The regulation of inhibin secretion has not been elucidated fully in male ruminants. The aim of this study was to determine the relative importance of FSH and testosterone concentrations, and FSH receptors, in the control of secretion of immunoactive inhibin in rams. In Expt 1, temporal changes in hormone concentrations and testicular FSH binding were determined for two groups of rams (n = 4) kept under opposite, alternating 4 month periods of long (16 h light:8 h dark) and short (8 h light:16 h dark) days. Testicular biopsies (1-2 g) were collected when the testes were regressed, redeveloping, redeveloped and regressing. In Expt 2, separate groups of rams (n = 4) kept under natural photoperiod (latitude 45 degrees 48 minutes N) were designated as controls or passively immunized (for 3 weeks) with sufficient oestradiol antiserum to increase testosterone secretion without altering LH and FSH; this was done when the testes were regressed (non-breeding season) and redeveloped (breeding season). In both groups of rams (Expt 1), 'seasonal' increases in FSH concentrations began a few weeks earlier than did increases in inhibin concentrations. FSH reached maximum concentrations during testicular recrudescence, whereas numbers of FSH receptors in the testis and circulatory inhibin concentrations did not reach peak values until the testes were fully developed. Numbers of FSH receptors per testis, but not FSH concentration, were positively correlated (r = 0.65) with inhibin concentrations across the four stages of the testicular cycle. Near the end of testicular recrudescence early in the breeding season (Expt 2), relatively high FSH concentration was associated with increased abundance of FSH receptor mRNA (90%) and number of receptors (45%) in the testis and increased inhibin concentrations (50%), compared with when the testes were regressed. Moderate, physiological increases in testosterone secretion in immunized rams did not affect inhibin in either season. These results indicate that: (i) FSH stimulation of immunoactive inhibin secretion by Sertoli cells as testes recrudesce is via increases in secretion (early) and cognate receptors (late); (ii) FSH upregulates the synthesis of its own receptor late in recrudescence; and (iii) the positive correlation (r = 0.70) observed between circulatory testosterone and immunoactive inhibin does not reflect a causal relationship.  相似文献

The aim of this study was to evaluate the distribution of inhibin/activin alpha, beta(A) and beta(B) subunits and follistatin in immature oocytes and in matured oocytes before and after IVF. Denuded oocytes were submitted to a whole-mount immunofluorescence procedure. Specimens were imaged and fluorescent intensities quantified by scanning laser confocal microscopy. Immunoreactivity for inhibin alpha subunit (both alpha(C) and pro-alpha regions), abundant in the ooplasm of immature oocytes, decreased after maturation (a 68% and 88% decrease, respectively; P < 0.001), but increased after IVF by 2- and 5.7-fold, respectively (P < 0.01). Intense staining for beta(A) was detected in immature oocytes (predominantly in the outer ooplasm and zona pellucida) but after maturation and fertilization it was localized mainly in the zona pellucida, perivitelline space and oolemma. Immunoreactivity for beta(A) in the ooplasm decreased by 58% after maturation (P < 0.001) but increased again by 75% after fertilization (P < 0.01). Immunoreactivity for beta(B) was localized mainly in the zona pellucida and did not change after maturation. However, immunoreactivity for beta(B) was not detected in the zona pellucida after fertilization, but remained unchanged in unfertilized oocytes. Immunoreactivity for follistatin was detected in the ooplasm and zona pellucida of immature oocytes but decreased progressively in the ooplasm after maturation (a 63% decrease; P < 0.001) and did not change after IVF. Examination of partially denuded cumulus-oocyte complexes confirmed abundant expression of alpha(C), pro-alpha, beta(A) and follistatin immunoreactivity in cumulus cells, whereas beta(B) subunit staining was weak or absent in cumulus cells, but intense in the zona pellucida. In conclusion, the present study shows that qualitative and quantitative changes in the distribution of inhibin/activin subunits and follistatin accompany oocyte maturation and fertilization. The possibility, indicated by these observations, that activin A and activin B may play distinct roles in bovine oocyte maturation and fertilization warrants further study.  相似文献

