Two experiments involving a total of 223 female and 46 male CD rats examined the influence of ovarian hormones on sensitization latencies at different periods in ontogeny and on the olfactory inhibition of maternal behavior, maternal nest building, and pup retrievals from a T-maze extension of the home cage. Ss ovariectomized before puberty failed to show a decline in sensitization latencies when tested 3.5 wks postoperatively; Ss ovariectomized during and after puberty had significantly longer latencies than sham operates when tested 8 or more weeks later. Interrupting olfaction by intranasal ZnSO? reduced latencies. Ovariectomized Ss built less compact maternal nests, and fewer retrieved pups from a –T maze than did sham operates. Estradiol benzoate prior to pup exposure significantly reduced sensitization latencies of ovariectomized Ss, increased the proportion retrieving pups from the maze, and tended to increase maternal nest ratings. Findings indicate that ovarian hormones, primarily estrogen, exert a tonic facilitatory influence on maternal responsiveness, beginning at puberty. Our research has demonstrated that the lateral habenular nucleus (Lhb) is necessary for the hormonal onset but not the postpartum maintenance of maternal behavior in the rat (K. P. Corodimas, J. S. Rosenblatt, & J. I. Morrell, 1992; K. P. Corodimas, J. S. Rosenblatt, M. E. Canfield, & J. I. Morell, 1993; T. Matthews-Felton, K. P. Corodimas, J. S. Rosenblatt, & J. I. Morell, 1995). To test the role of the Lhb in the nonhormonal onset of maternal behavior, we used the sensitization model in which the continual exposure of females to pups induces maternal behavior. Ovariectomized females received bilateral cytotoxic lesions of neurons of either the Lhb or the dorsal medial cingulate cortex-hippocampus, or they were unoperated. Maternal behavior, activity, and oromotor carrying capability were tested. Complete lesions of the neurons of the Lhb induced significant deficits in pup retrieval and nest building. Sniffing, licking, and crouching behaviors were unaltered. Activity and carrying ability were normal. These results indicate a role for the Lhb that extends to the nonhormonally dependent onset of maternal behavior, but they also indicate a more limited role than in the mediation of the hormonal onset of the behavior.  相似文献

Gave to sensitized virgins and postpartum lactating mothers, both exhibiting maternal behavior, donor litters that increased in age by 1 day, for 28 days, starting at the onset of maternal behavior. Ss were 18 Charles River rats. Each day Ss were tested for maternal behavior with 4-8 day old pups. Maternal care (i.e., nursing-crouching, retrieving, nest building, and licking and maternal withdrawal, rejection, and prevention of nursing were recorded. After the 9th day, Ss were also tested with the progressively older pups 10-28 days of age with which they were living. Virgins and lactating mothers showed generally similar patterns of maternal care although some differences were found, and they declined in maternal behavior toward the older pups in a similar manner. Maternal behavior did not decline in tests with younger pups. Results are interpreted as supporting the hypothesis that the decline as well as the maintenance of maternal behavior postpartum is nonhormonally mediated. In 16-day pregnant, hysterectomized-ovariectomized and estradiol benzoate-treated rats, cytotoxic lesions of the lateral habenula (Lhb) produced severe deficits in maternal behavior (K. P. Corodimas, J. S. Rosenblatt, M. E. Canfield, & J. I. Morrell, 1993). To determine if deficits could be found in parturient rats, females were bilaterally injected with kainic acid (KA) to produce cytotoxic lesions of the Lhb. Controls either received bilateral KA-induced lesions to the hippocampus or were unoperated. All females maintained their pregnancies, underwent parturition, and were tested for maternal behavior (pup retrieval, nursing, and nestbuilding), general activity, and oromotor carrying. Females with lesions of the Lhb had severe disruptions of all components of maternal behavior and postpartum estrus. These results extend previous findings that the Lhb is involved in the hormonal onset of maternal behavior and also demonstrate that the Lhb supports the display of postpartum estrus. The medial amygdala exerts an inhibition of maternal behavior in virgin rats, but neither the site to which it projects to exert this effect nor the neurotransmitter used in such a pathway is known. There is also evidence that the ventromedial nucleus of the hypothalamus exerts an inhibition of maternal behavior, and the medial amygdala projects to this nucleus, suggesting that it may receive a projection from the medial amygdala which is inhibitory for maternal behavior. Tachykinin injection into the hypothalamus inhibits reproductive behavior in male rats, and there is a tachykininergic projection from the medial amygdala to the ventromedial hypothalamus. Consequently, the present study was conducted to evaluate the hypothesis that the tachykinin neuropeptide K can inhibit maternal behavior after injection into the ventromedial hypothalamus. Female rats were primed to be maternal by pregnancy termination and estrogen injection. Four doses of the peptide (279 pmol, 186 pmol, 116 pmol, and 66 pmol) were bilaterally injected into the ventromedial hypothalamus, and all were effective in delaying the onset of maternal behavior. Evidence that neuropeptide K disrupts maternal behavior in animals that have already begun to be maternal is also presented. Site specificity of neuropeptide K's effect to within the region of the ventromedial hypothalamus was supported, as injection into the mediodorsal thalamus was without effect. Some possibly relevant neuroendocrine effects are addressed, as well as the possibility that tachykinins may act within the ventromedial hypothalamus to promote virgin female rats' fear of pup odors.  相似文献

