为快速检测食品中的玉米赤霉烯酮 (Zearalenone以下简称ZEN) ,建立了抗ZEN的单克隆杂交瘤细胞株。用ZEN BSA偶联物免疫 8~ 10周龄雌性BalB c小鼠后 ,取脾细胞与小鼠骨髓瘤细胞Sp2 0融合 ,经过 4~ 5次亚克隆建立了稳定分泌抗ZEN抗体的杂交瘤细胞株 ,分别命名为Z2A4、Z8D6。将上述 2种杂交瘤细胞分别注入BalB c小鼠腹腔 ,获得含抗ZEN单克隆抗体的腹水。将腹水用饱和硫酸铵盐析法纯化 ,得到Z2A4、Z8D6单克隆抗体。经鉴定 ,其亚类为IgG1,抗体腹水效价Z2A4和Z8D6分别为 1∶1 6× 10 6和 1∶3 2× 10 6;分子量均为 15 0 0 4 0 (15 0kD) ;参考工作浓度分别为1∶4 0 0 0 0和 1∶10 0 0 0 0 ;纯化后抗体IgG含量分别为 34和 39mg ml ;抗体与其它霉菌毒素无交叉反应(交叉反应率 <1% ) ,具有较高特异性 ;抗体亲和力常数Z2A4和Z8D6分别为 5 5 2× 10 8和 4 6 8×10 9mol L。  相似文献

研究筛选到抗黄曲霉毒素M1的单克隆抗体细胞株,用AFM1-oxim e-BSA偶联物免疫Balb/C小鼠后,取小鼠脾细胞与Sp2/0骨髓瘤细胞融合,经过3次克隆和亚克隆得到两株稳定分泌抗黄曲霉毒素M1抗体的杂交瘤细胞株,经过检测两株均为IgG1亚类,并且经过鉴定其各项性质均符合标准。  相似文献

为快速检测食物中的赭曲霉素A ,建立了抗OchratoxinA的单克隆杂交瘤细胞株。用OA -匙孔嘁血蓝素 (OA KLH)偶联物免疫 8~ 10周龄雌性Balb c小鼠后 ,取脾细胞与小鼠骨髓瘤细胞系Sp2 0融合 ,经过 3~ 4次亚克隆建立了 3个稳定分泌抗赭曲霉素A抗体的杂交瘤细胞株 ,分别命名为 10G7、10G10和 5G11。将上述 3株杂交瘤细胞分别注入Balb c小鼠腹腔 ,获得含抗赭曲霉素A单克隆抗体的腹水。将腹水用饱和硫酸铵法纯化 ,得到 10G7、10G10和 5G11单克隆抗体。其中10G10、5G11单克隆抗体的Ig亚类为IgG1,10G7单克隆抗体的Ig亚类为IgG2a。抗体腹水的稀释度为 1∶6 4× 10 6~ 1∶1 3× 10 8,参考工作浓度为 1∶1 0× 10 6~ 1∶3 2× 10 7。纯化后 10G7抗体的IgG含量为 16 4g L ,亲和常数为 9× 10 -8mol L。 10G7抗体与其它结构类似物无交叉反应 ,具有较高的特异性。  相似文献

为了制备高灵敏、高特异性的司帕沙星(SPFX)单克隆抗体,鉴定免疫学特性。采用蛋白质偶联技术DCC法制备免疫抗原SPFX-BSA和包被抗原SPFX-OVA,免疫BALB/c小鼠,用间接ELISA和阻断ELISA选出产生优质多克隆抗体的的小鼠进行细胞融合,经过多次亚克隆筛选出分泌高敏感的特异性抗SPFX单克隆抗体的杂交瘤细胞株,体内诱生腹水法制备抗SPFX的单克隆抗体并对其免疫学特性进行鉴定。结果表明,间接ELISA检测显示免疫的6只小鼠效价均达1.28:10~4以上,融合后筛选出四株杂交瘤细胞株4H4-C8,4H4-C4,4H4-B1和4H4-E9;其细胞上清效价分别为1:1.4×10~3,1:8.0×10~2,1:5.0×10~2,1:6.7×10~2;4H4-C8株腹水效价为1:2.6×10~6,抗SPFX的单克隆抗体对SPFX的IC_(50)为31μg/L,且具有较好的特异性。本研究成功合成SPFX人工抗原,制备出优质的单克隆抗体,为SPFX免疫学快速检测方法的建立奠定坚实的基础。  相似文献