The aim of this study was to examine the function of granulosa cells and hormone concentrations in follicular fluid in bovine ovarian follicles during selection of the first dominant follicle. Ovaries were obtained from beef heifers on days 1-5 after ovulation: follicles > 4 mm in diameter were dissected and follicular fluid and granulosa cells were collected from individual follicles. Oestradiol production by granulosa cells after culture with testosterone was used to determine aromatase activity and responsiveness to gonadotrophins was determined by cAMP production after culture with FSH or LH. Concentrations of oestradiol, progesterone and insulin-like growth factor binding proteins (IGFBPs)-4 and -5 were measured in follicular fluid. Follicles were classified as largest or smaller (days 1 and 2), or dominant or subordinate (days 3-5). Aromatase activity was greater in granulosa cells from the largest follicle than in granulosa cells from smaller follicles on days 1, 3, 4 and 5 (P < 0.05). Responsiveness to LH was not detected in granulosa cells on day 1, but from day 2 to day 5 cells from the largest follicle were significantly more responsive than cells from smaller follicles (P < 0.05). Responsiveness to FSH was detected in granulosa cells from all follicles from day 1 onwards and did not differ between cells from the largest follicle or smaller follicles on any day. Follicular fluid concentrations of oestradiol and the ratio of oestradiol:progesterone were greater and concentrations of IGFBP-4 and -5 were lower in the largest follicle than in smaller follicles from day 2 to day 5 (P < 0.05). In conclusion, selection of the dominant follicle is associated with increased granulosa cell aromatase activity followed by increased cAMP response to LH and follicular fluid oestradiol concentrations, and decreased follicular fluid concentrations of IGFBP-4 and -5 within 2 days after ovulation.  相似文献

n-3 Polyunsaturated fatty acids (n-3 PUFA) are important for the normal development and functioning of all organisms. Mammals lack the n-3 fatty acid desaturase required for the synthesis of α-linolenic acid (18:3n-3), and are therefore dependent on dietary sources to obtain this essential fatty acid. Currently, the richest source of dietary long-chain n-3 PUFA, eicosapentaenoic acid (20:5n-3) and docosahexaenoic acid (22:6n-3), are triacylglycerides extracted from rapidly declining marine resources. The nematode Caenorhabditis elegans synthesizes a wide range of PUFA and possesses the only known example of an n-3 fatty acid desaturase enzyme in the animal kingdom. Transgenic mice expressing the C. elegans n-3 desaturase under the control of the lactation-induced goat β-casein mammary gland promoter were generated via pronuclear microinjection. Significant increases in n-3 PUFA, decreases in n-6 PUFA, and an overall decrease in the n-6:n-3 PUFA ratio were observed in the milk produced by transgenic mice. Neonate mice consuming milk from transgenic females accumulated increased levels of docosahexaenoic acid in their brains. This transgenic model may provide useful information to address some basic questions of neonatal nutrition, and demonstrates one of the steps that would be required to increase the n-3 PUFA content of milk and dairy products endogenously. Increasing the proportion of n-3 PUFA in milk fat would help to improve the nutritional composition of an important component of the North American diet.  相似文献

Recent studies suggest that IGF-I is a crucial regulatory factor in follicular growth during early post-partum period. The aim of the present study was to determine in detail the changing profiles of metabolic and reproductive hormones in relation to ovulation of the dominant follicle (DF) of the first follicular wave post-partum in high-producing dairy cows. Plasma concentrations of related hormones in 22 multiparous Holstein cows were measured from 4 weeks pre-partum to 3 weeks post-partum, and the development of DF was observed with colour Doppler ultrasound. Thirteen cows showed ovulation by 15.2 days post-partum. Anovulatory cows showed higher GH and lower IGF-I levels than those in ovulatory cows during the peri-partum period. Each DF developed similarly, and a clear blood flow in the follicle wall was observed despite ovulation or anovulation. In addition, detailed endocrine profiles were analyzed in 9 out of the 22 cows. Five cows showed an increase in plasma oestradiol-17beta (E2) with follicular growth followed by E2 peak, LH surge and ovulation. In these cows, plasma IGF-I concentrations remained high until 10 days post-partum followed by a gradual decrease. Subsequently, the insulin level increased together with the E2 peak towards ovulation. These profiles were not observed in anovulatory cows. In conclusion, our data strongly support the concept that IGF-I and insulin represent 'metabolic signals' of the resumption of ovarian function post-partum in high-producing dairy cows. Moreover, we provide the first visual evidence that both ovulatory and anovulatory DFs of the first follicular wave post-partum are similarly supplied with active blood flow.  相似文献