Expanding on research showing that radio-frequency-induced lesions of the habenular complex disrupt the hormonal onset of maternal behavior in rats, the authors explored the importance of neurons in the lateral (Lhb) subdivision of the habenular complex for the onset of maternal behavior. On Day 12 of pregnancy, bilateral cytotoxic lesions were produced in the Lhb with kainic acid or, as a control, just dorsal in the medial hippocampus. A 3rd group had radio-frequency-induced Lhb and medial (Mhb) lesions. On Day 16 of pregnancy, Ss were hysterectomized–ovariectomized, given estradiol, and tested 48 hrs later for 10 days. Neuroanatomical tracing with fluoro-gold was then used to directly quantify the extent and location of Lhb neuron loss and to verify that Mhb neurons were intact. Cytotoxic lesions of the entire Lhb prevented the onset of all components of maternal behavior compared with controls. Ventricle cells from neonatal rats were cultured in a medium preventing cell proliferation on a modified Roller apparatus with a defined pericellular oxygen partial pressure (pO2) of 38 or 0.6 mm Hg for about one week. The cells were harvested after the second and eighth day of culture (corresponding to one or seven days of culture under a defined pO2) and prepared for electron microscopy. Electron micrographs of this material were examined by stereologic techniques. The obtained results showed some deviations of mitochondrial fine structure of heart muscle cells depending on oxygen supply. During cultivation with a pO2 of 0.6 mm Hg (hypoxic conditions) we observed a proportional increase in mitochondria without cristae and an increase in size accompanied by a more irregular shape. On the contrary, the mitochondria of cells cultured with a pericellular pO2 of 28 mm Hg, which we consider to be a normoxic condition, did not show any deviation of inner membrane arrangement, but an increase in number and a decrease of size during cultivation was apparent. The mitochondria-myofibrils ratio decreased during cultivation in both conditions of oxygen supply. The ratio between sarcoplasmatic reticulum and myofibrils decreased markedly at a pO2 of 0.6 mm Hg.  相似文献

We have determined the temporal pattern of plasma cocaine levels and increased activity that result from acute systemic injections of cocaine to female rats in two different endocrine and behavioral states, in nonmaternal virgins and in lactating maternal dams. Plasma levels of cocaine as well as ambulatory and rearing activity were determined every 30 min for a total of 300 min after subcutaneous injections of either 10, 20, or 40 mg/kg of cocaine. Virgin females had no prior drug history, whereas lactating, maternal dams had received two cocaine injections before activity testing. Within 30 min after an injection, cocaine in the plasma and activity were substantially elevated, and generally remained so for 270-300 min. Overall, plasma cocaine levels and activity were well correlated and followed a predictable dose-response pattern. The onset, peak, duration, and decline of activity corresponded generally to the onset, peak, duration, and decline of plasma cocaine. For virgins, mean ambulatory activity increased 2.5-4.0-fold over baseline, whereas in lactating females activity increased 5-11-fold over baseline. Stereotypy did not occur. Although the general responsivity of these females to cocaine was very similar to that reported for males, there are differences in the timing of peak activity and the return of activity to baseline when the virgins and the lactating dams are compared to each other and to reports by others on male rats. These data support the hypothesis that endocrine or behavioral state may influence the responsiveness of animals to cocaine.  相似文献

Three experiments examined whether the medial preoptic area (MPOA) is involved in the onset of maternal behavior in the rat. Exp I, with 54 female Charles River CD rats, investigated whether estradiol benzoate (EB) acts on the MPOA to facilitate the onset of maternal behavior in 16-day pregnant, hysterectomized, and ovariectomized Ss. When given EB implants in the MPOA, these Ss had significantly shorter latencies for the onset of maternal behavior than those implanted with cholesterol in the MPOA or with EB in the ventromedial hypothalamus, in mammillary bodies, or under the skin. Exp II, with 62 Ss, showed that estrogen-induced prolactin release was not involved in this facilitation. Exp III, with 35 Ss, showed that MPOA lesions disrupted the onset of maternal behavior induced by pup stimulation in virgin females. The experience of interacting with pups causes long-term changes in mothers' brains that mediate long-term changes in maternal behavior. As little as 1 hr of pup experience postpartum results in enhanced maternal responses