The attachment of anticancer agents to polymers is a promising approach towards reducing the toxic side‐effects and retaining the potent antitumour activity of these agents. A new tetrahydrophthalimido monomer containing 5‐fluorouracil (ETPFU) and its homopolymer and copolymers with acrylic acid (AA) and with vinyl acetate (VAc) have been synthesized and spectroscopically characterized. The ETPFU contents in poly(ETPFU‐co‐AA) and poly(ETPFU‐co‐VAc) obtained by elemental analysis were 21 mol% and 20 mol%, respectively. The average molecular weights of the polymers determined by gel permeation chromatography were as follows: Mn = 8900 g mol?1, Mw = 13 300 g mol?1, Mw/Mn = 1.5 for poly(ETPFU); Mn = 13 500 g mol?1, Mw = 16 600 g mol?1, Mw/Mn = 1.2 for poly(ETPFU‐co‐AA); Mn = 8300 g mol?1, Mw = 11 600 g mol?1, Mw/Mn = 1.4 poly(ETPFU‐co‐VAc). The in vitro cytotoxicity of the compounds against FM3A and U937 cancer cell lines increased in the following order: ETPFU > 5‐FU > poly(ETPFU) > poly(ETPFU‐co‐AA) > poly(ETPFU‐co‐VAc). The in vivo antitumour activities of all the polymers in Balb/C mice bearing the sarcoma 180 tumour cell line were greater than those of 5‐FU and monomer at the highest dose (800 mg kg?1). © 2002 Society of Chemical Industry  相似文献

Attachment of anticancer agents to polymers has been demonstrated to improve their therapeutic profiles. A new monomer containing camptothecin, 5‐norbonene‐endo‐2,3‐dicarboxylimidoundecanoyl‐camptothecin (NDUCPT) and its homopolymer and copolymer with acrylic acid (AA) were synthesized and spectroscopically characterized. The NDUCPT content in poly(NDUCPT‐co‐AA) obtained by elemental analysis was 51%. The average molecular weights of the polymers determined by gel permeation chromatography were as follows: Mn = 12 100, Mw = 23 400 g mol?1, Mw/Mn = 1.93 for poly(NDUCPT), Mn = 15 400, Mw = 28 300 g mol?1, Mw/Mn = 1.83 for poly(NDUCPT‐co‐AA). The IC50 value of NDUCPT and its polymers against U937 cancer cells was larger than that of CPT. The in vivo antitumour activity of all polymers in Balb/C mice bearing the sarcoma 180 tumour cell line was greater than that of CPT at a dose of 100 mg kg?1. A new monomer, vinyl‐(5‐fluorouracil)‐ethanoate (VFUE), was synthesized by reaction of 5‐fluorouracil (5‐FU) and vinyl iodoacetate. The homopolymer of VFUE and its copolymers with acrylic acid (A, A) and maleic anhydride (MAH) were prepared by photopolymerization. The synthesized VFUE and polymers were identified by FTIR, 1H NMR and 13C NMR spectroscopies. The contents of VFUE unit in poly(VFUE‐co‐AA) and poly(VFUE‐co‐MAH) were 21 mol% and 16 mol%, respectively. The number average molecular weights of the polymers determined by gel permeation chromatography were in the range 9600–17900 g mol?1. The in vitro cytotoxicities of the samples against a normal cell line decreased as follows: 5‐FU > VFUE > poly(VFUE) > poly(VFUE‐co‐AA) > poly(VFUE‐co‐MAH). The in vivo antitumour activities of the polymers against Balb/C mice bearing the sarcoma 180 tumour cells were greater than those of 5‐FU at all concentrations. The inhibition of simian virus 40 DNA replication by the samples was much greater than that of the control. © 2002 Society of Chemical Industry  相似文献

A new monomer, methacryloyl‐2‐oxy‐1,2,3‐propane tricarboxylic acid (MTCA), was synthesized from citric acid and methacrylic anhydride. Poly(methacryloyl‐2‐oxy‐1,2,3‐propane tricarboxylic acid) and poly(methacryloyl‐2‐oxy‐1,2,3‐propane tricarboxylic acid)‐co‐(maleic anhydride) were prepared by radical polymerizations. Terpoly(methacryloyl‐2‐oxy‐1,2,3‐propane tricarboxylic acid–maleic anhydride–furan) was obtained by in situ terpolymerization of MTCA and exo‐3,6‐epoxy‐1,2,3,6‐tetrahydrophthalic anhydride. The synthesized samples were identified by FTIR, 1H NMR and 13C NMR spectroscopies. The number‐average molecular weights of the fractionated polymers determined by GPC were in the range 14 900–16 600 and polydispersity indices were less than 1.14. The in vitro IC50 values of the monomer and polymers against cancer and normal cell lines were much higher than those of 5‐fluorouracil (5‐FU). The in vivo antitumour activities of the synthesized samples at a dosage of 0.8 mg kg−1 against mice bearing the sarcoma 180 tumour cell line decreased in the order terpoly(MTCA‐MAH‐FUR) > poly(MTCA‐co‐MAH) > poly(MTCA) > MTCA > 5‐FU. The synthesized samples inhibited DNA replication and angiogenetic activity more than did 5‐FU. © 2001 Society of Chemical Industry  相似文献

