It is necessary for the dairy industry to reduce calf morbidity and mortality, and the reliance on antibiotics to treat sick calves, to address the growing concern regarding antibiotic resistant bacteria. The primary objective of this study was to evaluate the effect that feeding dairy calves medium-chain fatty acids (MCFA) has on growth performance and health, and the secondary objective was to evaluate the effect of MCFA on energy status around weaning and the adaptive immune response following a vaccine challenge. Thirty-three Holstein bull calves (5 ± 1.6 d of age) were randomly assigned to 1 of 2 treatments. Control (CON) calves were fed milk replacer with no C8:0 or C10:0 oil added and MCFA calves were fed milk replacer with 0.5% of a combination of C8:0 or C10:0 oil added. Body weight and average daily gain were measured weekly. Feed efficiency (gain/feed) and the change in body condition score, hip width, hip height, heart girth, and paunch girth were calculated for the duration of the study. Fecal scores were recorded daily and all medical treatments were documented for the duration of the trial. On d 42, 49, and 56 of the study, a serum sample was collected from each calf and used to measure nonesterified fatty acids, β-hydroxybutyric acid, insulin, and glucose concentrations to evaluate energy status around weaning. A subset of 11 calves per treatment were enrolled in a vaccine challenge. At 21 ± 1.9 d of age (mean ± standard deviation) calves were vaccinated intramuscularly with 1 mL of endotoxin-free ovalbumin (OVA) mixed with aluminum hydroxide adjuvant. At 42 d of age (±1.9 d), blood samples were collected and used to analyze OVA-specific IgG1 and IgG2, and calves were vaccinated a second time. At 56 d of age (±1.9 d), blood samples were collected to analyze IgG1 and IgG2 as well as IFN-γ and IL-4 secreted from peripheral blood mononuclear cells (PBMC) treated with OVA or phytohemagglutinin. Data were analyzed as a completely randomized design with repeated measures when applicable. A tendency for greater daily fecal score was observed for MCFA calves compared with CON. At d 42 of the study, nonesterified fatty acid concentrations were greater in CON calves compared with MCFA. At 42 and 56 d of age, anti-OVA IgG1 concentrations for CON and MCFA calves were greater than prevaccination samples. This study suggests that feeding MCFA to calves affects the energy status of calves around weaning and vaccinating dairy calves with ovalbumin combined with an aluminum hydroxide adjuvant is an effective way to evaluate the adaptive immune responses.  相似文献

This study evaluated the effects of incremental tributyrin supplementation in pasteurized waste milk on growth performance, health, and blood metabolism of dairy calves before and after weaning. Forty-eight newborn female Holstein dairy calves (39.6 ± 2.75 kg; mean ± standard deviation) were blocked by age and randomly assigned to 3 treatments: pasteurized waste milk (1) without supplementation, (2) with 1 g/L of tributyrin products (unprotected solid powder; containing 35% tributyrin), or (3) with 2 g/L of tributyrin products. The calves were weaned on d 56 and were raised until d 77. Data were analyzed for the preweaning, postweaning, and overall periods. The results showed that starter intake and hay intake were not different among treatments in any period of the trial, but the crude protein intake tended to increase linearly with tributyrin supplementation during the overall period. Although tributyrin supplementation had no effects on body weight during preweaning and overall periods, body weight increased linearly with tributyrin supplementation postweaning. The average daily gain tended to increase linearly during postweaning and overall periods. No effects were observed on feed efficiency in any period. A positive linear relationship between body length and tributyrin supplementation was observed during the postweaning period, but no differences were found for the other body structural measurements in any period. The results of diarrhea showed that tributyrin concentration had a negative linear relationship with diarrhea frequency during preweaning and overall periods. The rectal temperature did not differ among treatments in any period, but a treatment × week effect for rectal body temperature was observed. For blood metabolism, tributyrin supplementation had no effects on insulin, growth hormone, total protein, albumin, or globulin. No differences were found in serum amyloid A concentration in any of the periods, yet haptoglobin concentration decreased linearly with increasing tributyrin concentration during postweaning and overall periods. Endothelin concentration showed a tendency to decrease linearly during preweaning and postweaning periods and decreased linearly with tributyrin supplementation during the overall period. An increasing tributyrin concentration was associated with a negative linear relationship with IL-1β concentration during the preweaning period, and no differences were found in the other periods. The concentration of IL-6 and tumor necrosis factor α were not different among treatments in any of the periods. These data suggest that increasing the concentration of tributyrin in pasteurized waste milk could increase growth performance and health of dairy calves, and incremental tributyrin supplementation could linearly reduce haptoglobin, endothelin, and IL-1β concentrations, indicating a positive effect of tributyrin on alleviating oxidative stress and inflammatory status of dairy calves. Calves fed pasteurized waste milk supplemented with tributyrin products (containing 35% tributyrin) at 2 g/L compared with 1 g/L of milk had more improved growth and health.  相似文献

