Y-21是从山东省淄博市蔬菜温室中分离到的一株细菌,通过室内平板对峙试验,对番茄灰霉病菌具有较强的拮抗性。本研究通过形态观察、生理生化反应和分子生物学方法对这株细菌进行系统鉴定。显微镜观察菌株形态为杆状;有芽孢;革兰氏染色呈阳性。采用经典的基因组提取方法提取基因组DNA,以其为模板,利用引物63F、1494R扩增16S rRNA基因,回收16SrRNA基因的PCR产物并与T载体连接,通过热激法将重组质粒转入大肠杆菌感受态细胞,利用蓝白斑筛选,挑出白斑,提取质粒,通过蓝白斑质粒的电泳图比对和质粒的PCR检测确定阳性克隆,将阳性克隆菌株送样测序。将测序结果输入BLAST进行比对,发现Y-21属于假单胞菌属,然后对其构建系统发育树。  相似文献

一株絮凝剂产生菌的16s rRNA鉴定和絮凝性能研究   总被引:2,自引:0,他引:2  
通过直接测序PCR产物的方法,获得了絮凝剂产生菌Lv1-2的16s rRNA基因序列,通过BLAST比对和系统进化树分析,将该菌鉴定为胶质芽孢杆菌(B.mucilaginosus)。对该菌产生的絮凝剂的热稳定性和助凝剂CaCl2的投入量进行了研究。结果表明,CaCl2有较好的促进絮凝的作用,絮凝率可提高10%;在30℃-90℃范围内絮凝剂有较好的热稳定性。  相似文献

Cellulose is the main structural component of lignocellulosic wastes that can be converted to sugars andbiofuels by cellulase. Due to wide applications of this enzyme in various industries around the world, cellulase isconsidered as the third industrial enzyme. The ability of thermophilic bacteria in the production of heat-stable cellulaseshas made them valuable tools in biotechnology. The aim of this study was isolation and molecular identification ofcellulolytic thermophile bacteria from Dig Rostam hot spring and investigating their cellulase activity. Samples weretaken from water and sediments of this hot spring, and cellulolytic bacteria were enriched in media containing celluloseas the only carbon source. The bacteria were incubated at 60°C, and single colonies were then isolated on solid media.Congo red assay was used as a quick test for the qualitative screening of cellulase activity. According to these qualitativeresults, four colonies named CDB1, CDB2, CDB3, and CDB4 were isolated, and their growth curve and some othercharacteristics were determined by biochemical assays. Moreover, endoglucanase, exoglucanase, and FPase activitiesof the isolates were investigated quantitatively. Results indicated that CDB1 exhibited the highest endoglucanase (0.096U/mL) and exoglucanase (0.156 U/mL) activities among other isolates. 16S rDNA partial sequencing indicated thatCDB1 had 99% similarity to the genus Anoxybacillus, and the other isolates showed the highest similarity to the genusGeobacillus. The cellulase gene of CDB1 isolate with the highest cellulase activity was also cloned, and its sequence isreported for the first time. Further studies on this thermophilic enzyme might be useful for industrial applications.  相似文献

We investigated the diversity and composition of microflora in feces of Lycopus lucidus Turcz.-fed mice. Inaddition, we evaluated the production of major cytokines (Interleukin-6 and -10) which are related to inflammationand fatty acid composition of several tissues. 16S ribosomal DNA sequencing-based microbiome taxonomic profilinganalysis was performed utilizing the EzBioCloud data base. Male mice fed on L. lucidus showed a significantlyreduced number of lactic acid bacteria and coliform in the feces compared with the control group (p < 0.05). 16SrDNA sequencing analysis of fecal samples showed that L. lucidus supplementation decreased the community ofharmful microflora (Enterobacteriaceae including Escherichia coli and Bacteroides sp.) in feces compared with thecontrol group (p < 0.05). There were no significant differences in mRNA expression of cytokine IL-6 and IL-10between the control and L. lucidus fed groups. The fecal fatty acid composition in the L. lucidus group hadpercentages of 4:0, 6:0, 8:0 and 10:0 in the intestine but those short chain fatty acids were not detected in the controlgroup. Our results showed that L. lucidus supplementation influenced gut environment by decreasing harmfulmicroflora and increased the percentages of several short fatty acids.  相似文献

The study was sought to enhance the synthesis of thermal stable β-cyclodextrin glycosyltransferase (β-CGTase)using potato wastewater as a low-cost medium and assess the degree to which it is efficient for industrial production ofβ-cyclodextrin (β-CD) from raw potato starch. Thermophilic bacteria producing β-CGTase was isolated from SaudiArabia and the promising strain was identified as Bacillus licheniformis using phylogenetic analysis of the 16S rRNAgene. Alginate-encapsulated cultures exhibited twice-fold of β-CGTase production more than free cells. Scanningelectron microscopy (SEM) of polymeric capsules indicated the potential for a longer shelf-life, which promotes therestoration of activity in bacterial cells across semi-continuous fermentation of β-CGTase production for 252 h. Theoptimal conditions for β-CGTase synthesis using potato wastewater medium were at 36 h, pH of 8.0, and 50°C with0.4% potato starch and 0.6% yeast extract as carbon and nitrogen sources, respectively. The purified enzyme showed aspecific activity of 63.90 U/mg with a molecular weight of ∼84.6 kDa as determined by SDS-PAGE analysis. The highenzyme activity was observed up to 60°C, and complete stability was achieved at 75°C. High levels of activity andstability were shown at pH 8.0, and the pH range from 7.0–10.0, respectively. The enzyme has an appreciable affinityfor raw potato starch with a Km of 5.7 × 10−6 M and a Vmax of 87.71 µmoL/mL/min. β-CD production was effectiveagainst 25 U/g of raw potato starch. The outcomes demonstrated its feasibility to develop a fermentation process byintegrating the cost-effective production of β-CGTase having distinctive properties for β-CD production with ecofriendly utilization of potato wastewater.  相似文献

Any vibration signal obtained from electromechanical systems contains a level of random changes. These random changes in the measured signal may be due to the random vibrations that can be related to the health of the machine for some faults such as dry bearing fault or bearing ageing. The presence of dry bearing fault, which is caused by the lack of lubricant, increases the level of random vibrations as compared to those obtained in healthy bearing machine. If these random vibrations could be isolated from the measured signal, useful information about bearing health may be obtained. Therefore, in this paper, signals (three line to line voltages, three currents, two vibration signals, four temperatures and one speed signal) obtained from the monitoring system are treated and analyzed using wavelet transform to correlate it to the dry bearing faults in induction machine. In this study, on-line analysis of the acquired signals has been performed using C++, while MATLAB has been used to perform the off-line analysis.  相似文献
