The fatty acid composition of triglycerides and phospholipids from rat liver was determined throughout the period of growth in the rat. Major changes in the triglyceride fatty acid composition were observed during the period studied. The triglycerides from fetal and newborn rats contained only a small percentage of polyunsaturated acids compared with suckling and weanling rats. During the suckling phase the liver triglycerides were rich in long chain polyunsaturated acids such as 20∶4, 20∶5, 22∶5, and 22∶6; once the pups were weaned, the percentage of these acids in the liver triglycerides fell. In these experiments, 18∶2 and 18∶3 were the only polyunsaturated acids in the maternal diet. However, the stomach contents of the suckling pups contained 18∶2 and 18∶3, as well as the long chain polyunsaturated acids. Radioactive 18∶3 and 22∶6 were fed to suckling pups, and the results suggested that the LCP in the rat liver triglycerides during the suckling period were derived from the long chain polyunsaturated acids in the dam's milk, rather than by synthesis from either 18∶2 or 18∶3 within the pups.  相似文献

Cholesterol esterification by acyl-CoA:cholesterol acyltransferase (ACAT; EC 2.3.1.26) has been studied in microsomes isolated from the mammary glands of rats in late pregnancy, in early and mid-lactation, and following premature weaning. The mammary glands were freeze-clamped and the microsomes prepared in the presence of phosphatase inhibitors to preserve the phosphorylation status of the enzyme. Optimal conditions were established for the assay of enzyme activity in the presence of endogenous cholesterol. Supplementation of the microsomes with exogenous cholesterol as a dispersion in Triton WR-1339 was shown to lead to an increase in enzyme activity. Incubation of microsomes with MgATP led to an increase in ACAT activity which could be reversed by treatment of the microsomes with a phosphoprotein phosphatase preparation from rat liver. The results suggested that ACAT activity in the mammary gland was activated by phosphorylation in a similar way to that observed for the hepatic enzyme. The mammary glands from pregnant animals contained a higher level of ACAT activity than did the glands of the lactating animals and this correlated with the higher cholesteryl ester content of the pregnant glands. The highest level of ACAT activity was found in the weaned animals but the cholesteryl ester content of the microsomes was lower than expected. The influence of progesterone levels and changes in feeding patterns during gestation were considered as factors in these variations in ACAT activity.  相似文献

In view of the previously reported changes in the fatty acid composition of maternal liver triacylglycerols in late pregnancy, changes in the composition of maternal liver triacylglycerol species were assessed in rats fed a semipurified diet during pregnancy. Between day 18 and day 21 of pregnancy, total maternal liver triacylglycerols increased by 50%. Triacylglycerol species with a total acyl carbon number (C) of 50 or 60 (C50, C60) remained unchanged while C48 and C52–C58 were relatively increased. The individual triacylglycerol species containing one, two or three linoleoyl moieties were incompletely recovered using a polar high temperature gas-liquid chromatography (GLC) column. Nevertheless, at day 21 compared to day 18, the linoleoyl-containing species were relatively increased by 62–463%, while tripalmitin was decreased by 38%. Our data suggest that despite an adequate intake of linoleic acid (25 g/kg in the diet), maternal hepatic triacylglycerol content of linoleic acid decreased during mid-pregnancy but increased significantly toward term possibly in preparation for the transfer of linoleic acid to the neonate during lactation.  相似文献

Cabbage leaves contain 0.16% total lipids of which 51.02% are neutral lipids, 40.78% glycolipids, and 8.18% phospholipids. The predominant fatty acids in the total lipid analysis are linolenic, linoleic, oleic, palmitic, and stearic acids. Linolenic, palmitic, tridecanoic, and oleic are the principal components in the neutral lipid fraction while glycolipids are composed mainly of linolenic, palmitic, lauric, myristic, and tricosanoic acids. Phospholipids are high in palmitic, linolenic, and linoleic acids. Both glucose and galactose were observed in the glycolipid fraction.  相似文献