to pups 10 days later. This experiment investigated the effects of lesions in multiple neural sites that have been implicated either in the actual expression of maternal behavior or in learning and memory within other behavioral contexts on the initiation and the long-term experience-based retention of maternal behavior. Electrolytic lesions were performed either before or after a 1-hr or 24-hr maternal experience. Rats sustaining lesions of the nucleus accumbens (NACC), whether administered before parturition and experience or immediately after a brief experience, failed to show a maternal experience effect. The behavioural and endocrine responses to a 10 min white noise stress have been characterized in female virgin and undisturbed lactating Sprague-Dawley rats. Animals were continuously video-taped and frequent blood samples were collected using an automated sampling system. Noise stress caused hypothalamo-pituitary-adrenal (HPA) activation, as indicated by a rapid increase in plasma corticosterone and ACTH in the virgins: corticosterone concentrations peaked 20 min after initiation of the stress before declining rapidly back to basal concentrations. In contrast, noise stress had no significant effect on either plasma corticosterone or ACTH concentrations in the lactating animals. However, 72 h after weaning the corticosterone response of the ex-lactating rats was of comparable magnitude, but longer duration to that seen in the virgins. Plasma prolactin concentrations were significantly higher in the lactating animals and declined in response to the noise whereas, a transient but reproducible increase was seen in the virgin group. In situ hybridization revealed a significantly lower basal expression of CRF mRNA in the paraventricular nucleus of lactating rats as compared to the virgins, but noise stress had no further effect. Virgin animals showed behavioural responses to the stress, including an increase in the total activity, exploratory behaviours (rearing) and displacement behaviours (grooming). Lactating animals also showed behavioural responses to the noise, but their activities were principally directed towards the pups. These data show that although lactating rats showed normal behavioural reactivity to a psychological stress they showed no statistically significant activation of the HPA axis, suggesting a dissociation of behavioural and neuroendocrine responses to this mild stress.  相似文献

Used 57 Sprague-Dawley virgin female rats to study the effects of olfactory bulbectomy under various conditions. Bilateral bulbectomy induced either cannibalism or short latency maternal behavior in naive Ss. Following unilateral bulbectomy or 2-stage bilateral bulbectomy, short latency maternal behavior appeared but cannibalism did not. Pregnancy and parturition overcame cannibalism after bilateral bulbectomy in inexperienced Ss, but not in females exhibiting cannibalism as virgins. Experienced (2 days postpartum) bulbectomized Ss had shorter latencies for reinduction of maternal behavior than experienced intact Ss. Results suggest that olfactory bulbs have 2 functions in relation to maternal behavior: sensory (olfaction) and nonsensory. Female response to olfactory bulbectomy may be due to anosmia facilitating maternal behavior, or to removal of nonsensory influences resulting in cannibalism. Carried out 3 experiments with virgin female Sprague-Dawley rats to isolate the effects of anosmia on maternal behavior from nonsensory effects of bulbectomy. Anosmia was produced peripherally by nasal infusion of zinc sulfate or centrally by lesioning the lateral olfactory tracts bilaterally. Discrimination tests verified anosmia produced by zinc sulfate. In nearly all cases anosmic Ss exhibited short latency maternal behavior when exposed to young pups. Odors from pups are viewed as delaying the onset of maternal behavior in virgins; females prevented from reacting to these odors are therefore able to receive and to respond more rapidly to those stimuli which elicit maternal behavior. Reports results of 3 experiments with a total of 92 postpartum lactating female Wistar rats. Medial preoptic area lesions severely disrupted maternal behavior, whereas lesions of the stria terminalis and medial cortico-hypothalamic tract knife cuts were without effect. Parasagittal knife cuts that severed the mediolateral connections of the preoptic-anterior hypothalamic continuum also severely disrupted maternal behavior. The lesions and knife cuts which disrupted maternal behavior had no effect on female sexual behavior. It is concluded that the medial preoptic area and its lateral connections are essential for the normal display of maternal behavior in postpartum lactating female rats. Evidence also indicates that independent neural mechanisms for the control of maternal behavior and sexual behavior exist within the hypothalamus of female rats. Pregnant rats (N?