Thirty multiparous Murciano-Granadina dairy goats milked once daily were used to study the lactational effects of an extended 24-mo kidding interval (K24; n = 14) compared with the traditional 12-mo kidding interval (K12; n = 16). Goats were divided into 2 groups at wk 29 of lactation balanced with respect to parity, milk yield, and somatic cell count. Over a period of 92 wk, K12 goats were mated twice, at wk 29 during the first lactation and at wk 79 during the second lactation, whereas K24 goats were mated once, at wk 79 of extended first lactation. The K12 goats were dried off from wk 14 to 21 of pregnancy (wk 43 to 50 of lactation). Milk yield was recorded from wk 2 to 92, and milk composition was studied from wk 29 to 92. Milk fatty acids were analyzed in milk samples taken at wk 39 (wk 10 of pregnancy) and 55 (wk 5 of subsequent lactation), when milk in udder compartments (cisternal and alveolar) was also evaluated. Average milk yield during the first 29 wk was 2.23 ± 0.13 L/d. Pregnancy reduced milk yield in K12 goats from wk 39 to 42 of lactation compared with K24 goats. During the dry period for K12 goats, milk yield of K24 goats averaged 1.53 ± 0.10 L/d. From wk 51 to 79, K12 goats produced 32% more milk than did K24 goats, but their milk contained lower fat and protein than that of K24 goats. No changes were detected for milk lactose and somatic cell count from wk 51 to 79. From wk 80 to 92, differences in milk yield and milk composition between groups were not significant. Milk of pregnant K12 goats contained higher C16:1 and conjugated C18:2 fatty acids, and had a higher desaturase index than milk of open K24 goats at wk 39. In the following lactation (wk 55), milk of K12 goats contained higher C18:2 and C18:3, and lower C16:0 fatty acids, resulting in a lower atherogenicity index compared with K24 goats. Cisternal milk at wk 39 was lower for K12 than K24 goats, whereas alveolar milk did not differ. In K12 goats, values of cisternal milk tripled, but alveolar milk only doubled at wk 55 (wk 5 of subsequent lactation) compared with wk 39, indicating the importance of the cistern in accommodating high milk yield in early lactation. Values of cisternal and alveolar milk did not differ between wk 39 and 55 for K24 goats. Fat content was higher for alveolar milk than cisternal milk for K12 goats at wk 55 and for K24 goats at wk 39 and 55. No differences in milk protein or lactose were detected between cisternal and alveolar milk. In conclusion, pregnancy reduced milk yield from wk 10 after conceiving onwards. Extended lactation did not significantly decrease milk yield (−8.2%), but increased milk components that may contribute to cheese yield, and may be a useful strategy for reducing metabolic stress in early lactation and for simplifying herd management in dairy goats.  相似文献

The objective of the experiment was to compare a silage-based control diet (C) with a nonforage diet (NF) in dairy goats throughout lactation in terms of animal performance and energy utilization. Eight Saanen goats were divided into two groups and fed C or NF, a commercial blend that included sunflower meal, cassava, coconut meal, and whole cottonseeds as the main ingredients that was characterized by a small particle size and a high crude protein content. In early, mid, and late lactation (44, 100, and 219 days in milk) the goats were individually tested for dry matter intake (DMI), digestibility, milk yield and composition, milk renneting properties, rumen and plasma parameters, and nitrogen and energy utilization (open circuit respiration chambers). During early and mid lactation, the NF fed goats had a very high DMI: 2946 and 2915 g/d, respectively. Nevertheless, milk yield was similar for the two treatments: 4369 vs. 4342 and 3882 vs. 3841 g/d for goats fed diets C and NF during the first and second periods, respectively. Milk fat content was not statistically different between the two diets. The protein content and rheological parameters of milk were similar for the two diets. Nonprotein nitrogen and urea levels in milk of goats fed NF were significantly higher than goats fed C. Ruminal ammonia and plasma urea nitrogen were also significantly increased by diet NF, due to its high protein content. Plasma glucose, beta-hydroxybutyrate, and nonesterified fatty acids and ruminal volatile fatty acids were not influenced by dietary treatment. Dietary NF significantly decreased energy digestibility (74.5 vs. 65.8%, on average for the lactation, for C and NF, respectively) and had a significantly lower metabolizability (metabolizable energy/intake energy; 66.6 vs 58.0%, on average); however, the efficiency of utilization of metabolizable energy was unaffected by the diet. In conclusion, goats were fed a nonforage diet during the entire lactation without detrimental effect on their health and productive performance.  相似文献

