免疫球蛋白G (Immunoglobulin G, IgG)的N-糖基化在炎症性疾病的发展中起着重要作用。本研究旨在评价IgG N-糖基在消化道癌症亚型中的诊断效能。从中国医学科学院肿瘤医院招募749名消化道癌症患者,包括食管癌(esophageal cancer, EC)、胃癌(gastric cancer, GC)、结直肠癌(colorectal cancer, CRC)和胰腺癌(pancreatic cancer, PC)患者。采用亲水交互高效液相色谱-超高效液相色谱(hydrophilic interaction liquid chromatography using ultra-performance liquid chromatography, HILIC-UPLC)分析血浆中IgG的N-糖基构成。采用Bio-Plex悬液芯片系统检测方法(Bio-Rad)进行炎症因子检测。采用典型相关分析(canonical correlation analysis, CCA)探索糖基和炎症因子之间的相关性。采用LASSO回归和logistic回归模型,基于检测到的糖基谱建立可用于区分胃肠...  相似文献

系统性红斑狼疮(systemic lupus erythematosus, SLE)是一种发病机制不明、临床表型异质性大的自身免疫性疾病。目前已有的SLE血清生物标志物灵敏度或特异性有限,使得SLE的早期精准诊断存在困难。在本研究中,通过对389例SLE患者及304例健康对照者进行深入的糖组学分析,鉴定出血清免疫球蛋白G(IgG)上的两种N-糖链能够作为SLE的诊断生物标志物。在其他易与SLE混淆的系统性自身免疫性疾病(如类风湿性关节炎、原发性干燥综合征或系统性硬化症)中,这两种生物标志物没有出现显著变化,提示这两种N-糖链生物标志物对诊断SLE具有特异性。值得注意的是,这两种N-糖链生物标志物被证明是自身抗体非依赖性的,并且适用于所有阶段的SLE患者。基于片段特异性糖链分析和糖肽分析,发现这两种N-糖链生物标志物位于IgG上的Fc区域,并与疾病活动性密切相关。而酶学分析结果则提示,SLE患者体内一系列糖转移酶的失调可能是观察到的糖链产生变化的原因。研究结果为基于血清IgG糖基化的高效人群筛查和SLE潜在的新致病因素提供了新的思路。  相似文献

采用一种不含有机溶剂的聚酰胺酸上浆剂实施上浆处理,为在T300级炭纤维(3 k)表面制备聚酰亚胺(PI)涂层提供一种绿色途径。通过傅里叶变换红外光谱和扫描电子显微镜对PI涂覆炭纤维的表面进行分析,结果表明,在炭纤维表面形成一层连续且均一的PI涂层。通过热重分析和单丝拉伸测试,考察炭纤维的热稳定性。与包含环氧上浆剂的炭纤维相比,在耐高温树脂的加工温度范围(300~400℃)内PI涂覆炭纤维表现出优异的热稳定性,起始分解温度与5%热失重温度分别高达567℃及619℃。此外,在空气中、400℃条件下氧化1 h后,PI涂覆炭纤维的拉伸强度仅下降6%,明显低于包含环氧上浆剂炭纤维(拉伸强度下降22%)。  相似文献

不加金属催化剂,以碱木素酚醛树脂(LPF)和硅粉作为原料在低温条件下合成SiC纳米线。利用SEM、TEM、XRD表征样品的形貌及显微结构,用热力学方法分析反应条件对SiC纳米线生长的影响。结果表明,SiC纳米线在1100℃左右开始生长,其由气-液-固生长机理控制,同时其生成温度比用商业酚醛树脂作为原料低。生成的SiC纳米线的直径为30~100nm并沿晶面的[1 1 1]方向生长。碱木素酚醛树脂中的无机盐在热解炭化过程中原位形成熔盐并起着液相催化剂球滴的作用,促进SiC纳米线的生长,并提出合成SiC纳米线的生长机理模型。  相似文献

