Seventeen fruits from Ecuador were analysed for total soluble phenolic compounds content and for antioxidant capacity, using three different methods (DPPH, FRAP and ABTS+). For the total phenolic content measured by the Folin–Ciocalteu method, three groups, having <100, 200–500 and >1000 mg GAE/100 g FW, were clearly distinguishable. Andean blackberry, capulí cherry peel and banana passion fruit were classified in the third group, with concentrations of 2167, 1494 and 1010 mg of GAE/100 g FW, respectively. Antioxidant capacity analyses revealed the same classes. FRAP and ABTS+ gave comparable results and were highly correlated (y = 0.691x + 6.78; r2 = 0.908). Spectrophotometric measurements showed that the Andean blackberry and capulí peel but not banana passion fruit contained high levels of anthocyanins (λmax = 520 nm).  相似文献

Phenolic compound profile and antioxidant activity of methanol, acetone, ethyl acetate, water and petroleum ether (b.p. 40–60 °C) extracts of Dorystoechas hastata, endemic to Turkey and being consumed as herbal tea by local inhabitants, have been investigated. HPLC–DAD analysis indicated the presence of chlorogenic, caffeic, p-coumaric, ferulic and rosmarinic acids as phenolic acids, quercetin, kaempferol and apigenin as flavonoids and carnosic acid and carnosol as diterpenoid antioxidants in the plant. Petroleum ether, methanol and water extracts of D. hastata were found to be very effective antioxidative extracts. Petroleum ether extract, having the highest amount of carnosic acid and carnosol contents (531.3 and 389.9 mg/g DW, respectively) among all solvent extracts, was found to have the highest antioxidative potential based on the individual antioxidant activity assays; DPPH, ABTS+, TBARS and total phenolic content, expressed as IC50, TEAC, EC50 and TPC values, respectively, and also on the principal component analysis. It exhibited significantly high TEAC (7.1 mM trolox) and low EC50 (54.5 μg/mL) values, indicating the strong potential in in vitro radical scavenging and in inhibiting lipid oxidation. Water extract, with its extremely low IC50 value of 4.9 μg/mL in DPPH radical scavenging and significant TEAC (4.8 mM trolox), EC50 (64.4 μg/mL) and TPC (116.7 mg GAE/g DW) values, was found to be the second highest in antioxidative potential among all extracts. TPC value of methanol extract (147.3 mg GAE/g DW) was found to be significantly higher than the other extracts studied. The results showed that D. hastata can be used as a potential antioxidative edible source due to its different classes of phenolic compounds and strong antioxidative capacity.  相似文献

Fermentation and roasting are the main causes of polyphenol degradation during the process for obtaining cocoa products. In the present study, a process for obtaining polyphenol-rich cocoa products on an industrial scale is described. The process avoids the fermentation and roasting steps and includes a step for the inactivation of the enzyme Polyphenol Oxidase (PPO), which helps preserve the polyphenol content present in the raw cocoa bean. In addition, our study evaluates the antioxidant capacity and characterizes the flavonoid profile of the polyphenol-rich cocoa products obtained from the natural polyphenol-rich cocoa cake. Using different protocols, we have obtained three cocoa extracts with high polyphenol content, namely extracts A (167 mg/g), B (374 mg/g) and C (787 mg/g). The scavenging capacity of the extracts was measured as their ability to bleach the stable radicals DPPH and ABTS+ while their antioxidant effect was evaluated with the FRAP assay. The results for A, B and C in the DPPH test expressed as Trolox equivalent (μmol)/mg dry weight of extract were 0.2, 1.4 and 3.0, respectively; in the ABTS test the results were 1.0, 4.7 and 9.8. The antioxidant capacity expressed as ascorbic acid equivalent (μmol)/mg dry weight of each product were 17.2, 76.1 and 207.7, respectively. The scavenging properties of cocoa powder against the superoxide anion, H2O2, HClO, and peroxynitrite were also determined. The IC50 (μg/mL) values in the hypoxanthine/xanthine oxidase test were 77.5, 12.3 and 10.3, for A, B and C, respectively, while as an HOCl scavenger the IC50 (μg/mL) values were 225.4, 73.2 and 21.5. As a peroxynitrite anion scavenger, only extract C had a relevant effect, with IC50 (μg/mL) values of 76.1 or 110.0 in the absence or presence of bicarbonate. None of the extracts tested showed activity in the hydrogen peroxide test, but B and C significantly increased the deoxyribose degradation in the absence of ascorbate. Likewise, none of the extracts inhibited the ferrous or copper chelating activity at 100 μg/mL, but they inhibited the lipid peroxidation in brain homogenates and human plasma through non-enzymatic generation systems, with extract C giving the best IC50 (μg/mL) values: 17.4 and 8.1 against lipid peroxidation in brain homogenates and human plasma, respectively. In conclusion, if the extractive protocol is well characterized, defined and optimized, cocoa could constitute a source of polyphenols for enriching foods, nutraceuticals and alimentary supplements.  相似文献

