A year-long collection of maize-based animal feed samples from the National Milling Company and mouldy maize collected from farmers fields near Lusaka were analysed for Fusarium mycotoxins. In the survey, 148 samples were tested for zearalenone, deoxynivalenol and nivalenol, and 60 samples for T-2 toxin and diacetoxyscirpenol. Zearalenone was present up to 0.6 mg kg?1 in 17% of the feed samples, and deoxynivalenol was found at I-0 mg kg?1 in 1.4 % of these samples. This is the first report of these toxins in animal feeds in Zambia. Zearalenone was also found in 57.6 % of the 33 mouldy maize samples collected at levels ranging from 0.08 to 6.0 mg kg?1 (mean concentration 1.11 mg kg?1), and 49.5% of these samples contained deoxynivalenol at levels ranging from 0.5 to 16.0 mg kg?1 (mean concentration 5.56 mg kg ?1). The occurrence of mycotoxins in 140 maize silages, 120 grass silages and 30 wheat silages produced in the Netherlands between 2002 and 2004 was determined using a liquid chromatography coupled with tandem mass spectrometry detection (LC-MS/MS) multi-method. Deoxynivalenol (DON) was detected above the limit of quantification (LOQ) of 250 μg kg?1 in 72% of maize and 10% of wheat silages. Average DON concentrations were 854 and 621 μg kg?1, respectively, and maximum concentrations 3142 and 1165 μg kg?1, respectively. Zearalenone was detected above the LOQ of 25 μg kg?1 in 49% of maize and 6% of grass silages. Average zearalenone concentrations were 174 and 93 μg kg?1, respectively, and maximum concentrations 943 and 308 μg kg?1, respectively. The incidences and average concentrations of DON and zearalenone in maize silage were highest in 2004. The incidence of other mycotoxins was low: fumonisin B1 and 15-acetyl-DON were detected in 1.4 and 5% of maize silages, respectively, and roquefortin C in 0.8% of grass silages. None of the silages contained aflatoxins, ochratoxin A, T2-toxin, HT2-toxin, sterigmatocystin, diacetoxyscirpenol, fusarenon-X, ergotamine, penicillinic acid, or mycophenolic acid. This study demonstrates that maize silage is an important source of DON and zearalenone in the diet of dairy cattle. Since the carryover of these mycotoxins into milk is negligible, their occurrence in feed is not considered to be of significant concern with respect to the safety of dairy products for consumers. Potential implications for animal health are discussed.  相似文献

Peanut, sorghum and maize samples were collected from the wholesale market in Mysore, India, over a period of one year (October 1984 to September 1985). The samples were analysed for the natural occurrence of T-2 toxin (T-2), diacetoxyscirpenol (DAS) and zearalenone by thin-layer chromatography, dermal toxicity test and gas chromatography. Of the total number of peanut samples analysed, 6.9% were positive for the toxic trichothecene(s) (T-2, up to 38.89 mg kg?1; DAS, up to 2.03 mg kg?1); 4.8% of total sorghum samples analysed contained T-2 up to 15 mg kg?1. Zearalenone was not found in any of the samples tested, and no toxins were detected in any of the maize samples. Samples marketed during winter and summer periods were contaminated with the toxins. All the toxin-positive samples except one peanut sample were found in produce stored for more than a week.  相似文献