The new monomer, 5′-O-methacryloyl-3′-azido-3′-deoxythymidine (MAZT), was synthesized by the reaction of methacryloyl chloride and 3′-azido-3′-deoxythymidine (AZT). Poly(MAZT) and copolymers of MAZT with vinyl acetate (VAc) and maleic anhydride (MAH) were synthesized by radical polymerizations. The synthesized MAZT and polymers were identified by 1H nuclear magnetic resonance (NMR), 13C NMR, elemental analysis and gel permeation chromatography. The quantities of MAZT units in poly(MAZT-co-VAc) and poly(MAZT-co-MAH) were 45 and 27 mol%, respectively. The weight average molecular weights of the polymers synthesized were in the range from 8800 to 17600. The in vitro cytotoxicities of samples against K562 human leukaemia cell line at 100 μg ml-1 decreased in the following order: poly(MAZT-co-MAH) > poly(MAZT-co-VAc) > poly(MAZT) > MAZT > AZT. The in vivo anti-tumour activities of the polymers synthesized against Balb/C mice bearing sarcoma 180 tumour cells were greater than those of 5-fluorouracil at all concentrations.  相似文献

A new monomer, 3,6‐endo‐methylene‐1,2,3,6‐tetrahydrophthalimidobutanoyl‐5‐fluorouracil (ETBFU), was synthesized by reaction of 3,6‐endo‐methylene‐1,2,3,6‐tetrahydrophthalimidobutanoyl chloride and 5‐fluorouracil. The homopolymer of ETBFU and its copolymers with acrylic acid (AA) or vinyl acetate (VAc) were prepared by photopolymerization using 2,2‐dimethoxy‐2‐phenylacetophenone as an initiator at 25 °C. The synthesized ETBFU and its polymers were identified by FTIR, 1H NMR and 13C NMR spectroscopies. The ETBFU content in poly(ETBFU‐co‐AA) and poly(ETBFU‐co‐VAc) was 43 and 14 mol%, respectively. The apparent number‐average molecular weight (Mn) of the polymers determined by GPC ranged from 8400 to 11 300. The in vitro cytotoxicity of the samples against mouse mammary carcinoma (FM3A), mouse leukaemia (P388), and human histiocytic lymphoma (U937) cancer cell lines decreased in the order 5‐FU ≥ ETBFU > poly(ETBFU) > poly(ETBFU‐co‐AA) > poly(ETBFU‐co‐VAc). Hyperbranched poly(silyl ester)s were synthesized via the A2 + B4 route by the polycondensation reaction. The solid poly(silyl ester) was obtained by the reaction of di‐tert‐butyl adipate and 1,3‐tetramethyl‐1,3‐bis‐β(methyl‐dicholorosilyl)ethyl disiloxane. The oligomers with tert‐butyl terminal groups were obtained via the A2 + B2 route by the reaction of 1,5‐dichloro‐1,1,5,5‐tetramethyl‐3,3‐diphenyl‐trisi1oxane with excess amount of di‐tert‐butyl adipate. The viscous fluid and soft solid poly(silyl ester)s were obtained by the reaction of the oligomers as big monomers with 1,3‐tetramethyl‐1,3‐bis‐β(methyl‐dicholorosilyl)ethyl disiloxane. The polymers were characterized by 1H NMR, IR, and UV spectroscopies, differential scanning calorimetry (DSC), and thermogravimetric analysis (TGA). The 1H NMR and IR analysis proved the existence of the branched structures in the polymers. The glass transition temperatures (Tg's) of the viscous fluid and soft solid polymers were below room temperature. The Tg of the solid poly(silyl ester) was not found below room temperature but a temperature for the transition in the liquid crystalline phase was found at 42°C. Thermal decomposition of the soft solid and solid poly(silyl ester)s started at about 130°C and for the others it started at about 200°C. Poly(butylene 2,6‐naphthalate) (PBN)/poly(ethylene glycol) (PEG) copolymers were synthesized by the two‐step melt copolymerization process of dimethyl‐2,6‐naphthalenedicarboxylate (2,6‐NDC) with 1,4‐butanediol (BD) and PEG. The copolymers produced had different PEG molecular weights and contents. The structures, thermal properties, and hydrophilicities of these copolymers were studied by 1H NMR, DSC, TGA, and by contact angle and moisture content measurements. In particular, the intrinsic viscosities of PBN/PEG copolymers increased with increasing PEG molecular weights, but the melting temperatures (Tm), the cold crystallization temperatures (Tcc), and the heat of fusion (ΔHf) values of PBN/PEG copolymers decreased on increasing PEG contents or molecular weights. The