We have developed an assay for the isolation and quantitation by gas chromatography-mass spectrometry (GC-MS) of free 9- and 13-hydroxyoctadecadienoic acid (9-HODE, 13-HODE) in the mammary glands of female mice. Internal standards consisting of 18O2-labeled analogs of 9- and 13-HODE are added to pulverized frozen tissue prior to extraction with ethanol. Nonlipid materials are removed in a chloroform/methanol/water extraction step. The remaining lipid material is methylated with ethereal diazomethane, and much of the nonoxygenated fatty acid methyl esters are removed via silica solid-phase extraction. Samples are either further derivatized with bis(trimethylsilyl)trifluoroacetamide to form the trimethylsilyl ethers for quantitative analysis by GC-MS or are analyzed as the methyl esters by chiral high-performance liquid chromatography to determine the enantiomeric distribution of the 9- and 13-HODE. The extraction and quantitation protocol was applied to the analysis of mammary glands for free 9- and 13-HODE from mice fed isocaloric diets containing 20% corn oil, 5% corn oil, or 20% beef tallow. Chiral analysis of the products showed higher production of 13(S)-HODE relative to 13(R)-HODE; the enantiomeric excess is most likely due to enzymatic production of 13-HODE superimposed on a background of autoxidative production of 13(R)-plus 9(S)- and 9(R)-HODE. In addition, the effect of sample handling and storage conditions on the formation of 9- and 13-HODE in the samples was assessed by exposing aliquots of a common pool of rat mammary gland tissue to specified conditions prior to analysis. This methodology will be important during investigations of the contribution of linoleate oxidation products to the enhancement of mammary tumorigenesis by dietary fat.  相似文献

A total of 82 dietary fats sold on the Bulgarian market in the period 1995—2000 were analyzed. The samples included 68 table margarines (50 of which were imported), 10 frying fats (6 imported) and 4 salad dressings (all imported). A validated analytical method, thin‐layer chromatography‐AgNO3‐densitometry, was used. It enabled direct determination of all fatty acid groups, differing by degree of unsaturation and double bonds geometry. Low levels of trans fatty acids (TFA) down to 0.1% of the total for mono trans‐trienoic (Tcct) and mono trans‐dienoic (Dct), and down to 0.2% for trans‐monoenoic (Mt) were quantitated, with an error under 3% and a standard deviation of 0.1—1.5. The total content of TFA in table margarines varied from 0 to 26.9% with a mean value of 8.6 ± 7.2% for imported and 1.6 ± 3.4% for Bulgarian samples. Saturated fatty acids (SFA) content varied from 11.5 to 45.7%, with a mean value of 25.4 ± 5.7% for imported and 26.9 ± 5.2% for Bulgarian margarines. A general trend of lower levels of TFA and SFA in imported margarines was observed over the studied period. Additionally, the content of individual saturated fatty acids was determined by gasliquid chromatography in 37 of all studied samples.  相似文献

The effect of dietary C18:1 trans fatty acids on back fat composition in pigs was investigated with special emphasis on conjugated linoleic acids (CLA). A total of 12 × 4 siblings of Large White and Swiss Landrace breed were housed in groups and fattened from 22 to 103 kg live weight. Pigs were fed a control diet (barley, wheat, soybean meal) or experimental diets which consisted of the control diet with a 5% replacement of olein or stearin fractions of pork fat, or partially hydrogenated fat. The hydrogenated fat was rich in C18:1 trans fatty acids but contained only negligible amounts of CLA. In contrast olein and stearin fractions contained far less C18:1 trans fatty acids but some CLA. In the control diet no C18:1 trans fatty acids and only traces of CLA were detected. The partially hydrogenated fat led to the highest CLA content in back fat (0.44%). Intermediate amounts of CLA were measured in pigs fed the fractionated pork fat (0.22/0.23%). In pigs fed the control diet, also small amounts of CLA were detected. The results indicate that CLA may be produced by endogenous Δ9‐desaturation out of dietary trans vaccenic acid in pigs.  相似文献

