The aim of the project was to calculate the apparent synthesis or destruction of cobalamin (vitamin B12) and its analogs in the rumen as well as their apparent intestinal disappearance in dairy cows. Four lactating cows were fed a diet supplemented with cobalt alone (0.76 mg/kg of DM; control) or with cobalt and vitamin B12 (cyanocobalamin, 500 mg/d; treated). In addition to cobalamin, the only biologically active molecule for the cow, 7 analogs were identified in duodenal and ileal digesta: cobinamide, which lacks the base, ribose, and phosphate groups; and 6 other molecules in which the base, 5,6-dimethylbenzimidazole, is replaced by cresol, 2-CH3-adenine, adenine, 2-CH3-S-adenine, or 5-OH-benzimidazole, or an unidentified cobamine. Small amounts of cobalamin and cobinamide were detected in the total mixed ration, but apparent synthesis of all forms took place in rumen. During the control period, cobalamin represented 38% of the total amounts of corrinoids produced in rumen. Approximately 11% of the average daily intake of cobalt was used for apparent ruminal synthesis of corrinoids, of which only 4% was incorporated into cobalamin. Only 20% of the supplement of cyanocobalamin was recovered at the duodenal level; cobinamide appeared to be the major product of degradation of supplementary cyanocobalamin in the rumen. During the control and treatment periods, there was an apparent intestinal disappearance of cobalamin and 5-OH-benzimidazole cobamide only; only the apparent intestinal disappearance of cobalamin differed between the 2 periods. Although cobalamin was not the major form synthesized by ruminal microflora and, even if supplementary cyanocobalamin was extensively destroyed by ruminal microflora, based on calculations of apparent intestinal disappearance, cobalamin seems to be the major form absorbed in the small intestine.  相似文献

There is little information on the vitamin D content of Australian red meat or on the possible influence of latitude on this content. To determine the content of vitamin D3 and 25-hydroxy-vitamin D3 (25OHD3), lamb and beef were analysed from 34° S with LC–IT-MS. To investigate the possible influence of latitude on vitamin D in meat, the lean meat and fat from five cuts of beef were analysed from 17° S and 41° S. Lamb contained 0.10 μg vitamin D3/100 g and 0.20 μg 25OHD3/100 g lean meat, while beef contained 0.12 μg vitamin D3 and 0.27 μg 25OHD3/100 g (lean meat). Latitude had no effect on the vitamin D3 (P = 0.21) or 25OHD3 (P = 0.29) content of lean beef, but fat from cattle in the 17° S latitude group contained significantly higher (P < 0.01) concentrations of vitamin D3 than fat from the 41° S group of cattle.  相似文献

The present experiment was undertaken to study the interactions between dietary supplements of rumen-protected methionine (RPM) and intramuscular injections of folic acid and vitamin B12, given from 3 wk before calving to 16 wk of lactation, on hepatic metabolism of lactating dairy cows. Sixty multiparous Holstein cows were assigned to 10 blocks of 6 cows each according to their previous milk production. Within each block, 3 cows were fed a diet calculated to supply Met as 1.83% of metabolizable protein, whereas the 3 other cows were fed the same diet supplemented with 18 g of RPM calculated to provide Met as 2.23% of metabolizable protein. Within each level of Met, the cows received no vitamin supplement or weekly intramuscular injections of 160 mg of folic acid alone or combined with 10 mg of vitamin B12. Liver biopsies were taken at 2, 4, 8, and 16 wk of lactation. Liver concentrations of folates and vitamin B12 were increased by their respective supplements but this response to vitamin supplements was altered by methionine supply. Concentrations of total lipids and triglycerides increased in livers of cows fed RPM, whereas concentrations of cholesterol ester, cholesterol, diglycerides, phosphatidylethanolamine, and phosphatidylcholine were not affected. Folic acid, alone or combined with vitamin B12, tended to increase the ratio of phosphatidylcholine to phosphatidylethanolamine. Gene expression of 5,10-methylene-tetrahydrofolate reductase, microsomal transfer protein, and phosphatidylethanolamine methyltransferase were higher in liver of cows fed RPM supplements. The relative mRNA abundance of 5,10-methylene-tetrahydrofolate reductase and methylmalonyl-CoA mutase were increased by the combined injections of folic acid and vitamin B12, whereas those of methionine synthase and methionine synthase reductase were not affected by treatments. These results suggest that increasing supply of methyl groups, as preformed labile methyl groups or through methylneogenesis, affected the methylation cycle but had a limited effect on dairy cow performance. The observed effects of the combined supplement of folic acid and vitamin B12 on lactational performance of dairy cows probably result from an improvement of energy metabolism during early lactation.  相似文献