Lysolecithin is an antiinflammatory emulsifier associated with improved apparent digestibility of total dietary fat and improved feed efficiency in dairy cattle. However, it is unknown if lysolecithin (LYSO) improves performance in calves. Moreover, since many conventional milk replacers use vegetable-sourced fat (e.g., palm oil), nutrient absorption and fecal score may be affected in neonatal calves. Thus, the objective of this study was to evaluate the effects of LYSO supplemented in milk replacer on performance, metabolites, and gut health of preweaned dairy calves. Holstein calves (n = 32) with adequate passive transfer were assigned in pairs (16 blocks) balanced by birth weight, date of birth, and sex at 1 d of age to randomly receive either LYSO (mixed in 2 milk replacer feedings at a rate of 4 g/d Lysoforte, Kemin Industries Inc., Des Moines, IA) or a milk replacer control (nothing added). Both treatments were fed 6 L/d milk replacer [22.5% crude protein, 16.2% crude fat (vegetable oil fat source) on a dry matter basis with 14% solids] by bucket in 2 daily feedings for 56 d. Calves were individually housed in wooden hutches and offered a commercial calf starter (24.6% crude protein and 13.9% neutral detergent fiber) and water by bucket ad libitum. Feed refusals and calf health was assessed daily. Weights and blood metabolites (glucose, total serum protein, albumin, creatinine, triglycerides, and cholesterol) were sampled weekly, and calves completed the study before weaning at 56 d of age. The effects of LYSO on calf average daily gain, feed efficiency, and blood metabolites were evaluated using a linear mixed model with time as a repeated measure, calf as the subject, and block as a random effect in SAS (SAS Institute Inc., Cary, NC). The effect of LYSO to improve the odds of abnormal fecal score was evaluated using a logistic model. Supplementation of LYSO increased average daily gain (control 0.28 ± 0.03 kg; LYSO 0.37 ± 0.03 kg; least squares means ± standard error of the mean) and increased feed efficiency (gain-to-feed; control 0.25 ± 0.03; LYSO 0.32 ± 0.03). Similarly, LYSO calves had a higher final body weight at d 56 (control 52.11 ± 2.33 kg; LYSO 56.73 ± 2.33 kg). Interestingly, total dry matter intake was not associated with LYSO despite improved average daily gain (total dry matter intake control 1,088.7 ± 27.62 g; total dry matter intake LYSO 1,124.8 ± 27.62 g). Blood glucose, albumin, creatinine, triglycerides, and cholesterol were not associated with LYSO. Indeed, only total serum protein had a significant interaction with LYSO and age at wk 5 and 6. Moreover, control calves had a 13.57 (95% confidence interval: 9.25–19.90) times greater odds of having an abnormal fecal score on any given day during the diarrhea risk period from d 1 to 28. The inclusion of LYSO as an additive in milk replacer in a dose of 4 g/d may improve performance, and calf fecal score, preweaning. Further research should investigate the mechanisms behind the effects of LYSO on fat digestibility in calves fed 6 L/d of milk replacer with vegetable-sourced fat.  相似文献

BACKGROUND: Human health may be improved if milk with a favorable fatty acid composition and Se concentration is ingested. The present study is to determine how a basal diet supplemented with daily 5 mg Se as Se‐enriched yeast (SY) or sodium selenite (SS) affects the fatty acid composition and Se concentration of bovine milk. The effects of Se form on blood Se concentration, erythrocyte glutathione peroxidase 1 (GPx1) activity, serum GPx3 activity and milk yield and component were also studied. RESULTS: Both Se forms, when compared to control group, increased Se concentrations of blood (P < 0.01) and milk (P < 0.01), erythrocyte GPx1 activity (P < 0.05) and milk percentages of polyunsaturated fatty acids (PUFA) (P < 0.05) and cis‐9,cis‐12 linoleic acid (P < 0.05). Cows supplemented with SY had higher Se levels in blood (P < 0.01) and milk (P < 0.01) and percentage of PUFA in milk (P < 0.05) when compared with those supplemented with SS. Milk yield, milk component and serum GPx3 activity were not significantly affected by Se form. CONCLUSION: Supplementation of diet with SY appears to be of more benefit than SS in producing favorable milk with high PUFA and Se concentrations. This study investigated the effects of feeding dairy calves starter diets containing 19% or 22% crude protein (CP) content on a dry matter basis and either supplemented or not with soybean oil (SBO, 0 vs. 3%, dry matter basis) on growth performance, digestibility, urinary nitrogen, and purine derivatives (PD) excretion. A total of 48 female Holstein dairy calves (mean 39.8 kg of body weight) were randomly distributed to experimental diets in a 2 × 2 factorial arrangement of treatments. The 4 dietary treatments were (1) starter diet without SBO supplement and 19% CP (NSBO-19CP), (2) starter diet without SBO supplement and 22% CP (NSBO-22CP), (3) starter diet with 3% SBO and 19% CP (SBO-19CP), and (4) starter diet with 3% SBO and 22% CP (SBO-22CP). Milk feeding value was similarly based on a constant protocol across experimental treatments and calves had ad libitum access to water and starter diets throughout the study. All calves were weaned on d 63 of age and remained in the study until d 83 of age. Calves supplemented with SBO had lower starter feed intake and average daily gain (ADG) and lower feed efficiency (FE) but had a higher fecal score indicating a higher likelihood of diarrhea occurrence compared with unsupplemented calves. Wither heights, digestibilities of organic matter, CP, and neutral detergent fiber were decreased, and ruminal volatile fatty acids tended to be reduced, and the molar proportion of ruminal butyrate (preweaning) and acetate (postweaning) reduced by supplemental SBO. The urinary allantoin and total PD excretion were reduced; however, urinary nitrogen excretion was increased when calves were supplemented with SBO. The CP amount did not affect starter feed intake, FE, or diarrhea occurrence rate, whereas the 22CP diets increased neutral detergent fiber digestibility, improved ADG (tendency), and increased allantoin and urinary PD excretion compared with the 19CP diets. The starter feed intake, ADG, FE, diarrhea occurrence rate, nutrient digestibility, and ruminal fermentation were not affected by the interaction between starter SBO and CP level; however, hip height and total PD in calves that received the SBO-22CP diets were higher than those fed the SBO-19CP diets. In conclusion, based on our experimental conditions, supplemental SBO could not be recommended for dairy calves. Furthermore, our findings indicate that SBO has negative effects on performance more attributed to reducing starter intake, digestibility, and ruminal volatile fatty acid concentration rather than because of a limitation of starter metabolizable protein supply and intestinal amino acid availability. Therefore, our results indicate that feeding the higher starter CP content is not a viable strategy to compensate for the negative effects of SBO supplementation on the growth performance of dairy calves.  相似文献

