Studies were carried out to determine the effect of dietary soybean trypsin inhibitor (SBTI) content on nutrient and energy digestibilities in growing pigs. Six barrows, average initial body weight (BW) 47·8±4·0 kg, were fitted with a simple T-cannula at the distal ileum and fed two diets according to a crossover design. Two maize starch-based diets were formulated to contain 200 g crude protein (CP) kg−1 from either Nutrisoy (a food-grade defatted soy flour) or autoclaved Nutrisoy. The contents of SBTI in the Nutrisoy and autoclaved Nutrisoy diets were 13·4 and 3·0 g kg−1, respectively. The experiment consisted of two periods of 12 days each. The average BW at the start of the first and second experimental periods were 53·3±3·7 and 61·0±5·1 kg, respectively. The average BW at the conclusion of the experiment was 71·8±7·6 kg. The ileal digestibilities of dry matter (DM), organic (OM), energy, CP and all amino acids measured were higher (P<0·01) in pigs fed the autoclaved Nutrisoy diet. The energy digestibility increased from 66·0 to 77·9%, and the CP digestibility increased from 37·4 to 77·1%. The increases in ileal digestibilities of the indispensable amino acids ranged from 27·0 (methionine) to 49·2 (leucine) percentage units. The increases in ileal digestibilities of the dispensable amino acids ranged from 30·2 (aspartic acid) to 50·8 (tyrosine) percentage units. The faecal digestibilities of all criteria measured were also higher (P<0·01) in pigs fed the autoclaved Nutrisoy diet. Furthermore, there was a greater (P<0·01) net disappearance (g kg−1 DM intake) of DM, OM, CP, energy and all amino acids in the large intestine of pigs fed the Nutrisoy diet. In conclusion, feeding diets containing high levels of SBTI decreased both ileal and faecal digestibilities of all criteria measured. The formation of SBTI-enzyme complexes is likely to be responsible for the reduction in protein digestion and amino acid absorption. © 1998 SCI.  相似文献

Two experiments were carried out to investigate the effect of feeding diets containing faba beans and peas on pancreatic secretions in young pigs fitted with a pancreatic pouch re-entrant cannula. In Experiment 1, eight barrows, with an average initial weight of 8·5 kg, were fed one of two maize starch-based diets formulated to contain 200 g crude protein (CP) kg−1 according to a completely randomised design. In one of the diets, soya bean meal was the sole protein source; in the other diet, soya bean meal and faba beans (cv Fibro; dark-flowering) each supplied 50% of the dietary CP. In Experiment 2, five barrows, with an average initial weight of 18·1 kg, were fed one of two diets according to a two-period change-over design. The diets were formulated to contain 150 g CP kg−1 with peas (cv Ascona or cv Radley) as the sole protein source. In both experiments, the experimental periods consisted of 8 days: 6 days adaptation followed by a 2 day collection of pancreatic juice. The soya bean meal diet and the faba bean diet contained 0.04 and 0.21% tannins (catechin equivalents), respectively. The trypsin inhibitor activities of the Ascona and Radley pea diets were 0·76 and 3·24 mg of trypsin inhibited per gram, respectively. In both experiments, diet did not affect ( P> 0·05) the pancreatic secretions of nitrogen, protein, amylase, lipase and chymotrypsin. In Experiment 1, specific trypsin activity was higher ( P< 0·05) in pancreatic juice from pigs fed the diet containing soya bean meal and faba beans (53700 units litre−1) than for those fed the soya bean meal diet (41200 units litre−1). However, there was no effect ( P> 0·05) when trypsin activity was expressed as total activity (units 24 h−1). It is concluded that the inclu-sion of faba beans and peas in diets for young pigs has only minor effects on exocrine pancreatic secretions.  相似文献

