Ganoderma tsugae Murrill was available in the form of mature and baby Ling chih, mycelia and fermentation filtrate. From these four forms, methanolic extracts were prepared and their antioxidant properties were studied. Methanolic extracts from mature and baby Ling chih showed high antioxidant activities (96.8% and 93.6%) at 20 mg ml−1, and had EC50 values of 0.53 and 1.11 mg ml−1, respectively. EC50 values in reducing power were 5.00, 2.28, 0.93 and 2.15 mg ml−1 for Ling chih, baby Ling chih, mycelia and filtrate, respectively. Methanolic extracts from mature and baby Ling chih scavenged 1,1-diphenyl-2-picrylhydrazyl radicals by 88.4% and 93.8% at 5 mg ml−1, whereas those from mycelia and filtrate scavenged by 85.7% and 79.3% at 10 mg ml−1, respectively. EC50 values in chelating ability on ferrous ions were 4.82, 3.05, 1.10 and 3.41 mg ml−1 for Ling chih, baby Ling chih, mycelia and filtrate, respectively. Total phenols were the major naturally occurring antioxidant components found in all methanolic extracts and in the range of 24.0–35.6 mg g−1. Based on EC50 values, G. tsugae was good in antioxidant properties except for the scavenging ability on hydroxyl radicals.  相似文献

Ganoderma tsugae Murrill are currently popular and used in the formulation of nutraceuticals and as functional foods. The non-volatile components in the form of mature and baby fruit bodies (Ling chih), mycelia and fermentation filtrate from submerged culture were studied. Mycelia and filtrate contained significantly higher moisture contents (10.3% and 19.8%) and higher contents of carbohydrates, reducing sugars, crude ash and crude protein. Four forms of G. tsugae contained from 7.65% to 10.1% dry weight of total soluble sugars and polyols. Total free amino acid contents ranged from 2.50 to 149 mg g−1 dry weight and in the descending order of filtrate, mycelia, baby Ling chih and Ling chih. Contents of monosodium glutamate-like components ranged from 0.16 to 26.0 mg g−1 whereas, contents of sweet components ranged from 0.50 to 24.6 mg g−1. The bitter components were predominant. Contents of total and flavour 5-nucleotides were high in filtrate (5.48 and 3.10 mg g−1, respectively). The umami intensities were expected to be in the descending order of filtrate, mycelia, baby Ling chih and Ling chih.  相似文献

Ganoderma tsugae Murrill (Ganodermataceae) were available in the form of mature and baby Ling chih, mycelia and fermentation filtrate. From these four forms, hot water extracted and hot alkali extracted polysaccharides were prepared and their antioxidant properties were studied. Polysaccharides showed good antioxidant activity as evidenced by their particularly low EC50 values (<0.1 mg/ml). At 20 mg/ml, both extracted polysaccharides from mycelia showed reducing powers of 0.41–0.52 whereas reducing powers of other polysaccharides were in the range of 0.87 to 1.14. At 20 mg/ml, scavenging abilities on 1,1-diphenyl-2-picrylhydrazyl radicals increased to 93.7–100%, except for that of the hot water extracted polysaccharide, from filtrate, being 74.9%. At 20 mg/ml, scavenging abilities of both extracted polysaccharides from mycelia on hydroxyl radicals were 13.9 and 24.4%, respectively whereas scavenging abilities of the other polysaccharides were in the range of 39.0–55.2%. At 10 mg/ml, the chelating abilities of polysaccharides from mature and baby Ling chih, mycelia and filtrate were 93.9–100%, 97.6–100%, 85.1–88.0% and 51.2%, respectively. Overall, both extracts of polysaccharides possessed good antioxidant properties except for scavenging ability on hydroxyl radicals and can be developed as a new dietary supplement and functional food.  相似文献

