Tissue engineering is a new approach for regeneration of damaged tissues. The current clinical methods such as autograft and allograft transplantation are not effective for repairing bone damages, mainly due to the limited available sources and the donor-site side effects. In this research, the nanocomposite poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV)/nano hydroxyapatite (nHA) scaffolds with different nHA ratios for bone regeneration were utilized. The diameter and porosity of scaffolds were approximately 200?nm and 74%, respectively. The degradability test of the scaffolds suggests a low degradation rate with total degradation of 30% after 3 months. Cytotoxicity result showed that cultured osteoblast cells (MC3T3) on nanocomposite scaffolds had superiority in terms of higher proliferation and attachment in comparison with PHBV scaffold. The protein expression of alkaline phosphatase illustrated that nanofibrous scaffold containing hydroxyapatite had the highest alkaline phosphatase activities as a result of better proliferation. These results recommend that PHBV/nHA scaffolds are suitable candidates for bone tissue engineering.  相似文献

A series of nanofibrous scaffolds were prepared by electrospinning of poly(vinyl alcohol) (PVA)/gelatin aqueous solution. PVA and gelatin was dissolved in pure water and blended in full range, then being electrospun to prepared nanofibers, followed by being crosslinked with glutaraldehyde vapor and heat treatment to form nanofibrous scaffold. Field emission scanning electron microscope (FESEM) images of the nanofibers manifested that the fiber average diameters decreased from 290 to 90 nm with the increasing of gelatin. In vitro degradation rates of the nanofibers were also correlated with the composition and physical properties of electrospinning solutions. Cytocompatibility of the scaffolds was evaluated by cells morphology and MTT assay. The FESEM images revealed that NIH 3T3 fibroblasts spread and elongated actively on the scaffolds with spindle‐like and star‐type shape. The results of cell attachment and proliferation on the nanofibrous scaffolds suggested that the cytotoxicity of all samples are grade 1 or grade 0, indicating that the material had sound biosafety as biomaterials. Compared with pure PVA and gelatin scaffolds, the hybrid ones possess improved biocompatibility and controllability. A series of herbal extract incorporated into poly(lactic acid) (PLA) composite nanofibrous scaffolds were successfully prepared by using electrospinning technique. Equisetum arvense extract (EE) and nanohydroxyapatite (nHA) in different quantities were loaded into PLA solution to fabricate composite nanofibrous webs under various electrospinning conditions. Uniform nanofibers were obtained with an average diameter of 157 ± 47 nm in the case of those containing the herbal extract. Characterization of the webs was carried out by means of Fourier transform infrared (FTIR) spectroscopy, field emission‐scanning electron microscopy (FESEM), energy‐dispersive X‐ray spectroscopy (EDX), and differential scanning calorimetry (DSC) techniques. Mechanical properties, porosity, and contact angle of the prepared webs were also determined. Releasing behavior was investigated in phosphate buffer solution (pH 7.2) medium. Moreover, cell studies and osteogenic capacity were assessed in vitro using human adipose tissue‐derived mesenchymal stem cell (AT‐MSC). Evaluations of cell attachment, spreading, and proliferation of AT‐MSC were done by SEM observation and thiazolyl blue (MTT) assay. Osteogenic differentiation capability of AT‐MSC on the nanofibrous webs was analyzed by alkaline phosphatase activity and calcium content assay. It was found that with the addition of nHA and EE to PLA nanofibrous webs, their surface hydrophobicity was reduced while the tensile strength and Young's modulus were increased satisfactorily. Regarding the samples containing EE and nHA, cellular adhesion was observed with flattened normal morphology. Osteogenic differentiation of AT‐MSC on PLA/nHA/EE webs showed the highest mineralization capacity after 3 weeks which, was about 1.8 and 3 times higher than that of PLA/nHA and tissue culture