Two experiments were carried out to investigate the effects of feeding diets containing faba beans and peas on the concentration, flow and composition of total, protein-bound and free amino acids (AA) in pancreatic juice collected from young pigs fitted with a pancreatic pouch re-entrant cannula. In Experiment 1, eight barrows, with an average initial weight of 8·5 kg, were fed one of two semi-purified diets formulated to contain 200 g kg−1 crude protein (CP) according to a completely randomised design. In one of the diets, soya bean meal was the sole protein source; in the other diet, soya bean meal and faba beans (cv Fibro; dark-flowering) each supplied 50% of the dietary CP. In Experiment 2, five barrows, with an average initial weight of 18·1 kg, were fed one of two diets formulated to contain 150 g kg−1 CP with peas (cv Ascona or cv Radley) as the sole protein source. There was no effect (P>0·05) of the diet, 8 h sampling period or experimental period on the concentration (mM), composition (% and g 16 g N−1) and flow (mmol 24 h−1) of total, protein-bound and free AA or on the percentage of protein-bound AA in pancreatic juice. The concentrations of total, protein-bound and free AA were higher in pancreatic juice collected from younger pigs in Experiment 1 than in pancreatic juice collected from older pigs in Experiment 2. However, the daily flows of total, protein-bound and free AA in pancreatic juice were similar in both experiments. The percentages of protein-bound AA in pancreatic juice were similar in both experiments. In conclusion, the concentrations, compositions and flows of total, protein-bound and free AA in pancreatic juice were not affected by diet, 8 h sampling period or experimental period.  相似文献

Six colour-flowering (Scirocco, Alfred, Carola, Condor, Tina and Herz Freya) and six white-flowering (Caspar, Albatros, Gloria, Tyrol, Vasco and Cresta) cultivars of Vicia faba were studied. The crude protein contents of colour- and white-flowering cultivars were 267±13·6 and 283±18·8 g kg−1, respectively, which did not differ significantly at P<0·05. The levels of lipids, crude fibre, starch and ash varied from 14 to 22 g kg−1, 88 to 143 g kg−1, 407 to 485 g kg−1 and 32 to 42 g kg−1, respectively. The calculated organic matter digestibility (OMD) and metabolisable energy (ME) of the white-flowering cultivars were significantly higher (P<0·001) than those of the colour-flowering cultivars (OMD: 889·1±26·6 g kg−1 vs 797·5±17·1 g kg−1; ME: 13·97±0·49 vs 12·30±0·34 MJ kg−1). In all cultivars, sulphur amino acids were lower than adequate concentration when compared with recommended amino acid pattern of FAO/WHO/UNO reference protein for a 2–5-year-old child. The in vitro rumen nitrogen degradability of colour-flowering cultivars was significantly lower (P<0·01) compared to that of white-flowering cultivars (71·4±9·3% vs 88·0±11·1%). Amongst colour-flowering varieties, the contents of total phenols (TP), tannins (T) and condensed tannins (CT) were highest in Alfred (28·3, 21·0 and 35·4 g kg−1, respectively). The contents of TP and T were similar (about 15 and 10 g kg−1, respectively) in Carola, Tina and Herz Freya, and the CT were in the order: Condor>Herz Freya>Carola. The CT were not detected in white-flowering varieties, T were virtually absent and TP were extremely low (4·0–4·9 g kg−1). The activities of other antinutritional factors (white- and colour-flowering cultivars, respectively: trypsin inhibitor activity 3·05±0·34 and 1·85±0·09 mg trypsin inhibited g−1; lectin 27·2±9·4 and 27·1±5·1 mg ml−1 assay medium producing haemagglutination; phytate 15·0±2·7 and 16·6±2·3 g kg−1) were very low. A strong negative correlation (r=-0·92, P<0·001) between tannins and in vitro rumen protein degradability was observed which suggested that tannins have adverse effect on protein degradability. Similarly negative correlations between tannin levels and metabolisable energy (r=-0·89; P<0·001) and organic matter digestibility (r=-0·89; P<0·001) were observed. The correlation coefficient between trypsin inhibitor activity and tannins was negative and highly significant (r=-0·88, P<0·001), whereas between tannins and saponins it was significantly positive (r=0·96, P<0·001). ©1997 SCI  相似文献