Dairy calf weaning results in blood ketone concentrations in excess of mature rates of use and can result in excretion of ketones in urine representing a loss of energy. Lasalocid is frequently supplemented as an anticoccidial agent in calf starters; however, in mature ruminants it is known to alter molar ratios of ruminal volatile fatty acids (VFA). Effects of weaning transition and postweaning ionophore supplementation on body weight, dry matter (DM) intake, average daily gain (ADG), and blood concentrations of glucose, acetoacetate, β-hydroxybutyrate (BHBA), lactate, pyruvate, nonesterified fatty acids (NEFA), VFA, insulin, and glucagon were examined using Jersey bull calves (n = 24) over 16 wk. Calves were blocked into groups of 2 according to birth date and weight and randomly assigned to receive either a commercial pelleted starter (control), or the same diet containing lasalocid (TRT; 83 mg/kg of DM). Calves were fed milk replacer from d 3 to 34 (d 3 to 20 = 454 g/d at 12% solids; d 21 to 34 = 568 g/d at 15% solids), from d 35 to 48 calves received both replacer (d 35 to 41 = 454 g/d; d 42 to 38 = 227 g/d) and free access to control or TRT starter, and from d 49 to 112 received ad libitum control or TRT. Body weight and jugular blood metabolite concentrations were measured and recorded weekly. Postweaning DM intake, average daily gain, and feed:gain did not differ between control and TRT calves. Glucose and NEFA concentrations did not differ between control and TRT, but declined with age. Insulin and glucagon concentrations did not differ between control and TRT, but glucagon concentrations increased with weaning. Total VFA significantly increased following introduction of solid feed at d 35 in both groups with an apparent 1-wk lag in TRT VFA increases compared with control. Jugular acetate and butyrate concentrations were greater in control calves than TRT calves during wk 7. Propionate concentrations did not differ between control and TRT at any time following weaning. Blood BHBA concentrations were greater in control than TRT during wk 8 and 9. Thus, consumption of starter supplemented with lasalocid delayed peak acetate and butyrate and lowered peak BHBA concentrations. However, supplementation at concentrations currently recommended for control of coccidiosis did not appear to be sufficient to enhance growth or efficiency during the wk 7 to 16 postweaning interval for this sample size.  相似文献

The ability to reduce incidence of disease in calves and improve early vaccination strategies is of particular interest for dairy producers. The n-3 fatty acids have been reported to reduce inflammatory diseases in humans but limited research has been done in calves. The objective of this study was to compare supplementation of n-3 fatty acids from fish and flax oil on gene expression of whole blood cells and growth of milk-fed Holstein calves. Forty-eight Holstein bull calves from a commercial dairy were randomly assigned to 1 of 3 diets beginning at 4 d old: (1) control milk replacer (MR) with all pork fat, (2) MR with 2% flax oil, and (3) MR with 2% fish oil. All MR were 17% fat, 27% crude protein on a dry matter (DM) basis, with all protein from whey sources. Calves were each fed 654 g DM of MR daily for the first 25 d and then 327 g/d for d 26, 27, and 28. On d 28, calves were challenged with a Pasteurella vaccine and the temperature response to the vaccine was recorded. Milk and feed intake and fecal scores were recorded daily, and body weight and hip width were recorded weekly. Blood was collected from all calves on d 25. One tube of collected blood was incubated with endotoxin (lipopolysaccharide; LPS) for 2 h and frozen with a second tube of control blood. Quantitative real-time PCR was used to assess the effects of LPS stimulation on cytokine gene expression. During the 28 d, calves supplemented with flax oil had a greater growth rate and feed efficiency than calves fed fish oil (0.52 ± 0.02 vs. 0.48 ± 0.02 g of gain:g of feed). Fish oil tended to decrease LPS stimulation of tumor necrosis factor-α expression. Flax oil, but not fish oil, decreased the expression of IL-4 and tended to decrease expression of osteopontin and IL-8. Flax oil tended to reduce the increase in rectal temperature in response to a Pasteurella vaccine. In conclusion, our data support the idea that supplementation with n-3 fatty acids affects cytokine gene expression.  相似文献

