This study is aimed at investigating the feasibility of casein glycomacropeptide (GMP) hydrolysates as potential prebiotics in yogurt. The growth performance of Bifidobacterium animalis subsp. lactis (Bb12) was determined in the presence of GMP hydrolysates produced by the action of proteolytic enzymes (trypsin and papain) with various degrees of hydrolysis. The growth‐promoting effect of GHP on Lactobacillus bulgaricus and Streptococcu thermophilus were also evaluated. Results showed that the optimum hydrolysis time of GMP with trypsin or papain for Bb12 growth promotion was 2 and 0.5 h, respectively. Compared with GMP and its trypsin hydrolysate, the GMP hydrolysate produced with papain (GHP) was the best for Bb12 growth, which obtained the highest viable count (9.3 log cfu mL?1) and the lowest pH of 4.69. The obtained GHP significantly improved the growth of S. thermophilus (P < 0.05), while it had little effect on the growth of L. bulgaricus (P > 0.05)., The viable count of Bb12 of the yogurt obtained with the addition of 1.5% GHP was about four times higher than that of the control without GHP addition. The growth‐promoting effect of GHP might be related to its high content of Glu, Leu and Ala while had no direct relationship with sialic acid content. 相似文献

Bovine glycomacropeptide (GMP) is a 7,000-Da glycopolypeptide released from κ-casein during cheese making. The O-glycan chains linked to GMP have many biological activities, but their utilization for nutraceutical products is limited due to their low content. To concentrate the functional glycan chains of GMP, we prepared sialylglycopeptide concentrate (SGC) from GMP-containing whey protein concentrate via proteolytic digestion of peptide chains and concentration of sialylglycopeptide by ultrafiltration using membranes with a molecular weight cut-off of 1,000 Da. The abundant saccharides detected in the prepared SGC were N-acetylneuraminic acid (Neu5Ac: 32.3% wt/wt), N-acetylgalactosamine (11.3%), and galactose (10.2%), which constitute O-glycans attached to GMP. The Neu5Ac content in SGC was found concentrated at approximately 4.8-fold of its content in GMP-containing whey protein concentrate (6.8%). Structural analysis of O-glycopeptides by liquid chromatography tandem mass spectrometry identified 88 O-glycopeptides. Moreover, O-acetylated or O-diacetylated Neu5Ac was detected in addition to the previously characterized O-glycans of GMP. Quantitative analysis of O-glycan in SGC by fluorescence labeling of chemically released O-glycan revealed that a disialylated tetrasaccharide was the most abundant glycan (76.6% of the total O-glycan). We further examined bifidogenic properties of SGC in vitro, which revealed that SGC served as a more potent carbon source than GMP and contributes to the growth-promoting effects on certain species of bifidobacteria. Overall, our study findings indicate that SGC contains abundant O-glycans and has a bifidogenic activity. Moreover, the protocol for the preparation of SGC described herein is relatively simple, providing a high yield of glycan, and can be used for large-scale preparation. 相似文献

采用气相色谱法对驼、牛、羊乳3 种乳中脂肪酸(fatty acids,FA)种类及含量进行比较。结果表明,3 种乳脂肪酸种类和含量差异显著。驼乳检测出33 种脂肪酸,棕榈酸相对含量最高((23.73±2.17)%);驼乳的多不饱和脂肪酸(polyunsaturated fatty acids,PUFA)相对含量((4.09±0.54)%)显著高于牛乳和羊乳;牛乳中检测出29 种脂肪酸,棕榈酸相对含量最高((27.44±2.45)%);羊乳检测出34 种脂肪酸,且油酸相对含量最高((30.89±3.57)%)。3 种乳中羊乳单不饱和脂肪酸(monounsaturated fatty acids,MUFA)含量最高((35.26±3.86)%)。此外,结合主成分分析和相关性分析方法,对驼乳的脂肪酸含量进行进一步分析,结果显示,驼乳的饱和脂肪酸(saturated fatty acids,SFA)∶MUFA∶PUFA为1∶0.60∶0.08,驼乳更接近联合国粮食及农业组织/世界卫生组织推荐SFA∶MUFA∶PUFA为1∶1∶1。PUFA与短链脂肪酸(short-chain fatty acids,SCFA)、长链脂肪酸(long-chain fatty acids,LCFA)呈负相关,与n-6 FA相关性最大(0.978);SCFA与LCFA呈正相关,n-6 FA与n-3 FA呈正相关。 相似文献