Five barrows were fitted with permanent pancreatic re-entrant cannulas to determine the influence of diet on exocrine pancreatic secretions in the weight range from 40 to 65 kg. The pigs were fed four diets. Diets 1 and 2 were based on corn starch and contained 18.6 and 16.9 % crude protein (dry matter basis) from soybean meal and canola meal, respectively. Diets 3 and 4 contained 93.9 % wheat and barley, respectively. In the same order, the crude protein contents (dry matter basis) of these diets were 14.4 and 11.2%, respectively. The experiment was carried out according to 4 × 4 Latin square design. Each experimental period comprised 12 days. Following an adaptation period of 8 days, pancreatic juice was collected from 08:00 on day 9 to 08:00 h on day 10 and from 08:00 on day 11 to 08:00 h on day 12. The pigs were fed twice daily, at 08:00 and 20:00 h, 800 g each meal. Despite large differences, up to 23%, between the overall mean values of the parameters that were measured there was no effect (P > 0.05) of diet on the volume of secretion of pancreatic juice, the pH and the secretion of total and TCA-precipitable nitrogen. Due to the high level of within-study variation there was also no effect (P > 0.05) of diet on the secretion of amino acids and the total activity of trypsin, chymotrypsin, α-amylase and lipase in pancreatic juice.  相似文献

Forty eight and ninety six pigs weaned at 22·6±2·6 days, were allocated to two trials to study the effect of feeding post-weaning diets ad libitum , either as dry pellets (DF) or as meal mixed with water in the ratio 2·5:1 (LF). The trials, lasting 28 days, differed only in the type of trough used to present the liquid feed to the pigs. In both trials the daily gain of LF pigs over the 4 weeks of the trial was significantly ( P< 0·001) better than that of the DF pigs. Daily gains for the two treatment groups were respectively 428 and 343±15 g day−1 in Trial 1 and 454 and 397±10 g day−1 in Trial 2. The feed conversion ratio (FCR) (corrected to fresh weight of dry feed) of LF pigs was significantly poorer ( P< 0·01) than that of DF pigs in both trials primarily as a result of feed wastage by the LF pigs. FCR for the DF and LF pigs was, respectively, 1·31 and 1·89±0·05 in Trial 1. In Trial 2 improvements in the trough design reduced the difference in FCR. FCR for the DF and LF treatments was 1·37 and 1·44±0·02 respectively. In Trials 1 and 2 LF pigs used on average 76% and 35% more water than DF pigs. LF pig produced 12% (Trial 1) and 5·7% (Trial 2) more effluent per unit of liveweight gain than DF pigs. Microbiological examination of the liquid feed system demonstrated that the system acted as a fermenter. Lactobacillus spp proliferated rapidly and became the dominant organism within 5 days and the populations remained relatively constant thereafter. The proliferation of lactobacilli reduced the pH of the liquid feed from approximately 6·0 to between 3·5 and 4·0. At the lower levels of pH the number of coliforms in the feed was significantly reduced.  相似文献

Raw mango ( Mangifera indica L) seed kernels were found to contain tannins (56·5 g kg−1 DM), cyanogenic glucosides (64 mg kg−1 DM), oxalates (42 mg kg−1 DM) and trypsin inhibitory activity (20 TIU g−1 DM). The contents of these anti-nutritive factors were lowered by both soaking and boiling treatments, but boiling was more effective. The in vitro protein digestibility (26·3%) and apparent metabolisable energy (7·88 MJ kg−1 DM) values of raw kernels were low, and these parameters were improved by soaking and boiling. The observed improvements paralleled reductions in tannin contents, indicating that tannins are largely responsible for the poor nutritive value of raw kernels. In experiment 1, diets containing 0, 50, 100, 150 and 200 g kg−1 raw mango seed kernels that replaced maize were fed to 7-day-old White Leghorn cockerels for 14 days. Inclusion of more than 50 g kg−1 raw kernels lowered ( P< 0·05) the weight gains, feed intake and feed efficiency of chicks. High level inclusion of raw kernels had toxic effects, as evidenced by increased mortality. In experiment 2, soaking and boiling treatments improved ( P< 0·05) feed intake of chicks fed on diets containing 100 g kg−1 kernels and reduced mortality. Feed/gain was unaffected by processing. Soaking had no effect, whereas boiling of kernels improved the weight gains. However, weight gains of chicks receiving diets containing 100 g kg−1 boiled kernels were numerically, though non-significantly, lower than those of the maize-control group. It is concluded that raw mango seed kernels are unsuitable as a feed ingredient in chick diets and, that soaking and boiling do not completely overcome the anti-nutritive effects of raw kernels.  相似文献