When four goats were subjected to total water deprivation for 48 h once in early lactation and once in mid lactation and their voluntary food intake and effects on milk secretion were observed, milk volume decreased and lactose and fat concentration increased during both periods. However, when the goats were then fed the mean of their own food intake at the two previous water deprivation experiments, and were left free access to water, it was found that decreased food consumption had only a minor influence on milk secretion. When six goats were subjected to 48 h of food deprivation and six to 48 h of water deprivation, milk secretion decreased, but the reduction was delayed and of smaller magnitude during water deprivation. During food deprivation lactose concentration decreased and the milk protein and fat increased; the fatty acid composition changed from a greater proportion of short-chain to more long-chain fatty acids. During water deprivation milk fat and lactose concentrations increased, and only after 44 h did a change from short-chain to a larger proportion of long-chain fatty acids become evident. These experiments indicate that not only food intake, but also water intake are concerned with milk production.  相似文献

Nulliparous goats were used to evaluate the effects of a standard protocol for inducing lactation with or without using a prolactin-releasing agent (reserpine). Estrus was synchronized and goats were submitted to daily s.c. injections of estradiol-17β and progesterone (0.5 and 1.25 mg/kg of body weight, respectively) for 7 d. The goats were divided into 2 groups and injected i.m. with 1 mg/d of reserpine (n = 7) or the vehicle (n = 7) on d 12, 14, 16, 18, and 20. Lactation was initiated by i.m. injections of dexamethasone (10 mg/d) from d 18 to 20. Goats were machine milked once daily from d 21 to 120, at which time they were mated with herd sires. Milk was measured and sampled daily during wk 1 of lactation and weekly thereafter. Udder traits were measured in all goats at d −2 (before the induction treatment) and on d 35 and 100 (during lactation). Goats initiated lactation on d 21 (100%) and milk yield increased thereafter. The milk yield of control and reserpine-treated goats increased as lactation advanced, peaking at wk 10 of lactation, when reserpine-treated goats yielded 1,079 ± 89 mL/d of milk compared with 850 ± 96 mL/d for control goats. Yet milk yield at the peak was only 55% of the peak milk yield observed in contemporary primiparous goats. The composition of initial milk (d 21) was different from that expected for colostrum. Milk composition stabilized after d 3 of lactation. There were no differences among groups for milk fat, protein, casein, or whey protein, but milk from control goats contained greater nonprotein nitrogen than that from reserpine-treated goats (0.48 ± 0.02 vs. 0.41 ± 0.02%). Teat length increased from 24.7 ± 1.1 to 34.5 ± 2.4 mm in control goats during mammogenesis (d −2 to 35), but stabilized in reserpine goats (25.2 ± 2.2 mm). The distance between teats (11.5 ± 0.4 cm), and the volume (922 ± 63 mL) and depth (15.6 ± 0.60 cm) of the udder increased similarly in both groups during mammogenesis and lactation. After mating, 82% of herdmates became pregnant, whereas only 21% of the lactation-induced goats conceived (1 reserpine-treated and 2 control goats). In conclusion, lactation induction was effective in nulliparous goats, but neither milk yield nor the side effects on fertility seem to support its recommendation.  相似文献