长双歧杆菌BBMN68是来源于中国广西巴马长寿老人肠道的益生菌。本研究利用生物信息学技术分析该菌株寡糖糖苷水解酶基因,并利用生长试验及RT-PCR方法验证菌株对寡糖的利用。结果显示,该菌株具有多种糖苷水解酶基因,具有分解含半乳糖、葡萄糖、N-乙酰葡萄糖胺、N-乙酰半乳糖胺、果糖、甘露糖、阿拉伯糖和木糖单体的寡糖的潜质。生长试验表明,菌株可在低聚木糖、低聚半乳糖、低聚异麦芽糖、低聚麦芽糖、低聚果糖中良好生长。RT-PCR试验结果表明,不同条件下,寡糖转运系统基因BBMN68_74,BBMN68_75,BBMN68_217与BBMN68_925等16个基因可被不同程度地诱导表达。 相似文献

Bifidobacterium longum subsp. infantis ATCC 15697 possesses five α-L-fucosidases, which have been previously characterized toward fucosylated human milk oligosaccharides containing α1,2/3/4-linked fucose [Sela et al.: Appl. Environ. Microbiol., 78, 795-803 (2012)]. In this study, two glycoside hydrolase family 29 α-L-fucosidases out of five (Blon_0426 and Blon_0248) were found to be 1,6-α-L-fucosidases acting on core α1,6-fucose on the N-glycan of glycoproteins. These enzymes readily hydrolyzed p-nitrophenyl-α-L-fucoside and Fucα1-6GlcNAc, but hardly hydrolyzed Fucα1-6(GlcNAcβ1-4)GlcNAc, suggesting that they de-fucosylate Fucα1-6GlcNAcβ1-Asn-peptides/proteins generated by the action of endo-β- N-acetylglucosaminidase. We demonstrated that Blon_0426 can de-fucosylate Fucα1-6GlcNAc-IgG prepared from Rituximab using Endo-CoM from Cordyceps militaris. To generate homogenous non-fucosylated N-glycan-containing IgG with high antibody-dependent cellular cytotoxicity (ADCC) activity, the resulting GlcNAc-IgG has a potential to be a good acceptor substrate for the glycosynthase mutant of Endo-M from Mucor hiemalis. Collectively, our results strongly suggest that Blon_0426 and Blon_0248 are useful for glycoprotein glycan remodeling. 相似文献

The addition of Bifidobacterium to goat milk has dual effects on health, for which various inherent nutrients of goat milk are retained and live probiotics are provided. We explored the effect of Bifidobacterium animalis ssp. lactis Probio-M8 (Probio-M8) on fermentation characteristics, formation of organic acid, sensory properties, and storage characteristics of fermented goat milk (with added 4.0% sucrose). Addition of Probio-M8 decreased the fermentation time and significantly increased the content of functional organic acids, such as acetic acid, and functional long-chain unsaturated fatty acids, including linoleic acid, α-linolenic acid, and docosahexaenoic acid. Furthermore, the contents of medium-chain and short-chain fatty acids, which are related to “goaty” flavor, were significantly lower in the Probio-M8 treatment compared with the control. The number of living Probio-M8 decreased from 8.27 log cfu/mL (1.80 × 108 cfu/mL) to 7.94 log cfu/mL (0.79 × 108 cfu/mL) after 28 d of storage. Titratable acidity and pH value did not differ between the control group and experimental group (containing Probio-M8). Sensory evaluation indicated a lower goaty flavor and odor in the Probio-M8 fermented milk. Our results suggest that the addition of the probiotic Probio-M8 could improve the sensory, physicochemical, and functional properties of fermented goat milk. 相似文献

采用气质联用法(GC-MS)测定东北区域不同泌乳期的母乳及牛乳和山羊乳常乳脂肪酸组成,并对其组成和含量进行差异分析,旨在为母乳脂质组学及以牛羊乳为基质的婴儿配方食品提供一定的理论基础。结果表明:母乳中主要脂肪酸为棕榈酸、油酸、亚油酸等,其中油酸含量最为丰富。饱和脂肪酸(SFA)、单不饱和脂肪酸(MUFA)和多不饱和脂肪酸(PUFA)含量在不同泌乳期存在差异,其中SFA差异显著(p<0.05),初乳、过渡乳和成熟乳SFA含量分别为36.16%、37.89%、38.10%。牛乳和山羊乳主要以SFA为主,山羊乳SFA含量最高(69.07%),SFA中辛酸和癸酸是羊乳的特征脂肪酸,其含量显著高于母乳和牛乳(p<0.05)。山羊乳中中链脂肪酸(MCFA)含量最高(21.03%),是牛乳的1.5倍。其中,母乳SFA:MUFA:PUFA的比例为1.41:1.29:1,牛乳为19.12:9.98:1,山羊乳为11.14:3.98:1,山羊乳脂肪酸组成在比例上更加接近母乳。山羊乳在婴儿配方食品开发方面有更高的优势和开发空间。 相似文献

