Smooth muscle myosin II contains two 17-kDa essential light chain isoforms (LC17gi and LC17nm) of which the relative contents differ among myosins. To understand the roles of LC17 isoforms in the functions of myosin, we performed an immunofluorescence microscopic examination of their localization in primary cultured cells isolated from rat aortic smooth muscle. To identify the isoforms, rabbit polyclonal antibodies were prepared against C-terminal nonapeptides corresponding to either LC17gi or LC17nm from porcine aortic smooth muscle myosin. These isoforms differ in only 5 amino acid residues within the C-terminal 9 residues. These antibodies specifically recognize each LC17 isoform on urea-PAGE of total rat aortic cell lysates. Immediately after plating, the smooth muscle cells stained heterogeneously with each antibody, indicating differing contents of LC17 isoforms among cells. On double staining 1-2 d cultures with both antibodies, LC17nm was detected diffusely throughout the cytoplasm, whereas LC17gi was concentrated in specific regions such as the cell periphery and the base of cytoplasmic processes. These results support the suggestion that myosin containing LC17gi is essential for force-generation by aortic smooth muscle and that myosin containing LC17nm may play an important role in maintaining smooth muscle tension. 相似文献

This paper presents a method for determining the dynamic impact factors for horizontally curved composite single- or multicell box girder bridges under AASHTO truck loading. The bridges are modeled as three-dimensional structures using commercially available software. The vehicle is idealized as a pair of concentrated forces, with no mass, traveling in two circumferential paths parallel to the curved centerline of bridges. An extensive parametric study is conducted, in which over 215 curved composite box girder bridge prototypes are analyzed. The key parameters considered in this study are: Number of cells, number of lanes, degree of curvature, arc span length, slope of the outer steel webs, number and area of bracing and top chord systems, and truck(s) speed and truck(s) positioning. Based on the data generated from the parametric study, expressions for dynamic impact factors for longitudinal moment, reaction, and deflection are proposed as function of the ratio of the arc span length to the radius of curvature. The results from this study would enable bridge engineers to design horizontally curved composite box girder bridges more reliably and economically. Furthermore, the results can be used to potentially increase the live-load capacity of existing bridges to prevent posting or closing of the bridge. 相似文献

A 39-year-old male complaining of shortness of breath on mild exertion. Radiographs revealed that a giant bulla occupied more than half the area of the right lung field. Thoracoscopic excision of the giant bulla was performed using some autosutures. After the emphysematous lesion was consolidated by laser ablation, it was sutured using PDS thread. The bulla in the left lung was similarly excised 3 weeks after the first procedure. The FEV1.0% improved from 72% to 89% after excision and laser ablation of a giant bulla and bullae. Thoracoscopic excision and laser ablation of a giant bulla appears to be an effective alternative to conventional thoracotomy. 相似文献

It is now 2 years since the commercial availability of G-CSF, and thus it seems appropriate to reassess the clinical merits of this cytokine in the context of current knowledge; such an evaluation seems best founded not only on an analysis of the biology of G-CSF itself, but also on a critical assessment of whether its biologic actions translate into a cost-effective alleviation of patient suffering and improved cancer cure. As is so often the case, the knowledge base upon which clinicians must rely to make prescribing decisions is smaller than we might wish, but a substantial body of existent information can provide a guide to clinical strategy until a number of ongoing trials give more concrete information. 相似文献

Biochemical features and sensitivity to chemotherapeutic agents of 190 strains of Staphylococcus isolated from pharinx and conjunctiva of ophtalmological patients and staff were studied. Each strain was investigated for: pigment, coagulase, hemolysis (alpha-toxin), phosphatase and penicillinase production and mannite fermentation. Twentyfour chemotherapeutic angents were used for sensitivity tests. The Authors emphasize that a positive coagulase test is the best laboratory evidence for pathogenicity of a given strain of Staphylococcus, and that the site of isolation of the examined strains is not related with their biochemical features and sensitivity to chemotherapeutic agents. 相似文献
