Background The pathogenesis of diabetic nephropathy (DN) is a complex pathophysiological process.Its precise mechanism is not fully known. In recent years it has been recognized that synthesis of various extracelluar matrix (ECM) components may increase, and that degradation of ECM may decrease in DN. It was reported heparin could inhibit mesangial cells proliferation in vitro. The main aim of this study is to explore whether heparin inhibits proliferation of mesangial cells grown in high glucose concentration and to measure the effect of heparin on matrix metalloproteinases (MMPs) expression in mesangial cells. Methods The medium contained either low glucose (5 mmol/L) or high glucose (25 mmol/L). The concentrations of heparin in the culture medium were 0, 25, 50,100, 200 or 400 μg/mL. A metabolic (WST-1) assay was used to measure mesangial cell proliferation and Western blot analysis was used to measure MMPs expression of mesangial cells. Results Normal human mesangial cell (NHMC) proliferation was higher in high glucose (HG) medium than in low glucose (LG) medium. They showed a 1.93 fold expansion after 72 h in high glucose in contrast to a 1.63 fold expansion in low glucose. In the presence of heparin, mesangial cells proliferation was inhibited, which was more obvious at high glucose concentrations than at low glucose concentrations. In high glucose, with heparin concentration of 50, 100, 200 and 400 μg/mL, the mesangial cells showed a 0. 61 fold, 0.52 fold, 0.52 fold and 0.41 fold reductions in cell number compared to cells grown without heparin. In low glucose, only concentrations of 200 μg/mL and 400 μg/mL showed reduction in cell number, namely 0.54 fold and 0.45 fold, when compared to cells grown without heparin. In Western blot analysis,MMP1, MMP2, MMP3 and MMP9 was expressed by mesangial cells expressed in both high and low glucose concentrations, which was more prominent in high glucose medium. Incubation of heparin further increased expression of MMP1, MMP2, MMP3 and MMP9. Conclusions This study suggests that glucose can accelerate mesangial cell proliferation while heparin can reduce proliferation, being more obvious at high glucose concentrations. Higher glucose concentrations led to increased MMP expression, which may take part in the regulation of mesangial matrix synthesis and degradation. Addition of heparin resulted in a corresponding increase in MMP expression, most notably at high glucose concentrations, indicating a potentially renoprotective role in DN. 相似文献

研究柠檬酸镧对肝癌细胞HepG2失巢凋亡的影响.采用聚羟乙基异丁烯酸阻断细胞与胞外基质联系的方法,建立体外细胞失巢生长模型.在贴壁和失巢条件下,用浓度0.1、0.01和0.001 mmol/L的柠檬酸镧溶液处理细胞.对于贴壁细胞,用碘化丙啶(propidium iodide,PI)染色检测细胞周期和凋亡率.对于失巢细胞,运用AnnexinV FITC-PI双染法检测失巢凋亡,用线粒体膜电位检测试剂盒JC-1(5,5′,6,6′-tetrachloro-1,1′,3,3′-tetraethyl-imidacarbocyanine iodide)荧光显微镜法检测线粒体膜电位.结果表明,在贴壁条件下,柠檬酸镧处理组的周期和凋亡率与对照组相比,不存在明显差异.而在失巢条件下,柠檬酸镧促进HepG2细胞失巢凋亡,降低了线粒体膜电位.结果显示,稀土元素可能具有作为新的抗癌药物的潜力. 相似文献

Physical model tests have been conducted by various researchers to investigate fault rupture propagation and ground deformation induced by bedrock faulting. However, the effects of pre-existing fracture on ground deformation are not fully understood. In this work, six centrifuge tests are reported to investigate the influence of pre-existing fracture on ground deformation induced by normal faulting in sand, clay and nine-layered soil with interbedded sand and clay layers. Shear box tests were conducted to develop a filter paper technique, which was adopted in soil model preparation to simulate the effects of pre-existing fracture in centrifuge tests. Centrifuge test results show that ground deformation mechanism in clay, sand and nine-layered soil strata is classified as a stationary zone, a shearing zone and a rigid body zone. Inclination of the strain localization is governed by the dilatancy of soil material. Moreover, the pre-existing fracture provides a preferential path for ground deformation and results in a scarp at the ground surface in sand. On the contrary, fault ruptures are observed at the ground surface in clay and nine-layered soil strata. 相似文献

为明确不同提取方法获得的玉米麸皮多糖对人结肠癌HT-29细胞增殖的抑制作用,采用MTT、倒置显微镜观察、分裂指数、集落形成方法测定细胞活力.MTT实验结果表明,水提玉米麸皮多糖可抑制结肠癌HT-29细胞的生长,随质量浓度和处理时间的增加细胞抑制率增加,0.02 g/L的多糖处理48 h其抑制率可达60.62%.另外两种多糖对HT-29细胞无明显的抑制效果.形态学观察结果表明,水提玉米麸皮多糖作用48 h,HT-29细胞有明显的形态学改变.同时,水提玉米麸皮多糖能够抑制HT-29细胞有丝分裂数和集落形成率,并呈时间和剂量效应. 相似文献