单针藻是制备生物柴油较好的原料。以产油单针藻为对象,研究了单针藻在异养培养中生长、代谢的特性及脂肪酸组成变化。结果表明,Richards模型能够较好拟合藻细胞的生长与培养时间的关系。建立了微藻细胞对硝酸盐、磷酸盐的代谢模型,得出藻细胞生物量对硝酸盐、磷酸盐的得率系数分别为5.319 g/g、500 g/g。生长过程中脂肪酸组成变化的分析表明,延长藻细胞的培养稳定期可增加C18∶1的含量。  相似文献

High concentrations of polyunsaturated fatty acids (PUFA) in meat have detrimental effects on its technical properties. The present study was carried out to investigate whether PUFA levels in pork can be reduced by increasing the concentrations of oleic acid in pig diets. To this end a bifactorial experiment was carried out with 48 female growing finishing pigs. Six different diets were used with two different concentrations of linoleic acid (12 vs. 24 g/kg) and three different concentrations of oleic acid (12 vs. 18 vs. 24 g/kg). The experiment started at a body weight (BW) of 58 kg and continued until 115 kg BW. The fatty acid composition of total lipids of backfat, perirenal fat and musculus (m.) longissimus dorsi was analysed. Concentrations of linoleic acid and total PUFA in backfat and perirenal fat were affected only by the dietary linoleic acid content but not at all by the dietary oleic acid content. Increasing the dietary concentration of oleic acid raised the level of oleic acid in those tissues at the expense of saturated fatty acids, suggesting competition between monounsaturated fatty acids and saturated fatty acids for incorporation into triglycerides. At the low dietary linoleic acid concentration, the percentages of linoleic acid and total PUFA in total lipids of m. longissimus dorsi were also unaffected by the dietary oleic acid content. In contrast, at the high dietary linoleic acid concentration, percentages of linoleic acid and total PUFA of the m. longissimus dorsi were reduced by increasing the dietary concentration of oleic acid, suggesting that oleic acid and linoleic acid compete for incorporation into muscle lipids. Thus, at high dietary linoleic acid levels the fatty acid composition of the m. longissimus dorsi was favourably affected by high dietary oleic acid concentrations; in backfat and perirenal fat, however, no beneficial effect of high dietary oleic acid levels was seen.  相似文献

The purpose of the study was to assess changes in the fatty acid composition of raw and grilled pig muscles after different storage periods. A total of 13 female and 12 castrated Pietrain×German Landrace pigs were fed a basal concentrate diet supplemented with 5% olive oil or 5% linseed oil during the growing‐finishing period. An entire cut of the pork loin with bone (15th rib to 5th lumbar vertebra) was stored at 5 °C for 48, 96 or 144 h. Simultaneous analyses of intramuscular fat and lipid composition were carried out on raw and grilled longissimus muscles following different storage intervals. Dietary inclusion of linolenic acid by linseed oil feeding effectively increased the long‐chain n‐3 fatty acids, whereas in the olive oil group the oleic acid in pork was higher. Mean total lipid ranged from 1.8 to 2.3% for raw and from 2.6 to 3.5% for grilled pork chops. The relative proportions of lauric acid, stearic acid and oleic acid significantly increased with storage time, while the percentages of linoleic, arachidonic, eicosapentaenoic acid and the sum of polyunsaturated fatty acids, especially n‐6 fatty acids, were decreased. Compared with raw muscle, grilling affected the relative fatty acid profile only slightly. Related to the original weight, storage and grilling increased the total fatty acid contents and the sum of saturated, monounsaturated, n‐6 and n‐3 fatty acids of loin chops, as a result of water losses.  相似文献

The aim of the present study was to identify and quantitatetrans isomers of C18 fatty acids in some French infant formulas. Twenty powdered infant formulas were purchased in pharmacies and supermarkets in order to assess theirtrans mono- and poly-unsaturated fatty acids content. The fatty acid profiles were examined using methyl and isopropyl ester derivatives. The combination of gas-liquid chromatography, high-performance liquid chromatography, and silver nitrate thin-layer chromatography was needed to describe the detailed fatty acid compositions of the samples, includingtrans isomers of unsaturated C18 fatty acids. All the samples containedtrans isomers of C18∶1 acid (mean level 1.97±0.28% of total fatty acids), with vaccenic acid being generally the major isomer (15 out of 20 samples), thus indicating the origin from bovine milk. All the formulas also contained various isomers of linoleic and α-linolenic acids, but at lower levels.Trans PUFA isomers are the same as those present in deodorized oils. In conclusion, all the infant formulas analyzed in this study contained sometrans fatty acids, including isomers of essential fatty acids. This should be taken into account in the dietary intake of the newborn.  相似文献

