Microwave energy was applied to inactivate the oxydoreductases peroxidase (POD, EC 1.11.1.7) and polyphenol oxidase (PPO, EC 1.14.18.1) in processed fruit products. Microwave blanching of papaya, strawberry and kiwi purees at various conditions of power and time produced inactivation of PPO and POD activities depending on the fruit and the heating conditions. Treatment at 850 W/60 s produced about 60% of POD inactivation for papaya and kiwi fruit. POD activity in strawberry, however, seemed to be more resistant to microwave inactivation; treatment at 850 W/60 s only achieved a loss of POD activity near 8%. Papaya oxidoreductases showed lower stability in the microwave treatments tested. Microwave blanching at 475 W/45 s produced about 75% inactivation of POD activity and nearly complete PPO inactivation. Kiwi fruit and strawberry purees exhibited similar inactivation of PPO – 32% at 475 W/30 s and 70% at 475 W/60 s. The decrease of PPO activity in both products was almost linear at constant power. This thermal treatment, however, directly affects the colour of the fruit pulps. Papaya, kiwi and strawberry purees suffered slight colour (CIE L* a*b*) changes. Carotenoid, chlorophyll and anthocyanin changes were evaluated by HPLC and related to objective colour. Microwave treatments produced small modifications of the quantitative and qualitative composition of carotenoids (in papaya) and anthocyanins (in strawberry). High hydrostatic pressure treatment (50-400 MPa) combined with heat treatment (20–60°C) effects on peroxidase (POD), polyphenoloxidase (PPO) and pectin methylesterase (PME) activities of fruit-derived products were studied. Assays were carried out on fresh orange juice and strawberry puree. Pressurization/depressurization treatments caused a significant loss of strawberry PPO (60%) up to 250 MPa and POD activity (25%) up to 230 MPa, while some activation was observed for treatments carried out in 250–400 MPa range for both enzymes. Optimal inactivation of POD was using 230 Mpa and 43°C in strawberry puree. Combinations of high pressure and temperature effectively reduced POD activity in orange juice (50%) to 35°C. The effects of high pressure and temperature on PME activity in orange juice were very similar to those for POD.  相似文献

The effects of refrigerated storage at 2 °C and 95% RH and processing treatments on the bioactive compounds and antioxidant capacities of ‘Marion’ and ‘Evergreen’ blackberries were investigated. During refrigerated storage, total phenolics (TPC), total monomeric anthocyanins (ACY), and radical scavenging activity (RSA) fluctuated in ‘Marion’, but TPC and ACY continuously declined in ‘Evergreen’. Oxygen radical absorbance capacity (ORAC) and ferric reducing antioxidant power (FRAP) decreased by 20% and increased by 19% in ‘Evergreen’, respectively after 7-d refrigerated storage, while no changes (P > 0.05) in ‘Marion’ were observed. Compared with frozen samples, freeze-dried ‘Evergreen’ had higher TPC (21%), ACY (5.5%), and RSA (14%), while hot-air dried “Marion” had lower ACY (56%), ORAC (37%), and FRAP (27%) and hot-air dried ‘Evergreen’ had lower TPC (37%), ACY (84%), and RSA (13%). ORAC and FRAP in canned ‘Marion’ was 21-61% lower than that of frozen samples. Jam also had lower TPC and ACY (67-84%), RSA (˜80%), and ORAC and FRAP (65-77%) values than frozen ones in both varieties (P < 0.05). The 6-mo post-process room storage had little effect on the bioactive compounds of frozen and freeze dried samples, but reduced ACY in hot-air dried, canned, and jam samples, and antioxidant capacities of all samples (P < 0.05).  相似文献

To investigate the relationship between bioactive compounds and antioxidant property, total phenolic content (TPC), total flavonoid content (TFC), individual and total isoflavone content of soymilk and their correlations with oxygen‐radical‐absorbing capacity (ORAC), ferric reducing antioxidant power (FRAP) and 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging capacity values were evaluated and compared. Results showed that TPC, TFC, isoflavones (including concentration and profiles) and antioxidation activity significantly varied among ten tested soybean cultivars. Significant correlations were established between DPPH and TFC (= 0.553, < 0.01), DPPH and TPC (= 0.753, < 0.01), FRAP and TFC (= 0.599, < 0.01) and FRAP and TPC (= 0.616, < 0.01). Positive linear correlations were found between subtotal, total isoflavones and ORAC. Unlike other isoflavone monomers, aglycone isoflavones correlated positively with DPPH and FRAP significantly. DPPH well correlated with FRAP, whereas neither DPPH nor FRAP correlated with ORAC. TPC, TFC, individual and total isoflavone values are potentially useful for soymilk antioxidant activity assessment.  相似文献