Agrocybe cylindracea (DC: Fr.) Mre. (Bolbitiaceae) was available in the form of fruit bodies, mycelia and fermentation filtrate. From these three forms, methanolic extracts were prepared and their antioxidant properties studied. The methanolic extract from fruit bodies showed high inhibitory ability on lipid oxidation (90.0-97.3%) at 5-20 mg/ml and had an EC50 value of 0.94 mg extract/ml. EC50 values in reducing power were 2.36, 6.89 and 5.00 mg extract/ml for fruit bodies, mycelia and filtrate, respectively. The scavenging ability of the methanolic extract from fruit bodies on 1,1-diphenyl-2-picrylhydrazyl radicals was 89.0% at 1 mg/ml whereas those from mycelia and filtrate was 91.4% and 94.9% at 10 mg/ml, respectively. The chelating abilities of methanolic extracts from fruit bodies, mycelia and filtrate on ferrous ions were 90.6%, 84.6% and 96.3% at 5 mg/ml, respectively. Total phenols were the major detected naturally occurring antioxidant components found in methanolic extracts from A. cylindracea and in the range of 15.55-23.47 mg/g. Based on EC50 values, A. cylindracea was good in antioxidant properties, except for the scavenging ability on hydroxyl radicals.  相似文献

For the first time, extracts from clove buds obtained by supercritical carbon dioxide extraction were screened for antioxidant and antibacterial activities. Additionally, antioxidant and antibacterial activities of extracts obtained by the supercritical extraction of the clove bud–oregano leaf mixtures were studied. Supercritical extract of pure clove had the highest eugenol (64%) and total phenolic content (530.56 mg GAE/gextract). All extracts had antioxidant activity comparable to synthetic antioxidants against 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals and formation of peroxides. Presence of 0.6% and 5% of oregano extract in the clove extracts obtained from the clove–oregano plant mixtures improved their antioxidant activity with respect to the extract from pure clove. Clove extract showed moderate antibacterial activities against selected Staphylococcus and Enterococcus bacterial strains. Presence of 50% of the oregano extract improved antibacterial activity of clove extract against all tested strains and resulted in a synergistic antibacterial activity against Methicillin-resistant Staphylococcus haemolyticus strain (MIC ? 1.25 μg/mL). Study demonstrated great potential of supercritical clove extract as natural functional ingredient and the possibility of increasing its antioxidant and antibacterial efficiencies in order to apply lower concentrations and to reduce undesirable flavour notes and toxicological effects in final products.  相似文献

Pleurotus citrinopileatus Sing. (Lentinaceae) was successfully cultivated and commercially available in Taiwan. The ethanolic, cold and hot water extracts were prepared from P. citrinopileatus fruit bodies, mycelia and fermentation filtrate and their antioxidant properties studied. For three samples, ethanolic extracts exhibited higher antioxidant activities than water extracts. Reducing powers of three extracts from fruit bodies were effective and 1.03-1.10 at 5 mg/ml. With regard to EC50 values of scavenging abilities on 1,1-diphenyl-2-picrylhydrazyl radicals, the effectiveness was in a descending order: ethanolic>hot water>cold water extracts. Scavenging abilities of water extracts from three samples on hydroxyl radicals were 53.4-80.1% at 20 mg/ml. Chelating abilities of cold and hot water extracts on ferrous ions were higher than those of ethanolic extracts. Contents of total phenols were in the descending order: fruit bodies (8.62-12.38 mg/g)>mycelia (5.84-7.85 mg/g)>filtrate (4.80-5.57 mg/g). Overall, three extracts from fruit bodies were more effective in antioxidant properties assayed than those from mycelia and filtrate. Ethanolic extracts were more effective in antioxidant properties assayed, except for scavenging abilities on hydroxyl radicals.  相似文献