Chlorine dioxide (CIO2) has been explored as a potential substitute for aqueous chlorine to clean seafood products. In an attempt to understand the interaction of CIO2 with organic compounds, duplicate fillets of Atlantic salmon (Salmo salar) and red grouper (Epinephelus morio) were treated for 5 min with freshly prepared aqueous CIO2 at 20, 40, 100, and 200 ppm total available CIO2 in 3.5% brine. Thiobarbituric acid (TBA) values and fatty acid composition were determined. CIO2-treated salmon and red grouper showed a dose-related increase in TBA; the 100 and 200 ppm groups had significantly (P<0.05) greater TBA values than controls and the 20 ppm group. Treated red grouper and salmon did not differ in percentage monounsaturated and polyunsaturated fatty acids compared to controls, although differences occurred with some individual fatty acids. Thus, CIO2 treatment did not greatly affect fatty acid composition of treated fillets.  相似文献

Lipid and fatty acid composition are considered to be key parameters that determine the nutritive quality of phytoplankton diets for zooplanktonic herbivores. The fitness, reproduction and physiology of the grazers are influenced by these factors. The trophic transfer of lipids and fatty acids from algal cells has been typically studied by using simple extraction and quantification approaches, which, as we argue here, do not reflect the actual situation in the plankton. We show that cell disruption, as it occurs during a predator's grazing on diatoms can drastically change the lipid and fatty acid content of the food. In some algae, a rapid depletion of polyunsaturated fatty acids (PUFAs) is observed within the first minutes after cell disruption. This fatty acid depletion is directly linked to the production of PUFA-derived polyunsaturated aldehydes (PUA); these are molecules that are thought to be involved in the chemical defence of the algae. PUA-releasing diatoms are even capable of transforming lipids from other sources if these are available in the vicinity of the wounded cells. Fluorescent staining reveals that the enzymes involved in lipid transformation are active in the foregut of copepods, and therefore link the depletion processes directly to food uptake. Incubation experiments with the calanoid copepod Temora longicornis showed that PUFA depletion in PUA-producing diatoms is correlated to reduced hatching success, and can be compensated for by externally added single fatty acids.  相似文献

The fatty acid composition was determined in 39 samples of beef, 20 samples of veal, and 34 samples of lamb, representative of the supply of ruminant meat in Denmark. Five cuts of beef and veal and three cuts of lamb with increasing fat content were selected, and analysis of the fatty acid methyl esters was performed by gas-liquid chromatography (GLC) on a polar 50-m capillary column CP Sil 88 with flame-ionization detection. Lamb had the highest content of saturated fatty acids (52.8±1.8 g/100 g fatty acids), higher than beef and veal (45.3±3.1 and 45.4±0.8 g/100 g fatty acids, respectively). Cis monounsaturated fatty acids were 49.2±3.1, 44.9±1.8, and 37.7±1.7, and polyunsaturated fatty acids were 3.3±0.7, 5.8±2.0, and 5.0±0.1 g/100 g fatty acids in beef, veal, and lamb, respectively. Beef contained 2.1±0.8 g trans C18:1 per 100 g fatty acids, about half that found in veal (4.0±1.2 g/100 g fatty acids) and lamb (4.5±0.6 g/100 g fatty acids). Trans C16:1 was 0.24±0.01, 0.14±0.02, and 0.79±0.02 g/100 g fatty acids in beef, veal, and lamb, respectively. Only small variations in trans and other fatty acids could be demonstrated between cuts. The overlap between cis and trans C18:1 by capillary GLC was verified by argentation-thin-layer chromatography followed by GLC, on three samples of veal and three samples of lamb. In veal 1.0 g, and in lamb 1.4 g trans C18:1 per 100 g fatty acids were hidden under the cis C18:1 peak. The mean intake of trans fatty acids from ruminant meat is estimated at 0.2 g/d.  相似文献