Metabolites of toxigenic fungi and bacteria occur as natural contaminants (e.g. mycotoxins) in feedstuffs making them unsafe to animals. The multi-toxin profiles in 58 commercial poultry feed samples collected from 19 districts in 17 states of Nigeria were determined by LC/ESI–MS/MS with a single extraction step and no clean-up. Sixty-three (56 fungal and seven bacterial) metabolites were detected with concentrations ranging up to 10,200?µg?kg-1 in the case of aurofusarin. Fusarium toxins were the most prevalent group of fungal metabolites, whereas valinomycin occurred in more than 50% of the samples. Twelve non-regulatory fungal and seven bacterial metabolites detected and quantified in this study have never been reported previously in naturally contaminated stored grains or finished feed. Among the regulatory toxins in poultry feed, aflatoxin concentrations in 62% of samples were above 20?µg?kg?1, demonstrating high prevalence of unsafe levels of aflatoxins in Nigeria. Deoxynivalenol concentrations exceeded 1000?µg?kg?1 in 10.3% of samples. Actions are required to reduce the consequences from regulatory mycotoxins and understand the risks of the single or co-occurrence of non-regulatory metabolites for the benefit of the poultry industry.  相似文献

Fusarium species (spp.) and fumonisin B1 (FB1) contaminations were monitored in maize and porridge consumed by a rural population of Limpopo Province, South Africa. Faecal samples were also analysed for FB1 as a means of estimating the degree of dietary exposure to this mycotoxin. In total, 142 samples of maize (n?=?54), porridge (47) and faeces (41) were screened for Fusarium spp. using a serial dilution technique followed by DNA sequencing, while FB1 was further screened and quantified by thin-layer chromatography (TLC) and high-performance liquid chromatography (HPLC), respectively. At least four species of Fusarium were identified, of which F. verticillioides was the most prevalent in all three sample types analysed. The contamination levels of FB1 were significantly higher in 87% of maize sampled (range?=?101–53,863?µg?kg?1) as compared with porridge (74% incidence rate; range?=?0.2–20?µg?kg?1) and faecal samples (100% incidence rate; range?=?0.3–464?µg?kg?1). Thus, it can be deduced that the level of human exposure to FB1 via the consumption of maize was high as several samples contained levels exceeding 1000?µg?kg?1, which was strongly supported by the levels found in faecal samples. Further data revealed that a high proportion of FB1 is destroyed or removed by processing maize into porridge. As maize porridge is consumed as a staple, the low levels found provide a means to limit exposure to FB1. Levels of FB1 found in the faeces which were higher indicate that other foods contaminated with the toxin are also consumed.  相似文献

A sensitive LC–MS/MS method for the simultaneous determination of type A, B and D trichothecenes in cereals is presented. The limits of detection ranged between 0.1 and 0.7 µg kg?1 for all analytes. The method was applied to 289 representatively drawn samples of wheat, rye and oat products. Ninety-four percent of the wheat samples (n = 130), 95% of the rye samples (n = 61) and 100% of the oat samples (n = 98) were contaminated with the type A trichothecenes T-2 and HT-2 toxin. Median levels of T-2/HT-2 (sum of the toxins) were 0.91, 0.53 and 8.2 µg kg?1, respectively. Highest levels were found in wheat bran (24 µg kg?1), rye kernels (3.1 µg kg?1) and oat flakes (85 µg kg?1). All wheat and rye samples and 75% of the oat samples were contaminated with the type B trichothecene deoxynivalenol. Median levels of this toxin were 23, 15 and 0.53 µg kg?1, respectively. Highest levels were found in wheat bran (1160 µg kg?1), rye kernels (288 µg kg?1) and oat flakes (55 µg kg?1). The type B trichothecene nivalenol was detected in 67% of the wheat samples, in 3% of the rye samples and in 24% of the oat samples with highest levels in wheat bran (96 µg kg?1), rye kernels (1.8 µg kg?1) and in oat flakes (17 µg kg?1), respectively. Levels of other type A and B trichothecenes played a minor role, although the rates of contamination were often high. Neither macrocyclic type D trichothecenes (satratoxin G and H, verrucarin A, roridin A) nor diacetylverrucarol and verrucarol (type A trichothecenes), were detected in any of the samples.  相似文献

