Different environmental conditions support optimal growth by Aphanizomenon and Microcystis in Ford Lake, Michigan, USA, based on weekly species biovolume and water chemistry measurements from June through October 2005-2007. Experimental withdrawal of hypolimnetic water through the outlet dam was conducted in 2006, with 2005 and 2007 acting as control years, to test theory regarding management of nuisance and toxic cyanobacteria. The dynamics of Aphanizomenon and Microcystis blooms in Ford Lake appear to be driven largely by NO3 concentrations, with higher levels shifting the advantage to Microcystis (P < 0.0001). Aphanizomenon was most successful with a mean TN:TP ratio (mol:mol) of 48.3:1, whereas Microcystis thrived with a mean ratio of 70.1:1. Withdrawal of hypolimnetic water successfully destabilized the water column and led to higher levels of NO3 and the near elimination of the Aphanizomenon bloom in 2006 (P < 0.0001). Selective withdrawal did not reduce Microcystis biovolume or microcystin toxicity. Microcystis biovolume and NO3 levels were positively correlated with microcystin toxin (P = 0.01) and jointly accounted for 30.5% of the variability in the data. Selective withdrawal may be a viable management option for improving water quality under certain circumstances. To fully address the problem of nuisance and toxic algal blooms in Ford Lake, however, an integrated approach is required that targets cyanobacteria biovolume dynamics as well as conditions suited for toxin production.  相似文献

Microcystis colonies are known to overwinter on the surface of the sediment of freshwater ecosystems. However, little is known about the genotypic and toxicological dynamics of Microcystis populations during this benthic life stage. In this study, we report a two-year-long survey of benthic populations of Microcystis, which had spent from a few days to more than six years in the sediment. In order to avoid any interaction with the planktonic proliferations, we chose two deeply buried benthic populations, which could be easily dated. Quantitative PCR on mcyB gene and protein phosphatase inhibition assays were performed to measure their toxic potential, and their genotypic structure was assessed by Capillary Electrophoresis-Single Strand Conformation Polymorphism (CE-SSCP), based on 16S-23S Intergenic Transcribed Spacer (ITS). The microcystin content of the cells seemed to change sharply during the first few months of benthic survival, whereas this content was low and decreased steadily after several years of benthic life. No genetic selection was observed in either the proportion of potentially toxic clones or the ITS sequences for any of the populations considered. From these results, the benthic life stage of Microcystis appears to preserve the structure and the composition of the population over a far larger time scale than classical overwintering period. Finally, some genotypes were common in both of the benthic populations, even though they originated from planktonic blooms that had developed five years apart, suggesting a major overlap of planktonic proliferations in successive years.  相似文献

This study investigates the toxicity and post-exposure effects of dissolved microcystin (MC-LR) on the dominant copepods of the upper San Francisco Estuary (SFE), where blooms of the toxic cyanobacteria Microcystis aeruginosa coincide with record low levels in the abundance of pelagic organisms including phytoplankton, zooplankton, and fish. The potential negative impact of Microcystis on the copepods Eurytemora affinis and Pseudodiaptomus forbesi has raised concern for further depletion of high quality fish food. Response of copepods to MC-LR (MC) was determined using a 48-h standard static renewal method for acute toxicity testing. Following exposure, a life table test was performed to quantify any post-exposure impacts on survival and reproduction. The 48-h LC-50 and LC-10 values for MC were 1.55 and 0.14 mg/L for E. affinis; and 0.52 and 0.21 mg/L for P. forbesi. Copepod populations recovered once dissolved MC was removed and cultures returned to optimal conditions, suggesting no post-exposure effects of MC on copepod populations. Dissolved microcystin above 0.14 mg/L proved likely to have chronic effects on the survival of copepods in the SFE. Since such high concentrations are unlikely, toxicity from dissolved microcystin is not a direct threat to zooplankton of the SFE, and other mechanisms such as dietary exposure to Microcystis constitute a more severe risk.  相似文献

