This study was carried out to determine the changes of volatile compounds composition during palm sugar production. The samples were collected at every 30 min interval for 4 h of heating process at 150 °C from a local traditional manufacturer in Kuala Pilah, Malaysia. The analyses were performed by gas chromatography–mass spectrometry after headspace solid phase micro extraction. The results showed that N-heterocyclic chemical class possessed the highest relative percentage area (RPA) 83.69%, followed by O-heterocyclic group with RPA of 14.5%. Main volatile compounds the determined were 5-methyl-6,7-dihydro-5H-cyclopenta pyrazine and 4-hydroxy-2,5-dimethyl-3(2H) furanone which were responsible for roasty and sweet caramel-like aroma notes, respectively. The pyrazine compounds increase exponentially with heating time but furan derivatives compounds were formed at a later stage, i.e. at 180 min of heating time and above 105 °C.  相似文献

Volatile organic acids in freeze-dried Cheonggukjang were quantified using a stable-isotope dilution assay (SIDA) according to the fermentation period. Five organic acids, acetic acid, propanoic acid, 2-methylpropanoic acid, butanoic acid, and 3-methylbutanoic acid, were identified using solid-phase microextraction (SPME) in conjunction with gas chromatography–mass spectrometry (GC–MS). The contents of volatile organic acids in Cheonggukjang were highly dependent on the fermentation period and they increased during fermentation. Moreover, the branched-chained organic acids (namely 2-methylpropanoic acid and 3-methylbutanoic acid) were formed earlier and were present at much higher contents than the corresponding straight-chained organic acids during Cheonggukjang fermentation.  相似文献

This study showed the relationship between tea leaf age, bud and first two leaves, and shade levels, on the relative concentrations of six major compounds of tea leaf, namely l-theanine, caffeine, and the major tea catechins; (−)-epigallocatechin gallate (EGCG), (−)-epigallocatechin (EGC), (−)-epicatechin (EC), and (−)-epicatechin gallate (ECG), all of which are reported to have positive effects on human health, as well as at the ferric reducing antioxidant power of bud and leaf extracts. The concentration of l-theanine and caffeine decreased as leaf age increased moving from bud to first and then second leaf, while the concentration of the four catechins increased from the bud to first and second leaves. In most cases this increase was generally relatively small but in the case of EGC it was 7 to 10-fold. Certain chemical components of freshly picked, minimally processed and essentially unoxidised tea may potentially be used as markers for age, quality, authenticity and area of growth.  相似文献

While beneficial health properties of tea leaves have been extensively studied, less attention has been given to that of flowers of the tea (Camellia sinensis) plant. In this work, the ethanolic extract and its ethyl acetate-soluble fraction (EEA) from the tea flowers were found to possess the potent antioxidant activity using 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging assay. The compounds present in EEA had comparatively strong DPPH scavenging activity and strongly contributed to the antioxidant activity of the tea flowers. From EEA, besides eight catechins, five flavonol glycosides were isolated and their structures were elucidated on the basis of mass spectrometry and nuclear magnetic resonance spectroscopy as myricetin 3-O-β-d-galactopyranoside, quercetin 3-O-β-d-galactopyranoside, kaempferol 3-O-β-d-galactopyranoside, kaempferol 3-O-β-d-glucopyranoside, and kaempferol 3-O-[α-l-rhamnopyranosyl-(1-6)-β-d-glucopyranoside]. In addition, epigallocatechin gallate and epicatechin gallate were found as the major active components responsible for the antioxidant activity of tea flowers through the use of a combination of preparative liquid chromatography separation and DPPH assay.  相似文献

The reputation of particular countries, and plantations within those countries, for the production of high quality tea means that these producers can ask a significantly higher price for their specific product than average. This leads to a temptation for unscrupulous producers to fraudulently label their product as coming from one of these areas to take advantage of this higher price. A simple method for the analysis and verification of the country and plantation of origin of tea would provide security to both tea growers and consumers and would largely stamp out this illegal activity. This paper details the use of complimentary organic and inorganic isotope techniques for the analysis of tea samples from Asia. The application of linear discriminant analysis of the isotope ratios and mineral concentrations permitted 97.6% correct classification of the tea samples using the following variables δD, δ13C, 49Ti, 53Cr, 59Co, 60Ni, 65Cu, 71Ga, 85Rb, 88Sr, 89Y, 93Nb, 111Cd, 133Cs, 138Ba, 139La, 140Ce, 141Pr, 153Eu, 203Tl, 208Pb and 209Bi.  相似文献

Wheat contributes significantly worldwide to antioxidant income with beneficial healthy effects. Two varieties of einkorn wheat (Triticum monococcum L. – Escana and Schwedisches Einkorn), two varieties of wild emmer [Triticum dicoccum Schuebl (Schrank)] (Rudico and Kahler Emmer) and three varieties of spring wheat (Triticum aestivum L. – Granny, Kärtner Früher and SW Kadrilj) were evaluated for tocols – tocopherols and tocotrienols by high performance liquid chromatography – electrospray ionisation tandem mass spectrometry (HPLC–ESI/MS/MS). Investigated emmer and einkorn wheat varieties have shown minor lower total tocols content in comparison with spring varieties, however significant typical levels of tocopherols and tocotrienols were found in these groups. In einkorn wheat varieties the major abundance of total tocols is represented by tocotrienols (Schwedisches Einkorn 65.0%, Escana 54.9%), in emmer wheat varieties the abundance of tocotrienols and tocopherols were similar (Rudico 48.9% of tocotrienols, Kahler Emmer 47.6%), whereas in spring bread wheat higher tocopherols (SW Kadrilj 54.5%, Kärtner Früher 60.2%, Granny 55.0%) were recorded.  相似文献