polystyrene as control, respectively. © 2017 Wiley Periodicals, Inc. In this study, polyvinyl alcohol (PVA) fibers were modified through an effective cross linking method. Adequate porosity and surface area are widely recognized as important parameters in the design of scaffolds for tissue engineering and therefore measurement of porosity is very important. Herein, porosity measurement of various surface layers of scaffold was done through a new method, and image analysis was used for this purpose. Scanning electron microscopy micrographs of nanofibrous scaffolds were converted to binary images using different thresholds and porosity of scaffold was measured in various layers. In addition, for ascertaining of cross linking of the PVA nanofibrous scaffolds, Fourier transform infrared spectroscopy analysis was employed. Also, the in vitro biodegradability of the nanofibrous scaffold was evaluated. The PVA crosslinked nanofibrous scaffold was found to exhibit the most balanced properties to meet all the required specifications for nerve tissue and was used for in vitro culture of nerve stem cells (PC12 cells). Finally, the results of the swelling behavior of the samples revealed that the cross linked PVA scaffold has a strong swelling about 450%.  相似文献

Silk sericin (SS) has been extensively used to fabricate scaffolds for tissue engineering. However, due to its inferior mechanical properties, it has been found to be a poor choice of material when being electrospun into nanofibrous scaffolds. Here, SS has been combined with poly(vinyl alcohol) (PVA) and electrospun to create scaffolds with enhanced physical properties. Crucially, these SS/PVA nanofibrous scaffolds were created using only distilled water as a solvent with no added crosslinker in an environmentally friendly process. Temperature has been shown to have a marked effect on the formation of the SS sol–gel transition and thus influence the final formation of fibers. Heating the spinning solutions to 70 °C delivered nanofibers with enhanced morphology, water stability and mechanical properties. This is due to the transition of SS from β‐sheets into random coils that enables enhanced molecular interactions between SS and PVA. The most applicable SS/PVA weight ratios for the formation of nanofibers with the desired properties were found to be 7.5/1.5 and 10.0/1.5. The fibers had diameters ranging from 60 to 500 nm, where higher PVA and SS concentrations promoted larger diameters. The crystallinity within the fibers could be controlled by manipulation of the balance between PVA and SS loadings. In vitro degradation (in phosphate buffer solution, pH 7.4 at 37 °C) was 30–50% within 42 days and fibers were shown to be nontoxic to skin fibroblast cells. This work demonstrates a new green route for incorporating SS into nanofibrous fabrics, with potential use in biomedical applications. © 2019 Society of Chemical Industry  相似文献

Cartilage tissue engineering is one of the interesting approaches used for repairing cartilage injuries. This study reports the fabrication of polyvinyl alcohol/alginate sulfate (PVA/ALG-S) nanofibrous mats as a functional support for chondrogenic differentiation of human bone marrow mesenchymal stem cells (hBM-MSCs). The PVA/ALG-S nanofibers were obtained through electrospinning of PVA solutions containing 10, 20, and 30 wt% of ALG-S. The appearance of a band at 833 cm−1 assigned to the symmetrical C O S vibration associated to a C O SO3 group confirmed the presence of ALG-S in nanofibrous mat. The SEM images illustrated the bead-free and smooth morphology of PVA/ALG-S nanofibers with a mean diameter of 185 ± 0.06 nm. The MTT assay of the hBM-MSCs seeded on scaffolds indicated the appropriate cytocompatibility of nanofibrous PVA/ALG-S scaffolds. Furthermore, the appropriate attachment and spreading of the hBM-MSCs based on SEM images, and their differentiation to the chondrocyte-like cells accompanied by a decrease in cell growth on MTT analysis and more color absorption in alician blue staining indicated the effective role of alginate sulfate on cell differentiation. Finally, the expression of Type II collagen by RT-PCR and immunocytochemistry analyses revealed the chondrogenic differentiation of hBM-MSCs on alginate sulfate nanofibers.  相似文献