The effects of conventional (CV) and microwave (MW) heating on total soluble solid (TSS) content, total anthocyanin content (TAC), total phenolic content (TPC) and antioxidant activity of raspberry juice obtained from two cultivars of Amol (AM) and Siyahkal (SK) were scrutinised. Antioxidant activity of the juices was measured by 2,2 diphenyl‐1‐picrylhydrazyl (DPPH) radical scavenging capacity and expressed as EC50 value. In the both methods, the concentration rate constant for the juices was significantly decreased by increasing operational pressure from 12 to 100 kPa (< 0.05). A first‐order reaction kinetic model was successfully fitted for the degradation of monomeric anthocyanins at all operating pressures. The results also showed that thermal treatment of MW compared CV in both SK and AM cultivars caused a lower decrease in the TAC, TPC and antioxidant activity. Moreover, the changes in trend of the antioxidant activities due to the thermal treatment were positively correlated with the TPC (= 0.74, < 0.05) and TAC (= 0.61, < 0.05).  相似文献

The objective of this work was to study the thermal and high pressure inactivation kinetics of polyphenol oxidase (PPO) and peroxidase (POD) in strawberry puree. PPO from two strawberry cultivars (‘Festival’ and ‘Aroma’) was found to be highly thermostable in strawberry puree with no significant inactivation even after 30 min treatment at 100 °C. In contrast, POD from the two cultivars displayed very high thermosensitivity with complete inactivation in less than 5 min at 70 °C. The thermal inactivation kinetics of strawberry POD was described by a biphasic model. The activation energies for the inactivation of the stable and the labile fractions were estimated to be 254.9 and 221.6 kJ/mol respectively. Combined high pressure–thermal treatment at pressures ranging from 100 to 690 MPa, temperatures ranging from 24 to 90 °C and treatment times between 5 and 15 min did not have significant effect on PPO while substantial inactivation of POD was observed. The inactivation kinetics of POD during combined high pressure–thermal processing was well described by first-order kinetics probably due to the inactivation of the labile fraction during the pre-heating and the compression phase.Industrial relevanceThe oxidative enzymes polyphenol oxidase and peroxidase cause the degradation of anthocyanins and other polyphenols in strawberry products, leading to discoloration and loss of antioxidant activity. In this work the thermal and high pressure inactivation of strawberry polyphenol oxidase and peroxidase was investigated so as to assess the suitability of high pressure processing as an alternative to thermal processing. Strawberry polyphenol oxidase was found to be highly resistant to both thermal and high pressure inactivation. Thus in order to maintain the quality of processed strawberry products, high pressure processing should be accompanied by additional measures such as exclusion of oxygen, refrigerated storage and the use of natural enzyme inhibitors.  相似文献

The aim of this study was to determine the concentrations of phenolic compounds and their antioxidant activity in different kinds of juice: clear, cloudy, and puree which were made from three different strawberry cultivars (Elkat, Kent, and Senga Sengana). The anthocyanins, p-coumaric acid, ellagic acid, quercetin, keampferol derivatives, (+)-catechin, proanthocyanidins content and degree of proanthocyanidin polymerization, were determined both in the fresh and after 6 months of storage at 4 and 30 °C. Freshly produced juices contained higher amounts of phenolics, especially of anthocyanins and proanthocyanidins, than those stored for 6 months at 4 and 30 °C. The processing of the clear juice showed the higher loss of all phenolic compounds. The antioxidant capacity was the smallest for clear, and the highest for the puree juices. This was assessed by measurements made with different antioxidant activity assays: ABTS and FRAP. The puree of strawberry juice had significantly higher levels of the phenolic compounds and showed more antioxidant activity than the clear and cloudy juices, before and after storage in all strawberry cultivars.  相似文献

Effect of rice bran protein extract (RBPE) on enzymatic browning inhibition in potato puree was studied by colour measurement and polyphenol oxidase (PPO) inhibition. RBPE inhibited browning in potato puree and showed a higher per cent potato PPO inhibition at pH 4.0 and 5.0 than those at pH 3.0, 6.0 and 7.0 (P ≤ 0.05). RBPE heated to 40 and 60 °C had browning inhibition in potato puree and per cent potato PPO inhibition at a similar extent to unheated RBPE (P > 0.05). Browning inhibition and per cent potato PPO inhibition of RBPE were decreased when it was heated to 80 °C (P ≤ 0.05). RBPE inhibited browning in potato puree higher than 5, 10 and 20 mm ascorbic acid and 5 and 10 mm citric acid (P ≤ 0.05). Regarding the kinetics study, RBPE exhibited a mixed‐type inhibition for potato PPO. Therefore, RBPE has a potential to be used as a natural antibrowning agent in the potato industry.  相似文献