We aimed to evaluate the effects of feeding super-conditioned corn at different temperatures on intake, growth performance, total-tract starch digestibility, rumen fermentation, blood metabolites, and feeding behavior of dairy calves. Thirty-six Holstein female dairy calves (40 ± 1.72 kg of body weight, ± SD) were randomly assigned to 1 of the following 3 treatments: (1) ground corn (control; CON; n = 12), (2) corn super-conditioned at 75°C (T-75; n = 12), and (3) corn super-conditioned at 95°C (T-95; n = 12). Three mash starter feeds with an identical nutritional composition were blended with 5% chopped alfalfa hay and fed to individually-housed calves from d 3 to 77 of their birth. All calves were fed 4 L/d of pasteurized whole milk twice daily since d 3 to 56, followed by 2 L/d of morning feeding from d 57 to 63 of age. Calves were weaned on d 63 and remained in the study until d 77. The T-75 and T-95 diets increased total-tract starch digestibility compared with the CON diet. Dry matter intake and weaning or final BW were not affected by treatments; however, average daily gain and feed efficiency increased in calves fed T-95 in the overall period. The T-95 diet increased withers height and tended to increase hip height compared with other diets, but feeding behavior did not change throughout the experimental period. Ruminal pH decreased in calves fed the T-95 diet compared with T-75 and CON diets. The molar proportion of ruminal propionate increased, whereas the acetate-to-propionate ratio tended to decrease in calves fed the T-95 compared with CON diet. Calves fed the T-95 diet had the highest blood glucose concentration, whereas a trend for increased insulin concentration was observed in calves fed T-95 compared with other diets. In conclusion, super-conditioning temperature of corn (T-95 vs. T-75 and CON) improved the average daily gain, feed efficiency, and skeletal growth, but did not influence dry matter intake during the first 77 d of age. Finally, the total-tract starch digestibility increased, whereas ruminal pH dropped during the postweaning period as super-conditioning temperature elevated.  相似文献

The objectives of this experiment were to investigate the effects of lauric (LA) and myristic (MA) acids on ruminal fermentation, production, and milk fatty acid (FA) profile in lactating dairy cows and to identify the FA responsible for the methanogen-suppressing effect of coconut oil. The experiment was conducted as a replicated 3 × 3 Latin square. Six ruminally cannulated cows (95 ± 26.4 DIM) were subjected to the following treatments: 240 g/cow per day each of stearic acid (SA, control), LA, or MA. Experimental periods were 28 d and cows were refaunated between periods. Lauric acid reduced protozoal counts in the rumen by 96%, as well as acetate, total VFA, and microbial N outflow from the rumen, compared with SA and MA. Ruminal methane production was not affected by treatment. Dry matter intake was reduced 35% by LA compared with SA and MA, which resulted in decreased milk yield. Milk fat content also was depressed by LA compared with SA and MA. Treatment had no effect on milk protein content. All treatments increased milk concentration of the respective treatment FA. Concentration of C12:0 was more than doubled by LA, and C14:0 was increased (45%) by MA compared with SA. Concentration of milk FA < C16 was 20% lower for LA than MA. Concentrations of trans 18:1 FA (except trans 12) and CLA isomers were increased by LA compared with SA and MA. Overall, the concentrations of saturated FA in milk fat were reduced, and that of > C16 FA and MUFA were increased, by LA compared with the other treatments. In this study, LA had profound effects on ruminal fermentation, mediated through inhibited microbial populations, and decreased DMI, milk yield, and milk fat content. Despite the significant decrease in protozoal counts, however, LA had no effect on ruminal methane production. Thus, the antimethanogenic effect of coconut oil, observed in related studies, is likely due to total FA application level, the additive effect of LA and MA, or a combination of both. Deoiled soy lecithin is a feed additive enriched in phospholipids. Our study evaluated the effects of dietary deoiled soy lecithin supplementation on (1) milk production and composition, (2) plasma and milk fatty acid (FA) content and yield, and (3) apparent FA digestibility and absorption in lactating dairy cows fed fractionated palm fat. In a split-plot Latin square design, 16 Holstein cows (160 ± 7 days in milk; 3.6 ± 1.2 parity) were randomly allocated to a main plot receiving a corn silage and alfalfa haylage-based diet with palm fat containing either moderate (MPA) or high palmitic acid (HPA) content at 1.75% of ration dry matter (72 or 99% palmitic acid, respectively; n = 8/palm fat diet). On each palm fat diet, deoiled soy lecithin was top-dressed at 0, 0.12, 0.24, or 0.36% of ration dry matter in a replicated 4 × 4 Latin square design. Following a 14-d covariate period, lecithin supplementation spanned 14 d, with milk and blood collected during the final 3 d. Milk composition and pooled plasma markers were measured. The statistical model included the fixed effects of palm fat type, lecithin dose, period, and the interaction between palm fat type and lecithin dose. The random effect of cow nested within palm fat group was also included. Lecithin linearly decreased dry matter intake. In cows fed HPA, lecithin feeding reduced milk fat content and tended to decrease milk fat yield. Although no changes in milk yield were observed, a quadratic reduction in 3.5% fat-corrected milk was observed with increasing lecithin dose. Lecithin linearly increased energy-corrected milk efficiency in cows fed MPA. Lecithin supplementation also decreased milk urea nitrogen, relative to unsupplemented cows. The proportion of 16-carbon FA in milk fat decreased linearly with lecithin dose, whereas 18-carbon FA increased linearly. Lecithin reduced de novo FA (<16-carbon) content and tended to increase preformed FA (>16-carbon) content in a linear manner. Compared with MPA, HPA diets reduced apparent total and 16-carbon FA digestibility and absorption. Deoiled soy lecithin feeding did not modify FA digestibility or absorption. Our observations suggest that soy lecithin feeding modifies rumen digestion to reduce dry matter intake and change milk composition.  相似文献