The level of gelatinases in surrounding body fluids of actively remodelling tissue is indicative of basement membrane and extracellular matrix degradation under various physiological and pathological circumstances. To elucidate the association of gelatinase with mammary tissue remodelling during gradual or acute involution, in the first trial, goats milked twice daily (lactation) and goats receiving decreased milking frequency (involution) served to provide a total of 12 milk samples and 11 mammary secretion samples, respectively. In the second trial, 6 cows served to provide samples of dry secretion in 3 consecutive weeks immediately following milk stasis. Gelatin zymography was applied for gelatinase phenotyping and quantification on milk, plasma and the degranulation medium/lysate of milk somatic cells. Results indicated that the most prevalent gelatinase subtype switched from gelatinase A in milk to gelatinase B in involution secretion. Mammary secretion of goats during involution contained marginally higher protein level, significantly lower casein ratio and greater specific capacity of gelatinase B compared with those of milk during lactation. Specific capacities of gelatinases A and B in plasma of goats were similar during lactation and involution, while gelatinase B capacity in degranulation medium/lysates based on unit number of goat somatic cell was significantly higher during involution than during lactation. Milk stasis of cows induced a significant increase in specific capacity of gelatinase B, but not gelatinase A, of dry secretion up to the third week. Results of both trials agree that regional selective accretion of gelatinase B in milk might have played a role in mammary tissue remodelling during involution induced by either decreasing milking frequency or milk stasis. It is suggested that infiltrated polymorphonuclear neutrophils are one of the potential contributors responsible for the accumulation of gelatinase B during involution.  相似文献

Resilience is the ability of an animal to cope with environmental disturbances, such as pathogens or negative energy balance. To improve resilience through breeding, we need resilience indicators. Functional longevity might be a good indicator of a dairy goat's lifetime resilience as it results from the ability to cope with and recover from all the challenges faced throughout its lifetime. The aim of this study was to validate the use of functional longevity as an indicator of resilience for selection. To address this question, we created 2 genetic lines of Alpine goats using hyperselected artificial insemination bucks with the most extreme estimated breeding values for functional longevity and the same milk yield performance. A total of 440 goats, 228 in the high longevity (high_LGV) and 221 in the low longevity (low_LGV) lines, were bred and monitored for 4 yr. Health treatments, serum IgG concentration as a proxy of passive immune transfer in early life, kidding, age, and reason of culling were systematically noted. Weight and body morphology were monitored. Weight and growth during the first year of life were similar in both goat lines. In contrast, the low_LGV goats had a lower weight during the beginning of first lactation than high_LGV goats. The milk fat-to-protein ratio was also significantly higher in low_LGV goats during first lactation. A multivariable Cox regression was fitted to the data to decipher survival at different stages of life in the 2 lines. The overall survival of high_LGV goats was significantly better than low_LGV goats (hazard ratio = 0.63, confidence interval = 0.47; 0.86) even after we included treatment, growth, serum IgG concentration at birth, and year effects in the model. The line effect was not constant over time; no significant effect was found during the first year, and the difference was observed after first kidding. This result suggested that survival at an early stage of life and during later productive life are under different genetic regulation. Altogether, this monitoring of the goat lines indicated that functional longevity-based selection helps to improve resilience by improving survival and mitigating some indicators of fat mobilization during early lactation.  相似文献

We investigated the effects of bovine somatotropin (bST) on mammary gland function and composition in the declining phase of lactation in goats. Sixteen Saanen goats, 180 +/- 11 days in milk (DIM), were divided equally into control and treated groups. The treated group received 120 mg/2 wk of slow-release bST for three cycles. Milk yield, milk composition, milk clotting measures, and plasmin-plasminogen activator activities were recorded weekly. Milk Na and K were determined in individual milk samples collected weekly during the third cycle. Blood samples were collected weekly during the second cycle and the plasma analyzed for nonesterified fatty acids (NEFA), glucose, and urea. At the end of the 6 wk, three goats from each group were slaughtered, and the udders were removed. Mammary gland weight, composition, and total DNA content were determined. The histological effects of bST on mammary tissue were investigated. The analyzed parameters included numbers of alveoli, corpora amylacea, apoptotic cells, and laminin fibronectin distribution and localization. An extensive morphological analysis on the epithelial and stromal components was performed. Milk yield was significantly higher in the treated group, fat content was not affected, but protein and nonprotein nitrogen were lower in treated goats milk. Treatment with bST did not influence milk pH but reduced coagulation time. Plasmin and plasminogen activator activities were not affected. Milk K levels were higher and the Na/K ratio was lower in treated animals. Plasma glucose, NEFA, and urea were unaffected. Mammary gland weight and total DNA were higher in treated than control animals, suggesting that with advancing lactation bST treatment maintains cells. Fat, protein, and collagen content of the mammary tissue did not differ between the groups. Treatment with bST significantly increased the number of lactating alveoli (LA) and significantly reduced the number of regressing alveoli (RA) and corpora amylacea, both within and outside the alveolar lumen. Laminin and fibronectin localization were not affected, and very few apoptotic cells were found in both treated and control samples. Our findings suggest that bST administration to dairy goats in late lactation can modulate mammary gland activity and improve lactation persistency; this is associated with maintained total mammary parenchyma weight and lactating alveoli.  相似文献