The hydroxycinnamic acid (HA) content of starting barley for brewers’ spent grains (BSG), whole BSG and phenolic extracts from BSG was measured using high performance liquid chromatography (HPLC) and correlated with antioxidant potential. The effect of BSG phenolic extracts on antioxidant activity of fruit beverages was also assessed (using the total phenolic content (TPC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) assays). The concentration of HA present in barley extract and BSG was in the order of ferulic acid (FA), p-coumaric acid (p-CA) derivatives, FA derivatives, p-CA, caffeic acid (CA) and CA derivatives. Results suggested that brewing and roasting decreased the HA content. Antioxidant activity was significantly (P < 0.05) correlated with caffeic acid (R2 = 0.8309) and total HA (R2 = 0.3942) concentrations. Addition of extracts to fruit beverages resulted in a significant (P < 0.05) increase in antioxidant activity of cranberry juice, measured by the FRAP assay. In vitro digestion significantly (P < 0.05) reduced TPC, DPPH and FRAP activity of the fruit beverages.  相似文献

Tea polyphenols possess antioxidant properties and have been shown to have a protective effect against several degenerative diseases. The study aimed to determine the amounts of polyphenols and antioxidant properties for teas grown in Sri Lanka, over a period of 10 months. Water extracts of freeze-dried fresh (unfermented) and fully-fermented tea leaves were made for a structured set of samples (fermented and unfermented teas from six plantations; teas representing two harvesting seasons from four plantations) collected from the main tea growing regions in Sri Lanka. Total phenolic content (TPC), the ferric reducing antioxidant power (FRAP) and the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activity were determined for each sample. The results highlight significant (P < 0.05) variations in antioxidant activity across the six plantations. FRAP and DPPH for both fermented and unfermented teas from the four highland plantations showed a significant (P < 0.05) interaction between season and plantation. A similar interaction between season and plantation was observed for total phenolics in unfermented teas from the four highland plantations. The variability of the total phenolics for fermented teas, however, was independent of seasonal variations. A significant correlation (r = 0.5, P < 0.05) was observed between FRAP and total phenolics.  相似文献

Abstract: Phenolic compounds and anthocyanins in muscadines have attracted much attention due to their diverse biological activities. With bioassays of antioxidant activities in terms of total phenolic content (TPC), total anthocyanin content (TAC), total procyanidin content (TPA), oxygen radical absorbance capacity (ORAC), and ferric reducing antioxidant power (FRAP) of different parts of the Noble muscadine, the butanol (BuOH) extract of the muscadine skin showed the highest TPC (317.91 ± 1.83 mg GAE/100 g FW), which might be ascribed to its high TAC of 227.06 ± 1.29 mg/100 g fresh weight (FW). The ethyl acetate (EtOAc) extract of the muscadine seed contained the highest TPA (55.30 ± 0.63 mg CE/100 g FW). Correlation analyses demonstrated a significant linear relationship of TPC and TAC compared to their ORAC and FRAP values within the range of R2 from 0.9283 to 0.9936, which suggested that phenolics and anthocyanins in the extracts contributed significantly to their antioxidant potential. Nineteen individual phenolics and 5 anthocyanins were identified by HPLC‐MS, which indicated different chemical profiles of anthocyanins and other phenolics in the muscadine extracts. Practical Application: The paper has provided rich information of bioactive phytochemical profiles in different solvent extracts and their correlation with the antioxidant activity in the muscadine that is a very special regional fruit in U.S. Its high content of phenolic compounds demonstrates that muscadine could be beneficial to human health.  相似文献

Riceberry bran protein hydrolysate (RBPH) was prepared from bran of purple‐pigmented Riceberry rice using enzymatic hydrolysis. The effect of enzyme type (Alcalase, Flavourzyme and Neutrase) and hydrolysis time (2, 4 and 6 h) on protein content, protein yield, total phenolic content (TPC), antioxidant activities (ABTS and FRAP) and molecular weight patterns of RBPH was investigated. The enzyme type significantly (< 0.05) affected the properties of RBPH whereas the hydrolysis time had no significant effect (P ≥ 0.05) on those properties. Flavourzyme was the most effective to increase protein yield, TPC and antioxidant activities compared to Alcalase and Neutrase. The optimal hydrolysis condition was 4 h using Flavourzyme which yielded 74.9% extracted protein. This hydrolysate contained peptides ranging from 16 to 64 kDa. The high antioxidant activity was related to negative charge peptides as shown by anion exchange chromatography. With high protein content and antioxidant properties, RBPH using Flavourzyme could be practically utilised in functional foods.  相似文献

