Four experiments were conducted to determine the effects and interactions of feeding different levels of vitamins A, cholecalciferol (vitamin D3), and E on broiler chicks. In Experiment 1, chicks were fed marginal vitamin D3 (500 IU/kg) and increasing dietary levels of vitamin A (5,000, 10,000, 20,000, 40,000, 80,000, and 160,000 IU/kg). Bone ash was reduced by 10,000 IU/kg of vitamin A in the diet and at vitamin A levels above 20,000 IU/kg of diet body weight was reduced. In Experiment 2, two levels of vitamin A (1,500 and 15,000 IU/kg) and six levels of vitamin E (10, 500, 1,000, 2,500, 5,000, and 10,000 IU/kg) were added to the basal diet. High levels of vitamins A and E significantly (P < 0.001) reduced bone ash. The vitamin A x E interaction was significant (P < or = 0.05) for rickets. In Experiment 3, the same two levels of vitamin A as Experiment 2 and six levels of vitamin D3 (500, 1,000, 1,500, 2,000, 2,500, and 3,000 IU/kg) were added to the basal diet that contained 10,000 IU/kg of vitamin E. Body weight and bone ash were increased by increasing vitamin D3 with a corresponding reduction (P < or = 0.05) in rickets. In Experiment 4, three levels of vitamin A (1,500, 15,000, and 45,000 IU/kg), three levels of vitamin D3 (500, 1,500, and 2,500 IU/kg), and three levels of vitamin E (10, 5,000, and 10,000 IU/kg) were added to the basal diet. Significant negative responses (P < or = 0.05) to increasing dietary vitamin A were observed for bone ash, rickets, and plasma and liver vitamin E. A significant (P < 0.001) increase in bone ash and plasma calcium with a corresponding reduction in rickets was observed by increasing vitamin D3. Increasing dietary vitamin E adversely affected (P < or = 0.01) bone ash, plasma calcium, and plasma and liver vitamin A concentrations. Diflubenzuron (Dimilin, TH-6040; N-[[(4-chlorophenyl)-amino] carbonyl]-2,6-difluorobenzamide) was fed to male broiler and layer chickens at levels of 0, 2.5, 25, and 250 ppm from 1 day to 98 days of age. Characteristics measured were body weights, grams of feed per gram of body weight, testes weights, liver weights, comb weights, and feet weights. There were no consistent significant differences among the control, 2.5, 25, or 250 ppm groups in any of the characteristics measured. There was a trend for higher body weights in the chickens fed diets containing diflubenzuron. 相似文献

Experiments were conducted to determine the nature of the interaction of high levels of vitamin A and vitamin E-selenium nutrition in the chicken. Results showed that chicks were protected from the vitamin E-selenium deficiency disease exudative diathesis (ED) by a high dietary level of vitamin A (1.0 X 10(6) IU/kg) which moderately depressed growth. A greater concentration (1.5 X 10(6) IU/kg) of vitamin A in the diets of hens fed a low vitamin E diet hastened their depletion of plasma tocopherols and increased plasma glutathione peroxidase (GSH-px) activity. At hatching the progeny of vitamin A-fed hens were severely depleted of plasma tocopherols but had normal plasma GSH-px activities. They showed increased susceptibility to ED when fed selenium-deficient, vitamin E-free diets for 2 weeks. Absorption studies using ligated duodenal loops or oral doses indicated that high-level dietary vitamin A promoted the enteric absorption of selenium but interfered with the absorption of vitamin E. The dual nature of these effects was related to the ED-protective influence of vitamin A when fed to chicks, and the ED-stimulative influence on progeny when vitamin A was fed to dams. 相似文献