An acidic peroxidase isoform, POD-A, with a molecular mass of 69.4 kDa and an isoelectric point of 3.5 was purified from papaya latex. Using o-phenylenediamine (OPD) as a hydrogen donor (citrate–phosphate as pH buffer), the optimum pH for the function of POD-A was 4.6, and the optimum temperature was 50 °C. The peroxidase activity of POD-A toward hydrogen donors was both pH- and concentration-dependent. Under optimal conditions, POD-A catalysed the oxidation of OPD at higher rates than pyrogallol, catechol, quercetin and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS). The chemical modification reagents N-bromosuccinimide and sodium azide significantly inhibited POD-A activity. The results of kinetic studies indicated that POD-A followed a ping-pong mechanism and had a Km value of 2.8 mM for OPD. Using CPC silica-immobilised POD-A for the determination of micromolar H2O2 in milk, the lower limit of determination was 0.1 μM, and the recoveries of added H2O2 were 96–109%.  相似文献

Post-mortem softening of fish tissue often results in low yield and decreased product quality. In this study, proteolytic profiles of trout stored 5 days on ice were obtained by SDS–PAGE. The link between protein band intensities and firmness of trout fillets was examined through a correlation study. In parallel, trout extracts were incubated with cathepsin B, cathepsin L and cathepsin D, alone or in combination, in order to evaluate the effect of each cathepsin on the texture-related proteins. Proteins from both myofibrillar (α-actinin, actin, MLC1, MLC2, and N-terminal 70 kDa MHC fragment) and sarcoplasmic (glycogen phosphorylase, creatine kinase, and TPI) fractions correlated closely with firmness. Cathepsins D, B and L affected, respectively, 10, 9 and 4 out of the 17 protein bands correlating with firmness, and most changes induced by cathepsin D were unfavourable to firmness. This implies that cathepsin D is likely to be involved in textural change of trout, due to breakdown of the muscle structure.  相似文献

Artichoke (Cynara scolymus L.) is an edible vegetable largely used in the Mediterranean diet and in folk medicine. The present paper discusses the analysis of the polyphenol content of leaves and outer bracts of Violetto di Toscana artichoke using different extraction procedures with the aim of establishing a correlation between polyphenol subclasses and antioxidant activity measured on human LDL oxidized by copper ions. HPLC/DAD and HPLC/MS analyses revealed that both the matrixes contain identical polyphenol subclasses, with mainly quantitative differences. The antioxidant effect of four artichoke extracts decreases in the following order when the sum of total phenolic compounds was considered: ethanolic extract from leaves (IC50 = 2.92 ± 0.46 μM); ethanolic extract from outer bracts (IC50 = 4.04 ± 0.21 μM); ethyl acetate extract from leaves (IC50 = 4.91 ± 0.11 μM); ethyl acetate extract from outer bracts (IC50 = 10.18 ± 1.6 μM). IC50 were also calculated considering the concentrations of single polyphenol subclasses. In both cases, the potency of antioxidant properties was not related to the amount of total polyphenols or the single subclasses.  相似文献

We investigated the metabolic profile and biological activities of the essential oil and polar extracts of Lavandula pedunculata subsp. lusitanica (Chaytor) Franco collected in south Portugal. Gas chromatography–mass spectrometry (GC–MS) analysis revealed that oxygen-containing monoterpenes was the principal group of compounds identified in the essential oil. Camphor (40.6%) and fenchone (38.0%) were found as the major constituents. High-performance liquid chromatography with diode array detection (HPLC–DAD) analysis allowed the identification of hydroxycinnamic acids (3-O-caffeoylquinic, 4-O-caffeoylquinic, 5-O-caffeoylquinic and rosmarinic acids) and flavones (luteolin and apigenin) in the polar extracts, with rosmarinic acid being the main compound in most of them. The bioactive compounds from L. pedunculata polar extracts were the most efficient free-radical scavengers, Fe2+ chelators and inhibitors of malondialdehyde production, while the essential oil was the most active against acetylcholinesterase. Our results reveal that the subspecies of L. pedunculata studied is a potential source of active metabolites with a positive effect on human health.  相似文献

Polyphenol concentrations, including anthocyanidins, flavonols, flavan-3-ols and stilbenes, were quantified by liquid chromatography/mass spectrometry in two cultivars of red grapes for daily consumption, which were subjected to different kinds of water supply and nitrogen fertilisation rates. Samples from the same vineyards were also analysed after a 6 week storage in a refrigerator and 6 week delayed harvesting. Berry skins and seeds were analysed separately.  相似文献

Lycium barbarum has been used as a traditional Chinese medicine (TCM) to nourish liver and kidney, and brighten the eye. The fruits are dried or freshly squeezed for their juice and concentrated for beverages. Among various constituents, a group of polysaccharides (LBP) with a Glycan-O-Ser glycopeptide structure has been most researched and considered to be important for the efficacy of L. barbarum. Studies indicate effects of L. barbarum on aging, neuroprotection, general well-being, fatigue/endurance, metabolism/energy expenditure, glucose control in diabetics, glaucoma, anti-oxidant properties, immunomodulation, anti-tumor activity and cytoprotection. In addition to TCM, L. barbarum can be sold as a dietary supplement or classified as a food based upon the long and safe traditional usage. This review is to provide background and updated information of chemical constituents and efficacies with safety including a new direction for research and current regulatory situation of L. barbarum.  相似文献
