Salmonella contamination continues to be one of the major concerns for the microbiological safety of raw poultry products. Application of more than one decontamination agent as a multihurdle intervention to carcasses in a processing line might produce greater reductions than one treatment alone due to different modes of action of individual antimicrobials. In this study, all possible two-way combinations and individual applications of ε-polylysine (EPL), lauric arginate (LAE), and acidic calcium sulfate (ACS) solutions were evaluated for their effects against Salmonella enterica serovars, including Enteritidis and Typhimurium, using a sterile membrane filter model system. The combinations that provided higher Salmonella reductions were further evaluated on inoculated chicken carcasses in various concentrations applied in a sequential manner. Sequential spray applications of 300 mg of EPL per liter followed by 30% ACS and of 200 mg of LAE per liter followed by 30% ACS produced the highest Salmonella reductions on inoculated chicken carcasses, by 2.1 and 2.2 log CFU/ml, respectively. Our results indicated that these sequential spray applications of decontamination agents are effective for decreasing Salmonella contamination on poultry carcasses, but further studies are needed to determine the effectiveness of these combinations over a storage period.  相似文献

Listeria monocytogenes is a food-borne pathogen with the ability to grow at refrigeration temperatures. Knowledge of the mechanisms involved in low temperature growth is incomplete and here we report the results of a metabolomics investigation of this. The small molecule contents of L. monocytogenes 10403S grown at 37 °C and 8 °C were compared by gas chromatography/mass spectrometry (GC/MS). Over 500 peaks were detected in both 37 °C and 8 °C-grown cells, and 103 were identified. Of the identified metabolites, the concentrations of 56 metabolites were increased (P < 0.05), while the concentrations of 8 metabolites were decreased at low temperature. Metabolites increasing in concentration at 8 °C included amino acids, sugars, organic acids, urea cycle intermediates, polyamines, and different compatible solutes. A principal component analysis (PCA) was used to visualize and compare the matrix containing the data in 6 samples, and this clearly identified the 37 °C and 8 °C metabolomes as different. The results indicated that an increase in solute concentrations in the cytoplasm was associated with low temperature adaptation, which may be a response to chill stress with the effect of lowering the freezing point of intracellular water and decreasing ice crystal formation.  相似文献

The antimicrobial effect of four smoke wood condensates against the psychrotrophic pathogensListeria monocytogenes and Aeromonas hydrophila was tested in vacuum-packed cold-smoked rainbow trout (Oncorhynchus mykiss) stored at refrigeration temperature. Salted trout samples were surface inoculated with L. monocytogenes or A. hydrophila, cold smoked, vacuum packaged and stored at 4 °C for 21 days. At different times of storage, samples were taken and analysed for L. monocytogenes and A. hydrophila by quantitative and qualitative procedures. All smoke extracts showed activity against A. hydrophila. However, only smoke extracts L1 and L2 were effective against L. monocytogenes. Although these extracts caused a reduction in viable cell counts of L. monocytogenes below the detectable level by surface plating procedure, this organism was detected by qualitative analysis in most of the samples. Finally, extracts L1 and L2 also reduced total viable cell counts to undetectable levels.  相似文献

Nisin (500 IU ml−1), EDTA (0.02 M), potassium sorbate (PS) (3%, w/v), sodium benzoate (SB) (3%, w/v) or sodium diacetate (SD) (3%, w/v); alone or in combination were used to dip uninoculated shrimps and shrimps inoculated with Listeria monocytogenes or Salmonella (∼4.0–5.0 log CFU g−1). Shrimps were then drip-dried, vacuum packaged and stored at 4 °C for 7 days. Untreated shrimps were used as a control. Numbers of L. monocytogenes, Salmonella and native background microflora were determined on uninoculated and inoculated shrimps on days 0, 3 and 7. Nisin–EDTA–PS and nisin–EDTA–SD significantly reduced (p < 0.05) L. monocytogenes numbers by 1.07–1.27 and 1.32–1.36 log CFU g−1, respectively, on day 0 and 3. However, all treatments failed to significantly reduce (p > 0.05) Salmonella counts on shrimps throughout storage. On day 7, numbers of aerobic bacteria, psychrotrophic bacteria and Pseudomonas on combined nisin–EDTA–salt of organic acids treated shrimps were significantly lower (p < 0.05) by 4.40–4.60, 3.50–4.01, and 3.84–3.99 log CFU g−1 respectively, as compared to the control. Dipping in organic acids solutions followed by vacuum packaging and chilled storage can help reduce L. monocytogenes and native microflora, but not Salmonella, on fresh shrimps.  相似文献