Chestnut (Castanea sativa) burs (CB) were processed by solvent extraction (with ethanol, toluene–ethanol or n-hexane) and/or isothermal autohydrolysis, and the potential as nutraceutics of the solubilized fractions of dietary fiber and antioxidants was evaluated. Among the studied alternatives, the most suited consisted on ethanol extraction, which yielded 21.0 g gallic acid equivalents (GAE)/100 g ethanolic CB extract, with EC50,DPPH = 0.38 g/L. Ethanol-extracted CB were treated with water at 120 °C, and the soluble fraction was mixed with ethanol to precipitate 8.8 g soluble fiber/100 g CB, with a Water Retention Capacity of 2.4 g/g and an oil absorption capacity of 23.7 g/g. The mother liquors from the above precipitation stage were concentrated and extracted with ethyl acetate to yield an organic phase containing soluble phenolics with high antioxidant activity (EC50,DPPH = 0.103 g/L) at a yield of 7 g/100 g CB. When the aqueous phase before the ethyl acetate extraction was treated with sulfuric acid, an additional antioxidant fraction (EC50,DPPH = 0.395 g/L) was obtained. The properties and characteristics of these extracts produced make them good candidates to be used as active ingredients.  相似文献

In the current study, an efficient method had been developed to acquire the antioxidant hydrolysate of monkfish muscle protein (MPH) using trypsin by an orthogonal (L9(3)4) test. Under the optimum conditions of enzymolysis time 4 h, enzyme-to-substrate ratio (E/S) 2%, enzymolysis temperature 40 °C and pH 8.0, the DH (Degree of hydrolysis) and hydroxyl radical scavenging activity of MPH reached 19.83 ± 0.82% and 58.05 ± 3.01%, respectively. By using ultrafiltration, gel filtration chromatography and reversed phase high performance liquid chromatography (RP-HPLC), three antioxidant pentapeptides were isolated from MPH, and their amino acid sequences were identified as Glu-Trp-Pro-Ala-Gln (MPH-P1), Phe-Leu-His-Arg-Pro (MPH-P2), and Leu-Met-Gly-Gln-Trp (MPH-P3) with molecular weights of 629.68 Da, 668.80 Da, and 633.77 Da, respectively. MPH-P1, MPH-P2, and MPH-P3 exhibited good scavenging activities on hydroxyl radical (EC50 0.269, 0.114 and 0.040 mg/ml), DPPH radical (EC50 2.408, 3.751, and 1.399 mg/ml), and superoxide anion radical (EC50 0.624, 0.101, and 0.042 mg/ml) in a dose-dependent manner. MPH-P3 was also effective against lipid peroxidation in the model system. The antioxidant activities of MPH-P1, MPH-P2, and MPH-P3 were due to their small sizes and the presence of antioxidant and hydrophobic amino acid residues within their sequences. The results of this study suggested that the protein hydrolysate and/or its isolated peptides might be effectively used as food additives for retarding lipid peroxidation occurring in foodstuffs.  相似文献

Partially purified polysaccharides (FP) and hot alkali extract (FNa) obtained from fruiting bodies of the wild basidiomycete Grifola frondosa were examined for their antimicrobial, antioxidant and cytotoxic activity. The structural properties of FP and FNa samples were investigated by FT-IR and high resolution 1H- and 13C-NMR spectroscopy. From a group of various G − and G + bacteria the antibacterial effects were highest against the G + B. cereus. FNa was the better antioxidant shown by the lower EC50 values of DPPH scavenging ability, ferric-reducing antioxidant power and ferrous ion-chelating ability. Ferric-reducing antioxidant power and ferrous ion-chelating ability were mostly linked to total polysaccharides, total- and β-glucan content, as well as total protein content. Both extracts displayed a moderate dose dependent antiproliferative action towards malignant human breast cancer MDA-MB-453, cervical adenocarcinoma HeLa and myelogenous leukemia K562 cells not observed in the non cancer derived MRC-5 fibroblasts. The highest effect was found in HeLa cells for FP extract. The mean diameter of Ca-alginate bead loading FP was 960.7 μm while the mean diameter of beads encapsulating FNa extract was 1051.7 μm.  相似文献