This study determined maize-user practices that influence the presence of fumonisin and aflatoxin contamination of maize in food consumed in the rural areas of Tanzania. Samples of the 2005 maize harvest in Tanzania were collected from 120 households and examined for fumonisins and aflatoxins. Information on whether the maize was sorted to remove defective (visibly damaged or mouldy) maize before storage and whether the damaged and mouldy maize or the non-dehulled maize was used as food was also collected. In addition, the percentage of defective kernels in the samples was determined. Ninety per cent of the households sorted out defective maize, 45% consumed the defective maize and 30% consumed non-dehulled maize. In 52% of the samples fumonisins were determined at levels up to 11,048 µg kg?1 (median = 363 µg kg?1) and in 15% exceeded 1000 µg kg?1; the maximum tolerable limit (MTL) for fumonisins in maize for human consumption in other countries. Aflatoxins were detected in 18% of the samples at levels up to 158 µg kg?1 (median = 24 µg kg?1). Twelve per cent of the samples exceeded the Tanzanian limit for total aflatoxins (10 µg kg?1). Aflatoxins co-occurred with fumonisins in 10% of the samples. The percentage defective kernels (mean = 22%) correlated positively (r = 0.39) with the fumonisin levels. Tanzanians are at a risk of exposure to fumonisins and aflatoxins in maize. There is a need for further research on fumonisin and aflatoxin exposure in Tanzania to develop appropriate control strategies.  相似文献

In Mexico, maize tortillas are consumed on a daily basis, leading to possible aflatoxin exposure. In a survey of 396 2-kg samples, taken over four sampling days in 2006 and 2007 from tortilla shops and supermarkets in Mexico City, aflatoxin levels were quantified by HPLC. In Mexico, the regulatory limit is 12?µg?kg?1 total aflatoxins for maize tortillas. In this survey, 17% of tortillas contained aflatoxins at levels of 3–385?µg?kg?1 or values below the limit of quantification (<LOQ) and, of these, 13% were >12?µg?kg?1 and 87% were below the regulatory limit. Average aflatoxin concentrations in 56 contaminated samples were: AFB1 (12.1?µg?kg?1); AFB2 (2.7?µg?kg?1); AFG1 (64.1?µg?kg?1) and AFG2 (3.7?µg?kg?1), and total AF (20.3?µg?kg?1).  相似文献

A survey was undertaken of a total of 120 farmers, 40 from each of the three studied agro-ecological zones of Tanzania, to determine local post-harvest management practices associated with aflatoxin (AF) and fumonisin (FB) contamination of maize. Data on practices (collected using a structured questionnaire) and maize samples were obtained from each of the 120 farmers. FB and AF contamination in the samples were analysed by HPLC. A total of 45% and 85% of maize samples were positive for AF and FB respectively, with levels ranging from 0.1 to 269 μg kg?1 for AF and from 49 to 18 273 μg kg?1 for FBs. Significant differences in contamination level were observed among the three agro-ecological zones. Farmers in the three agro-ecological zones practised similar practices in varying degrees. Drying, sorting and protecting maize against insect infestation are practices that showed significant association with AF or FB contamination of maize. Drying maize on mat/raised platform, sorting (damaged, discoloured and moulded grains) and application of synthetic insecticides during storage are practices that were associated with less contamination of maize with AF and FB. The results can be used to advise on effective post-harvest strategies for prevention of AF and FB contamination of maize in rural Tanzania.  相似文献