Effects of diets containing increasing levels of whole cotton seed (WCS) on feed intake, liveweight gain, feed conversion, feeding margin and carcass characteristics were studied. Diets contained 0, 5, 10, 15, 20 and 30% WCS and were fed ad libitum to growing lambs for 54 days. All diets were approximately isonitrogenous (163–186 g crude protein kg−1 dry matter) and isocaloric (19–20 MJ gross energy kg−1 of dry matter). During the first 28 days, four groups of 10 Karagouniko male lambs with initial liveweights of approximately 17·6 kg were fed diets containing 0, 5, 10 and 15% WCS and then, for the next 26 days, diets containing 0, 10, 20 and 30% WCS, respectively. Lambs fed the 15–30% WCS diet ingested about 20 mg kg liveweight−1 day−1 of free gossypol. At 54 days and liveweights of approximately 34·7 kg, five lambs from each treatment were slaughtered and carcass data were collected. There were significant differences in feed intake, liveweight gain, cooler shrink, rumen contents, intestinal fat and liver weight among the lambs fed these diets. Cooler shrink and intestinal fat, and perirenal fat and the combined weight of intestinal and perirenal fat, were significantly (P < 0·05) correlated. Pelleting of the WCS diets reduced the concentration of free gossypol in these diets. It was concluded that WCS was satisfactory as a feed ingredient for growing sheep and can be incorporated into ruminant diets as a source of energy and protein. The effect of feeding pigs weaned at 24±2·6 days, ad libitum , a diet mixed with water to provide four different dry matter concentrations of 149, 179, 224 and 255 g kg−1 (diets DM149, DM179, DM224, DM255, respectively) was investigated in a 28 day feeding trial, utilising a total of 96 pigs. The diets were delivered by an automated system, the storage and mixing vessel of which was replenished daily. In addition to the water supplied in the liquid diet water was freely available to the pigs at all times from nipple waterers. Dietary dry matter (DM) concentration had no significant effect on DM intake, weight gain or DM feed conversion ratio of the piglets. Decreasing feed DM had a noticeable effect on total volumetric (DM plus water) intake of the piglets. When feed dry matter was reduced from 255 to 224 g kg−1 piglets reduced their voluntary water intake from nipple waterers in order to maintain DM intake and total volumetric intake. At lower DM concentrations, the pigs maintained DM intake by increasing their total volumetric intake. Piglets continued to consume an average of 223 ml water per day from the nipple waterers even at the lowest DM concentration. On DM149 total volumetric intake was 30% of live weight. The effect of dry matter (DM) concentration of feed on the pattern of microbial activity of the liquid feed system was investigated. Liquid diets were colonised rapidly by naturally occurring Lactobacillus spp in the first 5 days. After 5 days the population stabilised and remained constant for the remainder of the trial period. DM concentration of liquid feed had little overall effect on the pattern of microbial activity. The proliferation of Lactobacillus spp resulted in acidification of the liquid feed, (pH range 4·1–4·2) for all four treatments and this was associated with a lowering of coliform numbers. There was a significant increase in the overall effluent output as the DM content of the diet decreased ( P< 0·01). It is concluded that weanling piglets will readily accept liquid feed with DM content in the range 255 to 149 g kg−1, and that DM content in this range has little effect on post weaning performance. However, in order to maintain DM intake without increasing effluent output, it is recommended that the DM content of liquid diets should not be reduced below 200 g kg−1 (equivalent to a 3·5:1 water to feed ratio).  相似文献

Using cannulated pigs and a standardised in vitro fermentation system the effect of diet and non‐starch polysaccharides (NSP) on the amount of energy available from microbial fermentation in the large intestine could be predicted. The available energy was calculated from the amounts of short‐chain fatty acids (SCFA) produced. Three diets were investigated: a low fibre diet based essentially on wheat flour (56 g NSP kg−1 feed) and two high fibre diets with added oat bran (93 g NSP kg−1 feed) or wheat bran (102 g NSP kg−1 feed). Colonic fermentation was estimated by in vitro fermentation of freeze‐dried ileal effluent collected from cannulated pigs. The in vitro fermentation method was optimised to use 10 g ileum content litre−1 incubated at pH 6.0 in a fermentor containing faecal slurry consisting of anaerobic mineral salts medium and 50 g litre −1 faeces from pigs fed the same diets as the cannulated pigs. The results demonstrate that it is very important to compensate for the faecal SCFA contribution when calculating the amount of SCFA produced from ileal digesta during in vitro fermentation. The amount of NSP digested in vitro was compared with data obtained from in vivo studies and there was a good agreement between in vivo and in vitro data. We concluded that the integrated in vivoin vitro method is a valuable technique to estimate the effect of diet and NSP on the amount of SCFA produced in the large intestine and when fed the three diets the microbial fermentation in the large intestine provided between 2.4–6.4% of the total available energy. © 1999 Society of Chemical Industry  相似文献