The fruits of Pilosocereus arrabidae (Cactaceae) is found in the Grumari shoal, located in the Rio de Janeiro state, Brazil and, several studies have already highlighted the importance of consuming its fruits. This work aims to investigate the physical, mineral and physicochemical properties of its fruits as well as to establish the knowledge about their chemical constituents and antioxidant properties. The peel (Pe) and pulp (Pu) extracts were obtained by maceration with the following solvents: hexane (HX), dichloromethane (DCL), ethyl acetate (EAC) and ethanol 70% (ET). The extracts were analyzed by Gas Chromatography coupled to Mass Spectrometry (GC–MS) and, by High Performance Liquid Chromatography (HPLC–DAD and HPLC–MS) for its chemical investigation. For the antioxidant activity investigations the ORAC, DPPH and ferrous ion chelating capacity (FICC) tests were performed. As results, we found higher yields for peels (72%) compared to pulps (28%). By the physical–chemical analyses we point out the fruits as a good source of fiber (pulp: 6.01 mg/100 g; peel: 8.02 mg/100 g). The minerals were analyzed by the method of issuing flames and indicated high levels of selenium (DRI for pulp and peel: 147%) and manganese (DRI pulp: 97.69% and DRI peel: 269.56%). The total flavonoid contents of the fruits performed by HPLC–DAD presented 0.45 μg equiv. in quercetin/mL of peel EAC extract and 0.25 μg equiv. in rutin/mL of pulp EAC extract. The antioxidant activities by the ORAC, FICC and DPPH methods indicated that the ET extracts showed antioxidant activities above the standards adopted for the tests. Among these, we highlight the ET extract of the pulp with EC50 of 17.57 ± 0.27 μg/mL, lower than Ginkgo biloba EGB761® (23.40 ± 0.04 mg/mL). By the FICC test the EAC extract of the peel and pulp showed 70.0% and 53.4% activity, respectively, at 500 mg/mL, higher than the standard quercetin (50.0%). By the HPLC–DAD and HPLC–MS methods there were detected, for the first time on this species, the presence of the following flavonoids on the EAC extracts: quercetin, rutin, catechin, dihydrokaempferol, quercetin 3 or 4′-O-glucoside, kaempferol and isorhamnetin. By the GC–MS analysis there were detected on the DCL extracts saturated fatty acids (palmitic and stearic) and, on the HX extracts, methylated sugars (peel) and menthol (pulp). To sum up, the fruits of P. arrabidae display antioxidant potential correlated to flavonoid presence, and, high levels of selenium, manganese and fibers, characteristics that can promote beneficial effects on human health.  相似文献

The antioxidant activities of brown pigment, extract of n-hexane and extract of supercritical carbon dioxide extraction of black sesame seeds were investigated in this study. Kinetics of anti-radical activity showed that the reaction between DPPH and brown pigment of sesame seed was rapid and reached the steady state in 10 min. Extracts from supercritical carbon dioxide extraction and n-hexane extraction reacted with DPPH slowly and the absorbance became stable after 35 min. However, α-tocopherol and trolox were rapid while the kinetic behaviour of BHA was intermediate. The brown pigment of sesame seed also showed a lower EC50 (13.5 μg ml−1) than the other two extracts and α-tocopherol, which was about 7–10 fold of the antioxidant activity of the supercritical carbon dioxide extract and the n-hexane extract. The ferric thiocyanate (FTC) method also showed that the brown pigment of sesame seed provided higher inhibition activity against lipid peroxidation at 200 μg ml−1 than did the supercritical carbon dioxide extract an n-hexane extract at 1 mg ml−1. However, the activities of the supercritical carbon dioxide extract and n-hexane extracts were significantly higher than that of α-tocopherol. In the linoleic acid system, the brown pigment of black sesame seed showed an equal antioxidant activity to BHA and higher than trolox and α-tocopherol. The results indicated that the brown pigment of sesame seed possessed excellent antioxidant activity.  相似文献

