In young calves on dairy farms the animal prevalence of extended-spectrum and AmpC β-lactamase-producing Escherichia coli (ESBL/AmpC-EC) is significantly higher compared with the animal prevalence in young stock and dairy cows. Hitherto it was unknown at what age antimicrobial resistant bacteria appear for the first time in the gut of calves on dairy farms, and how long these infections persist. The aim of this study was to examine the prevalence of ESBL/AmpC-EC, the number of excreted ESBL/AmpC-EC (in cfu/g of feces), as well as the ESBL/AmpC genotypes in young dairy calves (0–21 d of age) and the variation of these parameters between calves of different ages. Next to this, the course of shedding ESBL/AmpC-EC during the first year in dairy calves was studied. In a cross-sectional study, fecal samples from 748 calves, from 0 to 88 d of age, on 188 Dutch dairy farms were collected. The prevalence of calves testing positive for ESBL/AmpC-EC in a phenotypic assay was determined for different age categories (per 2 d of age). Positive samples were subjected to a semiquantitative test to determine the numbers of ESBL/AmpC-EC per gram of feces and for a selection of ESBL/AmpC-EC isolates the ESBL/AmpC genotype was determined. Ten of the 188 farms were selected for a longitudinal study based on the presence of at least 1 female calf with ESBL/Amp-EC in the cross-sectional study. These farms were additionally visited 3 times with a 4-mo interval. All calves that were sampled in the cross-sectional study were, if still present, resampled during the follow-up visits. Results show that from the day of birth ESBL/AmpC-EC can be present in the gut of calves. The phenotypic prevalence of ESBL/AmpC-EC was 33.3% in 0- to 21-d-old calves and 28.4% in 22- to 88-d-old calves. The prevalence of ESBL/AmpC-EC positive calves varied per age category among calves up to 21 d of age: significant increases and decreases at an early age were shown. Results of the longitudinal study show that after 4, 8, and 12 mo the prevalence of ESBL/AmpC-EC positive calves dropped to 3.8% (2/53), 5.8% (3/52), and 2.0% (1/49), respectively. This indicates that early gut colonization in young calves with ESBL/AmpC-EC is transient and does not lead to long-term shedding of these bacteria.  相似文献

The AmpC enzyme is normally expressed constitutively in Escherichia coli, and its overproduction confers resistance to cefoxitin. A newly reported AmpC, the extended-spectrum AmpC (ESAC), is related to resistance to cefepime, a fourth-generation cephalosporin. This enzyme presents more flexibility in the active site due to insertions, replacements, and deletions on AA sequences. Many isolates producing ESAC were reported in human clinical isolates, but E. coli ESAC producers were reported in animals only in France. The animal E. coli strains can produce this enzyme and possibly disseminate it to human and production environments. In our study, 3 strains of E. coli from milk and feces bovine samples, collected in Rio de Janeiro, Brazil, were suspected to produce ESAC. After excluding other mechanisms of resistance, the gene was sequenced to verify ESAC characteristics. These strains presented replacement of AA in omega and R2 loops, suggesting ESAC production. This is the first report to study ESAC E. coli in dairy farms in Brazil.  相似文献