Blooms of toxic cyanobacteria such as Microcystis aeruginosa periodically occur within wastewater treatment lagoons in the warmer months, and may consequently cause contamination of downstream water and outages of the supply of recycled wastewater. Lab-scale sonication (20 kHz) was conducted on suspensions of M. aeruginosa isolated from a wastewater treatment lagoon, and two other algal strains, Anabaena circinalis and Chlorella sp., to investigate cell reduction, growth inhibition, release of microcystin and sonication efficiency in controlling the growth of the M. aeruginosa. For M. aeruginosa, for all sonication intensities and exposure times trialled, sonication led to an immediate reduction in the population, the highest reduction rate occurring within the initial 5 min. Sonication for 5 min at 0.32 W/mL, or for a longer exposure time (>10 min) at a lower power intensity (0.043 W/mL), led to an immediate increase in microcystin level in the treated suspensions. However, prolonged exposure (>10 min) to sonication at higher power intensities reduced the microcystin concentration significantly. Under the same sonication conditions, the order of decreasing growth inhibition of the three algal species was: A. circinalis > M. aeruginosa > Chlorella sp., demonstrating sonication has the potential to selectively remove/deactivate harmful cyanobacteria from the algal communities in wastewater treatment lagoons.  相似文献

Short-term bacterial community composition (BCC) dynamics in response to accumulation and breakdown of Microcystis blooms were examined by conducting in situ mesocosm experiments with varying levels of Microcystis sp. biomass, ranging from 15 to 3217 μg/L as measured by chlorophyll-a concentration in the freshwater water column. The BCC was assessed by means of terminal restriction fragment length polymorphism (T-RFLP) of 16S ribosomal RNA genes followed by cloning and sequencing of selected samples. The results showed that the composition of both free-living and particle-attached bacterial communities changed during the accumulation and breakdown phases of a Microcystis bloom, and differences were also evident with different levels of Microcystis biomass. The relative abundance of bacteria affiliated with Micrococcineae and Legionellales increased in general after amendment with Microcystis. Significant correlation between the relative abundance of Micrococcineae and breakdown of Microcystis biomass was also observed. Canonical correspondence analysis (CCA) showed that the changes in the free-living and particle-attached bacterial community were mostly related to the changes in the concentrations of chlorophyll-a, dissolved organic carbon (DOC), dissolved oxygen (DO) and pH, which were mainly induced by the breakdown of Microcystis biomass. Overall, our study revealed the following: i) accumulation of Microcystis blooms and their breakdown have strong impacts on bacterial community composition; ii) there might be saprophytic association between Micrococcineae and decomposition of Microcystis biomass; iii) it is necessary to reveal potential associations between Legionellales organisms and Microcystis blooms in eutrophic freshwater lakes.  相似文献

Dissolved (extracellular) microcystin (MC) concentrations were determined at 3 sampling stations on Lake Kovada, Turkey. The dominant species of cyanobacteria found in August and September of 2006 were Microcystis aeruginosa, Synechococcus sp., Phormidium limosum, Phormidium formosa and Planktothrix limnetica. MC concentrations in water were measured by ELISA and MC variants were examined by HPLC-PDA. Quantitative analysis by HPLC indicated that five MC variants (MC-LR, -RR, -LA, -LW, -LF) were identified in water samples from Kovada Lake. The maximum concentration of dissolved MC-LW was 98.9 µg l− 1 in October. MC-LR was only detected in May at a concentration of 0.5 µg l− 1. The cross reactivity of the antibody (MC10E7) to variants such as MC-LA MC-LW & MC-LF was low. Hence the results determined by ELISA were lower than those determined by HPLC in September and October samples due to differences in the specificity of the antibody to MC variants. Total extraceullar MCs was quantified by ELISA and ranged from 0.73 to 48.5 µg MC-LR equivalents l− 1, which in some cases exceeded the WHO provisional Guideline Value for MC-LR in drinking water. This study confirms that the lakes of Turkey should be monitored for toxic cyanobacteria and for MCs to avoid or reduce the potential exposure of people to these health hazards.  相似文献