The present experiment was undertaken to determine if the effects of supplementary folic acid on lactational performance were caused by improved methylneogenesis and if the supply in vitamin B12 could affect this metabolic pathway. In this eventuality, supplementary Met, a major source of preformed methyl groups, should reduce the requirements for these vitamins. Sixty multiparous Holstein cows were assigned to 10 blocks of 6 cows each according to their previous milk production. Within each block, 3 cows were fed a diet estimated to supply Met as 1.83% metabolizable protein and 3 cows were fed the same diet supplemented with 18 g of rumen-protected methionine (RPM) to supply Met as 2.23% of metabolizable protein. Within each level of Met, cows received no vitamin supplement or weekly intramuscular injections of 160 mg of folic acid alone or combined with 10 mg of vitamin B12 from 3 wk before to 16 wk after calving. There was no treatment effect on dry matter intake during pre- and postcalving periods: 13.4 ± 0.4 and 21.8 ± 0.4 kg/d, respectively. Milk production was not affected by RPM supplementation. Folic acid and vitamin B12 given together tended to increase milk production during the 16 wk of lactation. This effect was more pronounced during the first 4 wk of lactation: 37.5, 37.7, and 40.3 ± 0.9 kg/d for cows receiving no vitamin supplement, folic acid alone, or folic acid combined with vitamin B12, respectively. Milk fat yield was not affected by treatments. Lactose, crude protein, and total solid yields were greater, in early lactation, in cows injected with folic acid and vitamin B12 together but this effect diminished as lactation progressed. Intramuscular injections of folic acid alone or combined with vitamin B12 tended to decrease plasma concentrations of homocysteine from 5.51 μM with no vitamin supplement to 4.54 and 4.77 ± 0.37 μM, respectively. Results of the present experiment suggest that the effects of the combined supplement of folic acid and vitamin B12 on lactational performance of dairy cows were not due to an improvement in methyl groups supply, because RPM supplement, a source of preformed methyl groups, did not alter the cow responsiveness to vitamin supplements.  相似文献

Biotin and vitamin B12 are coenzymes in reactions that are essential to propionate metabolism in dairy cows. The objective of the present studies was to determine whether an increased dietary supply of these vitamins would change the net flux of nutrients through the rumen, the portal-drained viscera (PDV), the total splanchnic tissues (TSP), and the liver. Four lactating cows equipped with ultrasonic flow probes around the right ruminal artery and the portal vein and catheters in the right ruminal vein, the portal vein, one hepatic vein, and one mesenteric artery were fed 12 times per day a mixed ration at 95% of ad libitum dry matter intake. Daily supplements of 500 mg of vitamin B12 + 20 mg of biotin or no vitamin supplement (study 1) or 500 mg of vitamin B12 alone or with 20 mg of biotin (study 2) were fed according to a crossover design with two 4-wk periods in each study. On the last day of each period, blood flow was recorded and blood samples were collected every 30 min for 4 h. In study 1, biotin and vitamin B12 given together increased milk production and milk protein yields compared with the control diet. The supplement increased appearance of the 2 vitamins across the PDV and TSP. It also reduced the net portal appearance of ammonia and total volatile fatty acids across the PDV. In study 2, compared with the 2 vitamins together, vitamin B12 alone increased glucose flux across PDV and TSP as well as its arterial concentration and PDV flux of ammonia. With the diet used in the present experiment, the major effects of the vitamin supplements seem to be mediated through changes in ruminal fermentation and gastrointestinal tract metabolism rather than by effects on hepatic metabolism.  相似文献

