目的:研究两种不同的淋巴细胞分离液对脐带血单核细胞的分离效果及后期对CIK细胞的诱导培养的影响。方法:分别使用国产TBD以及进口GE healthcare牌淋巴细胞分离液分离提取脐带血单核细胞,用悬浮细胞培养法诱导培养脐带血单核细胞形成CIK细胞,再用血球计数板及台盼兰染色法检测细胞密度及活率。结果:国产的TBD牌所分离的单核细胞分层较清楚,并且方便提取,而使用进口GE healthcare的淋巴细胞分离液提取的单核细胞数量较多,但经过后期诱导培养CIK细胞发现两种分离液提取的细胞培养到后期数量逐渐接近。结论:国产TBD牌淋巴细胞分离液在用于脐带血淋巴细胞分离提取时较进口GE healthcare牌经济,并且细胞提取方便,后期培养效果无明显差别。 相似文献

介绍了一种基于干式化学法的便携式血液检测系统.该系统以高精度数字信号处理器TM320VC5509A为控制核心,以线光斑半导体激光器为光源,以电荷耦合器(CCD)为光电转换器件,实现血细胞参数的检测.系统稳定性测试结果表明,血细胞参数的相对标准偏差均小于1.7%.与Sysmex XT-1800i血液分析仪的对比性测试实验表明,以红细胞压积(HCT)、血红蛋白(HGB)、淋巴及单核细胞(LM)、粒细胞(GRAN)、血小板(PLT)和白细胞(WBC)作为比对项目,相关系数分别为:0.991、0.972、0.952、0.986、0.979和0.985.该系统检测时间≤60 s,并具备测量记录显示、查询和删除等功能.该系统将在血液现场快速检测和急诊检查中有广泛的应用前景. 相似文献

A comparison of blood pressures measured by direct radial intra-arterial and indirect brachial automated oscillometric methods (Dinamap 1845), was performed in thirty hypertensive post-carotid endarterectomy patients. Five hundred fifty-eight "triplet" readings of systolic, diastolic, and mean blood pressure were compared. Mean differences of 18.92, -6.32, and -0.26 mm Hg, and correlation coefficients of 0.831, 0.724, and 0.776 were calculated for systolic, diastolic, and mean blood pressures, respectively. Mean differences were then calculated within incremental direct systolic pressure ranges of 20 mm Hg and found to become significantly larger with each increasing increment of pressure. These differences ranged from -0.8 to 12.6 and 20.0 to 53.3 mm Hg in the less than 160 and greater than 160 mm Hg groups, respectively, with direct intra-arterial systolic pressures significantly greater than indirect oscillometric systolic pressures in the latter group. The results demonstrate that in the hypertensive pressure range, a significant difference exists in systolic pressure measured by intra-arterial and automated oscillometric methods. 相似文献

The umbilical cord blood (UCB) is an important source of hematopoietic stem cells with great deal of interest in regenerative medicine. The UCB cells have been extensively studied as an alternative to the bone marrow transplants. The challenge is to define specific methods to purify and characterize these cells in different animal species. This study is aimed at morphological characterization of progenitor cells derived from UCB highlighting relevant differences with peripheral blood of adult in dog and cats. Therefore, blood was collected from 18 dogs and 5 cats' umbilical cords from fetus in various developmental stages. The mononuclear cells were separated using the gradient of density Histopaque-1077. Characterization of CD34+ cells was performed by flow cytometric analysis and transmission electron microscopy. Granulocytes (ancestry of the basophiles, eosinophiles, and neutrophiles) and agranulocytes (represented by immature lymphocytes) were identified. We showed for the first time the ultrastructural features of cat UCB cells. 相似文献

目的:通过本科室两种生化检测系统(BDS)对10项常规生化项目的检验结果进行比对分析和偏倚评估,探讨不同BDS对同一检测项目的结果是否具有可比性和偏倚可接受性。方法:根据美国临床实验室标准协会EP9-A2文件标准要求,每天随机选取8例新鲜血清样本在贝克曼AU2700[为参比系统]和XC8001[为试验系统]全自动生化分析仪上测定10项(谷丙转氨酶、谷草转氨酶、白蛋白、总蛋白、肌酐、尿素、总胆固醇、甘油三酯、高密度脂蛋白胆固醇和低密度脂蛋白胆固醇)常规生化项目,每例样本按正反序重复检测两次,连续测量5天,记录各测定值。计算Y与X之间的相关系数,直线回归方程和不同医学决定水平处的相对偏差。并以美国临床实验室改进修正案能力验证规则(CLIA,88)分析质量要求中规定可接受范围的1/2作为标准,判断两BDS之间的可比性和偏倚临床可接受性。结果:未发现有离群点出现,10个常规生化检测项目的r值均≥0.975,满足EP9-A2文件规定r≥0.975(或R2≥0.95)的要求。所检测数据经单因素方差分析,差异无统计学意义(P>0.05)。10个常规生化检测项目不同Xc的相对偏差SE%,结果均<可接受范围的1/2。结论:本院两种BDS通过方法学比对,对同一检测项目测定结果具有一致性,且各检测项目在各医学决定水平处出现偏倚临床是接受可接受性。 相似文献