Contamination of recreational waters with Escherichia coli and Enterococcus sp. is a widespread problem resulting in beach closures and loss of recreational activity. While E. coli is frequently used as an indicator of fecal contamination, and has been extensively measured in waterways, few studies have examined the presence of potentially pathogenic E. coli strains in beach waters. In this study, a combination of high-throughput, robot-assisted colony hybridization and PCR-based analyses were used to determine the genomic composition and frequency of virulence genes present in E. coli isolated from beach water in Avalon Bay, Santa Catalina Island, CA. A total of 24,493 E. coli isolates were collected from two sites at a popular swimming beach between August through September 2007 and from July through August 2008. All isolates were examined for the presence of shiga-like toxins (stx1/stx2), intimin (eaeA), and enterotoxins (ST/LT). Of the 24,493 isolates examined, 3.6% contained the eaeA gene, indicating that these isolates were potential EPEC strains. On five dates, however, greater than 10% of the strains were potential EPEC, suggesting that incidence of virulence genes at this beach has a strong temporal component. No STEC or ETEC isolates were detected, and only eight (<1.0%) of the potential EPEC isolates were found to carry the EAF plasmid. The potential EPEC isolates mainly belonged to E. coli phylogenetic groups B1 or B2, and carried the β intimin subtype. DNA fingerprint analyses of the potential EPEC strains indicated that the isolates belonged to several genetically diverse groups, although clonal isolates were frequently detected. While the presence of virulence genes alone cannot be used to determine the pathogenicity of strains, results from this study show that potential EPEC strains can be found in marine beach water and their presence needs to be considered as one of the factors used in decisions concerning beach closures.  相似文献

This is the first to conduct simultaneous determination of microcystin (MC) contaminations in multi-groups of vertebrates (fish, turtle, duck and water bird) from Lake Taihu with Microcystis blooms. MCs (-RR, -YR, -LR) in Microcystis scum was 328 μg g− 1 DW. MCs reached 235 μg g− 1 DW in intestinal contents of phytoplanktivorous silver carp, but never exceeded 0.1 μg g− 1 DW in intestinal contents of other animals. The highest MC content in liver of fish was in Carassius auratus (150 ng g− 1 DW), followed by silver carp and Culter ilishaeformis, whereas the lowest was in common carp (3 ng g− 1 DW). In livers of turtle, duck and water bird, MC content ranged from 18 to 30 ng g− 1 DW. High MC level was found in the gonad, egg yolk and egg white of Nycticorax nycticorax and Anas platyrhynchos, suggesting the potential effect of MCs on water bird and duck embryos. High MC contents were identified for the first time in the spleens of N. nycticorax and A. platyrhynchos (6.850 and 9.462 ng g− 1 DW, respectively), indicating a different organotropism of MCs in birds. Lakes with deaths of turtles or water birds in the literatures had a considerably higher MC content in both cyanobacteria and wildlife than Lake Taihu, indicating that toxicity of cyanobacteria may determine accumulation level of MCs and consequently fates of aquatic wildlife.  相似文献

It is generally agreed that the hepatotoxic microcystins (MCs) are the most abundant toxins produced by cyanobacteria in freshwater. In various freshwater lakes in East Africa MC-producing Microcystis has been reported to dominate the phytoplankton, however the regulation of MC production is poorly understood. From May 2007 to April 2008 the Microcystis abundance, the absolute and relative abundance of the mcyB genotype indicative of MC production and the MC concentrations were recorded monthly in five freshwater lakes in Uganda: (1) in a crater lake (Lake Saka), (2) in three shallow lakes (Lake Mburo, George, Edward), (3) in Lake Victoria (Murchison Bay, Napoleon Gulf). During the whole study period Microcystis was abundant or dominated the phytoplankton. In all samples mcyB-containing cells of Microcystis were found and on average comprised 20 ± 2% (SE) of the total population. The proportion of the mcyB genotype differed significantly between the sampling sites, and while the highest mcyB proportions were recorded in Lake Saka (37 ± 3%), the lowest proportion was recorded in Lake George (1.4 ± 0.2%). Consequently Microcystis from Lake George had the lowest MC cell quotas (0.03-1.24 fg MC cell−1) and resulted in the lowest MC concentrations (0-0.5 μg L−1) while Microcystis from Lake Saka consistently showed maximum MC cell quotas (14-144 fg cell−1) and the highest MC concentrations (0.5-10.2 μg L−1). Over the whole study period the average MC content per Microcystis cell depended linearly on the proportion of the mcyB genotype of Microcystis. It is concluded that Microcystis populations differ consistently and independently of the season in mcyB genotype proportion between lakes resulting in population-specific differences in the average MC content per cell.  相似文献