Twenty-four multiparous Holstein cows were assigned to 6 blocks of 4 cows each according to their previous milk production. Within each block, 2 cows were fed a diet estimated to supply methionine as 1.83% metabolizable protein, equivalent to 76% of methionine requirement, whereas the 2 other cows were fed the same diet supplemented daily with 18 g of rumen-protected methionine. Within each diet, the cows were administrated either no vitamin supplement or weekly intramuscular injections of 160 mg of folic acid plus 10 mg of vitamin B12. To investigate metabolic changes at 12 wk of lactation, glucose and methionine kinetics were measured by isotope dilution using infusions of 3[U-13C]glucose, [13C]NaHCO3 and 3[1-13C,2H3] methionine. Milk and plasma concentrations of folic acid and vitamin B12 increased with vitamin injections. Supplementary B-vitamins increased milk production from 34.7 to 38.9 ± 1.0 kg/d and increased milk lactose, protein, and total solids yields. Whole-body glucose flux tended to increase with vitamin supplementation with a similar quantitative magnitude as the milk lactose yield increase. Vitamin supplementation increased methionine utilization for protein synthesis through increased protein turnover when methionine was deficient and through decreased methionine oxidation when rumen-protected methionine was fed. Vitamin supplementation decreased plasma concentrations of homocysteine independently of rumen-protected methionine feeding, although no effect of vitamin supplementation was measured on methionine remethylation, but this could be due to the limitation of the technique used. Therefore, the effects of these B-vitamins on lactation performance were not mainly explained by methionine economy because of a more efficient methylneogenesis but were rather related to increased glucose availability and changes in methionine metabolism.  相似文献

Stability of vitamin D2 in milk was determined in vitamin D2 fortified milk. Reverse phase high pressure liquid chromatography was used to determine the vitamin D2 loss during processing, packaging and under light. The percentage losses during pasteurization, boiling and sterilization were demonstrated to be statistically insignificant. Milk was stored for seven days in both glass and plastic bottles under refrigerated temperature, non significant loss of vitamin D2 was observed, whereas, when stored in polyethylene pouches significant loss was observed as vitamin D2 decreased from 596.66 to 548.04 IU. This clearly indicated that vitamin D2 was sorbed up by polyethylene material during storage resulting in its loss. Milk samples were stored for 32 h under three different light intensities (14,852,970 and 4455 lux). Non significant loss of vitamin D2 was observed in glass packaging, whereas significant loss was observed in polyethylene pouches. In milk fortified with both calcium and vitamin D2, non significant effect of calcium was observed on the loss of vitamin D2.  相似文献

Fresh Shiitake mushrooms (Lentinula edodes), Oyster mushrooms (Pleurotus ostreatus), Button mushrooms (Agaricus bisporus), and Abalone mushrooms (Pleurotus cystidus) were irradiated with Ultraviolet-A (UV-A; wavelength 315–400 nm), Ultraviolet-B (UV-B; wavelength 290–315 nm), and Ultraviolet-C (UV-C; wavelength 190–290 nm). Irradiation of each side of the mushrooms for 1 h, was found to be the optimum period of irradiation in this conversion. The conversions of ergosterol to vitamin D2 under UV-A, UV-B, and UV-C were shown to be significantly different (p < 0.01). The highest vitamin D2 content (184 ± 5.71 μg/g DM) was observed in Oyster mushrooms irradiated with UV-B at 35 °C and around 80% moisture. On the other hand, under the same conditions of irradiation, the lowest vitamin D2 content (22.9 ± 2.68 μg/g DM) was observed in Button mushrooms.  相似文献