The fatty acids of liver lipids from rats raised on a fat free diet from the 30th to the 90th day after birth were analyzed with special regard to the detection of positional isomers of mono-, di-, tri-, and tetraenoic fatty acids. The methyl esters obtained after transesterification of total lipids were separated by argentation chromatography into five fractions: I saturated, II monoenoic, III dienoic, IV dienoic nonmethylene interrupted, V triand tetraenoic fatty acid esters. After hydroxylation of the double bonds with osmium tetroxide, the analysis of the poly-O-trimethylsilyl derivatives by gas liquid chromatography on S.C.O.T. columns combined with mass spectrometry revealed the presence of 19 monoenoic, 15 dienoic, and 9 trienoic as well as 3 tetraenoic fatty acid isomers including the normally occurring representatives of the (n−3), (n−6), (n−7), and (n−9) fatty acid families. The majority of the identified isomers can be coordinated to one of these families like 7–16∶1; 11–20∶1; 6,9–18∶2; 8,11–20∶2; 5,11–20∶2; 5,8,11–20∶3; 7,10,13–22∶3 to the (n−9) family, 11–18∶1; 13–20∶1; 5,11–18∶2; 7,13–20∶2; 6,11–18∶2; 6,9–16∶2; 8, 11–18∶2; 10,13–20∶2; 5,8,11–18∶3; 7,10,13–20∶3; 4,7,10,13–20∶4 to the (n−7) family and 11,14–20∶2; 5,11,14–20∶3; 6,9,12–18∶3; 8,11,14–20∶3; 5,8,11,14–20∶4; 7,10,13,16–22∶4 to the (n−6) family. All these naturally occuring isomers can be placed into a network of desaturation and chain elongation steps which allows certain conclusions about the substrate specificity of the Δ6-, Δ5-and Δ4-desaturase systems. The great number of isomers found in the (n−7) family indicates that the members of this family are actively metabolized in partial essential fatty acid deficiency.  相似文献

Recent literature on the composition of milk lipids is reviewed and discussed. Many additional exotic fatty acids, mainly branched chain, keto, hydroxy and isomers containing double bonds, have been identified, often with the aid of mass spectrometry. It is estimated that ca. 500 fatty acids have been found in milk lipids. Components of lipid classes have been isolated and their structure determined, e.g., glucosyl and lactosyl ceramide, sphingomyelin, ether lipids, etc. The fatty acid composition of “protected” milk is discussed. This milk, obtained from cows fed polyunsaturated oils encapsulated with formaldehyde-treated casein, contains about five times more linoleic acid than regular milk. The future of research on milk lipids is promising, as there are still many intriguing problems of separation and identification. Groups of normal and hepatoma (7288CTC) bearing rats were maintained on normal chow and fat-free diets for 4 weeks. Normal liver, host liver, and hepatoma neutral lipids were examined in detail and compared. Water content, unaffected by diet was: hepatoma, 82 percent; host liver, 71 percent; and normal liver, 67 percent. The fat-free diet had no effect upon the hepatoma neutral lipids but elevated the triglyceride level in normal and host liver, shifted the triglyceride carbon number distribution to lower mol wt, and elevated the percentage of monoenoic acids in triglycerides and cholesteryl esters. Host triglyceride concentrations were ca. half, and cholesterol levels were reduced moderately relative to normal liver values. Hepatoma cholesterol levels were higher and triglyceride concentrations lower than normal and host liver values. Hepatoma triglycerides differed dramatically from liver and were characterized by increased concentrations of high mol wt species and a fivefold increase in the percentage of C-20 and C-22 fatty acids. The percentage of C-20 and C-22 fatty acids in hepatoma cholesteryl esters also increased ca. fivefold relative to liver. The data indicate that the systems that regulate triglyceride and cholesteryl ester fatty acid composition in liver do not control the compositions of these lipid classes in this hepatoma. The unchanged high level of essential fatty acids in the hepatoma lipids from the fat-free fed animals demonstrates the hepatoma's ability to absorb and conserve specific fatty acids.  相似文献