A comparison of the role of two blue-green algae in THM and HAA formation   总被引:8,自引:0,他引:8  
The contribution of two blue-green algae species, Anabaena flos-aquae and Microcystis aeruginosa, to the formation of trihalomethanes (THMs) and haloacetic acids (HAAs) was investigated. The experiments examined the formation potential of these disinfection by-products (DBPs) from both algae cells and extracellular organic matter (EOM) during four algal growth phases. Algal cells and EOM of Anabaena and Microcystis exhibited a high potential for DBP formation. Yields of total THMs (TTHM) and total HAAs (THAA) were closely related to the growth phase. Reactivity of EOM from Anabaena was slightly higher than corresponding cells, while the opposite result was found for Microcystis. Specific DBP yields (yield/unit C) of Anabaena were in the range of 2-11 μmol/mmol C for TTHM and 2-17 μmol/mmol C for THAA, while those of Microcystis were slightly higher. With regard to the distributions of individual THM and HAA compounds, differences were observed between the algae species and also between cells and EOM. The presence of bromide shifted the dominant compounds from HAAs to THMs.  相似文献

An existing volunteer monitoring network in the state of Michigan was exploited to conduct a statewide survey of the cyanobacterial toxin, microcystin, and to test hypotheses about the interactive influences of eutrophication and dreissenid mussel invasion. A total of 77 lakes were sampled by citizen volunteers for microcystin, total phosphorus (TP) and chlorophyll a. Microcystin was measured in depth-integrated samples collected from the euphotic zone as well as in surface-water samples collected along the shoreline. Average microcystin in samples collected by volunteers was not different from samples collected side-by-side by professionals. Euphotic-zone microcystin was positively related to TP in lakes without dreissenids (uninvaded) but not in lakes with dreissenids (invaded). Regression-tree analysis indicated that euphotic-zone microcystin was eight times higher in the presence of dreissenids for lakes with TP between 5 and 10 μg L−1. In contrast, euphotic-zone microcystin was almost identical in invaded and uninvaded lakes with TP between 10 and 26 μg L−1. Across all lakes, microcystin concentrations at the surface were on average more than double, and in some cases an order-of-magnitude greater than, concentrations in the euphotic-zone. Given these results, it seems prudent to include dreissenid invasion status in forecasting models for microcystin, and to include shoreline sampling in monitoring programs aimed at assessing recreational exposure to cyanobacterial toxins.  相似文献

We have developed a rapid and robust technological solution including a membrane filtration and dissolution method followed by a molecular enrichment and a real-time PCR assay, for detecting the presence of Enterococcus sp. or Enterococcus faecalis/faecium per 100 mL of water in less than 5 h and we compared it to Method 1600 on mEI agar in terms of specificity, sensitivity, and limit of detection. The mEI and the Enterococcus sp.-specific assay detected respectively 73 (64.0%) and 114 (100%) of the 114 enterococcal strains tested. None of the 150 non-enterococcal strains tested was detected by both methods with the exception of Tetragenococcus solitarius for the Enterococcus sp. assay. The multiplexed E. faecalis/faecium assay efficiently amplified DNA from 47 of 47 (100%) E. faecalis and 27 of 27 (100%) E. faecium strains tested respectively, whereas none of the 191 non-E. faecalis/faecium strains tested was detected. By simultaneously detecting the predominant fecal enterococcal species, the E. faecalis/faecium-specific assay allows a better distinction between enterococcal strains of fecal origin and those provided by the environment than Method 1600. Our procedure allows the detection of 4.5 enterococcal colony forming units (CFU) per 100 mL in less than 5 h, whereas the mEI method detected 2.3 CFU/100 mL in 24 h (95% confidence). Thus, our innovative and highly effective method provides a rapid and easy approach to concentrate very low numbers of enterococcal cells present in a 100 mL water sample and allows a better distinction between fecal and environmental enterococcal cells than Method 1600.  相似文献