The present study investigated in vitro antimicrobial activity of Eryngium caeruleum essential oil (EEO) against five foodborne pathogenic bacteria based on microdilution and disk diffusion methods. Moreover, its effects on specific spoilage microorganisms, inoculated Listeria monocytogenes, and its sensory changes in minced fish were evaluated during 12 days of storage at refrigeration temperature. The results showed that Staphylococcus aureus and Escherichia coli were the most sensitive and the most resistant bacteria with a minimum inhibitory concentration of 0.125 and 1 mg/ml, as well as inhibition zones of 15.66 and 11.66 mm, respectively. Regarding the antimicrobial effect of EEO on the microbial profile and inoculated L. monocytogenes, treating with 0.4% EEO caused a significant decrease in the studied microorganisms when compared to the control group (p < 0.05). In addition, considering the sensory evaluation, the best scores were observed for the samples treated with 0.2% and 0.4% EEO. However, none of the groups obtained acceptable scores until the final day of storage except for the color attribute. In general, sensory evaluation and its correlation with microbial counting indicated that the treatment with 0.4% EEO was able to preserve the microbial quality of the minced fish at refrigeration temperature without any undesirable sensory effects.  相似文献

A combination of a factorial design and two central composite designs was used to assess quantitatively the effects and interactions of water activity (1–0·95) and pH (5·6–9·5) variations on the growth of Listeria monocytogenes in a meat broth at 10°C. At inoculation or at the beginning of the exponential phase, the cells were exposed to the addition of NaCl and acetic acid or NaCl and NaOH. The effects of abrupt fluctuating conditions on the generation and lag times were analysed using turbidity measurements. The data indicated that the cells exposed to osmotic and acid or alkaline variable conditions from the time of inoculation were less affected than cells exposed at the beginning of the exponential phase. In this last case, a lag phase could be induced and the growth recovery was different from those expected in the new environment. Generation time values were estimated by three available predictive models which describe the effects of temperature, salt concentration and pH on L. monocytogenes growth to highlight the potential problems of variable conditions.  相似文献

The quality of smoked and marinated anchovy was investigated in terms of sensory, chemical [total volatile basic nitrogen (TVB-N), thiobarbituric acid (TBA), peroxide value (PV), fatty acids, free fatty acids (FFA), and pH] and microbiological parameters (total aerobic count (TVC), coliform, Escherichia coli, Salmonella, Staphylococcus aureus) during 7 months of storage. The results of sensory analyses showed that the total appearance, odor-taste, and texture scores decreased gradually during storage period. The results of the chemical analyses also correlated well with the sensory analyses apart from TVBN value. TBA significantly increased (p<0.05) from 1.9 to 4.25 MA/kg after the storage of 6 month. Peroxide value (POV) showed significant fluctuations (p<0.05) during the storage period. Initial TVC of 3.8 log CFU/g increased to 6.2 log CFU/g at the end of storage period. According to results obtained from sensory, chemical, and microbiological analyses, shelf life of this product was about 6 month.  相似文献