The primary objectives of this study were to follow the temporal patterns of testicular LH and FSH receptor (LH-R and FSH-R) concentrations and affinity (Ka) during sexual maturation in bulls and to see if such patterns could help explain the control of rapid testicular growth that occurs after 25 weeks of age, when serum gonadotropin concentrations are low. Separate groups of Hereford x Charolais calves (n = 6) were castrated every 4 weeks from 5 to 33 weeks of age and at 56 weeks of age. A week prior to castrations, from 5 to 33 weeks of age, blood was collected every 15 min for 10 h. The transition from indifferent supporting cells to Sertoli cells in seminiferous tubules was rapid between 13 and 25 weeks and rapid testis growth occurred after 25 weeks of age. Serum LH and FSH concentrations were transiently elevated at 12 weeks of age (P < 0.05). LH-R concentrations decreased from 13 to 25 weeks of age and increased to 56 weeks of age (P < 0.05). LH-RKa decreased from 9 to 17 weeks of age, increased to 29 weeks of age and declined to 33 weeks of age (P < 0.05). FSH-R concentrations declined from 17 to 25 weeks of age then increased to 56 weeks of age (P < 0.05). FSH-RKa increased from 17 to 25 weeks of age (P < 0.05). High concentrations of gonadotropins and their receptors may be critical to initiate testis growth postnatally and support it after 25 weeks of age in the face of low serum gonadotropin concentrations.  相似文献

Mature Holstein cows were drenched daily with either 500 mL of water (control; n = 28) or propylene glycol (PPG; n = 28) from d 10 before parturition until d 25 postpartum. Follicular development was monitored thrice weekly by transrectal ultrasound. Blood samples were collected every 30 min from a subset of 10 cows per treatment on d −10, 2, and 25 to assess glucose and insulin response to treatments, and on d 10 postpartum, blood was collected every 10 min for 12 h to determine LH pulse profiles. Both insulin and glucose were elevated on d 2 and 25 following PPG administration, but only insulin was elevated on d −10. On d 10 postpartum, the number of LH pulses, mean LH, and pulse amplitude were not different between control and PPG cows. The proportion of first postpartum dominant follicles that became ovulatory, atretic, or cystic was not different between control and PPG cows. Despite evidence of improved metabolic status, PPG failed to increase LH pulse frequency, and failed to increase the proportion of first postpartum follicle waves resulting in ovulation. The dominant follicle of each cow was retrospectively categorized as being ovulatory (n = 17), nonovulatory high estradiol (n = 6), nonovulatory low estradiol (n = 24), or cystic (n = 8). Differences in dry matter intake and energy balance among cows in the different follicle categories were apparent as early as 3 wk before parturition. The nonovulatory low estradiol cows had lower pre- and postpartum dry matter intake and energy balance compared with ovulatory cows. The nonovulatory low estradiol cows also had postpartum metabolic hormone and metabolite profiles indicative of more severe negative energy balance.  相似文献