An experiment using nitrogen stable isotope tracer (15N) was conducted to track the fate of nitrogen derived from cyanobacterial blooms and the effectiveness with which the seasonal blooms are retained by vegetated and unvegetated sediment in a large shallow eutrophic lake (Lake Taihu, China). 15N enriched Microcystis was injected into both unvegetated sediment and sediment occupied by common reed (Phragmites australis) in the littoral zone. Nutrient retention by the vegetated sediment was greater than by the unvegetated sediment, resulting in higher δ15N in the sediment nitrogen pool. The labeled Microcystis material was also distributed deeper into the vegetated sediment than the unvegetated sediment. A portion of the Microcystis-derived nitrogen was quickly assimilated, appearing first in the belowground biomass and subsequently in the aboveground biomass of the reed plants. The labeled nitrogen was found to support new growth as evidenced by 15N enrichment of new leaves. This study indicates that common reed beds in the littoral zone may play an important role in retention of sedimented planktonic materials.  相似文献

We evaluated the use of DGGE fingerprinting to differentiate communities of Escherichia coli from animal and geographic sources. An initial screening of 15 gene candidates revealed the ability of three target genes (mdh, phoE and uidA-4) to effectively differentiate E. coli communities originating in horses, pigs, geese and goats. Cluster and jackknife analyses performed on the communities from a more extensive number of hosts (n = 150) including humans (via raw sewage), horses, pigs, geese and cows revealed that the internal accuracy of classification of E. coli community fingerprints to their origin was similar for each of the three genes (85-86%). Each of the three genes were tested for their ability to associate E. coli source- and sink communities in two settings featuring contaminated water; (i) a stream receiving municipal wastewater effluent and (ii) a pond inhabited by geese. For each gene, DGGE fingerprints effectively matched effluent- and downstream E. coli communities (98-100% similarity) and excluded upstream communities, while communities from goose fecal material were 77-79% similar to communities in pond water, indicating fecal inputs from geese. Furthermore, each gene discriminated against E. coli communities from hosts non-indigenous to either setting. DGGE analysis of E. coli communities appears to be a promising tool to augment existing efforts aiming to address the dynamics of bacteria pollution in complex, natural environments.  相似文献

We tested the hypothesis that zebra mussels (Dreissena polymorpha) have positive effects on the toxin-producing cyanobacterium, Microcystis aeruginosa, at low phosphorus (P) concentrations, but negative effects on M. aeruginosa at high P, with a large-scale enclosure experiment in an oligotrophic lake. After three weeks, mussels had a significantly positive effect on M. aeruginosa at ambient P (total phosphorus, TP ∼10 μg L−1), and a significantly negative effect at high P (simulating a TP of ∼40 μg L−1 in lakes). Positive and negative effects were strong and very similar in magnitude. Thus, we were able to ameliorate a negative effect of Dreissena invasion on water quality (i.e., promotion of Microcystis) by adding P to water from an oligotrophic lake. Our results are congruent with many field observations of Microcystis response to Dreissena invasion across ecosystems of varying P availability.  相似文献