High pressure processing (HPP) is a non-thermal technology used to activate or inactivate enzymes. This study investigated the effects of HPP (600 MPa for 5 or 30 min at 25 °C) on cocoyam, Peruvian carrot and sweet potato color, and the polyphenoloxidase (PPO) and peroxidase (POD) activities in tuber cubes, puree, and enzyme extract subjected to HPP. The results showed enzyme inactivation by HPP in cocoyam (up to 55% PPO inactivation in puree and 81% POD inactivation in extract) and Peruvian carrot (up to 100% PPO and 57% POD inactivation the extract). In contrast, enzyme activation was observed in sweet potato (up to 368% PPO and 27% POD activation in puree). The color results were compatible to enzyme activity: the color parameters remained unchanged in cocoyam and Peruvian carrot, which showed high PPO and POD inactivation after HPP. Furthermore, the impact of HPP on the enzymes was influenced by the matrix in which HPP was carried out, evidencing that the enzyme structure can be protected in the presence of other food constituents.Industrial relevanceThe enzymes PPO and POD are an important concern for vegetable processing, due its ability to induce browning after vegetables are cut. The HPP at 600 MPa for 5 or 30 min can be used to inactivate these enzymes in cocoyam and Peruvian carrot, guaranteeing the color and freshness of the tubers similar to the fresh cut vegetable.  相似文献

The total phenolic content (TPC) and antioxidant activity of fresh and dried Phyllanthus amarus plant materials were evaluated using the Folin-Ciocalteau method, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity and ferric reducing antioxidant power (FRAP) assays. Different drying treatments led to significant reduction (P<0.05) in antioxidant properties of P. amarus methanolic extracts, with microwave drying causing the highest decrease in TPC and antioxidant activity exhibited by the reduction in both radical scavenging activity and FRAP. On the other hand, boiling water extracts appeared to exhibit significantly stronger antioxidant potentials (P<0.05) even in dried plant materials due to greater solubility of compounds, breakdown of cellular constituents as well as hydrolysis of tannins. Its strong free radical scavenging activity suggests that it has great potential in the food industry as functional food ingredient.  相似文献

The assessment of geographical origin of honey is economically important for producers and consumers as every region may present particular quality characteristics. In this study, honeys from the seven different regions of Buenos Aires province (Argentina) were characterised by their antioxidant capacity (DPPH, FRAP), total phenolic content (TPC), mineral composition, colour and ash. Honeys showed significant differences among their antioxidant capacity (DPPH), ash, colour and mineral content ( 0.05). Besides, a good antioxidant activity and low amounts of Cu and Zn (<1.0–1.5 and 0.7–1.8 mg kg?1, respectively) were found in the samples. Significant Pearson's correlations ( 0.05) among the different parameters were found. Moreover, the linear discriminant analysis allowed the classification of honeys in their original groups with a prediction success of 98%. The present results suggest that honeys could be correctly classified by their geographical origin through their TPC, colour, ash and mineral concentrations.  相似文献

This study's objective was to characterize the effect of traditional and 3 newly devised (soaking+grinding) methods combined with cooking on the content and composition of phenolic substances, antioxidant, and antiproliferative properties of soymilk prepared from black soybean. Phenolic substances and antioxidant profile were characterized and antiproliferation of prostate cancer DU145 cells was conducted using a cell culture assay. Results indicated Grinding Method 4 produced significantly (P < 0.05) higher total phenolic content (TPC), total flavonoid content (TFC), condensed tannin content (CTC), and total isoflavone content in both raw and cooked black soymilk as compared to Method 1. Cooking soymilk reduced 23% to 38% of total phenolic substances. Raw black soymilk produced by Method 4 displayed the highest antioxidant capability, which was determined using ORAC, FRAP, and DPPH assays, and a higher antiprostate cell proliferation ability. Cooking only slightly reduced the potency to inhibit DU145 prostate cancer cells as IC50 value was increased from the average of about 4.0 mg/mL of raw soymilk extracts to 5.5 mg/mL of cooked soymilk extracts of all grinding methods. Overall, total isoflavone content was the only component that was negatively correlated with IC50 value (r = –0.93, P < 0.05) which indicates the ability to inhibit prostate cancer cell is associated with the increase in total isoflavone content, not with any other phenolic substances or antioxidant properties.  相似文献