食用煮熟的小龙虾可能导致哈夫病(HD),该病被认为是由不明毒素引起的,而病因尚不清楚。N-糖组的剖析将促进破译疾病的分子机制,而HD相关的糖基化从未被探索过。2019—2020年期间,本研究团队招募了来自武汉市疾病预防控制中心的90份HD患者和对照组血清样本。本文中,采用基于高通量的正交质谱对HD中血清和血清衍生的IgG的N-糖组谱进行了表征。数据显示,HD与总血清糖蛋白的核心岩藻糖基化和单半乳糖醇化升高有关。血清IgG水平是HD患者的良好指标。此外,IgG的差异半乳糖基化和唾液酸化与HD密切相关。值得注意的是,IgG1和IgG2的半乳糖基化和唾液酸化的变化具有亚类特异性。有意义的是,IgG2或IgG3/4的唾液酸化和半乳糖基化改变与HD的临床标志物密切相关。本研究表明差异化IgG N-糖基化与HD的关联,为这种罕见疾病的病因提供了新的见解。  相似文献

利用大气等离子喷涂制备了ZrO2涂层,采用扫描电子显微镜结合图像分析的方法对涂层的显微结构特征(总气孔率和大气率)进行了量化表征.同时利用扫描电子显微镜附带X射线扫描成像对涂层内部的微裂纹和大的分隔裂纹的三维分布进行了表征.据此建立了ZrO2涂层显微结构与热导率之间的关系.室温下,涂层N2的热导率较N1的低,因为涂层N2内气孔和微裂纹数较多;在1000℃,由于微裂纹的烧结作用导致热导率增大,所以涂层N2的热导率较N1反而高.高温下,涂层内由微裂纹汇聚而形成的大的分隔裂纹的存在则可以有效地抑制烧结,降低涂层的热导率.  相似文献

硝基苯选择性加氢反应是生产苯胺的重要方法,其关键在于开发一种具有高效选择性且低成本的绿色催化剂。本文基于葡萄糖水热炭化得到水热炭前驱体,结合尿素和硝酸钴一步高温热解制备了一种高度分散的富含Co纳米颗粒的氮掺杂多孔炭材料,将其作为催化剂用于硝基苯加氢反应。详细研究了热解温度对催化剂结构和催化活性的影响,证实了催化活性主要受催化剂的比表面积、Co掺杂量及其Co-Nx配位效应的影响。结果表明,将含10%Co源制备的前驱体800°C热解得到的催化剂(Co@NCG-800)表现出优异的硝基苯催化加氢性能,在以异丙醇为溶剂、100°C和1 MPa氢压下反应2.5 h时可实现硝基苯的转化,苯胺选择性高达99%。催化剂循环使用6次后的硝基苯转化率和苯胺选择性几乎保持不变。优异的循环稳定性能可归因于多孔炭材料中的氮元素和Co纳米颗粒的强相互作用,同时磁性Co纳米颗粒使得催化剂具有较好的分离特性和重复使用性。  相似文献

以聚乳酸、正硅酸乙酯和硝酸钙为原料,采用溶胶-凝胶方法制备了不同比例的聚乳酸/SiO2-CaO多孔复合薄膜.采用扫描电子显微镜和红外光谱仪对薄膜的微观结构和组成进行分析,并利用表面Zeta电位测定仪对其表面电位进行表征,通过模拟体液浸泡实验和MTT方法对薄膜的生物活性和细胞毒性进行评价.结果表明:复合薄膜在模拟体液中经过7d的浸泡,薄膜表面形成了环状结构的类骨磷灰石层;随着SiO2-CaO含量增加,复合薄膜表面微孔的孔径变小,薄膜表面Zeta电位变负,诱导类骨磷灰石沉积的能力增强;MTT实验证实复合薄膜对MG-63细胞没有毒性且有利于细胞的增殖.  相似文献