Chicken carcasses from a supermarket and from a poultry abattoir were examined using methods designed to isolate as many strains of campylobacters and related organisms as possible. Strains of arcobacter, but no campylobacters, were isolated from every carcass after enrichment. Campylobacter jejuni subsp. jejuni was isolated from all carcasses examined by direct plating and other Campylobacter-like strains were isolated from nine out of 15 abattoir carcasses by direct plating but not after enrichment. Only the Camp. jejuni subsp. jejuni strains could be identified to species level using a readily available identification scheme and/or a commercial identification kit. Examination of caecal contents from the 15 abattoir poultry yielded Camp. jejuni subsp. jejuni and Campylobacter-like strains from 15 and eight by direct plating, and from six and nine after enrichment, respectively. Four sites in the intestine of the abattoir birds (60 samples) were examined for arcobacters and only one strain was isolated. This indicates that arcobacters are probably not normal inhabitants of the poultry intestine. Poultry is a rich source of other campylobacteria besides the thermophilic Campylobacter spp. 相似文献

Experiments were conducted to determine the ability of a hydrated sodium calcium aluminosilicate (T-Bind) sorbent to reduce the toxicity of aflatoxins (AF) or T-2 toxin in male broiler chickens from day of hatch to 21 d of age. In Experiment 1, the sorbent was added at 0.250 or 0.375% to diets containing AF at 5 or T-2 toxin at 8 mg/kg of diet. When compared with controls, AF reduced BW gain by 27% and T-2 toxin reduced BW gain by 17%. The addition of the sorbent at 0.250 or 0.375%, in the absence of added mycotoxins, did not alter the performance of the chicks. The sorbent reduced the toxic effects of 5 mg AF/kg of diet on BW gain by 43% but did not significantly diminish the toxic effects of 8 mg T-2 toxin/kg of diet. The decreased efficiency of feed utilization and the increased relative organ weights caused by AF were significantly diminished to differing degrees by the sorbent. Oral lesions caused by T-2 toxin were not affected by the sorbent. In Experiment 2, the sorbent was added at 0.80% to a diet containing 8 mg T-2 toxin/kg of diet. The sorbent did not diminish the toxic effects of T-2 toxin when added at 0.80% of the diet. These data demonstrate that this specific sorbent can provide protection against the toxicity of AF, but not T-2 toxin, in young broiler chicks. 相似文献

Experiments were conducted to determine the efficacy of three inorganic sorbents, S1, S2, and S3, to reduce the toxicity of aflatoxins (AF) and T-2 toxin in male broiler chickens from day of hatch to 21 d of age. The compounds had been reported to bind to AF and T-2 toxin in vitro. S1 and S2 were the same basic compound that had been stored for different lengths of time following activation. In Experiments 1, 2, and 3, the appropriate diets were produced to contain no mycotoxins, the specific adsorbent at 0.5% of diet, AF alone at 5 mg/kg of diet, T-2 alone at 8 mg/kg of diet, AF at 5 mg/kg of diet plus the specific sorbent at 0.5% of diet, or T-2 at 8 mg/kg of diet plus the specific sorbent at 0.5% of diet. The specific sorbents used were: 1) Experiment 1, S1; 2) Experiment 2, S1 and S2; and 3) Experiment 3, S3. In Experiments 1 and 3, S1 and S3, respectively, showed no protection against AF or T-2 toxin as measured by BW gain, when compared to AF alone group. In Experiment 2, S1 showed no protection; however S2 reduced the effects of AF on BW gain by 25% as compared to AF alone diet. The data demonstrate that under the conditions of our experiment: 1) one of the sorbents provided some protection against aflatoxicosis; 2) there was variability in protection against aflatoxicosis between two different samples of the same sorbent that had been stored for different lengths of time following activation; 3) protection by the sorbents against the effects of T-2 toxin was not observed. 相似文献

This paper systematically reviews the results from epidemiologic studies investigating the hypothesis that breast cancer risk in postmenopausal women increases with increasing concentrations of estradiol in blood and with increasing urinary estrogen excretion rates. Data from 29 epidemiologic studies of endogenous hormones and postmenopausal breast cancer were used. The ratio of the average estrogen concentration in the women with breast cancer to that in the women without breast cancer (and its 95 percent confidence interval [CI]) was calculated for each study, and the results were summarized by calculating weighted averages of the log ratios. In six prospective studies of serum estradiol concentration, 329 women who subsequently developed breast cancer had, overall, a 15 percent (CI = 6-24 percent, P = 0.0003) higher mean concentration of estradiol in their blood than the 1,105 women who remained free of cancer. The results of these prospective studies did not differ significantly from each other (chi2 for heterogeneity = 8.7; degrees of freedom = 5; P > 0.1). Similar differences in mean estrogen levels were seen in the case-control studies which reported either estradiol concentrations in the blood or urinary estrogen excretion. However, the case-control studies showed significant heterogeneity among their results. The data from the prospective studies strongly suggest that breast cancer risk in postmenopausal women is associated with relatively high concentrations of endogenous estradiol. 相似文献