Saccharomyces cerevisiae products (SCP) have the potential to promote the growth and development of the gastrointestinal tract and immunity in young livestock animals. However, the effects of SCP supplementation on calves are inconsistent among the reported studies in the literature. Hence, we performed a meta-analysis to comprehensively assess the effects of SCP on the growth performance, ruminal fermentation parameters, nutrients digestibility, ruminal histological morphology, serum immune response, and fecal pathogen colony counts in calves. We searched the Web of Science, ScienceDirect, PubMed, and China National Knowledge Infrastructure for relevant studies published up to October 1, 2021. After screening against a set of criteria, the data of 36 studies were included in our meta-analysis (2,126 calves in total). We evaluated the quality of the data using sensitivity analysis and assessed publication bias. Our meta-analysis revealed several important findings. First, SCP supplementation increased the ruminal short-chain fatty acid concentration, ruminal papilla height, and fiber digestibility, pointing toward stimulation of the development of the rumen in calves. Second, SCP supplementation increased the serum concentrations of total protein, IgA, and IgG but decreased fecal pathogen colony counts, suggesting that SCP could help calves to promote immunity (especially maintaining circulating concentrations of immunoglobulins in preweaning calves) and resistance to pathogens. Third, a subgroup analysis between preweaning and postweaning calves showed that SCP increased average daily gain and dry matter intake preweaning but not postweaning, suggesting that SCP is better supplemented to preweaning calves to achieve the best results. Forth, based on the dose-response curve, 24 to 25 g/d might be the optimal dose range of SCP supplementation (into starter feed) preweaning to achieve the best overall effect, meanwhile, we need more studies to improve the consistency and accuracy of the dose-response curve prediction. Overall, SCP supplementation improved growth performance, rumen development, and immunocompetence in calves, particularly in preweaning calves.  相似文献

The objective of this study was to investigate the effects of the physical forms of starter and forage sources on feed intake, growth performance, rumen pH, and blood metabolites of dairy calves. Forty male Holstein calves (41.3 ± 3.5 kg of body weight) were used (n = 10 calves per treatment) in a 2 × 2 factorial arrangement of treatments with the factors being physical forms of starter (coarse mash and texturized) and forage source [alfalfa hay (AH) and wheat straw (WS)]. Individually housed calves were randomly assigned to 1 of the 4 dietary treatments, including (1) coarsely mashed (CM; coarse ground grains combined with a mash supplement) starter feed with AH (CM-AH), (2) coarsely mashed starter feed with WS (CM-WS), (3) texturized feed starter (TF; includes steam-flaked corn, steam-rolled barley combined with a pelleted supplement) with AH (TF-AH), and (4) TF with WS (TF-WS). Both starters had the same ingredients and nutrient compositions but differed in their physical forms. Calves were weaned on d 56 and remained in the study until d 70. All calves had free access to drinking water and the starter feeding at all times. No interaction was detected between the physical forms of starter feeds and forage source concerning starter intake, dry matter intake, metabolizable energy (ME) intake, average daily gain (ADG)/ME intake, ADG, and feed efficiency (FE). The preweaning and overall starter feed intake, dry matter intake, and ME intake were greater for calves fed TF starter diets than those fed CM starter diets. The ADG/ME intake was greater for calves fed TF starter diets than that fed CM starter. The FE was greater for calves fed TF starter diets compared with those fed CM starter during the preweaning, postweaning, and overall periods. The WS improved FE during the postweaning period compared with AH. The physical form of starter, forage source, and their interaction did not affect plasma glucose, triglycerides, and very low-density lipoprotein concentrations. Ruminal pH was greater for calves fed TF starter diets than those fed CM starter on d 30 of life. An interaction was observed between the physical forms of starter diets and forage source for β-hydroxybutyrate on d 28. These results showed that when starter diets contained similar ingredients and nutrient contents, processing calf starters to reduce the number of fine particles can improve the growth performance in dairy calves. The effects of supplementation with grass silage and replacement of some corn in the concentrate with soybean meal (SBM) on milk production, and milk fatty acid (FA) profiles were evaluated in a replicated 4 × 4 Latin square study using 16 dairy cows grazing pasture composed of ryegrass, Kentucky bluegrass, and white clover. Each experimental period lasted for 3 wk. The 4 dietary treatments were PC, 20 h of access to grazing pasture, supplemented with 6 kg/d of corn-based concentrate mixture (96% corn; C); PCSB, 20 h of access to grazing pasture, supplemented with 6 kg/d of corn- and SBM-based concentrate mixture (78% corn and 18% SBM; CSB); SC, 7 h of access to grazing pasture during the day and 13 h of ad libitum access to grass silage at night, supplemented with 6 kg/d of C concentrate; and SCSB, 7 h of access to grazing pasture during the day and 13 h of ad libitum access to grass silage at night, supplemented with 6 kg/d of CSB concentrate. The concentrate mixtures were offered twice each day in the milking parlor and were consumed completely. Grass silage supplementation reduced dietary crude protein and concentration of total sugars, and dietary SBM inclusion increased dietary crude protein concentration and decreased dietary starch concentration. Milk yield and energy-corrected milk were increased by SBM supplementation of cows with access to grass silage. Milk protein concentration was lower in cows offered grass silage, regardless of whether SBM was fed. Dietary SBM inclusion tended to increase milk fat concentration. Plasma urea N was reduced by silage feeding and increased by SBM supplementation. Supplementation with grass silage overnight could represent a useful strategy for periods of lower pasture availability. Dietary inclusion of SBM in solely grazing cows had no effects on milk production and composition, exacerbated the inefficient capture of dietary N, and increased diet cost. Grass silage supplementation affected milk FA profiles, increasing both the FA derived from de novo synthesis and those derived from rumen microbial biomass, and decreasing the sum of C18 FA (mostly derived from diet or from mobilization of adipose tissue reserves). Milk fat concentrations of conjugated linoleic acid cis-9, trans-11, vaccenic acid (18:1 trans-11), and linolenic acid (18:3n-3) were unaffected by grass silage supplementation, suggesting that partial replacement of pasture by unwilted grass silage does not compromise the dietary quality of milk fat for humans.  相似文献