Growth and virulence of pathogenicYersinia enterocoliticawere investigated on high (pH>6.0) and normal (pH<5.8) pH pork packaged in modified atmospheres and stored at 4°C. Modified atmospheres used in the study were vacuum packaging and saturated CO2. Pork was packaged in a high gas barrier packaging film and examined over a 30-day period. Phenotypic characteristics were used to detect the presence of the virulence plasmid ofY. enterocoliticaafter exposure to the pork packaging and storage regimen. Phenotypic characteristics ofY. enterocoliticaisolates from pork loin stored at 4°C for 30 days that were studied included Congo red uptake, calcium dependence and autoagglutination in methyl red Voges–Proskauer broth and tissue culture medium. Numbers ofY. enterocoliticaon the lean surface of high pH pork slices increased approximately 2.7logcfucm−2when vacuum packaged and stored at 4°C for 30 days. Storage of inoculated normal pH pork in 100% CO2resulted inY. enterocoliticaremaining in the lag phase over the storage period. Virulence ofY. enterocoliticawas maintained in 25 to 35% of isolates following storage for 30 days at 4°C in vacuum- and CO2-packaged meats and was not affected by pH of the pork loin.  相似文献

Traditional aged Cheddar cheese does not support Listeria monocytogenes growth and, in fact, gradual inactivation of the organism occurs during storage due to intrinsic characteristics of Cheddar cheese, such as presence of starter cultures, salt content, and acidity. However, consuming high-salt (sodium) levels is a health concern and the dairy industry is responding by creating reduced-salt cheeses. The microbiological stability of low-salt cheese has not been well documented. This study examined the survival of L. monocytogenes in low-salt compared with regular-salt Cheddar cheese at 2 pH levels stored at 4, 10, and 21°C. Cheddar cheeses were formulated at 0.7% and 1.8% NaCl (wt/wt) with both low and high pH and aged for 10 wk, resulting in 4 treatments: 0.7% NaCl and pH 5.1 (low salt and low pH); 0.7% NaCl and pH 5.5 (low salt and high pH); 1.8% NaCl and pH 5.8 (standard salt and high pH); and 1.8% NaCl and pH 5.3 (standard salt and low pH). Each treatment was comminuted and inoculated with a 5-strain cocktail of L. monocytogenes at a target level of 3.5 log cfu/g, then divided and incubated at 4, 10, and 21°C. Survival or growth of L. monocytogenes was monitored for up to 90, 90, and 30 d, respectively. Listeria monocytogenes decreased by 0.14 to 1.48 log cfu/g in all treatments. At the end of incubation at a given temperature, no significant difference existed in L. monocytogenes survival between the low and standard salt treatments at either low or high pH. Listeria monocytogenes counts decreased gradually regardless of a continuous increase in pH (end pH of 5.3 to 6.9) of low-salt treatments at all study temperatures. This study demonstrated that post-aging inoculation of L. monocytogenes into low-salt (0.7%, wt/wt) Cheddar cheeses at an initial pH of 5.1 to 5.5 does not support growth at 4, 10, and 21°C up to 90, 90, and 30 d, respectively. As none of the treatments demonstrated more than a 1.5 log reduction in L. monocytogenes counts, the need for good sanitation practices to prevent post-manufacturing cross contamination remains.  相似文献

The effects of ozone in aqueous solution on the shelf life of whole, vacuum-packaged rainbow trout, stored under refrigeration (4±0.5°C) were studied by monitoring the microbiological, chemical and sensory changes for a period of 15 days. Vacuum-packaged non-ozonated trout served as the control sample. Ozonation affected populations of bacteria namely, mesophilic aerobic bacteria, Pseudomonas spp. and H2S-producing bacteria until day 11 of storage, Brochothrix thermosphacta, lactic acid bacteria and Enterobacteriaceae until day 8 of storage. Trimethylamine (TMA) values of all rainbow trout samples remained low (<3 mg N/100 g) until day 11 of storage, and then increased to 12.2, 8.9 and 4.7 mg N/100 g for the control and the samples ozonated for 60 and 90 min, respectively on day 15 of storage. Total volatile basic nitrogen (TVB-N) values remained relatively constant (20–25 mg N/100 g) until day 11 of storage, but increased to 61.1, 37.6 and 39.4 mg N/100 g flesh for the control and ozonated specimen for 60 and 90 min, respectively on day 15 of storage. Thiobarbituric acid (TBA) values remained relatively constant (1–3 mg MA/kg flesh) until day 12 of storage but increased to 8.4, 6.4 and 3.8 mg MA/kg flesh on day 15 of storage for the control and the ozonated trout for 60 and 90 min, respectively. Sensory evaluation (odor, taste and texture) of cooked rainbow trout showed a very good correlation with bacterial populations. On the basis of both sensory and microbiological data, a shelf life of 10 and 12 days was obtained for ozonated, vacuum-packaged and refrigerated rainbow trout at 60 and 90 min, respectively.  相似文献