The objective of this study was to evaluate the effect of body condition score (BCS), body weight (BW), average daily weight gain (ADG), and age at first calving (AFC) of Holstein heifers on production and reproduction parameters in the 3 subsequent lactations. The data set consisted of 780 Holstein heifers calved at 2 dairy farms in the Czech Republic from 2007 to 2011. Their BW and BCS were measured at monthly intervals during the rearing period (5 to 18 mo of age), and the milk production and reproduction data of the first 3 lactations were collected over an 8-yr period (2005 to 2012). The highest milk yield in the first lactation was found in the group with medium ADG (5 to 14 mo of age; 0.949 to 0.850 kg of ADG). The highest average milk yield over lifetime performance was detected in heifers with the highest total ADG (≥0.950 kg/d). The difference in milk yield between the evaluated groups of highest ADG (in total and postpubertal growth ≥0.950 kg/d and in prepubertal growth ≥0.970 kg/d) and the lowest ADG (≤0.849 kg/d) was approximately 1,000 kg/305 d per cow. The highest milk yield in the first lactation was found in the group with the highest AFC ≥751 d, for which fat and protein content in the milk was not reduced. Postpubertal growth (11 to 14 mo of age) had the greatest effect on AFC. The group with lowest AFC ≤699 d showed a negative effect on milk yield but only in the first 100 d of the first parity. The highest ADG was detrimental to reproduction parameters in the first lactation. The highest BW at 14 mo (≥420 kg) led to lower AFC. Groups according to BCS at 14 mo showed no differences in AFC or milk yield in the first lactation or lifetime average production per lactation. We concluded that low AFC ≤699 d did not show a negative effect on subsequent production and reproduction parameters. Therefore, a shorter rearing period is recommended for dairy herds with suitable management.  相似文献

We previously established a bovine experimental model showing that the corpus luteum (CL) does not appear following aspiration of the preovulatory follicle before the onset of LH surge. Using this model, the present study aimed to determine the profile of follicular development and the endocrinological environment in the absence of CL with variable nadir circulating progesterone (P(4)) concentrations during the oestrous cycle in cattle. Luteolysis was induced in heifers and cows and they were assigned either to have the dominant follicle aspirated (CL-absent) or ovulation induced (CL-present). Ultrasound scanning to observe the diameter of each follicle and blood collection was performed from the day of follicular aspiration or ovulation and continued for 6 days. The CL-absent cattle maintained nadir circulating P(4) throughout the experimental period and showed a similar diameter between the largest and second largest follicle, resulting in co-dominant follicles. Oestradiol (E(2)) concentrations were greater in the CL-absent cows than in the CL-present cows at day -1, day 1 and day 2 from follicular deviation. The CL-absent cows had a higher basal concentration, area under the curve (AUC), pulse amplitude and pulse frequency of LH than the CL-present cows. After follicular deviation, the CL-absent cows showed a greater basal concentration, AUC and pulse amplitude of growth hormone (GH) than the CL-present cows. These results suggest that the absence of CL accompanying nadir circulating P(4) induces an enhancement of LH pulses, which involves the growth of the co-dominant follicles. Our results also suggest that circulating levels of P(4) and E(2) affect pulsatile GH secretion in cattle.  相似文献

The response of Graafian follicles to pre-ovulatory surge levels of FSH and LH in vivo triggers the terminal differentiation of granulosa cells and oocyte maturation. In polyovular species, the LH-driven signalling uses the epidermal growth factor (EGF)-like ligands AREG, EREG and BTC to promote oocyte maturation and cumulus expansion. This experimental series used a physiologically relevant ovine in vitro maturation (IVM) system to evaluate the impact of exposure to pre-ovulatory levels (100 ng/ml) of LH and FSH on ovine cumulus cell expression of EGF-like ligands in vitro. The serum-free sheep IVM system supported high levels (91.4%) of gonadotrophin-induced maturation of cumulus-enclosed oocytes and embryo development to the blastocyst stage (34.5%). Results were equivalent to a serum-based IVM system (85.1% IVM, 25.8% blastocyst rate; P>0.05) but were significantly different (P<0.05) to serum-free medium without gonadotrophins (69.5% IVM; 8.0% blastocyst rate). Ovine BTC was cloned and sequenced. Gonadotrophin-induced AREG, EREG, BTC and EGFR expressions were quantified in cumulus and mural granulosa cells during IVM. A rapid induction of AREG expression was apparent in both cell types within 30 min of gonadotrophin exposure in vitro. LHCGR (LHR) was detected in mural cells and FSHR in both cumulus and mural granulosa cells. The data confirm the involvement of AREG and EGFR during gonadotrophin-induced cumulus expansion, oocyte maturation and the acquisition of developmental competence by sheep oocytes matured in vitro.  相似文献