The objectives were to evaluate the effects of differential supplementation of Ca salts (CS) of fatty acids (FA) on plasma acute phase proteins and both FA composition and function (i.e., activity and cytokine production) of neutrophils, during the peripartum and breeding periods. Holstein cows were assigned randomly to receive either CS of palm (PO) or safflower (SO) oils from 30 d prepartum until 35 d postpartum (dpp) and CS of PO or fish oil (FO) from 35 to 160 dpp. Supplementation of CS of FA was at 1.5% of dietary dry matter. Cows (n = 32) were sampled three times weekly from parturition to 35 dpp for analyses of plasma concentrations of haptoglobin and fibrinogen. Cows (n = 47) were sampled for neutrophil phagocytic and oxidative burst activities toward Escherichia coli and Staphylococcus aureus, and neutrophil abundances of L-selectin and β2-integrin assessed by flow cytometry at 32 d prepartum, within 7 h after parturition, and 4 and 7 dpp. Profiles of FA in neutrophils and cytokine production (i.e., tumor necrosis factor alpha, TNF-α, and IL-1β) were assessed prepartum (n = 14), 35 (PO vs. SO; n = 26) and 85 dpp (PO vs. FO; n = 28). Plasma concentrations of haptoglobin and fibrinogen were greater for cows fed SO compared with PO. The percentage of neutrophils with phagocytic and oxidative burst activities was not affected by transition diets, but activities per neutrophil were greater in SO compared with PO diets at 4 (phagocytosis and oxidative burst) and 7 dpp (oxidative burst). Neutrophil abundance of L-selectin, but not β2-integrin, was greater in SO compared with PO at 4 and 7 dpp. Neutrophil productions of TNF-α and IL-1β were increased at 35 dpp in SO compared with PO diets, but production of TNF-α was attenuated in FO compared with PO at 85 dpp. Neutrophil ratios of n-6:n-3 FA were greater at 35 dpp in the SO diet and less at 85 dpp in FO compared with PO diets. In conclusion, cows supplemented with CS of SO had improved innate immunity (i.e., acute phase response and neutrophil function) to better cope with the bacterial challenges in the postpartum period. The objective of this study was to determine the effects of feeding an increased amount of extruded flaxseed with high proportions of n-3 fatty acids (FA) to transition dairy cows on performance, energy balance, and FA composition in plasma, adipose tissue, and milk fat. Multiparous Israeli-Holstein dry cows (n = 44) at 256 d of pregnancy were assigned to 2 treatments: (1) control cows were fed prepartum a dry-cow diet and postpartum a lactating-cow diet that consisted of 5.8% ether extracts; and (2) extruded flaxseed (EF) cows were supplemented prepartum with 1 kg of extruded flaxseed (7.9% dry matter)/cow per d, and postpartum were fed a diet containing 9.2% of the same supplement. The EF supplement was fed until 100 d in milk. On average, each pre- and postpartum EF cow consumed 160.9 and 376.2 g of C18:3n-3/d, respectively. Postpartum dry matter intake was 3.8% higher in the EF cows. Milk production was 6.4% higher and fat content was 0.4% U lower in the EF group than in the controls, with no differences in fat and protein yields. Energy balance in the EF cows was more positive than in the controls; however, no differences were observed in concentrations of nonesterified fatty acids and glucose in plasma. Compared with controls, EF cows had greater proportions of C18:3n-3 in plasma and adipose tissue. The proportion of n-3 FA in milk fat was 3.7-fold higher in the EF cows, and the n-6:n-3 ratio was decreased from 8.3 in controls to 2.3 in the EF cows. Within-group tests revealed that the C18:3n-3 content in milk fat in the EF cows was negatively correlated with milk fat percentage (r = –0.91) and yield (r = –0.89). However, no decrease in de novo synthesis of less than 16-carbon FA was found in the EF group, whereas C16:0 yields were markedly decreased. It appears that the enrichment of C18:3n-3 in milk fat was limited to approximately 2%, and the potential for increasing this n-3 FA in milk is higher for cows with lower milk fat contents. In conclusion, feeding increased amounts of C18:3n-3 during the transition period enhanced dry matter intake postpartum, increased milk production, decreased milk fat content, and improved energy balance. Increased amounts of EF considerably influenced the FA profile of plasma, adipose tissue, and milk fat. However, the extent of C18:3n-3 enrichment in milk fat was limited and was negatively correlated with milk fat content and yield.  相似文献