Cyanobacteria that produce the toxin microcystin have been isolated from many parts of the world. Most of these organisms are planktonic; however, we report on several microcystin-producing benthic filamentous cyanobacterial isolates from four drinking-water reservoirs in southern California (USA): Lake Mathews, Lake Skinner, Diamond Valley Lake (DVL), and Lake Perris. Some samples of benthic material from these reservoirs tested positive for microcystin by an ELISA tube assay, and all the positive samples had in common a green filamentous cyanobacterium 10-15microm in diameter. Seventeen unialgal strains of the organism were isolated and tested positive by ELISA, and 11 cultures of these strains were found to contain high concentrations of microcystin-LR (90-432microgL(-1)). The cultures were analyzed by protein phosphatase inhibition assay (PPIA) and HPLC with photodiode array detector (PDA) or liquid chromatography/mass spectrometry (LC/MS). Microcystin per unit carbon was determined for six cultures and ranged from 1.15 to 4.15microgmg(-1) C. Phylogenetic analysis of four cultures from Lake Skinner and DVL using cyanobacterial-specific PCR and sequencing of the partial 16S rRNA gene suggested the highest similarity to an unidentified cyanobacterium in the oscillatoriales, and to a Phormidium sp. Morphologically, some of the isolates were similar to Oscillatoria, and others resembled Lyngbya. The significance of these organisms lies in the relative scarcity of known toxin producers among freshwater benthic cyanobacteria, and also as a source of cell-bound microcystin in these reservoirs.  相似文献

The cyanobacterium Microcystis, which occurs as colonies of different sizes under natural conditions, can produce toxic microcystins (MCs). To monitor the toxicity and assess the risk of Microcystis blooms in Lake Taihu, it is important to investigate the relationship between MC production and Microcystis colony size. In this study, we classified Microcystis collected from Zhushan Bay of Lake Taihu during blooms into four classes with size of <50 μm, 50–100 μm, 100–270 μm and >270 μm and studied their differences in MC production and genetic structure. The results showed that colonies with size of <50, 50–100, 100–270 and >270 μm produced 12.2 ± 11.2%, 19.5 ± 7.9%, 61.3 ± 12.6%, and 7.0 ± 9.6% of total MC, respectively. The proportion of cell density of colonies with size of 50–100, 100–270 and >270 μm was positively correlated with MC concentration during blooms, while that of colonies with size of <50 μm was negatively correlated. The MC cell quota tended to be higher during blooms in colonies with larger size except that of colonies with size of 100–270 μm was higher than that of colonies with size of >270 μm from June 11 to September 16. Colonies with size of <50 μm showed the highest proportion of the less toxic MC congener MC-RR, and colonies with size of >100 μm showed higher proportion of the most toxic MC congener MC-LR than colonies with size of <100 μm. Real-time PCR indicated that larger colonies had higher proportion of potential toxic genotype. Principal component analysis of PCR-denaturing gradient gel electrophoresis profile showed that cpcBA and mcyJ genotype compositions were different between colonies with size of <50 μm and colonies with size of >50 μm, and cpcBA genotype composition was also different among colonies with size of 50–100 μm, 100–270 μm and >270 μm. These results indicated that MC cell quota and congener composition were different in Microcystis colonies with different sizes in Lake Taihu during blooms, and the differences in MC production in colonies with different size resulted chiefly from the difference in their genotype composition. Therefore, the authorities of water quality monitoring and drinking water supply service in Lake Taihu should be alert that the toxicity of Microcystis colony with different size was different during blooms, and the high abundance of colonies larger than 50 μm could be an indicator of relatively high bloom toxicity.  相似文献