Growth factors that are present in goat milk may be responsible for its beneficial effects on the digestive system as described in ancient Chinese medical texts. To develop a nutraceutical product rich in growth factors for promoting gastrointestinal health, it is essential to collect milk with consistently high growth factor activity. Therefore, we investigated the factors affecting growth factor activity in goat milk. Among the 5 breeds of dairy goats tested, milk from Nubian goats had the highest growth factor activity. Tight-junction leakage induced by a 24-h milking interval did not increase growth factor activity in the milk. Milk collected from pregnant does had a significantly higher growth factor activity than milk collected postpartum. Growth factor activity decreased during the first 8 wk of lactation, fluctuated thereafter, and then increased dramatically after natural mating. During wk 1 to 8, growth factor activity was inversely correlated with milk yield and week of lactation. No correlation was observed during wk 9 to 29. After natural mating of the goats, the growth factor activity in the milk correlated significantly with somatic cell count and conductivity (a measure of membrane permeability), and correlated inversely with milk yield. Based on the above data, goat milk with higher growth factor activity could be selectively collected from Nubian pregnant does.  相似文献

Variations in mammary glucose uptake were measured during the normal pregnancy-lactation cycle in dairy goats. In addition mammary glucose uptake was studied in response to somatotropin (ST) treatment in mid-lactation and acute increases in glucose concentration induced by sodium-propionate challenge in early lactation. Mammary glucose uptake was independent of arterial glucose, insulin and Insulin-like Growth Factor-1 (IGF-1) concentrations during lactation and during acute increases in arterial glucose concentration. Glucose uptake in the lactating mammary gland of the goat must therefore be carried out by an insulin-independent carrier, possible GLUT1, and glucose supply is not a limiting factor for uptake under in vivo conditions. Extraction of glucose uptake changed markedly during the normal course of lactation, following the overall changes in milk yield. Concentrations of glucose in skimmed milk, believed to reflect intracellular glucose concentration, changed in opposite directions, resulting in decreasing ratios of arterial: skimmed milk glucose concentration with progressing lactation. Thus, mammary synthetic capacity also involves a capacity for glucose uptake, which may be influenced by variations in glucose carrier numbers, as well as mammary metabolic activity (intracellular glucose concentration). In contrast to the situation during the normal course of lactation, ST stimulated milk yield, despite less efficient glucose extraction.  相似文献