An experiment was conducted to determine the efficacy of 3 adsorbents, Solis (SO; Novus International Inc.), NovasilPlus (NOV; Engelhard Corp.), and MTB-100 (MTB; Alltech), in reducing aflatoxin (AF) M1 concentrations in milk of dairy cows fed an AF-contaminated diet. Twelve early to mid lactation dairy cows averaging 163 d in milk were used in a 4 × 4 Latin square design with 3 replications. Cows were blocked by parity, body weight, and milk production and were provided ad libitum access to feed and water. Within each replicate, cows were randomly assigned to the 4 dietary treatments for 4 consecutive 7-d periods. Dietary treatments included AF [112 μg of AFB1/kg of diet dry matter (DM)]; AF + 0.56% SO; AF + 0.56% NOV; and AF + 0.56% MTB. Milk samples were collected on d 6 and 7 of each of the experimental periods. Feed intake, milk production, milk fat percentage, milk protein percentage, and linear somatic cell scores were not affected by dietary treatments and averaged 22.20 kg/d of DM, 33.87 kg/d, 3.78%, 2.95%, and 1.60, respectively, across all treatments. Transfer rates of AF from feed to milk averaged 2.65, 1.48, 1.42, and 2.52% for cows fed AF, AF + SO, AF + NOV, and AF + MTB, respectively. Daily AFM1 excretion in milk averaged 66, 37, 35, and 63 μg/d for cows fed AF, AF + SO, AF + NOV, and AF + MTB, respectively. The addition of SO and NOV to the AF diet resulted in a significant reduction in milk AFM1 concentrations (SO, 45%; NOV, 48%) and AFM1 excretion (SO, 44%; NOV, 46%). In contrast, MTB was not effective in reducing milk AFM1 concentrations (4%), AFM1 excretion (5%), or AF transfer from feed to milk (2.52%). Results indicated that SO and NOV at 0.56% of the diet were effective in reducing milk AFM1 concentrations in cows consuming a total mixed ration containing 112 μg of AFB1/kg of diet DM.  相似文献

This study was undertaken to determine the effect of a combined folic acid and vitamin B12 supplement given in early lactation on culling rate, metabolic disorders and other diseases, and reproduction in commercial dairy herds. A total of 805 cows (271 primiparous and 534 multiparous cows) in 15 commercial dairy herds were involved. Every 2 mo from February to December 2010 and within each herd, cows were assigned according to parity, previous 305-d milk production, and calving interval to 5 mL of either (1) saline 0.9% NaCl (control group) or (2) 320 mg of folic acid + 10 mg of vitamin B12 (vitamin group). Treatments were administered weekly by intramuscular injections starting 3 wk before the expected calving date until 8 wk after parturition. A total of 221 cows were culled before the next dry period. Culling rate was not affected by treatment and was 27.5%; culling rate was greater for multiparous (32.2%) than for primiparous cows (18.8%). Within the first 60 d in milk (DIM), 47 cows were culled, representing 21.3% of total culling, and no treatment effect was noted. Ketosis incidence based on a threshold ≥100 µmol/L of β-hydroxybutyrate in milk was 38.3 ± 2.9% for the vitamin group and 41.8 ± 3.0% for the control group and was not affected by treatment. The combined supplement of folic acid and vitamin B12 did not decrease incidence of retained placenta, displaced abomasum, milk fever, metritis, or mastitis. However, the incidence of dystocia decreased by 50% in multiparous cows receiving the vitamin supplement, although no effect was observed in primiparous cows. The first breeding postpartum for multiparous cows occurred 3.8 d earlier with the vitamin supplement compared with controls, whereas no treatment effect was seen for primiparous cows. Days open, first- and second-breeding conception rates, number of breedings per conception, and percentage of cows pregnant at 150 DIM were not affected by treatment. The reduced percentage of dystocia combined with the earlier DIM at first breeding for multiparous cows receiving the combined supplementation in folic acid and vitamin B12 indicates that the vitamin supplement had a positive effect in older cows.  相似文献