Cytosol binding protein for 1alpha,25-(OH)2D3 and 25-OHD3 was demonstrated in rat intestinal mucosa using sucrose density gradient ultracentrifugation analysis, Sephadex G-200 column chromatography and polyacrylamide disc gel electrophoresis. The binding protein has a sedimentation constant of 5-6S, a molecular weight of 100,000-120,000 daltons and a mobility in electrophoresis of Rf 0.42. Further analysis of binding behavior by DEAE-cellulose filter assays revealed that the bindig protein had a higher affinity for 25-OHD3 than for 1alpha,25-(OH)2D3, and that 1alpha,25-(OH)2D3 competed with 25-OHD3 for the same binding site on the protein. The apparent dissociation constant for 1alpha,25-(OH)2D3 and 25-OHD3 was 9.2 X 10(-9) M and 1.5 X 10(-9) M, respectively. The binding capacity was 3.1 pmoles/mg protein for both 1alpha,25-(OH)2D3 and 25-OHD3. The order of binding affinity was 25-OHD3 greater than 1alpha,25-(OH)2D3 greater than D3 = 1alpha-OHD3. 相似文献

The presence of Campylobacter and Salmonella on poultry meat products remains a significant public health concern. Previous research has indicated that feed withdrawal may significantly increase Salmonella contamination of broiler crops and that crop contents may serve as an important source of Salmonella carcass contamination at commercial processing. The present study evaluated the effect of preslaughter feed withdrawal on the incidence of Campylobacter isolation in crops of market-age commercial broiler chickens prior to capture and transport to the processing plant. The incidence of Campylobacter isolation from the crop was determined immediately before and after feed withdrawal in 40 7-wk-old broiler chickens obtained from each of nine separate broiler houses. Ceca were collected from broilers in six of the same flocks for comparison with the crop samples. Feed withdrawal caused a significant (P < 0.025) increase in Campylobacter-positive crop samples in seven of the nine houses sampled. Furthermore, the total number of Campylobacter-positive crops increased significantly (P < 0.001) from 90/360 (25%) before feed removal to 224/359 (62.4%) after the feed withdrawal period. Alternatively, feed withdrawal did not significantly alter the Campylobacter isolation frequency from ceca. Similar to our previous studies with Salmonella, the present results suggest that preharvest feed withdrawal increases the frequency of Campylobacter crop contamination and, thus, provides a source of Campylobacter contamination of carcasses at commercial processing. 相似文献

Biochemical changes that are associated with the growth phase of stretch-induced skeletal muscle hypertrophy are better understood than events that maintain the increased muscle mass. One purpose of this study was to determine whether changes that occur during the period of rapid muscle hypertrophy persist during periods when muscle growth plateaus or the rate of enlargement slows. Serum response factor (SRF), myogenin, MyoD, and actin mRNA expression patterns were examined. SRF protein interactions with serum response element-1 (SRE1) of the chicken skeletal alpha-actin gene were also characterized. Anterior latissimus dorsi (ALD) wet weight (132% and 122%) and total RNA concentration (29% and 19%) increased after 2 and 3 weeks of stretch overload, respectively. Myogenin mRNA per microgram RNA increased after 3 (775%), 6 (1073%), 14 (227%), and 21 days (133%) of stretch overload. At 6 days, myogenin mRNA levels were increased in the distal, middle and proximal regions of the ALD. Serum response factor (SRF) mRNA per microgram total RNA was not increased after 2 or 3 weeks of stretch overload. MyoD and skeletal alpha-actin mRNAs per microgram total RNA were also unchanged after 2 and 3 weeks of stretch. Gel mobility shift assays demonstrated that SRF bound to SRE1 from 14-day-stretched ALD nuclear extracts had an increased mobility compared to control, and this difference in mobility was maintained in nuclear extracts from ALD muscle whose mass was declining. These results indicate that the expression of myogenin mRNA and total RNA remains elevated during either slow or maintenance periods of stretch-induced increases in ALD mass, when SRF mRNA has returned to control levels. Additionally, stretch-induced alterations in SRF binding to SRE1, from the skeletal alpha-actin promoter, occur regardless of the rate of stretch-induced growth. 相似文献