The aim of this study was the evaluation of climate sensitivity via genomic reaction norm models [i.e., to infer cow milk production and milk fatty acid (FA) responses on temperature-humidity index (THI) alterations]. Test-day milk traits were recorded between 2010 and 2016 from 5,257 first-lactation genotyped Holstein dairy cows. The cows were kept in 16 large-scale cooperator herds, being daughters of 344 genotyped sires. The longitudinal data consisted of 47,789 test-day records for the production traits milk yield (MY), fat yield (FY), and protein yield (PY), and of 20,742 test-day records for 6 FA including C16:0, C18:0, saturated fatty acids (SFA), unsaturated fatty acids (UFA), monounsaturated fatty acids (MUFA), and polyunsaturated fatty acids (PUFA). After quality control of the genotypic data, 41,057 SNP markers remained for genomic analyses. Meteorological data from the weather station in closest herd distance were used for the calculation of maximum hourly daily THI. Genomic reaction norm models were applied to estimate genetic parameters in a single-step approach for production traits and FA in dependency of THI at different lactation stages, and to evaluate the model stability. In a first evaluation strategy (New_sire), all phenotypic records from daughters of genotyped sires born after 2010 were masked, to mimic a validation population. In the second strategy (New_env), only daughter records of the new sires recorded in the most extreme THI classes were masked, aiming at predicting sire genomic estimated breeding values (GEBV) under heat stress conditions. Model stability was the correlation between GEBV of the new sires in the reduced data set with respective GEBV estimated from all phenotypic data. Among all test-day production traits, PY responded as the most sensitive to heat stress. As observed for the remaining production traits, genetic variances were quite stable across THI, but genetic correlations between PY from temperate climates with PY from extreme THI classes dropped to 0.68. Genetic variances in dependency of THI were very similar for C16:0 and SFA, indicating marginal climatic sensitivity. In the early lactation stage, genetic variances for C18:0, MUFA, PUFA, and UFA were significantly larger in the extreme THI classes compared with the estimates under thermoneutral conditions. For C18:0 and MUFA, PUFA, and UFA in the middle THI classes, genetic correlations in same traits from the early and the later lactation stages were lower than 0.50, indicating strong days in milk influence. Interestingly, within lactation stages, genetic correlations for C18:0 and UFA recorded at low and high THI were quite large, indicating similar genetic mechanisms under stress conditions. The model stability was improved when applying the New_env instead of New_sire strategy, especially for FA in the first stage of lactation. Results indicate moderately accurate genomic predictions for milk traits in extreme THI classes when considering phenotypic data from a broad range of remaining THI. Phenotypically, thermal stress conditions contributed to an increase of UFA, suggesting value as a heat stress biomarker. Furthermore, the quite large genetic variances for UFA at high THI suggest the consideration of UFA in selection strategies for improved heat stress resistance.  相似文献

Neonatal Holstein calves were fed texturized calf starters containing 33% whole (WC), dry-rolled (DRC), roasted-rolled (RC), or steam-flaked (SFC) corn to investigate how corn processing method affects intake, growth, rumen and blood metabolites, and rumen development. In the first experiment, 92 Holstein calves (52 male and 40 female) were started at 2 +/- 1 d of age and studied for 42 d. Starter dry matter (DM) intake was measured and fecal scoring conducted daily. Growth and blood parameter measurements were conducted weekly. A subset of 12 male calves (3/treatment) was euthanized at 4 wk of age and rumen tissue sampled for rumen epithelial development measurements. Experiment 2 consisted of 12 male Holstein calves ruminally cannulated at 7 +/- 1 d of age. Rumen fluid and blood samples were collected during wk 2 to 6. In the first experiment, postweaning and overall starter and total DM intake were significantly higher in calves fed starter with DRC than RC or SFC. Postweaning and overall starter and total DM intake were significantly higher in calves fed starter with WC than SFC. Postweaning average daily gain was significantly greater in calves fed starter with DRC than SFC. Blood volatile fatty acid concentrations were significantly higher in calves fed starter with SFC than in calves fed all other treatments. Papillae length and rumen wall thickness at 4 wk were significantly greater in calves fed starter with SFC than DRC and WC, respectively. In experiment 2, calves fed starter with WC had higher rumen pH and lower rumen volatile fatty acid concentrations than calves fed all other starters. Rumen propionate production was increased in calves receiving starter with SFC; however, rumen butyrate production was higher in calves fed starter with RC. Results indicate that the type of processed corn incorporated into calf starter can influence intake, growth, and rumen parameters in neonatal calves. Calves consuming starter containing RC had similar body weight, feed efficiency, and rumen development but increased structural growth and ruminal butyrate production when compared with the other corn processing treatments.  相似文献