Cortisol has long been used as a marker of the stress response in animals. Cortisol can be analyzed from different media, most notably from the blood, saliva, and feces; however, the collection of cortisol from some of these media requires invasive procedures or excessive handling of the animals. Furthermore, it is not possible to capture long-term increases in circulating concentrations of cortisol from the blood, saliva, or feces. Hair cortisol has been found to be a reliable alternative for measuring chronic stress. With this emerging measure, appropriate sampling methodology must be developed and validated. The aim of this study was to determine the effects of hair color, sampling location, and processing method on cortisol concentrations in hair from lactating black and white Holstein cows (n = 18). Furthermore, we aimed to measure the hair growth rates at different body locations (n = 12) and test hair cortisol levels when resampled over short intervals (n = 37). Both black- and white-colored hair was collected from the shoulder, top line, hip, and tail switch of Holsteins; due to breed characteristics only white hair was harvested from the tail switch. All samples were cleaned with water and isopropanol, and then ground in a ball mill or finely cut with scissors once dry. Cortisol was extracted with methanol before being measured using a commercially available ELISA kit. Concentrations of cortisol were greater in white than in black hair (7.8 ± 1.1 vs. 3.8 ± 1.1 pg/mg). When only white samples were analyzed, hair from the tail switch had more cortisol than hair from the shoulder (11.0 ± 1.2 vs. 6.2 ± 1.2 pg/mg), whereas no difference was found when compared with the hip and top line. Samples ground with a ball mill had greater concentrations of cortisol extracted than those minced with scissors (10.4 ± 1.2 vs. 4.7 ± 1.2 pg/mg). The growth rate of hair was significantly greater at the tail switch compared with the hip and shoulder (0.51 ± 0.05 vs. 0.04 ± 0.05 vs. 0.03 ± 0.05 mm/d). When hair was collected every 3 wk after calving, a tendency was detected for multiparous cows to have greater concentrations of hair cortisol and significantly greater concentrations of cortisol on d 0 and 21 after calving compared with d 42, 84, and 126. In Holsteins, the hair on the tail switch is always white, grows more rapidly than other sites, and is sensitive enough to capture changes in cortisol over intervals as short as 3 wk, making it the ideal location for measuring hair cortisol.  相似文献

Management of dairy cow productivity requires monitoring of their nutritional status by visual observation. It has been suggested that changes in hair coat appearance are among the indicators of nutritional state in dairy cows. Temporal changes in the skin morphology in cows, however, have not been reported. In this study, we examined the changes in the skin of dairy cows that occur during the peripartum period. Seven pluriparous cows were used. Skin samples were collected at 28 d before the due date and 28 d and 56 d after calving for morphological examination. Hair follicle width was 108.8 ± 5.9 µm (±SD) in the dry period, 95.5 ± 5.5 µm at 28 d after calving, and 104.2 ± 5.3 µm at 56 d postpartum. The percentages of anagen hair follicles during these 3 periods were 41.4 ± 3.4, 18.5 ± 3.4, and 32.3 ± 3.3%, respectively. The corresponding sebaceous gland sizes were 8,362.0 ± 707.6, 7,800.0 ± 831.4, and 9,186.8 ± 962.6 µm2, respectively. Hair follicle width was positively correlated with percentage of anagen hair follicles. The thickness of epidermal and proliferation rate of epidermal cell were also correlated. However, the hair follicle width, sebaceous gland size and cell proliferation rate, and thickness and proliferation rate of epidermal cells did not show any marked changes.  相似文献