Salmonella spp. are a leading cause of foodborne illness. Mathematical models that predict Salmonella survival and growth on food from a low initial dose, in response to storage and handling conditions, are valuable tools for helping assess and manage this public health risk. The objective of this study was to develop and to validate the first predictive microbiology model for survival and growth of a low initial dose of Salmonella on chicken during refrigerated storage. Chicken skin was inoculated with a low initial dose (0.9 log) of a multiple antibiotic-resistant strain of Salmonella Typhimurium DT104 (ATCC 700408) and then stored at 4 to 12 °C for 0 to 10 days. A general regression neural network (GRNN) model that predicted log change of Salmonella Typhimurium DT104 as a function of time and temperature was developed. Percentage of residuals in an acceptable prediction zone, from -1 (fail-safe) to 0.5 (fail-dangerous) log, was used to validate the GRNN model by using a criterion of 70% acceptable predictions. Survival but not growth of Salmonella Typhimurium DT104 was observed at 4 to 8 °C. Maximum growth of Salmonella Typhimurium DT104 during 10 days of storage was 0.7 log at 9 °C, 1.1 log at 10 °C, 1.8 log at 11 °C, and 2.9 log at 12 °C. Performance of the GRNN model for predicting dependent data (n=163) was 85% acceptable predictions, for predicting independent data for interpolation (n=77) was 84% acceptable predictions, and for predicting independent data for extrapolation (n=70) to Salmonella Kentucky was 87% acceptable predictions. Thus, the GRNN model provided valid predictions for survival and growth of Salmonella on chicken during refrigerated storage, and therefore the model can be used with confidence to help assess and manage this public health risk.  相似文献

The survival of Salmonella, Escherichia coli O157:H7, and Listeria monocytogenes was determined on almonds and pistachios held at typical storage temperatures. Almond kernels and inshell pistachios were inoculated with four- to six-strain cocktails of nalidixic acid-resistant Salmonella, E. coli O157:H7, or L. monocytogenes at 6 log CFU/g and then dried for 72 h. After drying, inoculated nuts were stored at -19, 4, or 24°C for up to 12 months. During the initial drying period after inoculation, levels of all pathogens declined by 1 to -log CFU/g on both almonds and pistachios. During storage, moisture content (4.8%) and water activity (0.4) of the almonds and pistachios were consistent at -19°C; increased slowly to 6% and 0.6, respectively, at 4°C; and fluctuated from 4 to 5% and 0.3 to 0.5 at 24°C, respectively. Every 1 or 2 months, levels of each pathogen were enumerated by plating; samples were enriched when levels fell below the limit of detection. No reduction in population level was observed at -19 or 4°C for either pathogen, with the exception of E. coli O157:H7-inoculated almonds stored at 4°C (decline of 0.09 log CFU/g/month). At 24°C, initial rates of decline were 0.20, 0.60, and 0.71 log CFU/g/month on almonds and 0.15, 0.35, and 0.86 log CFU/g/month on pistachios for Salmonella, E. coli O157:H7, and L. monocytogenes, respectively, but distinct tailing of the survival curves was noted for both E. coli O157:H7 and L. monocytogenes.  相似文献