为阐明双峰驼乳中乳脂球膜蛋白(milk fat globule membrane proteins,MFGMP)的组成以及与牛乳MFGMP之间的差异,以牛乳和双峰驼乳中的MFGMP为研究对象,采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳、双向电泳和液相色谱-串联质谱(liquid chromatograph-tandem mass spectrometry,LC-MS/MS)技术对其中的高丰度蛋白质进行表征,并对双峰驼乳MFGMP提取方法进行选择。结果显示,双峰驼乳和牛乳MFGMP中含量最高的蛋白组分依次为嗜乳脂蛋白、黄嘌呤氧化还原酶/脱氢酶、乳凝集素(MFG-E8或PAS6/7)、脂肪分化相关蛋白(ADRP或PLIN2),3 种方法分析双峰驼乳和牛乳中MFGMP中高丰度蛋白组分的结果具有一致性。基于LC-MS/MS技术对双峰驼乳的MFGMP进行鉴定,并将对鉴定出的蛋白质进行基因本体功能注释和京都基因与基因组百科全书路径分析,发现这些蛋白主要是细胞膜、线粒体、内质网膜等组分,参与的生物途径有翻译和翻译的起始、氧化还原过程、细胞内蛋白质转运、蛋白质折叠、细胞氧化还原稳态等,具有结合活性和分子催化等分子功能。 相似文献

为阐明不同泌乳期间牛乳乳脂肪球膜蛋白的差异,选定牛初乳和牛常乳中的乳脂肪球膜蛋白作为研究对象,利用定量蛋白质组学技术对其中的蛋白质进行表征,并对两组间的丰度差异蛋白进行筛选及生物信息学分析。本研究共鉴定到763 种蛋白,其中197 种为两组共有蛋白,进一步从其中筛选出80 种丰度差异蛋白,包括41 种上调蛋白和39 种下调蛋白(牛初乳/牛常乳)。对上述丰度差异蛋白进行生物信息学分析发现,这些蛋白参与的主要细胞组分为细胞外泌体、细胞外空间、细胞外区域等,参与的主要代谢通路为代谢途径、嘌呤代谢、苯丙氨酸代谢、酪氨酸代谢、丙酮酸代谢、糖酵解/糖异生等,并进一步筛选出31 种与其他蛋白存在相互作用的关键蛋白,包括结合珠蛋白、2-磷酸-D-甘油酸水解酶等。研究结果将有助于了解泌乳期间乳蛋白成分的差异及其功能性,为牛乳制品的精深加工奠定基础。 相似文献

为阐明牛乳蛋白质N-糖基化,采用糖蛋白质组学技术,在牛初乳和牛常乳乳清中共鉴定到154 个N-糖蛋白和246 个糖基化位点,其中,牛初乳鉴定到117 个N-糖蛋白和183 个糖基化位点;牛常乳鉴定到109 个N-糖蛋白和145 个糖基化位点。初乳中丛生蛋白(P17697)糖基化位点N-283表达量最高,常乳中α-乳白蛋白糖基化位点N-93表达量最高。考虑定量差异及有无差异,在牛初乳和牛常乳乳清中共鉴定到129 个糖蛋白的190 个差异表达糖基化位点。基因本体论功能注释表明,差异表达糖蛋白参与的生物学过程是生物调节、刺激性反应、多细胞生物过程、定位、免疫系统过程等;主要分布为细胞外区域和细胞器;主要的分子功能是结合作用、催化活性和分子功能调节。差异表达糖蛋白参与的代谢通路主要是补体与凝血级联、金黄色葡萄球菌感染和溶酶体等。此外,通过蛋白互作分析,找到一些具有高连接度的重要糖蛋白。本研究丰富了牛乳N-糖蛋白质组成及其糖基化位点信息,阐明了牛乳乳清N-糖基化的功能,为评价和改善牛乳品质、研发婴幼儿配方乳中糖蛋白质的改良及功能食品的生产提供了理论依据。 相似文献

乳清富含多种功能特性和生物活性的蛋白质,本研究利用SDS-PAGE电泳将牛初乳与牛乳中乳清蛋白质的组成部分进行分离鉴定,发现牛初乳与牛乳中乳清蛋白质的组成存在较大的差异,且在牛初乳乳清中鉴定出290种蛋白,牛乳乳清中鉴定出325种蛋白。由GO功能注释分析发现,在生物过程中,牛初乳乳清蛋白在细胞定位建立和细胞定位中的作用略高于牛乳乳清蛋白。在分子功能上酶抑制活性作用是牛初乳乳清蛋白和牛乳中乳清蛋白的主要分子功能。在细胞组成上牛初乳乳清蛋白参与较多的是细胞外部分和细胞外空隙,与牛乳乳清蛋白相比参与的细胞组成大体相同。通过KEGG代谢通路分析可知,牛初乳和牛乳乳清蛋白均参与过补体及凝血级联反应通路。对牛初乳乳清蛋白组成进行研究,不仅能够增加牛初乳的利用率,并且为日后以乳清蛋白作为原料生产乳制品提供理论依据。 相似文献