We studied the effect of four rapeseed oils with different fatty acid profiles on parameters implicated in the pathogenesis of atherosclerosis in humans in a model experiment with hamsters. The hamsters were divided into seven groups and fed a semi‐synthetic, cholesterol‐enriched diet (5 g/kg diet) containing 15% of the fat in question for a period of six weeks. The following rapeseed oils were used: (1) conventional rapeseed oil (6% saturated fatty acids [SFA], 64% monounsaturated fatty acids [MUFA], 18% linoleic acid [LA], 9% α‐linolenic acid [ALA]), (2) linoleic acid‐rich rapeseed oil (6% SFA, 61% MUFA, 28% LA, 2% ALA), (3) oleic acid‐rich rapeseed oil (6% SFA, 74% MUFA, 11% LA, 5% ALA), (4) myristic acid‐rich rapeseed oil (11% myristic acid, 35% SFA, 44% MUFA, 14% LA, 5% ALA). Sunflower oil, olive oil and lard were used as control fats. The concentrations of the lipids in the plasma, in the lipoprotein fractions and in the liver, the fatty acid composition of various tissues, the tocopherol status and the susceptibility of low‐density lipoproteins (LDL) to in vitro‐oxidation were determined. The concentrations of total cholesterol found in the plasma and in the LDL fraction and the ratios of LDL to HDL were similar after feeding the four different types of rapeseed oil, sunflower oil and olive oil. Lard produced the highest concentrations of cholesterol in plasma and the LDL fraction and the highest ratio of LDL to HDL. Feeding conventional, oleic acid‐ and myristic acid‐rich rapeseed oils resulted in markedly lower ratios of arachidonic to eicosapentaenoic acid in the lipids of the liver and the erythrocytes. This is considered beneficial for the formation of eicosanoids. The lag‐time before the onset of peroxidation of the LDL lipids, induced by copper ions, was not statistically significant between the seven hamster groups suggesting that the susceptibility of LDL to lipid peroxidation was similar after feeding all types of fat. Considering all parameters obtained in the used hamster model it is obvious that all four rapeseed oils are at least as favourable as olive oil or sunflower oil.  相似文献

Various phospholipids may act as antioxidants or prooxidants. This study investigated the effects of three phospholipid classes and their fatty acid composition on antioxidant activity. Antioxidant properties of sphingomyelin, phosphatidylcholine, and phosphatidylethanolamine from salmon and menhaden oil were measured by oxidation induction time. An antioxidant activity index was determined in these systems with the Rancimat 617. Fatty acid profiles of the individual phospholipids and total oils were determined by gas-liquid chromatography before and after oxidation. The index was significantly (P<0.05) influenced by the headgroup and fatty acid composition of the phospholipid. Lipids with a choline headgroup had oxidation induction times greater than 60 h in the salmon oil system. The choline-containing phospholipid also offered better (P<0.05) protection from oxidation to the n-3 and total polyunsaturated fatty acids in salmon oil. Phospholipids containing more saturated fatty acids had longer oxidation induction times (>84 h) and higher antioxidant index (>9). Chainlength of the fatty acids may have contributed to the observed index, as phospholipids with longer chains (i.e., C18 and above) had longer oxidation induction times. Phospholipids tested in this study had little or no antioxidant activity in menhaden oil, nor did they offer protection to n-3 or total polyunsaturated fatty acids in this oil. These findings suggest that fatty acid profiles of individual oils may influence the antioxidant index of each phospholipid.  相似文献

In the present study, the lipid amount and fatty acid profile of different Sparidae species, including gilthead seabream, juvenile gilthead seabream, annular seabream, white seabream, common two-banded seabream were evaluated. Fish were seasonally collected from Köyceğiz Lagoon (Muğla, South Western Turkey) from June 2018 to June 2019 and after collection, the sex of each specimen was recorded. According to the results of the study, the highest lipid amount was found in female annular seabream individuals as 8.09 ± 0.78% in November and the lowest lipid amount was found in male juvenile gilthead seabream as 0.98 ± 0.12% in March. Palmitic acid and oleic acid were determined as the most abundant SFA and MUFA for all species, respectively. The highest value of DHA, which was the predominant PUFA was assessed as 15.33 ± 0.26% in female white seabream in November whereas the lowest value (3.83 ± 0.36%) was found in gilthead seabream in December. The n-6/n-3 ratio was determined between 0.27 ± 0.00 (for male common two-banded seabream in July)-1.20 ± 0.03 (for male gilthead seabream in December) and it followed within the range of healthy values for all species. As a conclusion, it was found that values of lipid and fatty acid profiles among the examined Sparidae species vary among the season of collection. The results of the study gave the seasonal nutritional values of four economically-important Sparidae species that being rich in healthy polyunsaturated fatty acids such as DHA, have beneficial in human nutrition.  相似文献