Objective Study on the effect of NF-κB in ox-LDL injured HUVEC-304 and the intervention with the serum of Tongmai Decoction. Methods Tongmai Decoction herbage-contained serum was made by the serum pharmacology methods, then the HUVEC-304 cells were divided into 5 groups and incubated for 24 h: 1) normal group;2) fetal calf serum group; 3) ox-LDL group; 4) simvastatin group; 5) Tongmai Decoction group. RT-PCR was used to determine the mRNA of NF-κB in the cells, Western Blot Assay was used to determine the protein content of NF-κB in the cells. Results 1) Compared with the cells in normal group and serum control group , the endothelial cells in ox-LDL group are seriously injured, while those in Tongmai Decoction group grow better; 2) Compared with the cells in normal group and fetal calf serum group, the expressions of NF-κB gene and protein are up-regulated in ox-LDL group(P<0.05, P<0.01); 3) Compared with the cells in ox-LDL group, the expression of NF-κB gene and protein in Tongmai Decoction group are significantly down-regulated, there is no difference between Tongmai Decoction group and simvastatin group(P>0.05). Conclusion Tongmai Decoction can protect the injured endothelial cells, up-regulate the expressions of NF-κB gene and protein content. 相似文献

To explore the effect of puerarin on insulin receptor (IR), insulin receptor substrate-1 (IRS-1) and protein expression of protein kinase B(PKB) in the P13K pathway of the glucose consumption, transportation and insulin signal transduction in 3T3-L1 adipocytes with insulin resistance. The insulin resistance 3T3-L1 adipocytes model was established by free fatty acid induction. The model cells were managed with puerarin in different concentrations. Glucose consumption was detected with glucose oxidase method, glucose transportation rate was determined by 2-deoxy-3H glucose ingesting method, and the IR, IRS-1 and PKB expression were determined by Western blot. Glucose consumption and transportation were significantly decreased in the model adipocytes, but increased after treated with puerarin (P<0.01). Moreover, the level of tyrosine phosphorylation of IR subunit β was higher in the puerarin treated groups, and that of IRS-1 was higher in the group treated with low dose puerarin than that in the model group. The 3T3-L1 adipocytes of insulin resistance model could be induced by free fatty acid successfully, puerarin could promote the glucose utilization in them to alleviate the insulin resistance, which may be related with the action in advancing the tyrosine phosphorylation of IR and IRS-1. 相似文献

对人膀胱癌T24细胞进行体外培养,采用MTT方法观察不同浓度人参皂苷Compound K处理细胞24、48、72h后对细胞增殖的抑制作用,利用Hoechst33342对细胞进行染色,荧光显微镜观察细胞形态学变化,应用流式细胞术观察人参皂苷Compound K对T24细胞凋亡的诱导作用,并应用WesternBlotting检测不同浓度人参皂苷Compound K处理后T24细胞中caspase-3及相关蛋白的表达情况。结果显示,人参皂苷Compound K对膀胱癌T24细胞的增殖具有较强的抑制作用,呈浓度和时间依赖关系,并诱导肿瘤细胞凋亡,同时caspase-3活化型表达量增加。说明人参皂苷Compound K能够显著抑制膀胱癌T24细胞的活力,有效地诱导T24细胞凋亡。 相似文献

基于线性计算结果讨论不同阻尼矩阵对高土坝地震反应计算误差的影响。首先,通过理论分析,说明了土石坝地震反应分析采用常用阻尼模型的局限性。然后采用不同特征频率建立土坝的有限元模型的不同阻尼矩阵,分别采用显式及隐式算法对高土坝进行时域内的线性地震反应计算。最后,和频域内计算结果进行对比,分析了不同阻尼矩阵建模方式对计算结果精度的影响。 相似文献

Stable and single-dispersed hydroxyapatite (HAP) nanoparticles were synthesized with ultrasonic-assisted method. HAP nanoparticles were characterized by dynamic light scattering, XRD (X-ray diffraction) and TEM (Transmission Electron Microscopy). The effect of HAP nanoparticles on the K562 human myelogenous leukemia cell line was investigated by MTT assay and cell count test, and the mechanism was studied through the changes of cell cycle and ultrastructure. The results showed that HAP nanoparticles inhibited the proliferation of K562 cells dramatically in vitro. HAP nanoparticles entered the cytoplasm of K562 cells and the cells were arrested at G/M phase, thus, the cells died directly. 相似文献

为系统研究焊接参数的变化对焊缝成形和熔深的影响规律,探索脉冲激光焊接最佳工艺参数,采用脉冲激光工艺对SiCp/6061Al复合材料进行焊接试验研究。结果表明,脉冲激光焊接过程中,激光功率、脉冲频率、焊接速度、离焦量等参数的变化,均能对焊缝成形和熔深产生显著影响,进而影响焊缝的微观组织和宏观的拉伸强度。其中激光功率和焊接速度是最主要的影响因素。 相似文献

The in situ synthesis method for titanium matrix composites (TMCs) has obvious technical and economical advantages over other traditional methods. Ultrafine reinforcement particles were formed in situ by chemical reaction between elements or between elements and compounds. Using the approach, contamination at the composite matrix/reinforcement particle interface did not occur, interface bonding was good, and the reinforcement particle was thermodynamically stable. The stage of development of the preparation process for in situ TMCs as well as the thermodynamic analysis of the possible in situ reaction systems was described. 相似文献