In an attempt to correlate the incorporation of fatty acids (FA) of different chain length into liver and biliary lipids’ isolated rat livers were perfused for 2 h with Krebs-Ringer bicarbonate containing 1% albumin and 10 μmol of [1-14C]-labeled FA: C2’ C8’ C10’ C12’ C16’ and C18∶1. One to 1.36 μmol of medium-chain fatty acids (MCFA’ C8’ C10’ and C12) and 6.6 μmol of long-chain FA (LCFA) were incorporated into liver lipids’ 40% of the latter into phosphatidylcholine (PC). 14C-acetate (13 nmol) was incorporated into biliary cholesterol; 14C-MCFA contributed only 3.2–5 nmol; LCFA did not lead to newly synthesized cholesterol. Newly synthesized liver PC (2.75 to 3.25%) and newly synthesized liver cholesterol (6.5 to 10%) were secreted into bile. The specific radioactivity of biliary PC after infusion of all-saturated FA was 3.8–6.8 times higher than that of liver PC; for C18∶1 it was only 1.7-fold. The specific radioactivity of biliary cholesterol’ as compared to liver cholesterol’ was 12 times higher for C2 and five times higher for MCFA. This indicates that a considerable proportion of the newly synthesized lipids was secreted into bile prior to significant mixing with preexisting liver PC and cholesterol pools. liver PC contained 8% of unchanged 14C−C12; while 14C−C10 was not detected. Biliary PC’ in contrast’ contained 18% of unchanged 14C−C12 and 3% 14C−C10. These results suggest that after prolonged infusion of medium-chain triacylglycerols/longchain triacylglycerols to patients’ biliary PC may become enriched with MCFA. In addition’ the oxidation of these FA may provide C-2 units which increase cholesterol synthesis.  相似文献

Total lipids in medium and low erucic acid cul-tivars of rapeseed(Brassica napus var. Sinus and Janpol, resp.) were fractionated into polar and non-polar constituents. Triglycerides, diglycerides, mono-glycerides, free fatty acids, sterol esters, sterols, phos-pholipids and glycolipids were quantitated and their fatty acid compositions determined. Triglycerides and phospholipids constituted 92 and 3.4%, resp., of the total lipid from each cultivar. Triglycerides were lower in saturated fatty acids but higher in monoun-saturated acids and linolenic acid than other lipid fractions. Phospholipids and glycolipids were higher in linoleic acid content than other lipid classes. Generally, the reduction in long chain, monoenoic, fatty acids was associated with a corresponding increase in oleic acid in most low erucic acid frac-tions.  相似文献

Phosphatidylcholine, -inositol and -ethanolamine represented 48, 18 and 8%, respectively, of the total phospholipids in medium and low erucic acid rapeseed. Oleic acid was the principal fatty acid in phosphatidylcholine and -ethanolamine but all components contained high levels of linoleic and saturated acids. There were eight components in the glycolipid fraction of which digalactosyl diglyceride, monogalactosyl diglyceride, sterol glycoside, esterified sterol glucoside and cerebrosides were identified. Defatted flours contained 2.0–3.5% of bound lipid which was composed primarily of phospholipids and glycolipids whereas neutral lipids were also a major component in the bound lipids of protein concentrates. The storage properties of these protein products would be adversely affected by the polyunsaturated fatty acids in these bound fractions.  相似文献

Mouse urine samples from different pregnancy and lactation periods were examined by capillary gas chromatography to assess variations in the volatile signals that may affect the endocrine function of other females. Statistically significant changes in the excretion of certain urinary volatiles were observed; from 26 readily quantifiable constituents, 14 appear to be under the endocrine control. These selected components, positively identified through mass spectrometry and retention data, and the synthetic standards are ketones, unsaturated alcohols, esters, and cyclic vinyl ethers.  相似文献

Lipids from bovine optic nerve were analyzed. The total content of 16.5% by weight included 27.2% nonpolar lipids, 26.1% glycolipids, and 46.7% phospholipids by weight. Free cholesterol was the major component of the nonpolar lipid fraction. The cerebrosides, 73.5% of total glycolipids, were separated by thin layer chromatography (TLC) into two bands (upper and lower) that were present in equal proportion. Cerebroside sulfates comprised about 27.5% of total glycolipids. Gangliosides were also detected in the glycolipid fraction. In order of predominance, choline glycerophospholipids, ethanolamine glycerophospholipids, ethanolamine plasmalogens, serine glycerophospholipids, sphingomyelins, and inositol glycerophospholipids were the major phospholipids. Palmitoyl (16∶0), stearoyl (18∶0), and oleoyl (18∶1) groups were the major acyl groups in all neutral and phospholipid classes. However, ethanolamine glycerophospholipids, serine glycerophospholipids, and inositol glycerophospholipids contained a large percentage of 22∶6 (docosahexaenoyl) group. The major alk-1-enyl groups of the plasmalogens were 16∶0, 18∶0, and 18∶1. Steroyl (18∶0), lignoceroyl (24∶0), and nervonoyl (24∶1) were the major acyl groups in all sphingolipids. Lower cerebroside band and cerebroside sulfates contained large amount of hydroxylignoceroyl (cerebronoyl) and hydroxynervonoyl groups. A detailed analysis of the lipids of spider silk is given for the first time. Extracts of the silk from the golden orb weaver, Nephila clavipes, were studied by gas chromatography, mass spectrometry, and chemical derivatizations. The major group of the lipids consisted of methyl-branched 1-methoxyalkanes (methyl ethers) with up to four methyl groups in the chain (chain length between C28 and C34), which are unique to spiders. The position of the methyl branches was determined by conversion into cyanides, which allowed easy location of methyl branches. The second-largest group included alkanes with a wide structural variety; 2-methyl-branched, even-numbered hydrocarbons predominated. A general numerical method for the estimation of retention indices of alkanes and their derivatives is presented. Further components of the web included alkanols and alkanediols, fatty acids, and glyceryl ethers. Some comments on the biosynthesis of these compounds are also given.  相似文献

