Low level intake of drugs from the ingestion of contaminated feed may lead to residue problems in food animals. Sulfadimidine (SDD) was used as a model to determine the residue risk at various doses in laying hens. The drug was administered as a single intravenous injection (100 mg kg-1 body weight, BW), as a single oral dose (100, 30, 10, 3, 1 mg kg-1 BW) and via medicated feed for 7 consecutive days (30, 10, 3 mg kg-1 BW). Drug levels were determined with HPLC-UV for plasma, yolk and albumen. Pharmacokinetic values, which were calculated using a first-order one-compartment model, residue levels and transfer rates into the eggs were found to be dose-dependent. Even low doses of 3 and 1 mg kg-1 BW resulted in measurable residues in yolk and albumen 1 day after a single oral administration. After ingestion of medicated feed at 3 mg kg-1 BW, mean drug levels at 0.14 ± 0.01 µg g-1 were found in albumen and at 0.09 ± 0.01 µg ml-1 in plasma. Generally, the residue levels in albumen and plasma were higher than in yolk. These findings demonstrate a residue risk for the consumer even after low level intake of drugs. 相似文献

A liquid chromatography–tandem mass spectrometry method, recently developed, validated and accredited, was used to screen for metronidazole, ronidazole dimetridazole ipronidazole, ternidazole, tinidazole, ornidazole carnidazole and three hydroxy metabolites (hydroxy-metronidazole, HMMNI and hydroxy-ipronidazole) in Irish retail egg samples. The method used had decision limits (CCα) in the range 0.33–1.26?µg?kg?1 and detection capabilities (CCβ) ranging 0.56–2.15?µg?kg?1 for all analytes. Internal standard-corrected recovery, calculated for the various analytes, ranged 87.2–106.2%, while the coefficient of variance, expressed as % CV, ranged 3.7–11.3%. The method was applied to 160 samples of caged, free range and organic hen and duck eggs available on the Irish retail market as well as two incurred proficiency test egg samples. No nitroimidazole residues were detected in the survey samples above the CCα and the results achieved for the two proficiency test samples were acceptable when compared with the assigned values. 相似文献

The depletion of doxycycline (DC) residues in eggs was determined after oral drug administration by drinking water to laying hens. The antibiotic was supplied to birds for 5 consecutive days and the eggs were collected during medication and 18 days after withdrawal. DC residues were determined by LC-MS/MS. DC was isolated from eggs with a solution of 0.02 M of oxalic acid (pH 4), 0.1 M Na2EDTA and acetonitrile. The limit of detection (LOD) and limit of quantification (LOQ) of the method were 2 and 5 µg kg–1, respectively. Analyses were performed on whole egg, egg white and yolk separately. DC was detectable 24 h after the beginning of administration. The concentration of antibiotic increased daily, resulting in the highest DC concentration in whole eggs at the first day of the withdrawal period. Thirteen days after withdrawal, the content of DC in whole eggs was below the LOQ of the method. However, some differences were found in the depletion curve of DC between egg white and yolk. Residues of DC in egg white were much higher during treatment and 1 day after withdrawal, but later the concentration in egg white decreased fairly rapidly and a higher DC content in egg yolk was observed. The depletion period was shorter for egg white than for yolk, and DC was detected in the egg white until 12 days after withdrawal and 2 days more in egg yolk than in white. DC reached a peak faster in egg white, but the residues were detectable for longer period in the yolk. 相似文献

Probabilistic and deterministic estimates of the acute and chronic exposure of the Danish populations to dithiocarbamate residues were performed. The Monte Carlo Risk Assessment programme (MCRA 4.0) was used for the probabilistic risk assessment. Food consumption data were obtained from the nationwide dietary survey conducted in 2000–02. Residue data for 5721 samples from the monitoring programme conducted in the period 1998–2003 were used for dithiocarbamates, which had been determined as carbon disulphide. Contributions from 26 commodities were included in the calculations. Using the probabilistic approach, the daily acute intakes at the 99.9% percentile for adults and children were 11.2 and 28.2 μg kg?1 body weight day?1, representing 5.6% and 14.1% of the ARfD for maneb, respectively. When comparing the point estimate approach with the probabilistic approach, the outcome of the point estimate calculations was generally higher or comparable with the outcome of the probabilistic approach at the 99.9 percentile (consumers only). The chronic exposures for adults and children were 0.35 and 0.76 μg kg?1 body weight day?1 at the 99.9 percentile, representing 0.7% and 1.5%, respectively, of the acceptable daily intake for mancozeb and maneb at 50 μg kg?1 body weight. 相似文献