The objectives of this cohort study were to identify a cut point on a previously described 6-level ultrasound score (USS6) at which average daily gain (ADG) is affected and to determine whether there is any additional benefit of using a clinical respiratory score. Calves from a commercial herd in Ohio were enrolled at entry to an automated calf feeder barn at (mean ± standard deviation) 21 ± 6 d of age (n = 308). Calves that survived until 50 d (n = 233) were included in the analyses. Twice-weekly health exams included a clinical respiratory score (CRS), USS6 (0–5, based on lung mass involved), and body weight. For the CRS, the nose, eyes, ears, cough, and rectal temperature were assigned a score (0–3), and calves were considered positive (CRS+) when at least 2 areas scored ≥2. For analysis, USS6 and CRS status were based on a calf's first bovine respiratory disease event identified during the study period. The first multivariable linear model was fit to determine whether USS6 was associated with ADG and controlled for CRS. We detected no difference in ADG among calves with USS6 scores of 2, 3, 4, and 5. Based on this finding, we proposed a simplified 2-level ultrasound score (USS2; without lung consolidation or with lung consolidation ≥1 cm2). A second multivariable model was fit to assess the association between USS2 and ADG; this model controlled for CRS, birth weight category, breed, and cohort. Calves with lung consolidation (n = 169) had lower ADG than calves without lung consolidation (n = 64; 0.73 vs. 0.85 kg/d, respectively). Calves that were CRS+ (n = 61) had lower ADG than calves that were CRS? (n = 172; 0.74 vs. 0.84 kg/d, respectively). Although CRS did not affect the relationship between USS2 and ADG, both CRS and USS2 are necessary to explain variation in ADG. We simplified USS6 and proposed USS2 based on how lung consolidation affected ADG. A simplified 2-level ultrasound score may be more practical for veterinarians to identify calves that may be at risk for poor growth. The effect on ADG was similar between calves with lung consolidation and calves identified as CRS+. Therefore, both thoracic ultrasonography and CRS should be used to identify calves with all types of respiratory disease that affect growth. However, this study represents calves in group housing from 21 to 50 d of age on 1 farm with high disease incidence. We encourage studies that investigate the effects of lung consolidation and CRS on ADG in different management systems.  相似文献

The objective of the study was to update a prior meta-analysis on the efficacy of the treatment of bovine endometritis with PGF. Clinical endometritis in cattle is defined as the presence of a purulent or mucopurulent vaginal discharge after 20 or 26 d, respectively, postpartum. Although the prevalence and impact of this disease are well known, the questions of whether and how to treat endometritis are still controversially discussed. Hence, to ensure timeliness of scientific results, we updated an existing meta-analysis published in 2013 that showed no treatment effect of PGF in cases of bovine endometritis. As in the prior meta-analysis, a comprehensive literature search was conducted. After applying specific inclusion and exclusion criteria, 3 trials remained that could be added to the 6 trials in the initial meta-analysis. Data for each trial were analyzed using the meta-analysis software Review Manager (version 5.3; The Nordic Cochrane Centre, Copenhagen, Denmark). Estimated effect sizes of PGF were calculated on calving to first service interval (CFSI) and calving to conception interval (CCI). As revealed by the initial meta-analysis, PGF treatment still had no effect on both reproductive parameters. Heterogeneity remained substantial for CFSI and CCI and even increased for CCI compared with the initial meta-analysis. Subgroup analysis, however, did not reveal a source of heterogeneity. In updating the meta-analysis, publication bias for trials reporting CCI became even more pronounced, suggesting that studies with higher standard error (i.e., usually smaller studies) that reported a prolonging effect on CCI after a PGF treatment were less likely to be published. Finally, although the number of trials published on the chosen topic and their comparability are still limited, this update of our meta-analysis now provides results that are based on approximately twice the number of included cows. The overall outcome, however, has not substantially changed and a positive effect of a PGF treatment in case of clinical endometritis could not be shown. Therefore, we do not recommend a treatment of bovine endometritis with PGF as a means to improve reproductive performance.  相似文献