Hypogonadism is a major manifestation of zinc deficiency in both humans and animals. In this study, we try to elucidate the main role of zinc in regulating the testicular function. Ninety male weanling albino rats were divided into three groups: control, zinc-deficient (ZD), and zinc-supplemented (ZS) groups. Six rats from each group were sacrificed every two weeks over a total period of ten weeks. Serum levels of follicle-stimulating hormone (FSH), luteinizing hormone (LH), prolactin (PRL) and testosterone (T) were estimated by radioimmunoassay technique. Serum zinc concentrations were also determined. Data revealed that serum zinc concentrations were significantly low in ZD group compared to both control and ZS groups. Normal FSH and LH but low PRL and T levels were demonstrated in sera of ZD group compared to both control and ZS groups. We came to the following conclusions: zinc deficiency has no effect on the pituitary gonadotropin output. Zinc deficiency affects the testicular function either directly through its effect on the testicular steroidogenesis and/or indirectly through its effect on the pituitary synthesis and/or secretion of PRL.  相似文献

Ten mares were studied from February (winter anoestrus) to their second ovulation in the breeding season to investigate the relationship between resumption of ovarian cyclicity in the spring and circulating concentrations of FSH, inhibin A and inhibin isoforms containing pro- and -alphaC immunoreactivity. An additional four mares were studied during one oestrous cycle. Growth and regression of ovarian follicles were monitored by transrectal ultrasonography. The frequency of blood sampling varied from three times a week to once a day, depending on the follicular activity present. Concentrations of FSH, oestradiol, inhibin A and pro- and -alphaC isoforms were low during deep winter anoestrus when minimal follicular activity was present in the ovaries. During spring transition, an increase in FSH concentration preceded the emergence of each follicular wave. Concentrations of inhibins were significantly higher (P < 0.05) during growth of anovulatory follicles in spring transition than during winter anoestrus. Plasma concentrations of oestradiol and inhibin A were significantly higher (P < 0.001, P < 0.05, respectively) during the growth of preovulatory follicles than during the growth of transitional anovulatory follicles, but concentrations of inhibin pro-alphaC isoforms did not differ between the two types of follicle. During the oestrous cycle, there was a significant inverse relationship (P < 0.001) between concentrations of FSH and the inhibins. Plasma inhibin pro-alphaC isoforms, but not inhibin A, reached a peak on the day of ovulation. The results strongly indicate that FSH regulates growth of spring anovulatory and preovulatory follicles. Inhibins are likely to contribute to negative feedback on the release of FSH from the pituitary gland both during the transitional period and the breeding season in mares.  相似文献

We aimed to test the effects of ad libitum feeding of whole milk (WM) or milk replacer (MR) versus restrictive feeding of MR during the first 4 wk of life on growth performance and on milk yield in the first lactation. We studied 57 German Holstein calves (29 females, 28 males) from birth until d 110 of life (trial 1). The 28 females from trial 1 were further studied during their first lactation (trial 2). In trial 1, all calves were randomly allocated at birth to 1 of 3 groups: MR-res [n = 20, 6.78 kg MR (11.5% solids)/calf per day], MR-ad lib (n = 17, 13.8% solids) or WM-ad lib (n = 20). All calves received colostrum ad libitum from their dam until d 3 of age. From d 4 to 27, calves were fed according to their group regimen. From d 28 to 55, all calves received MR-res feeding and were then gradually weaned until d 69. We recorded body weight (until d 110) and feed intake (amount, metabolizable energy, and frequency of liquid feed intake until weaning). We estimated the profitability of the different feeding regimens, taking into account income from milk yield (trial 2) and feed costs during rearing. In trial 1, the calves from WM-ad lib and MR-ad lib had total metabolizable energy intakes 2.02- and 1.65-fold greater than the MR-res group during the first 4 wk of life. During this period, concentrate intake did not differ among groups, but tended to be greater in WM-ad lib than in MR-ad lib calves from d 28 to 69. The MR-res calves visited the automatic feeders more often than the ad libitum-fed groups during differential feeding, but 70% of the visits were unrewarded (<10% in the ad libitum-fed calves). When all calves were fed at the MR-res level, the average proportion of unrewarded visits was 65% in all groups. Average daily gain and body weight were greater among MR-ad lib and WM-ad lib calves than among MR-res animals during the first 4 wk of life, but not from d 1 to 110. In trial 2, age at first calving, dry matter intake, and body weight over the first 10 mo of lactation were not different among groups, nor was milk composition. Milk yields (305 d) were numerically but not statistically greater in the ad libitum-fed groups during the first lactation (+765 kg for WM-ad lib vs. MR-res; +612 kg for MR-ad lib vs. MR-res). Feeding WM-ad lib and MR-ad lib was 1.37- and 1.21-fold more costly than MR-res, respectively, but amounted to 18, 15, and 13% of the total estimated feed costs until first calving in WM-ad lib, MR-ad lib, and MR-res, respectively. Our study confirms that ad libitum feeding is an attractive measure for rearing dairy calves, both for animal welfare and—with the caveat of a small sample size in trial 2 that led to insufficient power—economic profit from milk.  相似文献