Fumonisin B1 (FB1) and fumonisin B2 (FB2) are the most abundant fumonisins (FBs) occurring worldwide in maize, infected mainly by Fusarium verticillioides and F. proliferatum. A total of 307 corn kernel samples were collected from 45 districts of Gansu, Shandong, Ningxia and the Inner Mongolia provinces of the north and northwest China. The samples were analysed for FB1 and FB2 by high-performance liquid chromatography. The FBs (FB1+ FB2) incidence rate in samples from Gansu, Shandong, Ningxia and Inner Mongolia were 31.5%, 81.1%, 46.2% and 53.6%, respectively. Average FBs concentration was 703 μg/kg and the concentrations ranged from ≤11 to 13,110 μg kg?1. Results were compared with the European Commission (EC) regulation for FB1+ FB2 in unprocessed maize for human consumption of 4 mg kg?1. Contamination in 17 samples was higher than these levels. More than 80% of the samples from Liaocheng county, which is located in the Shandong province, were contaminated with FBs, with a mean total FB concentration of 2496 μg/kg. The result was significantly different from that of the Inner Mongolia (1399 μg/kg), Ningxia (373 μg/kg) and Gansu (175 μg/kg). Average exposure to FBs (0.12 μg/kg body weight/day) is within the provisional maximum tolerable daily intake of 2.0 mg/kg of body weight set by the Joint Food and Agriculture Organization and World Health Organization Expert Committee on Food Additives.  相似文献

Aflatoxicosis has repeatedly affected Kenyans, particularly in the eastern region, due to consumption of contaminated maize. However, save for the cases of acute toxicity, the levels of sub-lethal exposure have not been adequately assessed. It is believed that this type of exposure does exist even during the seasons when acute toxicity does not occur. This study, therefore, was designed to assess the exposure of households to aflatoxins through consumption of maize and maize products. Twenty samples each of maize kernels, muthokoi and maize meal were randomly sampled from households in Kibwezi District of Makueni County in Eastern Kenya and analysed for aflatoxin contamination. The samples were quantitatively analysed for aflatoxin contamination using HPLC. The uncertainty and variability in dietary exposure was quantitatively modelled in Ms Excel using Monte Carlo simulation in @Risk software. Aflatoxins were found in 45% of maize kernels at between 18 and 480 μg kg–1, 20% of muthokoi at between 12 and 123 μg kg–1, and 35% of maize meal at between 6 and 30 μg kg–1. The mean dietary exposure to aflatoxin in maize kernels was 292 ± 1567 ng kg?1 body weight day?1, while the mean dietary exposure to aflatoxin in maize meal and muthokoi were 59 ± 62 and 27 ± 154 ng kg?1 body weight day?1 respectively. The results showed that the amount and frequency of consumption of the three foods is the more important contributing factor than the mean aflatoxin concentration levels, to the risk of dietary exposure to aflatoxins.  相似文献

This study reports for the first time in Serbia the occurrence of patulin in apple-based food and the risk assessment associated with patulin intake by infants and preschool children. In total, 214 samples of infant fruit juices (48), infant purée (66), and juices for children (small package with straw, 100) were collected over 3 years (2013–15) and analysed using HPLC with ultraviolet detection. Patulin was found in 43.8% of infant juices and 16.7% of infant purée, with all values below the legal limit of 10 μg kg?1 (maximum 8.3 and 7.7 μg kg?1, respectively). The proportion of contaminated samples among fruit juices for children was 43.0%, with the highest patulin concentration at 30.2 μg kg?1, not exceeding the maximum allowed level of 50 μg kg?1. Risk assessment of patulin intake by Serbian infants and preschool children, conducted by deterministic and probabilistic approaches, revealed a hazard quotient well below 1, indicating a tolerable exposure level and no health concern.  相似文献

Total mercury (THg) levels in 440 pairs of milled rice samples and brown rice samples from 15 major rice grain-producing provinces of China were measured and the associated health risk via rice consumption for different age categories of Chinese population was also assessed. THg contents were measured by a direct mercury analyser and the limit of detection (LOD) was 1.5 μg kg?1. The THg levels for milled rice samples and brown rice samples varied from non-detected to 17.8 μg kg?1 and 1.5 to 25.4 μg kg?1, respectively, with a mean level of 3.4 μg kg?1 and 4.9 μg kg?1, respectively. The THg levels in all milled and brown rice samples were generally low, except three brown rice samples having concentrations above the legally set value for cereals (20 μg kg?1 Hg). THg intakes for different age categories were estimated according to THg content and corresponding rice consumption and the associated health risk was evaluated by the corresponding provisional tolerable weekly intake (PTWI) for THg (5.0 μg kg?1 bw week?1), which was established by the Joint FAO/WHO Expert Committee on Food Additives (JECFA). The 50th percentile of the THg intakes via milled rice and brown rice consumption for different age categories was in the range 0.09–0.19 μg kg?1 bw week?1 and 0.14–0.27 μg kg?1 bw week?1, respectively, well below the PTWI, suggesting that the associated health risk is relatively low. However, the 99.9th percentile of the THg intakes for 2–4-year-old children amounted up to 20.6% of the PTWI (milled rice) and 29.5% of the PTWI (brown rice), which deserves attention.  相似文献