The objective of this study was to identify the calf-level colostrum management practices associated with an adequate transfer of passive immunity (TPI; defined as serum Brix refractance ≥8.4% in the first week of life) in small-sized herds. A total of 818 calves from 61 commercial Holstein dairy farms were included in this observational cross-sectional study. For each calf, sex, colostrum delivery method, colostrum volume fed at first meal, and time to first feeding (delay between birth and first colostrum meal) were noted. Blood and colostrum samples were collected to estimate the serum and colostrum quality using Brix refractometry. To quantify the level of bacterial contamination in colostrum samples, total bacteria count and total coliform count (TCC) were measured using the Petrifilm (3M, St. Paul, MN) culture system. In this study, 68% of calves had an adequate TPI (≥8.4%). For data distribution, the 25th, 50th, and 75th percentiles were 1.3, 2.8, and 3.3 L for the colostrum volume fed at the first meal; 20.9, 23.5, and 26.5% Brix; and 1.1, 3.1, and 6.5 h for the time to first feeding of colostrum, respectively. The odds of adequate TPI were 2.6 times higher in calves receiving ≥2.5 L colostrum at their first meal, 2.9 times higher in calves receiving colostrum with ≥24.5% Brix, and 1.6 times higher in calves receiving colostrum within 3 h after birth, than in calves not meeting these criteria. In the present study, median bacterial contamination distribution (interquartile range) in the first colostrum meal was 14,000 cfu/mL (3,000–83,000 cfu/mL) for total bacteria count, and 0 cfu/mL (0–1,000 cfu/mL) for TCC. Total bacteria count and TCC were not associated with the odds of adequate TPI in the final model. Overall, these results suggest that specific calf-level colostrum management practices are associated with adequate TPI in small- to medium-sized dairy herds.  相似文献

The establishment of pregnancy following insemination is the primary definition of fertility in most dairy systems. Highly fertile cows establish pregnancy sooner after calving and require fewer inseminations than lower-fertility cows. Pregnancy occurs through a series of individual events in sequence. In postpartum cows, for example, the uterus involutes, estrous cycles are re-established, estrus is expressed and detected, sperm are deposited in the reproductive tract and capacitate, ovulation occurs and is followed by fertilization, and the corpus luteum forms and produces sufficient progesterone to maintain pregnancy. The oviduct supports early cleavage and the uterus establishes a receptive environment for the developing pregnancy. Each individual event is theoretically heritable and these events collectively contribute to the phenotype of pregnancy after insemination. Across most dairy systems, genetic selection for fertility in cows is primarily based on reduced days from calving to pregnancy (i.e., days open). Dairy systems differ with respect to reproductive management applied to cows, which may affect the relative importance of individual components to the overall fertility of the cow. In some systems, cows are inseminated after detected estrus with minimal intervention. In these systems, days open effectively captures the summation of the individual components of fertility. More intensive systems use hormonal treatments (e.g., PGF, GnRH) followed by timed artificial insemination (AI). Timed AI does not invalidate days open but the individual components that contribute to days open may be more or less important. Selection of cows for days open within populations that are managed differently may place different pressures on the individual components of fertility. Ensuring uniform performance of future cows across a variety of reproductive management systems may require a greater understanding of the underlying genetics of the individual components of fertility.  相似文献

In this study, we aimed to assess trends in antimicrobial resistance and to investigate the characteristics of extended-spectrum β-lactamase (ESBL)-producing isolates from bovine mastitic milk from 2012 to 2015. A total of 374 Escherichia coli isolates were analyzed (154 in 2012, 113 in 2013, 76 in 2014, and 31 in 2015). No consistent trends in antimicrobial resistance of E. coli isolates occurred during the 4-yr period. The most frequently observed resistance was tetracycline (23.3%), followed by streptomycin (17.1%), ampicillin (16.6%), neomycin (11.8%), and trimethoprim/sulfamethoxazole (11.2%). Multidrug resistance was observed in 15.5% of isolates. Among these isolates, 15 (4.0%) carried one or more blaCTX-M and AmpC ESBL genes from 11 different farms, including blaCTX-M-15 at 4 farms, blaCTX-M-3 at 2 farms, blaCTX-M-1 at 3 farms, and blaCMY-2 at 3 farms. This study is the first report of blaCTX-M-3-producing E. coli in dairy milk. Transfer of ESBL was observed in 3 blaCTX-M-3-producing isolates, 1 blaCTX-M-1-producing isolate, and all 3 blaCMY-2-producing isolates. Almost all blaCTX-M-15 and blaCTX-M-1 genes possessed an insertion sequence, ISECP1, upstream of the blaCTX-M gene. Identical pulsed-field gel electrophoresis profiles were also observed in blaCTX-M-producing E. coli from the same farm. These results suggested that ESBL might spread by both clonal and horizontal spread in dairy farms in South Korea. Although no significant changes occurred in the antimicrobial resistance of E. coli during the 4-yr study period, the resistance rates and presence of ESBL were high compared with those in other countries. Thus, these findings suggest the importance of control measures for E. coli, particularly ESBL-producing bacteria, on dairy farms to reduce treatment failure and transmission to humans.  相似文献