In a recent study, we observed that starch-rich diets used in mid lactation induced lower milk production persistency and higher body fat accumulation in dairy ewes compared with dairy goats. Because these species differences could be linked to hormonal mechanisms that drive energy partitioning, in the same experiment, we explored the evolution of metabolic and hormonal status during lactation to test this hypothesis. Twenty mature Sarda dairy ewes and 20 mature Saanen goats [15–134 ± 11 d in milk (DIM), mean ± SD] were compared simultaneously. In early lactation, each species was allocated to one dietary treatment: high-starch diet [HS: 20.4% starch, on dry matter (DM) basis], whereas from 92 ± 11 DIM, each species was allocated to 1 of 2 dietary treatments: HS (20.0% starch, on DM basis) and low-starch (LS: 7.8% starch, on DM basis) diets. Blood samples were collected in the morning to analyze glucose, nonesterified fatty acids (NEFA), growth hormone (GH), insulin, and insulin-like growth factor I (IGF-I). Data were analyzed using the PROC MIXED procedure of SAS with repeated measurements (SAS Version 9.0). The HS and LS diets applied in mid lactation did not affect metabolic status of the animal within species; thus, only a comparison between species was carried out. From early to mid lactation, plasma glucose concentration was higher in ewes than in goats (54.57 vs. 48.35 ± 1.18 mg/dL), whereas plasma NEFA concentration was greater in goats than in ewes (0.31 vs. 0.25 ± 0.03 mmol/L). Goats had higher plasma GH concentration and lower plasma insulin content than ewes (4.78 vs. 1.31 ng/mL ± 0.47; 0.11 vs. 0.26 μg/L ± 0.02). Plasma IGF-I concentration did not vary between species. The comparison of metabolic and hormonal status of lactating Sarda dairy ewes and Saanen goats, carried out by studying simultaneously the 2 species in the same stage of lactation and experimental conditions, suggests that the higher insulin and glucose concentration observed in Sarda ewes explains why they partitioned more energy toward body reserves than to the mammary gland, especially in mid lactation. This can justify the negative effect of high-starch diets in mid-lactating Sarda ewes. Conversely, the highest GH and NEFA concentration observed in Saanen goats explain why they partitioned more energy of starch diets toward the mammary gland than to body reserves and justify the positive effect of high-starch diet in mid lactation. Together, these different responses contribute to explain why specialized dairy goats, such as the Saanen breed, have a higher milk production persistency than specialized dairy sheep breeds, such as the Sarda.  相似文献

Lactoperoxidase and thiocyanate content in goat milk from two Spanish breeds throughout 5 mo of lactation has been investigated. Mean lactoperoxidase for milk from Verata and Murciano-Granadina goats was .95 and 2.15 units/ml, respectively. Very low concentrations, .03 units/ml for Verata and .20 units/ml for Murciano-Granadina, were found for the first 24 h after kidding. Highest concentrations were detected at the end of lactation (135 to 150 d) for Verata and in midlactation (60 to 75 d) for Murciano-Granadina goats. Mean thiocyanate content was 5.76 ppm for Verata goats, without a distinct maximum throughout lactation, and 3.20 ppm for Murciano-Granadina goats, with maximum levels at final lactation stages. Activation of the lactoperoxidase system might be a useful procedure in preserving raw goat milk quality by addition of low levels of thiocyanate and hydrogen peroxide.  相似文献

The effects of once (1X) vs. twice (2X) daily milking throughout lactation on milk yield, milk composition, somatic cell count (SCC), and udder health were studied in 32 Murciano-Granadina dairy goats. Goats were assigned at wk 2 of lactation to two treatment groups; once daily milking at 0900 (1X, n = 17), or twice daily milking at 0900 and 1700 (2X, n = 15). Milk yield was recorded weekly until wk 28, and milk composition and SCC were evaluated for each individual udder half at each milking at wk 2 and 4 of lactation and then, monthly until the end of the experiment. Once daily milking resulted in an 18% reduction in the yield of 4% fat-corrected milk compared to twice daily milking (1.61 vs. 1.95 L/d, respectively). This reduction was more marked from wk 2 to 12 than in mid and late lactation. Response to milking frequency also varied according to parity number where goats of less than four parities suffered more milk yield losses during 1X than older goats. Milk of 1X goats contained higher percentages of total solids (13.6 vs. 12.9%), fat (5.10 vs. 4.62%) and casein (2.57 vs. 2.35%) than milk of 2X goats, but milk protein percentage did not differ between treatments (3.28 vs. 3.20%). Yields of total solids, fat, protein and casein tended to be higher for 2X than 1X. Milk SCC did not differ between treatments. We conclude that application of once daily milking in Murciano-Granadina dairy goats moderately reduced milk yield without negative effects on milk composition and udder health. Losses in milk yield would be reduced if 1X is practiced during mid- or late lactation and in older goats. An increase in labor productivity and a higher farmer's standard of living is also expected.  相似文献