A liquid chromatography–tandem mass spectrometry (LC–MS/MS) method, which has recently been developed and validated, was used for the identification and quantification of polyether ionophore, macrolide and lincosamide residues in commercial eggs sold in the metropolitan area of Rio de Janeiro, Brazil. The method was applied to 100 samples and the results showed a high incidence of polyether ionophore residues (25%). Salinomycin was detected in 21% of samples, but only two non-compliant results (5.3 and 53 µg?kg?1) were found if maximum limits (tolerances) established by European Union were adopted in Brazil and if a method decision limit (CCα) of 3.4?µg?kg?1 was considered. In 8% of analyzed samples, more than one studied coccidiostat was found. The lincosamide, lincomycin, and the macrolide, tylosin, were detected at trace levels in 4 and 1% of the samples, respectively. Lasalocid, clarithromycin and erythromycin were not found. 相似文献

Eggs were used to study the determination and depletion of amoxicillin (AMO) residues after oral dosing hens (25.0 mg kg–1, 50.0 mg kg–1 body weight), once daily for five days. A reverse-phase high-performance liquid chromatography with fluorescence detection (RP-HPLC-FLD) method was developed to determine AMO residues in albumen, yolk and whole egg. By using pre-column derivatisation, an improved liquid–liquid extraction procedure was developed for sample preparation. AMO were extracted from eggs with acetonitrile. The extract solution was extracted using saturated methylene chloride. The supernatant was reacted with salicylaldehyde under acidic and heating conditions. Limits of detection (LODs) were 1.2 ng g–1 and limits of quantification (LOQs) were 3.9 ng g–1 for AMO. Recoveries of AMO from samples fortified at levels of 5.0–125.0 ng g–1 ranged from 79.1% to 88.5% in albumen, 78.6–83.6% in yolk and 78.3–85.1% in whole egg, with coefficients of variation of ≤7.3%. The maximum concentrations of AMO in albumen, yolk and whole egg were found to occur at 1.5, 2.5, 1.5 days after withdrawal of medication respectively. AMO was not detectable in albumen at 7.5 days after final administration of AMO, at 10.5–11.5 days in yolk and 10.5 days in whole egg after administration of two oral doses. The method was applied during the residue study of AMO in order to formulate a reasonable withdrawal period to ensure food safety. 相似文献

A probabilistic estimation of dietary exposure to cypermethrin residues for the Chinese population was performed. Cypermethrin residue data were obtained from the national food contamination monitoring program for 2001–2006, encompassing 14,096 samples from 36 commodities with a detection rate of 10.4%. Food consumption data were gathered from the national nutrition and health survey conducted in 2002, comprising 65,915 consumers aged 2–100 years and 3701 children of 2–6 years old. The whole country was roughly divided into six regions and the ranges of the median and of P99.9 exposure estimated for these regions were 0.018–0.026 and 3.131–7.095?µg?kg?1?bw?day?1, respectively. Pak-choi and Chinese cabbage contributed 33.9 and 13.2%, respectively, to the cypermethrin intake for the general population, while pak-choi and citrus covered 30.7 and 22.5% of the total intake for children, respectively. The exposure of the rural population was higher than urban populations. Rural areas mainly located in the plains of central China had among the highest exposure of the six regions, accounting for 17.7% of the ARfD at P99.9, while the 99.99th percentile of exposure for children, especially rural children, far exceeded the ARfD, which is a cause for concern. 相似文献