Subclinical ketosis is a common metabolic disorder affecting dairy cattle that results in a greater risk for the development of subsequent metabolic and infectious disease. Canwest Dairy Herd Improvement (DHI; Guelph, ON, Canada) has begun to use an infrared test (MilkoScan FT600, Foss Analytical A/S, Hillerød, Denmark) applied to metered composite milk samples to detect β-hydroxybutyrate (BHB) levels as a herd surveillance test for hyperketonemia. However, the test has not been compared with the gold standard, serum BHB as determined in a reference diagnostic laboratory. The objective of this cross-sectional diagnostic accuracy study was to validate the DHI milk BHB test to identify cows with hyperketonemia as determined by quantification of BHB in serum. A total of 316 cows from 17 dairy herds in southwestern Ontario had a milk and blood sample taken. Milk was collected at a routine DHI test, and blood from the same cow was sampled within 24 h of the milk test. The BHB concentration in milk was determined using the DHI milk BHB test, and serum was sent to the Animal Health Laboratory at the University of Guelph (Guelph, ON, Canada). A nonparametric receiver operating characteristic curve was generated to compare DHI milk BHB concentrations with serum BHB concentrations. Overall, a total of 34 cows (11%) had a level of serum BHB ≥1.2 mmol/L. The concentration of DHI milk BHB was moderately correlated with the concentration of serum BHB, yielding a coefficient of determination value of 0.61. The optimal cut point for determining hyperketonemia (≥1.2 mmol/L) on the DHI milk BHB test was ≥0.14 mmol/L, yielding a sensitivity of 81% and specificity of 92%. The performance of the DHI milk BHB test varied depending on the days in milk (DIM) of the cows tested, with a higher specificity being found in cows that were ≤25 DIM compared with cows tested >25 DIM. If the herd-level prevalence of hyperketonemia was ≥14%, the DHI milk BHB test had an improved sensitivity when compared with a herd-level prevalence of <14%. This study demonstrates that the DHI milk BHB test is a reliable measure for evaluating hyperketonemia using routine DHI milk samples and could be used as a herd-level monitoring tool for ketosis when evaluating nutritional management or preventative medicine strategies.  相似文献

The objective of this study was to identify herd-level colostrum management factors associated with the adequate transfer of passive immunity (TPI; defined as serum Brix refractance ≥8.4% in the first week of life). A total of 59 commercial Holstein dairy farms were included in this observational cross-sectional study. In every participating herd, a minimum of 14 Holstein calves were sampled to measure their TPI using a digital Brix refractometer. Colostrum samples fed to each of these calves were collected to estimate IgG concentration (colostrum quality) using a digital Brix refractometer and bacterial contamination using the Petrifilm (3M, St. Paul, MN) culture system. Dairy producers completed a questionnaire on colostrum management to assess on-farm practices. The study outcome was the prevalence of adequate TPI calculated based on the proportion of adequate TPI (defined with an individual threshold ≥8.4% Brix) on the total samples tested within each herd. According to the threshold determined in a previous study investigating the influencing colostrum management factors to achieve adequate TPI at the calf level, the prevalence of an adequate colostrum volume fed at first meal (≥2.5 L), the prevalence of adequate colostrum quality (≥24.5% Brix), the prevalence of an adequate time to first feeding (delay between birth and the first colostrum meal, ≤3 h), the prevalence of low aerobic bacterial contamination (≤20,000 cfu/mL), the prevalence of low coliform contamination (≤1,000 cfu/mL), and the prevalence of females were calculated. The herd-level prevalence of adequate TPI ranged from 24% to 100%, with a median of 68%. The median herd prevalences of an adequate colostrum volume fed at first meal, of adequate colostrum quality, of an adequate time to first feeding, of low aerobic bacterial contamination, of low coliform contamination, and of females, were 71, 42, 41, 64, 88, and 61%, respectively. In the final model, the prevalence of adequate TPI was associated with the prevalence of an adequate colostrum volume fed at first meal and the prevalence of an adequate time to first feeding. In summary, management practices varied greatly between farms and influenced the prevalence of adequate TPI.  相似文献