The preventive action of vitamin E (Vit. E)-containing liposomes on cataractogenesis was examined in male Wistar rats (five weeks old) fed a 25% galactose diet. Vit. E-containing liposomes prepared with dipalmitoylphosphatidylcholine were instilled into both eyes three times a day over a 45-day period. Cataract appeared at 18-day galactose feeding and developed gradually thereafter. Simultaneous Vit. E-containing liposome instillation delayed this cataractogenesis. Lenses of 18-day galactose-fed rats showed decreases in Vit. E and reduced glutathione (GSH) contents and Na+, K(+)-ATPase activity and increases in lipid peroxide (LPO), galactitol, and water contents. Lenses of 45-day galactose fed rats showed decreases in GSH content and Na+,K(+)-ATPase activity and increases in Vit. E, LPO, galactitol, and water contents. Serum Vit. E and cholesterol levels decreased in 18-day galactose-fed rats, while both levels increased in 45-day galactose-fed rats. Simultaneous Vit. E-containing liposome instillation prevented these changes except for the changes of lenticular galactitol and water contents and serum Vit. E and cholesterol levels. These results indicate that simultaneously instilled Vit. E-containing liposomes can delay cataractogenesis in young adult rats fed a 25% galactose diet mainly by the antioxidative action of Vit. Extended posthatch holding (in the hatcher) has been reported to dehydrate chicks, reduce broiler performance, and depress immune response. Nevertheless, some commercial hatcheries are increasing incubation time in an attempt to minimize possible bacterial contamination of incompletely healed navels. The objectives of this study were to determine the effects of posthatch holding on broiler performance and the effects of bird density and additive stress on performance and immune response. Twelve hundred broiler eggs (58 to 70 g) were incubated. Chicks were removed from the hatcher after 528 h of incubation, banded, and weighed. Half of the chicks were returned to the hatcher for an additional 24 h (HELD). Both hatcher treatments were placed at two densities (.07 and .12 m2 per bird). Individual BW were taken at 21 and 43 d of placement and 43 d of age. The HELD chicks weighed significantly less than controls at time of placement. At 21 d postplacement the HELD broilers were significantly heavier than controls, but were similar by 43 d. Total feed conversion was not affected in the HELD treatment, but birds in the .07 m2 per bird density were less efficient in terms of total feed conversion. Chick holding time and density seemed to affect antibody titers at 5 wk. Although holding chicks in the hatcher for 24 h did not clinically dehydrate chicks or affect performance, it decreased immune response. The aim of this work was to investigate the local effects of 1:25 dihydroxychole-calciferol, one of the most potent osteoclastic activity stimulants. Tests were conducted on 32 Wistar albino rats divided into 3 groups: an experimental (appliance + drug, n = 11), control (appliance only, n = 11) and a no therapy group (n = 10) which was used for purposes of interpreting histological findings in the others. To assess the influence of age on vitamin A intestinal and liver metabolism in humans, the postprandial plasma concentrations of intestinal-originated vitamin A, i.e., retinyl esters, and liver-originated vitamin A, i.e., retinol, were compared in eight young (20-30 years old) and eight elderly (64-72 years old) healthy men. Plasma and chylomicron retinyl esters and retinol concentrations were measured for up to 24 h following the intake of a test meal that contained 23,300 RE retinyl palmitate. The chylomicron retinyl palmitate response (area under the curve) was not significantly different between the two groups, but its peak was slightly delayed (1 h) in the elderly men. The proportion of the different retinyl esters secreted in the chylomicrons was not significantly different between the two groups. The postprandial plasma retinol concentration did not change in the young participants, whereas it significantly increased in the elderly. These results suggest that vitamin A intestinal absorption and retinol intestinal esterification processes are not markedly modified in the elderly, whereas the chylomicron clearance and the regulation of postprandial plasma retinol concentration are apparently altered in these subjects. 相似文献

