One hundred and twenty-seven lambs were fed as follows: lambs and dams grazing alfalfa (Gr); the same as Gr but lambs had access to concentrate (Gr + S); ewes grazed and lambs received milk and concentrate until weaning and thereafter concentrate and straw (Rat-Gr); ewes and lambs were stall-fed (Ind). Lambs were slaughtered at 22-24 kg live weight and fat and M. rectus abdominis colour was measured instrumentally and subjectively. The evolution of the instrumental colour and texture of M. longissimus lumborum was also recorded.There were significant differences in the instrumental colour of subcutaneous fat amongst feeding systems, but no differences were observed upon visual appraisal. On average, M. rectus abdominis colour in the four feeding systems was classed as pink. Differences in longissimus thoracis colour at 0 h disappeared at 24 h of air exposure and there were no differences in pH or cooking losses. In this study, polyphenol oxidase (PPO) was isolated from fresh lettuce. Its optimum temperature and pH were found to be 40 °C and 7.0, respectively. Lettuce PPO was shown to use catechin, catechol, chlorogenic acid, caffeic acid and gallic acid as substrates. Among the substrates used, the greatest substrate specificity was observed with chlorogenic acid. Lettuce PPO was sensitive to some inhibitors. Ascorbic acid, cysteine, oxalic acid and citric acid were tested as potential inhibitors of lettuce PPO. Cysteine was the most effective inhibitor. Total phenol and total antioxidant activity contents were also determined in the presence of these inhibitors at room and refrigerator temperatures. Ascorbic acid and cysteine increased the total antioxidant activity of lettuce while citric and oxalic acids had no effect on the total antioxidant activity. Thirty Holstein cows grazing mixed clover-grass pastures for 12 wk from May through July were fed a grain supplement containing solvent-extracted soybean meal (SES), or mechanically extracted soybean meal (MES) to determine whether differences in supplemental 18:2n6 fatty acid intake altered secretion of unsaturated fatty acids. Groups of 10 cows each were fed in two equal feedings a supplement (7.3 kg/d) containing ground corn plus either 1.8 kg of SES, 2.2 kg of MES, or 2.2 kg of MES plus 30 g of methionine hydroxy analog (Alimet; MESM). Fatty acid content (% of DM) of grass and clover in pastures averaged 1.9 and 1.5%, respectively. Concentration of 18:3n3 was higher in grass compared with clover (532 vs. 454 mg/g of total fatty acids). Yield of milk (32 kg/d average) and milk components did not differ by supplements. Total blood plasma fatty acids (mg/ml) during wk 4 were higher due to MESM (1.0) compared with MES (0.6) or SES (0.6). Cows fed MESM or MES had greater concentrations of 18:2n6, trans11-18:1, and cis9,trans11-18:2 in plasma compared with cows fed SES. The additional trans11-18:1 was found exclusively in plasma triglycerides, whereas the additional cis9,trans11-18:2 was found in plasma phospholipids and free fatty acids. Daily yields of 18:2n6, trans11-18:1 and cis9,trans11-18:2 in milk fat were greater for cows fed MES or MESM compared with SES. Results indicate yields of trans11-18:1, cis9, trans11-18:2, 18:2n6, and 18:3n3 in milk fat of pasture-fed cows were enhanced by feeding a grain supplement containing mechanically extracted, rather than solvent-extracted, soybean meal. 相似文献

Forty male Ile-de-France lambs (10 blocks of 4 homologous lambs) were used to study the effects of four feeding systems on muscle fatty acids (FA): raising and finishing on cool-season grasses (G), raising on the same grasses and stall-finishing, indoors, on concentrates and hay, respectively, for 22 (GSS) or 41 days (GSL), and stall-feeding indoors on concentrates and hay during both growing and finishing periods (S). Twenty-four lambs only (6 blocks) were retained for comparison of growth performances, lipid content in the longissimus thoracis muscle (LT) and their FA composition according to treatment. The 16 other lambs (4 blocks) were removed from the comparison, due to a large spread in the growth of the lambs towards the end of the trial. No significant effects of treatment were seen on the rate of growth (221, 228, 243 and 245±SE 8.0g/d, respectively, for G, GSS, GSL and S groups), and the lipid contents of the LT (2.22, 2.16, 2.17 and 2.52±SE 0.11g/100g fresh tissue). Grazing, lowered n-6 PUFA (polyunsaturated fatty acids), and increased n-3 PUFA and C18:2 c9t11 (conjugated linoleic acid cis9, trans11) compared to concentrate feeding. The main effects of grazing were not removed by a short period of finish indoors on concentrate (GSS group), but C20:4 n-6 and C22:6 n-3 contents achieved the lowest contents in this group, with significant differences from the values observed for GSL and S groups (C20:4 n-6) or from the three other groups (C22:6 n-3). After a longer period of finish on concentrate (GSL group), C18:3 n-3 (linolenic acid), C18:2 c9t11 and long chain (LC) n-3 PUFA were brought to the levels observed in the S group. In terms of adequacy for human health, the C18:2 n-6/C18:3 n-3 ratios were favourably low in the four groups (2.6, 3.6, 4.9 and 5.2±SE 0.7, respectively, for G, GSS, GSL and S groups), the level observed in the case of G group being significantly lower than for the three other groups and the level observed for GSS group being significantly lower than for the GSL and S groups. 相似文献

The impact of gamma-irradiation on 5′-nucleotides and on the free amino acids tyrosine and phenylalanine in fresh mushrooms was studied. After irradiation the samples were freeze-dried to avoid enzyme induced chemical changes. Three 5′-nucleotides could be detected using HPLC–UV and LC–ESI-MS: adenosine 5′-monophosphate (AMP), guanosine 5′-monophosphate (GMP) and guanosine 5′-diphosphate (GDP). Irradiation significantly reduced (p = 0.05) the GDP concentration (22%). AMP showed a marked reduction (46%) only at 5 kGy. GMP, tyrosine, and phenylalanine were not affected by gamma-irradiation. 相似文献

Consumer awareness, pesticide and fertiliser contaminations, and environmental concerns have resulted in significant demand for organically grown farm products. The present study evaluates the influence that organic and conventional farming practices exert on the total phenolic content in eggplant samples from two cultivars, Blackbell (American eggplant) and Millionaire (Japanese eggplant), grown by conventional and organic farming practices with similar climatic conditions. In addition, plant-to-plant variation in phenolic content was determined from eight eggplant samples collected from different plants grown at various field positions. Samples were separately assayed for phenolic content by the two independent procedures, HPLC/LC–MS and Folin–Ciocalteu assay. The results of the HPLC analysis showed significant plant-to-plant variation (% RSD ranged from 22% to 48%) in 5-caffeoylqunic acid content (the most abundant phenolic acid) in eight eggplant samples collected from eight different plants belonging to the same cultivar and grown under similar conditions. The cv. Millionaire showed a higher total phenolic content compared to the Blackbell cultivar. We did not observe significant consistent trend in the phenolic content of eggplant samples grown with organic and conventional farming practices with both eggplant cultivars. We, however, observed a strong correlation (R2 = 0.87) between total phenolic content as assayed by Folin–Ciocalteu assay and 5-caffeoylqunic acid, a major phenolic acid in eggplant extract. This study concludes that multiple repetitive analyses of plant products collected from different plants grown over various time periods (seasons) at different locations should always be carried to unambiguously prove the impact of growing conditions on phenolic content or antioxidant activity. 相似文献