An exploratory study on the management of undesirable cyanobacteria blooms with respect to off-flavor problems using an integrated vertical-flow constructed wetland (CW) was performed at a small commercial-scale channel catfish farm from 2004 to 2007. The results of the three-year experiment indicated that water treatment by the CW could reduce the possibility of dominance by undesirable cyanobacteria species that often cause off-flavor problems. A detailed investigation in 2007, showed that the concentrations of geosmin, MIB (2-methylisoborneol), and β-cyclocitral in the water of the recirculating pond (4.3 ng L−1, U.D. (undetected) and 0.2 ng L−1, respectively) treated by the CW were significantly lower than those in the control pond (152.6 ng L−1, 63.3 ng L−1 and 254.8 ng L−1, respectively). In addition, the relationships among the cyanobacteria species, the off-flavor compounds and ten environmental variables were explored by canonical correspondence analysis (CCA). The results showed that Oscillatoria sp., Oscillatoria kawamurae and Microcystis aeruginosa were the main sources of off-flavor compounds in the catfish ponds. The successful manipulation of undesirable cyanobacteria species potentially resulted in lower concentrations of odorous compounds in the water of the recirculating pond. An investigation of the concentrations of geosmin and MIB in catfish fillets showed that the levels of odorous compounds were below the OTC (odor threshold concentration) values in the recirculating pond but were above the OTC values from July to October in the control pond. Water recycling by the CW could potentially be one of the best management practices to control off-flavor occurrences in aquaculture.  相似文献

Animal agriculture in watersheds produces manure bacteria that may contaminate surface waters and put public health at risk. We measured fecal indicator bacteria (commensal Escherichia coli and fecal enterococci) and manure pathogens (Salmonella and E. coli 0157:H7), and physical-chemical parameters in pond inflow, within pond, pond outflow, and pond sediments in three ponds in agricultural watersheds. Bishop Pond with perennial inflow and outflow is located in the Piedmont, and Ponds A and C with ephemeral inflow and outflow in the Coastal Plain of Georgia. Bromide and chloride tracer experiments at Bishop Pond reflected a residence time much greater than that estimated by two models, and indicated that complete mixing within Bishop Pond was never obtained. The long residence time meant that fecal bacteria were exposed to solar UV-radiation and microbial predation. At Bishop Pond outflow concentrations of fecal indicator bacteria were significantly less than inflow concentrations; such was not observed at Ponds A and C. Both Salmonella and E. coli 0157:H7 were measured when concomitant concentrations of commensal E. coli were below the criterion for surface water impairment indicating problems with the effectiveness of indicator organisms. Bishop Pond improved down stream water quality; whereas, Ponds A and C with ephemeral inflow and outflow and possibly greater nutrient concentrations within the two ponds appeared to be less effective in improving down stream water quality.  相似文献

The ubiquitous application and release of antibiotics to the environment can result in bacterial antibiotic resistance, which in turn can be a serious risk to humans and other animals. Southeast Asian countries commonly apply an integrated recycling farm system called VAC (Vegetable, Aquaculture and Caged animal). In the VAC environment, antibiotics are released from animal and human origins, which would cause antibiotic-resistant bacteria (ARB). This study evaluated occurrence of ARB in the VAC environment in northern Vietnam, with quantitative analysis of antibiotic pollution. We found that sulfonamides were commonly detected at all sites. In dry season, while sulfamethazine was a major contaminant in pig farm pond (475-6662 ng/l) and less common in city canal and aquaculture sites, sulfamethoxazole was a major one in city canal (612-4330 ng/l). Erythromycin (154-2246 ng/l) and clarithromycin (2.8-778 ng/ml) were the common macrolides in city canal, but very low concentrations in pig farm pond and aquaculture sites. High frequencies of sulfamethoxazole-resistant bacteria (2.14-94.44%) were found whereas the occurrence rates of erythromycin-resistant bacteria were lower (< 0.01-38.8%). A positive correlation was found between sulfamethoxazole concentration and occurrence of sulfamethoxazole-resistant bacteria in dry season. The sulfamethoxazole-resistant isolates were found to belong to 25 genera. Acinetobacter and Aeromonas were the major genera. Twenty three of 25 genera contained sul genes. This study showed specific contamination patterns in city and VAC environments and concluded that ARB occurred not only within contaminated sites but also those less contaminated. Various species can obtain resistance in VAC environment, which would be reservoir of drug resistance genes. Occurrence of ARB is suggested to relate with rainfall condition and horizontal gene transfer in diverse microbial community.  相似文献
