A serial of star‐shaped poly(ε‐caprolactone)‐b‐poly(ethylene oxide) (SPPCL‐b‐PEO) block copolymers with porphyrin core were successfully synthesized from ring‐opening polymerization (ROP) of ε‐caprolactone (CL) initiated with porphyrin core, followed by coupling reaction with a hydrophilic polymer poly(ethylene oxide) (PEO) shell. The structure of this novel copolymer were synthesized and thoroughly characterized by Nuclear Magnetic Resonance (NMR), Gel Permeation Chromatography (GPC), Fourier Transform Infrared Spectroscopy (FTIR). Notably, the as‐prepared porphyrin‐cored star‐shaped copolymer could self‐assembly into different structures determined by transmission electron microscopy (TEM) and dynamic lighting scattering (DLS), which provides the great potential of using this well‐defined photodynamic therapy material for drug delivery system. In this article, a new kind of biodegradable poly(ε‐caprolactone)‐poly(ethylene glycol)‐poly(ε‐caprolactone)‐based polyurethane (PCEC‐U) copolymers were successfully synthesized by melt‐polycondensation method from ε‐caprolactone (ε‐CL), poly(ethylene glycol) (PEG), 1,4‐butanediol (BD), and isophorone diisocyanate (IPDI). The obtained copolymers were characterized by 1H‐nuclear magnetic resonance (1H‐NMR), FTIR, and gel permeation chromatography (GPC). Thermal properties of PCEC‐U copolymers were studied by DSC and TGA/DTG under nitrogen atmosphere. Water absorption and hydrolytic degradation behavior of these copolymers were also investigated. Hydrolytic degradation behavior was studied by weight loss method. 1H‐NMR and GPC were also used to characterize the hydrolytic degradation behavior of PCEC‐U copolymers. The molecular weight of PCL block and PEG block in soft segment and the content of hard segment strongly affected the water absorption and hydrolytic degradation behavior of PCEC‐U copolymers. © 2009 Wiley Periodicals, Inc. One‐step synthesis of star copolymers by reversible addition–fragmentation chain transfer (RAFT) and ring‐opening polymerization (ROP) by using a novel dual initiator is reported. Triarm block copolymers comprising one polystyrene (or polyacrylamide) arm and two poly(β‐butyrolactone) arms were synthesized in one‐step by simultaneous RAFT polymerization of styrene (St) (or acrylamide, designated as AAm) and ROP of β‐butyrolactone (BL) in the presence of a novel trifunctional initiator, 1,2‐propanediol ethyl xanthogenate (RAFT‐ROP agent). This dual initiator was obtained through the reaction of 3‐chloro‐1,2‐propanediol with the potassium salt of ethyl xanthogenate. The principal parameters such as monomer concentration, initiator concentration, and polymerization time that affect the one‐step polymerization reaction were evaluated. The characterization of the products was achieved using Fourier‐transform infrared spectroscopy (FTIR), 1H‐nuclear magnetic resonance (1H‐NMR), 13C‐nuclear magnetic resonance (13C‐NMR), Gas chromatography–mass spectrometry (GC–MS), gel‐permeation chromatography (GPC), thermogravimetric analysis (TGA), and fractional precipitation (γ) techniques. © 2010 Wiley Periodicals, Inc. In this study, a novel well‐defined epoxy mid‐chain functional macromonomer of poly(ε‐caprolactone) (PCL) has been synthesized by ring‐opening polymerization (ROP) of ε‐caprolactone (ε‐CL) and epoxidation on workup with 3‐chloroperoxybenzoic acid. The ROP of ε‐CL monomer in bulk at 110°C, by means of a dihydroxy functional initiator namely, 3‐cyclohexene‐1,1‐dimethanol in conjunction with stannous‐2‐ethylhexanoate, (Sn(Oct)2), yielded a well‐defined PCL with a cyclohexene mid‐chain group. The epoxidation of the cyclohexene (CH) mid‐chain group of PCL was performed using 3‐chloroperoxybenzoic acid. GPC, IR, and 1H‐NMR analyses revealed that a low‐polydispersity macromonomer of PCL with the desired cyclohexene oxide (CHO) functionality at the mid‐chain was obtained. In this article, we describe the synthesis and solution properties of PEG‐b‐PTMC star block copolymers via ring‐opening polymerization (ROP) of trimethylene carbonate (TMC) monomer initiated at the hydroxyl end group of the core PEG using HCl Et2O as a monomer activator. The ROP of TMC was performed to synthesize PEG‐b‐PTMC star block