In this study, the effects of temperature on the fatty acids profile and the effects of temperature on the degree of unsaturation of fatty acids of Oreochromis niloticus were investigated. The analysis was performed by gas chromatography. The study showed that there were large temperature variations (10.0–32.0°C) during the study period (January–December). The highest crude fat content was found in January (3380 mg/100 g) and the lowest in June (2050 mg/100 g). The fatty acids profile showed significantly different diversity (p < 0.05). Total saturated fatty acid (∑SFA) content ranged from 409.54 to 1297.61 mg/100 g, monounsaturated fatty acid (∑MUFA) from 207.68 to 665.81 mg/100 g, and polyunsaturated fatty acid (∑PUFA) from 175.12 to 972.23 mg/100 g. The ∑MUFA and ∑PUFA concentrations were highest in January and lowest in June, and the ∑SFA concentration was lowest in January and highest in June. EPA and DHA contents were highest in January (198.96 mg/100 g) and lowest in June (48.76 mg/100 g). The contents of omega-3 (653.17 mg/100 g) and omega-6 fatty acids (252.54 mg/100 g) were highest in January and lowest in June (ω-3; 106.43 and ω-6; 60.91 mg/100 g). It concluded that the degree of unsaturation of fatty acids increases with decreasing temperature. In this study, the nutritional quality of the FAs profile was assessed using lipid quality indices. The indices indicating dietary quality of lipids by their values: Atherogenic index (0.47), thrombogenic index (0.38), hypocholesterolemic to hypercholesterolemic (3.00), meat fat quality (6.78), ω6/ω3 ratio (0.39), PUFA/SFA (2.37), MUFA/SFA (1.62), PUFA/MUFA (1.46), and PUFA + MUFA/SFA (3.99). These values are within the recommended range, indicating that the lipid profile of O. niloticus has high nutritional quality, which can be further improved by harvesting the fish during the winter season. Due to the nutritional importance of O. niloticus, the culture of this species could have significant interest to the people of Karachi, especially the coastal communities. To promote the nutritional diet in local population, the government should support the aquaculture of Nile tilapia.  相似文献

Black cumin, Nigella sativa L. belonging to Ranunculaceae, is a valuable medicinal plant because of the curative effects of seed and oil. Despite studies on black cumin, there are few studies on the effects of nitrogen on fatty acid composition, no records on thymoquinone yield, and the thymoquinone content of the seed oil. In this study, the effects of different nitrogen doses on the seed oil yield, thymoquinone ratio/yield, and fatty acid compositions were investigated. Field studies with nitrogen doses (0, 40, 60, 80, and 100 kg/ha) were studied using the randomized plot design. As a result of the research, nitrogen doses significantly affected the oil and thymoquinone yields while insignificant on the thymoquinone ratio. The highest oil yield with 501.6 kg/ha was obtained from 80 kg/ha of the nitrogen doses, while the highest thymoquinone yield with 10.24 kg/ha was obtained from the control plots. Thymoquinone yields were in the same statistical group with other nitrogen doses, except for means of 100 kg/ha nitrogen doses. Linoleic, oleic, and palmitic acids were major fatty acids of black cumin, and their variations were insignificant in nitrogen applications. The values of butyric, capric, and linolenic acids varied significantly in different nitrogen doses. As a result of the study, it was determined that the highest oil yield was obtained from 80 kg/ha nitrogen application. It has been determined that nitrogen doses above 60–80 kg/ha should be avoided for the seed oil and thymoquinone yields in black cumin.  相似文献