制备抗沙丁胺醇单克隆抗体,鉴定其特异性并建立间接竞争ELISA检测方法.以合成的沙丁胺醇完全抗原免疫Balb/c小鼠,利用杂交瘤技术进行细胞融合,筛选克隆得到稳定分泌抗沙丁胺醇抗体的单克隆杂交瘤细胞,命名为4E4和3A2.经鉴定,两个抗体亚类都为IgG,,其腹水纯化后效价分别达到2.56 ×105和1.28 ×105.以效价高且特异性较好的4E4细胞株获得的抗体建立间接竞争ELISA方法,经条件优化后得到标准曲线,检测范围为4.76~84.99ng/mL,IC50为20.12ng/mL,检测限为3.25ng/mL,与克伦特罗交叉反应率为253.62%,与其它结构类似物没有明显交叉反应.本研究制备的单克隆抗体和建立的间接ELISA方法,可用于开发同时检测沙丁胺醇和克伦特罗的试剂盒.  相似文献

通过碳化二亚胺法(EDC法)制备免疫原OTA-BSA,并免疫小鼠,经杂交瘤技术建立产抗赭曲霉毒素A(OTA)单克隆抗体的杂交瘤细胞株1株。通过体内诱生腹水法制备腹水,纯化后对其亚型、效价、灵敏性、亲和性、特异性等免疫学特性进行鉴定;单抗亚类为IgG1型,间接ELISA效价1:6.4×105,IC50为112.8pg/mL,亲和常数为9.74×1011L/mol,与赭曲霉毒素B交叉反应率为5.6%,与其他常见真菌产物交叉小于0.003%;本实验制备的抗OTA单克隆抗体具有高效、敏感、特异等特点,适合OTA的免疫学快速检测。  相似文献

为建立一种简便、快速、准确的双酚A检测方法,采用活性酯法将双酚酸与牛血清蛋白、卵清蛋白偶联制备人工抗原免疫BALB/c小鼠。选择抗血清效价和灵敏度高的BALB/c小鼠,采用聚乙二醇法制备杂交瘤细胞,获得一株可分泌双酚A单克隆抗体的杂交瘤细胞株。通过方阵滴定和反应条件优化,建立一种基于单克隆抗体的双酚A间接竞争酶联免疫分析法。该检测方法在0.5 ng/mL~50 ng/mL内有良好的线性关系,最低检测限IC10为0.5 ng/mL,半数抑制率IC50为16.65 ng/mL,水样中加标回收率为89.72%~105.25%。该单克隆抗体效价高、特异性强,该检测方法灵敏度高。  相似文献

利用细胞融合技术制备小鼠抗油质蛋白杂交瘤细胞,通过间接ELISA方法筛选稳定分泌特异性抗体的杂交瘤细胞株。获得10株杂交瘤细胞,分别命名为1B2、2D8、2F3、3A4、3D7、3F8、4A10、4F12、6A12、6E7,利用Ig亚类鉴定试剂盒鉴定各单克隆抗体的Ig亚型,除1B2、3D7、3F8为IgG1类外其余均为IgG2a类型。将杂交瘤细胞株注射入小鼠腹腔获取腹水,应用Protein A亲和层析法对腹水进行纯化。单抗效价有9株在2.0×105以上。ELISA和Western blotting分析表明这些单抗均能特异性识别芝麻过敏原油质蛋白。  相似文献

目的 建立免疫学快速检测减肥类保健食品中非法添加的酚酞, 制备酚酞单克隆抗体并进行评价。方法 利用碳二亚胺(carbodiimide, EDC)法合成免疫原和包被原, 用免疫原免疫Balb/C小鼠, 取小鼠脾脏与SP2/0鼠骨髓瘤细胞融合。采用竞争结合双阳性两步筛选法, 筛选出能分泌特异性抗体的杂交瘤细胞, 利用有限稀释亚克隆方法得到单株细胞; 采用体内诱生法制备腹水型单克隆抗体。利用辛酸-饱和硫酸铵法对腹水型抗体进行纯化, 利用酶联免疫吸附法鉴定纯化后的抗体。结果 成功合成了酚酞-BSA免疫原和酚酞-OVA包被原, 筛选获得酚酞杂交瘤细胞株FT/BSA/2019, 单克隆抗体的效价1×105。结论 本研究初步建立了特异性高的酚酞单克隆抗体的制备方法。  相似文献

Since grapevine ( Vitis spp .) rootstock material is being traded increasingly as disbudded woody material a lack of distinctive morphological features on such material necessitates an alternative and reliable means of identification. Methods described here were developed for rapid and efficient extraction of DNA from woody samples rich in phenolic compounds and polysaccharides, and for subsequent identification of varieties by RAPD PCR. Using these methods, and with the application of only one selected RAPD primer, we were able to differentiate sixteen rootstock varieties, including the seven varieties most commonly used in Germany. Problems commonly encountered with reproducibility of RAPD patterns were avoided by choosing primers with a dinucleotide sequence and a high G/C content that allowed a rather high annealing temperature of 45°C. Methods described here should also be useful for other horticultural crops, especially those with woody tissues rich in phenolic compounds and polysaccharides.  相似文献