Germ cell production and organization of the testicular epithelium in a prosimian species, the grey mouse lemur, Microcebus murinus, was investigated to extend knowledge of comparative primate spermatogenesis. In addition, semen samples collected from adult male lemurs (body weight 53-92 g; n = 16) by rectal probe electroejaculation were evaluated using computer-assisted morphometric and kinematic analysis of spermatozoa. Epididymidal spermatozoa were collected from six animals after hemicastration; the testes were weighed and prepared for stereological analysis and flow cytometry. The relative testis mass (as a percentage of body weight) ranged between 1.17 and 5.6%. Twelve stages of testicular seminiferous epithelium as described for macaques were applied and only a single stage was observed in most of the seminiferous tubule cross-sections. On average (mean SD), a single testis contained 1870 +/- 829 x 10(6) germ cells and 35 +/- 12 x 10(6) Sertoli cells. Germ cell ratios (preleptotene:type B spermatogonia = 2, round spermatid:pachytene = 3; elongated spermatid:round spermatids = 1) indicated high spermatogenic efficacy. Sperm head dimensions and tail lengths of the ejaculated and epididymidal spermatozoa were similar. Percentages of defects (neck/mid-piece and tail) were low ( 10%) and similar for ejaculated and epididymidal spermatozoa. Spermatozoa were highly motile, characterized by extensive lateral head displacement, but relatively low progressive motility. In conclusion, the grey mouse lemur has unusually large testes with a highly efficient spermatogenic process and large sperm output. These features, together with the high proportion of morphologically normal and highly motile spermatozoa in the ejaculates, indicate that Microcebus murinus is a species in which sperm competition after ejaculation is likely to occur. The predominantly single spermatogenic stage system seems to be an ancestral feature among primates.  相似文献