The effects of dry-curing with low concentrations of salt and sugar on the quality of rainbow trout (Oncorhynchus mykiss) fillets were studied over a 16-day storage period at 4°C. Physicochemical characteristics (total volatile base nitrogen (TVB-N), biogenic amines (BAs), ATP-related compounds and K value), microbial populations (total aerobic counts (TAC), H2S-producing bacteria, Pseudomonas, Aeromonas, and lactic acid bacteria) and sensory (SA) indicators were analyzed. Fish samples were subjected to the following treatments: untreated (control), dry-cured with 1.3% salt (T1), and dry-cured with 1.3% salt that also contained 0.9% sugar (T2). SA indicated that the sensory quality of the control, T1, and T2 was unacceptable after 8, 12, and 12 days of storage, respectively. Moreover, the cadaverine concentration of treated samples was significantly lower (P < 0.05) than the control after storing for 8 days. T1 and T2 were significantly higher (P < 0.05) than the control in inosine 5-monophosphate (IMP) content from the 2th to 10th day. Cadaverine, putrescine, hypoxanthine, and TAC were correlated significantly (P < 0.01) with SA, and these seemed to be reliable indicators of fish quality. The results indicated that curing treatments extended the shelf life of rainbow trout fillets, which could be exploited by processors.  相似文献

The effect of rosemary extract, chitosan and α-tocopherol, added individually or in combination, on microbiological parameters and lipid oxidation of fresh pork sausages stored for 20 days at 4 °C was investigated. Microbiological determinations included the enumeration of Enterobacteriaceae, Pseudomonas spp., total viable count, yeasts and moulds and lactic acid bacteria, while pH values were also determined. Lipid oxidation was evaluated through measurement of primary (peroxide value) and secondary (malondialdehyde concentration) oxidation products. Chitosan addition resulted in significant (P ? 0.05) inhibition of microbial growth, while the lowest microbial counts were obtained in the samples containing both chitosan and rosemary, indicating a possible synergistic effect. Chitosan and its combinations with either α-tocopherol or rosemary, and especially the latter combination, showed also the most intense antioxidative effect (P ? 0.05), at least when compared to α-tocopherol alone or the controls, while use of rosemary extract alone had a comparable (P > 0.05) antioxidative effect with chitosan and its combinations. Shelf life of samples containing chitosan was almost doubled compared to the remaining samples. In conclusion, the best antimicrobial and antioxidative effects were obtained from the combination of chitosan with the rosemary extract.  相似文献

The effects of curdlan (2%, 3% and 4%) on the gel properties of Alaska surimi using high-temperature treatment were examined in this study. Curdlan treatment improved the gel strength of surimi in a concentration-dependent manner. Based on the results of dynamic rheology and differential scanning calorimetry (DSC), curdlan promoted the stability of proteins in the gel. Moreover, curdlan facilitated the interaction between gel proteins and actomyosin, thereby preventing the aggregation and denaturation of protein and increased the thermal transition temperature. The results of scanning electron microscopy showed that the addition of curdlan induced the formation of a more ordered and denser gel matrix and the fibrils in the three-dimensional network became more delicate. Therefore, surimi samples with curdlan may hold more moisture and exhibit improved transfer of free water to bound water, leading to a higher water-holding capacity (WHC).  相似文献

The effects of turmeric extract (T), shallot extract (Sh), and their combination (T + Sh) on the quality of vacuum-packaged rainbow trout (Oncorhynchus mykiss) were examined during refrigerated storage (4 ± 1 °C) over a period of 20 d. Fish samples were divided into 4 batches; 3 batches were treated by dipping for 30 min in aqueous solution of turmeric extract (1.5%; v/v), shallot extract (1.5%; v/v), or turmeric and shallot extract combination (1.5%+ 1.5% v/v), while the fourth batch was dipped in distilled water as a control sample. The control and the treated fish samples were analyzed periodically for microbiological (total viable count, psychrotrophic count), chemical (total volatile base nitrogen [TVB-N], peroxide value [PV], and thiobarbituric acid [TBA] value), and sensory characteristics. The results indicated that the effect of the T, Sh, and T + Sh on the fish samples were to enable the good quality characteristics to be retained longer and to extend the shelf life during the refrigerated storage.  相似文献