The primary objective of this study was to evaluate the repeatability of measurements of β-hydroxybutyrate (BHB) concentrations in capillary blood obtained from minimal-invasive puncturing of different sites at the external vulva skin of dairy cows. A total of 240 multiparous cows sampled once within 21 d in milk were enrolled in the study. Capillary blood samples were obtained from the right and left labium of the external vulva skin and additionally from a site close to the first puncture. Samples were analyzed by using an electronic hand-held device [FreeStyle Precison Neo (FSP-Neo), Abbott GmbH & Co., Wiesbaden, Germany)]. Additionally, samples from a coccygeal vessel were analyzed as reference in a laboratory. Mean deviations (± standard deviation) between the 3 capillary sampling sites ranged from 0.01 ± 0.14 (left vs. right labium) to 0.02 ± 0.13 mmol/L (left or right labium vs. additional site). The BHB concentrations in capillary blood compared with the reference showed a mean deviation of 0.14 ± 0.18 mmol/L. The Spearman correlation coefficient between BHB concentrations in capillary blood and coccygeal blood was 85.4%. Mixed model analyses showed no significant effect of sampling site or squeezing the skin during sampling procedures on the differences toward the reference. Based on laboratory BHB concentrations of ≥1.2 and ≥1.4 mmol/L, respectively, the prevalence of hyperketonemia was 10.4 (n = 25) and 5.0% (n = 12). Thresholds to detect hyperketonemia with the FSP-Neo were optimized by receiver operating characteristic analyses. Based on a laboratory BHB concentration of ≥1.2 mmol/L thresholds were determined as 1.1 (left labium, 2nd puncture) and 1.0 mmol/L (right labium), with sensitivities between 94 and 99% and specificities between 89 and 97%. Based on a serum BHB concentration of 1.4 mmol/L, receiver operating characteristic analyses resulted in optimized cutoffs of 1.4 mmol/L for all sampling sites at the vulva, with sensitivities of 100% and specificities between 96 and 99%. In conclusion, vulvar capillary blood is suitable for accurate measuring of BHB concentrations by using the FSP-Neo. After adjusting the thresholds, the FSP-Neo provides very good test characteristics independent of the capillary sampling site or squeezing the skin and allows a practical and reliable screening tool for dairy cows.  相似文献

We identified 199 Staphylococcus aureus isolates from quarter milk samples of 1,289 dairy cattle between 2014 and 2018. About 66% of the isolates were resistant to at least 1 antimicrobial agent; the highest rate of resistance was to penicillin, followed by resistance to ampicillin, erythromycin, and sulfadimethoxine. We obtained 30 methicillin-resistant S. aureus (MRSA) strains from 6 farms in 3 provinces. The MRSA strains exhibited a significantly higher resistance rate to most of the tested antimicrobials than the oxacillin-susceptible strains. The MRSA strains represented 5 genotypes: ST72-t324-SCCmec IV (n = 14), ST30-t1752-SCCmec IV (n = 8), ST188-t189-SCCmec NT (n = 6), ST188-t2284-SCCmec NT (n = 1), and NT-NT-SCCmec IV (n = 1). One of the ST188 MRSA strains represented a novel staphylococcal protein A (spa) type (t2284). In addition, 7 of the 8 ST30 MRSA strains were Panton-Valentine leukocidin (PVL)–positive and carried various staphylococcal enterotoxin encoding genes. This is the first report of PVL-positive ST30 MRSA-t1752-SCCmec IV from bovine mastitis in Korea. All of ST72-t324-SCCmec IV MRSA strains carried staphylococcal enterotoxin and leukotoxin encoding genes. They were also sensitive to most of the tested non-β-lactam antimicrobials. In contrast, ST188-t189 MRSA strains were resistant to multiple antimicrobials and predominantly carried the leukotoxin encoding gene. Taken together, these findings may indicate that dairy cows could be a major source for spreading MRSA strains, and contaminated milk could be a vehicle for transmission. Suitable hygienic measures should be established in dairy farms and processing plants to limit the likelihood of introducing MRSA into the food chain.  相似文献