The aim of this study was to produce high‐quality meat from lambs under different feeding conditions, as measured by the accumulation of n‐3 fatty acids and conjugated linoleic acids (CLA) in muscle and subcutaneous fat. In total, 13 male crossbred lambs (Black Head×Gotland), each at 24 kg live weight, were divided into two feeding groups. Lambs were kept either on pasture (pasture grazing, n = 6) or in the stable (concentrate feeding, n = 7). The linolenic acid (C18:3n‐3) contained in the grass was absorbed and deposited into the different lipid classes of muscle and subcutaneous fat. The proportion of total n‐3 fatty acids in the different lipids of grazing lambs was significantly (p = 0.05) higher compared to that in concentrate‐fed lambs. The n‐6/n‐3 ratio (mean ± SEM) in muscle of grazing lambs was 1.2 ± 0.09 in contrast to 2.3 ± 0.09 (p = 0.05) of the animals kept in the stable. In subcutaneous fat, this ratio was 0.9 ± 0.2 in lambs kept on pasture versus 3.5 ± 0.2 (p = 0.05) after indoor keeping. The relative concentration of C18:1trans‐11 in total muscle lipids, phospholipids, triacylglycerols and subcutaneous fat was significantly increased by grass feeding compared to concentrate feeding. Significant influences of feeding were shown for saturated fatty acids. In concentrate‐fed lambs, a lower content of saturated fatty acids was detected. The proportion of CLAcis‐9,trans‐11 (1.9 ± 0.2% vs. 1.1 ± 0.1% in muscle, 2.5 ± 0.2% vs. 1.4 ± 0.2% in subcutaneous fat, 0.7 ± 0.04% vs. 0.4 ± 0.04% in phospholipids) in lambs was significantly (p = 0.05) higher after grazing than after concentrate feeding, respectively.  相似文献

The effect of a diet containingtrans-fatty acids (tFA) on the fatty acid composition and fat accumulation in adipose tissue was investigated in mice. Male C57BI/6J mice were fed Control or Trans Diets that were similar, except that 50% of the 18∶1, which was allcis in the Control Diet, was replaced bytFA in the Trans Diet. At selected ages, body weight, epididymal fat pad weight, perirenal fat yield, adipose tissue cellularity and fatty acid composition were examined. Over the time period studied (2–24 mon), the proportion of 18∶0 and 16∶0 tended to decrease whilecis-18∶1 levels increased. Compared to the Control Diet, the Trans Diet resulted in adipose tissue lipids with higher percentages of 14∶0 and 18∶2n−6 and lower percentages ofcis-18∶1 and 20∶4n−6. In polar lipids,tFA replaced saturated fatty acids, whereastFA replacedcis-18∶1 in the nonpolar lipids. Body weights at 16 and 24 mon of age and epididymal fat pad weights at 8–24 mon of age were lower in mice fed the Trans Diet as compared to those fed the Control Diet. At the ages studied, the Trans Diet also resulted in lower values for perirenal fat weights, triacylglycerol to polar lipid ratios, and adipose cell size. The data suggest that chronic consumption oftFA affects lipid metabolism and results in decreased fat accumulation in murine adipose tissue.  相似文献

The influence of the distribution of polyunsaturated fatty acids on the glycerol backbone of dietary triacylglycerols on the fatty acid profile of adipose tissue and muscle phospholipids was investigated in growing‐finishing pigs (48) and broiler chicken (84). The animals were fattened on barley/soybean meal diets supplemented with a blend of soybean oil and beef tallow, either in the ratio 3:1 w/w (high‐PUFA) or 1:3 w/w (low‐ PUFA). Part of the high‐ and low‐PUFA blends was chemically interesterified to randomly distribute all fatty acids over the three positions of the glycerol. Thus, two sets of diets of identical overall fatty acid composition, but differing in the distribution of fatty acids in the triacylglycerols, were fed. Growth performance and carcass composition were neither affected by fatty acid composition nor by randomisation of dietary fats in either animal species. Apparent digestibility of energy was slightly lower in pigs fed the low‐PUFA blends. Fatty acid profile of subcutaneous fat of pigs and broilers as well as of internal body fat (lamina subserosa) and muscle phospholipids of pigs varied according to the dietary fatty acid composition but was not affected by randomisation of dietary fats. These findings are explained in terms of the hydrolysis of TAG during transport of lipids from enterocytes to adipose tissue cells and the continuous lipolysis and re‐esterification of fatty acids that take place in adipose tissue cells.  相似文献