A total of 178 milk samples (94 of buffalo and 84 of cow) were randomly taken from Punjab and the North West Frontier Province (NWFP) of Pakistan (n?=?89 in each province) and analysed for the presence of aflatoxin M1 (AFM1) by HPLC-FLD. From Punjab about 46% of buffalo's and 49% of cow's milks were contaminated with AFM1 as compared with 52% and 51% for milk samples from NWFP, respectively. Overall, the mean AFM1 concentration was 0.046?µg?kg?1 with a maximum of 0.350?µg?kg?1. All samples complied with the Codex Alimentarius limit of 0.50?µg?kg?1 for AFM1 in milk, but 16.3% of samples exceeded the European Union maximum level of 0.05?µg?kg?1. Another set of 415 buffalo's and cow's milk samples (213 morning milks and 202 evening milks) were analysed. Statistical analysis revealed significant differences (p?<?0.05) between mean AFM1 concentrations in milk during the morning (0.043?µg?kg?1) and the evening (0.028?µg?kg?1) lactation times.  相似文献

A total of 64 samples of sorghum (37 Tunisian sorghum samples and 27 Egyptian sorghum samples) were collected during 2011–2012 from markets in Tunisia. Samples were analysed for contamination with aflatoxin B1, ochratoxin A and zearalenone by High-Performance Liquid Chromatography Coupled with Fluorescence Detection (HPLC-FLD). Aflatoxin B1 was found in 38 samples in the range 0.03–31.7 µg kg?1. Ochratoxin A was detected in 24 samples with concentrations ranging from 1.04 to 27.8 µg kg?1. Zearalenone was detected in 21 samples and the concentration varied between 3.7 and 64.5 µg kg?1. ANOVA analysis of the influence of the country of origin on the incidence and concentration of mycotoxins in the samples studied showed no significant difference (P > 0.05) between the two batches of samples for each of the three mycotoxins studied. The studied mycotoxins contaminate sorghum and may also co-exist because of the diversity of the mycobiota in this cereal.  相似文献

The aim was to evaluate the distribution of aflatoxins and fumonisins in fractions derived from the dry-milling of contaminated maize. Two maize lots with different contamination levels were processed and sampled: the first (maize 1) had aflatoxin B1 (AFB1) and fumonisin B1 (FB1) levels of 3.6 and 5379 µg kg?1, respectively; the second (maize 2) had corresponding levels of 91.1 and 8841 µg kg?1, respectively. The cleaning step reduced AFB1 and FB1 levels by 8 and 11% in maize 1 and by 57 and 34% in maize 2. The subsequent removal of bran and germ led to a further decrease in contamination levels in the products destined for human consumption. In the latter, AFB1 was uniformly distributed, while FB1 was concentrated in the finer size fractions. Contamination of raw maize 1 (3.6 µg kg?1) was below the European Union AFB1 limit of 5 µg kg?1 for unprocessed maize, but among the final products only coarse flour (1.7 µg kg?1) was within the European Union limit of 2 µg kg?1, while grits and fine flour showed higher levels (2.7 and 2.5 µg kg?1, respectively). As regards cleaned maize, a different distribution of the two toxins was observed in the kernels: AFB1 contamination was more superficial and concentrated in germ, while FB1 contamination affected the inner layers of the kernels.  相似文献