Free fatty acid receptors (FFAR) play significant roles in various physiological processes, including energy metabolism, through interaction with their ligands, fatty acids. To determine whether the receptors FFAR1 and FFAR2 are involved in the regulation of liver metabolism during the peripartal period, we selected 13 German Holstein multiparous dairy cows and grouped them as high β-hydroxybutyrate (H-BHB; n = 8) or low β-hydroxybutyrate (L-BHB; n = 5) according to their individual maximum plasma BHB concentration observed within wk 2 or 3 postpartum (H-BHB: >1 mmol/L and L-BHB: <0.77 mmol/L). The selected cows had a milk yield of more than 10,000 kg/305 d during a previous lactation. The cows were fed a total mixed ration according to their requirements during the far-off dry period [5.9 MJ of net energy for lactation (NEL)/kg of dry matter (DM), crude protein (CP) 126 g/kg of DM], close-up dry period (6.5 MJ of NEL/kg of DM, CP 137 g/kg of DM), and lactation (7 MJ of NEL/kg of DM, CP 163 g/kg of DM). Blood samples were taken weekly, from d ?34 to d 40 relative to parturition. Liver biopsies were taken on d ?34, ?17, 3, 18, and 30 relative to parturition and at slaughter (d 40). The protein abundance of FFAR1 was lower during the whole peripartal period in the H-BHB group. The abundance of FFAR2 increased over time and tended to be higher in H-BHB cows. The abundance of FFAR1 might be associated with imbalances of liver metabolism in peripartal dairy cows.  相似文献

Reports of β-lactam-resistant Staphylococcus aureus in artisanal goat cheese are increasing, and this phenomenon is relevant to public health. The objective of the present study was to determine the prevalence of S. aureus strains carrying the blaZ and mecA resistance genes, as well as the genes encoding the staphylococcal enterotoxins SEA, SEB, SEC, SED, SEE, and TSST-1 in artisanal coalho cheese made from goat milk produced in northeastern Brazil. We used biochemical and molecular tests to characterize 54 S. aureus isolates found in artisanal coalho cheese collected from commercial establishments producing animal products in 11 municipalities of Pernambuco State, Brazil. A PCR analysis revealed that 42.6% (23/54) of the isolates were positive for the blaZ gene, and 7.4% (4/54) were resistant to methicillin by phenotypic testing. We did not detect mecA or any genes encoding enterotoxins. The presence of S. aureus carriers of the blaZ gene and the identification of methicillin-resistant S. aureus strains are of concern for the health of consumers of this type of cheese.  相似文献

Megasphaera elsdenii is a bacterial species of the rumen that can utilize lactate to produce butyrate, a key volatile fatty acid often implicated in driving calf rumen development. Because lactate is abundant in the rumen of young calves, administration of M. elsdenii to increase butyrate production and thus promote calf rumen development is an appealing possibility. The main objective of this study was to determine whether M. elsdenii administration to calves via oral drench at 14 d of age affected its long-term establishment at 70 d postadministration. Ruminal volatile fatty acid and lactate profiles and blood glucose and β-hydroxybutyrate concentrations were also examined to determine potential influence on rumen metabolism. Six neonatal Holstein heifer calves were blocked on d 1 by body weight (41.3 ± 1.8 kg) and total serum protein (5.23 ± 0.16 g/dL) and assigned to either the M. elsdenii (n = 3) or control (n = 3) treatment groups. On d 14, calves in the M. elsdenii group orally received 25 mL of a commercially available M. elsdenii suspension, whereas calves in the control group received 25 mL of the same product that had been autoclaved. Rumen contents and blood samples were collected weekly from each animal until 84 d of age. The oral administration of M. elsdenii at 14 d did not increase the abundance of M. elsdenii 70 d postdosing, alter rumen fermentation, or change blood metabolites associated with butyrate. These results suggest that a single administration of the M. elsdenii probiotic may not affect the rumen establishment of the organism.  相似文献