thermal stabilities of the copolymers were unaffected by PEG content or molecular weight. Hydrophilicities as determined by contact angle and moisture content measurements were found to be significantly increased on increasing PEG contents and molecular weights. © 2006 Wiley Periodicals, Inc. Poly(vinyl alcohol) was modified by UV radiation with dimethyl amino ethyl methacrylate (DMAEMA) monomer to get poly(dimethyl amino ethyl methacrylate) modified poly(vinyl alcohol) (PVADMAEMA) membrane. The PVADMAEMA membranes were characterized by Fourier transform infrared spectroscopy. The tensile strength and elongation of PVADMAEMA membranes were measured by Universal Testing Machine. The results of X‐ray diffraction (XRD) and differential scanning calorimetry (DSC) showed that (1) the crystalline area in PVADMAEMA decreased with increasing the content of poly(dimethyl amino ethyl methacrylate) in the membrane. (2) Only one glass transition temperature (Tg) was found for the various PVADMAEMA membranes. It means that poly(dimethyl amino ethyl methacrylate) and PVA are compatible in PVADMAEMA membrane. (3)The Tg of the membrane is reduced with increasing the content of poly(dimethyl amino ethyl methacrylate) in the membrane. The water content on the PVADMAEMA membranes was determined. It was found that the water content on the PVADMAEMA membrane increased with increasing the content of poly(dimethyl amino ethyl methacrylate). The changes of properties enhanced the permeability of 5‐Fluorouracil (5‐Fu) through the PVADMAEMA membranes. A linear relationship between the permeability and the weight percent of poly(dimethyl amino ethyl methacrylate) in the PVADMAEMA membrane is found. It is expressed as P (cm/s) = (9.6 ± 0.4) × 10?5 + (8.8 ± 0.6) × 10?5 W x , where P is the permeability of 5‐Fu through the membrane and Wx is the weight percent of poly(dimethyl amino ethyl methacrylate) in the PVADMAEMA membrane. © 2011 Wiley Periodicals, Inc. Low molecular weight poly(acrylic acid‐co‐vinyl aminomethylene phosphonic acid)s were prepared by consecutively applying the Hofmann degradation and the Mannich reaction to polyacrylamide and poly(acrylamide‐co‐acrylic acid)s. 1H‐NMR, 31P‐NMR, and microanalysis were used for structural analyses. These polymers were tested as anti‐scalent and they showed better anti‐scalent effect than commercial poly(acrylic acid)s. A series of novel aromatic poly(ester‐ether‐imide)s with inherent viscosity values of 0.44–0.74 dL g?1 were prepared by the diphenylchlorophosphate‐activated direct polycondensation of an imide ring‐containing diacid namely 5‐(4‐trimellitimidophenoxy)‐1‐trimellitimido naphthalene ( 1 ) with various aromatic dihydroxy compounds in the presence of pyridine and lithium chloride. Owing to comparison of the characterization data, an ester‐containing model compound ( 2 ) was also synthesized by the reaction of 1 with phenol. The model compound 2 and the resulted polymers were fully characterized by FT‐IR and NMR spectroscopy. The ultraviolet λmax values of the poly(ester‐ether‐imide)s were also determined. The resulting polymers exhibited an excellent organosolubility in a variety of high polar solvents such as N,N‐dimethylacetamide, N,N‐dimethylformamide, dimethyl sulfoxide, and N‐methyl‐2‐pyrrolidone. They were soluble even in common less polar organic solvents such as pyridine, m‐cresol, and tetrahydrofuran on heating. Crystallinity of the polymers was estimated by means of wide‐angle X‐ray diffraction. The resulted polymers exhibited nearly an amorphous nature. From differential scanning calorimetry thermograms, the polymers showed glass‐transition temperatures between 221 and 245°C. Thermal behaviors of the obtained polymers were characterized by thermogravimetric analysis, and the 10% weight loss temperatures of the poly(ester‐ether‐imide)s were found to be over 410°C in nitrogen. © 2009 Wiley Periodicals, Inc. A new monomer, 1,2,3‐tris(ethoxycarbonyl)‐2‐propyl acrylate (TPA), was synthesized by reaction of acryloyl chloride and triethyl citrate. The homopolymer of TPA and its copolymers with acrylic acid (AA), vinyl acetate (VAc) and maleic anhydride (MAH) were prepared by polymerization using lauroyl peroxide (LPO) at 70 °C for 24 h. The structures of TPA and its polymers were identified