Two experiments were conducted to determine the milk loss of high-yielding Alpine goats resulting from once-daily milking (ODM) and its relationship to udder cisternal size. We investigated the effects of application of this management strategy on milk yield, composition, and technological parameters: lipolysis, fat globule size, and cheese yield. In a second experiment, we investigated the effect of repeated periods of ODM management during lactation. Goats at the beginning of both experiments were at 25 d in milk on average and were previously milked twice daily (twice-daily milking; TDM). In experiment 1, which was conducted for 2 periods (P) of 9 wk (P1, P2), 48 goats were grouped (1, 2, 3, and 4) according to milk yield, parity, and somatic cell count (SCC). Over the 2 periods, goats from group 1 were managed with TDM and those from group 2 were managed with ODM. In group 3, goats were assigned to TDM during P1 and ODM during P2, conversely, those in group 4 were assigned to ODM in P1 and TDM in P2. During P1, the 12 goats from group 3 underwent 2 distinct morning machine milkings to measure milk repartition (cisternal and alveolar) in the udder based on the “atosiban method.” On P1 plus the P2 period of 18 wk, milk loss caused by ODM (compared with TDM) was 16%. In our condition of 24-h milk accumulation, there was no correlation between milk loss and udder cisternal size. Milk fat content, fat globule size, or apparent laboratory cheese yield was not modified by ODM, but milk protein content (+2.7 g/kg), casein (+1.8 g/kg), milk soluble protein concentration (+1.0 g/kg), and SCC increased, whereas lipolysis decreased (−0.3 mEq/100 g of oleic acid). In experiment 2, which was conducted for 4 periods (P1, P2, P3, P4) of 5 wk each, 8 goats, blocked into 2 homogenous groups (5 and 6), were used to study the effects of a double inversion of milking frequency (TDM or ODM) for 20 wk of lactation. Milk loss was 17% and ODM did not modify milk fat or protein contents, SCC, casein, or milk soluble protein concentration, but lipolysis was decreased (−0.3 mEq/100 g of oleic acid). Neither experiment showed the effects of period of ODM management on milk yield, milk fat or protein content, SCC, fat globule size, lipolysis, casein, milk soluble protein concentration, or apparent laboratory cheese yield.  相似文献

Nine West African dwarf (Fouta djallon), Red Sokoto (Maradi) and Saanen lactating goats, hand-milked, were used for these studies, which lasted 12 weeks. The Saanen goat's milk was obtained weekly from the Western State Ministry of Agriculture and Natural Resources Urban Dairy Farm at Iwo Road, while similar samples were collected from the herd at the University of Ibadan Teaching and Research Farm. The goats were balanced for stage of lactation, namely early, mid- and late-lactation. They were each maintained on giant star grass (Cynodon nlemfuensis) ad lib. and 1 kg dairy concentrate mixture daily. The mean contents, with standard deviations, of the milks of Saanen, West African dwarf and Red Sokoto goats at mid-lactation respectively were: (in percentages), total solids 12-25 plus or minus 0-078, 18-18 plus or minus 0-392, 15-85 plus or minus 0-077; solids-not-fat (SNF) 8-91 plus or minus 0-42, 10-48 plus or minus 0-471, 10-53 plus or minus 0-140; butterfat 3-34 plus or minus 0-141, 7-78 plus or minus 0-621, 5-32 plus or minus 0-098; protein 3-04 plus or minus 0-139, 5-30 plus or minus 0-292, 4-74 plus or minus 0-021; lactose 4-56 plus or minus 0-167, 5-19 plus or minus 0-176, 4-77 plus or minus 0-053; energy (kj/g dried milk), 21-08 plus or minus 1-11, 25-51 plus or minus 1-74, 22-17 plus or minus 0-51. Breed differences were significant (P less than 0-01). The milk of West African dwarf goats contained more of these milk components than the milk of the Red Sokoto or Saanen goats. The Red Sokoto goats gave milk of higher content than did Saanen goats, even at an early stage of lactation. Butterfat, protein, lactose and energy values were significantly affected by stages of lactation (P less than 0-05) and tended to rise with advancing lactation; this was particularly so with West African dwarf goats.  相似文献