Using a mixed linear animal model, genetic parameters were estimated for clinical mastitis (MAST), lactation average somatic cell score (LSCS), and milk production traits in the first 3 lactations of more than 200,000 Swedish Holstein cows with first calving from 1995 to 2000. Heritability estimates for MAST (0.01 to 0.03) were distinctly lower than those for LSCS (0.10 to 0.14) and production traits (0.23 to 0.36). The genetic correlation between MAST and LSCS was high for all lactations (mean 0.70), implying that selection for low LSCS will reduce the incidence of mastitis. Undesirable genetic relationships with production were found for both MAST and LSCS with genetic correlations ranging from 0.01 to 0.45. This emphasizes the need for including udder health traits in the breeding goal. Dairy cows face metabolic challenges in the transition from late pregnancy to early lactation. The energy demands for the growing fetus and the onset of milk production are increasing but voluntary feed intake often decreases around parturition and cannot meet these demands. This energy balance, among others, can change the oxidative status. Oxidative stress occurs when antioxidant defense mechanisms are not sufficient to cope with the increasing generation of reactive oxygen species. Our objectives were to investigate (1) the effect of parity on the oxidative status of dairy cows (n = 247) in late pregnancy and early lactation; and (2) the effect of different inclusion rates of concentrate feeding (150 vs. 250 g/kg of energy-corrected milk) during early lactation on 2 farms including 87 cows in total. In addition, we aimed to compare the oxidative status across the 2 farms using equal portions of concentrate feeding. For these purposes, we measured concentrations of the derivatives of reactive oxygen metabolites (dROM) and the ferric reducing ability (FRAP) in serum on d −50, −14, +8, +28, and +100 relative to calving. Furthermore, we calculated the oxidative status index (OSi) as dROM/FRAP × 100. Data were analyzed using a linear mixed model. Cows in the first and second lactations had greater dROM, FRAP, and OSi than cows in their third and greater lactations. Hence, supporting the antioxidative side of the balance might be of particular importance in the first and second lactations. Feeding different amounts of concentrates did not affect dROM, FRAP, or OSi under our experimental conditions, suggesting that the relatively small differences in energy intake were not affecting the oxidative status. Comparing farms, cows from one farm were notable for having greater dROM and lower FRAP, resulting in a greater OSi compared with cows on the other farm. Milk yield showed a time by farm interaction with 7% less milk on d 100 on the farm with the greater OSi. Moreover, cows on that farm had 1.4-fold greater β-hydroxybutyrate concentrations. Our results emphasize the value of assessing oxidative status with regard to both the pro- and antioxidative sides, and support the association between oxidative and metabolic status. Further investigations are needed to determine the applicability of OSi as a prognostic tool during early lactation and to determine which factors have the greatest influence on oxidative status.  相似文献

Our objective was to determine the dose-response effects of calcium salts of palm fatty acids (CSPF) on nutrient digestibility and production responses of early-lactation dairy cows grazing on tropical pastures and to evaluate carryover effects throughout mid and late lactation. Forty multiparous dairy cows (Jersey × Holstein) with (mean ± standard error of the mean) 20 ± 1.69 kg of milk/d and 20 ± 5.0 d in milk were used in a randomized complete block design. During the treatment period, all cows were kept in a grazing system. The treatments were offered for 90 d (treatment period) and consisted of 4 increasing levels of CSPF: 0 (0 kg/d), 0.2 (0.2 kg/d), 0.4 (0.4 kg/d), and 0.6 (0.6 kg/d). Each treatment had 10 animals. Increasing CSPF from 0 to 0.6 kg/d replaced an equivalent amount of a corn-based concentrate supplement offered at 10 kg/d on an as-fed basis (8.96 kg/d as a dry matter basis). All cows were housed and received a diet without fat inclusion fed as total mixed ration once a day from 91 to 258 d of the experiment (carryover period). During the treatment period, increasing CSPF linearly decreased dry matter intake (1.20 kg/d), linearly increased neutral detergent fiber digestibility (3.90 percentage units), and quadratically increased total fat digestibility (6.30 percentage units at 0.4 kg/d CSPF). Increasing CSPF linearly increased the yields of milk (4.10 kg/d), milk fat (0.11 kg/d), milk lactose (0.19 kg/d), energy-corrected milk (ECM; 3.30 kg/d), and feed efficiency (ECM/dry matter intake, 0.34 kg/kg), and linearly decreased milk protein content (0.38 g/100 g), body weight change (0.05 kg/d), and body condition score (0.37). We observed interactions between CSPF and time during the carryover period. Overall, CSPF supplementation linearly increased or tended to increase milk yield until 202 d of the experiment with a similar pattern observed for all the other yield variables. In conclusion, supplementing CSPF from 0 to 0.6 kg/d during 90 d increased neutral detergent fiber and total fat digestibility and the yields of milk, milk fat, and ECM in early-lactation dairy cows grazing on tropical pastures. Most production measurements linearly increased during the treatment period, indicating that 0.6 kg/d CSPF was the best dose. Also, supplementing CSPF from 0 to 0.6 kg/d for 90 d during early lactation had positive carryover effects across mid and late lactation.  相似文献