The present experiment was undertaken to determine the effects of dietary supplements of folic acid and vitamin B12 given from 3 wk before to 8 wk after calving on lactational performance and metabolism of 24 multiparous Holstein cows assigned to 6 blocks of 4 cows each according to their previous milk production. Supplementary folic acid at 0 or 2.6 g/d and vitamin B12 at 0 or 0.5 g/d were used in a 2 × 2 factorial arrangement. Supplementary folic acid increased milk production from 38.0 ± 0.9 to 41.4 ± 1.0 kg/d and milk crude protein yield from 1.17 ± 0.02 to 1.25 ± 0.03 kg/d. It also increased plasma Gly, Ser, Thr, and total sulfur AA, decreased Asp, and tended to increase plasma Met. Supplementary B12 decreased milk urea N, plasma Ile, and Leu and tended to decrease Val but increased homocysteine, Cys, and total sulfur AA. Liver concentration of phospholipids was higher in cows fed supplementary B12. Plasma and liver concentrations of folates and B12 were increased by their respective supplements, but the increase in plasma folates and plasma and liver B12 was smaller for cows fed the 2 vitamins together. In cows fed folic acid supplements, supplementary B12 increased plasma glucose and alanine, tended to decrease plasma biotin, and decreased Km of the methylmalonyl-coenzyme A mutase in hepatic tissues following addition of deoxyadenosylcobalamin, whereas it had no effect when cows were not fed folic acid supplements. There was no treatment effect on plasma nonesterified fatty acids as well as specific activity and gene expression of Met synthase and methylmalonyl-coenzyme A mutase in the liver. Ingestion of folic acid supplements by cows fed no supplementary B12 increased total lipid and triacylglycerols in liver, whereas these supplements had no effect in cows supplemented with B12. The increases in milk and milk protein yields due to folic acid supplements did not seem to be dependent on the vitamin B12 supply. However, when vitamin B12 was given in combination with folic acid, utilization of the 2 vitamins seems to be increased, probably more so in extrahepatic tissues. Metabolic efficiency seems also to be improved as suggested by similar lactational performance and dry matter intake for cows fed supplementary folic acid but increased plasma glucose and decreased hepatic lipids in cows fed folic acid and vitamin B12 together.  相似文献

Apparent ruminal synthesis and duodenal flow of thiamin, riboflavin, niacin, vitamin B6, folates, and vitamin B12 were evaluated in an experiment using 15 ruminally and duodenally cannulated lactating Holstein cows fed a basal diet, according to a crossover design, supplemented or not with 56 g/d of Saccharomyces cerevisiae fermentation product. Duration of the treatment period was 28 d. The basal ration had 28% neutral detergent fber, 30% starch and 16.5% crude protein; forages were corn silage (67% of forage dry matter) and alfalfa silage (33% of forage dry matter). Concentrations of B vitamins were analyzed in feed and duodenal digesta. Apparent ruminal synthesis of each B vitamin was calculated as the duodenal flow minus the intake. Under the present experimental conditions, a dietary supplement of Saccharomyces cerevisiae fermentation product had no effect apparent synthesis of B vitamins in the rumen or on the amounts of these vitamins reaching the duodenum and available for absorption by the dairy cow.  相似文献

Physical restrictions at the feed barrier may cause skin lesions, especially on the necks of cows. Thus, some aspects of the functionality of feed barrier design can be assessed by observing skin lesions on the necks of dairy cattle. In this study, 232 herds, mainly of the breed Norwegian Red (94%), were investigated between September 2006 and June 2007. The herds were categorized into 2 groups. One data set, based on 1,148 cows (115 herds), was from farms using feed barriers with post-and-rail designs (86 herds) and combinations of barriers (29 herds). The second data set, which included 1,168 cows (117 herds), was from farms using only vertical feed barriers such as tombstone, diagonal, and headlock. Skin lesions were observed on the necks of 21% of the 2,335 cows observed. Forty-two percent of cows had neck lesions in herds with post-and-rail barriers, whereas 4% of cows had neck lesions in herds with vertical barriers and 30% of cows had neck lesions in herds with a combination of both horizontal and vertical barriers. Alternating logistic regression was used to identify factors associated with skin lesions on the necks of cows. The estimates were transformed into odds ratios (OR). In herds in which post-and-rail design barriers were used, the following risk factors were associated with skin lesions: manger front lower than 63 cm versus higher (OR = 3.6), no feed stalls versus feed stalls (OR = 10.9), mixed rations versus no mixed ration (OR = 3.8), cows later than 30 DIM versus earlier (OR = 1.9), cows with low milk production versus those producing 1,000 kg higher (OR = 1.4), feeding frequency of less than 3 times daily versus 3 and more times daily (OR = 1.9), using only post-and-rail feed barriers (OR = 1.8) versus using combinations, and farmer disagreement with the statement “animals experience physical pain as humans do” (OR = 1.9) versus agreement. Diagonal barriers, categorized as vertical barriers, increased the association of skin lesions with increasing manger front heights. Feed barrier design was associated with skin lesions on the necks of cows, particularly for barriers of a post-and-rail design. To minimize the risk of neck lesions, the top rail height of post-and-rail barriers should be above 109 cm, especially for cows taller than 126 cm, and the manger front should not be lower than 63 cm. Feed stalls, feeding frequency of 3 or more times daily, and the feeding of unmixed rations were also identified as preventive factors.  相似文献