Seventeen pregnant multiparous Murciano-Granadina dairy goats, kept in a semi-intensive exploitation system with once daily milking throughout lactation and 1 kidding per year (milk yield, 577 L/300 d), were used to study the effects of dry-off period length on performance during the subsequent lactation. Goats were mated at wk 29 of lactation and were assigned to 2 experimental groups according to dry-off treatment: goats that were dried off 56 d before expected kidding (D56; n = 9) and goats without dry-off (D0; n = 8). After parturition, kids were removed from their mothers and weighed before suckling. Goats were hand milked to obtain colostrum and were machine milked thereafter. Colostrum was sampled for composition and IgG analysis. Milk yield was recorded weekly during the preceding and subsequent lactations. Udders were biopsied in a sample of goats at d −65 (late lactation), d −49 (during dry-off), and d 48 (early lactation) to kidding (d 0). Apoptotic and proliferating cells in mammary tissues were detected immunohistochemically. Five goats (63%) in the D0 group dried off spontaneously at 27 ± 4 d before kidding and were considered separately (D27). The rest of the D0 goats yielded 0.86 L/d from d −56 to kidding. Goats kidded 2.25 kids/goat, but the D0 kids had smaller birth weights (1.7 kg) than the D27 (2.2 kg) and D56 (2.1 kg) kids. Colostrum of the D0 goats contained less IgG (5.6 mg/mL) than the D27 (32.9 mg/mL) and the D56 (42.4 mg/mL) goats. In the subsequent lactation (210 d), the D0 goats produced less milk (1.78 L/d) than the D27 (2.51 L/d) and D56 (2.24 L/d) goats, with no detectable difference between the D27 and D56 goats. Apoptosis and proliferation indices increased from 0.51 and 2.09%, at d −65, to 1.75 and 7.12% at d −49 (d 7 of dry-off) in D56 goats. Despite differences in daily milk yield during early lactation (d 48) between the D0, D27, and D56 treatments (1.73, 2.68, and 2.53 L/d, respectively), no differences in apoptosis or proliferation indices were detected (D0: 0.65 and 2.48%; D27: 0.68 and 1.37%; and D56: 0.71 and 2.95%), indicating that duration of the dry period did not affect mammary cell turnover during the subsequent lactation. Omitting the dry period between lactations reduced the quality of colostrum and had negative effects on milk yield in dairy goats. Goats dried off spontaneously for 27 d were as productive as goats dried off for 56 d, indicating that less than 2 mo of dry-off may be sufficient in practice.  相似文献

Milk fat globule (MFG) size and phospholipids (PL) content and composition were determined in milk collected at 65 (pretreatment), 110, 135 and 170 days of lactation from goats randomly assigned to grazing in Mediterranean brushland or fed clover hay indoors, in addition to concentrate. Daily feed intake and dietary contents of neutral detergent fibre and acid detergent fibre were higher in grazing goats, associated with milk richer in fat, with larger MFGs and 20% higher PL content. Smaller MFGs, produced by all confinement groups, was associated with 15 μg g−1 fat higher milk PL content. The greatest effect was found in the Damascus goats, with over 44% higher PL concentration, on milk fat basis, in the confined compared with grazing group. Our understanding of how PL content is modulated by the interaction between genetic background and nutrition will enable to achieve either PL-rich milk or PL-enriched milk fat.  相似文献

The chemical composition of Norwegian bulk collected goats' milk from the University herd was analysed during one lactation period (30 weeks, 20 samples during 1983). There was considerable variation in chemical composition during the year. Fat content decreased over the first 4 months of lactation and increased during the mountain pasture period. Protein concentration decreased during the first 4 months, and then increased until the end of lactation. Lactose concentration decreased throughout lactation. Casein nitrogen (casein N) was highest at mid lactation and lowest at the beginning and end of lactation. beta-Lactoglobulin N showed the opposite trend. Citrate content showed a significantly quadratic decrease and total ash content an increase with advancing lactation. Mutual significant correlations between total P, K, Na, Ca and Mg were calculated, and all increased throughout lactation. There was significant positive correlation between concentrations of individual medium-chain fatty acids and stage of lactation. They remained more or less constant during the first part of the lactation, decreased to minima when the goats were on pasture, and increased during the last phase of lactation. Concentration of C16 fatty acid was negatively correlated with C18 and C18:1. Goat flavour intensity score and quality flavour score were highest at mid lactation, and positively correlated with the acid degree value.  相似文献