Five castrated male Iberian pigs (100 ± 2 kg body weight) fitted with T‐shaped cannulas at the terminal ileum were used to determine ileal digestibility of legume seed meals. The diets were based on defatted soybean, lupin or chickpea seed meals and contained similar levels of digestible energy (14.2–15.1 kJ g?1) and protein (107 g kg?1). Protein‐free and a hydrolysed casein diets were used to study endogenous ileal amino acid flows. Chromium oxide (10 g kg?1 diet) was added to the diets as an indigestible marker. Ileal flows in pigs fed the hydrolysed casein diet were different (p < 0.05) in amino acid contents and composition from those on the protein‐free diet. Ileal sialic acid flows in pigs fed lupin‐ or chickpea‐based diets were higher (p < 0.05) than those of animals fed soybean or casein diets. Among essential amino acids, only the apparent ileal digestibilities of phenylalanine and valine in lupin meal were lower (p < 0.05) than those in soybean. Apparent ileal digestibilities of lupin aspartate and proline, together with chickpea aspartate, were also lower (p < 0.05) than those of soybean. True ileal digestibility of nitrogen in pigs fed lupin or chickpea meals, calculated according to values from animals fed the protein‐free diet, was lower (p < 0.05) than that for soybean or casein. Among individual essential amino acids, only the true ileal digestibility of phenylalanine in lupin was lower (p < 0.05) than that in soybean. True ileal digestibility of nitrogen calculated according to values obtained with pigs fed a hydrolysed casein diet was not different among soybean, lupin or chickpea meals. Among essential amino acids, only the true ileal digestibilities of isoleucine and lysine in chickpea were lower (p < 0.05) than those of soybean. It is concluded that true ileal nitrogen and amino acid digestibilities of lupin and chickpea meals are comparable to those of defatted soybean in Iberian pigs. The effect of feeding newly weaned pigs acidified liquid diets was investigated. The control diet was acidified to about pH 4 with lactic acid (LA). A second diet of the same formulation was acidified to about pH 4 by fermentation with Pediococcus acidilactici (PA). Forty‐eight weaner pigs (weight 7 kg±1 kg, age 24±4 days) were allocated to the two dietary treatments according to a randomised block design and fed ad libitum for 28 days. Food intake, daily gain and water intake were recorded, and a microbial assessment of the liquid diet was conducted. Reducing pH<4.0 in either of the liquid diets was effective in eliminating coliform bacteria. There were no significant differences in any of the performance parameters measured. The average daily liveweight gain overall was 474 and 496±17.8 g d−1 for PA and LA, respectively, with a feed conversion ratio overall of 1.15 and 1.11±0.025 for PA and LA, respectively. Fermentation of liquid diets for newly weaned piglets could provide a more cost effective means of acidifying diets than the use of organic acids. Reducing the pH of the liquid diet to 4.00 by fermentation with Pediococcus acidilactici was a cost effective method of eliminating enteropathogens and spoilage organisms from the diet. © 1999 Society of Chemical Industry  相似文献

Disappearance of cis-18:1(n-9) from ruminal in vitro cultures supplemented with either oleic acid or oleamide was measured over 48 h to determine if the amide resisted biohydrogenation. Oleamide added to the substrate maintained higher concentrations of cis-18: 1(n-9) in the microbial cultures at 24 and 48 h of incubation compared to substrates with added oleic acid. Disappearance rates of cis-18: 1(n-9) from the cultures, which were calculated as a measure of biohydrogenation, were 0·064 and 0·025 h−1 for the oleic acid and oleamide supplements, respectively. Four sheep were fed four diets (control, 42 g kg−1 oleic acid, 23 g kg−1 oleamide, and 45 g kg−1 oleamide) in a 4×4 Latin square to determine how the amide affected fatty acid digestibility. Total tract digestibilities of protein and fibre were not affected (P>0·05) by either oleic acid or oleamide compared to the control diet. Fatty acid and energy digestibilities were not changed (P>0·05) by oleic acid, but were increased (P<0·05) when oleamide was added to the sheep diets at 45 g kg−1. These results show that oleamide resists ruminal biohydrogenation without impairing fatty acid digestibility. © 1998 SCI.  相似文献