A biomimetic nanofibrous poly(L ‐lactide) scaffold strengthened by nanohydroxyapatite particles was fabricated via a thermally induced phase separation technique. Scanning electron microscopy results showed that nanohydroxyapatite particles uniformly dispersed in the nanofibrous poly(L ‐lactide) scaffold (50–500 nm in fiber diameter) with slight aggregation at a high nHA content, but showed no influence on the interconnected macroporous and nanofibrous structure of the scaffold. The nanofibrous poly(L ‐lactide) scaffold presented a specific surface area of 34.06 m2 g?1, which was much higher than that of 2.79 m2 g?1 for the poly(L ‐lactide) scaffold with platelet structure. Moreover, the specific surface area of the nanofibrous scaffold was further enhanced by incorporating nanohydroxyapatite particles. With increasing the nanohydroxyapatite content, the compressive modulus and amount of bovine serum albumin adsorbed on the surface of the nanofibrous composite scaffold were markedly improved, as opposed to the decreased crystallinity. In comparison to poly(L ‐lactide) scaffold, both the nanofibrous poly(L ‐lactide) and poly(L ‐lactide)/nanohydroxyapatite scaffolds exhibited a faster degradation rate for their much larger specific surface area. The culture of bone mesenchymal stem cell indicated that the composite nanofibrous poly(L ‐lactide) scaffold with 50 wt % nanohydroxyapatite showed the highest cells viability among various poly(L ‐lactide)‐based scaffolds. The strengthened biomimetic nanofibrous poly(L ‐lactide)/nanohydroxyapatite composite scaffold will be a potential candidate for bone tissue engineering. © 2012 Wiley Periodicals, Inc. J. Appl. Nanofibrous biocomposite scaffolds of chitosan (CS), PVA, and hydroxyapatite (HA) were prepared by electrospinning. The scaffolds were characterized by FTIR, SEM, TEM, and XRD techniques. Tensile testing was used for the characterization of mechanical properties. Mouse fibroblasts (L929) attachment and proliferation on the nanofibrous scaffold were investigated by MTT assay and SEM observation. FTIR, TEM, and XRD results showed the presence of nanoHA in the scaffolds. The scaffolds have porous nanofibrous morphology with random fibers in the range of 100–700 nm diameters. The CS/PVA (90/10) fibrous matrix (without HA) showed a tensile strength of 3.1 ± 0.2 MPa and a tensile modulus 10 ± 1 MPa with a strain at failure of 21.1 ± 0.6%. Increase the content of HA up to 2% increased the ultimate tensile strength and tensile modulus, but further increase HA up to 5–10% caused the decrease of tensile strength and tensile modulus. The attachment and growth of mouse fibroblast was on the surface of nanofibrous structure, and cells' morphology characteristics and viability were unaffected. A combination of nanofibrous CS/PVA and HA that mimics the nanoscale features of the extra cellular matrix could be promising for application as scaffolds for tissue regeneration, especially in low or nonload bearing areas. © 2008 Wiley Periodicals, Inc. Hydroxyapatite (HA), the bone mineral and Cissus quadrangularis (CQ), a medicinal plant with osteogenic activity, are attaining increasing interest as a potential therapeutic agent for enhanced bone tissue regeneration. In the present study a synergistic effect of these two agents were analyzed by fabricating PCL‐CQ‐HA nanofibrous scaffolds by electrospinning and compared with PCL‐CQ and PCL (control) nanofibrous scaffolds. Morphology, composition, hydrophilicity, and mechanical properties of the electrospun PCL, PCL‐CQ, PCL‐CQ‐HA nanofibrous scaffolds were examined by Field emission scanning electron microscopy (FESEM), Fourier transform infrared spectroscopy (FTIR), Contact angle and Tensile tests, respectively. The response of human foetal osteoblast cells on these scaffolds were evaluated using MTS assay, alkaline phosphatase activity, alizarin red staining, and osteocalcin expression for bone tissue regeneration. While the observed cellular response to both groups of scaffolds was better than for the control PCL scaffold, the PCL‐CQ‐HA nanofibrous scaffolds provided the most favorable substrate