copolymers with one, two, four, and eight arms. The PEG‐b‐PTMC star block copolymers with same ratio of between hydrophobic PTMC and hydrophilic PEG segments were obtained in quantitative yield and exhibited monomodal GPC curves. The amphiphilic PEG‐b‐PTMC star block copolymers formed spherical micelles with a core–shell structure in an aqueous phase. The mean hydrodynamic diameters of the micelles increased from 17 to 194 nm with increasing arm number. As arm number increased, the critical micelle concentration (CMC) of the PEG‐b‐PTMC star block copolymers increased from 3.1 × 10?3 to 21.1 × 10?3 mg/mL but the partition equilibrium constant, which is an indicator of the hydrophobicity of the micelles of the PEG‐b‐PTMC star block copolymers in aqueous media, decreased from 4.44 × 104 to 1.34 × 104. In conclusion, we confirmed that the PEG‐b‐PTMC star block copolymers form micelles and, hence, may be potential hydrophobic drug delivery vehicles. © 2008 Wiley Periodicals, Inc. Lanthanide metal (II) 2,6‐di‐tert‐butylphenoxide complexes (ArO)2Ln(THF)3 (Ln = Sm 1 , Yb 2 ) alone have been developed to catalyze the ring‐opening polymerization of trimethylenecarbonate (TMC) and random copolymerization of TMC and ε‐caprolactone (ε‐CL) for the first time. The influence of reaction conditions, such as initiator, initiator concentration, polymerization temperature, and polymerization time, on monomer conversion, molecular weight, and molecular weight distribution of the resulting PTMC was investigated. It was found that the divalent complex 1 showed higher activity for the polymerization of TMC than complex 2 . The random structure and thermal behavior of the copolymers P(TMC‐co‐CL) have been characterized by 1H NMR, 13C NMR, GPC, and DSC analysis. © 2007 Wiley Periodicals, Inc. Two series of poly(ε‐caprolactone)‐b‐poly(ethylene glycol)‐b‐poly(ε‐caprolactone) triblock copolymers were prepared by the ring opening polymerization of ε‐caprolactone in the presence of poly(ethylene glycol) and dibutylmagnesium in 1,4‐dioxane solution at 70°C. The triblock structure and molecular weight of the copolymers were analyzed and confirmed by 1H NMR, 13C NMR, FTIR, and gel permeation chromatography. The crystallization and thermal properties of the copolymers were investigated by wide‐angle X‐ray diffraction (WAXD) and differential scanning calorimetry (DSC). The results illustrated that the crystallization and melting behaviors of the copolymers were depended on the copolymer composition and the relative length of each block in copolymers. Crystallization exothermal peaks (Tc) and melting endothermic peaks (Tm) of PEG block were significantly influenced by the relative length of PCL blocks, due to the hindrance of the lateral PCL blocks. With increasing of the length of PCL blocks, the diffraction and the melting peak of PEG block disappeared gradually in the WAXD patterns and DSC curves, respectively. 2,5‐Dibromo‐1,4‐(dihydroxymethyl)benzene was used as initiator in ring‐opening polymerization of ε‐caprolactone in the presence of stannous octoate (Sn(Oct)2) catalyst. The resulting poly(ε‐caprolactone) (PCL) macromonomer, with a central 2,5‐dibromo‐1,4‐diphenylene group, was used in combination with 1,4‐dibromo‐2,5‐dimethylbenzene for a Suzuki coupling in the presence of Pd(PPh3)4 as catalyst or using the system NiCl2/bpy/PPh3/Zn for a Yamamoto‐type polymerization. The poly(p‐phenylenes) (PPP) obtained, with PCL side chains, have solubility properties similar to those of the starting macromonomer, ie soluble in common organic solvents at room temperature. The new polymers were characterized by 1H and 13C NMR and UV spectroscopy and also by GPC measurements. The thermal behaviour of the precursor PCL macromonomer and the final poly(p‐phenylene)‐graft‐poly(ε‐caprolactone) copolymers were investigated by thermogravimetric analysis and differential scanning calorimetry analyses and compared. Hexa‐armed star‐shaped poly(ε‐caprolactone)‐block‐poly(L ‐lactide) (6sPCL‐b‐PLLA) with dipentaerythritol core were synthesized by a two‐step ring‐opening polymerization. GPC and 1H NMR data demonstrate that the polymerization courses are under control. The molecular weight of 6sPCLs and 6sPCL‐b‐PLLAs increases