利用高分辨质谱技术获取牛初乳、牛常乳、羊初乳和羊常乳的乳清蛋白组轮廓,基于其非标记定量强度建立偏最小二乘判别分析模型。结果发现牛初乳和牛常乳在蛋白质组成方面较羊初乳和羊常乳更相似,同时筛选出羊乳中丰度较高的9 种蛋白用作标志物区分这4 组乳样。生物信息学分析发现羊乳中的高丰度蛋白大部分与免疫应答和代谢过程有关,说明羊乳更有助于新生儿建立抗微生物感染的免疫系统。该研究可加深对羊乳蛋白的认识,对牛乳及其制品的营养改良和母乳替代品的生产具有重要意义。 相似文献

Johne's disease and bovine tuberculosis are diseases of economic, public health, and animal welfare importance. The single intradermal cervical comparative tuberculin (SICCT) test, which is used to determine bovine tuberculosis status as part of eradication schemes in the United Kingdom and some other countries, has been reported to interfere with the results of the widely used ELISA to detect antibodies against Mycobacterium avium ssp. paratuberculosis (MAP) in milk. Better understanding of the relationship between SICCT and MAP tests can improve management and control of Johne's disease. The aim of this study was to characterize the relationship between SICCT testing and milk ELISA performance and to assess whether the immunological response to the SICCT test is different for MAP-infected cows and noninfected cows. We used repeated MAP milk ELISA test results of a cohort of 805,561 cows in the United Kingdom between 2010 and 2018 that had milk ELISA tests within 90 d of SICCT testing to identify cows likely to be infected. We then assessed, separately, for cows deemed to be MAP-infected and noninfected, the association between MAP test results and proximity to SICCT testing by means of survival analysis and generalized additive mixed models. The results were used to quantify the effect SICCT testing may have on performance of milk ELISA tests conducted soon after SICCT testing. At high prevalence levels (20%) of MAP in the infected herd, overall accuracy of the milk ELISA is not reduced when testing occurs within 14 d from SICCT testing. Milk ELISA values of cows deemed to be infected were highest when MAP testing was closer in time to SICCT testing, suggesting the SICCT test enhances antibody response for MAP in infected cows. This corresponds to higher sensitivity of the MAP milk ELISA when testing within 30 d of the SICCT test. For cows deemed to be noninfected, the effect of previous SICCT testing was delayed compared with infected cows, with MAP milk ELISA values peaking at around 15 d post-SICCT testing. For both, MAP-infected and noninfected cows, interference from SICCT test diminished 30 d after SICCT testing, suggesting post 30 d to be the most appropriate time for evaluating the milk ELISA for MAP after SICCT testing. Our results provide strong evidence that the effect of the SICCT test on serological response against MAP is different for MAP-infected versus noninfected cows and that, as a result of this distinct effect, it is possible to improve interpretation of MAP milk ELISA test results (higher accuracy) by taking into consideration time since SICCT testing. 相似文献

The effects of milk products fermented by Bifidobacterium longum strain BL1, a probiotic strain, on blood lipids in rats and humans were studied. Rats were fed a cholesterol-enriched experimental diet, supplemented with lyophilized powders of 1) acid milk (control), 2) milk fermented with a mixed culture of ordinary yogurt starters composed of Streptococcus thermophilus and Lactobacillus delbrueckii subsp. bulgaricus (SL), and 3) bifidobacterium milk fermented with the probiotic B. longum strain BL1, respectively. The bifidobacterium milk feeding brought about significant lowering of the serum concentrations of total cholesterol, low-density lipoprotein cholesterol, and triglycerides, in comparison with the control, while no change in high-density lipoprotein cholesterol concentration was observed. On the other hand, supplementation with SL milk resulted in only slight, nonsignificant decreases in serum lipid concentrations in comparison with the control. In the human study, 32 subjects with serum total cholesterol ranging from 220 to 280 mg/dl were randomly assigned to two treatments: 1) intake of a low-fat drinking yogurt prepared with ordinary yogurt starters composed of S. thermophilus and L. delbrueckii subsp. bulgaricus (P-group) and 2) intake of a low-fat drinking yogurt prepared with the two ordinary yogurt starters plus B. longum strain BL1 (B-group). After intake for 4 wk at 3 x 100 ml/day, reduction of serum total cholesterol was observed in approximately half of the B-group subjects; a particularly significant decrease in serum total cholesterol was found among subjects with moderate hypercholesterolemia (serum total cholesterol > 240 mg/dl). However, the serum lipid concentrations in the P-group subjects were almost stable during the experimental periods. The present results indicate the potential of the probiotic B. longum strain BL1 in serum lipid improvement. 相似文献