The feeding value of feed grade chickpeas (Kabuli and Desi type) for ruminants and pigs was determined in two studies. Two ruminally fistulated cows were used in the first study to determine the effects of moist heat treatment (127 °C for 10 min) on the ruminal protein degradability of feed grade chickpeas. Heat treatment reduced (P < 0.05) soluble crude protein and increased (P < 0.05) the amount of crude protein associated with neutral detergent fibre for both types of chickpeas. Ruminal degradability of crude protein for Kabuli and Desi chickpeas was reduced by 39 and 33% (P < 0.05) respectively as a result of heating. In the second study, 64 crossbred pigs were each assigned to one of four dietary treatments in a factorial (4 treatments × 2 sexes) design experiment. The experimental barley/wheat‐based diets were supplemented with either soybean meal (control), Kabuli chickpeas (300 g kg−1), Desi chickpeas (300 g kg−1) or field peas (300 g kg−1). Dry matter and gross energy digestibility coefficients, determined using the indicator method, were lower (P < 0.05) for the Desi‐ than for the Kabuli‐supplemented diet. However, no differences were observed in dry matter or gross energy digestibility between the Kabuli chickpea‐supplemented diet and the control or the field pea‐supplemented diet. Diets supplemented with chickpeas or field peas had a lower (P < 0.05) crude protein digestibility than the control diet. Pigs fed either of the chickpea‐supplemented diets during the growing phase gained less (P < 0.05) than pigs fed the control diet. However, during the finishing phase and over the entire experiment, dietary treatment had no effect on pig performance. There were no significant differences in carcass traits between pigs fed any of the four diets. It was concluded that moist heat treatment is an effective method to increase the rumen escape protein value of chickpeas and that the inclusion of feed grade Kabuli or Desi chickpeas in finishing diets up 300 g kg−1 had no detrimental effects on pig performance. © 2000 Society of Chemical Industry  相似文献

Three experiments, the first with rats, the second with young pigs and the third with growing pigs, were carried out to compare the nutritional value of cv Lysimax, a newly developed high-lysine barley, to cv Lami, a conventional barley grown in Denmark. In the diets for rats and growing pigs the barleys were the sole protein source while in the experiment with young pigs the barleys made up to 649 g kg-1 of the respective diets. The lysine content of cv Lysimax was 52·6% higher than that of cv Lami; 4·99 g 16 g N-1 compared to 3·27 g 16 g N-1. The contents of threonine, tryptophane, methionine and cystine of cv Lysimax were 3·80, 1·41, 1·80 and 2·27 g 16 g N-1, respectively; being 18·4, 8·5, 9·8 and 5·1% higher than in cv Lami. Conversely, the contents of glutamic acid and proline in cv Lysimax were 15·79 and 7·40 g 16 g N-1, respectively, being 36·1 and 40·0% lower than in cv Lami. In the experiment with rats, the higher lysine content and the lower content of glutamic acid and proline in cv Lysimax resulted in a 34·8% increase (P<0·05) in biological value (BV); 0·867 for cv Lysimax compared to 0·643 for cv Lami. True protein digestibilities and energy digestibilities in rats fed cv Lysimax were 0·019 and 0·010 units lower (P<0·05) than for cv Lami. For young pigs the increased lysine content and the lower content of glutamic acid and proline of cv Lysimax increased (P<0·05) nitrogen (N) retention from 10·9 to 12·4 g day-1, respectively, and increased (P<0·05) apparent BV from 0·690 to 0·734, respectively. The digestibility of energy was 0·030 units lower (P<0·05) for the diets with cv Lysimax than for the diets with cv Lami. Likewise, with growing pigs the increased lysine content and the lower content of glutamic acid and proline increased (P<0·05) N retention from 6·4 to 10·8 g day-1 and apparent BV from 0·271 to 0·495, respectively. The apparent ileal digestibilities of lysine, threonine, tryptophane, methionine and cystine were similar (P>0·05) between the barleys. It is concluded that the development of high-lysine barley varieties could be beneficial for meeting the requirements of essential amino acids for monogastrics. Moreover, N excretion into the environment was substantially reduced due a reduction in the prolamine fraction which is rich in glutamic acid and proline. © 1997 SCI.  相似文献