The methanolic extracts of 16 Helichrysum species were investigated for their in vitro antioxidant, radical scavenging and antimicrobial activities. All the extracts showed strong antioxidant and radical scavenging activity. The highest total antioxidant capacity as ascorbic acid equivalents (AAE) of 194.64 mg/g dry extract was obtained for Helichrysum noeanum in the phosphomolybdenum assay. The highest IC50 value (7.95 μg/ml) was observed for the extract of Helichrysum stoechas subsp. barellieri in the DPPH assay. The total phenolic contents of the extracts ranged from 66.74 to 160.63 mg gallic acid equivalents (GAE)/g dry extract. The major component present in the extracts was identified as chlorogenic acid followed by apigenin-7-glucoside and apigenin by HPLC analysis. All the extracts showed significant antimicrobial activity against microorganisms containing 13 bacteria and two yeasts in the agar diffusion method.  相似文献

The aim of this study was to determine the antioxidant potency and total phenolic and flavonoid contents of kiwifruit (Actinidia deliciosa) in vitro by analysing the radical scavenging activity of lyophilised water extract from kiwifruit (LEK) for 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS), 1,1-diphenyl-2-picryl-hydrazyl (DPPH), N,N-dimethyl-p-phenylenediamine (DMPD), and superoxide anion radical (O2) as well as the total reducing power by FRAP and CUPRAC assays and the metal chelating activities. LEK showed efficient radical scavenging activity with DPPH, ABTS, DMPD, and O2 radicals; ferric (Fe3+) and cupric (Cu2+) ion reducing power and metal chelating activities. Moreover, the amounts of phenolic compounds, such as caffeic acid, ferulic acid, syringic acid, ellagic acid, catechol, pyrogallol, p-hydroxy benzoic acid, vanillin, p-coumaric acid, gallic acid, quercetin, ??-tocopherol and ascorbic acid, in LEK were quantified by LC-MS-MS. The results show that pyrogallol (2070.0 mg/kg LEK) is the main phenolic compound responsible for the antioxidant and radical scavenging activities of LEK. Finally, total phenolic and flavonoid contents were determined as gallic acid (GAE) and quercetin equivalents (QE). The GAE and QE values in LEK were 16.67 ± 2.83 ??g GAE/mg and 12.95 ± 0.52 ??g QE/mg, respectively. The results suggest that consumption of kiwifruit (A. deliciosa) can be beneficial effects due to its antioxidant properties.  相似文献

Solid–liquid extraction and response surface methodology were used to optimise conditions for the antioxidant activity of grape cane extracts. The independent processing variables were ethanol concentration, temperature and solvent to solid ratio. Ethanol concentration and temperature significantly affected antioxidant activity measured by the Trolox equivalent antioxidant capacity (TEAC) assay and the oxygen radical absorbance capacity using fluorescein (ORACFL) method (p  0.01), whereas the solvent to solid ratio did not significantly affect the activity (p > 0.05). Antioxidant activity of the extracts, determined by the TEAC assay, varied from 85.6 to 238.6 μmol Trolox equivalents/g of dry sample. ORACFL values ranged from 308.4 to 1302.7 μmol Trolox equivalents/g of dry sample. Ethanol concentrations of 40.4% and 55.4% were optimal for the highest antioxidant activities measured by the TEAC assay and the ORACFL method, respectively. The optimal temperature was 83.6 °C. Antioxidant activity correlates with total phenolic content of extracts.  相似文献

A total of 38 commonly consumed vegetable samples, including 3 kale, 1 rhubarb, 3 spinach, 3 broccoli, 2 green bean, 5 carrot, 10 tomato, and 11 potato samples, were investigated for their total phenolic contents (TPC) and antioxidant properties. The measured antioxidant properties included free radical scavenging activities against DPPH, superoxide anion radical (O2), ABTS+ and peroxyl radical, and Fe2+ chelating capacity. The tested vegetables differed in their antioxidant properties and TPC, although all vegetables had significant antioxidant activities and contained significant levels of phenolics. The TPC and the measured antioxidant properties were correlated to each other. The Fe+2 chelating capacity and the scavenging capacities against ABTS+, DPPH and O2 were reported for these commonly consumed vegetables for the first time. Results from this study suggest that kale, spinach, broccoli, and rhubarb are the better dietary sources of natural antioxidant activities and phenolic compounds. Furthermore, kale, broccoli and spinach produced in Colorado may have greater antioxidant contents than those grown at other locations.  相似文献