with increasing molar ratio of monomer to initiator, and the molecular weight distribution is in the range of 1.03–1.10. The investigation of the melting and crystallization demonstrated that the values of crystallization temperature (Tc), melting temperature (Tm), and the degree of crystallinity (Xc) of PLLA blocks are increased with the chain length increase of PLLA in the 6sPCL‐b‐PLLA copolymers. On the contrary, the crystallization of PCL blocks dominates when the chain length of PLLA is too short. According to the results of polarized optical micrographs, both the spherulitic growth rate (G) and the spherulitic morphology are affected by the macromolecular architecture and the length of the block chains. © 2010 Wiley Periodicals, Inc. A new monomer, exo‐3,6‐epoxy‐1,2,3,6‐tetrahydrophthalimidocaproic acid (ETCA), was prepared by reaction of maleimidocaproic acid and furan. The homopolymer of ETCA and its copolymers with acrylic acid (AA) or with vinyl acetate (VAc) were obtained by photopolymerizations using 2,2‐dimethoxy‐2‐phenylacetophenone as an initiator at 25 °C. The synthesized ETCA and its polymers were identified by FTIR, 1H NMR and 13C NMR spectroscopies. The apparent average molecular weights and polydispersity indices determined by gel permeation chromatography (GPC) were as follows: Mn = 9600 g mol?1, Mw = 9800 g mol?1, Mw/Mn = 1.1 for poly(ETCA); Mn = 14 300 g mol?1, Mw = 16 200 g mol?1, Mw/Mn = 1.2 for poly(ETCA‐co‐AA); Mn = 17 900 g mol?1, Mw = 18 300 g mol?1, Mw/Mn = 1.1 for poly(ETCA‐co‐VAc). The in vitro cytotoxicity of the synthesized compounds against mouse mammary carcinoma and human histiocytic lymphoma cancer cell lines decreased in the following order: 5‐fluorouracil (5‐FU) ≥ ETCA > polymers. The in vivo antitumour activity of the polymers against Balb/C mice bearing sarcoma 180 tumour cells was greater than that of 5‐FU at all doses tested. © 2001 Society of Chemical Industry  相似文献

Combination of cationic, redox free radical, and thermal free radical polymerizations was performed to obtain linear and star polytetramethylene oxide (poly‐THF)‐polymethyl methacrylate (PMMA)/polystyrene (PSt) multiblock copolymers. Cationic polymerization of THF was initiated by the mixture of AgSbF6 and bis(4,4′ bromo‐methyl benzoyl) peroxide (BBP) or bis (3,5,3′,5′ dibromomethyl benzoyl) peroxide (BDBP) at 20°C to obtain linear and star poly‐THF initiators with Mw varying from 7,500 to 59,000 Da. Poly‐THF samples with hydroxyl ends were used in the methyl methacrylate (MMA) polymerization in the presence of Ce(IV) salt at 40°C to obtain poly(THF‐b‐MMA) block copolymers containing the peroxide group in the middle. Poly(MMA‐b‐THF) linear and star block copolymers having the peroxide group in the chain were used in the polymerization of methyl methacrylate (MMA) and styrene (St) at 80°C to obtain PMMA‐b‐PTHF‐b‐PMMA and PMMA‐b‐PTHF‐b‐PSt linear and star multiblock copolymers. Nuclear magnetic resonance spectroscopy (NMR) characterization of the statistical copolymers of this study showed that the poly(ε‐caprolactone‐co‐L‐lactide)s, with ε‐caprolactone (ε‐CL) molar contents ranging from 70 to 94% and ε‐CL average sequence length (lCL) between 2.20–9.52, and the poly(ε‐caprolactone‐co‐δ‐valerolactone)s, with 60 to 85% of ε‐CL and lCL between 2.65–6.08, present semi‐alternating (R→2) and random (R~1) distribution of sequences, respectively. These syntheses were carried out with the aim of reducing the crystallinity of poly(ε‐caprolactone) (PCL), needed to provide mechanical strength to the material but also responsible for its slow degradation rate. However, this was not achieved in the case of the ε‐caprolactone‐co‐δ‐valerolactone (ε‐CL‐co‐δ‐VAL). Non‐isothermal cooling treatments at different rates and isothermal crystallizations (at 5, 10, 21 and 37°C) were conducted by differential scanning calorimetry (DSC), and demonstrated that ε‐CL copolymers containing δ‐valerolactone (δ‐VAL) exhibited a larger crystallization capability than those of L‐lactide (L‐LA) and also arranged into crystalline structures over shorter times. The crystallization enthalpies of the ε‐CL‐co‐δ‐VAL copolymers during the cooling treatments and their heat of fusion (ΔHm) at the different