for cell proliferation and mineralization. The results showed that PCL‐CQ‐HA nanofibrous scaffolds had appropriate surface roughness for the osteoblast adhesion, proliferation, and mineralization comparing with other scaffolds. The observed investigation of physicochemical and biological properties suggests that the CQ‐HA loaded PCL nanofibrous scaffolds serve as a potential biocomposite material for bone tissue engineering. © 2013 Wiley Periodicals, Inc. Tissue engineering involves the fabrication of three‐dimensional scaffolds to support cellular in‐growth and proliferation. Ideally, the scaffolds should be similar to the native extracellular matrix (ECM). Electrospun polymer nanofibrous scaffolds are appropriate candidates for ECM mimetic materials since they mimic the nanoscale properties of ECM. Electrospun polymer nanocomposites based on poly(lactide‐co‐glycolide) (PLGA)/poly(vinyl alcohol) (PVA) and organically modified montmorillonite (OMMT) were prepared by a solution intercalation technique followed by electrospinning. The morphology of fibrous scaffolds based on these nanocomposites was investigated using scanning electron microscopy. The scaffolds showed highly porous structure within the nanofibres of diameters ranging from 400 to 700 nm. X‐ray diffractometry gave evidence of good dispersion of the OMMT in the blends with exfoliated morphology. Measurements of the water uptake and water contact angle of the fibrous scaffolds indicated significant improvement in the hydrophilicity of the scaffolds. Evaluations of the mechanical properties and unrestricted somatic stem cell culture of the electrospun fibrous nanocomposite scaffolds revealed that the PLGA90/PVA10/1.5% OMMT and PLGA90/PVA10/3% OMMT samples are the most useful from the tissue engineering application viewpoint. In this study, vascular stents were fabricated from poly (lactide-ɛ-caprolactone)/collagen/nano-hydroxyapatite (PLCL/Col/nHA) by electrospinning, and the surface morphology and breaking strength were observed or measured through scanning electron microscopy and tensile tests. The anti-clotting properties of stents were evaluated for anticoagulation surfaces modified by the electrostatic layer-by-layer self-assembly technique. In addition, nano-composite scaffolds of poly (lactic-co-glycolic acid)/polycaprolactone/nano-hydroxyapatite (PLGA/PCL/nHA) loaded with the vascular stents were prepared by thermoforming-particle leaching and their basic performance and osteogenesis were tested in vitro and in vivo. The results show that the PLCL/Col/nHA stents and PLGA/PCL/nHA nano-composite scaffolds had good surface structures, mechanical properties, biocompatibility and could guide bone regeneration. These may provide a new way to build vascularized-tissue engineered bone to repair large bone defects in bone tissue engineering.  相似文献

Nanofibrous biocomposite scaffolds of poly(vinyl alcohol) (PVA) and graphene oxide (GO) were prepared by using electrospinning method. The microstructure, crystallinity, and morphology of the scaffolds were characterized through X‐ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), and scanning electron microscopy (SEM). The mechanical properties were investigated by tensile testing. Moreover, Mouse Osteoblastic Cells (MC3T3‐E1) attachment and proliferation on the nanofibrous scaffolds were investigated by MTT [3‐(4,5‐dimeth‐ylthiazol‐2‐yl)‐2,5‐diphenyl tetrazolium bromide] assay, SEM observation and fluorescence staining. XRD and FTIR results verify the presence of GO in the scaffolds. SEM images show the three‐dimensional porous fibrous morphology, and the average diameter of the composite fibers decreases with increasing the content of GO. The mechanical properties of the scaffolds are altered by changing the content of GO as well. The tensile strength and elasticity modulus increase when the content of GO is lower than 1 wt %, but decrease when GO is up to 3 and 5 wt %. MC3T3‐E1 cells attach and grow on the surfaces of the scaffolds, and the adding of GO do not affect the cells' viability. Also, MC3T3‐E1 cells are likely to spread on the PVA/GO composite scaffolds. Above all, these unique features of the PVA/GO nanofibrous scaffolds prepared by electrospinning would open up a wide variety of future applications in bone tissue engineering and drug delivery systems. © 2012 Wiley Periodicals, Inc. 兼具良好孔隙率和原位任意塑形固化的可注射复合多孔骨修复材料在临床不规则骨缺损的治疗方面显示出巨大的优势。本研究通过优化双组分设计,以水为发泡剂制备可注射纳米羟基磷灰石/聚氨酯(nHA/PU)复合多孔支架。利用扫描电子显微镜(SEM)、傅里叶变换红外光谱(FTIR)、X射线衍射(XRD)、力学测试及Gillmore针测试等手段对制备的支架进行结构形貌、化学组成、力学性能和固化时间表征。结果表明,本研究制备的可注射nHA/PU复合多孔支架孔隙率高、孔隙贯通性好,孔径分布在100~700μm,适宜细胞和组织向孔内生长;添加20% nHA显著提高了PU支架的力学强度,但降低了支架的孔隙率;可注射支架在8h固化,适宜临床操作。本研究制备的可注射nHA/PU复合多孔支架在不规则骨缺损修复领域具有较大的应用潜力。  相似文献