This study was undertaken to evaluate the relationship between plasma and milk concentrations of vitamin B12 as well as the relationship between plasma or milk concentrations of vitamin B12 and plasma concentration of free fatty acids (FFA) or blood concentration of β-hydroxybutyrate (BHB) of early lactating Ayrshire (AY) and Holstein (HO) cows. A total of 44 dairy herds (7 AY and 37 HO herds) and 62 AY (21 in first, 19 in second, and 22 in third and more lactations) and 228 HO (51 in first, 74 in second, and 103 in third and more lactation) cows between 3 and 40 d in milk were involved in the study. Hand-stripped milk samples and blood samples were taken 6 h after the morning milking. Milk and plasma samples were analyzed for vitamin B12 concentration and plasma samples were analyzed for FFA concentration. A handheld device was used for blood BHB concentration determination. Thresholds for elevated plasma FFA concentration and hyperketonemia were set at ≥0.70 and ≥1.2 mmol/L, respectively. Vitamin B12 concentration in milk of AY primiparous cows [2,557 (1,995–3,276) pg/mL] was lower than in milk from HO primiparous cows [3,876 (3,356–4,478) pg/mL], whereas no difference was observed among other parities and breeds. Regardless of breeds, plasma concentration of vitamin B12 of first and second parities was lower than plasma concentration of third and more lactation cows. Milk vitamin B12 concentration was positively correlated with plasma vitamin B12 concentration, but the relationship was stronger for AY (ρ averaging 0.63) than for HO cows (ρ averaging 0.36). For AY and HO breeds, a significant relationship between milk or plasma vitamin B12 concentrations and plasma FFA concentration (ρ between 0.29 and 0.59) was observed. Moreover, cows with elevated plasma FFA concentration had greater milk and plasma vitamin B12 concentrations than cows with normal plasma FFA concentration. No relationship between vitamin B12 concentration in milk or plasma and blood BHB concentration and hyperketonemia was noted. In summary, milk is not well correlated with plasma vitamin B12 concentration for HO. It could be hypothesized that elevated plasma concentration of FFA could have a negative effect on the use of vitamin B12 by cow cells, which increases the concentration of the vitamin in plasma and its secretion in milk.  相似文献