Agrocybe cylindracea (DC: Fr.) Mre. was available in the form of fruit bodies, mycelia and fermentation filtrate. From these three forms, hot-water extracts were prepared and their antioxidant properties were studied. Antioxidant activities of hot-water extracts from fruit bodies, mycelia and filtrate were 63.6%, 81.6% and 56.8% at 20 mg ml−1, respectively. EC50 values in reducing power were 2.72, 3.97 and 3.09 mg ml−1 whereas those in scavenging abilities of 1,1-diphenyl-2-picrylhydrazyl radicals were 0.62, 1.66 and 0.82 mg ml−1 for fruit bodies, mycelia and filtrate, respectively. At 20 mg ml−1, the scavenging abilities of hydroxyl radicals were 80.1%, 57.0% and 54.3% for fruit bodies, mycelia and filtrate, respectively. With regard to EC50 values in chelating abilities on ferrous ions, the hot-water extract from filtrate was better than that from mycelia. Total phenols were the major naturally occurring antioxidant components found in hot-water extracts and in the range of 23.74–30.16 mg g−1. From EC50 values obtained, it can be concluded that hot-water extracts from three forms of A. cylindracea were good in antioxidant properties.  相似文献

Mushrooms have become attractive as functional foods and as a source of physiologically beneficial bioactive compounds. Herein, we describe and compare the chemical constituents (phenolic compounds, macronutrients, sugars, fatty acids, tocopherols and ascorbic acid) of four wild edible mushrooms widely appreciated in gastronomy: Armillaria mellea (Vahl) P. Kumm., Calocybe gambosa (Fr.) Donk, Clitocybe odora (Fr.) P. Kumm., Coprinus comatus (O.F. Müll.) Pers. Furthermore, the antioxidant activity of their water soluble polysaccharidic and ethanolic fractions was studied by three different in vitro assays. C. comatus revealed the highest concentrations of sugars (43.23/100 g dry weight), PUFA (77.46%), phenolic compounds (45.02 mg/kg), tocopherols (301.03 μg/100 g) and, among all of the fractions tested, its ethanolic fraction showed the highest antioxidant activity (EC50 < 2.6 mg/ml). C. odora revealed one of the highest ascorbic acid (172.65 mg/100 g) contents and its water soluble polysaccharidic fraction showed the best antioxidant properties (EC50 < 3.6 mg/ml) among the polysaccharidic fractions. The studied mushrooms species could potentially be used in well-balanced diets and as a source of bioactive compounds.  相似文献

Passion fruit seeds are currently discarded on the pulp processing but are known for their high piceatannol and scirpusin B contents. Using pressurized liquid extraction (PLE), these highly valuable phenolic compounds were efficiently extracted from defatted passion fruit bagasse (DPFB). PLE was performed using mixtures of ethanol and water (50 to 100% ethanol, w/w) as solvent, temperatures from 50 to 70 °C and pressure at 10 MPa. The extraction methods were compared in terms of the global yield, total phenolic content (TPC), piceatannol content and the antioxidant capacity of the extracts. The DPFB extracts were also compared with those from non-defatted passion fruit bagasse (nDPFB). Identification and quantification of piceatannol were performed using UHPLC–MS/MS. The results showed that high TPC and piceatannol content were achieved for the extracts obtained from DPFB through PLE at 70 °C and using 50 and 75% ethanol as the solvent. The best PLE conditions for TPC (70 °C, 75% ethanol) resulted in 55.237 mg GAE/g dried and defatted bagasse, whereas PLE at 70 °C and 50% ethanol achieved 18.590 mg of piceatannol/g dried and defatted bagasse, and such yields were significantly higher than those obtained using conventional extraction techniques. The antioxidant capacity assays showed high correlation with the TPC (r > 0.886) and piceatannol (r > 0.772). The passion fruit bagasse has therefore proved to be a rich source of piceatannol and PLE showed high efficiency to recover phenolic compounds from defatted passion fruit bagasse.  相似文献

Citharexylum solanaceum is a native fruit from Brazil, which both bioactive compounds and antioxidant potential were not yet investigated. Thus, the freeze-dried extracts of seed and pulp + skin of C. solanaceum fruits were obtained after solid-liquid extraction with ethanol and their bioactive compounds composition, namely phenolic compounds and carotenoids, were determined. The antioxidant capacity of both extracts against physiologically relevant reactive oxygen species (ROS) and reactive nitrogen species (RNS) was further investigated. Both C. solanaceum extracts showed high contents of phenolic compounds; however, pulp + skin extract presented 2.4-times more phenolic compounds (33.54 mg/g) than the seed extract (14.09 mg/g). Verbascoside (phenylpropanoid) was the major compound identified in both extracts (11–25 mg/g). Regarding the carotenoid composition, all-trans-lutein (14–42 μg/g) and all-trans-β-carotene (13–44 μg/g) were the major compounds in both extracts. The high content of phenolic compounds and carotenoids in pulp + skin extract might explain its higher scavenging capacity against all the ROS/RNS as compared to seed extract. In general, both extracts showed better scavenging capacity for the RNS than for the ROS. Our results indicate that C. solanaceum fruits can be explored as an important natural source of antioxidant compounds.  相似文献