The objective of this study was to evaluate the effects of feeding gallic acid on the growth, nutrient digestibility, plasma metabolites, rumen fermentation, and bacterial community in the rumen fluid and feces of preweaning calves. Thirty-six female Holstein calves with similar ages (means ± SD; 3.1 ± 1.39 d) and body weights (40.8 ± 2.87 kg) were randomly assigned to receive 3 treatments. Calves were fed 1 of 3 treatments as follows: basal diet with no gallic acid (control), 0.5 g/kg gallic acid in starter diet (low), and 1 g/kg gallic acid in starter diet (high). The results showed that feeding gallic acid increased growth by improving the starter intake and average daily gain of the calves. The fecal score tended to decrease in a linear manner with the addition of gallic acid. Total-tract apparent protein digestibility tended to increase linearly with feeding gallic acid. Feeding gallic acid led to a linear increase in the plasma total protein and β-hydroxybutyrate levels. In addition, feeding gallic acid linearly increased catalase and total antioxidant capacity levels and decreased malondialdehyde and tumor necrosis factor-α concentrations. The concentrations of total volatile fatty acids, propionate, butyrate, and valerate in the rumen fluid increased linearly with the addition of gallic acid, resulting in a linear pH reduction. Feeding gallic acid linearly increased the relative abundances of Prevotella_1, Saccharofermentans, and Prevotellaceae_UCG-001 and linearly decreased the relative abundance of Prevotella_7 in the rumen fluid. The Shannon index of ruminal bacterial communities linearly increased by feeding gallic acid. Feeding gallic acid linearly increased the relative abundances of Ruminococcaceae_UCG-005, Bacteroides, and Christensenellaceae_R-7_group in the feces. In summary, feeding gallic acid improved growth, antioxidant function, and rumen fermentation and altered the bacterial community in the rumen fluid and feces of preweaning dairy calves.  相似文献

Interest in changing the milk fatty acid profile is growing. However, little is known about the genetic variability of milk fatty acids in the US Holstein population. Therefore, genetic parameters for milk fatty acids were estimated using a single-trait, mixed, linear animal model on 592 individual milk samples from 233 daughters of 53 sires in a cow herd genetically representative of the US Holstein population. Heritability (h2) and repeatability (r) estimates ± standard errors for yields of individual fatty acids ranged from 0.00 ± 0.08 (C4:0) to 0.43 ± 0.13 (C12:0) for heritabilities and from 0.21 ± 0.05 (C18:1) to 0.43 ± 0.05 (C12:0) for repeatabilities. Saturated (h2 = 0.23 ± 0.12; r = 0.36 ± 0.05) and de novo synthesized fatty acids (C6:0 to C14:0; h2 = 0.30 ± 0.13; r = 0.40 ± 0.05) had numerically higher estimates than did monounsaturated (h2 = 0.09 ± 0.09; r = 0.22 ± 0.05) and polyunsaturated fatty acids (h2 = 0.08 ± 0.09; r = 0.27 ± 0.05). For relative proportions of individual fatty acids, the greatest heritability and repeatability estimates were obtained for C8:0 (h2 = 0.18 ± 0.12; r = 0.36 ± 0.05), C10:0 (h2 = 0.22 ± 0.13; r = 0.46 ± 0.05), C12:0 (h2 = 0.18 ± 0.12; r = 0.46 ± 0.05), C16:0 (h2 = 0.09 ± 0.12; r = 0.48 ± 0.05), C16:1 (h2 = 0.49 ± 0.13; r = 0.49 ± 0.05), and C18:0 (h2 = 0.24 ± 0.11; r = 0.39 ± 0.05). Our results suggest the existence of genetic variability of milk fatty acids, in particular of medium-and long-chain fatty acids (C8:0 to C18:0), which could be used to improve the nutritional and textural properties of milk fat by selective breeding.  相似文献

Most cows encounter a state of negative energy balance during the periparturient period, which may lead to metabolic disorders and impaired fertility. The aim of this study was to assess the potential of milk fatty acids as diagnostic tools of detrimental levels of blood plasma nonesterified fatty acids (NEFA), defined as NEFA concentrations beyond 0.6 mmol/L, in a data set of 92 early lactating cows fed a glucogenic or lipogenic diet and subjected to 0-, 30-, or 60-d dry period before parturition. Milk was collected in wk 2, 3, 4, and 8 (n = 368) and blood was sampled weekly from wk 2 to 8 after parturition. Milk was analyzed for milk fatty acids and blood plasma for NEFA. Data were classified as “at risk of detrimental blood plasma NEFA” (NEFA ≥0.6 mmol/L) and “not at risk of detrimental blood plasma NEFA” (NEFA <0.6 mmol/L). Concentrations of 45 milk fatty acids and milk fat C18:1 cis-9-to-C15:0 ratio were subjected to a discriminant analysis. Milk fat C18:1 cis-9 revealed the most discriminating variable to identify detrimental blood plasma NEFA. A false positive rate of 10% allowed us to diagnose 46% of the detrimental blood plasma NEFA cases based on a milk fat C18:1 cis-9 concentration of at least 230 g/kg of milk fatty acids. Additionally, it was assessed whether the milk fat C18:1 cis-9 concentrations of wk 2 could be used as an early warning for detrimental blood plasma NEFA risk during the first 8 wk in lactation. Cows with at least 240 g/kg of C18:1 cis-9 in milk fat had about 50% chance to encounter blood plasma NEFA values of 0.6 mmol/L or more during the first 8 wk of lactation, with a false positive rate of 11.4%. Profit simulations were based on costs for cows suffering from detrimental blood plasma NEFA, and costs for preventive treatment based on daily dosing of propylene glycol for 3 wk. Given the relatively low incidence rate (8% of all observations), continuous monitoring of milk fatty acids during the first 8 wk of lactation to diagnose detrimental blood plasma NEFA does not seem cost effective. On the contrary, milk fat C18:1 cis-9 of the second lactation week could be an early warning of cows at risk of detrimental blood NEFA. In this case, selective treatment may be cost effective.  相似文献