The simultaneous effect of electrospun scaffold alignment and polymer composition on chondrogenic differentiation of human bone marrow mesenchymal stem cells (hBMMSC) is investigated. Aligned and randomly oriented polycaprolactone/poly(lactic-co-glycolic acid) (PLGA) hybrid electrospun scaffolds with two different ratios are fabricated by electrospinning. It is found that aligned nanofibrous scaffolds support higher chondrogenic differentiation of hBMMSCs compared to random ones. The aligned scaffolds show a higher expression level of chondrogenic markers such as type II collagen and aggrecan. It is concluded that the aligned nanofibrous scaffold with higher PLGA ratio could significantly enhance hBMMSC proliferation and differentiation to chondrocytes.  相似文献

Corneal ulcer, which is brought on by a breach in the epithelial barrier, is a dangerous infection of the avascular corneal stroma. New treatment strategies are needed, suppressing the aggressive nature of the disease and including a combination of different drugs. In this study, vancomycin (VAN) and fluconazole (FLU) dual-drug loaded dual-layered polyvinyl alcohol and gelatin (PVA/GEL) nanofibrous patches are produced by electrospinning. Scanning electron microscopy (SEM) images show smooth surfaces are obtained for both pure and drug-loaded nanofibrous patches. The tensile test results report that loading the FLU and VAN separately into the PVA/GEL patches decrease both the tensile strength and elongation at break and it is further reduced when combining two drug-loaded layers in one patch. According to drug release results, the FLU and VAN-loaded nanofibrous patches show a controlled release profile extending up to 96 h. Moreover, PVA/GEL/FLU, PVA/GEL/VAN, and PVA/GEL/FLU/VAN nanofibrous patches display significant antimicrobial activity against Candida albicans and Staphylococcus aureus. SEM, 4'-6diamidynofenyloindol (DAPI) staining, and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay show that PVA/FLU and PVA/GEL/FLU/VAN nanofibrous patches have a superior effect on NIH3T3 cell spreading and proliferation. The novelty of this study lays in the development of a potential dual drug rapid treatment for corneal ulcers of aggressive nature.  相似文献

Three-dimensional silk fibroin impregnated poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) nanofibrous scaffolds with or without hydroxyapatite (HAp) were prepared by wet-electrospinning method followed by freeze-drying. Scaffolds with cotton wool-like structure have the average fiber diameter of 450–850?nm with 80–85% porosity. In-vitro cell culture tests using MG-63 osteosarcoma human cells revealed improved cell viability, alkaline phosphatase (ALP) activity and total cellular protein amount on the silk impregnated scaffolds compared to PHBV and HAp/PHBV scaffolds after 10 days of cell culture. Immunohistochemical analyses on the silk impregnated scaffolds showed that HAp triggered cell penetration and type I collagen production. Besides, HAp mineralization tendency increased with a decrease in percent crystallinity of the scaffolds comprising HAp and silk after 4 weeks of incubation in simulated body fluid. Consequently, cotton wool-like HAp/PHBV-SF scaffold would be a promising candidate as a bone-filling material for tissue regeneration.  相似文献