Our objective was to describe management and herd characteristics of the transition period on freestall dairy herds in the northeastern United States using an on-farm survey and prospective cohort design. Enrolled herds (n = 72) had a median of 900 milking cows (range: 345–2,900) and a rolling herd average of 12,674 kg (standard deviation ± 1,220 kg), and 87.2% (n = 82/94) of fresh pens were milked at least 3×/d. The prevalence of herds with ≥15% of sampled cows with elevated concentrations of nonesterified fatty acids prepartum (≥0.27 mmol/L, 2–14 d before parturition) and postpartum [primiparous: ≥0.60 mmol/L, multiparous: ≥0.70 mmol/L, 3–14 d in milk (DIM)], β-hydroxybutyrate postpartum (≥1.2 mmol/L, 3–14 DIM), and haptoglobin postpartum (≥1 g/L, 0–12 DIM) was 51%, 51%, 51%, and 57%, respectively. In most herds, cows were moved to a calving pen when showing signs of labor (73.6%; n = 53/72) instead of 0 to 3 d before expected calving (26.4%, n = 19/72). Cows remained in the calving or maternity pen for a median (range) time of 2 (0–24) h after parturition before moving to the next pen. Primiparous cows remained in the first pen moved to after parturition for a longer period than multiparous cows [median (range) days: 12 (1.5–25) vs. 6 (1.5–22)]. Approximately 20% of herds had routine vaccinations administered in the maternity or calving pen, first pen after parturition, or both. Almost all herds (n = 69/72) performed fresh cow health checks; however, only 53% (n = 38/72) locked up all fresh cows daily. More herds housed primiparous and multiparous cows in separate pens during the far-off dry (65.3%; n = 47/72) and high-lactation (81.9%; n = 59/72) periods compared with the close-up dry (31.9%; n = 23/72) and fresh periods (27.8%; n = 20/72). At least half of the pens observed during the far-off dry, close-up dry, and fresh periods had a stocking density <100%. Approximately one-third of pens observed during the far-off dry period had feed pushed up ≤4×/d compared with approximately 15 to 20% of pens observed during the close-up dry, fresh, and high-lactation periods. More than half of the total mixed ration samples acquired from the far-off and close-up dry period visits had greater than the recommended proportion of particles in the 19-mm screen of the Penn State Particle Separator. The results of this observational study illustrated the range of management practices used in freestall herds in this region and lay the groundwork for future hypothesis-driven studies using this sampled population.  相似文献

The objective of this observational study was to assess the relationship between herd-level prevalence of hyperketolactia (HPH) with management practices of the transition period and herd milk production. Dairy herds (n = 71) were selected based on their inclusion in a herd management risk assessment study (August 2014–March 2018) using a Vital 90 (Elanco) Risk Assessment tool (one assessment per farm). Data from multiple milk recording test-days (Dairy Herd Improvement, DHI; Lactanet) were included in the analysis. Tests performed within ±6 mo relative to each farm's risk assessment date were included (10 ± 2 SD tests per farm). The majority of the farms were located in Ontario (83%). For each farm DHI test, the data set included herd average milk yield (kg/cow per day), average milk fat and protein (%), average somatic cell count (cells/mL), average days in milk (DIM), number of cows tested for ketosis, number of ketosis-positive tests (milk β-hydroxybutyrate ≥0.15 mmol/L), and proportion of cows by parity groups. Overall HPH (5–21 DIM) was calculated based on data available per farm (sum of all positive tests within 5–21 DIM/sum of all cows tested within 5–21 DIM). Each farm average was obtained by considering all test-days. A logit-transformation was applied to hyperketolactia prevalence. Linear regression models (PROC GLM and MIXED of SAS, Version 9.4) were used to predict herd HPH (milk β-hydroxybutyrate ≥0.15 mmol/L within 5 to 21 DIM; the outcome of interest). Four initial models (far-off, close-up, and fresh periods, and DHI) were separately built to assess associations between their variables and HPH; a final model considered variables selected in the initial models. Univariable (liberal P < 0.25) followed by multivariable models were used to build specific models for each period of the risk assessment. Herd prevalence of hyperketolactia was 27 ± 14%, with an average herd size of 141 ± 110 cows. The final HPH model (R2 = 24.8%) included weighted milk yield, the proportion of primiparous cows, water access in the close-up period, and access to rest areas or stall access in the fresh period. Herd prevalence of hyperketolactia was negatively associated with milk yield [odds ratio, OR = 0.96 (95% confidence interval 0.92–0.99)] and proportion of primiparous cows [OR = 0.98 (0.96–0.99)]. The odds of hyperketolactia were greater with poor water access and quality (<5 cm of linear access per cow; dirty water; only 1 water location in pen) than with ≥10.2 cm of linear access per cow; clean water; >2 water locations in pen [1.23 (1.11–2.39)] in the close-up period. The odds of hyperketolactia were greater in farms providing limited access to rest areas in the fresh period than in farms providing constant access to rest areas, without dead-ends [1.64 (1.03–2.80)]. In Canadian dairy herds, HPH in early lactation was associated with certain transition-period management practices and was negatively associated with herd productivity.  相似文献