An internet website (http://cpf.jrc.it/smt/) has been produced as a means of dissemination of methods of analysis and supporting spectroscopic information on monomers and additives used for food contact materials (principally packaging). The site which is aimed primarily at assisting food control laboratories in the European Union contains analytical information on monomers, starting substances and additives used in the manufacture of plastics materials. A searchable index is provided giving PM and CAS numbers for each of 255 substances. For each substance a data sheet gives regulatory information, chemical structures, physico-chemical information and background information on the use of the substance in particular plastics, and the food packaging applications. For monomers and starting substances (155 compounds) the infra-red and mass spectra are provided, and for additives (100 compounds); additionally proton NMR are available for about 50% of the entries. Where analytical methods have been developed for determining these substances as residual amounts in plastics or as trace amounts in food simulants these methods are also on the website. All information is provided in portable document file (PDF) format which means that high quality copies can be readily printed, using freely available Adobe Acrobat Reader software. The website will in future be maintained and up-dated by the European Commission's Joint Research Centre (JRC) as new substances are authorized for use by the European Commission (DG-ENTR formerly DGIII). Where analytical laboratories (food control or other) require reference substances these can be obtained free-ofcharge from a reference collection housed at the JRC and maintained in conjunction with this website compendium.  相似文献

The characterization of the aromatic profile of several apricot cultivars with molecular tracers in order to obtain objective data concerning the aromatic quality of this fruit was undertaken using headspace–solid phase microextraction (HS–SPME). Six apricot cultivars were selected according to their organoleptic characteristics: Iranien, Orangered, Goldrich, Hargrand, Rouge du Roussillon and A4025. The aromatic intensity of these varieties measured by HS–SPME–Olfactometry were defined and classified according to the presence and the intensity of grassy, fruity and apricot like notes. In the six varieties, 23 common volatile compounds were identified by HS–SPME–GC–MS. Finally, 10 compounds, ethyl acetate, hexyl acetate, limonene, β-cyclocitral, γ-decalactone, 6-methyl-5-hepten-2-one, linalool, β-ionone, menthone and (E)-hexen-2-al were recognized by HS–SPME–GC–O as responsible of the aromatic notes involved in apricot aroma and considered as molecular tracers of apricot aromatic quality which could be utilized to discriminate apricot varieties.  相似文献

The advent of the functional barrier concept in food packaging has brought with it a requirement for fast tests of permeation through potential barrier materials. In such tests it would be convenient for both foodstuffs and materials below the functional barrier (sub-barrier materials) to be represented by standard simulants. By means of inverse gas chromatography, liquid paraffin spiked with appropriate permeants was considered as a potential simulant of sub-barrier materials based on polypropylene (PP) or similar polyolefins. Experiments were performed to characterize the kinetics of the permeation of low molecular weight model permeants (octene, toluene and isopropanol) from liquid paraffin, through a surrogate potential functional barrier (25 μm-thick oriented PP) into the food simulants olive oil and 3% (w/v) acetic acid. These permeation results were interpreted in terms of three permeation kinetic models regarding the solubility of a particular model permeant in the post-barrier medium (i.e. the food simulant). The results obtained justify the development and evaluation of liquid sub-barrier simulants that would allow flexible yet rigorous testing of new laminated multilayer packaging materials.  相似文献

This paper describes the second part of a project undertaken to develop certified mussel reference materials for paralytic shellfish poisoning toxins. In the first part two interlaboratory studies were undertaken to investigate the performance of the analytical methodology for several PSP toxins, in particular saxitoxin and decarbamoyl-saxitoxin in lyophilized mussels, and to set criteria for the acceptance of results to be applied during the certification exercise. Fifteen laboratories participated in this certification study and were asked to measure saxitoxin and decarbamoyl-saxitoxin in rehydrated lyophilized mussel material and in a saxitoxin-enriched mussel material. The participants were allowed to use a method of their choice but with an extraction procedure to be strictly followed. The study included extra experiments to verify the detection limits for both saxitoxin and decarbamoyl-saxitoxin. Most participants (13 of 15) were able to meet all the criteria set for the certification study. Results for saxitoxin.2HCl yielded a certified mass fraction of <0.07 mg/kg in the rehydrated lyophilized mussels. Results obtained for decarbamoyl-saxitoxin.2HCl yielded a certified mass fraction of 1.59+/-0.20 mg/kg. The results for saxitoxin.2HCl in enriched blank mussel yielded a certified mass fraction of 0.48 +/- 0.06 mg/kg. These certified reference materials for paralytic shellfish poisoning toxins in lyophilized mussel material are the first available for laboratories to test their method for accuracy and performance.  相似文献