isothermal temperatures were very large (i.e. ΔHc > 53 Jg?1) and in some cases, unrelated to the copolymer composition. In some compositions, such as the 60 : 40, Wide Angle X‐ray Scattering (WAXS) proved that that these two lactones undergo isomorphism and co‐crystallize in a single cell. © 2015 Wiley Periodicals, Inc. This study describes the synthesis of amphiphilic ABC‐triblock copolymers comprising a central pseudopoly(4‐hydroxy‐L ‐proline) segment and terminal hydrophilic poly(ethylene glycol)methyl ether as well as hydrophobic poly(ε‐caprolactone) blocks. Differential scanning calorimetry, 1H‐NMR spectroscopy, and gel permeation chromatography are used to characterize the copolymers. The thermal properties (Tg and Tms) of the triblock copolymers depend on the composition of polymers. Larger amounts of ε‐CL incorporated into the macromolecular backbone increased Tg and Tms. Fluorescence spectroscopy, transmission electron microscopy, and dynamic light scattering are utilized to investigate their micellar characteristics in the aqueous phase. Observations showed a higher critical micelle concentration with higher hydrophilic components in the copolymers. The micelle exhibited a core‐shell‐corona and/or vesicle shape, and the average size was less than 300 nm. Star‐shaped copolymers poly(ε‐caprolactone)‐bolck‐poly(ε‐benzyloxycarbonyl‐l ‐lysine) (SPPCL‐b‐PZLLs) with porphyrin core were synthesized by a sequential ring‐opening polymerization (ROP) of CL and Nε‐Benzyloxycarbonyl‐l ‐lysine N‐Carboxyanhydride. After the deprotection of benzyloxycarbonyl groups in polylysine blocks, the star‐shaped amphiphilic copolymers SPPCL‐b‐PLLs were obtained. These amphiphilic copolymers can self‐assemble into micelles or aggregates in aqueous solution. Investigation shows that the morphology of micelles/aggregates varied according to the change of pH values of media, indicating the pH‐responsive property of SPPCL‐b‐PLL copolymers. Furthermore, associated with conjugated porphyrin cores, the SPPCL‐b‐PLL copolymers micelles showed a certain degree of Photodynamic Therapy (PDT) effects on tumor cells, suggesting its potential application as carrier for hydrophobic drug with additional therapeutic ability of inherent porphyrin segments. © 2013 Wiley Periodicals, Inc. The main aims of the work reported here were to synthesize and characterize a new 2,2′‐ethylidene‐bis(4,6‐di‐tert‐butylphenol) (EDBPH2)‐based bimetal yttrium complex, Y(EDBP)2(DME)Na(DME)3 (1c; where DME is ethylene glycol dimethyl ether), which was employed as an efficient initiator for the ring‐opening polymerization of ε‐caprolactone (ε‐CL). From single‐crystal X‐ray diffraction, the solid structure of this new bimetal initiator was well established. Experimental results show that 1c initiates the ring‐opening polymerization of ε‐CL to afford poly(ε‐CL) with a narrow molecular weight distribution (Mw/Mn = 1.09–1.36, 65 °C). Based on an in situ NMR study, a plausible coordination–insertion mechanism is then proposed. The bimetal complex 1c can be used as an initiator for the ring‐opening polymerization of ε‐CL with some living characteristics. A study of the mechanism reveals that DME displacement in 1c by ε‐CL is involved in the initiation process and the propagation may proceed through three pathways by Na? O insertion or Y? O insertion. Novel polyesters, poly[(ε‐caprolactone)‐co‐(N‐trityl‐L ‐serine‐β‐lactone)]s, were prepared by copolymerizing ε‐caprolactone (CL) with N‐trityl‐L ‐serine‐β‐lactone (TSL) using ZnEt2 as the catalyst. The number‐average molecular weights were determined which ranged from 2.7 × 104 to 4.9 × 104 Da with dispersity values ranging from 1.6 to 1.8. The structures of the copolymers were investigated by means of 1H NMR, 13C NMR and infrared spectroscopies, thermogravimetric analysis and differential scanning calorimetry. The results indicated that CL and TSL monomer units were randomly distributed within the copolymer backbone structures and the ratios of TSL to CL in the copolymers were close to those in the feeds. After removal of the trityl group under mild condition, a new polyester with side amino groups provided by serine units was obtained. L929 cell culturing test indicated good biocompatibility of the polyester with or without protective groups. © 2012 Society of Chemical Industry  相似文献