Phenolic compounds, antioxidant and antibacterial activities of defatted Moringa oleifera seed flour (DMF) were investigated. The total phenolic and flavonoid contents were measured using colorimetric methods. Free phenolic acid and flavonoid profiles were analyzed by high performance liquid chromatography, while antioxidant capacities were evaluated using scavenging assays of 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), ferric reducing antioxidant power and total antioxidant capacity. The results showed that extractability of phenolic compounds was significantly higher (p < 0.05) in bound phenolic extract (4173.00 ± 32.22 mg gallic acid equivalents (GAE)/100 g) than in free phenolic extract (780.00 ± 14.2 mg GAE/100 g) and it showed higher antioxidant and antimicrobial activities. The IC50 value for DPPH radical scavenging activity was 0.9 ± 0.05 and 14.9 ± 0.07 mg/mL for bound phenolic and free phenolic extracts, respectively. Bound phenolic extract was more effective (minimum inhibitory concentration (MIC), 0.06–0.157%) than free phenolic extract (MIC, 0.117–0.191%) against tested bacteria. Ten phenolic compounds (gallic acid, p-coumaric acid, ferulic acid, caffeic acid, protocatechuic acid, cinnamic acid, catechin, epicatechin, vanillin and quercetin) were identified and quantified in both extracts. These natural plant phenolics from Moringa seeds could be a good source of antioxidants and antibacterials for food and pharmaceutical industries.  相似文献

In the present study the total phenols, flavonoids, carotenoids and capsaicinoids content, the in vitro antioxidant and hypoglycemic activities of extracts of air-dried fruits from two Capsicum annuum cultivars were investigated. A different composition between the two cultivars was evidenced. C. annuum var. acuminatum medium extract presented a major content of phenols, carotenoids, capsaicin and dihydrocapsaicin while C. annuum var. acuminatum big extract is characterized by the highest quercetin, luteolin and kaempferol content with 68.0, 87.6 and 29.7 μg/g dried weight, respectively, analyzed by HPLC. Medium pepper showed the highest radical scavenging activity in DPPH assay (IC50 of 85.3 μg/ml) while big pepper have an interesting activity in ABTS assay (IC50 of 16.4 μg/ml) and the highest inhibition of linoleic acid oxidation with an IC50 value of 1.2 μg/ml after 30 min of incubation. A selective inhibitory activity against α-amylase was demonstrated for C. annuum var. acuminatum big lipophilic fraction (IC50 values of 8.7 μg/ml). The obtained results suggest that C. annuum cultivars could be used as valuable flavor with functional properties for foods.  相似文献

Tea and herbal infusions have been studied for their polyphenolic content, antioxidant activity and phenolic profile. The total phenolics recovered by ethyl acetate from the water extract, were determined by the Folin–Ciocalteu procedure and ranged from 88.1 ± 0.42 (Greek mountain tea) to 1216 ± 32.0 mg (Chinese green tea) GAE (Gallic acid equivalents)/cup. The antioxidant activity was evaluated by two methods, DPPH and chemiluminescence assays, using Trolox and quercetin as standards. The EC50 of herbal extracts ranged from 0.151 ± 0.002 mg extract/mg DPPH (0.38 quercetin equivalents and 0.57 Trolox equivalents), for Chinese green tea, to 0.77 ± 0.012 mg extract/mg DPPH (0.08 quercetin equivalents and 0.13 Trolox equivalents), for Greek mountain tea. Chemiluminescence assay results showed that the IC50 ranged from 0.17 ± 3.4 × 10−3 μg extract/ml of the final solution in the measuring cell (1.89 quercetin and 5.89 Trolox equivalents) for Chinese green tea, to 1.10 ± 1.86 × 10−2 g extract/ml of the final solution in the measuring cell (0.29 quercetin and 0.90 Trolox equivalents) for Greek mountain tea. The phenolic profile in the herbal infusions was investigated by LC-DAD-MS in the positive electrospray ionization (ESI+) mode. About 60 different flavonoids, phenolic acids and their derivatives have been identified.  相似文献