by FTIR, 1H NMR, 13C NMR spectroscopies, and elemental analysis. The number average molecular weights and polydispersity indices of the synthesized polymers determined by GPC were in the range 4200–23 000 g mol?1 and 1.1–2.1, respectively. The IC50 values of the synthesized samples against cancer cell lines were greater than those of 5‐fluorouracil (5‐FU). The percentage inhibition values of SV40 DNA replication were 82.2 for TPA, 34.3 for poly (TPA), 81.9 for poly(TPA‐co‐AA), 82.0 for poly(TPA‐co‐VAc), 35.6 for poly(TPA‐co‐MAH) and 12.7 for 5‐FU. The inhibitions of SV40 DNA replication and antiangiogenesis for the synthesized TPA and its polymers are much greater than those of the control. © 2001 Society of Chemical Industry  相似文献

Poly(amides) and poly(imides) containing the heteroatoms Si or Ge in the main chain and bonded to four carbon atoms were synthesized and characterized by IR and 1H, 13C and 29Si NMR. The acid dichlorides bis(4‐chloroformylphenyl)‐dimethylgermane, bis(4‐chloroformylphenyl)‐diphenylgermane, bis(4‐chloroformylphenyl)‐dimethylsilane, and bis(4‐chloroformylphenyl)‐diphenylsilane were synthesized from the ditolyl derivatives, which were oxidized to the respective diacids. The dianhydrides bis(3,4‐dicarboxyphenyl)‐dimethylgermane dianhydride, bis(3,4‐dicarboxyphenyl)‐diphenylgermane dianhydride, bis(3,4‐dicarboxyphenyl)‐dimethylsilane dianhydride, and bis(3,4‐dicarboxyphenyl)‐diphenylsilane dianhydride were synthesized from the dixylyl derivatives, which were oxidized to the tetraacids. Fully aromatic diamines also containing Si or Ge were synthesized from 4‐bromo‐N,N‐bis(trimethylsilyl)‐aniline and diphenyl‐dichlorosilane or germane. The ditolyl and dixylyl derivatives were synthesized from 4‐bromo‐toluene or 4‐bromo‐xylene and dimethyl‐ or diphenyl‐dichlorogermane, dimethyl‐ or diphenyl‐dichlorosilane. The glass transition temperatures and the thermal stability were determined showing in general that the polymers with Si atom in the main chain presented higher values of both parameters due to the higher ionic character of the C? Pyromellitic dianhydride (benzene‐1,2,4,5‐tetracarboxylic dianhydride) (1) was reacted with several amino acids in acetic acid and the resulting imide‐acid [N,N′‐(pyromellitoyl)‐bis‐L ‐amino acid diacid] (4a–4d) was obtained in high yield. The direct polycondensation reaction of these diacids with 4,4′‐thiobis(2‐tert‐butyl‐5‐methylphenol) (5) was carried out in a system of tosyl chloride(TsCl), pyridine, and N,N‐dimethyl formamide (DMF) to give a series of novel optically active poly(esterimide)s. Step‐growth polymerization was carried out by varying the time of heating and the molar ratio of TsCl/diacid, and the optimum conditions were achieved. These new chiral polymers were characterized with respect to chemical structure and purity by means of specific rotation experiments, FTIR, 1H‐NMR, X‐ray diffraction, elemental, and thermogravimetric analysis (TGA) field emission scanning electron microscopy (FE‐SEM) techniques. These polymers are readily soluble in many polar organic solvents like DMF, N,N‐dimethyl acetamide, dimethyl sulfoxide, N‐methyl‐2‐pyrrolidone, and protic solvents such as sulfuric acid. TGA showed that the 10% weight loss temperature in a nitrogen atmosphere was more than 390°C; therefore, these new chiral polymers have useful levels of thermal stability associated with good solubility. Furthermore, study of the surface morphology of the obtained polymers by FE‐SEM showed that each polymers exhibit nanostructure morphology. © 2011 Wiley Periodicals, Inc. The two poly(silyl ester)s containing 2,2‐bis(p‐dimethylsiloxy‐phenyl)propane units in the polymer backbones have been prepared via polycondensation reaction of di‐tert‐butyl adipate and di‐tert‐butyl fumarate with 2,2‐bis(p‐chloro dimethylsiloxy‐phenyl)propane to give tert‐butyl chloride as the condensate. The polymerizations were performed under nitrogen at 110°C for 24 h without addition of solvents and catalysts to obtain the poly(silyl ester)s with weight average molecular weights typically ranging from 5000 to 10,000 g/mol. Characterization of the poly(silyl ester)s included 1H NMR and 13C NMR spectroscopies, infrared spectroscopy, ultraviolet spectroscopy, differential scanning calorimetry, thermogravimetric