Emerging extraction techniques, including pulsed electric field (PEF) and ultrasound (US), are attracting considerable interest in the recovery of bioactives. Though, limited work has focused on PEF application as pre-treatment for US assisted extraction to enhance the release of phenolics from herbs. Hence, the present study investigated the use of an optimized PEF pre-treatment to enhance the recovery of phenolics from fresh rosemary and thyme by-products in a subsequent US assisted extraction step. Total phenolic content (TPC), 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and ferric reducing antioxidant power (FRAP) were assessed as an index of extraction efficacy. Qualitative and quantitative analyses were performed through liquid chromatography-mass spectrometry analyses to evaluate the influence of the methods on individual phenolic compounds and the formation of potential derivatives. The results indicated that in a number of cases PEF pre-treatment enhanced (p < 0.05) the recovery of phenolic compounds and antioxidant capacity compared to US individually.  相似文献

The aim of this study was to evaluate the potency of polyphenolic extract from black radish (Raphanus sativus L. var. niger) root peel waste (PEBRRP) as an antioxidant food additive. PEBRRP was found a powerful antioxidant extract, in which had 172.9 ± 11.5, 462.72 ± 3.05, and 796.51 ± 10.4 mg TE per g dw of total antioxidant capacity (TAC) by CUPRAC, DPPH, and FRAP assays, respectively. The total phenolic content (TPC) and total flavonoid content (TFC) of PEBRRP were found as 305.51 ± 5.2 mg GAE per g dw and 171.58 ± 6.2 mg CAE per g dw, respectively. The ferulic acid (28.02 ± 4.5 mg per g dw) and epicatechin (19.82 ± 3.6 mg per g dw) were identified as the main polyphenols in undigested PEBRRP. The bioaccessibility of total phenolic and total flavonoid compounds of PEBRRP was found approximately 5%, and the antioxidant capacity of bioaccessible PEBRPP fraction was determined 6% in CUPRAC, 36% in DPPH and 47% in FRAP methods.  相似文献

比较蓝莓花、嫩茎、嫩叶及老叶总多酚、总黄酮、原花青素的含量,并研究其抗氧化性能差异。结果表明:嫩叶中总多酚含量最高,为43.77 mg GAE/g(以干质量计,下同),分别是花、嫩茎和老叶的3.45、1.42、1.12倍;老叶中总黄酮、原花青素含量最高,为68.01 mg RE/g、23.29 mg CAE/g,分别是花的2.57倍和4.61倍、嫩茎的1.79倍和1.23倍、嫩叶的1.03倍和1.98倍。抗氧化性能中,老叶的铁还原能力最大,其次是嫩茎和花,嫩叶较差;清除·OH的能力为嫩茎嫩叶花老叶;清除DPPH自由基能力和总抗氧化能力均为嫩茎嫩叶老叶花甲醇提取物,均高于对照品;蓝莓各部分提取物对DPPH自由基的清除能力最强,总抗氧化能力次之,清除·OH能力和铁还原能力较差。  相似文献

为了避免食品电场加工中因电极使用而产生电化学反应造成食品污染和电极腐蚀的问题,采用交变磁通代替电极作为激励源,在果泥料液中诱导形成感应电场(induced electric field,IEF),实现对蓝莓果泥的IEF处理:分析激励电压、温度和时间对蓝莓果泥多酚氧化酶(polyphenol oxidase,PPO)和过氧化物酶(peroxidase,POD)活性及总花青素含量的影响。结果表明,100 V激励电压产生的IEF可减缓PPO和POD酶活的受热损失。150和250 V激励电压下,IEF与高温钝化PPO和POD具有协同作用。IEF可提高花青素分子的热稳定性且电场强度越高稳定作用越强。与未处理果泥相比,经85℃、250 V激励电压的IEF处理30 min的果泥总花青素含量得到最大提升(12. 55%),PPO和POD可被完全灭活。该条件下获得的蓝莓果泥表观黏度与鲜榨果泥更为接近,且储藏期内(4℃,8周)的菌落总数维持在1 lg(CFU/mL)以下,显著低于常规热处理(0 V)和未经热处理的果泥。研究为蓝莓果泥的IEF处理提供了数据参考。  相似文献