A new monomer, 3,6‐endo‐methylene‐1,2,3,6‐tetrahydrophthalimidobutanoyl‐5‐fluorouracil (ETBFU), was synthesized by reaction of 3,6‐endo‐methylene‐1,2,3,6‐tetrahydrophthalimidobutanoyl chloride and 5‐fluorouracil. The homopolymer of ETBFU and its copolymers with acrylic acid (AA) or vinyl acetate (VAc) were prepared by photopolymerization using 2,2‐dimethoxy‐2‐phenylacetophenone as an initiator at 25 °C. The synthesized ETBFU and its polymers were identified by FTIR, 1H NMR and 13C NMR spectroscopies. The ETBFU content in poly(ETBFU‐co‐AA) and poly(ETBFU‐co‐VAc) was 43 and 14 mol%, respectively. The apparent number‐average molecular weight (Mn) of the polymers determined by GPC ranged from 8400 to 11 300. The in vitro cytotoxicity of the samples against mouse mammary carcinoma (FM3A), mouse leukaemia (P388), and human histiocytic lymphoma (U937) cancer cell lines decreased in the order 5‐FU ≥ ETBFU > poly(ETBFU) > poly(ETBFU‐co‐AA) > poly(ETBFU‐co‐VAc). Diblock copolymers of poly(L ‐lactide)‐block‐poly(methyl methacrylate) (PLLA‐b‐PMMA) were synthesized through a sequential two‐step strategy, which combines ring‐opening polymerization (ROP) and atom transfer radical polymerization (ATRP), using a bifunctional initiator, 2,2,2‐trichloroethanol. The trichloro‐terminated poly(L ‐lactide) (PLLA‐Cl) with high molecular weight (Mn,GPC = 1–12 × 104 g/mol) was presynthesized through bulk ROP of L ‐lactide (L ‐LA), initiated by the hydroxyl group of the double‐headed initiator, with tin(II) octoate (Sn(Oct)2) as catalyst. The second segment of the block copolymer was synthesized by the ATRP of methyl methacrylate (MMA), with PLLA‐Cl as macroinitiator and CuCl/N,N,N′,N″,N″‐pentamethyldiethylenetriamine (PMDETA) as catalyst, and dimethyl sulfoxide (DMSO) was chosen as reaction medium due to the poor solubility of the macroinitiator in conventional solvents at the reaction temperature. The trichloroethoxyl terminal group of the macroinitiator was confirmed by Fourier transform infrared spectroscopy (FTIR) and 1H‐NMR spectroscopy. The comprehensive results from GPC, FTIR, 1H‐NMR analysis indicate that diblock copolymers PLLA‐b‐PMMA (Mn,GPC = 5–13 × 104 g/mol) with desired molecular composition were obtained by changing the molar ratio of monomer/initiator. DSC, XRD, and TG analyses establish that the crystallization of copolymers is inhibited with the introduction of PMMA segment, which will be beneficial to ameliorating the brittleness, and furthermore, to improving the thermal performance. © 2010 Wiley Periodicals, Inc. Reverse iodine transfer polymerizations (RITP) of 2‐h‐ydroxyethyl acrylate (HEA) were performed in N,N‐dimethylformamide at 75°C using AIBN as initiator. Poly(2‐hydroxyethyl acrylate) (PHEA) with Mn = 3300 g mol?1 and Mw/Mn <1.5 were obtained. Homopolymerization of styrene in RITP was also carried out under similar conditions using toluene as solvent. The resulting iodo‐polystyrene (PS‐I) with (Mn, SEC = 607 g mol?1, polydispersity index (PDI) = 1.31) was used as a macroinitiator for the synthesis of amphiphilic block copolymers based on HEA with controlled well‐defined structure. Poly(styrene‐b‐2‐hydroxyethyl acrylate) (PS‐b‐PHEA) with Mn = 13,000 g mol?1 and polydispersity index (Mw/Mn) = 1.4 was obtained, copolymer composition was characterized using 1H‐NMR and FTIR, whereas SEC and gradient HPLC were used to confirm the formation of block copolymer and the living character of polymer chains. © 2012 Wiley Periodicals, Inc. Multi‐arm star polystyrenes with hyperbranched polyester (HP3) core were prepared by atom transfer radical polymerization (ATRP). The structures of the polymers were investigated with FTIR and 1H NMR. GPC results showed that the resultant polymers had relatively broad polydispersity indices that arouse from the macromolecular initiator (HP3‐Br). The thermal properties were studied using differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA). DSC analysis indicated that polystyrene star polymers had only the glass transition temperatures (Tg), which changes with the weight ratio of multi‐functional macroinitiator‐to‐monomer.