Effects of the forage-to-concentrate ratio on apparent ruminal synthesis of thiamine, riboflavin, niacin, vitamin B6, folates, and vitamin B12 were evaluated in an experiment using 14 ruminally and duodenally cannulated Holstein cows. The experiment was a crossover design with two 15-d treatment periods and a 14-d preliminary period in which cows were fed a diet intermediate in composition between the treatment diets. Treatments were diets containing low-forage (44.8% forage, 32.8% starch, 24.4% neutral detergent fiber) or high-forage (61.4% forage, 22.5% starch, 30.7% neutral detergent fiber) concentrations. Both diets were formulated with different proportions of the same ingredients. Concentrations of B vitamins were analyzed in feed and duodenal digesta. Apparent ruminal synthesis of each B vitamin was calculated as the duodenal flow minus the intake. The high-forage diet had the highest concentrations of riboflavin, niacin, vitamin B6, and folates, whereas the low-forage diet had the highest thiamine concentration. Vitamin B12 in the diets was under the level of detection. Consequently, despite a reduction in dry matter intake when the cows were fed the high-forage diet, increasing dietary forage concentration increased or tended to increase intakes of riboflavin, niacin, and vitamin B6 but reduced thiamine and folate intakes. Increasing dietary forage concentration reduced apparent ruminal degradation of thiamine and apparent ruminal synthesis of riboflavin, niacin, and folates and increased ruminal degradation of vitamin B6, but had no effect on ruminal synthesis of vitamin B12. As a consequence, increasing the forage-to-concentrate ratio had no effect on the amounts of thiamine, riboflavin, and vitamin B12 reaching the small intestine but decreased the amounts of niacin, vitamin B6, and folates available for absorption. Apparent ruminal syntheses of riboflavin, niacin, folates, and vitamin B12 were correlated positively with the amount of starch digested in the rumen and duodenal flow of microbial N, whereas these correlations were negative for thiamine. Apparent ruminal syntheses of thiamine and vitamin B6 were negatively correlated with their respective intakes, whereas folate intake was positively correlated with its synthesis in the rumen.  相似文献

Although taking body temperature by rectal thermometer is the method most commonly used to identify sick cows in the postpartum period, no data on the repeatability of this measure are available. The overall objective of this study was to evaluate variability of rectal temperatures in dairy cows considering different factors (intra- and interinvestigator repeatability, different thermometers, penetration depth into the rectum, and defecation). High coefficients of correlation (r = 0.98) and small differences between values of rectal temperatures (observer A = 39.3 ± 1.0°C and observer B = 39.4 ± 1.0°C) provide evidence that rectal temperature was a repeatable measure in dairy cows. Testing was carried out using 4 different digital thermometers: GLA M750 (GLA Agricultural Electronics, San Luis Obispo, CA), MTI8101 (SES Scala Electronics, Stahnsdorf, Germany), MT1831 (Microlife AG, Widnau, Switzerland) and Domotherm TH1 (Uebe Medical GmbH, Wertheim, Germany). Thermometers were inserted into the rectum to a certain depth (GLA M750 and MTI8101 = 11.5 cm; MT1831 = 8.4 cm; Domotherm TH1 = 7.7 cm) and a measure was finished when a visual or acoustic signal was emitted by the thermometer. The measures could be influenced by the procedure itself (up to 0.5°C), type of thermometer (up to 0.3°C), and the penetration depth (11.5 cm or 6.0 cm in one of the experiments) into the rectum (up to 0.4°C difference between a penetration depth of 11.5 cm and 6.0 cm in one of the experiments). Differences in rectal temperature before and after defecation were minor (<0.1°C). These results indicate that some care is required in generalizing rectal measures of body temperature.  相似文献