An experiment was conducted to determine the effect of different window coverings on broiler pigmentation. Equal numbers of male and female broilers were grown in open-type houses. The control house had open sides, while each treatment house had window coverings with either 42% or 89% light transmission properties. Birds from houses with window coverings deposited the same amount of pigment in the skin as did birds from the open house as indicated by blood xanthophyll level and excitation purity. Skin color scores, however, as well as dominant wavelengths and luminosities of shanks from these birds, indicated that pigmentation of birds grown in the house with 42% light transmission window coverings was significantly inferior to that observed for birds grown in either the open house or the house with 89% light transmission window covering. It was concluded that in situations where pigmentation is of concern, care should be taken in selecting window coverings which will not adversely affect pigmentation. 相似文献

The present experiments were undertaken to investigate the effects of environmental temperatures on growth, abdominal fat content, rate of muscle protein turnover, and heat production in tube-fed intact male broiler chickens. Plasma concentrations of thyroxine (T4), triiodothyronine (T3), and corticosterone (CTC) were also examined. Chicks (15 d old) were kept at different environmental temperatures (16, 19, 22, 25, 28, 31, and 34 degrees) and given the experimental diet (200 g crude protein/ kg, 13.57 MJ/kg metabolizable energy) by tube three times daily throughout the 12 d experimental period. In the hot conditions, except for 34 degrees, body-weight gain was significantly higher than in the cold conditions. Thus, food conversion ratios (food:gain ratios) were lower when the birds were exposed to the hot conditions other than 34 degrees. Likewise, abdominal fat content was significantly increased, and heat production was lower in the groups kept under the hot conditions other than 34 degrees. The rate of skeletal muscle protein turnover and plasma concentration of CTC were decreased when the birds were exposed to hot conditions other than 34 degrees, suggesting a role of CTC in the regulation of muscle protein turnover. Plasma concentrations of T4 and T3 were significantly decreased as environmental temperature increased. These results clearly show that plasma concentrations of thyroid hormones and CTC are associated with accelerated muscle protein turnover and heat production. 相似文献

We analyzed retrospectively the outcome of 169 patients in chronic hemodialysis (CHD), divided into four groups: 1) 24 patients with diabetic nephropathy (age 53.7 +/- 11 years); 2) 19 with polycystic kidney disease (age 55.3 +/- 9 years) 3) 43 patients older than 60 when starting chronic hemodialysis with etiologies different from diabetes and polycystic kidney disease (age 69.2 +/- 5.8 years) and 4) 83 patients younger than 60 with diverse etiologies (age 42.8 +/- 12.4 years). In groups 1, 2 and 3 serum creatinine, arterial hypertension at the beginning, morbility, mortality and its causes were registered. In group 1, the prevalence of severe diabetic retinopathy and cardiovascular disease at the beginning were also analyzed. In all groups survival was determined. Of the diabetics, 92% presented severe diabetic retinopathy and 88% cardiovascular disease. The prevalence of hypertension was 100, 74 and 67% in groups 1, 2 and 3, respectively (p = 0.13). Twelve diabetics died before the first year of treatment; there was no difference in creatinine, age, cardiovascular disease, severe retinopathy and hypertension with those who lived more than one year. The percentage of time in risk hospitalized and the days/patients/year hospitalized were significantly different between group 1 and 3 and group 2 (p < 0.001). Patients were hospitalized for similar causes in groups 1 and 3: the initiation of CHD, cardiovascular and neurological diseases. The main causes of death in groups 1 and 3 were: cardiovascular disease and sudden death at home. Survival was better in group 2 compared with group 1 (p = 0.0014) but was similar between groups 1 and 3 (p = 0.21) even though there was a difference of 15 years between them. The Cox's proportional hazard model identified as risk factors diabetes, age, year of starting chronic hemodialysis and hospitalization episodes, adjusted for covariates. The outcome of diabetic patients in chronic hemodialysis showed high morbidity and mortality and was quite similar to that of elderly patients. 相似文献