The capability of goats to maintain milk production during water deprivation is remarkable and not yet fully understood. The aim of the present study was to investigate whether intravenous infusions of hypertonic NaCl cause release of both vasopressin and oxytocin and whether the peptides, in combination with the hyperosmolality, affect milk flow and milk composition. Six Swedish domestic landrace goats in their first to third lactation were milked every 30 min during experiments. Hypertonic NaCl (HNaCl) or isotonic NaCl (IsoNaCl) were infused for 90 min. Goats were not allowed to drink during infusions. Plasma vasopressin concentration increased during HNaCl infusions, and did not change in response to IsoNaCl infusions. Plasma oxytocin concentration did not change during either infusion. Milk flow was maintained during the infusions. Milk fat concentration decreased in the three samples taken before onset of the infusions, but then increased gradually during HNaCl infusions, while it continued to fall during the IsoNaCl infusions. Milk osmolality followed the rise in plasma osmolality during the HNaCl infusions and did not change in IsoNaCl experiments. Milk lactose concentration increased throughout both series of experiments, the concentration being higher during HNaCl infusions. Milk protein concentration did not change during HNaCl infusions, but fell in the IsoNaCl experiments. It is concluded that the hyperosmolality in combination with elevated plasma vasopressin levels did not disturb the secretory activity of the mammary cells, but rather facilitated emptying of the alveolar milk. Such a mechanism may help to explain the sustained milk production in water deprived goats.  相似文献

Milk yield is greatly influenced by the size of the mammary secretory cell population. In rats, proliferation of cells during early lactation may account for as much as three-quarters of the increase in daily milk yield between parturition and peak lactation; the remainder is due to increased activity of existing cells. Conversely, in goats, all the initial decline in milk yield after peak lactation can be attributed to loss of secretory cells. Increased frequency of milking enhances milk yield and reduces secretory cell loss, whereas goats hemimastectomized at peak lactation undergo compensatory changes in the remaining gland, which include a complete maintenance of cell number for at least 18 wk. Cell proliferation is increased in both cases, showing that mammary growth can occur during established lactation. The significance of this observation is discussed in relation to ways in which the milk yield of dairy animals might be increased.  相似文献

In ruminants, milk yield can be affected by treatment with growth hormone (rbGH) and/or changes in frequency of milking. Frequent milkings encourage the maintenance of lactation, whereas infrequent milkings result in mammary involution. Our objective was to evaluate the influence of rbGH treatment and milking frequency on mammary gland morphology and milk composition. After adaptation to twice-daily milkings, six Saanen goats in late lactation were milked once daily from one udder-half and thrice-daily from the other udder-half. Concurrently, three of the six goats received daily injections of rbGH. After 23 d of treatment, milking frequency significantly affected milk yield (+8% vs. -26% for thrice- vs. once-daily milking). Additionally, treatments of rbGH increased milk yield from thrice-daily milked udder-halves (+19%), but failed to abate the reduction in milk yield from once-daily milked udder-halves (-31%). Mammary glands were heavier in the frequently milked udder-halves and in GH-treated goats. Based on histological and DNA analysis of mammary tissues, it was determined that milking frequency clearly affected epithelial cell numbers and alveolar diameter, whereas rbGH induced a potential cell hypertrophy and only a tendency to increase and/or maintain the mammary cell number. RNA concentration and kappa casein gene expression were not affected by treatments. In udder-halves milked once-daily, low casein:whey protein ratios, high Na+:K+ ratios, and high somatic cell counts (SCC) were indicative of changes in epithelial permeability, which rbGH treatment facilitated. The present data suggest that milking frequency and exogenous treatments of rbGH use different cellular mechanisms to influence mammary gland morphology and milk production.  相似文献