Serum amyloid A3 (SAA3) is the predominant SAA isoform secreted by mammary epithelial cells in dairy cows; it is also expressed in bovine adipose tissue (AT). The adipokine SAA3 is linked to obesity and insulin resistance of AT and the respective inflammatory response, at least in mice. Dietary treatment with conjugated linoleic acids (CLA) reportedly also affects insulin sensitivity and inflammatory status in monogastrics. Both SAA3 and CLA thus seem to alter similar functions. Based on changes in insulin sensitivity and the inflammatory status throughout lactation, we hypothesized that the mRNA abundance of SAA3 in various tissues might be regulated as well and that CLA could be a modulator of SAA3 mRNA expression. In 2 trials, 21 pluriparous and 25 primiparous Holstein cows were fed 100 g/d of a CLA or a control fat supplement from d 1 to 182 or 105 postpartum, respectively. Biopsies from liver and subcutaneous (s.c.) AT from pluriparous cows and samples from 3 different visceral AT and 3 s.c. AT, muscle, mammary gland, and liver tissue from slaughtered primiparous cows were obtained. In an adipocyte cell culture system, cell samples were collected during differentiation of bovine preadipocytes at d 0, 2, 6, 8, 10, 12, and 13 relative to the onset of differentiation. The SAA3 mRNA abundance in tissues and in differentiating bovine preadipocytes was measured by real-time PCR. The presence of the SAA protein was confirmed by Western blotting. Treatment with CLA yielded only few and inconsistent effects on SAA3 mRNA abundance. In both trials, SAA3 mRNA peaked at d 1 postpartum in all tissues except in mesenteric AT, in which the change was not significant. The highest SAA3 mRNA expression was observed in the mammary gland, followed by omental AT. The SAA protein was present in the visceral and s.c. AT depots investigated. Adipocytes as one source of SAA3 were confirmed by the SAA3 mRNA profile in differentiating adipocytes. The longitudinal changes observed point to SAA3 being involved in the inflammatory situation around parturition.  相似文献

The effect of fat supplements (FS) providing different proportions of saturated (SFA) and unsaturated (UFA) fatty acids on supply, apparent ruminal synthesis (ARS), and duodenal flow (DF) of some B vitamins (thiamine, riboflavin, niacin, vitamin B6, folates, and vitamin B12) were evaluated in an experiment using 8 ruminally and duodenally cannulated lactating Holstein cows. The experiment was a replicated 4 × 4 Latin square design with 21-d treatment periods. The 4 treatments were a control diet without fatty acid supplement and 3 diets with 2.5% additional fatty acids from supplements containing (1) SFA, (2) an intermediate mixture of SFA and UFA, or (3) UFA. All diets were served as a total mixed ration once daily at 115% of the expected intake. B-vitamin concentrations were analyzed in feed and duodenal digesta. Apparent ruminal synthesis of each B vitamin was calculated as the DF minus the intake. B-vitamin concentrations were similar among the 4 treatments; consequently, daily intake of the vitamins followed the same pattern as dry matter intake. Adding FS decreased B-vitamin intakes (except vitamin B12), as did increasing the proportion of UFA. Riboflavin and niacin DF and ARS, expressed as total daily amount or per unit of dry matter intake, were not affected by FS, but increasing the proportion of UFA decreased riboflavin and niacin DF and ARS. Fat supplements decreased DF of vitamin B6, expressed either as total daily amount or per unit of dry matter intake. No treatment effects were observed on total daily folate DF and ARS. However, when expressed per unit of dry matter intake, folate DF and ARS were greater when cows were fed FS and they increased linearly with the proportion of UFA in the supplement. Inclusion of fat supplements into the dairy cow diet had a limited effect on the fate of most B vitamins in the rumen although increasing the proportion of UFA in the FS linearly decreased apparent synthesis of riboflavin and niacin in the rumen and the amounts of these vitamins reaching the small intestine.  相似文献
