素进行了筛选,所选取的6个相关因素为:初始pH、培养温度、发酵时间、装液量、接种量、摇床转速。在此基础上,采用响应曲面法(Response Surface Methodology,RSM)对影响玉米黑粉菌发酵胞外多糖的关键影响因素培养温度、发酵时间和接种量的最佳水平范围作了进一步的研究,通过对二次多项回归方程求解得知,培养温度26.37℃、发酵时间7.03d和接种量9.49%时,胞外多糖产量的最大预测值为1650.92μg/mL。 相似文献