=?17) were treated either throughout gestation (Gestational Day 1–20) with 30 mg/kg per day (chronic cocaine) or with 1 15-mg/kg dose immediately following parturition (acute cocaine). Chronic and acute cocaine treatment delayed or diminished the postpartum onset of some components of maternal behavior, and chronically treated dams were significantly more aggressive toward a male intruder than acute cocaine-treated or saline-treated dams. Cocaine increased the latency to crouch over pups and decreased crouch duration during a 30-min observation period that immediately followed parturition. Latencies to nest build were also longer in more chronic cocaine-treated dams than in saline controls. On Day 6 postpartum, 83% of chronic cocaine-treated dams pinned and attacked an intruder male 8 or more times during a 10-min observation period, whereas only 4% of acute cocaine-treated and none of the saline-treated dams exhibited this much aggression. Examined the effects of word frequency and list length on the long-term serial position curve in 2 experiments, using a total of 68 undergraduates. In Exp I, the object was to find a distractor activity that would be sufficient to eliminate the recency effect in conventional free recall. In Exp II, whether list length would show a similar pattern of effects in a continuous-distractor paradigm was examined. Results demonstrate that word frequency and list length had the same effects on the serial-position curve in the continuous-distractor paradigm of delayed recall that they had previously been shown to have in immediate recall. High word frequency and shorter lists led to improved recall of preterminal items but did not influence recall of terminal items. Results suggest that the same processes underlie recency effects in the 2 paradigms and that accounts that attribute recency effects to primary (or short-term) memory are inadequate. The frequency and sequencing of aggressive behaviors by naive female hamsters has been found to change during series of brief encounters, probably because of the lack of stable dominance relations. Such initial encounters seem most representative of interactions likely in free-ranging hamsters and have been emphasized in studies of the hormonal mediation of female aggression. The present 4 experiments, conducted with a total of 72 random-bred female hamsters, found that nonestrous females exhibited intense aggression toward conspecifics of either sex. Estrous females were not aggressive and spent much time in lordosis, indicative of sexual receptivity. While oil-injected adrenalectomized-ovariectomized females fought at high levels, comparable with intact nonestrous females, the combination of 17-β-estradiol benzoate and progesterone suppressed fighting completely. In contrast, replacement of estradiol, progesterone, or testosterone propionate individually had no consistent effect. Hypophysectomized females also fought at high levels, indicating that pituitary hormones are not required for vigorous aggression. Further, individual anterior pituitary hormones did not produce marked changes in fighting. Male laboratory rats were raised in two colonies, each of 27 rats, and then given intraventricular injections of the norepinephrine neurotoxin 6-hydroxydopamine (6-OHDA) or the serotonin neurotoxin 5,6-dihydroxytryptamine (5,6-DHT) or saline. They were then returned to their enclosure and behavior during the next 50 days was observed. Shortly after neurotoxin injections the 6-OHDA rats spent more time in the burrows than controls and when out were inactive. The 5,6-DHT rats in contrast spent more time in the open than controls, ran more in activity wheels, approached humans, and fought more. Fighting, mounting, and hoarding in the colony gradually increased for 25 days; during this time the status of the 6-OHDA animals fell progressively whereas the 5,6-DHT animals increased in dominance. Social behavior returned to more normal levels after 50 days. Several successive stages of behavioral alterations occur following neurotoxin injections.  相似文献

This study investigated the effects of early-rearing experiences on responsiveness to pups and on the pattern of c-fos activation in the brain of juvenile female rats. From Days 4 to 20, littermate females were reared with their mothers (MR) or artificially (AR). AR rats received minimal licking-like tactile stimulation (AR-min) or maximal stimulation (AR-max). On Day 20, rats were exposed to pups for 4 or 8 days, exposed to a playmate for 4 or 8 days, or left in isolation for 4 or 8 days. Compared with MR rats, pup-exposed AR rats engaged in less pup licking, and all AR rats showed significant reductions in c-fos immunoreactivity in the medial preoptic area and the parietal and piriform cortices. The AR-min group showed the greatest difference in Fos-lir compared with the MR groups. Possible mechanisms that mediate the effects of rearing on the development of neural circuits underlying maternal behavior are discussed. Examined the evidence derived from 2 types of studies-those employing lesion techniques and those manipulating hormonal levels. The hormone studies suggest that prolactin, progesterone, and estrogen-and maybe another, as yet unidentified substance-play a coordinated role in the initiation of maternal behavior. Though the lesion studies are inconclusive, the evidence suggests that lesions of the hypothalamic-pituitary system, which maintains the hormonal balance, produce deficits in the motivational aspects of maternal behavior, whereas lesions of the limbic system produce deficits in the integration and organization of the behavior. It is suggested that researchers specify the deficits they find and assess the possible disruption of other behavior patterns, so that valid conclusions can be reached about functional specificity in the control of maternal behavior.