目的:探讨血液净化模式治疗终末期肾病的应用效果,并分析其优势特点。方法:选择我院2019年10月至2020年12月终末期肾病患者42例,对照组21例采用常规的血液透析治疗,观察组21例采用高通量血液透析(HFHD)的方法,比较两组的肾功能指标、炎症以及心脏功能指标变化情况。结果:相比对照组,观察组的肾功能指标尿素氮(BUN),血清肌酐(Cr)清除率明显提高,炎症指标IL-6、IL-18、TNF-α表达水平显著降低,心脏功能的四项指标均显著改善(P<0.05),表明治疗效果好。结论:HFHD模式可显著改善患者的肾功能和炎症反应,同时能够减轻心脏负荷,可作为临床治疗终末期肾病的途径之一。 相似文献

建立了液相色谱-串联质谱法同时定量检测干血斑中4种临床常用免疫抑制剂的方法。打孔裁取直径8mm的干血斑样本,经甲醇-乙腈混合溶液提取和沉淀蛋白后取上清液氮气吹干,甲醇溶液复溶后进样分析。选用C18色谱柱分离,甲醇与水(均含2mM乙酸铵和0.1%甲酸)为流动相梯度洗脱,在电喷雾正离子电离(ESI+)和多反应监测(MRM)模式下检测。结果表明:环孢菌素A在20~1000ng/mL范围内,西罗莫司、他克莫司和依维莫司在2.0~100ng/mL范围内线性良好。该方法对病患友善(样本采血量仅30μL),提取回收率高(73.6%~110.8%),其准确度、精密度(CV)、残留效应和基质效应均符合生物样品的分析要求。该方法快速、高效、灵敏度高、精确度好,适用于干血斑中同时测定环孢菌素A、西罗莫司、他克莫司和依维莫司浓度。 相似文献

Cellular behavior is influenced by micro-concentration gradients existing in their immediate spatio-environment. A practical real-time measurement system, composed of miniaturized long-lasting sensors placed within a two-dimensional bioreactor and a 32-channel data acquisition system for ammonia, is presented in this paper aiming to generate the spatio-temporal ammonia concentration profiles for the whole culture process. Our results demonstrate the feasibility of the system and its applicability in elucidating the cell growth micro-environment within a bioreactor, which can potentially facilitate the intelligent control of ammonia levels in cell culture systems. The presented system is flexible enough to interface other sensors for generating their spatio-temporal profiles of the sensed cell culture parameters. 相似文献

水质参数监测是工厂化水产养殖的重要组成部分,是工厂化养殖向自动化、大规模、高产量和高质量发展的关键环节。针对我国工厂化养殖水质监测手段落后、自动化水平低的现状,研制了一套适合国情的在线多参数水质监测系统,可对水温、水位、酸碱度(PH值)、溶解氧含量4个水质参数进行连续在线监测。以一台PC工业控制计算机作为现场上位机,以多台单片机作为现场鱼池的下位机,一旦发生故障,养殖池发生环境变化,能够及时报警,从而实现了对养殖现场中鱼类生长的实时监控。实际应用结果表明,系统运行稳定、安全可靠,测量数据的精确度完全可以达到工厂化渔业生产的要求。 相似文献

This work aimed to study the beneficial role of human umbilical cord blood-derived mesenchymal stem cellconditioned medium (MSC-CM) in hypoxia-induced apoptosis in H9c2 cardiomyoblasts, in which the serine/heroinekinases (Akt) pathway would be involved. For this, CM was collected by culturing MSCs in serum-free DMEMmedium for 24 h, and paracrine factors were analyzed by protein chip. H9c2 cells were divided into the followinggroups: control group, hypoxia group, MSC-CM intervention group (CM group), MSC-CM + Akt phosphorylationinhibitor (LY294002) group (LY group). Apoptosis of the H9c2 cells was tested with chromatin dye Hoechst 33342and FITC-conjugated Annexin V apoptosis detection kit by flow cytometer after a hypoxia/serum deprivation (H/SD)for 24 h. The apoptosis-related proteins were evaluated by Western blot. MSC-CM displayed significantly elevatedlevels of growth factors, anti-inflammatory, and anti-apoptosis cytokines. On Hoechst 33342 apoptosis staining, theH9c2 cell morphology displayed a lower proportion of apoptosis in the CM group than those in the hypoxia group,while apoptosis was increased in LY group. Flow cytometer analysis revealed the apoptosis ratio in the CM group waslower than the hypoxia group (12.34 ± 2.00% vs. 21.73 ± 2.58%; p < 0.05), while the LY group was significantly higher(22.54 ± 3.89%). Active caspase-3 expression was increased in hypoxia group than control group (p < 0.05), butdecreased in CM group (p < 0.01). Umbilical cord blood-derived mesenchymal stem cell-conditioned media secretemultiple paracrine factors that are able to inhibit hypoxia-induced H9c2 cardiomyoblasts apoptosis, and in which theactivation of Akt phosphorylation is involved to achieve the protective effect. 相似文献