Pregnant rats were fed a high carbohydrate diet containing either 1% trilinolein or 1% trilinolein with 0.2% methyl sterculate from 18 day gestation to 21 day postpartum. The pups were weaned at 21 days and continued on the same diet for an additional 10 days. The microsomal stearyl CoA desaturase activities of the liver were effectively inhibited. Liver triglycerides showed increases in the saturated fatty acids concentrations at the expense of the corresponding monoenes. The concentration ofcis 6–7 octadecenoic acid was elevated. In liver phospholipids, the concentration of stearic acid was increased without a corresponding decrease in the oleic acid content. A drastic decrease in the nervonic acid (24∶1, n−9) concentration of liver sphingomyelin was observed. The lipids of the brain did not contain sterculic acid, and brain desaturase activity was unaffected. There was no significant change in the concentration of monoenoic acids from 16∶1 to 22∶1. However, nervonic acid was decreased by 32%. These results suggest that brain nervonic acid may be derived from a precursor other than oleic acid.  相似文献

The effect of dietary calcium (Ca) level on maternal zinc (Zn) nutritional status was studied. Female Wistar rats, weighing 250-350 g, were fed during pregnancy and lactation with an experimental diet containing/100 g different levels of calcium: 0.2 g (low calcium: LCa), 0.6 g (normal calcium: NCa) or 0.9 (high calcium: HCa). Maternal blood samples were drawn from the tail at delivery and at the end of lactation. Laboratory determinations were: Zn in whole blood (WB) at delivery and weaning; Zn (ZnF) and Ca (CaF) in the ashed femur at weaning. The results (mean +/- SEM) were: ZnWB (microgram/ml) at delivery and weaning: LCa: 8.73 +/- 1.05; 12.8 +/- 2.02; NCa: 3.49 +/- 0.19; 3.73 +/- 0.37; HCa: 3.21 +/- 0.19; 3.85 +/- 0.27. CaF (mg/100 mg): LCa: 19.2 +/- 0.8; NCa: 21.4 +/- 0.6; HCa: 20.4 +/- 1.1. ZnF (microgram/100 mg): LCa: 30.2 +/- 0.9; NCa: 24.1 +/- 0.3; HCa: 24.1 +/- 0.9. ZnWB was significantly higher in LCa (p < 0.0001) regarding NCa and Hca. ZnF showed an increase and CaF a decrease in LCa regarding NCa and HCa (p < 0.0001). There were no significant differences in ZnWB, ZnF and CaF between NCa and HCa These results show that: there was no detrimental effect when dietary Ca content was increased by 50% above the normal requirements of the rat.; low dietary Ca during pregnancy and lactation produced an increase of Zn utilization, reflected in maternal blood Zn and in ZnF content.  相似文献