Nanoengineered biodegradable constructs based on synthetic and natural polymers enriched with hydroxyapatite (HA) nanoparticles have been found to mimic the extracellular matrix of bone tissue. The main objective of this study was to create biocomposite nanostructured scaffolds by incorporating collagen and HA nanoparticles into poly(L-lactic acid)-co-poly(?-caprolactone) by electrospinning. The fiber diameter of the composite PLCL/Col and PLCL/Col/HA fibers was smaller compared to PLCL. In vitro biocompatibility of the scaffolds studied using human fetal osteoblasts and EDX analysis showed high deposition of calcium on PLCL/Col/HA. The results shows that PLCL/Col/HA nanofibrous constructs have huge potential as substrates for bone regeneration.  相似文献

In the present study, polylactic acid (PLA)/polyethylene glycol (PEG)/multiwalled carbon nanotube (MWCNT) electrospun nanofibrous scaffolds were prepared via electrospinning process and their applications for the anticancer drug delivery system were investigated. A response surface methodology based on Box–Behnken design (BBD) was used to evaluate the effect of key parameters of electrospinning process including solution concentration, feeding rate, tip–collector distance (TCD) and applied voltage on the morphology of PLA/PEG/MWCNT nanofibrous scaffolds. In optimum conditions (concentration of 8.15%, feeding rate of 0.2 mL/h, voltage of 18.50 kV and TCD of 13.0 cm), the minimum experimental fiber diameter was found to be 225 nm which was in good agreement with the predicted value by the BBD analysis (228 nm). In vitro drug release study of doxorubicin (DOX)‐loaded nanofibrous scaffolds, higher drug content induced an extended release of drug. Also, drug release rate was not dependent on drug/polymer ratio in different electrospun nanofibrous formulations. The equation of Mt = c0 + kt0.5was used to describe the kinetic data of DOX release from electrospun nanofibers. The cell viability of DOX‐loaded nanofibrous scaffolds was evaluated using 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide, a tetrazole assay on lung cancer A549 cell lines. We propose that DOX‐incorporated PLA/PEG/MWCNT nanofibrous scaffold could be used as a superior candidate for antitumor drug delivery. © 2014 Wiley Periodicals, Inc. J. Appl. Polym. Many polymeric materials have been developed and introduced for bone regeneration. Especially, their nanofibrous forms are mostly applied for artificial extracellular matrices. Polymeric materials in their nanofibrous form show some potent properties such as high surface‐to‐volume ratio, tunable porosity, and ease of surface functionalization. Benefiting from the properties of their main polymer and additives, they can provide new opportunities for cell seeding, proliferation, and new 3D‐tissue formation. This article focuses on most cited polymeric nanofibrous scaffolds fabricated by electrospinning and recent achievements. They were divided into two main categories: natural (collagen, silk, keratin, gelatin, chitosan, and alginate) and synthetic (e.g., polycaprolactone, polylactic acid, and polyglycolic acid) polymers. The role of several additives like hydroxyapatite, bone morphogenetic proteins (BMPs), tricalcium phosphate, and collagen type I in improving the adhesion, differentiation, and tissue formation of stem cells were discussed. Nanofibrous scaffolds were obtained by co‐electrospinning poly (3‐hydroxybuty‐rate‐co‐3‐hydroxyvalerate) (PHBV) and fibroin regenerated from silk in different proportions using 1,1,1,3,3,3‐hexafluoro‐2‐isopropanol (HFIP) as solvent. Field emission scanning electron microscope (FESEM) investigation showed that the fiber diameters of the nanofibrous scaffolds ranged from 190 to 460 nm. X‐ray diffraction (XRD) and Fourier transform infrared spectroscopy analysis (FT‐IR) showed that the main structure of silk fibroin (SF) in the nanofibrous scaffold was β‐sheet. Compared to the PHBV nanofibrous scaffold, the surface hydrophilicity and water‐uptake capability of the PHBV/SF nanofibrous scaffold with 50/50 were improved. The results of cell adhesion experiment showed that the fibroblasts adhered more to the PHBV/SF nanofibrous scaffold with 50/50 than the pure PHBV nanofibrous scaffold. The proliferation of fibroblast on the PHBV/SF nanofibrous scaffold with 50/50 was higher than that on the pure PHBV nanofibrous scaffold. Our results indicated that the PHBV/SF nanofibrous scaffold with 50/50 may be a better candidate for biomedical applications such as skin tissue engineering and wound dressing. POLYM. ENG.