In this study, we investigated the potential of aflatoxin B1 (AFB1) production by five Aspergillus flavus strains previously isolated from sorghum grains on cereals (barley, maize, rice, wheat and sorghum), oilseeds (peanuts and sesame) and pulses (greengram and horsegram). Five strains of A. flavus were inoculated on all food grains and incubated at 25 °C for 7 days; AFB1 was extracted and estimated by enzyme‐linked immunosorbent assay. All A. flavus strains produced AFB1 on all food grains ranging from 245.4 to 15 645.2 μg kg?1. Of the five strains tested, strain Af 003 produced the highest amount of AFB1 on all commodities ranging from 2245.2 to 15 645.2 μg kg?1. Comparatively, the AFB1 accumulation was high on rice grains ranging from 3125.2 to 15 645.2 μg kg?1, followed by peanuts ranging from 2206.2 to 12 466.5 μg kg?1. Less AFB1 accumulation was observed in greengram and sesame seeds ranging from 645.8 to 2245.2 and 245.4 to 2890.6 μg kg?1, respectively. Our results showed that all food grains tested are susceptible to A. flavus growth and subsequent AFB1 production.  相似文献

No nitrosamines could be detected above a level of 1 μg kg?1 in five samples of indirectly dried skim-milk powders, 20 samples of directly-fired skim-milk powders and 10 samples of directly-fired butter-milk powders, selected at random from eight processing plants in New Zealand over a period of 8 months continuous production. The maximum level of N-nitrosodimethylamine (NDMA) observed in samples of milk proteins (lactic casein and lactalbumin) was 0.3 μg kg?1 (in one sample) while most of a further 10 samples showed traces of NDMA at the limit of detection of 0.1 μg kg?1.  相似文献

An easy‐to‐use and simplified liquid chromatography‐tandem mass spectrometry (LC‐ESI (+)‐MS/MS) method was adopted to determine the amount of acrylamide in thirty‐two samples of potato chips purchased on the South Italian market in 2009. Extract produced from matrices was directly analysed without derivatisation, the use of isotopically labelled acrylamide or the use of clean up cartridges. Quality analytical data were reported. The limit of detection and limit of quantification were 6 μg kg?1 and 18 μg kg?1, respectively and recovery values ranged from 90.7–96.3%. The relative standard deviation (RSD) ranged between 2.1% and 5.8%. The values ranged between 27 and 1400 μg kg?1 and the arithmetic mean acrylamide content resulted 363 μg kg?1. According to Foot et al. [Food Additives and Contaminants, 24 (2007)(S1), 37] and considering 500 μg kg?1 as the minimum level possible with the actual available mitigation tools, the number of samples showing an acrylamide level higher than 500 μg kg?1 resulted to be 22%. This paper gives a contribution to data collection in response to the monitoring activity of EFSA and following the Commission Recommendation 2007/331/EC: really, the number of Italian samples of potato chips and potato products for which acrylamide values was reported in the ‘results on the monitoring of acrylamide’ of EFSA ( http://www.efsa.europa.eu/cs/BlobServer/Report/datex_report_acrylamide_en,0.pdf?ssbinary=true , 2009) issued on 30 April 2009 is very limited.  相似文献

Thirty-seven different samples of canned sardines and other fish sold in the United Kingdom were analysed for their furan content using a validated automated headspace gas chromatography–mass spectrometry procedure. All 37 samples contained detectable furan, with an average level of 26 μg kg?1. The maximum furan content was in canned fish containing tomato sauce, which had an average of 49 μg kg?1 and in canned fish packed with lemon which had an average of 55 μg kg?1. All fish in brine or in oil contained less than 20 μg kg?1 furan. Furan levels recorded in fish packed in extra virgin olive oil were low with an average of 2 μg kg?1.  相似文献