The liver and plasma lipids and fatty acid composition of rats fed synthetic diets of differing fat type and content were studied. All animals were starved for 48 hr and then refed a high carbohydrate, fat-free diet for 48 hr. They were then divided into three groups and fed for an additional 48 hrs the following: group 1, the fat-free diet; group 2, a diet containing 44% of calories from corn oil; and group 3, a diet containing 44% calories from completely hydrogenated soybean oil. The total lipid concentration of the liver in the animals on the fat-free diet was elevated at 72 and 96 hr. The addition of either saturated or unsaturated fat in the diet at 48 hr prevented this accumulation. The total phospholipid and cholesterol concentrations of the liver were relatively uninfluenced by any diet in this study. Plasma total fatty acid concentration was elevated at 72 hr in the animals on a fat-free diet compared to those fed the stock diet, starved for 48 hr or fed the fat-containing diets. By 96 hr, however plasma fatty acid concentrations in all groups were similar to those in animals fed only the stock diet. The release of de novo synthesized fatty acids into plasma from the liver was strongly inhibited by dietary fat, either saturated or polyunsaturated. With the fat-free diet there was a significant increase in the saturated and monounsaturated fatty acids in both liver and plasma. The addition of corn oil to the diet facilitated a reversion of the fatty acid composition in liver and plasma to that found in the animals fed the stock diet ad libitum, but saturated fat did not. No effect of diet on the fatty acid composition of the red cells was observed during the course of this study. Exogenous saturated fatty acids, although similar chemically to the fatty acids synthesized by the liver, may have physiological actions that differ from endogenously synthesized fat.  相似文献

Recent publications on the composition of human milk are reviewed. The importance of proper sampling is discussed. Fat contents of 2.6–4.5% and cholesterol amounts of 200–650 mg/100 g fat were reported. The phytosterols in milk were increased by the consumption of these sterols. Phytosterols could contribute to the “total cholesterol” in milk if analyses are done colorimetrically. The fatty acid composition is remarkably uniform unless bizarre diets are consumed; the amounts of linoleic acid vary the most. Phospholipids contained more long chain polyunsaturated fatty acids than triacylglycerols. Striking differences were found in the compartmentalization of fatty acids into liver lipid fractions. The saturated fatty acids—lauric, myristic, palmitic and stearic—were incorporated into phosphoglycerides at faster rates with increasing chain lengths, while triglyceride incorporation was almost uniform. The degree of incorporation of the unsaturated fatty acids into phosphoglycerides (structural) compared to triglyceride (storage and energy) was the converse of their oxidation rates. The incorporation of oleic, linoleic and α-linolenic acids was mainly into triglyceride, whereas dihomo-γ-linolenic acid and arachidonic acid were preferentially incorporated into phosphoglycerides. The data suggest that distribution of each fatty acid is different depending on its destination for structural or energy function.  相似文献

Analyses of the fatty acid content and composition of various lung lipids were conducted in rats 1 day, 5 days, and 12 days after birth and in adult animals in order to define more clearly the specific lipid peroxidizing system found in neonatal rat lungs. Lipid peroxidation occurs in the 900×g supernatant fraction of rat lung homogenates in an age-dependent manner independent of the addition of any factor and is maximal at 5 days of age. No lipid peroxidation is evident in similar preparations of either newborn or adult lung tissue. As the animals develop, arachidonic and docosahexaenoic acids, fatty acids which are both highly susceptible to lipid peroxidation in the presence of a suitable catalyst, decrease gradually when measured as the percentage of the total fatty acids in the triglyceride fraction of the lung. The total quantity of triglycerides, however, is significantly lower in lungs from 1-day-old rats than at any other age. The fatty acid composition and total quantity of both lung phospholipids and lung free fatty acids do not show similar changes. Following in vitro incubation of the 900×g supernatant fraction of peroxidizing lung homogenates, an appreciable decrease in the amount of arachidonic and docosa-hexaenoic acid could be detected in lung triglycerides. Less extensive decreases were observed in the phospholipid fraction. No changes in these components were observed in newborn or adult animals. The addition of triarachidonin to the 900×g supernatant fraction of lung homogenates resulted in increased malondialdehyde release at all ages tested while added arachidonic acid increased the formation of malondialdehyde only in 5- and 12-day-old rat lung preparations. The addition of triolein, cholesterol arachidonate, and diarachidonyl phosphatidylcholine had no effect on malondialdehyde formation at any age. The age-dependent lipid peroxidation observed after in vitro incubation of rat lung homogenate preparations, therefore, may result from the relatively high concentration of triglycerides containing polyunsaturated fatty acids present in the neonatal tissue. As the susceptible polyunsaturated fatty acids of lung triglycerides are replaced by less unsaturated species, this activity may diminish concomitantly.