Twenty‐four Large White × Landrace pigs were individually fed, from 50 to 90 kg live weight, either a control (CONT) diet containing palm oil or one of three diets based on full‐fat rapeseed (250 g kg−1) (diets RD). The RD diets were supplemented with 0, 200 or 500 mg DL ‐α‐tocopherol acetate (ATA) kg−1 diet (diets RD0, RD200 and RD500 respectively). Diets were formulated to be isonitrogenous and isocaloric. Daily live weight gain was significantly increased (p < 0.01) in pigs fed diet RD500. Plasma AT concentration was significantly increased by dietary supplementation with 200 mg ATA kg−1 but showed no further significant increase by supplementation with 500 mg ATA kg−1. At slaughter, after 45 days, carcass weights were increased for the RD500 group but dressing percentage was unaffected. ATA supplementation significantly reduced drip loss on days 4 and 5–7 in fresh muscle and on days 1 and 4 in frozen muscle. The concentrations of calcium, sodium and potassium in drip loss fluid collected on days 1 and 4 from fresh muscle were not significantly affected by treatment or by time of collection and did not suggest any change in the relative contribution of intra‐ and extracellular fluid to total drip loss. Plasma enzyme activities related to tissue damage (creatine kinase, alanine aminotransferase, aspartate aminotransferase and lactate dehydrogenase) were not influenced by dietary treatments. © 2000 Society of Chemical Industry  相似文献

Binding interactions of human serum albumin (HSA) with sunset yellow (SY) and allura red (AR), two food colorants, were investigated at the molecular level through fluorescence and UV absorption as well as molecular docking. The collective results of the study under the simulated physiological conditions proposed a static type of binding occurring between the two dyes and HSA. When compared with AR (293 K: Ksv = (4.21 ± 0.36) × 104 L·mol−1; Kb = (0.30 ± 0.23) × 106 L·mol−1), SY (293 K: Ksv = (6.80 ± 0.10) × 104 L·mol−1; Kb = (3.11 ± 2.01) × 106 L·mol−1) had stronger quenching ability and higher affinity for HSA due to less steric hindrance. It can be deduced that the energy transfer from HSA to the two dyes occurred with high probability based on the Förster resonance energy transfer theory (r < 7 nm, 0.5 R0 < r < 2.0 R0). The spectral analysis suggested that the formation of the dye-HSA complex resulted in the change in microenvironment around Tyr and Trp residues and in the secondary structure of the protein. According to molecular docking simulation, the two structural analogs almost bound to the same site of HSA, near Sudlow's Site I, but significant difference existed in the number and location of hydrogen bond (H-bond) formed between the dyes and HSA. From the molecular docking along with the thermodynamic parameters (AR: ΔHo = −(58.79 ± 15.24) kJ·mol−1, ΔSo = −(115.1 ± 31.10) J·mol−1·K−1; SY: ΔHo = −(52.24 ± 3.15) kJ·mol−1, ΔSo = −(50.07 ± 11.14) J·mol−1·K−1), it could be inferred that H-bond and van der Waals forces were the major binding forces involved in formation of the dye-HSA complexes.  相似文献