analysis (TGA), gel permeation chromatography, and Ubbelohde viscometer. The glass transition temperatures (Tg) of the obtained polymers were above zero because of the introducing 2,2‐bis(p‐dimethylsiloxy‐phenyl)propane units in the polymer backbones. The TGA/DTG results showed that the obtained poly(silyl ester)s were stable up to 180°C and the residual weight percent at 800°C were 18 and 9%, respectively. © 2006 Wiley Periodicals, Inc. Sulfonated dimethyl fumarate (SDMF) was prepared with dimethyl fumarate (DMF) and sodium hydrogensulfite (NaHSO3). Sodium sulfonate side‐chain poly(ethylene oxide) (SPEO) was synthesized by grafting sodium sulfonate onto the chain of PEO with molecular weights of 400, 600, 800, and 1000. SPEO was used subsequently in step‐growth polymerization to give a polyurethane ionomer (SPU). Samples were characterized by element analysis, FTIR, 1H‐NMR, EDX mapping, X‐ray, gel permeation chromatography, and impedance analysis. The SPUs exhibited an amorphous structure. A series of poly(ether–ester) copolymers were synthesized from poly(2,6 dimethyl‐1,4‐phenylene oxide) (PPO) and poly(ethylene terephthalate) (PET). The synthesis was carried out by two‐step solution polymerization process. PET oligomers were synthesized via glycolysis and subsequently used in the copolymerization reaction. FTIR spectroscopy analysis shows the coexistence of spectral contributions of PPO and PET on the spectra of their ether–ester copolymers. The composition of the poly(ether–ester)s was calculated via 1H NMR spectroscopy. A single glass transition temperature was detected for all synthesized poly(ether–ester)s. Tg behavior as a function of poly(ether–ester) composition is well represented by the Gordon‐Taylor equation. A series of aliphatic poly(ether–ester)s based on flexible poly(tetramethylene oxide) (PTMO) and hard poly (butylene succinate) (PBS) segments were synthesized by the catalyzed two‐step transesterification reaction of dimethyl succinate, 1,4‐butanediol, and α,ω‐hydroxy‐terminated PTMO (Mn = 1000 g/mol) in the bulk. The content of soft PTMO segments in the polymer chains was varied from 10 to 50 mass %. The effect of the introduction of the soft segments on the structure, thermal, and physical properties, as well as on the biodegradation properties was investigated. The composition and structure of the aliphatic segmented copolyesters were determined by 1H NMR spectroscopy. The molecular weights of the polyesters were verified by viscometry of dilute solutions and polymer melts. The thermal properties were investigated using DSC. The degree of crystallinity was determined by means of DSC and WAXS. Biodegradation of the synthesized copolyesters, estimated in enzymatic degradation tests on polymer films in phosphate buffer solution with Candida rugosa lipase at 37°C, was compared with hydrolytic degradation in the buffer solution. Viscosity measurements confirmed that there was no change in molecular weight of the copolyesters leading to the conclusion that the degradation mechanism of poly(ester–ether)s based on PTMO segments occurs through the surface erosion. © 2007 Wiley Periodicals, Inc. A series of chiral poly(ether sulfone)s containing (S)‐binaphthyl unit were synthesized via nucleophilic substitution polycondensation. The as‐synthesized polymers were characterized by NMR. The molecular weights of these polymers were calculated by viscosity and gel permeation chromatography. The thermal stability was measured by TGA. The optical activities of the polymers were studied by circular dichroism and optical rotation. It was found that introducing more reactive 2,2′‐biphenol monomer to partly substitute the binaphthol monomer could increase the molecular weight of the polymer from 4.1 × 104 to 9.8 × 104 g mol?1. Meanwhile, the thermal stability increased and the decomposition temperature rose from 447 to 518 °C, with the optical activity of the polymer well preserved. However, the value of the specific optical rotation decreased with increasing biphenol to binaphthol molar ratio. In addition, the optical rotation direction of the polymer was contrary to that of the pristine binaphthol monomer, which indicated that the chiral polymers formed a secondary structure. © 2013 Society of Chemical Industry  相似文献