A number of 100 Pu-erh tea samples from the 2013 harvest in Yunnan Province (China) were analysed for 74 pesticides. A total of 11 pesticides were detected. At least one pesticide was detected in 56% of the samples. None of these samples contained the 74 monitored pesticides at concentrations above the Chinese maximum residual levels. Imidacloprid, bifenthrin and acetamiprid were most frequently found, with percentages of 53%, 46% and 31%, respectively. These were also the top three pesticides with maximum concentrations of 140, 246 and 672 μg kg?1, respectively. Residual levels of the monitored pesticides showed no significant correlation with the production time or area of Pu-erh tea. Whereas a high incidence of pesticide residues was detected in Pu-erh tea, the contamination levels observed do not pose any serious health risks. 相似文献

Distribution of semduramicin in hen eggs and tissues after administration of cross-contaminated feed

Semduramicin is an ionophore coccidiostat used in the poultry industry as a feed additive. Cross-contamination of feeds for non-target animals with semduramicin is unavoidable. However, it is not known whether undesirable residues of semduramicin may occur in food after cross-contaminated feed is administered to animals. The aim of the work was to determine the levels of semduramicin in hen eggs (yolks and albumen) and tissues (liver, muscle, spleen, gizzard, ovarian yolks and ovaries) after administration of feed contaminated with 0.27 mg kg?1 of this coccidiostat. The residues were determined using LC-MS/MS. The distribution pattern confirmed the high lipophilicity of semduramicin. Residues were found mainly in egg yolks (28.8 µg kg?1), ovarian yolks (19.5 µg kg?1) and liver (2.57 µg kg?1), while hens’ muscle was free from semduramicin (LOD = 0.1 µg kg?1). BACKGROUND: The objective of this study was to determine the effects of sources of supplemental fat on laying performance, egg quality, and fatty acid composition of egg yolk. RESULTS: Two hundreds Isa Brown layers were assigned randomly to be fed ad libitum a standard commercial layer feed (BD), basal diet plus 2% tallow (T), basal diet plus a mixture of 1% tallow and 1% flaxseed oil (MTFO), basal diet plus 2% sunflower oil (SO), or basal diet plus 2% flaxseed oil (FO), which were offered for 8 weeks. Each diet was given to five groups, each containing 10 hens. The feed efficiency and egg production were 2.78 and 53.51% for BD; 2.30 and 63.47% for T; 2.45 and 60.14% for MTFO; 2.29 and 64.30% for SO, and 2.62 and 61.18% for FO groups, respectively. Dietary fat supplementation affected the laying performance but had no significant effects on egg quality parameters. The fatty acid composition of egg yolk lipids were significantly affected by dietary fatty acid composition. The supplemental tallow increased palmitic fatty acid. The proportions of linoleic and arachidonic fatty acids in egg yolks for layers fed the SO diet were higher than in the BD group and those on diets containing other fats. Concentrations of oleic and omega‐3 fatty acids were the highest in layers fed the FO diet during the laying period. The authors performed exposure and risk assessments based on surveillance studies of retail foods in Japan that were undertaken during the past six years (2004–2010). The exposure to ochratoxin A (OTA) and fumonisins (FBs) in different age groups, including toddlers and young children (1–6 years old), older children (7–14 years old), adolescents (15–19 years old) and adults (over 20 years old) was simulated, and the risk of these mycotoxins was evaluated by comparing the provisional maximum tolerated daily intake (PMTDI) for FBs and the provisional maximum tolerated weekly intake (PMTWI) for OTA established by the FAO/WHO Joint Export Committee on Food Additives. The exposure assessment for both mycotoxins in each age group in Japan indicated that the highest exposure occurred in toddlers and children, but in all cases the percentage of the PMTWI and PMTDI at the 99th percentile of exposure was less than 35% for OTA and 10% for FBs. 相似文献