Bioactive compounds obtained from Coriolus versicolor (Trametes versicolor (L.: Fr) Lloyd, 1920.) mushrooms cultivated in a stirred-tank bioreactor were tested to determine their antimicrobial potential. Extracellular polysaccharides were isolated from the fermentation broth by ethanol precipitation. A methanol extract was prepared from mycelium. The cultivation conditions applied during the fermentation process provided for significant biomass 6.63 ± 0.31 g dry weight L 1 and yield of extracellular polysaccharides (EPS) (0.74 ± 0.12 g L 1). Microscopic analyses revealed that the mycelium grew predominately in the form of fluffy pellets. The methanol extract demonstrated very good activity against all the tested Gram-positive bacteria. Bacillus spizizeni and Staphylococcus epidermidis were the most sensitive strains (minimum inhibitory concentration (MIC) < 0.3125 mg mL 1). Among the Gram-negative bacteria, Yersinia enterocolitica had the lowest MIC value, 5 mg mL 1. Microbicidal activity of mycelia methanol extract was established in seven out of ten tested Gram-negative bacteria strains with minimum bactericidal concentration (MBC) values ranged from 20 to 40 mg mL 1. Enterococcus faecalis and Staphylococcus aureus showed higher sensitivity to the extracellular polysaccharides (MIC values 2.5 mg mL 1). FTIR analysis revealed a more complex chemical composition of the methanol extract compared to EPS, which might explain the better antibacterial activity of the methanol extract.Our results suggested that the submerged cultivation of Coriolus versicolor followed by ethanol precipitation of EPS and the methanol extraction of mycelia can be a promising process to obtain biological active compounds with significant antimicrobial activity.Industrial RelevanceMushrooms contain a large number of chemicals with potential use as antimicrobial compounds.One of the biggest challenges for providing biologically active compounds from mushrooms is short-term process standardization with a low risk for contamination. Submerged culture cultivation is the best choice for providing antimicrobial compounds from mushrooms. The submerged culture method represents an effective and energy-efficient means to produce novel antibacterial compound from mushrooms. Antibacterial activity testing revealed that methanol extract and isolated exopolysaccharides exhibited strong antibacterial activity, especially against Gram-positive bacteria.  相似文献

The objective of this study was to evaluate the effect of fermentation on the phenolic, vitamin C and total flavonoid contents and antioxidant properties of okra seed. The okra seeds were removed from the pod of matured and fibrous okra that cannot be easily cut with kitchen knife, and was allowed to undergo chance fermentation for 120 h. Samples were taken for antioxidant analysis at 24 h, 72 h and 120 h, respectively. The aqueous extracts of fermented and unfermented okra seeds were obtained and subsequently used for the analysis. The phenolic, vitamin C and total flavonoid contents and the antioxidant properties (ferric reducing antioxidant power (FRAP) and 1, 1-diphenyl-2 picrylhydrazyl (DPPH) free radical-scavenging ability) of the extracts were determined. The results revealed that fermented okra seeds had significantly (p < 0.05) higher phenolic content, vitamin C, total flavonoid and non-flavonoid contents and showed greater antioxidant activities than unfermented okra seed. Okra seeds fermented for 24 h exhibited the highest ferric reducing antioxidant power of 980 mgAAE/100 g and the least IC50 of 2.27 mg/ ml for DPPH free radical-scavenging ability than the others. Okra seeds fermented for 24 hours had significantly (p < 0.05) higher antioxidant activities. The product from fermented okra seeds could be used for the production of functional foods.  相似文献