In experiment 1, 224 pigs (initially 8.85 kg and 28 ± 2 days of age) were used in a 2 × 2 factorial arrangement of treatments to investigate the interaction between lactofeed (LF70) (860 g kg?1 whey permeate, 140 g kg?1 soya bean meal) level (175 g kg?1 and 350 g kg?1) and avilamycin (0 and 40 mg kg?1) inclusion in piglet starter diets. Pigs were fed starter diets from day 0 to day 22 and a transition diet was fed from day 23 to day 39. The inclusion level of LF70 in the transition diet was 75 g kg?1 and 150 g kg?1. Pigs fed 350 g kg?1 LF70 had a higher daily gain (ADG) (p < 0.01) and an improved feed conversion ratio (FCR) (p < 0.05) during the starter period than the pigs fed 175 g kg?1 LF70. Pigs fed medicated diets had a higher ADG (p < 0.05) and an improved FCR (p < 0.05) than the non‐medicated fed pigs during the starter period. There was an increase in feed intake (AFI) (p < 0.05) during the transition period with increasing levels of LF70. There was an improvement in FCR during the transition period with the inclusion of avilamycin (p < 0.01). There was a significant interaction (p < 0.01) between LF70 and avilamycin for ADG during the transition period. The inclusion of avilamycin at 175 g kg?1 LF70 inclusion had no effect (p > 0.05) on ADG. However at 350 g kg?1 LF70 inclusion the pigs offered medicated diets had a higher ADG (p < 0.001) compared with non‐medicated diets. The inclusion level of LF70 in the transition diet was 75 g kg?1 and 150 g kg?1 and of ZnO was 2 g kg?1. There was a significant increase (p < 0.05) in ADG with increasing levels of LF70 during the starter period. The inclusion of ZnO during the starter period resulted in an increase in ADG (p < 0.001) and FCR (p < 0.05) compared with no ZnO inclusion. Neither the inclusion of zinc oxide not of LF70 had an effect (p > 0.05) on performance during the transition period. In conclusion the supplementation of starter diets with increasing levels of LF70, ZnO and avilamycin resulted in increased ADG and improved FCR. The objective of this study was to determine the effects of processing and dietary inclusion level of flaxseed on broiler performance and nutrient utilisation. Flaxseed was included in the diet fed to day‐old broilers for the first 3 weeks as whole seed, ground seed, autoclaved whole seed, ground autoclaved whole seed or whole seed pelleted with the other ingredients, at levels of 0, 100, 120 and 140 g kg?1. Chicks fed the pelleted flaxseed‐containing diets had heavier body weights, consumed more feed and had better feed/gain ratios than those fed the other flaxseed‐containing diets during the 3 weeks period (P < 0.01). The flaxseed level in the diet also had very significant (P < 0.01) effects on body weight and feed/gain ratio at the end of weeks 1 and 3, with the diets containing 100 g kg?1 flaxseed resulting in better performance than the other flaxseed‐containing diets. Among the flaxseed containing diets, the pelleted diets led to higher apparent ether extract digestibilities, with values of 778 and 770 g kg?1 for the diets containing 100 and 140 g kg?1 flaxseed respectively. This better utilisation of ether extract by young broilers may explain the significantly (P < 0.05) higher AMEn (apparent metabolisable energy) value of 2924 kcal kg?1 for the pelleted 140 g kg?1 flaxseed‐containing diet. This was 15.4–17.5% higher than for the diets with the same level of flaxseed but provided as raw or autoclaved whole seed. The pelleting of flaxseed allowed an inclusion rate of 100 g kg?1 without any reduction in the performance and nutrient utilisation of broilers. The aim of the study was to determine whether endogenous nitrogen and amino acid excretions at the terminal ileum change over time in the growing pig fed a protein‐free diet for 8 days. Seven entire male pigs with an overall mean live weight of 81.6 kg (SEM 3.3 kg) and surgically implanted post‐valve T caecum cannulas were fed a semi‐synthetic casein‐based diet for 8 days. Food was withheld from the pigs for 24 h, after which they were fed a protein‐free diet for a further 8 days at a rate of 10% of metabolic body weight per day. Chromic oxide was included in the protein‐free diet as an indigestible marker. Ileal digesta were collected continuously from 13:00 to 18:00 h on each day of the experimental period. Endogenous ileal nitrogen flows were determined for each pig each day the protein‐free diet was given, and endogenous ileal amino acid flows for the first and eighth days. There were no significant (P > 0.05) effects of the duration of feeding of the protein‐free diet on endogenous ileal total nitrogen or amino acid flows, except for the amino acids glycine and cysteine, the flows of which significantly decreased over the 8 day period (P < 0.01 and P < 0.05 for glycine and cysteine respectively), from (mean ± SEM) 1639 ± 217 to 892 ± 212 µg g−1 dry matter intake (DMI) for glycine and from 173 ± 13 to 127 ± 19 µg g−1 DMI for cysteine. The relative contributions (moles of each amino acid as a proportion of total moles of amino acids) of threonine, glycine and cysteine decreased significantly (P < 0.05) and that of proline increased significantly (P < 0.05) during the 8 days that the protein‐free diet was fed to the pigs. © 2000 Society of Chemical Industry  相似文献