This study aimed to assess the exposure of the Serbian adult population to As, Cd, Pb and Hg through the consumption of eggs and egg-based products and to assess health risks. Coccidiostat residues were also examined, and their contribution to acceptable daily intake was calculated. Egg samples were collected from 2018 to 2020. The consumption survey was conducted during 2020. A Monte Carlo simulation was performed to estimate the intake of toxic elements based on one-day and seven-day recalls of egg consumption. The results obtained showed that the adult population in Serbia is not exposed to health risks from the studied toxic elements when consuming eggs and egg-based products. The presence of anticoccidial agents was detected in 22% of egg samples at concentrations above the limits prescribed by current regulations in Serbia (which do not allow detectable levels of coccidiostats in eggs), with the main risk of exposure being to coccidiostat residues associated with maduramycin. 相似文献

BACKGROUND: This aim of this study was to investigate the effects of supplementing laying hen diets with different n‐3 fatty acid sources (linseed oil, fish oil and microalgae) on egg quality and fatty acid profile. The addition of inorganic selenium (Se) and iodine (I) and their effects on egg characteristics and Se concentration were also evaluated. RESULTS: A significantly lower egg weight was found in the control group and in groups fed a diet containing fish oil or a diet containing microalgae plus Se and I. Yolks from all supplemented diet groups exhibited a significant reduction in n‐6 fatty acid content compared with the control group. The docosahexaenoic acid (DHA) content of yolks was higher (P < 0.01) in all supplemented diet groups. Selenium supplementation increased the Se concentration (P < 0.001) in eggs. CONCLUSION: The results of this study provide evidence that an improvement in egg weight and in the DHA content of yolks can be achieved by feeding hens a microalgae‐rich diet, which avoids the unpleasant flavours associated with fish oil supplementation. The use of food industry by-products for feeding is restricted by European Union by-product regulation (1774/2002). However, the actual public health risks involved in the use of such products are poorly recognized. This study focuses on bovine milk rejected at the dairy because of a positive result in antimicrobial drug testing and thereafter used as feed for finisher pigs in Finland. In theory, this current practice could expose pork consumers to antimicrobial drug residues. Raw bulk milk samples originating from rejected lots were analysed with a multiresidue method detecting five beta-lactams, including benzylpenicillin. Based on the probabilistic simulation model developed, concentrations of benzylpenicillin in pork invariably remained below 1% of the maximum residue limit. Therefore, the use of this by-product as feed with the current practice was considered to pose only a negligible risk to consumers of pork products. 相似文献

BACKGROUND: The objective of this study was to determine the effects of dietary black cumin seed on performance, egg traits, egg cholesterol content and egg yolk fatty acid composition in laying hens during a 12 week period. For this purpose a total of 160 Lohmann Brown laying hens 36 weeks of age were allocated to four dietary treatments with one control group and three treatment groups. Black cumin seed (Nigella sativa L.) was used at the level of 5, 10 and 15 g kg?1 in the diets of the first, second and third treatment groups, respectively. RESULTS: Dietary treatments did not significantly affect body weight, feed intake, egg production, egg quality characteristics and blood parameters. Diets containing 10 and 15 g kg?1 black cumin seed increased egg weight (P < 0.01), improved feed efficiency (P < 0.01) and decreased egg yolk cholesterol, saturated fatty acids (% of total fatty acid methyl esters) and the ratio of saturated to unsaturated fatty acids (P < 0.05) compared to the diet of control group. CONCLUSION: Dietary black cumin seed at the level of 10 and 15 g kg?1 had beneficial effects on egg weight, feed efficiency, egg cholesterol content and egg yolk fatty acid composition. Morpholine is a common chemical used as emulsifier in the preparation of wax coatings for some fruit to help them remain fresh and protect against insects and fungal contamination. It has been reported that morpholine has acute toxic effects on rodents. In the present study, morpholine concentrations were analysed in fruits (citrus fruits, apples, strawberries and grapes) and juices (apple juice and orange juice) in order to determine dietary exposure among the Chinese population. A total of 732 fruit and juice samples were collected during 2015–2016, which covered major foods in China. Fruit and juice consumption data were taken from China National Nutrient and Health Survey (2002) and include data from 16,407 fruit or juice consumers. It was found that mean dietary exposure to morpholine residues from fruits and/or juices for general Chinese consumers and children 2–6 years old were 0.42 and 1.24 µg/kg bw/day, respectively. The 97.5% intake in general Chinese consumers and children 2–6 years old were 2.25 and 6.90 µg/kg bw/day, respectively. The primary food sources of the morpholine dietary intake of general Chinese consumers were citrus fruits (57.4%) and apples (40.8%). These findings suggested that dietary exposure to morpholine in the Chinese population was lower than the acceptable daily intake of morpholine, and there are no health concerns. 相似文献

The aim of this study was to evaluate the residues of 140 pesticides in raspberries from north-eastern Poland (2005–2010). Gas chromatography with electron capture detector (GC-ECD) and nitrogen phosphorous detector (GC-NPD) was used. Among the 128 samples, 66 (51.6%) were found to detect residues: 14.1% contained one pesticide and around 38% multiple pesticide residues. The most frequently detected were pyrimethanil residues (36.0%). Twenty-seven (21.1%) raspberry samples exceeded the maximum residue limits. The estimated daily intakes ranged from 0.003% to 3.183% of the acceptable daily intake (ADI) for adults 0.008% and 9.7% for toddlers, respectively. The most critical case is procymidone, the acute risk was 180.9% of acute reference dose (ARfD) for toddlers and for adults (83% of ARfD) which is high. 相似文献

To estimate of food and nutrient intakes, 24-h recalls are frequently used in dietary assessment. However intake data collected for a short period are a limited estimator of long-term usual intake. An important limitation of such data is that the within-person variability tends to inflate the intake distribution leading to a biased estimation of extreme percentiles. Statistical models, named usual-intake models, that separate the within-person variability from the between-persons variability, have lately been implemented. The main objectives of this study were to highlight the potential impact that usual-intake models can have on exposure estimate and risk assessment and to point out which are the key aspects to be considered in order to run these models properly and be sure to interpret the output correctly. To achieve the goal we used the consumption data obtained by the French dietary survey INCA2 and the concentration data collected during the French TDS2, using Monte Carlo Risk Assessment (MCRA) software, release 8.0. For the three substances included in this study (cadmium, acrylamide and sulphites), the exposure of the upper percentiles was significantly reduced when using usual-intake models in comparison with the results obtained in the observed individual mean models, even if in terms of risk assessment the impact of using usual-intake models was limited. From the results it appears that the key aspects to consider when using usual-intake models are: (1) the normality of the log-transformed intake distribution, (2) the contribution per single food group to the total exposure, and (3) the independency of food consumption data on multiple days. In conclusion, usual-intake models may have an impact on exposure estimates although, referring to the results, it did not bring any changes in terms of risk assessment, but further investigations are needed. 相似文献

Animals intended for human food may absorb pesticides from residues in their feed, water or during direct/indirect exposure in the course of pest control. The objective of the present investigation was to monitor organochlorine pesticide residues in poultry feed, chicken muscle and eggs at a selected poultry farm. The samples were Soxhlet extracted for 8 h in 200 mL hexane–acetone (1:1, v/v) mixture. The clean‐up of the samples was performed by silica gel column chromatography and analysis was done on a gas chromatograph equipped with an electron capture detector. The mean total hexachlorocyclohexane (HCH) and dichlorodiphenyltrichloroethane (DDT), endosulfan sulfate and heptachlor epoxide residues were 0.65, 0.91, 0.42 and 0.02 mg kg?1, respectively, in feed while respective values for chicken muscle were 0.11, 0.24, 0.10 and 0.07 mg kg?1. Higher residues were encountered in eggs as compared to muscle. None of the muscle samples exceeded maximum residue limits (MRL) for organochlorine pesticides, while all egg samples had values above the MRL for HCH and heptachlor epoxide and seven egg samples exceeded MRL for DDT residues. The results indicated that poultry feed could be one of the major sources of contamination for chicken and eggs. These residues are present despite complete ban on the use of technical HCH and DDT for agricultural purposes in India. Copyright © 2005 Society of Chemical Industry 相似文献
